Objective: The ongoing outbreak of the respiratory disease coronavirus disease 2019 (COVID‐19) is currently presenting a major global health threat. This pandemic is unprecedented in recent human history. The objective of this study was to examine the relationship between cycle quantitation (Cq) and laboratory parameters in COVID‐19 patients, aiming to determine if Cq levels can provide valuable insights into the COVID‐19 disease. Methods: This study involved 234 participants who were divided into case and control groups. Real‐time PCR tests were used to diagnose COVID‐19 cases in the study participants. Blood tests, including complete blood count, C‐reactive protein (CRP), erythrocyte sedimentation rate (ESR), lactate dehydrogenase (LDH), D‐dimer, IgG, and IgM, were also conducted. Statistical analysis was performed using SPSS 22 software. Results: The findings showed that COVID‐19‐positive cases had significantly higher levels of the neutrophil‐to‐lymphocyte ratio (NLR), platelet‐to‐lymphocyte ratio (PLR), D‐dimer, ESR, CRP, and LDH compared to normal cases. Additionally, the case group had significantly lower lymphocyte and platelet counts. There was a statistically significant positive correlation between Cq levels and lymphocyte count (r =.124, p =.014). Conversely, there was a statistically significant inverse correlation between Cq levels and NLR (r = −.208, p =.017). Furthermore, the evaluation of hematological, inflammatory, and biochemical indexes in COVID‐19 patients using the receiver‐operating characteristics curve demonstrated statistically appropriate sensitivity and specificity. Conclusion: Our outcomes indicated a significant association between Cq levels and PLR, NLR, D‐dimer, CRP, and ESR in COVID‐19 patients. Consequently, including the report of laboratory parameters alongside Cq values offers a promising prognosis. [ABSTRACT FROM AUTHOR]

Cancer is the second most common cause of death globally and is a major public health concern. Managing this disease is difficult due to its multiple stages and numerous genetic and epigenetic changes. Traditional cancer diagnosis and treatment methods have limitations, making it crucial to develop new modalities to combat the increasing burden of cancer. The clustered regularly interspaced short palindromic repeats (CRISPR)‐CRISPR‐associated protein 9 (Cas9) system has transformed genetic engineering due to its simplicity, specificity, low cytotoxicity, and cost‐effectiveness. It has been proposed as an effective technology to enhance cancer diagnosis and treatment strategies. This article presents the most recent discoveries regarding the structure, mechanism, and delivery methods of the highly powerful genome editing tool, CRISPR‐Cas9. In terms of diagnosis, the article examines the role of CRISPR‐Cas9 in detecting microRNAs and DNA methylation, and discusses two popular gene detection techniques that utilize the CRISPR‐Cas system: DNA endonuclease‐targeted CRISPR trans reporter and specific high sensitivity enzymatic reporter unlocking. Regarding treatment, the article explores several genes that have been identified and modified by CRISPR‐Cas9 for effective tumorigenesis of common cancers such as breast, lung, and colorectal cancer. The present review also addresses the challenges and ethical issues associated with using CRISPR‐Cas9 as a diagnostic and therapeutic tool. Despite some limitations, CRISPR‐Cas9‐based cancer diagnosis has the potential to become the next generation of cancer diagnostic tools, and the continuous progress of CRISPR‐Cas9 can greatly aid in cancer treatment. The problems of drug resistance in bacteria have become one of the daily challenges of the clinical treatment of patients, which inevitably forces us to use agents other than common antibiotics. Among these, we can take help from different properties and applications of nanoparticles (NPs). In this work, we evaluate the antibacterial activity of biosynthesized selenium nanoparticles (SeNPs) against standard strains of multidrug‐resistant Pseudomonas aeruginosa and Acinetobacter baumannii. The production of biosynthesized SeNPs was proved by ultraviolet‐visible, Fourier transform infrared, X‐ray diffractometer, Field Emission Scanning Electron Microscopy, Dynamic light scattering, and Zeta potential methods. The cytotoxicity effect of SeNPs was investigated by MTT assay. Disk diffusion agar (DDA) and minimum inhibitory concentration (MIC) tests were performed on the mentioned bacteria using different classes of standard antibiotics and SeNPs separately. The impact of SeNPs combined with the desired antibiotics for better treatment of these infections was evaluated by checkerboard assay to determine the synergism effect. After the confirmation results based on the biosynthesis of SeNPs, both standard bacterial strains were susceptible to SeNPs and had a zone of inhibition using the DDA test. Also, the results of MICs showed that biosynthesized SeNPs in lower concentrations than antibiotics cause no growth of bacteria. On the other hand, according to the checkerboard assay, SeNPs had a synergistic effect with conventional antibiotics. The antibacterial sensitivity tests demonstrated the inhibition of bacterial growth in the presence of lower concentrations of SeNPs than common antibiotics. This property can be exerted in future applications to solve the drug resistance obstacle of microorganisms in bacterial diseases. [ABSTRACT FROM AUTHOR]

In this project, we employed ethanolic (EMI) and aqueous (AMI) extracts of mango (Mangifera indica L., Anacardiaceae) fruit seeds as a modulator of antibiotic resistance against multidrug-resistant (MDR) Pseudomonas aeruginosa and Acinetobacter baumannii to evaluate natural compounds isolated from by-products or waste of edible plants. We also investigated the effect of these extracts alone and in combination with standard classes of antibiotics in the desired strains. M. indica seeds were processed and exploited using ethanol and water. The minimum inhibitory concentrations (MICs) of clinical isolates were examined against EMI and AMI extracts, followed by seven antibiotics of ceftazidime, ciprofloxacin, penicillin, amikacin, meropenem, ampicillin, and colistin. The checkerboard method evaluated the synergistic action between mango kernel extract (EMI) and seven antibiotics. EMI extract significantly revealed antimicrobial properties against MDR A. baumannii and P. aeruginosa with synergistic effects with the applied antibiotics. The considerable antibacterial efficacy of ethanolic extract of M. indica seeds can have great curative value as antibacterial drugs against infections caused by MDR P.aeruginosa and A. baumannii. The biosynthesis of nanoparticles stands as an uncomplicated, biocompatible, inexpensive, and secure approach for replacing the available chemical and physical methods. In this paper, Selenium nanoparticles (Se-NPs) and silver nanoparticles (Ag-NPs) were biosynthesized using Nepeta and Berberine plant extracts, respectively. UV–Vis, FT-IR, and FESEM/EDAX techniques were applied for confirming the synthesis of nanoparticles. In addition, the method of Minimum Inhibitory Concentration (MIC) was utilized to perform a sensitivity test on fungal species' leachates through the usage of Duplicate. The application biosynthesized Se-NPs and Ag-NPs at concentrations of 1 µg/mL and below caused no growth of fungal agents. However, antifungal drugs at concentrations higher than 1 µg/mL and even up to 64 µg/mL inhibited the growth of these drug-resistant fungi. A comparison of the results showed that even these nanoparticles in some cases have a better effect than the group of antifungal drugs. Therefore, the usage of biosynthesized nanoparticles is considered to be more convenient and economical due to the increasing drug resistance of opportunistic and pathogenic microorganisms towards the exerted target drugs.
