Abstract To evaluate the dosimetric benefits and clinical feasibility of deep inspiratory breath-hold (DIBH) combined with volumetric modulated arc therapy (VMAT) in left-sided postmastectomy radiotherapy (PMRT). Eligible patients with left-sided breast cancer undergoing DIBH-based PMRT were prospectively included. Chest wall, supra/infraclavicular fossa, and/or internal mammary node irradiation (IMNI) were planned with a prescription dose of 43.5 Gy in 15 fractions. VMAT plans were designed on free breathing (FB)—and DIBH-CT to compare dosimetric parameters in heart, left anterior descending artery (LAD) and lung. Cone-beam computed tomography (CBCT) was performed before and after treatment to evaluate inter- and intra-fractional setup errors. Heart position and dose variations during treatment were estimated by fusing CBCT with DIBH-CT scans.Twenty patients were included with 10 receiving IMNI. Abstract Physics informed neural network (PINN) demonstrates powerful capabilities in solving forward and inverse problems of nonlinear partial differential equations (NLPDEs) through combining data-driven and physical constraints. In this paper, two PINN methods that adopt tanh and sine as activation functions, respectively, are used to study data-driven solutions and parameter estimations of a family of high order KdV equations. Compared to the standard PINN with the tanh activation function, the PINN framework using the sine activation function can effectively learn the single soliton solution, double soliton solution, periodic traveling wave solution, and kink solution of the proposed equations with higher precision. The PINN framework using the sine activation function shows better performance in parameter estimation. In addition, the experiments show that the complexity of the equation influences the accuracy and efficiency of the PINN method. The outcomes of this study are poised to enhance the application of deep learning techniques in solving solutions and modeling of higher-order NLPDEs.

Abstract In recent years, defending against adversarial examples has gained significant importance, leading to a growing body of research in this area. Among these studies, pre-processing defense approaches have emerged as a prominent research direction. However, existing adversarial example pre-processing techniques often employ a single pre-processing model to counter different types of adversarial attacks. Such a strategy may miss the nuances between different types of attacks, limiting the comprehensiveness and effectiveness of the defense strategy. To address this issue, we propose a divide-and-conquer reconstruction pre-processing algorithm via multi-classification and multi-network training to more effectively defend against different types of mainstream adversarial attacks. The premise and challenge of the divide-and-conquer reconstruction defense is to distinguish between multiple types of adversarial attacks. Our method designs an adversarial attack classification module that exploits the high-frequency information differences between different types of adversarial examples for their multi-classification, which can hardly be achieved by existing adversarial example detection methods. In addition, we construct a divide-and-conquer reconstruction module that utilizes different trained image reconstruction models for each type of adversarial attack, ensuring optimal defense effectiveness. Extensive experiments show that our proposed divide-and-conquer defense algorithm exhibits superior performance compared to state-of-the-art pre-processing methods.

Abstract Integrating flexible electronics and photonics can create revolutionary technologies, but combining these components on a single polymer device has been difficult, particularly for high-volume manufacturing. Here, we present a robust chiplet-level heterogeneous integration of polymer-based circuits (CHIP), where ultrathin polymer electronic and optoelectronic chiplets are vertically bonded at room temperature and shaped into application-specific forms with monolithic Input/Output (I/O). This process was used to develop a flexible 3D integrated optrode with high-density microelectrodes for electrical recording, micro light-emitting diodes (μLEDs) for optogenetic stimulation, temperature sensors for bio-safe operations, and shielding designs to prevent optoelectronic artifacts. CHIP enables simple, high-yield, and scalable 3D integration, double-sided area utilization, and miniaturization of connection I/O. Systematic characterization demonstrated the scheme’s success and also identified frequency-dependent origins of optoelectronic artifacts. We envision CHIP being applied to numerous polymer-based devices for a wide range of applications.

Objective: To evaluate whether improved progression-free survival (PFS) from radiotherapy (RT) translates into an overall survival (OS) benefit for diffuse large B-cell lymphoma (DLBCL). Methods: A systematic literature search identified randomized controlled trials (RCTs) and retrospective studies that compared combined-modality therapy (CMT) with chemotherapy (CT) alone. Weighted regression analyses were used to estimate the correlation between OS and PFS benefits. Cohen's kappa statistic assessed the consistency between DLBCL risk-models and PFS patterns. Furthermore, the benefit trend of RT was analyzed by fitting a linear regression model to the pooled hazard ratio (HR) according to the PFS patterns. Results: For both 7 RCTs and 52 retrospective studies, correlations were found between PFS HR (HRPFS) and OS HR (HROS) at trial level (r = 0.639–0.876), and between PFS and OS rates at treatment-arm level, regardless of CT regimens (r = 0.882–0.964). Incorporating RT into CT increased about 18% of PFS, and revealed a different OS benefit profile. Patients were stratified into four CT-generated PFS patterns (>80%, >60–80%, >40–60%, and ≤40%), which was consistent with risk-stratified subgroups (kappa > 0.6). Absolute gain in OS from RT ranged from ≤5% at PFS >80% to about 21% at PFS ≤40%, with pooled HROS from 0.70 (95% CI, 0.51–0.97) to 0.48 (95% CI, 0.36–0.63) after rituximab-based CT. The OS benefit of RT was predominant in intermediate- and high-risk patients with PFS ≤ 80%. Conclusion: We demonstrated a varied OS benefit profile of RT to inform treatment decisions and clinical trial design.

Daphnetin, a coumarin derivative isolated from Daphne odorifera, has anti-tumor effects. The MAPK, STAT3, and NF-κB signaling pathways are closely related to the pathogenesis of lung cancer. To investigate the effect of daphnetin on anti-lung adenocarcinoma A549 cells and its mechanism. The anti-tumor effects of daphnetin on the proliferation, clone formation, migration, and invasion of A549 lung adenocarcinoma cells were investigated. The results showed that daphnetin inhibited the proliferation, colony formation, migration, and invasion of A549 cells through the MAPK/STAT3/NF-KB pathway, and mainly inhibited the clonal formation and migration of A549 cells through the JNK pathway. These results provide a new research direction and theoretical basis for the use of daphnetin in the inhibition of lung adenocarcinoma.

Background: Chemoradiotherapy (CRT) is the main treatment for elderly patients with non-metastatic rectal cancer who are ineligible for or decline surgery, but the optimal modality remains unclear. Objectives: This study was to validate the safety and efficacy of comprehensive geriatric assessment (CGA) guided radiotherapy in older patients. Design: An exploratory analysis of a single-arm, multicenter, Phase II trial. Methods: Patients aged over 70 and diagnosed with rectal cancer were enrolled and evaluated by CGA. CGA-guided radiotherapy was individually conducted in a multidisciplinary setting. Patients in fit, intermediate, and frail groups were scheduled to receive CRT, long-course radiotherapy, and short-course radiotherapy (SCRT) alone respectively. Patients who were unfit for or refused surgery were analyzed for acute toxicities and survival outcomes. Results: In a total of 109 enrolled patients, 47 individuals who did not undergo surgery were included, with 26, 9, and 12 categorized into fit, intermediate, and frail groups. Only 11 (23.4%) grade 3 or above toxicities were observed overall. Within a median follow-up of 69.0 months, the 3-year overall survival (OS), progression-free survival (PFS), and cancer-specific survival (CSS) rates were 44.3% (95% CI: 32.1%–61.2%), 25.5% (95% CI: 15.7%–41.6%) and 61.0% (95% CI: 47.8%–77.6%) in total. The 5-year OS, PFS, and CSS reached 15.0% (95% CI: 7.4%–30.3%), 14.6% (95% CI: 7.3%–29.4%), and 36.2% (95% CI: 22.0%–59.4%), with no significant difference among the three subgroups. SCRT ( p

We report a general flash arc synthesis (FAS) method to batch fabricate alloy micro/nanocrystals of up to eight components on a wafer-scale. Combined with experiments and first-principle calculations, we show that the shape and size distribution of prepared micro/nanocrystals is determined by both the adhesion energy and diffusion coefficient of metal atoms on the highly oriented pyrolytic graphite (HOPG) substrate. Moreover, we show that surface atomic steps in the HOPG substrate can induce self-assembly and anchoring of metal micro/nanocrystals. The proposed method can essentially prepare alloy micro/nanocrystals with any number of components due to the high cooling rate of micro/nanocrystals, which could lead to the quick development of new materials by directly alloying on the nanoscale and facilitate the applications of alloy micro/nanocrystals.

