Hot spring environments encompass broad physicochemical ranges, in which temperature and pH account for crucial factors shaping hot spring microbial community and diversity. However, the presence of photosynthetic microbial mats adjacent to boiling hot spring vents, where fluid temperatures extend beyond photosynthetic capability, questions the microbial profiles and the actual temperatures of such adjacent mats. Therefore, this study aims to characterize thermophilic microbial communities at Pong Dueat Pa Pae hot spring using next-generation sequencing, including investigating hot spring mineralogy. Results suggest that Pong Dueat Pa Pae hot spring precipitates comprise mainly silica which also acts as the main preservative medium for microbial permineralization. Molecular results revealed the presence of cyanobacterial and Chloroflexi species in the thick, orange and green subaerial mats surrounding the vents, suggesting the mats would be at least 30 °C cooler than source vents despite constantly receiving geyser splashes. Bacterial abundance was considerably higher than archaeal (97.9% versus 2.1%). Cyanobacterial (mainly Synechococcus and Leptolygbya) and Chloroflexi species (mainly Roseiflexus) accounted for almost half (40.04%) of the bacterial community, while DHVEG-6 and Thaumarchaeota comprised dominant members (> 90%) of the archaeal fraction. This research aimed to: 1) gather the needs of Thai and foreign tourists in using the Decision Support System for Hot Springs Tourism in Surat Thani; 2) Design and develop the Decision Support System using the concept of an agile method; and 3) Evaluate the effectiveness of the Decision Support System implementation. The agile method enabled the System to be developed to support both the Android and IOS in Thai and English. The users’ requirement was collected by using questionnaires with 30 Thai and 30 foreign tourists. The users’ evaluation data was collected by using questionnaires with 400 Thai and 400 foreign tourists. The results found that: 1) Thai and foreign tourists need information in seven aspects. Mo st tourists use iPhones and need similar information. 2) The design of the Decision Support System consists of searching travel routes, distance calculation, nearby attractions, and evaluation assessment function. 3) Foreign tourists had a slightly high er average score of overall opinions than Thai tourists which is at the highest level in all aspects. The Thai and foreign tourists found no significant difference with the overall opinions in using the Decision Support System (p> .05). In conclusion, the DSS for Hot Springs Tourism in Surat Thani was successfully developed for both Thai and foreign tourists. The findings indicate the system's effectiveness in supporting tourists and its potential to enhance tourism experiences in Surat Thani.

Sodium hyaluronate acid (HA) comprises N-acetylglucosamine and D-glucuronic acid, and distinguishes itself with superior qualities in skin regeneration, providing moisturizing and anti-aging benefits. The combination of HA with HS water is widely applied across ophthalmology, pneumology, nutrition, and cosmetics. This study delved into the application of HA in cosmetology, with a focus on its interaction with HS water and its effects on moisture retention and promoting wound healing. In particular, with the alkaline pH levels of the Yumoto HS, HA molecules may undergo dissociation to be ionized resulting in a negatively charged polymer and interacting with positively charged ions in the HS water through electrostatic interactions. The shifted peaks in the FTIR result and zeta potential shifts to a less negative region in the case of HA-HS compared to HA-DI indicate an ionic interaction between HS water and HA. Moisture tests confirmed the sustained hydration when HA is dissolved in HS water, underscoring its potential to improve skin hydration at certain concentrations, specifically at 0.5% and 1%. Additionally, MTT assay results demonstrated that HS water stimulates the growth of fibroblast cells compared to distilled water, implying its potential beneficial effect in wound healing. These findings suggested the multifaceted benefits of HAHS in skincare, highlighting its role in enhancing skin hydration and potentially accelerating wound healing processes, thus presenting avenues for the development of advanced cosmeceutical formulations.

