Fast growing Kariba weed causes major problems and pollution on freshwater and shellfish aquaculture systems by interfering with nutrient uptake of crops, restricting sunlight penetration, and decreasing water quality due to massive biomass of Kariba weed remnants. Solvothermal liquefaction is considered an emerging thermochemical technique to convert waste into high yield of value-added products. Solvothermal liquefaction (STL) of Kariba weed as an emerging contaminant was performed to investigate the effects of different types of solvents (ethanol and methanol) and Kariba weed mass loadings (2.5–10 % w/v) on treating and reducing the weed via conversion into potentially useful crude oil product and char. Up to 92.53 % of Kariba weed has been reduced via this technique. The optimal conditions for crude oil production were found to be at 5 % w/v of mass loading in methanol medium, resulting in a high heating value (HHV) of 34.66 MJ/kg and yield of 20.86 wt%, whereas the biochar production was found to be optimum at 7.5 % w/v of mass loading in methanol medium, resulting in 29.92 MJ/kg of HHV and 25.38 wt% of yield. The crude oil consisted of beneficial chemical compounds for biofuel production such as hexadecanoic acid, methyl ester (65.02 peak area %) and the biochar showed high carbon content (72.83 %). In conclusion, STL as a remediation for emerging Kariba weed is a feasible process for shellfish aquaculture waste treatment and biofuels production.

Background The purification of keratinase is essential to enhance the catalytic efficiency of keratinase enzyme for industrial applications. Biphasic system incorporated with electric fields is an effective purification tool for the recovery of biomolecules. Ionic liquids (IL) with good conductivity are ideal phase-forming components for biphasic system which promotes the separation of charged biomolecules via electromigration. Methods This study aimed to evaluate the effect of electric fields on enhanced recovery of Kytococcus sedentarius TWHKC01 keratinase using IL-based biphasic system. Findings Keratinase was recovered with biphasic system incorporated with anode in salt-rich bottom phase and cathode in IL-rich top phase operated at 5 V for 5 min. A total of 95.82% ± 0.17 of keratinase was attained with the biphasic system at pH 9 comprising of 20% (w/w) 1‑butyl‑3-methylimidazolium tetrafluoroborate ((Bmim)BF4) and 25% (w/w) ammonium sulphate loaded with 15% (w/w) K. sedentarius TWHKC01 crude feedstock. The selectivity (S) and purification fold (PFT) of keratinase were increased from 7.25 ± 1.50 to 10.87 ± 0.61 and 5.94 ± 0.22 to 8.22 ± 0.44, respectively with the incorporation of electrodes, demonstrating the efficiency of electric fields in enhancing the purity of recovered keratinase.

Melanoma is an extremely aggressive form of skin cancer that can spread to the lungs, brain, and liver, among other vital organs. Melanoma cells, unlike any other cancer cells, can produce significant amounts of melanin by a process known as melanogenesis, causing them to become heavily pigmented. Melanogenesis, specifically the melanin pigment, is well known for its ability to protect the skin from the harmful effects of UV light, which can lead to the development of skin cancer. Nevertheless, uncontrolled melanogenesis plays a role in the advancement of melanotic melanoma, and melanin pigments can reduce the effectiveness of radiotherapy and immunotherapy. Therefore, studies are being performed that focus on inhibiting melanogenesis to prevent melanoma metastasis. However, it is worth noting that, in addition to its UV‑protective function, melanin also plays a role in preventing melanoma metastasis. Microphthalmia‑associated transcription factor (MITF) and melanin have been demonstrated to attenuate the aggressiveness of melanoma by suppressing numerous essential metastatic processes. Eumelanin and pheomelanin (two types of melanin), which cause oxidative stress, can also prevent melanoma progression in the early stages. Hence, it is vital to explore the role of inducing melanogenesis rather than inhibiting melanogenesis in preventing melanoma metastasis.

