16. Prediction of folding transition-state position (βT) of small, two-state proteins from local secondary structure content.

Abstract

Folding kinetics of proteins is governed by the free energy and position of transition states. But attempts to predict the position of folding transition state on reaction pathway from protein structure have been met with only limited success, unlike the folding-rate prediction. Here, we find that the folding transition-state position is related to the secondary structure content of native two-state proteins. We present a simple method for predicting the transition-state position from their α-helix, turn and polyproline secondary structures. The method achieves 81% correlation with experiment over 24 small, two-state proteins, suggesting that the local secondary structure content, especially for content of α-helix, is a determinant of the solvent accessibility of the transition state ensemble and size of folding nucleus. 17. Template imprinting amphoteric polymer for the recognition of proteins

Abstract

A selective recognizing of proteins with amphoteric polymer is essential for application in protein separations. An amphoteric polymer was synthesized by copolymerization of the two functional monomers with the crosslinker in the presence of the template proteins. After extraction of the template protein, the polymer was applied as chromatographic stationary phase, showing high resolution capacity. The molecular imprinting technique allows the formation of specific recognition sites in amphoteric polymers through the use of template protein. Protein is preferentially bound to a polymer made in the presence of the same protein. The recognition ability of an amphoteric polymer is attributed to imprinted cavities that complement proteins in shape and electrostatic interactions. © 2004 Wiley Periodicals, Inc. Abstract

4-(N,N-Diallylamino)pyridine (DAAP), N,N-diallylaminobenzene (DAAB), N,N,N',N'-tetrallyl-4,4'-diaminobenzidine (AAB), N,N,N',N'-tetrallyl-4,4'-diaminodiphenyl sulfone (AABS), and N,N,N',N'-tetrallyl-4,4'-diaminodiphenyl ether (AABE) were prepared by sodium substitution and N-allylation. Moreover, linear polyDAAP, poly(DAAP-co-DAAB), and network poly(DAAP-co-AAB), poly(DAAP-co-AABS), and poly(DAAP-co-AABE), all being polymers containing supernucleophilic groups, were synthesized in the cyclopolymerization. © 2000 John Wiley & Sons, Inc.