Geothermal features, such as hot springs and mud volcanoes, host diverse microbial life, including many extremophile organisms. The physicochemical parameters of the geothermal feature, such as temperature, pH, and heavy metal concentration, can influence the alpha and beta diversity of microbial life in these environments, as can spatiotemporal differences between sites and sampling. In this study, water and sediment samples were collected and analyzed from eight geothermal sites at Yellowstone National Park, including six hot springs, a mud volcano, and an acidic lake within the same week in July 2019, and these geothermal sites varied greatly in their temperature, pH, and chemical composition. All samples were processed and analyzed with the same methodology and taxonomic profiles and alpha and beta diversity metrics determined with 16S rRNA sequencing. These microbial diversity results were then analyzed with respect to pH, temperature, and chemical composition of the geothermal features. Results indicated that predominant microbial species varied greatly depending on the physicochemical composition of the geothermal site, with decreases in pH and increases in dissolved heavy metals in the water corresponding to decreases in alpha diversity, especially in the sediment samples. Similarly, sites with acidic pH values had more similar microbial populations (beta diversity) to one another than to relatively neutral or alkaline pH geothermal sites. This study suggests that pH and/or heavy metal concentration is a more important driver for microbial diversity and population profile than the temperature for these sites and is also the first reported microbial diversity study for multiple geothermal sites in Yellowstone National Park, including the relatively new mud volcano Black Dragon’s Caldron, which erupted in 1948. Northeastern Algeria boasts numerous hot springs, yet these hydrothermal sites remain largely unexplored for their microbial ecology. The present study explores the bacterial abundance and diversity within two distinct Algerian hot springs (Hammam Saïda and Hammam Debagh) and investigates the link between the prevailing bacteria with geochemical parameters. High-throughput 16S rRNA gene sequencing of water and sediment samples revealed a bacterial dominance of 99.85–91.16% compared to Archaea (0.14–0.66%) in both springs. Interestingly, Saïda hot spring, characterized by higher temperatures and sodium content, harbored a community dominated by Pseudomonadota (51.13%), whereas Debagh, a Ca–Cl-SO4 type spring, was primarily populated by Bacillota with 55.33%. Bacteroidota displayed even distribution across both sites. Additional phyla, including Chloroflexota, Deinococcota, Cyanobacteriota, and Chlorobiota, were also present. Environmental factors, particularly temperature, sodium, potassium, and alkalinity, significantly influenced bacterial diversity and composition. These findings shed light on the interplay between distinct microbial communities and their associated geochemical properties, providing valuable insights for future research on biogeochemical processes in these unique ecosystems driven by distinct environmental conditions, including potential applications in bioremediation and enzyme discovery. This study explores the potential of machine learning algorithms for earthquake prediction, utilizing fluid chemical anomaly data from hot springs. Six hot springs, located within an active fault zone along the southeastern coast of China, were carefully chosen as hydrochemical monitoring sites for an extended period of two and a half years. Using this data, a prediction model integrating six algorithms was developed to forecast M ≥ 5 earthquakes in Taiwan. The model's performance was validated against recorded earthquake events, and the factors influencing its predictive capability were analyzed. Our comprehensive analysis conclusively demonstrates the superiority of machine learning algorithms over traditional statistical methods for earthquake prediction. Additionally, including sampling time in the data sets significantly improves the model's predictive performance. However, it is important to note that the model's predictive performance varies across different hot spring and indicators type, highlighting the importance of identifying optimal indicators for specific scenarios. The model parameters, including the anomaly detection rate (P) and earthquake response time threshold (M), significantly impact the model's predictive capabilities. Therefore, adjustments are needed to optimize the model's performance for practical use. Despite limitations such as the inability to differentiate pre‐earthquake anomalies from post‐earthquake anomalies and pinpoint the precise location of earthquakes, this study successfully showcases the potential of machine learning algorithms in earthquake prediction, paving the way for further research and improved prediction methods. This investigation aimed to identify the bioremediation potential of Mn2+-resistant bacterial strains cultured from the Surya Kund hot spring, Yamunotri, Uttarakhand. In this study, eight heavy metal-resistant isolates belonging to two phyla, i.e., Firmicutes and Proteobacteria, were investigated for their Mn2+biosorption potential. The metal tolerance potential of all the thermophilic bacterial strains was determined by MIC. Bioremediation assay of these metal-resistant strains was performed for Mn2+ through the live and dead biomass of the bacterial cell. The evaluation of the bioremediation rate of metal ions through bacteria was done by AAS. All the selected bacterial strains were evaluated with effective biosorption rates for Mn2+. Acinetobacter sp. LSN-10 (YII-1) has been showing the highest potential for the removal of Mn2+ in both live (41.202%) and dead biomass (64.721%) conditions. The bioremediation rate of dead biomass was observed quite higher in comparison to bioremediation through live bacterial cells in the maximum number of isolates. This study may provide a new eco-friendly and cost-effective approach to dealing with metal toxicity. Background: At present, research on facile, green synthesis of nanoparticles has significantly increased because of its fast, one-step, cost-effective, time-efficient, and non-toxic nature. In this study, we have reported a single-step green synthesis of copper nanoparticles using cell wall polysaccharides of a hot spring origin, thermotolerant Bacillus species. Result: Copper nanoparticles were characterized using UV-visible spectrophotometry, fluorescence and Fourier transform infrared spectroscopy, scanning electron microscopy with energy dispersive spectroscopy, particle size, and zeta potential analyses. UV-visible spectra of synthesized copper nanoparticles exhibited a band cantered between 220–235 nm, characteristic spectra of copper oxide nanoparticles. Infrared spectra showed the band at 490-530 cm−1 corresponding to metal-oxygen or copper nanoparticle vibration, supporting the presence of copper oxide nanoparticles in the monoclinic phase. The energy dispersive spectra of copper nanoparticles exhibited a strong signal from elemental copper. The dynamic Light Scattering pattern confirmed the nanoparticle nature of the studied sample. These nanoparticles showed preferential activity against gram-negative pathogens, Salmonella typhi and Escherichia coli. The thermodynamic nature of the nanoparticles is also established for its antibacterial actions. Conclusions: The antibacterial action and its thermodynamics reinforce the possible use of copper nanoparticles as an alternative to commercially available antimicrobials. Abstract A type of high molecular weight bioactive polymers called exopolysaccharides (EPS) are produced by thermophiles, the extremophilic microbes that thrive in acidic environmental conditions of hot springs with excessively warm temperatures. Over time, EPS became important as natural biotechnological additives because of their noncytotoxic, emulsifying, antioxidant, or immunostimulant activities. In this article, we unravelled a new EPS produced by Staphylococcus sp. BSP3 from an acidic (pH 6.03) San Pedro hot spring (38.1 °C) located in the central Andean mountains in Chile. Several physicochemical techniques were performed to characterize the EPS structure including Scanning electron microscopy–energy dispersive X-ray spectroscopy (SEM–EDS), Atomic Force Microscopy (AFM), High-Performance Liquid Chromatography (HPLC), Gel permeation chromatography (GPC), Fourier Transform Infrared Spectroscopy (FTIR), 1D Nuclear Magnetic Resonance (NMR), and Thermogravimetric analysis (TGA). It was confirmed that the amorphous surface of the BSP3 EPS, composed of rough pillar-like nanostructures, is evenly distributed. The main EPS monosaccharide constituents were mannose (72%), glucose (24%) and galactose (4%). Also, it is a medium molecular weight (43.7 kDa) heteropolysaccharide. NMR spectroscopy demonstrated the presence of a [→ 6)-⍺-d-Manp-(1 → 6)-⍺-d-Manp-(1 →] backbone 2-O substituted with 1-⍺-d-Manp. A high thermal stability of EPS (287 °C) was confirmed by TGA analysis. Emulsification, antioxidant, flocculation, water-holding (WHC), and oil-holding (OHC) capacities are also studied for biotechnological industry applications. The results demonstrated that BSP3 EPS could be used as a biodegradable material for different purposes, like flocculation and natural additives in product formulation. Graphical Abstract

