Abstract

Let $G=\left\langle S|R_{A}\right\rangle $ be a semigroup with generating set $ S$ and equivalences $R_{A}$ among $S$ determined by a matrix $A$. This paper investigates the complexity of $G$-shift spaces by yielding the topological entropies. After revealing the existence of topological entropy of $G$-shift of finite type ($G$-SFT), the calculation of topological entropy of $G$-SFT is equivalent to solving a system of nonlinear recurrence equations. The complete characterization of topological entropies of $G$-SFTs on two symbols is addressed, which extends [Ban and Chang, arXiv:1803.03082] in which $G$ is a free semigroup.

Abstract

Three-dimensional green biomass can indicate the benefits of urban ecosystems. Sixty representative plant communities (20 m × 20 m) in seven parks were selected as sample plots in Nanping City, China. The structural characteristics, three-dimensional green biomass per square meter (PGB), and ecological benefit value of the plant communities in each plot were measured. The results showed that the PGB of Jiufengshan Park was the highest, reaching 5.60 m³·m-2. The Random forest model revealed that the coverage, diameter at breast height (DBH), and tree number had a highly significant effect on PGB (P < 0.01), and tree crown width had a significant effect on the PGB (P < 0.05). Therefore, the PGB of parks can be improved by improving the coverage, DBH, tree number, and tree crown width, which would help to increase the ecological benefit value and well - being of human beings. 19. MicroRNA governs bistable cell differentiation and lineage segregation via a noncanonical feedback

Abstract

Positive feedback driven by transcriptional regulation has long been considered a key mechanism underlying cell lineage segregation during embryogenesis. Using the developing spinal cord as a paradigm, we found that canonical, transcription-driven feedback cannot explain robust lineage segregation of motor neuron subtypes marked by two cardinal factors, Hoxa5 and Hoxc8. We propose a feedback mechanism involving elementary microRNA-mRNA reaction circuits that differ from known feedback loop-like structures. Strikingly, we show that a wide range of biologically plausible post-transcriptional regulatory parameters are sufficient to generate bistable switches, a hallmark of positive feedback. Through mathematical analysis, we explain intuitively the hidden source of this feedback. Using embryonic stem cell differentiation and mouse genetics, we corroborate that microRNA-mRNA circuits govern tissue boundaries and hysteresis upon motor neuron differentiation with respect to transient morphogen signals. Our findings reveal a previously underappreciated feedback mechanism that may have widespread functions in cell fate decisions and tissue patterning.

29. Vaccination to reduce severe COVID-19 and mortality in COVID-19 patients: a systematic review and meta-analysis.

Abstract

OBJECTIVE: The outbreak of coronavirus disease (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARSCoV-2), has resulted in the death of up to 5 million people worldwide, with a mortality rate of approximately 2%. Wearing masks, maintaining social distance, tracking, and isolating close contacts are not sufficient to control the epidemic. The effectiveness of vaccines is affected by the willingness of people to be vaccinated. Therefore, in this review, we aimed to examine the efficacy of different types of vaccines in reducing hospitalization rates, disease severity, and mortality. MATERIALS AND METHODS: We searched five databases (Embase, PubMed, Cochrane, EBSCO, and CEPS) for related research on September 3, 2021. We used a random-effects model for analysis. RESULTS: Seven studies were identified, involving 1,366,700 participants (689,967 participants in the vaccinated group and 676,733 participants in the non-vaccinated group). There were 292 significant incidents (56 in the vaccinated group and 236 in the non-vaccinated group) with a risk ratio of 0.12 and a 95% confidence interval of 0.040-0.363. Compared with no vaccine, all types of vaccines can effectively prevent the rate of severe illness. CONCLUSIONS: We evaluated whether different brands of vaccines or types of COVID-19 vaccines could prevent the risk of severe illness after diagnosis. The analysis showed that all types of vaccines can effectively prevent severe disease. Implementing epidemic prevention guidelines and obtaining vaccines in different countries can improve vaccine protection and reduce COVID-19-related deaths worldwide. [ABSTRACT FROM AUTHOR]

Abstract

The osteogenic potential of mesenchymal stromal cells (MSCs) varies among different tissue sources. Strontium enhances the osteogenic differentiation of bone marrow-derived MSCs (BM-MSCs), but whether it exerts similar effects on placental decidual basalis-derived MSCs (PDB-MSCs) remains unknown. Here, we compared the influence of strontium on the proliferation and osteogenic differentiation of human PDB- and BM-MSCs in vitro. We found that 1 mM and 10 mM strontium, but not 0.1 mM strontium, evidently promoted the proliferation of human PDB- and BM-MSCs. These doses of strontium showed a comparable alkaline phosphatase activity in both cell types, but their osteogenic gene expressions were promoted in a dose-dependent manner. Strontium at doses of 0.1 mM and 1 mM elevated several osteogenic gene expressions of PDB-MSCs, but not those of BM-MSCs at an early stage. Nevertheless, they failed to enhance the mineralization of either cell type. By contrast, 10 mM strontium facilitated the osteogenic gene expression as well as the mineralization of human PDB- and BM-MSCs. Collectively, this study demonstrated that human PDB- and BM-MSCs shared a great similarity in response to strontium, which promoted their proliferation and osteogenic differentiation in a dose-dependent manner.

