Propofol has become a microtubule‐stabilizing drug for prostate cancer (PC) therapy, but propofol resistance impairs the therapeutic effect. This study aimed to explore the regulatory mechanism of propofol in the pathogenesis of PC through mechanisms involving N6‐methyladenosine (m6A) modification. The changes in PC cell malignancy were evaluated by means of transwell, cell counting kit 8 (CCK‐8), western blotting and tumour xenograft model assays. Long noncoding RNA TRHDE‐AS1 and m6A methyltransferase METTL14 expression levels were determined via reverse transcription quantitative polymerase chain reaction (RT‐qPCR). The m6A modification of TRHDE‐AS1 which was mediated by METTL14 was confirmed by conducting methylated RNA immunoprecipitation (MeRIP) assay. We observed that propofol (200 μM) inhibited PC cell malignancy in vivo and in vitro, elucidating that it impaired cell proliferation, migration and tumour growth but induced apoptosis. TRHDE‐AS1 expression was observed to be lower in PC cells and tissues, and propofol induced TRHDE‐AS1 upregulation in PC cells. Propofol was capable of reversing the tumour‐promoting effect of TRHDE‐AS1 knockdown in PC cells. Additionally, METTL14 was upstream of TRHDE‐AS1 to induce m6A modification of TRHDE‐AS1 in PC cells. Collectively, our results show that propofol prevents PC progression by upregulating TRHDE‐AS1 expression and METTL14 is involved in the m6A modification of TRHDE‐AS1. These findings suggest that TRHDE‐AS1 may be a potential therapeutic target for the improvement of propofol's therapeutic effect. [ABSTRACT FROM AUTHOR]

Polyureas have been widely applied in many fields, such as coatings, fibers, foams and dielectric materials. Traditionally, polyureas are prepared from isocyanates, which are highly toxic and harmful to humans and the environment. Synthesis of polyureas via non-isocyanate routes is green, environmentally friendly and sustainable. However, the application of non-isocyanate polyureas is quite restrained due to their brittleness as the result of the lack of a soft segment in their molecular blocks. To address this issue, we have prepared polyester polyureas via an isocyanate-free route and introduced polyester-based soft segments to improve their toughness and endow high impact resistance to the polyureas. In this paper, the soft segments of polyureas were synthesized by the esterification and polycondensation of dodecanedioic acid and 1,4-butanediol. Hard segments of polyureas were synthesized by melt polycondensation of urea and 1,10-diaminodecane without a catalyst or high pressure. A series of polyester polyureas were synthesized by the polycondensation of the soft and hard segments. These synthesized polyester-type polyureas exhibit excellent mechanical and thermal properties. The synthesis of hybrid star-shaped polymers was carried out by atom transfer radical polymerization of n-butyl acrylate from a well-defined multifunctional titanium-oxo-cluster initiator. Conditions were identified to prevent possible side reactions among monomer, polymer, and the titanium-oxo-cluster ligands. Polymerizations provided linear first-order kinetics and the evolution of the experimental molecular weight is also linear with the conversion. 1H DOSY NMR and cleavage of the polymeric branches from the multifunctional initiator by hydrolysis were used to (i) prove the star-shaped structure of the polymer, and (ii) demonstrate that the shoulder observed on size exclusion chromatograms is not due to a noncontrolled polymerization but to ungrafting of polymeric branches during analysis. Rheological properties of the hybrid star-shaped poly( n-butyl acrylate) were studied in the linear regime and show that the Ti-oxo-cluster not only increases significantly the viscosity of the polymer relative to its ungrafted arm but has a rheological signature which is qualitatively different from that of stars with organic cores suggesting that the Ti cluster reduces significantly the molecular mobility of the star. © 2011 Wiley Periodicals, Inc. To study the effect of parecoxib sodium in alleviating inflammation in burned rats and restoring cognitive function in burned rats. 30 SPF grade SD rats were randomly divided into 6 groups: (1) Blank control group (Group C). (2) Sham surgery group (Group Sham). (3) Second-degree burn model (Group B). (4) Low-dose (1 mg/kg/d) parecoxib sodium (Group L+B). (5) Medium-dose (10 mg/kg/d) parecoxib sodium (Group M+B). (6) High-dose (20 mg/kg/d) parecoxib sodium (Group H+B). ELISA measures inflammatory factor IL-2, IL-6, TNF-α and IFN-γ, cognitive function factor NSE, cortisol and S-100β. Combined with water maze and dark avoidance experiments to further verify the recovery of cognitive function in rats. The contents of IL-2, TNF-α and IL-6 in Group M+B were significantly lower than those in Group Sham (P<0.05), and the content of IFN-γ was significantly lower than that in Group Sham (P<0.05). The cognitive markers NSE, S-100β and cortisol levels in Group M+B were significantly higher than those in Group Sham at 2h, 1d, 5d and 10d after operation (P<0.05). In the Group M+B dark-avoidance experiment, the number of probes and errors were not significantly different than those in Group Sham and Group C (P>0.05), and the number of times Group M+B found a platform in the water maze experiment and crossed the platform was second only to Group B and Group C. Parecoxib sodium can effectively reduce inflammation in burn rats and promote cognitive recovery in burn rats, and the optimal dose of parecoxib sodium for burn rats is 10 mg/kg., (© The Author(s) 2024. Background: Whether neuromuscular block (NMB) affects Intra-abdominal pressure (IAP) and cognition in Prostate cancer (PC) patients with Robotic-assisted laparoscopic radical prostatectomy (RALRP) remains unclear. Here we aimed to compare the effects of deep and moderate NMB on the IAP, inflammation, and cognition., Methods: The Moderate neuromuscular block (MNMB) group (N = 44) and Deep neuromuscular block (DNMB) group (N = 47) were recruited. Intra-abdominal pressure was adjusted to meet RALRP requirements. The expression of pro-inflammatory factors was measured by ELISA. MMSE scores were recorded before the operation, 1 day and 1 week after the operation., Results: Significant decreases in IAP (p < 0.001) and IL-1β, IL-6, TNF-a, and S-100β (p ≤ 0.01) expressions were found in the DNMB group. The MMSE score in the DNMB group was higher than that in the MNMB group on day one (p = 0.046). The incidence of nausea and vomiting was lower in the DNMB group than that in the MNMB group (p = 0.013)., Conclusions: DNMB reduces IAP and inflammation and improves post-operative cognitive functions in PC patients with RALRP., (© 2023 John Wiley & Sons Ltd.)

Objective: The objective is to compare the effect of general anesthesia (GA) and monitored anesthesia care (MAC) on clinical outcomes in patients with endovascular therapy for vertebrobasilar occlusion stroke., Methods: 139 patients undergoing endovascular therapy for vertebrobasilar stroke, were recruited. The patients were randomized into GA group and MAC group (about 1:1 ratio). GA group received general anesthesia and MAC group received monitored anesthesia care during endovascular therapy. The primary outcome measure was the shift in the degree of disability among the 2 groups as measured by the modified Rankin scale score (mRS) at 90 days (80-100 days). Secondary end points included infarct volume and related complications., Results: The patients were assigned randomly (about 1:1 allocation) to GA group (n=72) and MAC group (n=67). The primary outcome of functional independence measured by 90-day mRS score was not significantly different between the 2 groups (median (IQR), 2 (1-3) vs. 3 (1-4); P=0.316). Final infarct volume was smaller in the GA group than in the MAC group (median (IQR), 27.60 (13.75-83.52) vs. 33.60 (26.85-92.95); P=0.045). There were no differences with statistical significance in rates of successful reperfusion (modified Thrombolysis in Cerebral Ischemia (mTICI) 2b-3) between 2 groups (73.61% vs. 76.12%; P=0.734). Early neurological outcomes measured by the 24-hour National Institutes of Health Stroke Scale scores (NIHSS) showed that 11 (interquartile range (IQR), 3-22) in GA group and 11 (interquartile range (IQR), 7-25) in MAC group, but were not statistically significant. There was no statistical difference in postoperative complications between the two groups., Conclusion: For patients who underwent endovascular therapy for vertebrobasilar occlusion strok caused by occlusions in the posterior circulation, MAC appears to be as effective as GA.