Hot springs are known to harbor potentially unique microorganisms due to the extreme temperatures in which they thrive and their biotechnologically important enzymes that are active at high temperature, which are beneficial for various industries. Sabah, Malaysia, houses several hot springs, yet knowledge of their microbiological diversity remains limited. Here, the raw sequence data of bacterial communities in a hot spring through metagenomic analysis are revealed. The data were obtained by collecting water and sediment samples from Poring Hot Spring (PHS) in Ranau, Sabah, and their bacterial diversity was analyzed using 16S rRNA amplicon sequencing targeting the V3-V4 regions. The analysis identified bacterial diversity in both water and sediment samples, with 35 phyla, 76 families, and 90 genera in water, and 38 phyla, 114 families, and 128 genera in sediment. Proteobacteria dominated the water samples (87 %), while Cyanobacteria were most abundant in sediment samples (51 %). The most abundant genera in water were Tepidimonas, Hydrogenophilus and Methylothermus, whereas Geitlerinema, Calothrix and Nitrospira dominated the sediment. Sediment samples exhibited higher bacterial richness and diversity compared to water samples, as indicated by α-diversity analysis. This paper aims to update our understanding of the carbon cycle in the Himalayas, the most intense collisional orogeny globally, by providing new insight into its impact on Cenozoic climate cooling through the use of isotopic variations in both organic and inorganic carbon and an isotopic mass balance model. Our results from 20 selected hot springs show that the relative contributions of dissolved carbon from the mantle, metamorphic decarbonization, aqueous dissolution, and soil organic matter are approximately 2%, 82%, 6%, and 10%, respectively. Approximately 87% ± 5% of CO2 generated in the deep crust precipitates as calcite, while approximately 5.5% ± 1% of this carbon is converted to biomass through microbial chemosynthesis at depths less than 2 km. Our random forests approach yielded a metamorphic carbon flux from the entire Himalayan orogenic belt of approximately 2.7 ∼ 4.5 × 1012 mol/yr. The minor CO2 released into the atmosphere (2.5 ∼ 4.2 × 1011 mol/yr) is comparable to the carbon consumption driven by Himalayan weathering. This paper provides new insights into deep carbon cycling, notably that approximately 93% of deeply sourced carbon is trapped in the shallow crust, rendering orogenic processes carbon neutral and possibly acting as one of the major triggers of long‐term climate cooling in the Cenozoic. However, the carbon sources and sinks and the mechanisms underlying their impacts on climate change are not fully understood. This paper reports the chemistry of aqueous solutions, carbon isotopic compositions of dissolved inorganic carbon (DIC) and dissolved organic carbon (DOC), and helium and carbon isotopic compositions of free gases from hot springs in southern Tibet. The carbon isotope fractionation model between gas and aqueous solutions revealed that the carbon released from depth underwent six different stages. Based on the random forests machine learning algorithm, we estimated the metamorphic carbon flux across the Himalayan orogenic belt. Furthermore, only a small fraction (7%) of deep carbon is eventually released into the atmosphere, and this volume is the same magnitude as the amount of carbon absorbed by silicate weathering on the Tibetan Plateau. This paper emphasizes that the carbon sequestration process in the upper crust may have resulted in nearly carbon‐neutral geological bodies in the Himalayan orogenic belt, thus playing a critical role in Cenozoic climate cooling. The development of sustainable and environmentally friendly industrial processes is becoming very crucial and demanding for the rapid implementation of innovative bio-based technologies. Natural extreme environments harbor the potential for discovering and utilizing highly specific and efficient biocatalysts that are adapted to harsh conditions. This review focuses on extremophilic microorganisms and their enzymes (extremozymes) from various hot springs, shallow marine vents, and other geothermal habitats in Europe and the Caucasus region. These hot environments have been partially investigated and analyzed for microbial diversity and enzymology. Hotspots like Iceland, Italy, and the Azores harbor unique microorganisms, including bacteria and archaea. The latest results demonstrate a great potential for the discovery of new microbial species and unique enzymes that can be explored for the development of Circular Bioeconomy. Different screening approaches have been used to discover enzymes that are active at extremes of temperature (up 120 °C), pH (0.1 to 11), high salt concentration (up to 30%) as well as activity in the presence of solvents (up to 99%). The majority of published enzymes were revealed from bacterial or archaeal isolates by traditional activity-based screening techniques. However, the latest developments in molecular biology, bioinformatics, and genomics have revolutionized life science technologies. Post-genomic era has contributed to the discovery of millions of sequences coding for a huge number of biocatalysts. Both strategies, activity- and sequence-based screening approaches, are complementary and contribute to the discovery of unique enzymes that have not been extensively utilized so far. This study elucidates methods for isolation and biochemical identification of thermophilic bacteria isolated from the Gauri Kund hot spring of Uttarakhand, India. Heavy metals are discharged from various industries and man-made activities into the soil and water resources. Increased toxic concentrations of heavy metals are responsible for disturbing metabolism and may cause lethal and chronic diseases in humans and plants. Therefore, this study aims to isolate and identify some metalresistant bacterial strains of thermophiles. A total of 3 iron (Fe) and 3 manganese (Mn) resistant bacterial strains were isolated. These strains were capable of tolerating the presence of Fe, and Mn metal ions. Additionally, the metal tolerance limit of isolated strains was determined by minimum inhibitory concentration (MIC). The thermophilic strain GA7 showed the highest MIC (475 μg/lt.) against Mn ions. However, for more accurate results, molecular characterization is also required and reported in another study with the results of a detailed bioremediation assay. The unique cell wall structures and thermoadaptation mechanisms of thermophilic bacteria help in heavy metal Bioremediation. It is concluded that bacterial bioremediation is a very good, eco-friendly, and cost-effective biotechnological approach to reduce heavy metal contamination from the soil and water bodies. As a crucial enzyme for cellulose degradation, β-glucosidase finds extensive applications in food, feed, and bioethanol production; however, its potential is often limited by inadequate thermal stability and glucose tolerance. In this study, a functional gene (lq-bg5) for a GH1 family β-glucosidase was obtained from the metagenomic DNA of a hot spring sediment sample and heterologously expressed in E. coli and the recombinant enzyme was purified and characterized. The optimal temperature and pH of LQ-BG5 were 55 °C and 4.6, respectively. The relative residual activity of LQ-BG5 exceeded 90% at 55 °C for 9 h and 60 °C for 6 h and remained above 100% after incubation at pH 5.0–10.0 for 12 h. More importantly, LQ-BG5 demonstrated exceptional glucose tolerance with more than 40% activity remaining even at high glucose concentrations of 3000 mM. Thus, LQ-BG5 represents a thermophilic β-glucosidase exhibiting excellent thermal stability and remarkable glucose tolerance, making it highly promising for lignocellulose development and utilization. The energy inside the Earth can not only be released outward through earthquakes and volcanoes but also can be used by humans in the form of geothermal energy. Is there a correlation between different forms of energy release? In this contribution, we perform detailed seismic and geothermal research in the Beijing area. The results show that the geothermal resources in Beijing belong to typical medium-low temperature geothermal resources of the sedimentary basin, and some areas are controlled by deep fault activities (e.g., Xiji geothermal well (No. 17)). The heat sources are upper mantle heat, radioactive heat in granite, and residual heat from magma cooling. The high overlap of earthquakes and geothermal field locations and the positive correlation between the injection water and earthquakes indicate that the exploitation and injection water will promote the release of the earth's energy. The energy releases are partitioned into multiple microearthquakes, avoiding damaging earthquakes (ML ≥ 5) due to excessive energy accumulation. Therefore, the exploitation of geothermal resources may be one way to reduce destructive earthquakes. A type of high molecular weight bioactive polymers called exopolysaccharides (EPS) are produced by thermophiles, the extremophilic microbes that thrive in acidic environmental conditions of hot springs with excessively warm temperatures. Over time, EPS became important as natural biotechnological additives because of their noncytotoxic, emulsifying, antioxidant, or immunostimulant activities. In this article, we unravelled a new EPS produced by Staphylococcus sp. BSP3 from an acidic (pH 6.03) San Pedro hot spring (38.1 °C) located in the central Andean mountains in Chile. Several physicochemical techniques were performed to characterize the EPS structure including Scanning electron microscopy–energy dispersive X-ray spectroscopy (SEM–EDS), Atomic Force Microscopy (AFM), High-Performance Liquid Chromatography (HPLC), Gel permeation chromatography (GPC), Fourier Transform Infrared Spectroscopy (FTIR), 1D Nuclear Magnetic Resonance (NMR), and Thermogravimetric analysis (TGA). It was confirmed that the amorphous surface of the BSP3 EPS, composed of rough pillar-like nanostructures, is evenly distributed. The main EPS monosaccharide constituents were mannose (72%), glucose (24%) and galactose (4%). Also, it is a medium molecular weight (43.7 kDa) heteropolysaccharide. NMR spectroscopy demonstrated the presence of a [→ 6)-⍺-d-Manp-(1 → 6)-⍺-d-Manp-(1 →] backbone 2-O substituted with 1-⍺-d-Manp. A high thermal stability of EPS (287 °C) was confirmed by TGA analysis. Emulsification, antioxidant, flocculation, water-holding (WHC), and oil-holding (OHC) capacities are also studied for biotechnological industry applications. Geothermal features, such as hot springs and mud volcanoes, host diverse microbial life, including many extremophile organisms. The physicochemical parameters of the geothermal feature, such as temperature, pH, and heavy metal concentration, can influence the alpha and beta diversity of microbial life in these environments, as can spatiotemporal differences between sites and sampling. In this study, water and sediment samples were collected and analyzed from eight geothermal sites at Yellowstone National Park, including six hot springs, a mud volcano, and an acidic lake within the same week in July 2019, and these geothermal sites varied greatly in their temperature, pH, and chemical composition. All samples were processed and analyzed with the same methodology and taxonomic profiles and alpha and beta diversity metrics determined with 16S rRNA sequencing. These microbial diversity results were then analyzed with respect to pH, temperature, and chemical composition of the geothermal features. Results indicated that predominant microbial species varied greatly depending on the physicochemical composition of the geothermal site, with decreases in pH and increases in dissolved heavy metals in the water corresponding to decreases in alpha diversity, especially in the sediment samples. Similarly, sites with acidic pH values had more similar microbial populations (beta diversity) to one another than to relatively neutral or alkaline pH geothermal sites. This study suggests that pH and/or heavy metal concentration is a more important driver for microbial diversity and population profile than the temperature for these sites and is also the first reported microbial diversity study for multiple geothermal sites in Yellowstone National Park, including the relatively new mud volcano Black Dragon’s Caldron, which erupted in 1948.

