Background: Osteoclasts are integral to the advancement of osteoporosis (OP), and their generation under conditions of oxidative stress (OS) involves various pathways. However, the specific mechanism through which the natural antioxidant kaempferol (KAE) mitigates the influence of OS on osteoclasts remains somewhat uncertain. This study aims to evaluate the effect of KAE on osteoclast formation under OS and explore its possible mechanism. Methods: Zebrafish were used to observe the effects of KAE on OP and OS. OP and OS "double disease targets" network pharmacology were used to predict the action target and mechanism of KAE on OP under OS. The effects of KAE on osteoclast differentiation induced by OS were evaluated using RWA264.7 cells induced by LPS. To elucidate the potential mechanism, we detected the expression of related factors and target genes during induction. Results: The presence of KAE exhibited potential in improving the conditions of OP and OS in zebrafish. KAE can reduce the OS of RAW 264.7 cells stimulated by LPS, inhibit the formation of osteoclasts, and change the level of related factors of OS, and reduce the increase of TRAP. The utilization of network pharmacology and target gene expression assay revealed that KAE exerted a down‐regulatory effect on the expression of proto‐oncogene tyrosine protein kinase (SRC), nuclear factor kappa‐B (NF‐κB), Serine/Threonine Kinase‐1 (AKT1), Nitric Oxide Synthase 3 (NOS3) and Matrix Metallopeptidase‐2 (MMP2). Conclusion: Based on the results of this study, KAE may effectively mitigate OS and impede the formation of osteoclasts through the SRC/NF‐κB‐AKT/NOS3 axis. [ABSTRACT FROM AUTHOR]

Aims Coagulase (Coa), a crucial virulence factor of Staphylococcus aureus (S. aureus), is considered a vital target for anti-virulence strategies. The research aimed to discover a natural compound capable of inhibiting S. aureus infection by targeting the virulence factor Coa. Methods and results The study showed that sinensetin at a concentration of 128 μg mL−1 effectively inhibited both Coa-induced coagulation and biofilm formation in S. aureus. However, western blot results indicated that sinensetin did not impact the expression of Coa protein, suggesting that sinensetin may directly target Coa to counteract the virulence of S. aureus. Thermal shift assay results demonstrated that sinensetin enhanced the thermal stability of Coa, supporting the theory of direct binding. Molecular docking and point mutation experiments identified two key binding sites for sinensetin to Coa as R73A-Coa and R204A-Coa. In vivo studies on mice revealed that sinensetin not only reduced lung tissue damage caused by S. aureus infection, but also decreased inflammatory factors in the lung lavage fluid. Furthermore, combining sinensetin with oxacillin improved the survival rates of the Galleria mellonella and mice. In mobile edge computing (MEC), mobile devices can choose to offload their tasks to edge servers for execution, thereby effectively reducing the completion time of tasks and energy consumption of mobile devices. However, most of the data transfer brought by offloading relies on wireless communication technology, making the private information of mobile devices vulnerable to eavesdropping and monitoring. Privacy leakage, especially the location and association privacies, can pose a significant risk to users of mobile devices. Therefore, protecting the privacy of mobile devices during task offloading is important and cannot be ignored. This paper considers both location privacy and association privacy of mobile devices during task offloading in MEC and targets to reduce the leakage of location and association privacy while minimizing the average completion time of tasks. To achieve these goals, we design a privacy-preserving task offloading scheme to protect location privacy and association privacy. The scheme is mainly divided into two parts. First, we adopt a proxy forwarding mechanism to protect the location privacy of mobile devices from being leaked. Second, we select the proxy server and edge server for each task that needs to be offloaded. In the proxy server selection policy, we make a choice based on the location information of proxy servers, to reduce the leakage risk of location privacy. In the edge server selection strategy, we consider the privacy conflict between tasks, the computing ability, and location of edge servers, to reduce the leakage risk of association privacy plus the average completion time of tasks as much as possible. Simulated experimental results demonstrate that our scheme is effective in protecting the location privacy and association privacy of mobile devices and reducing the average completion time of tasks compared with the-state-of-art techniques. A highly sensitive surface plasmon resonance (SPR) sensor composed of a photonic crystal fiber (PCF) with the D-type structure is designed and analyzed by the full-vector finite element method (FEM). Indium tin oxide (ITO) is adopted as the plasmonic substance on account of the low cost and controllable infrared range (1500–2600 nm). By optimizing the structural parameters, the sensor shows a maximum wavelength sensitivity of 46,000 nm/RIU and average sensitivity of 13,166.67 nm/RIU for analyte refractive indexes between 1.355 and 1.385. This PCF combining a circular layout and D-type structure offers excellent sensitivity while the deposition and manufacturing complexity can be reduced. Circular RNAs were recently identified as a novel type of noncoding RNAs. An increasing number of reports have demonstrated their essential regulatory roles in various biological processes and human diseases, including cancer. However, the role of circRNA in cervical cancer (CC) remains largely unknown. In the current study, we investigated the physiological functions of circ_0067934 during CC development and progression. We found that circ_0067934 was overexpressed in CC tissues and cell lines. Circ_0067934 upregulation was associated with advanced stage, lymph node metastasis, and poor prognosis in CC patients. Knockdown of circ_0067934 suppressed the proliferation, colony formation, migration, invasion, and epithelial‐mesenchymal transition of CC cells in vitro. Circ_0067934 loss also inhibited CC tumor growth in vivo. Mechanistically, silencing circ_0067934 increased miR‐545 expression. MiR‐545 repressed EIF3C expression through targeting its 3′‐untranslated region. MiR‐545 suppressed the proliferation, migration, and invasion of CC cells, whereas restoration of EIF3C could rescue the effects of circ_0067934 knockdown. Taken together, our findings revealed that circ_0067934 promotes CC progression via miR‐545/EIF3C axis. Our study may provide a new insight into the pathogenesis of CC. Ovarian cancer is a leading cause of cancer-related women mortality in China. In recent years, the molecular mechanisms involved in ovarian carcinoma development and/or progression have been intensely studied, and several genes have been identified. Deleted in Colorectal Carcinoma ( DCC), is an important tumor suppressor gene, which is inactivated in many kinds of tumors, and its function(s) is not clarified. Even though the lost expression of DCC occurred in later stages of multistep colorectal carcinogenesis, its contribution to the onset or progression of ovarian cancer is not fully understood. To investigate DCC expression in ovarian cancer, we studied 254 clinical samples by RT-PCR. Our results revealed that 52% malignant ovarian cancer did not express DCC gene. By contrast, DCC expression was observed in all normal ovary tissues and 80% benign ovarian tumors. Obviously, there was a significant correlation between DCC expression and ovarian cancer, especially in the epithelial ovarian cancer. The present study also suggested that the loss expression of DCC occurred more frequently in the cases of later clinical stage, higher pathological grade, and poorer prognosis. In the other part of this study, we further explored DCC expression after transfection in two kinds of ovarian caner cell lines, namely SKOV3 cell and HO-8910 cell, using RT-PCR and immunocytochemistry. The results indicated that DCC expressed in SKOV3- DCC and HO-8910 -DCC cells, and ultrastructural analysis showed the appearance of apoptotic features in them. Furthermore, cell growth was markedly down-regulated in above groups of cells, indicating that transfection with the DCC constructs can suppress the growth of tumor cells. In conclusion, our results suggest an association of lost expression of DCC with the ovarian cancer, and DCC gene may inhibit the growth of ovarian carcinoma cells.