This investigation sought to understand how physical activity, sleep quality, and depression interrelate among university students in a particular region of China. It involved a diverse group of 2363 students from three institutions, using a cross-sectional approach with well-established measurement tools. These tools demonstrated strong reliability and validity, with Cronbach's alpha values indicating robust internal consistency. The data revealed that higher levels of physical activity were inversely related to depression and directly related to better sleep quality, highlighting the potential of exercise and sleep improvement to reduce depressive symptoms among this demographic.

ObjectiveThe goal of this research was to reveal the association between living alone and depressive symptoms in older adults. It also aims to explore the mediating role of social activity and adult children’s relationship satisfaction. Ultimately, the study seeks to add to the body of knowledge for lowering the risk of depression among older people and promoting positive aging.MethodsBased on information from the 2020 China Health and Retirement Longitudinal Study, the ordinary least square (OLS) regression model and propensity score matching (PSM) were used to investigate the association between living alone and depressive symptoms in older adults and to explore possible heterogeneity in different groups. Utilizing the SPSS PROCESS macro application, the mediation model was constructed. Abstract Background Maedi-visna virus (MVV) is a lentivirus that infects monocyte/macrophage lineage cells in sheep, goats, and wild ruminants and causes pneumonia, mastitis, arthritis, and encephalitis. The immune response to MVV infection is complex, and a complete understanding of its infection and pathogenesis is lacking. This study investigated the in vivo transcriptomic patterns of lung tissues in sheep exposed to MVV using the RNA sequencing technology. Result The results indicated that 2,739 genes were significantly differentially expressed, with 1,643 downregulated genes and 1,096 upregulated genes. Many variables that could be unique to MVV infections were discovered. Gene Ontology analysis revealed that a significant proportion of genes was enriched in terms directly related to the immune system and biological responses to viral infections. Kyoto Encyclopedia of Genes and Genomes analysis revealed that the most enriched pathways were related to virus-host cell interactions and inflammatory responses. Numerous immune-related genes, including those encoding several cytokines and interferon regulatory factors, were identified in the protein-protein interaction network of differentially expressed genes (DEGs). The expression of DEGs was evaluated using real-time polymerase chain reaction and western blot analysis. CXCL13, CXCL6, CXCL11, CCR1, CXCL8, CXCL9, CXCL10, TNFSF8, TNFRSF8, IL7R, IFN-γ, CCL2, and MMP9 were upregulated. Immunohistochemical analysis was performed to identify the types of immune cells that infiltrated MVV-infected tissues. B cells, CD4+ and CD8+ T cells, and macrophages were the most prevalent immune cells correlated with MVV infection in the lungs. Conclusion Overall, the findings of this study provide a comprehensive understanding of the in vivo host response to MVV infection and offer new perspectives on the gene regulatory networks that underlie pathogenesis in natural hosts.

Abstract Background Aldehyde dehydrogenases (ALDHs) are a family of enzymes that catalyze the oxidation of aldehyde molecules into the corresponding carboxylic acid, regulate the balance of aldehydes and protect plants from the poisoning caused by excessive accumulation of aldehydes; however, this gene family has rarely been studied in cotton. Results In the present study, genome-wide identification was performed, and a total of 114 ALDH family members were found in three cotton species, Gossypium hirsutum, Gossypium arboreum and Gossypium raimondii. The ALDH genes were divided into six subgroups by evolutionary analysis. ALDH genes in the same subgroup showed similar gene structures and conserved motifs, but some genes showed significant differences, which may result in functional differences. Chromosomal location analysis and selective pressure analysis revealed that the ALDH gene family had experienced many fragment duplication events. Cis-acting element analysis revealed that this gene family may be involved in the response to various biotic and abiotic stresses. The RT‒qPCR results showed that the expression levels of some members of this gene family were significantly increased under salt stress conditions. Gohir.A11G040800 and Gohir.D06G046200 were subjected to virus-induced gene silencing (VIGS) experiments, and the sensitivity of the silenced plants to salt stress was significantly greater than that of the negative control plants, suggesting that Gohir.A11G040800 and Gohir.D06G046200 may be involved in the response of cotton to salt stress. Conclusions In total, 114 ALDH genes were identified in three Gossypium species by a series of bioinformatics analysis. Gene silencing of the ALDH genes of G. hirsutum revealed that ALDH plays an important role in the response of cotton to salt stress.

Abstract Background The purpose of this study was to evaluate the safety and efficacy of preoperative concurrent chemoradiotherapy (preCRT) for locally advanced rectal cancer in older people who were classified as “fit” by comprehensive geriatric assessment (CGA). Methods A single-arm, multicenter, phase II trial was designed. Patients were eligible for this study if they were aged 70 years or above and met the standards of “fit” (SIOG1) as evaluated by CGA and of the locally advanced risk category. The primary endpoint was 2-year disease-free survival (DFS). Patients were scheduled to receive preCRT (50 Gy) with raltitrexed (3 mg/m2 on days 1 and 22). Results One hundred and nine patients were evaluated by CGA, of whom eighty-six, eleven and twelve were classified into the fit, intermediate and frail category. Sixty-eight fit patients with a median age of 74 years were enrolled. Sixty-four patients (94.1%) finished radiotherapy without dose reduction. Fifty-four (79.3%) patients finished the prescribed raltitrexed therapy as planned. Serious toxicity (grade 3 or above) was observed in twenty-four patients (35.3%), and fourteen patients (20.6%) experienced non-hematological side effects. Within a median follow-up time of 36.0 months (range: 5.9-63.1 months), the 2-year overall survival (OS), cancer-specific survival (CSS) and disease-free survival (DFS) rates were 89.6% (95% CI: 82.3-96.9), 92.4% (95% CI: 85.9-98.9) and 75.6% (95% CI: 65.2-86.0), respectively. Forty-eight patients (70.6%) underwent surgery (R0 resection 95.8%, R1 resection 4.2%), the corresponding R0 resection rate among the patients with positive mesorectal fascia status was 76.6% (36/47). Conclusion This phase II trial suggests that preCRT is efficient with tolerable toxicities in older rectal cancer patients who were evaluated as fit based on CGA. Abstract Background Leukemias driven by activated, chimeric FGFR1 kinases typically progress to AML which have poor prognosis. Mouse models of this syndrome allow detailed analysis of cellular and molecular changes occurring during leukemogenesis. We have used these models to determine the effects of leukemia development on the immune cell composition in the leukemia microenvironment during leukemia development and progression. Methods Single cell RNA sequencing (scRNA-Seq) was used to characterize leukemia associated neutrophils and define gene expression changes in these cells during leukemia progression. Results scRNA-Seq revealed six distinct subgroups of neutrophils based on their specific differential gene expression. In response to leukemia development, there is a dramatic increase in only two of the neutrophil subgroups. These two subgroups show specific gene expression signatures consistent with neutrophil precursors which give rise to immature polymorphonuclear myeloid-derived suppressor cells (PMN-MDSCs). Analysis of gene expression in these precursor cells identified pathways that were specifically upregulated, the most pronounced of which involved matrix metalloproteinases Mmp8 and Mmp9, during leukemia progression. Pharmacological inhibition of MMPs using Ilomastat preferentially restricted in vitro migration of neutrophils from leukemic mice and led to a significantly improved survival in vivo, accompanied by impaired PMN-MDSC recruitment. As a result, levels of T-cells were proportionally increased. In clinically annotated TCGA databases, MMP8 was shown to act as an independent indicator for poor prognosis and correlated with higher neutrophil infiltration and poor pan-cancer prognosis. Conclusion We have defined specific leukemia responsive neutrophil subgroups based on their unique gene expression profile, which appear to be the precursors of neutrophils specifically associated with leukemia progression. An important event during development of these neutrophils is upregulation MMP genes which facilitated mobilization of these precursors from the BM in response to cancer progression, suggesting a possible therapeutic approach to suppress the development of immune tolerance.