Antimicrobial resistance of pathogenic bacteria to currently used antibiotics killed an estimated 1.27 million people in 2019 and it is believed that this toll will increase to 10 million per year by 2050. This remains one of the most urgent threats to human health and the hunt for novel antimicrobial natural products (NPs) from microorganisms is essential in efforts to combat this threat, but recent efforts have been unsuccessful. The investigation of extreme ecological niches, such as hot springs, has gained recent interest due to their unique ecosystems, increasing the chance for novel antimicrobial NP discovery. The Roman Baths (Bath) is the UK’s only hot spring and it remains unexplored in terms of microbial diversity. In this study, samples of water, biofilm and sediment were taken from the King’s Spring and the Great Bath, and 16 S rRNA metagenomic sequencing was performed for bacterial and archaeal community exploration. Additionally, samples were used for culture-dependent isolation of antimicrobial NP producing isolates. At the phylum level, data from water samples revealed a shift from an archaeal dominant community in the King’s Spring (Crenarchaeota, Aenigmarchaeota and Nanoarchaeota) to bacteria in the Great Bath (Cyanobacteria and Proteobacteria). Archaea were still present in the biofilm and sediment of the hot spring but consistently the bacterial phyla Cyanobacteria, Bacteroidota, Proteobacteria and Nitrospirota were dominant at both sites. The antimicrobial NP producing candidate phyla Actinobacteria and Myxococcota were found in all samples. From initial isolation experiments, 297 isolates were purified, of which 15 showed broad spectrum activity against human pathogens, though interestingly these were not from target genera in the Actinobacteria. From these data, there is clear potential for novel NP discovery from the Roman Baths, as has been demonstrated from other thermal hot springs globally.