A green bio-separation alternative can be performed with a non-toxic and biodegradable aqueous biphasic system (ABS) composed of short-chain aliphatic alcohol-based top phase (1-propanol and 2-propanol) and carbohydrate-based bottom phase (glucose, sucrose, and maltose). A model protein, bovine serum albumin (BSA) was adopted to determine the effects of types and concentration of phase-forming components; protein concentration; and system pH on the protein partition efficiency in the ABS. Results showed that the 1-propanol/maltose ABS gave an overall better partition efficiency of BSA to the alcohol-rich top phase compared to the 1-propanol/sucrose ABS, 1-propanol/glucose ABS, and 2-propanol/sugar ABS attributed to the lower hydrophilicity of 1-propanol and the stronger sugaring-out effect exerted by the maltose. A maximum partition coefficient (K) of 20.01 ± 0.05 and recovery yield (Y) of 95.42% ± 0.01 of BSA were obtained with the 35% (w/w) 1-propanol/22% (w/w) maltose ABS at pH 5.0 which contained 10% (w/w) BSA. The K and Y of BSA in 1-propanol/maltose ABS was further enhanced with the addition of 3% (w/w) of ionic liquids, 1-butyl-3-methylimidazolium bromide ([Bmim]Br) as the adjuvants which provides the protein stabilizing effect. The Fourier Transform Infrared Spectrum (FTIR) analysis revealed that the protein structure remained unaltered upon the separation process.

Keratinase is widely applied in the bioconversion of feather waste to produce value-added bioproducts. The wide applications of industrial keratinase have urged the advancement in producing keratinase with high productivity and yields. The efficiency of aqueous two-phase electrophoresis system (ATPES) to recover extracellular Kytococcus sedentarius TWHKC01 keratinase was investigated. The operation time of 10 min and voltage of 10 V were applied for the recovery of K. sedentarius TWHKC01 keratinase with the PEG 1000/sulfate ATPES. Keratinase was recovered in the PEG-rich phase of the ATPES consists of 23% (w/w) PEG 1000, 18.5% (w/w) ammonium sulfate of pH 9, 20% (w/w) crude feedstock and with 3% (w/w) of NaCl added. Highest partition coefficient (Ke) of 4.63 ± 0.17, recovery yield (YT) of 78.75% ± 0.61, and purification fold (PFT) of 3.89 ± 0.84 were obtained in the optimal ATPES coupled with cathode electrode in top phase and anode electrode in the bottom phase. The introduction of electric field to the ATPS has enhanced the selectivity (S) of the system towards the extracellular keratinase by two-fold from 2.79 ± 0.46 to 5.68 ± 0.43, demonstrating the efficiency of ATPES in separating the keratinase from crude feedstock when compared to conventional two-phase system.

The aims of this study were to compare the effectiveness of different drying methods and to investigate the effects of adding a series of individual protectant such as skim milk, sucrose, maltodextrin, and corn starch for preserving Lactobacillus acidophilus FTDC 3081 cells during spray and freeze-drying and storage at different temperatures. Results showed a remarkable high survival rate of 70–80% immediately after spray- and freeze-drying in which the cell viability retained at the range of 109 to 1010 CFU/mL. After a month of storage, maltodextrin showed higher protective ability on both spray- and freeze-dried cells as compared to other protective agents at 4°C, 25°C, and 40°C. A complete loss in viability of spray-dried L. acidophilus FTDC 3081 was observed after a month at 40°C in the absence of protective agent.

Carotenoids serve as one of the most important group of naturally-occurring lipid-soluble pigments which exhibit great biological activities such as antioxidant, anti-inflammatory and provitamin A activities. Owing to their advantageous health effects, carotenoids are widely applied in various industries. Microbial carotenoids synthesis therefore has attracted increasing attention in recent years. In the present study, a marine microorganism originally isolated from seawater in northern Taiwan was determined to be a strain of Gordonia terrae based on its 16S rRNA gene sequence. The strain G. terrae TWRH01 has the ability to synthesize and accumulate the intracellular pigments was identified by gas chromatography–mass spectrometry (GC–MS). The biochemical production characteristics of this strain were studied by employing different fermentation strategies. Findings suggested that G. terrae TWRH01 can actively grow and efficiently synthesize carotenoids in medium adjusted to pH 7 containing 16 g L−1 sucrose as the carbon source, 16 g L−1 yeast extract as the nitrogen source, 0.6 M NaCl concentration, and supplemented with 0.45% (v/v) 1 M CaCl2. Results revealed that the optimization of fermentation yielded 15.29 g L−1 dry biomass and 10.58 μmol L−1 relative β-carotene concentration. According to GC–MS analysis, the orange-red colored pigments produced were identified as carotenoid derivatives, mainly echinenone and adonixanthin 3′-β- d -glucoside. Therefore, the new bacterial strain showed a highly potential bioresource for the commercial production of natural carotenoids.