Abstract To investigate the safety and efficacy of the neoadjuvant chemoradiotherapy (NCRT) followed by neoadjuvant consolidation chemotherapy (NCCT) and surgery for locally advanced gastric cancer (GC) or gastroesophageal junction (GEJ) adenocarcinoma. Patients diagnosed as locally advanced GC or Siewert II/III GEJ adenocarcinoma with clinical stage T3-4 and/or N positive were prospectively enrolled. Patients underwent NCRT (45 Gy/25 fractions) with concurrent S-1, followed by NCCT (4 to 6 cycles of the SOX regimen) 2 to 4 weeks after NCRT. Gastric cancer radical resection with D2 lymph node dissection was performed 4 to 6 weeks after the total neoadjuvant therapy. The study was conducted from November 2019 to January 2023, enrolling a total of 46 patients. During the NCRT, all patients completed the treatment without dose reduction or delay. During the NCCT, 32 patients (69.6%) completed at least 4 cycles of chemotherapy. Grade 3 or higher adverse events in NCRT (5 cases) were non-hematological. During the course of NCCT, a notable occurrence of hematological toxicities was observed, with grade 3 or higher leukopenia (9.7%) and thrombocytopenia (12.2%) being experienced. A total of 28 patients (60.9%) underwent surgery, achieving R0 resection in all cases. A significant proportion of cases (71.4%) exhibited pathological downstaging to ypT0-2, while 10 patients (35.7%) demonstrated a pathologic complete response (pCR). The total neoadjuvant therapy comprising NCRT followed by NCCT and surgery demonstrates a low severe adverse reactions and promising efficacy, which could be considered as a viable treatment for locally advanced GC or GEJ adenocarcinoma. Abstract Background Magnetic resonance (MR)‐guided ultra‐hypofractionated radiotherapy with whole‐pelvic irradiation (UHF‐WPRT) is a novel approach to radiotherapy for patients with high‐risk (HR) and very high‐risk (VHR) prostate cancer (PCa). However, the inherent complexity of adaptive UHF‐WPRT might inevitably result in longer on‐couch time. We aimed to estimate the delivered dose, study the feasibility and safety of adaptive UHF‐WPRT on a 1.5‐Tesla MR‐Linac. Methods Ten patients with clinical stage T3a‐4N0‐1M0‐1c PCa, who consecutively received UHF‐WPRT, were enrolled prospectively. The contours of the target and organ‐at‐risks on the position verification‐MR (PV‐MR), beam‐on 3D‐MR(Bn‐MR), and post‐MR (after radiotherapy delivery) were derived from the pre‐MR data by deformable image registration. The physician then manually adjusted them, and dose recalculation was performed accordingly. GraphPad Prism 9 (GraphPad Prism Software Inc.) was utilized for conducting statistical analyses. Results In total, we collected 188 MR scans (50 pre‐MR, 50 PV‐MR, 44 Bn‐MR, and 44 post‐MR scans). With median 59 min, the mean prostate clinical target volume (CTV)‐V100% was 98.59% ± 2.74%, and the mean pelvic CTVp‐V100% relative percentages of all scans was 99.60% ± 1.18%. The median V29 Gy change in the rectal wall was −2% (−18% to 20%). With a median follow‐up of 9 months, no patient had acute Common Terminology Criteria for Adverse Events (CTCAE) grade 2 or more severe genitourinary (GU) or gastrointestinal (GI) toxicities (0%). Conclusion UHF‐RT to the prostate and the whole pelvis with concomitant boost to positive nodes using an Adapt‐To‐Shape (ATS) workflow was technically feasible for patients with HR and VHR PCa, presenting only mild GU and GI toxicities. The estimated target dose during the beam‐on phase was clinically acceptable based on the 3D‐MR–based dosimetry analysis. AbstractPlatelet-rich plasma (PRP) holds promise as a therapeutic modality for wound healing; however, immediate utilization encounters challenges related to volume, concentration, and consistency. Cryopreservation emerges as a viable solution, preserving PRP’s bioactive components and extending its shelf life. This study explores the practicality and efficacy of cryopreserved platelet-rich plasma (cPRP) in wound healing, scrutinizing both cellular mechanisms and clinical implications. Fresh PRP and cPRP post freeze-thaw underwent assessment in macrophage, fibroblast, and endothelial cell cultures. The impact of cPRP on active component release and cell behavior pertinent to wound healing was evaluated. Varied concentrations of cPRP (1%, 5%, 10%) were examined for their influence on cell polarization, migration, and proliferation. The results showed minimal changes in cPRP’s IL-1β levels, a slight decrease in PDGF-BB, and superior effects on macrophage M2 polarization and fibroblast migration, while no statistical significance was observed in endothelial cell angiogenesis and proliferation. Remarkably, 5% PRP exhibited the most significant stimulation among all cPRP concentrations, notably impacting cell proliferation, angiogenesis, and migration. The discussion underscores that cPRP maintains platelet phenotype and function over extended periods, with 5% cPRP offering the most favorable outcomes, providing a pragmatic approach for cold storage to extend post-thaw viability and amplify therapeutic effects.

ABSTRACTS-amlodipine, a commonly prescribed antihypertensive agent, is widely used in clinical settings to treat hypertension. However, the potential adverse effects of long-term S-amlodipine treatment on the liver remain uncertain, given the cautionary recommendations from clinicians regarding its administration in individuals with impaired liver function. To address this, we conducted a study using an eight-week-old male rat model and administered a daily dose of 0.6 ~ 5 mg/kg of S-amlodipine for 7 weeks. Our findings demonstrated that 1.2 ~ 5 mg/kg of S-amlodipine treatment induced liver inflammation and associated dysfunction in rats, further in vitro experiments revealed that the observed liver inflammation and dysfunction were not attributable to direct effects of S-amlodipine on the liver. Metagenome sequencing analysis revealed that S-amlodipine treatment led to alterations in the gut microbiome of rats, with the bloom of E. coli (4.5 ~ 6.6-fold increase) and a decrease in A. muciniphila (1,613.4 ~ 2,000-fold decrease) and B. uniformis (20.6 ~ 202.7-fold decrease), subsequently causing an increase in the gut bacterial lipopolysaccharide (LPS) content (1.4 ~ 1.5-fold increase in feces). S-amlodipine treatment also induced damage to the intestinal barrier and increased intestinal permeability, as confirmed by elevated levels of fecal albumin; furthermore, the flux of gut bacterial LPS into the bloodstream through the portal vein resulted in an increase in serum LPS content (3.3 ~ 4-fold increase). LPS induces liver inflammation and subsequent dysfunction in rats by activating the TLR4 pathway. This study is the first to show that S-amlodipine induces liver inflammation and dysfunction by perturbing the rat gut microbiome. These results indicate the adverse effects of S-amlodipine on the liver and provide a rich understanding of the safety of long-term S-amlodipine administration.

In recent years, new energy vehicles (NEVs) have taken the world by storm. A large number of NEV batteries have been scrapped, and research on NEV battery recycling is important for promoting the sustainable development of NEVs. Battery recycling is an important aspect of the sustainable development of NEVs. In this study, we conducted an in-depth analysis of the current status of research on NEV battery recycling from a new perspective using bibliometric methods and visualization software. This study shows that research targeting the recycling of NEV batteries is growing rapidly, and collaborative networks exist among researchers from different countries, institutions, and fields. The focus of research has shifted from lead-acid batteries to lithium batteries, and the supply chain and circular economy related to NEV battery recycling is an emerging research hotspot. Based on our analysis, we propose that the government should establish policies to improve the recycling networks at the collection stage and provide subsidies to attract consumers. Enterprises should develop low-cobalt and cobalt-free technologies, utilize green solvents, and develop new battery swap modes. The establishment of an information platform is conducive to the further development of collaborative networks.