Introduction: β-Glucosidase serves as the pivotal rate-limiting enzyme in the cellulose degradation process, facilitating the hydrolysis of cellobiose and cellooligosaccharides into glucose. However, the widespread application of numerous β-glucosidases is hindered by their limited thermostability and low glucose tolerance, particularly in elevated-temperature and high-glucose environments. Methods: This study presents an analysis of a β-glucosidase gene belonging to the GH1 family, denoted lqbg8, which was isolated from the metagenomic repository of Hehua hot spring located in Tengchong, China. Subsequently, the gene was cloned and heterologously expressed in Escherichia coli BL21(DE3). Post expression, the recombinant β-glucosidase (LQBG8) underwent purification through a Ni affinity chromatography column, thereby enabling the in-depth exploration of its enzymatic properties. Results: LQBG8 had an optimal temperature of 70°C and an optimum pH of 5.6. LQBG8 retained 100 and 70% of its maximum activity after 2-h incubation periods at 65°C and 70°C, respectively. Moreover, even following exposure to pH ranges of 3.0–10.0 for 24 h, LQBG8 retained approximately 80% of its initial activity. Notably, the enzymatic prowess of LQBG8 remained substantial at glucose concentrations of up to 3 M, with a retention of over 60% relative activity. The kinetic parameters of LQBG8 were characterized using cellobiose as substrate, with Km and Vmax values of 28 ± 1.9 mg/mL and 55 ± 3.2 μmol/min/mg, respectively. Furthermore, the introduction of LQBG8 (at a concentration of 0.03 mg/mL) into a conventional cellulase reaction system led to an impressive 43.7% augmentation in glucose yield from corn stover over a 24-h period. Molecular dynamics simulations offered valuable insights into LQBG8’s thermophilic nature, attributing its robust stability to reduced fluctuations, conformational changes, and heightened structural rigidity in comparison to mesophilic β-glucosidases. Discussion: In summation, its thermophilic, thermostable, and glucose-tolerant attributes, render LQBG8 ripe for potential applications across diverse domains encompassing food, feed, and the production of lignocellulosic ethanol. During the investigation of the freshwater diatoms from Tibet, a monoraphid species was observed from a hot spring near Anduo County, located on a plateau in the central portion of Tibet. This species shares the diagnostic features of Crenotia, such as the valve bent along the transapical axis, striae uniseriate to biseriate from centre to the apices and areolae with special structures located at the end of each stria. We compared the morphological characters of this new species with the others in this genus and show it to be new; it is named Crenotia tibetia sp. nov. This species has small valves with slightly protracted ends with nearly capitate apices, lanceolate axial area, central area unilaterally expanded to the margin, striae uniseriate to biseriate, but, in some valves, the striae are only uniseriate. Areolae are round small to irregular in shape and, at the end of each stria, there is a horseshoe-shaped areola present. Observations of developing valves show all the striae begin biseriate, then they become covered by silica to form uniseriate striae. The continuous requirement for pre-bleaching processes on kraft pulp, employing a range of compatible enzymes, aims to mitigate the pollution caused by chemical bleaching agents. In the present study, the laccase-producing bacterium Bacillus licheniformis BK-1 was isolated from the Bakreshwar hot spring in India and tested for laccase production using different lignocellulosic substrates. The isolate was found to produce maximum laccase (8.25 IU/mL) in the presence of rice bran as a substrate, followed by 5.14 IU/mL using sawdust over a 48 h period. Laccase production doubled when medium parameters were optimized using a central composite design. The bleaching of rice straw pulp was accomplished using a laccase, xylanase (previously extracted from the same bacteria), and laccase–xylanase mixture. The mix-wood kraft pulp treated with the enzyme mixture at pH 7.0 and 50 °C temperature for up to 180 min reduced the chlorine amount by 50% compared to the control. The results also revealed that the enzyme mixture improved the pulp's optical (brightness 10.39%) and physical (tear index 39.77%, burst index 22.82%, and tensile strength 14.28%) properties with 50% chlorine dose. These exceptional properties underscore the enzyme mixture's suitability for pulp pre-bleaching in paper manufacturing, offering a safer and more environmentally friendly process. We study the effects of centralized health management based on hot spring resorts on the physical examination index and sleep quality of people at high risk of chronic diseases. We recruited 114 volunteers at high risk of chronic diseases. We then divided them into 57 in the intervention group and 57 in the control group. The intervention group collectively received 4 weeks (28 days) of comprehensive health management interventions at Tongjing Hotspring Resort, including regular schedules, balanced diet, appropriate exercise, targeted health education, etc. The main outcomes are physical examination indicators (height, weight, waist circumference, blood pressure, lipids, and glucose) and sleep quality. Both groups underwent a questionnaire and physical examination at baseline, 2 weeks and 4 weeks. Intragroup comparisons grouped by exposure criteria showed decreases in BMI, waist circumference, triglycerides, total cholesterol, and blood glucose in the intervention group at both 2 and 4 weeks (all P < 0.05); however, in the control group, only triglycerides decreased at 4 weeks (P < 0.05). Intergroup comparisons showed BMI and waist circumference were significantly lower in the intervention group than in the control group at 4 weeks (all P < 0.05). Intragroup comparisons of insomnia severity index (ISI) scores showed a significant decrease in the intervention group at both 2 and 4 weeks (all P < 0.001) with no significant change in the control group (P > 0.05). Intergroup comparisons showed that the insomnia severity index (ISI) scores were significantly higher in the intervention group than in the control group at baseline (P = 0.006) but became significantly lower than the control group at 2 and 4 weeks (all P < 0.001). Thus, this pattern significantly improved BMI, waist circumference, triglycerides, and sleep in the intervention group. Background and Objectives: Rinaie Marwah hot spring Kishtwar (RMHSK) is one of the geothermal springs located at 33°51'51"N 75°32'07"E with an elevation of 2134 meters above sea level in Jammu and Kashmir, India. We aimed to study the microbial diversity of this geothermal spring using metagenomics. Materials and Methods: In the present study, physiochemical parameters including temperature (65-75oC), pH (6. 9-8. 8), hardness (250 ppm), and mineral content was measured along with the microbial diversity using Illumina MiSeq metagenome-based 16s amplicon sequencing (V3-V4). The sequence reads were classified taxonomically into 31 phyla, 71 classes, 152 orders, 256 families, 410 genus, and 665 species. QIIME 2 (Quantitative Insights into Microbial Ecology), an extensible, powerful, and decentralized analytical tool, was used for taxonomic analysis. Results: Bacteroidota (32. 57%) was the dominant phylum, Bacteroidia (32. 51%) the dominant class, Bacteroidales (16. 6%) the dominant order, and Lentimicrobiaceae (14. 23%) was the dominant family per the abundance analysis. Shannon (2. 28) and Chao 1 (87. 0) diversity indices support the existence of higher microbial diversity in RMHSK (50717 OTUs). We examined the movement patterns of American Coots relative to water temperature and heat index where a geothermally heated stream enters Utah Lake, UT, USA during winter using a thermal imager onboard an Unmanned Aerial System (UAS). Observations of Coots in different water temperatures (n = 2354) were compiled during 31 flights in January–March, 2020. Coots were actively swimming during 82.3% of the observations, fleeing during 8.1%, and sedentary in water during 9.6%. The population spent more time swimming in cooler water and were sedentary for longer in warmer water. The only instance of uniform behavior occurred during an anomalously cold period in which Coots stood huddled in the warmest available water. Using a linear model framework, we found that Coot time spent increased with water temperature when heat index decreased. Coots' swim paths deviated away from warmer water more than cooler water and more so as heat index increased. We suggest that Coots use the spring water primarily for ice-free habitat and maintain metabolic rates via a combination of thermal conductance, caloric ingestion, and exercise, but only exploit the heated water strictly for thermoregulation during anomalously cold periods. We discuss the benefits and hindrances of using UASs amidst a rapidly expanding assortment of sensors and aircraft available to researchers. Given wetlands' immediate conservation concern, ecosystem services they provide, and support of unique species assemblages, our research provides valuable insight into documenting wildlife's use of wetlands with relatively low scientific take and human interference. Various microorganisms exist in environments, and each of them has its optimal growth temperature (OGT). The relationship between genomic information and OGT of each species has long been studied, and one such study revealed that OGT of prokaryotes can be accurately predicted based on the fraction of seven amino acids (IVYWREL) among all encoded amino-acid sequences in its genome. Extending this discovery, we developed a 'Metagenomic Thermometer' as a means of predicting environmental temperature based on metagenomic sequences. Temperature prediction of diverse environments using publicly available metagenomic data revealed that the Metagenomic Thermometer can predict environmental temperatures with small temperature changes and little influx of microorganisms from other environments. The accuracy of the Metagenomic Thermometer was also confirmed by a demonstration experiment using an artificial hot water canal. The Metagenomic Thermometer was also applied to human gut metagenomic samples, yielding a reasonably accurate value for human body temperature. The result further suggests that deep body temperature determines the dominant lineage of the gut community. The hot spring Vranjska Banja is the hottest spring on the Balkan Peninsula with a water temperature of 63–95 °C and a pH value of 7.1, in situ. According to the physicochemical analysis, Vranjska Banja hot spring belongs to the bicarbonated and sulfated hyperthermal waters. The structures of microbial community of this geothermal spring are still largely unexplored. In order to determine and monitor the diversity of microbiota of the Vranjska Banja hot spring, a comprehensive culture-independent metagenomic analysis was conducted in parallel with a culture-dependent approach for the first time. Microbial profiling using amplicon sequencing analysis revealed the presence of phylogenetically novel taxa, ranging from species to phyla. Cultivation-based methods resulted in the isolation of 17 strains belonging to the genera Anoxybacillus, Bacillus, Geobacillus, and Hydrogenophillus. Whole-genome sequencing of five representative strains was then performed. The genomic characterization and OrthoANI analysis revealed that the Vranjska Banja hot spring harbors phylogenetically novel species of the genus Anoxybacillus, proving its uniqueness. Moreover, these isolates contain stress response genes that enable them to survive in the harsh conditions of the hot springs. The results of the in silico analysis show that most of the sequenced strains have the potential to produce thermostable enzymes (proteases, lipases, amylases, phytase, chitinase, and glucanase) and various antimicrobial molecules that can be of great importance for industrial, agricultural, and biotechnological applications. Hot spring biofilms are stable, highly complex microbial structures. They form at dynamic redox and light gradients and are composed of microorganisms adapted to the extreme temperatures and fluctuating geochemical conditions of geothermal environments. In Croatia, a large number of poorly investigated geothermal springs host biofilm communities. Here, we investigated the microbial community composition of biofilms collected over several seasons at 12 geothermal springs and wells. We found biofilm microbial communities to be temporally stable and highly dominated by Cyanobacteria in all but one high-temperature sampling site (Bizovac well). Of the physiochemical parameters recorded, temperature had the strongest influence on biofilm microbial community composition. Besides Cyanobacteria, the biofilms were mainly inhabited by Chloroflexota, Gammaproteobacteria, and Bacteroidota. In a series of incubations with Cyanobacteria-dominated biofilms from Tuhelj spring and Chloroflexota- and Pseudomonadota-dominated biofilms from Bizovac well, we stimulated either chemoorganotrophic or chemolithotrophic community members, to determine the fraction of microorganisms dependent on organic carbon (in situ predominantly produced via photosynthesis) versus energy derived from geochemical redox gradients (here simulated by addition of thiosulfate). We found surprisingly similar levels of activity in response to all substrates in these two distinct biofilm communities, and observed microbial community composition and hot spring geochemistry to be poor predictors of microbial activity in the study systems. Bacterial strain VB3, an aerobic bacterium, was isolated from the water of Vashisht hot spring, Himachal Pradesh, India. The lysogenic bacterial strain VB3's draft genome sequences comprise approximately 6.06 MB DNA and 5607 predicted genes with a G + C content of 60.56%. The 16S rRNA sequencing assigned the bacterial strain VB3 to the genus Pseudomonas. Pseudomonas sp. VB3 was a Gram-negative, rod-shaped bacteria demonstrating catalase, oxidase, citrate utilization, cellulase, and lipase activities. Genome mining revealed fourteen gene clusters coding for the biosynthesis of bioactive secondary metabolites. PHASTER analysis identified two intact and three incomplete putative prophage-related sequence regions in the draft genome. The reduction in the growth rate of the strain VB3 on mitomycin-C treatment was concurrent with the detection of the clearance zone in the spot assay. [ABSTRACT FROM AUTHOR]