42. Interrupting TGF-β1/CCN2/integrin-α5β1 signaling alleviates high mechanical-stress caused chondrocyte fibrosis.

Abstract

OBJECTIVE: Mechanical-stress has been reported to trigger cartilage fibrosis, in which transforming growth factor (TGF)-β and connective tissue growth factor (CCN2) are involved. However, the function of integrin-a5β1, a cytomembrane receptor, on mechanical-stress related fibrosis has not yet been elucidated. This study aims to reveal the interaction of TGF-β1/CCN2/integrin-α5β1 in the mechanical-stress induced chondrocyte (CH) fibrosis. PATIENTS AND METHODS: We used different levels (5% and 10%) of cyclic tension simulation (CTS) to stretch CHs and observed the gene expression of TGF-β1/CCN2/integrin-α5β1 as well as the fibrous related genes containing collagen I/II/III, Runx2, MMP13, and ADAMTS-5 by real-time polymerase chain reaction (RT-PCR) or immunofluorescence. We used the siRNA or the corresponding antagonist of TGF-β1, CCN2, integrin-α5β1 during the CTS to clear the effect of them in the fibrosis progress. In addition, to verify the crosstalk between TGF-β1, CCN2, and integrin-α5β1, we used the recombinant human (rh)-TGF-β1 and CCN2 to culture CHs without CST. RESULTS: 24 hours-10% CTS was sufficient to induce a decrease of collagen II and increase the collagen I/III, Runx2, MMP13, and ADAMTS-5 gene expression. Under CTS, TGF-β1 silencing resulted in a decline of CCN2, integrin-α5β1, and alleviated the CHs fibrosis. Apart from this, blocking CCN2 or integrin-α5β1 expression also contributed to the suppression of 10% CTS induced CHs fibrosis. Meanwhile, the exogenic protein supplement raised the cellular TGF-β1 or CCN2 expression and increased the integrin-α5β1 mRNA level. However, the downregulation of TGF-β1 or CCN2 did not affect integrin-α5β1 expression, whether the CTS exited or not. CONCLUSIONS: High mechanical-stress induces CHs fibrosis via the activation of TGF-β1/CCN2/integrin-α5β1 signaling, and interrupting the TGF-β1, CCN2, or integrin-α5β1 expression can alleviate the fibrous process. 45. Abnormal blink reflex recovery cycle in manifesting and nonmanifesting carriers of the DYT1 gene mutation

Abstract

The aim of this study is to evaluate the brainstem function in DYT1 carriers manifesting clinical dystonia (MDYT1) and those without clinical symptoms (NMDYT1). Motor cortical inhibition and plasticity were found to be abnormal in MDYT1, whereas these were less abnormal in NMDYT1. However, the spinal reciprocal inhibition was abnormal in MDYT1, but normal in NMDYT1. Moreover, protein accumulation and perinuclear inclusion bodies were found in the brainstem, but not in other brain areas, in DYT1 patients. Therefore, we designed this study to investigate the brainstem physiology using the blink reflex (BR) recovery cycle test in MDYT1 and NMDYT1. We recruited eight MDYT1, five NMDYT1, and nine age-matched healthy controls. The BR recovery cycle was assessed with paired stimuli that induced the BR in a random order at interstimulus intervals of 250, 500, and 1000 ms. A two-way analysis of variance showed a significant difference between MDYT1, NMDYT1, and the healthy control (P=0.004). Post-hoc analysis showed that this was because of a significantly lower inhibition of R2 in MDYT1 and NMDYT1 compared with the controls (two-way analysis of variance: P=0.003 and 0.021, respectively). There was no difference between MDYT1 and NMDYT1 (P=0.224). The tested brainstem circuits were equally involved in MDYT1 and NMDYT1. The finding is in agreement with the pathological findings in DYT1 carriers. Together with previous findings in the motor cortex and the spinal cord, the brainstem may lie closer to the pathogenesis of dystonia than the motor cortex in DYT1 gene carriers.