Objectives To evaluate asthma characteristics and treatment patterns, including short-acting β2-agonist (SABA) prescriptions, in primary and specialist care in the Singapore cohort of the SABA use IN Asthma (SABINA III) study.Design Cross-sectional, observational study.Setting Multicentre study conducted at five sites across Singapore.Methods In patients with asthma (aged ≥12 years), data on demographics, disease characteristics and asthma treatment prescriptions were collected using electronic case report forms. Patients were classified by investigator-defined asthma severity (guided by 2017 Global Initiative for Asthma recommendations) and practice type (primary/specialist care).Results Of the 205 patients analysed (mean (SD) age, 53.6 (16.8) years; female, 62%), 55.9% were enrolled by specialists and 44.1% by primary care physicians. Most study patients (80.5%) had moderate-to-severe asthma (86.0% in specialist care and 74.4% in primary care). In the 12 months before study enrolment, 18.0% of patients experienced ≥1 severe exacerbation. Asthma was well or partly controlled in 78.0% of patients. Overall, 17.1% of all patients were overprescribed SABA (≥3 SABA canisters/year) in the preceding 12 months, and overprescription was greater in specialist versus primary care (26.3% vs 5.6%). Only 2.9% of patients were prescribed SABA monotherapy, while 41.0% received SABA in addition to maintenance therapy. Among the latter, 40.5% were overprescribed SABA. Overall, a higher percentage of patients prescribed ≥3 SABA canisters (vs 0–2 SABA canisters) were assessed as having uncontrolled asthma during the study visit (42.9% vs 17.6%). Maintenance therapy in the form of inhaled corticosteroids (ICS) or ICS/long-acting β2 agonist fixed-dose combinations were prescribed to 14.1% and 84.9% of patients, respectively, in the 12 months before enrolment.Conclusions In this Singapore cohort, ~17% of all patients and more than 40% of patients prescribed SABA in addition to maintenance therapy were overprescribed SABA. These findings emphasise the need to align clinical practices with the latest evidence-based treatment recommendations.Trial registration NCT03857178.

Flexible psammophytes play an important role in controlling soil wind erosion and desertification, owing to their characteristics. Although flexibility of psammophytes has been considered in previous studies, the interaction between flexible psammophytes and the surrounding airflow field still remained unclear. In this study, we used the Young’s modulus to describe plant flexibility and conducted a 3D computational fluid dynamics simulation using a standard k-ε model and a fluid–structure interaction model. Taking Caragana korshinskii (Caragana), a typical psammophyte, as the research object, we constructed 3D geometric models with different diameters to simulate the airflow field around the flexible psammophytes. By comparing with the simulation results of rigid plants and simulation results of flexible plants at different wind speeds, we could verify the rationality of the simulation method. Based on the simulation results, the maximum swing amplitude of the model and the Young’s modulus were found to have a negative correlation, presenting an exponential functional relationship with good fitting. The relationship between the actual Young’s modulus of the plant branches and that of different diameter models in the numerical simulation was also established. This study is expected to improve our basic understanding of the interaction between flexible psammophytes and the surrounding airflow field, and provide some qualitative reference for the numerical simulation of the airflow field around flexible psammophytes.

The photoelectric conversion efficiency of photovoltaic thermal (PVT) systems is a key concern in solar energy research. The widespread use of continuous nanofluid cooling in PVT/NPCM (Nano-Enhanced Phase Change Material) systems leads to heightened energy consumption. In order to improve efficiency of the PVT/NPCM systems, a two-dimensional transient heat transfer numerical model is established to analyze the PVT/NPCM system with intermittent flow cooling. Corresponding govern equations with boundary conditions are proposed, and numerically solved by using the finite element method. Simulation results are compared with experimental data, showing a deviation of less than 7 %. The findings indicate that the intermittent cooling reduces the flow energy consumption required to drive 2220 L of nanofluid compared to continuous cooling over a 7-h period. Furthermore, under intermittent cooling conditions, the average electrical efficiency stands at approximately 19.7 %. Notably, the electrical efficiency of PVT/NPCM systems employing intermittent flow cooling closely aligns with those employing continuous flow cooling, exhibiting a maximum deviation of merely 0.0348 %. Additionally, intermittent flow cooling emerges as a favorable choice under solar radiation intensities surpasses 1000W/m2, enabling a significant reduction in overall energy consumption while maintaining commendable cooling performance.

Video synthetic aperture radar (video SAR) has been successfully applied in many fields and the registration of the video SAR images has been proven to be a crucial step in their preprocessing. However, video SAR images exhibit more severe image differences because of the unique imaging mechanism and the immature imaging methods. This results in existing registration methods failing to achieve satisfactory registration outcomes for video SAR images. The convolutional neural network (CNN) can contribute to improving registration performance. Nevertheless, CNN-based registration methods must be driven by a large amount of labeled data, which is impractical for video SAR images. Therefore, to tackle these problems, we propose an unsupervised end-to-end deep registration method for video SAR images. First, an end-to-end deep registration model (DRM) is proposed to improve the registration performance for video SAR images. In the proposed DRM, the offset field is utilized to indirectly calculate the registered parameters and we construct a CNN, MUnet, to regress the offset field accurately. We also develop a differentiable H-transform and a differentiable spatial transformation to implement the mapping from end to end while allowing DRM to backpropagate the losses during the training phase. Meanwhile, we borrow intensity-based methods to further optimize the registration results. Furthermore, we propose an unsupervised deep training strategy that can use the generated pseudodata with pseudolabel to train the proposed DRM in the absence of large amounts of labeled data. Experiment results on multiple data demonstrate the effectiveness of the proposed registration method.

Image deblurring is one of the fundamental tasks in image processing tasks, which can provide the necessary support for advanced tasks such as image recognition. In this paper, we propose a new deblurring model, named Efftformer model, which is specialized in the blurring elimination of motion blur. The model focuses on the recovery of detail information and edge information to provide more effective image information and better basic support for the realization of advanced tasks. In Efftformer model, firstly, we introduce a frequency domain based ReLU residual stream, which allows network to learn blur kernel level information for better restoration of original image. Secondly, we propose a cross-connection channel attention module (CCAM) to explore an effective fusion approach in multiple scales adaptively, which helps decoders to restore original image well by aggregating the semantic information in different scales. Considering the effectiveness of edge information in image recognition tasks, we enhanced the edge information in recovered image by performing a Sobel filter as well as an auxiliary edge loss function. We conducted experiments on different motion blur datasets and compared them with state-of-the-art algorithms. The experimental results show that Efftformer model proposed in this paper achieves comparable even superior performance to the state-of-the-art algorithms.

Abstract Background Scutellaria baicalensis Georgi has been extensively used as a medicinal herb in China for over 2000 years. They may be intentionally or inadvertently substituted or blended with comparable species in the local market, threatening clinical medication safety. Molecular markers are effective tools to prevent misidentification and eliminate doping and falsification among Scutellaria plants. This study screened four highly variable regions to identify Scutellaria and its adulterants. In addition, a phylogenetic analysis was performed using the complete cp genome combined with published Scutellaria species samples. Moreover, a comparative analysis of the cp genomes was conducted to investigate the cp genome evolution of S. baicalensis. Results The complete cp genome of five species of Scutellaria was sequenced for the first time, and four previously published Scutellaria species were re-sequenced. They all exhibited a conserved quadripartite structure in their cp genomes, including two distinct regions, namely a small and large single copy region, respectively, and two inverted repeats encompassing the majority of ribosomal RNA genes. Furthermore, the nine species exhibited high conservation from aspects of the genome structure, codon usage, repeat sequences, and gene content. Four highly variable regions (matK-rps16, ndhC-trnV-UAC, psbE-petL, and rps16-trnQ-UUG) may function as potential molecular markers for differentiating S. baicalensis from its adulterants. Additionally, the monophyly of Scutellaria was ascertained and could be reclassified into two subgenera, subgenus Anaspis and subgenus Scutellaria, as evidenced by the phylogenetic analyses on sequences of cp genome and shared protein-coding sequences. According to the molecular clock analysis, it has been inferred that the divergence of Scutellaria occurred at approximately 4.0 Mya during the Pliocene Epoch. Conclusion Our study provides an invaluable theoretical basis for further Scutellaria species identification, phylogenetics, and evolution analysis.