The lignocellulose bioconversion process is an eco-friendly and green-economy alternative technology that allows the reduction of pollution and global warming, so it is necessary for thermophilic and thermostable hydrolytic enzymes from natural sources. This research aimed to isolate cellulolytic and xylanolytic microbial consortia from Huancarhuaz hot spring (Peru) from sludge or in situ baiting cultured with or without sugarcane bagasse. According to the hydrolytic activities consortium T4 from in situ baiting was selected. It was cultivated in submerged fermentation at 65 °C, pH 6.5 for eight days using LB supplemented with sugar cane bagasse (SCB), pine wood sawdust (PWS), CMC, xylan of birchwood, or micro granular cellulose. Crude extract of culture supplemented with SCB (T4B) showed better endoglucanase and xylanase activities with higher activities at 75 °C and pH 6. In these conditions, cellulase activity was kept up to 57% after 1 h of incubation, while xylanase activity was up to 63% after 72 h. Furthermore, this crude extract released reduced sugars from pretreated SCB and PWS. According to metagenomic analysis of 16S rDNA, Geobacillus was the predominant genus. It was found thermostable genes: a type of endoglucanase (GH5), an endo-xylanase (GH10), and alkali xylanase (GH10) previously reported in Geobacillus sp. strains. Finally, Huancarhuaz hot spring harbors a genetic microbial diversity for lignocellulosic waste bioconversion in high temperatures, and the T4B consortium will be a promising source of novel extreme condition stable enzymes for the saccharification process.

BACKGROUND AND OBJECTIVES: Medicinal plants growing in geothermal areas have been reported to possess relatively high concentrations of bioactive secondary metabolites, which have been attributed to the adaptive heat stress response. Nonetheless, the exploitation of their medicinal benefits is limited by potentially life-threatening concentrations of heavy metals. Chromolaena odorata Linn, also called seurapoh, is a well-known medicinal plant that can absorb and accumulate heavy metals from the soil. This present study aimed to investigate the contents of mercury, lead, cadmium, and arsenic in the ethanolic extract of Chromolaena odorata Linn leaves collected from a geothermal area in Aceh Province, Indonesia.METHODS: Three hot springs, namely Ie-Suum, Ie-Jue, and Ie-Brouk, located in the same geothermal area, Seulawah Agam, were selected as the sampling points, and three samples of Chromolaena odorata were collected at each point. Extraction was carried out by means of maceration employing ethanol solvent. The heavy metal contents of each extract were determined using priorly validated atomic absorption spectrometry and graphite furnace atomic absorption spectrometry. The maximum thresholds of each heavy metal from the Indonesian Agency for Drug and Food Control were adopted during data interpretation.FINDINGS: The results revealed the presence of trace levels of arsenic (0.0482 ± 0.004 – 0.0639 ± 0.007 milligram per kilogram) and lead (0.0219 ± 0.004 – 0.0672 ± 0.006 milligram per kilogram), which did not exceed Indonesian maximum safety thresholds (≤5 and ≤10 milligram per kilogram, respectively). Mercury in all samples was not observable (limit of detection = 0.018 milligram per liter). Cadmium was observed in almost all samples with a concentration range of 0.0219 ± 0.005–1.1472 ± 0.006 milligram per kilogram, which exceeded the maximum threshold (0.3 milligram per liter).CONCLUSION: Heavy metal contamination in the ethanolic extract of geothermal Chromolaena odorata leaves has been attributed to volcanic activities. Among the heavy metals of concern, cadmium was the only one with a concentration exceeding the safety limit. The presence of cadmium in the extract at a high concentration could cause its translocation to the human body, eventually leading to multiple organ damage. Therefore, the extract of Chromolaena odorata leaves collected from a geothermal area should be consumed with caution for possible cadmium intoxication.

This study presents a comprehensively analysis of geothermal characteristics in the Xianshuihe geothermal area along the Sichuan-Tibet Railway, using temperature logging, temperature monitoring and thermal conductivity measurement, and regional geothermal geological survey data. The research focuses on the geothermal background, geothermal field, and their potential impact on the surrounding tunnels. The investigation reveals that the average heat flow value in the study area is approximately 73.0 mW/m2, significantly higher than the average terrestrial heat flow in mainland China (​​62.5 mW/m2). This high terrestrial heat flow signifies a distinct thermal background in the area. In addition, geothermal anomalies in the area are found to be closely associated with the distribution of hot springs along NW faults, indicating a strong control by the Xianshuihe fault zone. The study concludes that the region's favorable conditions for geothermal resources are attributed to the combination of high terrestrial heatflow background and water-conducting faults. However, these conditions also pose a potential threat of heat damage to the tunnels along the Sichuan-Tibet Railway. To evaluate the risk, the research takes into account the terrestrial heat flow, thermal conductivity of the tunnel surrounding rocks, characteristics of the regional constant temperature layer, as well as the distribution of hot springs and faults. The analysis specifically focuses on the thermal damage risk of Kangding 1# tunnel and 2# tunnel passing through the study area. Based on the findings, it is determined that Kangding 1# tunnel and 2# tunnel have relatively low risk of heat damage, as they have avoided most of the high temperature anomaly areas. However, several sections of the tunnels do traverse zones with low to medium temperatures, where surface rock temperatures can reach up to 45°C. Therefore, these regions should not be neglected during the construction and operation of the tunnel project, and mitigation measures may be necessary to address the potential heat-related challenges in the area.