Ectoine is a zwitterionic amino acid derivative that can be naturally sourced from halophilic microorganisms. The increasing demands of ectoine in various industries have urged the researches on the cost-effective approaches on production of ectoine. Ionic liquids-based aqueous biphasic system (ILABS) was applied to recover Halomonas salina ectoine from cells hydrolysate. The 1-butyl-3-methylimidazolium tetrafluoroborate (Bmim)BF4 was used in the ILABS and the recovery efficiency of ILABS to recover ectoine from H. salina cells lysate was evaluated by determining the effects of phase composition; pHs; crude loading and additional neutral salt (NaCl). The hydrophilic ectoine was targeted to partition to the hydrophilic salt-rich phase. A total yield (YB) of 96.32% ± 1.08 of ectoine was obtained with ILABS of phase composition of 20% (w/w) (Bmim)BF4 and 30% (w/w) sulfate salts; system pH of 5.5 when the 20% (w/w) of crude feedstock was applied to the ILABS. There was no significant enhancement on the ectoine recovery efficiency using the ILABS when NaCl was added, therefore the ILABS composition without the additional neutral salt was recommended for the primary purification of ectoine. Partition coefficient (KE) of 30.80 ± 0.42, purity (PE) of 95.82% and enrichment factor (Ef) of 1.92 were recorded with the optimum (Bmim)BF4/sulfate ILABS. These findings have provided an insight on the feasibility of recovery of intracellular biomolecules using the green solvent-based aqueous system in one single-step operation.

Ectoine production using inexpensive and renewable biomass resources has attracted great interest among the researchers due to the low yields of ectoine in current fermentation approaches that complicate the large-scale production of ectoine. In this study, ectoine was produced from corn steep liquor (CSL) and soybean hydrolysate (SH) in replacement to yeast extract as the nitrogen sources for the fermentation process. To enhance the bacterial growth and ectoine production, biotin was added to the Halomonas salina fermentation media. In addition, the effects addition of surfactants such as Tween 80 and saponin on the ectoine production were also investigated. Results showed that both the CSL and SH can be used as the nitrogen source substitutes in the fermentation media. Higher amount of ectoine (1781.9 mg L-1) was produced in shake flask culture with SH-containing media as compared to CSL-containing media. A total of 2537.0 mg L-1 of ectoine was produced at pH 7 when SH-containing media was applied in the 2 L batch fermentation. Moreover, highest amount of ectoine (1802.0 mg L-1) was recorded in the SH-containing shake flask culture with addition of 0.2 μm mL-1 biotin. This study demonstrated the efficacy of industrial waste as the nutrient supplement for the fermentation of ectoine production.

Ionic liquid-based aqueous biphasic system (ILABS) has emerged as an attractive green approach for the extraction and separation of various biomolecules. The growing market demands of cytochrome c (cyt c) due to its vast uses in medical applications have urged the search for cost-efficient approaches for the production and purification of cyt c. In this study, the feasibility of ILABS to recover cyt c from simulated Saccharomyces cerevisiae cell cultures was investigated by evaluating the effects of phase composition, pH, and additives concentration on the recovery efficiency of cyt c. The ILABS was developed using the hydrophilic ionic liquids, 1-hexyl-3-methylimidazolium bromide ((C6mim)Br) and potassium citrate. The optimal separation conditions for recovery of cyt c from simulated cells cultures were attained with ILABS of pH 9 comprising of 28% (w/w) of (C6mim)Br and 24% (w/w) of potassium citrate and the addition of 0.2% (w/w) NaCl at room temperature (25 °C). Cyt c was recovered in the IL-rich phase with partition coefficient (Kc) of 364.00 ± 1.09, recovery yield (Y) of 99.76% ± 0.03 and selectivity (S) of 223.31 ± 0.56. The results suggest that ILABS can efficiently recover cyt c from microbial fermentation broth with high recovery yields and separation efficiency.