Abstract Background Microglia-mediated neuroinflammation in Alzheimer’s disease (AD) is not only a response to pathophysiological events, but also plays a causative role in neurodegeneration. Cytoplasmic cysteinyl-tRNA synthetase (CARS) is considered to be a stimulant for immune responses to diseases; however, it remains unknown whether CARS is involved in the pathogenesis of AD. Methods Postmortem human temporal cortical tissues at different Braak stages and AD patient-derived serum samples were used to investigate the changes of CARS levels in AD by immunocytochemical staining, real-time PCR, western blotting and ELISA. After that, C57BL/6J and APP/PS1 transgenic mice and BV-2 cell line were used to explore the role of CARS protein in memory and neuroinflammation, as well as the underlying mechanisms. Finally, the associations of morphological features among CARS protein, microglia and dense-core plaques were examined by immunocytochemical staining. Results A positive correlation was found between aging and the intensity of CARS immunoreactivity in the temporal cortex. Both protein and mRNA levels of CARS were increased in the temporal cortex of AD patients. Immunocytochemical staining revealed increased CARS immunoreactivity in neurons of the temporal cortex in AD patients. Moreover, overexpression of CARS in hippocampal neurons induced and aggravated cognitive dysfunction in C57BL/6J and APP/PS1 mice, respectively, accompanied by activation of microglia and the TLR2/MyD88 signaling pathway as well as upregulation of proinflammatory cytokines. In vitro experiments showed that CARS treatment facilitated the production of proinflammatory cytokines and the activation of the TLR2/MyD88 signaling pathway of BV-2 cells. The accumulation of CARS protein occurred within dense-core Aβ plaques accompanied by recruitment of ameboid microglia. Significant upregulation of TLR2/MyD88 proteins was also observed in the temporal cortex of AD. Conclusions The findings suggest that the neuronal CARS drives neuroinflammation and induces memory deficits, which might be involved in the pathogenesis of AD.

The prediction of equilibrium components for chemical reactions is a considerable section in the metallurgical industry. According to the ion and molecule coexistence theory (IMCT), a modified mass action concentration model based on a thermodynamic database is proposed in this paper, which complys with the law of mass conservation and can be applied in the batching process for Al-Ti-Ca-oxide system that originates from SHS (Self-propagating High-temperature Synthesis) metallurgy. The trend for slag and alloy component under different batching conditions are in good agreement with experiment, while the difference between the theoretical calculation and experiment can be attributed to the deviation from the thermodynamic equilibrium. The modified mass action concentration model with melts and slag can be used to predict the composition and content of the system when equilibrium is achieved at a certain temperature under a specific material ratio, which is conducive to reducing the cost of the experiment and predicting the operability of the actual process. Moreover, it is believed that this thermodynamic insight may has certain application prospects in these metallurgical procedure based on the equilibrium process.

Abstract Background In Singapore, there is currently scarce population-based research informing the recent trends of asthma-related healthcare burdens. In this study, we investigated the past 25-year trends of asthma-related hospitalisations, emergency department (ED) visits and deaths in Singapore and projected the future burdens from 2023 to 2040. Methods We acquired annually-measured data from the Singapore Ministry of Health Clinical and National Disease Registry, containing 25-year asthma-related hospitalisation and death rates as well as 15-year ED visit rates. We conducted change-point analysis and generalised linear modelling to identify time intervals with stable trends and estimate asthma-related healthcare utilisation and mortality rates. To project future asthma-related burdens, we developed a probabilistic model which combined projections of future population size with the estimated rate outcomes from the last stable period. Results Our results show that the asthma hospitalisation rate in Singapore had remained at approximately 80 episodes per 100,000 from 2003 to 2019 and are likely to grow by 1.7% each year (95% CI: 0.7, 5.0%), leading to a total of 163,633 episodes from 2023 to 2040 which corresponds to an estimated $103,075,820 based on 2022 USD. Besides, Singapore’s asthma-related ED visit rate was 390 per 100,000 in 2019 and is expected to decline by 3.4% each year (95% CI: − 5.8, 0.0%), leading to a total of 208,145 episodes from 2023 to 2040 which corresponds to USD$15,053,795. In contrast, the 2019 asthma-related mortality rate in Singapore was approximately 0.57 per 100,000 and is likely to stay stably low (change per year: -1.3, 95% CI: − 11.0, 4.3%). Between 2023 and 2040, Singapore’s estimated total number of asthma-related deaths is 638 episodes. Conclusions Currently, the burden of asthma acute care in Singapore is high; Singapore’s asthma-related hospitalisation and ED visit rates are relatively higher than those of other developed economies, and its asthma admission rate is expected to increase significantly over time, possibly indicating excess resource use for asthma. The established national asthma programme in Singapore, together with recent efforts in reinforcing primary care at the national level, provides opportunities to reduce avoidable asthma admissions.

Sugarcane smut, a serious disease caused by the fungus Sporosorium scitamineum, can result in 30% to 100% cane loss. The most affordable and efficient measure of preventing and handling sugarcane smut disease is to select disease-resistant varieties. A comprehensive evaluation of disease resistance based on the incidence of smut disease is essential during the selection process, necessitating the rapid and accurate identification of sugarcane smut. Traditional identification methods, which rely on visual observation of symptoms, are time-consuming, costly, and inefficient. To address these limitations, we present the lightweight sugarcane smut detection model (YOLOv5s-ECCW), which incorporates several innovative features. Specifically, the EfficientNetV2 is incorporated into the YOLOv5 network to achieve model compression while maintaining high detection accuracy. The convolutional block attention mechanism (CBAM) is added to the backbone network to improve its feature extraction capability and suppress irrelevant information. The C3STR module is used to replace the C3 module, enhancing the ability to capture global large targets. The WIoU loss function is used in place of the CIoU one to improve the bounding box regression’s accuracy. The experimental results demonstrate that the YOLOv5s-ECCW model achieves a mean average precision (mAP) of 97.8% with only 4.9 G FLOPs and 3.25 M parameters. Compared with the original YOLOv5, our improvements include a 0.2% increase in mAP, a 54% reduction in parameters, and a 70.3% decrease in computational requirements. The proposed model outperforms YOLOv4, SSD, YOLOv5, and YOLOv8 in terms of accuracy, efficiency, and model size. The YOLOv5s-ECCW model meets the urgent need for the accurate real-time identification of sugarcane smut, supporting better disease management and selection of resistant varieties.

The meat industry is closely related to people’s daily lives and health, and with the growing global population and increasing demand for meat, the development of efficient pig farming technology is particularly important. However, China’s pig industry still faces multiple challenges, such as high labor costs, high biosecurity risks, and low production efficiency. Therefore, there is an urgent need to develop a fast, accurate, and non-invasive method to estimate pig body data to increase production efficiency, enhance biosecurity measures, and improve pig health. This study proposes EfficientVit-C model for image segmentation and cascade several models to estimate the weight of pigs. The EfficientVit-C network uses a cascading group attention module and improves computational efficiency through parameter redistribution and structured pruning. This method uses only one camera for weight estimation, reducing equipment costs and maintenance expenses. The results show that the improved EfficientVit-C model can segment pigs accurately and efficiently the mAP50 curve convergence is 98.2%, the recall is 92.6%, and the precision is 96.5%. The accuracy of pig weight estimation is 100 kg +/− 3.11 kg. On the Jetson Orin NX platform, the average time to complete image segmentation for each 640*480 resolution image was 4.1 ms, and the average time required to complete pig weight estimation was 31 ms. The results show that this method can quickly and accurately estimate the weight of pigs and provide guidance for the subsequent weight evaluation procedures of pigs.