Introductionβ-Glucosidase serves as the pivotal rate-limiting enzyme in the cellulose degradation process, facilitating the hydrolysis of cellobiose and cellooligosaccharides into glucose. However, the widespread application of numerous β-glucosidases is hindered by their limited thermostability and low glucose tolerance, particularly in elevated-temperature and high-glucose environments.MethodsThis study presents an analysis of a β-glucosidase gene belonging to the GH1 family, denoted lqbg8, which was isolated from the metagenomic repository of Hehua hot spring located in Tengchong, China. Subsequently, the gene was cloned and heterologously expressed in Escherichia coli BL21(DE3). Post expression, the recombinant β-glucosidase (LQBG8) underwent purification through a Ni affinity chromatography column, thereby enabling the in-depth exploration of its enzymatic properties.ResultsLQBG8 had an optimal temperature of 70°C and an optimum pH of 5.6. LQBG8 retained 100 and 70% of its maximum activity after 2-h incubation periods at 65°C and 70°C, respectively. Moreover, even following exposure to pH ranges of 3.0–10.0 for 24 h, LQBG8 retained approximately 80% of its initial activity. Notably, the enzymatic prowess of LQBG8 remained substantial at glucose concentrations of up to 3 M, with a retention of over 60% relative activity. The kinetic parameters of LQBG8 were characterized using cellobiose as substrate, with Km and Vmax values of 28 ± 1.9 mg/mL and 55 ± 3.2 μmol/min/mg, respectively. Furthermore, the introduction of LQBG8 (at a concentration of 0.03 mg/mL) into a conventional cellulase reaction system led to an impressive 43.7% augmentation in glucose yield from corn stover over a 24-h period. Molecular dynamics simulations offered valuable insights into LQBG8’s thermophilic nature, attributing its robust stability to reduced fluctuations, conformational changes, and heightened structural rigidity in comparison to mesophilic β-glucosidases.DiscussionIn summation, its thermophilic, thermostable, and glucose-tolerant attributes, render LQBG8 ripe for potential applications across diverse domains encompassing food, feed, and the production of lignocellulosic ethanol.

Background Sleep-wake rhythm is highly correlated with core body temperature, and bathing in the hot spring can effectively manipulate core body temperature to improve sleep quality. However, there is still a lack of experimental research on the influencing factors of utilizing hot spring facilities to positively affect sleep. Methods In this study, we aimed to explore the factors improving sleep quality via hot spring bathing in terms of both subjective (self-rating scales) and objective (actigraphy) measures, 33 insomniacs were recruited and randomly divided into the afternoon (AG) and evening bathing groups (EG). Results The results showed that after the intervention, subjective sleep quality was improved significantly in both groups, and the evening bathing group had a better performance. In contrast, neither group had a significant change in objective sleep measures. Moreover, we found that hot spring spa therapy is particularly effective in reducing pre-sleep thoughts inventory and shortening sleep onset latency, and it is more applicable to people with difficulty falling asleep. Conclusion This study identified the timing and individual's behaviour traits as the important factors, and these formed practical guidance for sleep improvement by spa therapy.

Background and Objectives: Rinaie Marwah hot spring Kishtwar (RMHSK) is one of the geothermal springs located at 33°51′51′′N 75°32′07′′E with an elevation of 2134 meters above sea level in Jammu and Kashmir, India. We aimed to study the microbial diversity of this geothermal spring using metagenomics. Materials and Methods: In the present study, physiochemical parameters including temperature (65-75oC), pH (6. 9-8. 8), hardness (250 ppm), and mineral content was measured along with the microbial diversity using Illumina MiSeq metagenome-based 16s amplicon sequencing (V3-V4). The sequence reads were classified taxonomically into 31 phyla, 71 classes, 152 orders, 256 families, 410 genus, and 665 species. QIIME 2 (Quantitative Insights into Microbial Ecology), an extensible, powerful, and decentralized analytical tool, was used for taxonomic analysis. Results: Bacteroidota (32. 57%) was the dominant phylum, Bacteroidia (32. 51%) the dominant class, Bacteroidales (16. 6%) the dominant order, and Lentimicrobiaceae (14. 23%) was the dominant family per the abundance analysis. Shannon (2. 28) and Chao 1 (87. 0) diversity indices support the existence of higher microbial diversity in RMHSK (50717 OTUs). Conclusion: The microbial diversity of RMHSK is reported for the first time through a metagenomic study. Identification of microorganisms with characteristics that are relevant to industries.

Thermostable α-amylase is a commercial enzyme that has found several biotechnological applications in recent years. This prompts researchers to check out the hot ecosystems the least explored to find producing microorganisms. Thermal waters are a poorly studied ecological niche compared to other ecosystems. Actinobacteria are known for their unique metabolic abilities to produce the most innovative bio-molecules. In this study, the isolation by conventional microbiological methods, allowed to obtain 13 thermophilic actinobacteria from a hot spring of TLEGHMA located in the eastern Algerian. All these isolates were characterized morphologically and by physiological methods. Molecular identification by sequencing of the rRNA16s gene, allowed to assign them to the genus Streptomyces sp. Among these isolates, 9 actinobacteria showed abilities to produce a thermostable α-amylase active at a temperature of 55°C. Two isolates named TA3 and TA4 are however, the most successful. They were assigned to Streptomyces albidofalvus and Streptomyces cavourensis respectively. they are able to produce a thermostable α-amylase at 55°C with an activity rate of 110.33U/ml by strain TA3 and 224U/ml by strain TA4 and with an optimum of activity in a pH equal to 9. These results show that these thermophilic Streptomyces from these hot waters, are a very important source of thermostable and alkalophilic α-amylase. [ABSTRACT FROM AUTHOR]

