Abstract

Background Insulin dysregulation (ID) is an equine endocrine disorder, which is often accompanied by obesity and various metabolic perturbations. The relationship between weight variations and fluctuations of the insulin response to oral glucose tests (OGT) as well as the metabolic impact of ID have been described previously. The present study seeks to characterize the concomitant metabolic impact of variations in the insulin response and bodyweight during repeated OGTs using a metabolomics approach. Methods Nineteen Icelandic horses were subjected to five OGTs over one year and their bodyweight, insulin and metabolic response were monitored. Analysis of metabolite concentrations depending on time (during the OGT), relative bodyweight (rWeight; defined as the bodyweight at one OGT divided by the mean bodyweight across all OGTs) and relative insulin response (rAUCins; defined accordingly from the area under the insulin curve during OGT) was performed using linear models. Additionally, the pathways significantly associated with time, rWeight and rAUCins were identified by rotation set testing. Results The results suggested that weight gain and worsening of ID activate distinct metabolic pathways. The metabolic profile associated with weight gain indicated an increased activation of arginase, while the pathways associated with time and rAUCins were consistent with the expected effect of glucose and insulin, respectively. Overall, more metabolites were significantly associated with rWeight than with rAUCins.

Abstract

The rapid development of precocial goats in the first weeks after birth requires an adequate adaptation of phosphate transport systems to maintain the P homeostasis at each developmental stage. Here we examined the age-related development of [Na.sup.+]-[P.sub.i] transport systems in small intestines, kidneys, and parotid glands of goats. Kinetic parameters were determined by brush-border membrane vesicle uptake studies, and relative expression of NaPi type II mRNA and protein was recorded by molecular biological methods. High intestinal [P.sub.i] transport capacity was already present on the first day of life. Within the first 3 wk of life there seemed to be a change in the type of [Na.sup.+]-dependent [P.sub.i] transporter, and NaPi IIb was expressed increasingly up to the fifth month of life. Renal [Na.sup.+]-[P.sub.i] transport capacity was also high at birth, and this was associated with high expression levels of NaPi IIa mRNA, indicating the important role of this transporter for renal [P.sub.i] reabsorption. At weaning an increase in both intestinal and renal [Na.sup.+]-[P.sub.i] transport balanced the increasing requirements for [P.sub.i] to establish the endogenous [P.sub.i] cycle. Salivary [P.sub.i] concentration and parotid NaPi II mRNA rose markedly to guarantee an adequate [P.sub.i] supply for rumen microbes. We concluded that the high demand for [P.sub.i] in young goats was assured by high basal [Na.sup.+]-[P.sub.i] transport capacity of small intestines and kidney expressed continuously during ontogenesis. ontogenesis of sodium-phosphate transport; ruminants; small intestine; kidney; parotid gland; NaPi type II cotransporters

Abstract

Due to intensive intracellular metabolism of short-chain fatty acids, ruminal epithelial cells generate large amounts of D-[beta]-hydroxybutyric acid, acetoacetic acid, and lactic acid. These acids have to be extruded from the cytosol to avoid disturbances of intracellular pH ([pH.sub.i]). To evaluate acid extrusion, [pH.sub.i] was studied in cultured ruminal epithelial cells of sheep using the pH-sensitive fluorescent dye 2', 7'-bis(2-carboxyethyl)-5(6)-carboxyfluorescein. Extracellular addition of D-[beta]-hydroxybutyrate, acetoacetate, or lactate (20 mM) resulted in intracellular acidification. Vice versa, removing extracellular D-[beta]-hydroxybutyrate, acetoacetate, or lactate after preincubation with the respective monocarboxylate induced an increase of [pH.sub.i]. Initial rate of phi decrease as well as of [pH.sub.i] recovery was strongly inhibited by pCMBS (400 [micro]M) and phloretin (20 [micro]M). Both cultured cells and intact ruminal epithelium were tested for the possible presence of proton-linked monocarboxylate transporter (MCT) on both the mRNA and protein levels. With the use of RT-PCR, mRNA encoding for MCT1 isoform was demonstrated in cultured ruminal epithelial cells and the ruminal epithelium. Immunostaining with MCT1 antibodies intensively labeled cultured ruminal epithelial cells and cells located in the stratum basale of the ruminal epithelium. In conclusion, our data indicate that MCT1 is expressed in the stratum basale of the ruminal epithelium and may function as a main mechanism for removing ketone bodies and lactate together with [H.sup.+] from the cytosol into the blood. intracellular pH; short-chain fatty acids; intracellular metabolism

Abstract

Endogenous [P.sub.i] recycling is a characteristic feature of the P homeostasis in ruminants. A pronounced salivary [P.sub.i] secretion into the rumen is balanced by a high intestinal [P.sub.i] absorption and an almost complete renal [P.sub.i] reabsorption. In monogastric animals, the major [P.sub.i] transport mechanism across the apical membrane of the enterocyte is an [Na.sup.+]-dependent transport mediated by NaPi cotransporter type IIb. In ruminants, an [Na.sup.+]-, as well as an [H.sup.+]-dependent, [P.sub.i] transport system seems to exist in the small intestines. Therefore, morphological localization, type of ionic dependence, and ability to adapt to dietary P or Ca restriction of duodenal and jejunal [P.sub.i] transport were characterized in goats. In the duodenum, there was an [H.sup.+]-dependent, [Na.sup.+]-sensitive [P.sub.i] transport system that did not belong to the NaPi type II family and was not influenced by dietary P or Ca restriction. In contrast, in the jejunum, there was an [Na.sup.+]-dependent, [H.sup.+]-sensitive [P.sub.i] transport mainly mediated by NaPi IIb.