BackgroundCoronary heart disease (CHD) is the leading cause of death in both developed and many developing countries. Exercise training is a fundamental component of cardiac rehabilitation programs for patients with CHD. This study aims to investigate the effects of a Tai Chi rehabilitation program, which is provided through a hybrid online and offline mode, on oxidative stress and inflammatory responses in patients with CHD.MethodsA total of 34 patients with coronary heart disease were randomly assigned to two groups: an experiment group (n = 14, age 62.07 ± 9.076 years) and a control group (n = 20, age 61.90 ± 9.700 years). The experiment group underwent a 12-week Tai Chi cardiac rehabilitation program (TCCRP), while the control group followed a conventional exercise rehabilitation program (CERP) consisting of 1-h sessions, 3 times per week, for a total of 36 sessions. Participants were studied at baseline and post-intervention. The main assessments include the levels of Malondialdehyde (MDA), Superoxide dismutase (SOD), Tumor necrosis factor (TNF-α) and Interleukin-10 (IL − 10) in blood samples. Multidrug-resistant bacterial infections are a major global health challenge, especially the emergence and rapid spread of methicillin-resistant Staphylococcus aureus (MRSA) urgently require alternative treatment options. Our study has identified that a magnolol derivative 6i as a promising agent with significant antibacterial activity against S. aureus and clinical MRSA isolates (MIC = 2–8 μg/mL), showing high membrane selectivity. Unlike traditional antibiotics, 6i demonstrated rapid bactericidal efficiency and a lower propensity for inducing bacterial resistance. Compound 6i also could inhibit biofilm formation and eradicate bacteria within biofilms. Mechanistic studies further revealed that 6i could target bacterial cell membranes, disrupting the integrity of the cell membrane and leading to increased DNA leakage, resulting in potent antibacterial effects. Meanwhile, 6i also showed good plasma stability and excellent biosafety. Notably, 6i displayed good in vivo antibacterial activity in a mouse skin abscess model of MRSA-16 infection, which was comparable to the positive control vancomycin. These findings indicated that the magnolol derivative 6i possessed the potential to be a novel anti-MRSA infection agent.

BackgroundMembers of the ACR gene family are commonly involved in various physiological processes, including amino acid metabolism and stress responses. In recent decades, significant progress has been made in the study of ACR genes in plants. However, little is known about their characteristics and function in maize.MethodsIn this study, ACR genes were identified from the maize genome, and their molecular characteristics, gene structure, gene evolution, gene collinearity analysis, cis-acting elements were analyzed. qRT-PCR technology was used to verify the expression patterns of the ZmACR gene family in different tissues under salt stress. In addition, Ectopic expression technique of ZmACR5 in Arabidopsis thaliana was utilized to identify its role in response to salt stress.ResultsA total of 28 ZmACR genes were identified, and their molecular characteristics were extensively described. Two gene pairs arising from segmented replication events were detected in maize, and 18 collinear gene pairs were detected between maize and 3 other species. Through phylogenetic analysis, three subgroups were revealed, demonstrating distinct divergence between monocotyledonous and dicotyledonous plants. Analysis of ZmACR cis-acting elements revealed the optional involvement of ZmACR genes in light response, hormone response and stress resistance. Expression analysis of 8 ZmACR genes under salt treatment clearly revealed their role in the response to salt stress. Ectopic overexpression of ZmACR5 in Arabidopsis notably reduced salt tolerance compared to that of the wild type under salt treatment, suggesting that ZmACR5 has a negative role in the response to salt stress.ConclusionTaken together, these findings confirmed the involvement of ZmACR genes in regulating salt stress and contributed significantly to our understanding of the molecular function of ACR genes in maize, facilitating further research in this field.

Abstract Imaging the crustal structure and state of the Himalayan Orogenic Belt (HOB) is of great significance for understanding crustal deformation and its tectonic response in the collision front of the Indian‐Eurasian plates. To avoid the influence from the anomalies underneath the north‐south trending rifts, a three‐dimensional electrical resistivity model across the central HOB was obtained between the Dingjie‐Shenzha and Yadong‐Gulu rifts. No lower‐crustal conductor was found in the southern Lhasa Terrane, and the lower crust with a moderate resistivity value of approximately 100 Ωm is interpreted to be the residue of previous partial melting beneath the Gangdese porphyry copper deposits. The middle crust between the Main Himalayan Thrust and Southern Tibet Detachment System is electrically resistive to the north of the north Himalayan gneiss domes (NHGD) belt and conductive to the south. The middle crust in the northern Himalayas may have undergone partial melting and crystallization during southward migration and duplexing. There is a strong deep‐shallow coupling relationship in the central HOB, where an increase in the underthrusting angle of the Indian crust has resulted in a southward turn of the NHGD belt.

Background: Scarce evidence exists for clinical target volume (CTV) definitions of regional lymph nodes (LNs) in intrahepatic cholangiocarcinoma (iCCA) or combined hepatocellular-cholangiocarcinoma (cHCC-CCA). We investigated the mapping pattern of nodal recurrence after surgery for iCCA and cHCC-CCA and provided evidence for the nodal CTV definition. Methods: We retrospectively reviewed the medical records of patients with iCCA or cHCC-CCA who underwent surgery between 2010 and 2020. Eligibility criteria included patients pathologically diagnosed with iCCA or cHCC-CCA after surgery and a first recurrent event in regional LNs during follow-up. All recurrent LNs were registered onto reference computed tomography images based on the vascular structures to reconstruct the node mapping. Fifty-three patients were eligible. LN regions were classified into four risk groups. Results: Hepatic hilar and portal vein-vena cava were the most common recurrent regions, with recurrence rates of 62.3 % and 39.6 % (high-risk regions), respectively. Recurrence rates in the left gastric, diaphragmatic, common hepatic, superior mesenteric vessels, celiac trunk, and paracardial regions ranged from 15.1 % to 30.2 % (intermediate-risk regions). Abstract Background Knee injuries are prevalent, and early diagnosis is crucial for guiding clinical therapy. MRI is the diagnostic gold standard for bone marrow edema (BME) in patients with acute knee injuries, yet there are still limitations. Dual-energy CT, a possible viable replacement, is being explored (DECT). Methods We systematically retrieved studies from EMBASE, Scopus, PUBMED, and the Cochrane Library and collected gray literatures. In accordance with the PRISMA-DTA standards, a systematic review was conducted between the study's initiation and July 31, 2021, utilizing an MRI reference standard and at least 10 adult patients with acute knee injuries to evaluate the diagnostic effectiveness of DECT for diagnosing BME. Two reviewers collected the study's details independently. For the meta-analysis, a bivariate mixed-effects regression model was utilized, and subgroup analysis was employed to determine the sources of variability. Results The research included nine studies that examined 290 individuals between the ages of 23 and 53 with acute knee injuries who had DECT and MRI. Overall, the sensitivity, specificity, and AUC of the BME were 85% (95% confidence interval [CI]: 77–90%), 96% (95% CI: 93–97%), and 0.97 (95% CI: 0.95–0.98), respectively. To account for the assumed diversity of research, there were no statistically significant differences between the comparison groups in terms of specificity and sensitivity. Conclusion DECT is a viable alternative to MRI for individuals with acute knee injuries when MRI is inappropriate or unavailable.

Abstract Purpose To report the planning benchmark case results of the POTENTIAL trial—a multicenter, randomized, phase 3 trial—to evaluate the value of internal mammary nodal (IMN) irradiation for patients with high-risk breast cancer. Methods All participating institutions were provided the outlines of one benchmark case, and they generated radiation therapy plans per protocol. The plans were evaluated by a quality assurance team, after which the institutions resubmitted their revised plans. The information on beams arrangement, skin flash, inhomogeneity corrections, and protocol compliance was assessed in the first and final submission. Results The plans from 26 institutions were analyzed. Some major deviations were found in the first submission. The protocol compliance rates of dose coverage for the planning target volume of chest wall, supraclavicular fossa plus axilla, and IMN region (PTVim) were all significantly improved in the final submission, which were 96.2% vs. 69.2%, 100% vs. 76.9%, and 88.4% vs. 53.8%, respectively. For OARs, the compliance rates of heart Dmean, left anterior descending coronary artery V40Gy, ipsilateral lung V5Gy, and stomach V5Gy were significantly improved. In the first and final submission, the mean values of PTVim V100% were 79.9% vs. 92.7%; the mean values of heart Dmean were 11.5 Gy vs. 9.7 Gy for hypofractionated radiation therapy and 11.5 Gy vs. 11.0 Gy for conventional fractionated radiation therapy, respectively. Conclusion The major deviations were corrected and protocol compliance was significantly improved after revision, which highlighted the importance of planning benchmark case to guarantee the planning quality for multicenter trials.

