Abstract

PURPOSE: Identifying molecular and immune features to guide immune checkpoint inhibitor (ICI)-based regimens remains an unmet clinical need. EXPERIMENTAL DESIGN: Tissue and longitudinal blood specimens from phase III trial S1400I in patients with metastatic squamous non-small cell carcinoma (SqNSCLC) treated with nivolumab monotherapy (nivo) or nivolumab plus ipilimumab (nivo+ipi) were subjected to multi-omics analyses including multiplex immunofluorescence (mIF), nCounter PanCancer Immune Profiling Panel, whole-exome sequencing, and Olink. RESULTS: Higher immune scores from immune gene expression profiling or immune cell infiltration by mIF were associated with response to ICIs and improved survival, except regulatory T cells, which were associated with worse overall survival (OS) for patients receiving nivo+ipi. Immune cell density and closer proximity of CD8+GZB+ T cells to malignant cells were associated with superior progression-free survival and OS. The cold immune landscape of NSCLC was associated with a higher level of chromosomal copy-number variation (CNV) burden. Patients with LRP1B-mutant tumors had a shorter survival than patients with LRP1B-wild-type tumors. Olink assays revealed soluble proteins such as LAMP3 increased in responders while IL6 and CXCL13 increased in nonresponders. Upregulation of serum CXCL13, MMP12, CSF-1, and IL8 were associated with worse survival before radiologic progression. CONCLUSIONS: The frequency, distribution, and clustering of immune cells relative to malignant ones can impact ICI efficacy in patients with SqNSCLC. High CNV burden may contribute to the cold immune microenvironment. Soluble inflammation/immune-related proteins in the blood have the potential to monitor therapeutic benefit from ICI treatment in patients with SqNSCLC.

Abstract

A double-layered perovskite oxide Sr$_3$Mo$_2$O$_7$ is considered a "hidden ladder" system with wide and narrow bands near the Fermi level, for which high-$T_{\rm c}$ superconductivity is expected. However, the difficulty in synthesis, especially in the preparation of samples without oxygen deficiency, can hinder the observation of superconductivity. In this study, we constructed a double-layer SrMoO$_3$ block through artificial superlattices with the insulating SrTiO$_3$ block, (SrMoO$_3$)$_m$/(SrTiO$_3$)$_t$ ($m = 2, 4$; $t = 4$). First-principles calculations for bilayered SrMoO$_3$ ($m = 2$) exhibit a wide-narrow band structure near the Fermi level, which bears a close resemblance to Sr$_3$Mo$_2$O$_7$. The dispersion along the $k_z$ direction is strongly suppressed by increasing the number of the SrTiO$_3$ layers, $t$. However, no superconductivity is observed down to 0.1 K. Abstract

Given the existing COVID-19 pandemic worldwide, it is critical to systematically study the interactions between hosts and coronaviruses including SARS-Cov, MERS-Cov, and SARS-CoV-2 (cause of COVID-19). We first created four host-pathogen interaction (HPI)-Outcome postulates, and generated a HPI-Outcome model as the basis for understanding host-coronavirus interactions (HCI) and their relations with the disease outcomes. We hypothesized that ontology can be used as an integrative platform to classify and analyze HCI and disease outcomes. Accordingly, we annotated and categorized different coronaviruses, hosts, and phenotypes using ontologies and identified their relations. Various COVID-19 phenotypes are hypothesized to be caused by the backend HCI mechanisms. To further identify the causal HCI-outcome relations, we collected 35 experimentally-verified HCI protein-protein interactions (PPIs), and applied literature mining to identify additional host PPIs in response to coronavirus infections. The results were formulated in a logical ontology representation for integrative HCI-outcome understanding. Using known PPIs as baits, we also developed and applied a domain-inferred prediction method to predict new PPIs and identified their pathological targets on multiple organs. Overall, our proposed ontology-based integrative framework combined with computational predictions can be used to support fundamental understanding of the intricate interactions between human patients and coronaviruses (including SARS-CoV-2) and their association with various disease outcomes., Comment: 32 pages, 1 table, 6 figures

12. Giant enhancement of THz wave emission underdouble pulse excitation of thin water flow

Abstract

Simultaneous measurements of THz wave and hard X-ray emission from thin and flat water flow when irradiated by double femtosecond laser pulses (800 nm, 35 fs, transform-limited, 0.5 kHz, delay times up to 15 ns) were carried out at the transmission and the reflection sides of the flow with THz time-domain spectroscopy and Geiger counters. THz wave emission spectra show their dynamic peak-shifts toward the low frequency with the highest intensity enhancements more than 1.5 x 10^3 times in |E|^2 at the delay time of 4.7 ns between the two pulses, while X-ray intensity enhancements are limited to about 20 times at 0 ns under the same experimental conditions. The mechanisms for the spectral changes and the intensity enhancements in THz wave emission are discussed from the viewpoint of laser ablation on the water flow induced by the pre-pulse irradiation., Comment: 11 pages, 4 figures

