Abstract

Purpose: To develop a circulating tumor DNA (ctDNA) methylation signature for early detection of metastatic relapse and surveillance of therapeutic efficacy in stage III CRC. Perimental design: The deferentially methylated locations (DMLs) between 52 normal intestinal mucosa and 134 stage III CRC tissue and between plasma from 96 stage I/II CRC patients and 73 stage IV CRC patients were detected by high-throughput targeted methylation sequencing. CtDNA methylation signature was established by random forest with DMLs shared by plasma and tissue samples. This signature was then applied to predict the risk of metastatic disease recurrence in blood samples from 50 stage III CRC patients and dynamic monitor the recurrence risk in serial blood samples from 4 CRC patients. Results: 2237 and 1185 hypermethylation markers were identified in CRC tissue and plasma from stage IV CRC patients, respectively. Our signature achieved the sensitivity of 84.21% and specificity of 81.81% in distinguishing late CRC from early CRC in the validation cohort. Moreover, this signature yielded high sensitivity (87.5%) and specificity (94.12%) for predicting metastatic disease relapse in a cohort of 50 stage III CRC patients, and could be an independent risk factor for relapse in those patients (HR, 18.7; 95% CI, 5.3-66.2; P = 9.86e-6). The analyses of serial blood samples of CRC showed that this model was more sensitive to monitor treatment response than imaging examination and serum carcinoembryonic antigen (CEA). Conclusions: We identify a ctDNA methylation signature for stratifying risk of recurrence in stage III CRC and non-invasively monitoring treatment response.Statement of translational relevanceWe successfully developed a ctDNA methylation signature for risk assessment of metastatic disease recurrence in stage III CRC patients and surveillance of therapeutic efficacy of CRC. Abstract

The article discusses a study which determined the morphology of perfluorosulfonic ionomer (PFSI) molecules in various concentrations. The study involved the measurement of PFSI using rheological dynamic light scattering (DLS) and scanning electron microscopy (SEM). Ionomer molecules decreased in size at higher concentrations of PFSI in N,N-dimethylformamide (DMF). A high temperature and high solution concentration preparation of the PFSI for fusing the particles of the membrane is recommended.

44. Controlled“Living” Radical Polymerization of Methyl Methacrylate Catalyzed by Cobalt Acetate

Abstract

Using cobalt acetate as a catalyst, the controlled“living” polymerization of MMA was carried out. PMMA with a predicted molecular weight and a low polydispersity $(\overline M _{\rm w} /\overline M _{\rm n} = 1.26)$was obtained. Using the obtained PMMA as a macroinitiator, a chain extension reaction was carried out. This increased the molecular weight of PMMA from 23 600 to 36 100, in good agreement with the predicted value, and the polydispersity index became lower $(\overline M _{\rm w} /\overline M _{\rm n} = 1.22)$. This indicated the “living” character of the polymerization. Bulk polymerization of styrene catalyzed by cobalt acetate using the obtained polystyrene as a macroinitiator yielded welldefined PSbPMMA.

45. Exploration of multigene, multistep and multi-pathway models of nasopharyngeal and colorectal carcinogenesis

Abstract

Abstract: Objective: To construct tree models for nasopharyngeal carcinoma (MPC) and colorectal carcinoma (CC) and explore the oncogeneic process of NPC and CC. Methods: Based on the software that Desper et al. developed, tree models were constructed for CC from the comparative genomic hybridization (CGH) data of 118 CC patients and for NPC from the CGH data of 140 southern Chinese patients, respectively. Results: Tree models for CC suggested that loss of 18q and gain of 20q were important early events in colorectal carcinogenesis. As changes in - 18q occurred prior to those in —17p, a cause-effect relationship might exist between them. Tree models for NPC suggested that loss of 3p was an important early event in nasopharyngeal carcinogenesis, and deletion of 11q. 14q. 16q and 9p were also nonrandom genetic events in carcinogenesis. suggesting that there might be tumor -associated genes existing on these chromosome arms. The tree model also indicated the existence of oncogenes on the short arm of chromosome 12. Conclusion: Constructing tree models based on the CGH data to demonstrate the initiation and progression of NPC might help elucidate its multigene, multistep and muitipathway development. It may provide valuable clues to explore the mechanism of tumorigenesis.

46. Ultrasensitive detection of IgE levels based on magnetic nanocapturer linked immunosensor assay for early diagnosis of cancer

Abstract

Rapid and accurate detection of immunoglobulin E (IgE) in serum and reduction of serum dosage are of great significance for clinical detection. Herein, we described a rapid magnetic separation of IgE from patient serum based on Fe3O4@SiO2-NTA@026 sdab as the capture probe and multiple horseradish peroxidase (HRP)-labeled antibodies linked gold nanoparticles (AuNPs) as chemiluminescence (CL) signal amplifier for ultrasensitive detection of total IgE. Results showed that the limit of detection of our immunosensor system in serum samples was 0.03 kU/L, which is lowest in comparison with current methods, and far lower than that of ImmunoCAP for IgE detection (0.1 kU/L). Furthermore, our immunosensor possessed satisfied repeatability and accuracy, as well as good stability. In comparison with the ImmunoCAP for the quantitative detection of IgE, highly consistent results were achieved in 20 serum samples. Specially, this method was also successfully utilized for assessing the IgE traces in breast cancer patients, which provides a new idea for the diagnosis of early cancer. Therefore, we believe that such versatile immunosensor will offer an alternative method for the on-site monitoring and determination of various IgE-related diseases.

