8. Axial Compressive Behavior of Cross-Shaped CFST Stub Columns with Steel Bar Truss Stiffening

Abstract

Concrete-filled steel tube (CFST) columns have been widely used in residential buildings due to their high bearing capacity, good ductility, and reliable seismic performance. However, conventional circular, square, or rectangular CFST columns may protrude from the adjacent walls, resulting in inconvenience in terms of the arrangement of furniture in a room. In order to solve the problem, special-shaped CFST columns, such as cross-shaped, L-shaped, and T-shaped columns, have been suggested and adopted in engineering practice. These special-shaped CFST columns have limbs with the same width as the adjacent walls. However, compared with conventional CFST columns, the special-shaped steel tube provides weaker confinement to the infilled concrete under axial compressive load, especially at concave corners. The parting at concave corners is the key factor affecting the bearing capacity and ductility of the members. Therefore, a cross-shaped CFST column with steel bar truss stiffening is suggested. In this paper, 12 cross-shaped CFST stub columns were designed and tested under axial compression loading. The effects of steel bar truss node spacing and column–steel ratio on the failure mode, bearing capacity, and ductility were discussed in detail. The results indicate that the columns with steel bar truss stiffening can change the final deformation mode of the steel plate from single-wave buckling to multiple-wave buckling, and the failure modes of columns also subsequently change from single-section concrete crushing failure to multiple-section concrete crushing failure. The steel bar truss stiffening shows no obvious effect on the axial bearing capacity of the member but significantly improves the ductility. The columns with a steel bar truss node spacing of 140 mm can only increase the bearing capacity by 6.8% while nearly doubling the ductility coefficient from 2.31 to 4.40. The experimental results are compared with those of six design codes worldwide. The results show that the Eurocode 4 (2004) and the Chinese code CECS159-2018 can be safely used to predict the axial bearing capacity of cross-shaped CFST stub columns with steel bar truss stiffening.

16. CRISPR/Cas9-mediated NlInR2mutants: Analyses of residual mRNA and truncated proteins

Abstract

CRISPR/Cas9 technology is a powerful genome manipulation tool in insects. However, little is known about whether mRNA and protein of a target gene are completely cleared in homozygous mutants. This study generated homozygous mutants of the insulin receptor gene 2 (NlInR2) in the brown planthopper (Nilaparvata lugens) using CRISPR/Cas9 genome editing. Both frameshift mutants, E5_D17 and E6_I7, differentiated towards long wings, but there were differences in wing morphology, with E5_D17 showing wing deformities. Subsequent investigations revealed the presence of residual expression of NlInR2mRNA in both mutants, as well as the occurrence of spliceosomes featuring exon skipping splicing in E5_D17. Additionally, the E5_D17 exhibited the detection of N-terminally truncated NlInR2 protein. RNA interference experiments indicated that the knockdown of NlInR2expression in the E5_D17 mutant line increased the proportion of wing deformities from 11.1 to 65.6%, suggesting that the residual NlInR2mRNA of the E5_D17 mutant might have retained some genetic functions. Our results imply that systematic characterization of residual protein expression or function in CRISPR/Cas9-generated mutant lines is necessary for phenotypic interpretation.

22. A rapid UPLC-MS/MS method for the determination of oleanolic acid in rat plasma and liver tissue: application to plasma and liver pharmacokinetics.

Abstract

A reliable high-throughput ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was developed and validated for oleanolic acid (OA) determination in rat plasma and liver tissue using glycyrrhetic acid as the internal standard (IS). Plasma and liver homogenate samples were prepared using solid-phase extraction. Chromatographic separation was achieved on a C18 column using an isocratic mobile phase system. The detection was performed by multiple reaction monitoring mode via positive electrospray ionization interface. The calibration curves showed good linearity ( R2 > 0.9997) within the tested concentration ranges. The lower limit of quantification for plasma and liver tissue was ≤0.75 ng/mL. The intra- and inter-day precision and accuracy deviations were within ±15% in plasma and liver tissue. The mean extraction recoveries ranged from 80.8 to 87.0%. In addition, the carryover, matrix effect, stability and robustness involved in the method were also validated. Abstract

The indolyl-4(3 H )-quinazolinone core is an important structural motif in functional molecules. However, few methods exist for its direct modification, which limits its potential application. Reported herein is a palladium-mediated amination of halogen-containing indolyl-4(3 H )-quinazolinones with a variety of primary and secondary amines via the corresponding palladium oxidative addition complexes. The protocol allows the facile synthesis of indolyl-4(3 H )-quinazolinone derivatives with amino groups at all the positions of the benzene ring in moderate to good yields with mild reaction conditions and good functional group tolerance. Furthermore, the antitumor activity of these products was evaluated.
