The transport of dissolved organic carbon (DOC) from the soils to inland waters plays an important role in the global carbon cycle. Widespread increases in DOC concentrations have been observed in surface waters over the last few decades, affecting carbon balances, ecosystem functioning and drinking water treatment. However, the primary hydrological controls on DOC mobilization are still uncertain. The aim of this study was to investigate the role of microtopography in the riparian zone for DOC export and DOM quality. DOC concentration and DOM quality in the shallow groundwater of a riparian zone and in streamflow in a forested headwater catchment was investigated using fluorescence and absorbance characteristics. We found higher DOC concentrations with a higher aromaticity in the microtopographical depressions, which were influenced by highly dynamic shallow groundwater levels, than in the flat forest soil. As a result of the frequent wet-dry cycles in the upper soil layers, aromatic DOC accumulated in the shallow groundwater within and below the microtopographical depressions. Rising groundwater levels during precipitation events led to the connection of the microtopographical depressions to the stream, resulting in a change toward more aromatic DOC in the stream. Increasing stream DOC concentrations were accompanied by increasing concentrations of iron and aluminum, suggesting the coupled release of these metals with DOC from the riparian zone. Our results highlight the importance of the interplay between microtopography and groundwater level dynamics in the riparian zone for DOC export from headwater catchments.

Dissolved organic matter (DOM) composition in streams reflects the dynamic interplay between DOM sources, mobilization mechanisms, and biogeochemical transformations within soils and receiving water bodies. The information regarding DOM sources being mobilized during baseflow can improve our ability to predict hydrological and biogeochemical responses to environmental changes, with implications for catchment management strategies. The objective of this study was to characterize the spatial changes in DOM composition along a headwater stream during baseflow and to link the findings in-stream with possible DOM sources in the catchment. DOM was monitored over 1.5 years at three sites in the Große Ohe catchment (19.2 km2), within the Bavarian Forest National Park, using UV-Vis absorption indicators of aromaticity (SUVA) and molecular weight (E2:E3) from high-frequency probe measurements. Additionally, discrete samples were collected and analyzed by ultrahigh-resolution Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR-MS). At baseflow conditions, DOC concentrations – a proxy of DOM amount – ranged from 1.5 to 4.7 mg L-1 and were similar along the slope gradient. However, DOM quality exhibited clear spatial patterns, with overall high aromatic and low molecular weight DOM in the lower part of the catchment. Moreover, molecular data revealed that oxygen-rich, aromatic compounds increased their abundance at high DOC concentrations in both upper and lower part of the catchment, with also additional input of oxygen-depleted, aromatic compounds identified in the lower part. In contrast, nitrogen-rich, aliphatic compounds were negatively correlated with DOC concentration, indicating a higher contribution of deep groundwater flows.

Understanding the controls on event-driven dissolved organic carbon (DOC) export is crucial as DOC is an important link between the terrestrial and the aquatic carbon cycles. We hypothesized that topography is a key driver of DOC export in headwater catchments because it influences hydrological connectivity, which can inhibit or facilitate DOC mobilization. To test this hypothesis, we studied the mechanisms controlling DOC mobilization and export in the Große Ohe catchment, a forested headwater in a mid-elevation mountainous region in southeastern Germany. Discharge and stream DOC concentrations were measured at an interval of 15 min using in situ UV-Vis (ultraviolet–visible) spectrometry from June 2018 until October 2020 at two topographically contrasting subcatchments of the same stream. At the upper location (888 m above sea level, a.s.l.), the stream drains steep hillslopes, whereas, at the lower location (771 m a.s.l.), it drains a larger area, including a flat and wide riparian zone. We focus on four events with contrasting antecedent wetness conditions and event size. During the events, in-stream DOC concentrations increased up to 19 mg L−1 in comparison to 2–3 mg L−1 during baseflow. The concentration–discharge relationships exhibited pronounced but almost exclusively counterclockwise hysteresis loops which were generally wider in the lower catchment than in the upper catchment due to a delayed DOC mobilization in the flat riparian zone. The riparian zone released considerable amounts of DOC, which led to a DOC load up to 7.4 kg h−1. The DOC load increased with the total catchment wetness. We found a disproportionally high contribution to the total DOC export of the upper catchment during events following a long dry period. We attribute this to the low hydrological connectivity in the lower catchment during drought, which inhibited DOC mobilization, especially at the beginning of the events. BACKGROUND Diffuse lower WHO grade II and III gliomas (LGG) are slowly progressing brain