BACKGROUND AND OBJECTIVES: Medicinal plants growing in geothermal areas have been reported to possess relatively high abundance of bioactive secondary metabolites concomittant to the adaptive heat stress response. Nonetheless, the exploitation of their medicinal benefits is limited by the possible healththreatening concentrations of heavy metal contamination. Chromolaena odorata Linn, or also called as seurapoh, is a well-known medicinal plant but could absorb and accumulate heavy metal from the soil. Herein, this present study aimed to investigate the contents of Hg, Pb, Cd, and As in ethanolic extract of Chromolaena odorata Linn leaves collected from a geothermal area in Aceh of Indonesia. METHODS: Three hot springs (Ie-Suum, Ie-Jue, and Ie-Brouk) located in the same geothermal area Seulawah Agam was selected as the sampling points, where three samples of Chromolaena odorata were collected in each sampling point. Extraction was carried out by means of maceration employing ethanol solvent. The contents of heavy metals in each extract were determined by priorly validated atomic absoption spectrometry and graphite furnace atomic absorption. FINDINGS: The results revealed that arsenic (0.0482 ± 0.004 - 0.0639 ± 0.007 miligram per kilogram) and lead (0.0219 ± 0.004 - 0.0672 ± 0.006 miligram per kilogram) were found in trace levels and did not exceed Indonesian maximum safety thresholds (=5 and =10 miligram per kilogram, respectively). The presence of mercury in all samples was not observable (limit of detection= 0.018 miligram per liter). Cadmium was observed in almost all samples with a concentration range of 0.0219 ± 0.005 - 1.1472 ± 0.006 miligram per kilogram exceeding the maximum threshold (0.3 miligram per liter). CONCLUSION: Contamination of heavy metals in the ethanolic extract of geothermal Chromolaena odorata leaves is thought to be originated from volcanic activities. Among the heavy metals of concern, cadmium was the only one with concentration exceeding the safety limit. The presence of cadmium in the extract with high concentration could act cause its translocation to human body which eventually lead to multiple organ damages. Therefore, the extract of geothermal C. odorata leaves collected from a geothermal area should be consumed with caution for possible cadmium intoxication. [ABSTRACT FROM AUTHOR]

β-glucosidase is a key enzyme in the degradation of lignocellulosic biomass, which is responsible for the conversion of oligosaccharides from cellulose hydrolysates to glucose. However, its required high temperatures and the presence of inhibitors have limited its use in industry. In this study, a new β-glucosidase gene, named thbg2, was obtained from the metagenome Ruidian Hot Spring, Tengchong City, Yunnan Province, southwestern China. The gene was synthesized, cloned, heterologously expressed, and enzymatically characterized. Its optimum temperature and pH were 60 °C and pH 5.6, respectively. ThBg2 exhibited more than 60% relative activity in temperatures ranging from 40 °C to 70 °C and across a pH of 4.0–6.6. It maintained 100% relative activity after incubation at either 50 °C for 24 h or 60 °C for 12 h and more than 80% residual activity after incubation at pH 4.0–6.0 for 24 h. Moreover, it maintained more than 80% relative activity in the presence of heavy metal ions, ethanol, SDS etc. Furthermore, glucose yields from corn stalks increased by 20% after ThBg2 (0.05 mg/mL) was added to the commercial cellulase reaction system. Overall, this work identified a thermophilic and inhibitor-tolerant β-glucosidase with potential applications in commercial lignocellulose utilization and the bioenergy industry. The taxonomic position of strain EF45031T, isolated from the Neungam Carbonate hot spring, was examined using the polyphasic taxonomic approach. Strain EF45031T shared the highest percentage of 16S rRNA gene sequence with Brachybacterium nesterenkovii CIP 104813 T (97.7%). The average nucleotide identity (ANI), average amino acid identity (AAI), and digital DNA–DNA hybridization (dDDH) values between strain EF45031T and the type strains B. nesterenkovii CIP 104813 T and B. phenoliresistens Phenol-AT were 77.0%, 69.15%, 21.9% and 75.73%, 68.81%, 20.5%, respectively. Phylogenomic analysis using an up-to-date bacterial core gene (UBCG) set revealed that strain EF45031T belonged to the genus Brachybacterium. Growth occurred between 25 and 50 ℃ at pH 6.0–9.0 and could tolerate salinity up to 5% (w/v). Strain had anteiso-C15:0 and anteiso-C17:0 as major fatty acids. Menaquinone-7 (MK-7) was the predominant respiratory menaquinone. The polar lipids were diphosphatidylglycerol, phosphatidylglycerol, three aminolipids, and two unidentified glycolipids. The cell-wall peptidoglycan contained meso-diaminopimelic acid as a diagnostic diamino acid. The genome comprised 2,663,796 bp, with a G + C content of 70.9%. Stress-responsive periplasmic chaperone/protease coding genes were identified in the genome of EF45031T and were not detected in other Brachybacterium species. The polyphasic taxonomic properties indicate that the strain represents a novel species within the genus Brachybacterium, for which the name Brachybacterium sillae sp. nov. is proposed. GAL08 are bacteria belonging to an uncultivated phylogenetic cluster within the phylum Acidobacteria. We detected a natural population of the GAL08 clade in sediment from a pH-neutral hot spring located in British Columbia, Canada. To shed light on the abundance and genomic potential of this clade, we collected and analyzed hot spring sediment samples over a temperature range of 24.2-79.8°C. Illumina sequencing of 16S rRNA gene amplicons and qPCR using a primer set developed specifically to detect the GAL08 16S rRNA gene revealed that absolute and relative abundances of GAL08 peaked at 65°C along three temperature gradients. Analysis of sediment collected over multiple years and locations revealed that the GAL08 group was consistently a dominant clade, comprising up to 29.2% of the microbial community based on relative read abundance and up to 4.7 × 105 16S rRNA gene copy numbers per gram of sediment based on qPCR. Using a medium quality threshold, 25 single amplified genomes (SAGs) representing these bacteria were generated from samples taken at 65 and 77°C, and seven metagenome-assembled genomes (MAGs) were reconstructed from samples collected at 45-77°C. Based on average nucleotide identity (ANI), these SAGs and MAGs represented three separate species, with an estimated average genome size of 3.17 Mb and GC content of 62.8%. Phylogenetic trees constructed from 16S rRNA gene sequences and a set of 56 concatenated phylogenetic marker genes both placed the three GAL08 bacteria as a distinct subgroup of the phylum Acidobacteria, representing a candidate order (Ca. Frugalibacteriales) within the class Blastocatellia. Metabolic reconstructions from genome data predicted a heterotrophic metabolism, with potential capability for aerobic respiration, as well as incomplete denitrification and fermentation. In laboratory cultivation efforts, GAL08 counts based on qPCR declined rapidly under atmospheric levels of oxygen but increased slightly at 1% (v/v) O2, suggesting a microaerophilic lifestyle.

As a crucial enzyme for cellulose degradation, β-glucosidase finds extensive applications in food, feed, and bioethanol production; however, its potential is often limited by inadequate thermal stability and glucose tolerance. In this study, a functional gene (lq-bg5) for a GH1 family β-glucosidase was obtained from the metagenomic DNA of a hot spring sediment sample and heterologously expressed in E. coli and the recombinant enzyme was purified and characterized. The optimal temperature and pH of LQ-BG5 were 55 °C and 4.6, respectively. The relative residual activity of LQ-BG5 exceeded 90% at 55 °C for 9 h and 60 °C for 6 h and remained above 100% after incubation at pH 5.0–10.0 for 12 h. More importantly, LQ-BG5 demonstrated exceptional glucose tolerance with more than 40% activity remaining even at high glucose concentrations of 3000 mM. Thus, LQ-BG5 represents a thermophilic β-glucosidase exhibiting excellent thermal stability and remarkable glucose tolerance, making it highly promising for lignocellulose development and utilization.