Abstract

Objective: To investigate the effect of exosomes derived from Epstein-Barr virus (EBV)-positive nasopharyngeal carcinoma (NPC) cells on lymphangiogenesis and lymph node metastasis of NPC., Methods: Exosomes from NP69 cells and EBV-positive HK1 (HK1-EBV) cells were obtained by ultracentrifugation and identified by Western blotting and nanoparticle tracking analysis. Dio dye phagocytosis test was performed to observe exosome uptake by lymphatic endothelial cells. Lymphatic endothelial cells were treated with exosomes from nasopharyngeal epithelium (NP69), HK1-EBV, and C666-1 cells or exosome-free supernatant of HK1-EBV and C666-1 cells, and tube formation and migration of the cells were observed. In a nude mouse model of popliteal lymph node metastasis of NPC, the effects of normal saline, NP69 cell-derived exosomes, HK1-EBV cell-derived exosomes, exosome-free supernatant of HK1-EBV cells, and HK1-EBV exosome-free supernatant protein on lymphangiogenesis and lymph node metastasis of the tumor were observed., Results: The exosomes obtained by ultracentrifugation contained abundant exosome-specific proteins and showed a normal size range. The exosomes from NPC cells and NP69 cells could be taken up by lymphatic endothelial cells. Compared with the blank control and exosomes form NP69 cells, exosomes derived from HK1-EBV and C666-1 cells significantly promoted tube formation and migration of lymphatic endothelial cells ( P < 0.05), and the exosomes and exosome-free supernatant of HK1-EBV and C666-1 cell produced similar effects ( P > 0.05). In the tumor-bearing nude mice, exosomes derived from HK1-EBV cells significantly promoted metastasis of NPC cells and local lymphangiogenesis compared with the blank control, NP69 cell-derived exosomes and exosome-free supernatant of HK1-EBV cells ( P < 0.05)., Conclusions: Exosomes from EBV-positive NPC cells can significantly promote lymphangiogenesis and lymph node metastasis of NPC.

Abstract

Objective: To screen differentially expressed genes and identify potential signaling pathway in Asian people with breast cancer., Methods: Five gene microarray datasets of Asian people with breast cancer, GSE6367, GSE9309, GSE15852, GSE33447 and GSE45255, were downloaded from GEO. Microarrays with 318 breast cancer and 60 normal breast tissues were used for analysis of differentially expressed genes and pathway. 32 pairs of breast cancer patients' specimens were used to validate the differentially expressed genes by real-time PCR., Results: Analysis of the large sample of microarray data identified 436 differentially expressed genes in breast cancer tissues, while 259 of these genes were up-regulated and the other 177 down-regulated. Pathway analysis showed that metabolism-related signaling pathway may be involved in the development of breast cancer in Asian people. The expressions of KRT19, ADIPOQ, CFD, RBP4, LPL, ABCA8 and CD36 genes were confirmed by real-time PCR., Conclusion: This study shows differential gene expression profile and potential signaling pathway in Asian people with breast cancer. CD36 gene may be closely related to the Asian breast cancer. ABCA8 gene may be a new disease gene in Asian breast cancer.

Abstract

Objective: To analyze the dysregulated genes among the differentially expressed genes in 41 nasopharyngeal biopsy samples and identify their protective transcriptional factors., Methods: The differentially expressed gene profiles were obtained by analyzing both types I and II nasopharyngeal carcinoma (NPC&#95;I and NPC&#95;II, respectively) using EXCEL and Bioinformatics tools. The transcriptional factors were further studied only when (1) the difference in the binding sites of the differentially expressed genes between NPC&#95;I and NPC&#95;II groups was statistically significant, (2) the expressions of the transcription factors were correlated with the gene expressions in the samples, and (3) the transcription factors affected at least 40% of the expression of the related genes., Results: In NPC&#95;I samples, 80 transcription factors were found to be up-regulated, in which RUNX3, GATA3, NR3C1, NRF1, RXRA, SMAD7, TBP, and ZBTB6 were positive factors and HLF and MTF1 were negative factors, involved in the regulation of the genes in T cell receptor signaling pathway. No eligible transcription factors were found in association with down-regulated genes in NPC&#95;I compared to NPC&#95;II gene expression profiles., Conclusions: The over-expressed genes in NPC&#95;I are mainly related to immune responses, and we found 8 positive factors and 2 negative factors that regulate the genes in T cell receptor signaling pathway. The 10 transcription factors may serve as potential therapeutic targets for NPC&#95;I. We failed to identify any transcription factors associated with down-regulated genes in NPC&#95;I relative to NPC&#95;II possibly as a result of multiple factors that affect the differential gene expressions in NPC&#95;II including the transcription factors, DNA phosphorylation and modification, chromosome variation and environmental factors.