16. Correlated emission of X-ray and sound from water film irradiated by femtosecond laser pulses

Abstract

Simultaneous measurements of hard X-ray by a Geiger counter and audible sound (10 Hz-20kHz) by a microphone from a thin water film in air were carried out under intense single and double pulse irradiations of femtosecond laser (35 fs, 800 nm, 1 kHz). Emission profiles of X-ray and sound under the single pulse irradiation by changing the water film position along the laser incident direction (Z-axis) show the same peak positions with a broader emission in sound (403{\mu}m at FWHM) than in X-ray (37{\mu}m). Under the double pulse irradiation condition with the time delay at 0 ps and 4.6 ns, it was clearly observed that the acoustic signal intensity is enhanced in associated with X-ray intensity enhancements. The enhancements can be assigned to laser ablation dynamics such as pre-plasma formation and transient surface roughness formation induced by the pre-pulse irradiation. For the acoustic signal under the double-pulse irradiation with the time delay, there was a weak dependence observed on the pre-pulse irradiation position at the laser focus. It is consistent with a long breakdown filament formation which makes the microphone-detection less position-sensitive.

43. THz Radiation from Subwavelength Volumes on Water Surfaces and in Air: Effects of Mass Density Gradients and External Magnetic Fields

Abstract

High-intensity ∼10 PW/cm2laser pulses with a 35 fs pulse duration are tightly focused into ∼ 8 μm in diameter focal spots to induce ionization and dielectric breakdown of 1) water microfilms and 2) air in the focal region and generate X-ray and THz waves. At the used focusing, there was no filamentation, and energy deposition took place within the geometrical confinement of the focal volume in air and on the water surface in the case of water films, and we explored very different mass-density conditions of medium for THz generation. The shockwave-induced mass-density gradients in air and at the front side of the water surface enhanced the X-ray and THz radiation when a prepulse (one or two) was time and space synchronized with the main pulse. At the optimized separation in space and the corresponding timing of the main pulse, X-ray and THz emissions were observed to be orders of magnitude stronger. Visualization of the main pulse interaction with the prepulse-triggered shockwave front was optically resolved and its handedness was measured in transmission and reflection from water targets. The mechanisms of X-ray and THz radiation are explained, and the effect of an externally applied magnetic field in different orientations is tested. It was observed that magnetic fields up to 0.6 T had only a minor effect on current transients, which were responsible for THz emission from the water microfilm target.

44. Parent–child math talk and early math interest: comparing the effects of written versus hands-on materials.

Abstract

This study investigated the impact of written and hands-on teaching materials on parent–child math talk and children’s interest in mathematics. Thirty parents and their preschool children participated in two play sessions, one using written material and the other using hands-on material, each lasting 12 min. At the end of each session, the children’s interest levels were assessed, and the frequency of math-related conversations was analysed. Regardless of the materials used, discussions primarily revolved around fundamental numerical concepts, with little mention of practical measurement units and conventional nominatives or number comparisons involving the application of mental representation. Ordinal and division concepts were only mentioned by parents in the written scenario rather than in the hands-on one. Overall, using written material led to doubling math discourse compared to hands-on material. However, children showed slightly higher interest levels in the hands-on scenario, where parent–child math talk categories also exhibited greater synchrony. Abstract

Laser machining by ultra-short (sub-ps) pulses at high intensity offers high precision, high throughput in terms of area or volume per unit time, and flexibility to adapt processing protocols to different materials on the same workpiece. Here, we consider the challenge of optimization for high throughput: how to use the maximum available laser power and larger focal spots for larger ablation volumes by implementing a fast scan. This implies the use of high-intensity pulses approaching ∼PW/cm2 at the threshold where tunneling ionization starts to contribute to overall ionization. A custom laser micromachining setup was developed and built to enable high speed, large-area processing, and easy system reconfiguration for different tasks. The main components include the laser, stages, scanners, control system, and software. Machining of metals such as Cu, Al, or stainless steel and fused silica surfaces at high fluence and high exposure doses at high scan speeds up to 3 m/s were tested for the fluence scaling of ablation volume, which was found to be linear. The largest material removal rate was 10 mm3/min for Cu and 20 mm3/min for Al at the maximum power 80 W (25 J/cm2 per pulse). Modified surfaces are color-classified for their appearance, which is dependent on surface roughness and chemical modification.