Abstract Currently, studies of ancient faunal community networks have been based mostly on uniformitarian and functional morphological evidence. As an important source of data, taphonomic evidence offers the opportunity to provide a broader scope for understanding palaeoecology. However, palaeoecological research methods based on taphonomic evidence are relatively rare, especially for body fossils in lacustrine sediments. Such fossil communities are not only affected by complex transportation and selective destruction in the sedimentation process, they also are strongly affected by time averaging. Historically, it has been believed that it is difficult to study lacustrine entombed fauna by a small-scale quadrat survey. Herein, we developed a software, the TaphonomeAnalyst, to study the associational network of lacustrine entombed fauna, or taphocoenosis. TaphonomeAnalyst allows researchers to easily perform exploratory analyses on common abundance profiles from taphocoenosis data. The dataset for these investigations resulted from fieldwork of the latest Middle Jurassic Jiulongshan Formation near Daohugou Village, in Ningcheng County of Inner Mongolia, China, spotlighting the core assemblage of the Yanliao Fauna. Our data included 27,000 fossil specimens of animals from this deposit, the Yanliao Fauna, whose analyses reveal sedimentary environments, taphonomic conditions, and co-occurrence networks of this highly studied assemblage, providing empirically robust and statistically significant evidence for multiple Yanliao habitats.

Photoacoustics can provide a direct measurement of light absorption by microalgae depending on the photosynthesis pigment within them. In this study, we have performed photoacoustic flowmetry on living microalgae cells to measure their flow characteristics, which include flow speed, flow angle, flow direction, and, more importantly, the photoacoustic absorption spectrum, all by observing the photoacoustic Doppler power spectra during their flowing state. A supercontinuum pulsed laser with a high repetition frequency is used as the light source: through intensity modulation at a specified frequency, it can provide wavelength-selectable excitation of a photoacoustic signal centered around this frequency. Our approach can be useful to simultaneously measure the flow characteristics of microalgae and easily discriminate their different species with high accuracy in both static and dynamic states, thus facilitating the study of their cultivation and their role in our ecosystem.

Moso bamboo (Phyllostachys edulis), renowned for its rapid growth, is attributed to the dynamic changes in its apical meristem. The CLAVATA3/EMBRYO SURROUNDING REGION-RELATED (CLE) family genes are known to play crucial roles in regulating meristem and organ formation in model plants, but their functions in Moso bamboo remain unclear. Here, we conducted a genome-wide identification of the CLE gene family of Moso bamboo and investigated their gene structure, chromosomal localization, evolutionary relationships, and expression patterns. A total of 11 PheCLE genes were identified, all of which contained a conserved CLE peptide core functional motif (Motif 1) at their C-termini. Based on Arabidopsis classification criteria, these genes were predominantly distributed in Groups A–C. Collinearity analysis unveiled significant synteny among CLE genes in Moso bamboo, rice, and maize, implying potential functional conservation during monocot evolution. Transcriptomic analysis showed significant expression of these genes in the apical tissues of Moso bamboo, including root tips, shoot tips, rhizome buds, and flower buds. Particularly, single-cell transcriptomic data and in situ hybridization further corroborated the heightened expression of PheCLE1 and PheCLE10 in the apical tissue of basal roots. Additionally, the overexpression of PheCLE1 and PheCLE10 in rice markedly promoted root growth. PheCLE1 and PheCLE10 were both located on the cell membrane. Furthermore, the upstream transcription factors NAC9 and NAC6 exhibited binding affinity toward the promoters of PheCLE1 and PheCLE10, thereby facilitating their transcriptional activation. In summary, this study not only systematically identified the CLE gene family in Moso bamboo for the first time but also emphasized their central roles in apical tissue development. This provides a valuable theoretical foundation for the further exploration of functional peptides and their signaling regulatory networks in bamboo species.

Biomimetic bone-active coatings composed of inorganic nano-hydroxyapatite (i.e., nHA) and organic silk fibroin (i.e., SF) are layer-by-layer deposited on Mg-Zn-Ca alloy by a controllable ultrasonic spray method. Meanwhile, plasma activation is developed as a promising strategy to pretreat magnesium surfaces, which facilitates the direct adhesion of coatings with enhanced bonding interfaces. In this work, we engineer the nHA/SF composite coatings with excellent mechanical properties and adhesion force. The optimized parameters of ultrasonic spray bring significant influence on the surface morphologies of coatings. Assisted by hybrid plasma of oxygen and nitrogen (i.e., O2/N2 plasma), the activated Mg-Zn-Ca surfaces are uniformly covered by a robust and compact nHA/SF composite coating, establishing a multifunctional system with superior corrosion resistance and biological performance. Interestingly, secondary oxygen plasma treatment of nHA/SF coatings (A-nHA/SF) promotes the hydrophilicity, leading to a rapid self-repair effect from surface damage. The improvement of anti-corrosion and self-repair provides a dependable platform for better cell adhesion, proliferation, spreading and differentiation. These favorable factors contribute to the preferable in vivo biocompatibility and the promotion of newly formed bones for the A-nHA/SF-coated Mg implants. This study lays important foundations for coating strategy on biomedical magnesium alloy as multifunctional osteogenic system in bioactive implantable applications.

Abstract Transient ischemic attack (TIA) was clinically divided into anterior circulation (AC) or posterior circulation (PC). Previous study reported that ABCD2 score could predict the stroke risk after AC-TIA but might have limitation for PC-TIA. We aimed to classify TIA depending on neuroimaging and assess the value of ABCD2 score for predicting stroke risk in different territories. Research data was from TIA database of the First Affiliated Hospital of Zhengzhou University. TIA patients with acute infarction on diffuse weighted imaging [that is, transient symptoms with infarction (TSI)] were divided into anterior and posterior circulation groups according to the location of infarction. The outcome was recurrent stroke within 7 and 90 days. The predictive power of ABCD2 score was determined using area under receiver operator characteristic curve (AUC) analyses. Overall, 382 AC-TSI and 112 PC-TSI patients were included. There were 38 (9.9%) AC-TSI patients and 11(9.8%) PC-TSI patients who had recurrent stroke at 7 days, and 66 (17.3%) AC-TSI patients and 19 (17.0%) PC-TSI patients who had recurrent stroke within 90 days. At 7 days, the AUC for ABCD2 score was 0.637 (95% confidence interval CI 0.554–0.720) in anterior circulation and 0.683 (95% CI 0.522–0.845) in posterior circulation. The C statistics for ABCD2 score in the two groups were not statistically significant (Z = − 0.499; P = 0.62). Similar result was found when the outcome time-point was set at 90 days. ABCD2 score could predict the short-term risk of recurrent stroke after AC-TSI and PC-TSI, and had similar predictive abilities for AC-TSI and PC-TSI.

Abstract Techniques to study brain activities have evolved dramatically, yet tremendous challenges remain in acquiring high-throughput electrophysiological recordings minimally invasively. Here, we develop an integrated neuroelectronic array that is filamentary, high-density and flexible. Specifically, with a design of single-transistor multiplexing and current sensing, the total 256 neuroelectrodes achieve only a 2.3 × 0.3 mm2 area, unprecedentedly on a flexible substrate. A single-transistor multiplexing acquisition circuit further reduces noise from the electrodes, decreases the footprint of each pixel, and potentially increases the device’s lifetime. The filamentary neuroelectronic array also integrates with a rollable contact pad design, allowing the device to be injected through a syringe, enabling potential minimally invasive array delivery. Successful acute auditory experiments in rats validate the ability of the array to record neural signals with high tone decoding accuracy. Together, these results establish soft, high-density neuroelectronic arrays as promising devices for neuroscience research and clinical applications.