tumors, many of which eventually transform into a more aggressive type. LGG is characterized by widespread genetic and transcriptional heterogeneity, yet little is known about the heterogeneity of the DNA methylome, its function in tumor biology, coupling with the transcriptome and tumor microenvironment and its possible impact for tumor development. METHODS We here present novel DNA methylation data of an LGG-cohort collected in the German Glioma Network containing about 85% isocitrate dehydrogenase (IDH) mutated tumors and performed a combined bioinformatics analysis using patient-matched genome and transcriptome data. RESULTS Stratification of LGG based on gene expression and DNA-methylation provided four consensus subtypes. We characterized them in terms of genetic alterations, functional context, cellular composition, tumor microenvironment and their possible impact for treatment resistance and prognosis. Glioma with astrocytoma-resembling phenotypes constitute the largest fraction of nearly 60%. They revealed largest diversity and were divided into four expression and three methylation groups which only partly match each other thus reflecting largely decoupled expression and methylation patterns. We identified a novel G-protein coupled receptor and a cancer-related 'keratinization' methylation signature in in addition to the glioma-CpG island methylator phenotype (G-CIMP) signature. These different signatures overlap and combine in various ways giving rise to diverse methylation and expression patterns that shape the glioma phenotypes. The decrease of global methylation in astrocytoma-like LGG associates with higher WHO grade, age at diagnosis and inferior prognosis. Improving the understanding of the controls on subsurface stormflow generation has been the goal of numerous experimental and modeling studies. However, the effect of the spatial variability of throughfall on soil moisture patterns and subsurface stormflow (SSF) generation has not yet been studied in detail. The objectives of this study are threefold: (1) to investigate the influence of a spatially variable throughfall pattern on soil moisture; (2) to investigate if soil moisture patterns reflect a balance between a throughfall and bedrock topography patterns; and (3) to investigate how this balance changes when soil depth, storm size and slope angle are varied. Virtual experiments are used to address these questions. A virtual experiment is a numerical experiment driven by collective field intelligence. It provides a learning tool to investigate the effect of individual processes in a complex system. In our virtual experiment we combined spatial throughfall data from the Huewelerbach catchment in Luxembourg with the topography of a well-studied hillslope within the Panola Mountain Research Watershed, Georgia, USA. We used HYDRUS-3D as a modeling platform. The virtual experiment shows that throughfall patterns influence soil moisture patterns, but only during and shortly after a storm. With a semi-variogram analysis we showed how the effective range of the soil moisture pattern (i.e., the main descriptor of a spatial pattern in case of a small nugget to sill ratio), is similar to the effective range of the throughfall pattern during the storm and gradually returns to the effective range of the bedrock topography after throughfall has ceased. The same analysis was carried out to investigate how this balance changes due to changes in storm size, soil depth and slope. The analysis showed that the throughfall pattern is more important during large storms on gentle slopes. BACKGROUND: Outcome after renal transplantation in children has been variable. We undertook a retrospective study of our experience over the past five years. STUDY DESIGN: From January 1, 1988, to October 15, 1992, 60 renal transplantations were performed upon 59 children at the Children's Hospital of Pittsburgh. Twenty-eight (47 percent) of the kidneys were from cadaveric donors, and 32 (53 percent) were from living donors. The recipients ranged in age from 0.8 to 17.4 years, with a mean of 9.8 ± 4.8 years. Forty-six (77 percent) recipients were undergoing a first transplant, while 14 (23 percent) received a second or third transplant. Eight (13 percent) of the patients were sensitized, with a panel reactive antibody of more than 40 percent. Eleven of the 14 patients undergoing retransplantation and seven of the eight patients who were sensitized received kidneys from cadaveric donors. Thirty- three (55 percent) patients received cyclosporine-based immunosuppression, and 27 (45 percent) received FK506 as the primary immunosuppressive agent. RESULTS: The median follow-up period was 36 months, with a range of six to 63 months. The one- and four-year actuarial patient survival rate was 100 and 98 percent. The one- and four-year actuarial graft survival rate was 98 and 83 percent. For living donor recipients, the one- and four-year actuarial patient survival rate was 100 and 100 percent; for cadaveric recipients, it was 100 and 96 percent. Corresponding one- and four-year actuarial graft survival rates were 100 and 95 percent for the living donor recipients and 96 and 69 percent for the cadaveric recipients. Patients on cyclosporine had a one- and four-year patient survival rate of 100 and 97 percent, and patients on FK506 had a one- and three-year patient survival rate of 100 and 100 percent.