Abstract Background Pumpkin seeds are major oil crops with high nutritional value and high oil content. The collection and identification of different pumpkin germplasm resources play a significant role in the realization of precision breeding and variety improvement. In this research, we collected 75 species of pumpkin from the Zhejiang Province of China. 35,927 near-infrared hyperspectral images of 75 types of pumpkin seeds were used as the research object. Results To realize the rapid classification of pumpkin seed varieties, position attention embedded three-dimensional convolutional neural network (PA-3DCNN) was designed based on hyperspectral image technology. The experimental results showed that PA-3DCNN had the best classification effect than other classical machine learning technology. The classification accuracy of 99.14% and 95.20% were severally reached on the training and test sets. We also demonstrated that the PA-3DCNN model performed well in next year’s classification with fine-tuning and met with 94.8% accuracy. Conclusions The model performance improved by introducing double convolution and pooling structure and position attention module. Meanwhile, the generalization performance of the model was verified, which can be adopted for the classification of pumpkin seeds in multiple years. This study provided a new strategy and a feasible technical approach for identifying germplasm resources of pumpkin seeds.

Objectives: To investigate the prognostic capacity of baseline 18F-fluorodeoxyglucose (18F-FDG) positron emission tomography/computed tomography (PET/CT) metabolic parameters in extranodal natural killer/T-cell lymphoma (ENKTCL), and the influence of relative thresholds (RT) and absolute thresholds (AT) selection on prognostic capacity. Materials and methods: Metabolic tumor volume (MTV)-based parameters were defined using RTs (41 % or 25 % of maximum standardized uptake value [SUVmax]), ATs (SUV 2.5, 3.0, 4.0, or mean liver uptake) in 133 patients. Metabolic parameters were classified into avidity-related parameters (SUVmax, mean SUV [SUVmean], standard deviation of SUV [SUVsd]), volume-related parameters (RT-MTV), and avidity- and volume-related parameters (total lesion glycolysis [TLG] and AT-MTV). The prognostic capacity of the metabolic parameters and the effects of different threshold types (RT vs. AT) were evaluated. Results: All metabolic parameters were moderately associated with prognosis. However, the area under the receiver operating characteristic curve of MTV and TLG was slightly higher than that of avidity-related parameters for predicting 5-year progression-free survival (PFS) (0.614–0.705 vs. 0.563–0.609) and overall survival (OS) (0.670–0.748 vs. 0.562–0.593). Abstract This study aimed to predict the 5‐year overall survival (OS) benefit of pola‐R‐CHP versus R‐CHOP in the POLARIX trial based on the 2‐year event‐free survival (EFS) and progression‐free survival (PFS) rates in diffuse large B‐cell lymphoma (DLBCL). We identified randomized controlled trials (RCT) published before 31 May 2023. The correlation between the logarithmic (log) hazard ratio (HR) for EFS (HREFS) or PFS (HRPFS) and the HR for OS (HROS) was estimated at the trial‐level. Correlation analysis was performed between 2‐year PFS or EFS and 5‐year OS rates at the treatment arm‐level. Linear regression models were used to calculate the 5‐year OS of pola‐R‐CHP and R‐CHOP. In the included 20 RCTs, a linear correlation between HREFS (r = 0.765) or HRPFS (r = 0.534) and HROS was observed at the trial‐ level. Two‐year EFS (r = 0.918) or 2‐year PFS (r = 0.865) correlated linearly with 5‐year OS. Linear regression analysis between 2‐year EFS/PFS and 5‐year OS gave estimated 5‐year OS rates between pola‐R‐CHP and R‐CHOP of 6.4% and 6.3%, respectively. Two‐year EFS and PFS are feasible early endpoints in patients with DLBCL treated primarily with immunochemotherapy. The pola‐R‐CHP regimen is expected to improve 5‐year OS.

The building automation system (BAS) of a subway is the core component of monitoring and managing urban rail transit systems. For the current problems such as low control efficiency, insufficient accuracy, and poor stability of metro BAS, this article proposes a cooperative control framework based on an improved fuzzy proportional-integral-derivative (PID) algorithm. Firstly, the concept of an integrated supervisory control system (ISCS) for subways is introduced by summarizing the previously implemented engineering construction and combining it with advanced automation technology. The system’s overall design under the ISCS framework is also improved by integrating it with the fire alarm system (FAS) with the BAS as the core unit of the reliance. Then, an improved seeker optimization algorithm (ISOA) is employed to optimize the parameters of the fuzzy PID control algorithm to achieve a coordinated control of the system based on considering the time lag problem. Finally, the accuracy, efficiency, and stability of the coordinated control response of the BAS under the ISCS framework are tested experimentally. The results suggest that the proposed cooperative control solution of BAS employing the improved fuzzy PID algorithm has good control accuracy and response efficiency and can also ensure the BAS’s higher stability in the coordinated control process, which thus greatly improves the automation level of the subway and provides a safer and more reliable high-performance for the ISCS of the subway in the urban rail transportation industry.

The efficiency of hydrogen production from waste activated sludge (WAS) through combining anaerobic digestion and microbial electrolysis cell (AD-MEC) systems has been increasing. With advancements in materials science, the cost of MEC anode materials has decreased, making this technology potentially economically viable. By analyzing data from 3611 centralized wastewater treatment plants (WWTPs) in 276 prefecture-level cities in China, theoretical application scenarios and environmental benefits of hydrogen production in these WWTPs via AD-MEC were evaluated. Additionally, a theoretical economic model is constructed to validate the economic feasibility of AD-MEC technology. The key findings suggested that the theoretical potential for hydrogen production from WAS in China is 152,610.5 tons/year. The amount of CO2 emitted by natural gas, fuel diesel buses, or power electric public transportation could reduce approximately 1.51, 3.2, and 2.4 million tons/year, respectively, through utilizing hydrogen instead of the above energy sources. Due to the broader temperature adaptability of hydrogen fuel cells, this emission reduction effect was more pronounced in colder, high-latitude regions. Economic analysis revealed that, considering sludge reduction, the AD-MEC system could achieved a return on investment of 13%. This study demonstrated that with the rapid progression in urbanization, AD-MEC systems could contribute to reducing WAS treatment costs and providing clean hydrogen for carbon emission reductions in urban areas.

To investigate the genetic mechanisms underlying the reproductive traits (time to flowering and maturity) in wheat and identify candidate genes associated, a phenotypic analysis was conducted on 239 wheat accessions (lines) from around the world. A genome-wide association study (GWAS) of wheat heading and maturity phases was performed using the MLM (Q+K) model in the TASSLE software, combined with the Wheat 55K SNP array. The results revealed significant phenotypic variation in heading and maturity among the wheat accessions across different years, with coefficients of variation ranging from 0.96% to 1.97%. The phenotypic data from different years exhibited excellent correlation, with a genome-wide linkage disequilibrium (LD) attenuation distance of 3 Mb. Population structure analysis, evolutionary tree analysis, and principal component analysis indicated that the 239 wheat accessions formed a relatively homogeneous natural population, which could be divided into three subgroups. The GWAS results identified a total of 293 SNP marker loci that were significantly associated with wheat heading and maturity stages (P ≤ 0.001) in different environments. Among them, nine stable SNP marker loci were consistently detected in multiple environments. These marker loci were distributed on wheat chromosomes 1A、1B、2D、3A、5B、6D and 7A. Each individual locus explained 4.03%-16.06% of the phenotypic variation. Furthermore, through careful analysis of the associated loci with large phenotypic effect values and stable inheritance, a total of nine candidate genes related to wheat heading and maturity stages were identified. These findings have implications for molecular marker-assisted selection breeding programs targeting specific wheat traits at the heading and maturity stages. In summary, this study conducted a comprehensive GWAS of wheat heading and maturity phases, revealing significant associations between genetic markers and key developmental stages in wheat. The identification of candidate genes and marker loci provides valuable information for further studies on wheat breeding and genetic improvement targeted at enhancing heading and maturity traits.