Garcinia mangostana pericarp is a good source of natural antioxidants with numerous functional properties. The conventional approaches for the recovery of antioxidants from Garcinia mangostana pericarp require long processing time and high temperature, which may cause degradation or loss of bioactivity of antioxidants, and often result in low recovery efficiency. In this study, the extraction of antioxidants from Garcinia mangostana pericarp was investigated using a polyethylene glycol (PEG)/citrate aqueous biphasic system (ABS) with the addition of surfactants. The optimum condition for the recovery of antioxidants was achieved in PEG 1000/citrate ABS of pH 8 with tie-line length (TLL) of 48.3% (w/w), volume ratio (VR) of 1.6, 0.2% (w/w) sample loading and addition of 1.0% (w/w) Tween 85. The antioxidants were recovered in the PEG-rich top phase with a high K value of 18.23 ± 0.33 and a recovery yield of 92.01% ± 0.09. The findings suggested that the addition of surfactants to polymer/salt ABS can enhance the recovery of antioxidants from Garcinia mangostana pericarps by conserving the antioxidative properties.

Aqueous biphasic flotation (ABF) integrates aqueous biphasic system (ABS) and solvent sublation for recovery of target biomolecules. The feasibility of the alcohol/salt ABF for exclusive partition of cytochrome c to one specific phase of the system was investigated. Aliphatic alcohols of different carbon chain length (ethanol, 1-propanol and 2-propanol) and salts (sulfate, phosphate and citrate) were used for the phase formation. The effects of phase composition, concentration of sample loading, pH, flotation time and flow rate of the system on the partition efficiency of cytochrome c were determined. Cytochrome c was exclusively partitioned to the alcohol-rich top phase of the ABF of 18% (w/w) ethanol and 26% (w/w) ammonium sulfate with pH 6 and 20% (w/w) of sample loading. Highest partition coefficient (K) of 6.85 ± 0.21 and yield (YT) of 99.40% ± 0.02 were obtained with optimum flotation rate of 10 mL/min and flow rate of 10 min.

This review provides a quantitative description of the nano-adsorbent processing and its viability against wastewater detoxification by extracting heavy metal ions. The impact of nano-adsorbent functionalities on specific essential attributes such as the surface area, segregation, and adsorption capacity were comprehensively evaluated. A detailed analysis has been presented on the characteristics of nanomaterials through their limited resistance to adsorb some heavy metal ions. Experimental variables such as the adsorbent dosage, pH, substrate concentration, response duration, temperature, and electrostatic force that influence the uptake of metal ions have been studied. Besides, separate models for the adsorption kinetics and isothermal adsorption have been investigated to understand the mechanism behind adsorption. Here, we reviewed the different adsorbent materials with nano-based techniques for the removal of heavy metals from wastewater and especially highlighted the nano adsorption technique. The influencing factors such as pH, temperature, dosage time, sorbent dosage, adsorption capacities, ion concentration, and mechanisms related to the removal of heavy metals by nano composites are highlighted. Lastly, the application potentials and challenges of nano adsorption for environmental remediation are discussed. This critical review would benefit engineers, chemists, and environmental scientists involved in the utilization of nanomaterials for wastewater treatment.

Food loss or waste is a far-reaching problem and has indeed become a worrying issue that is growing at an alarming rate. Fruits and vegetables are lost or wasted at the highest rate among the composition of food waste. Furthermore, the world is progressing toward sustainable development; hence, an efficient approach to valorise fruit and vegetable waste (FVW) is necessary. A simple phenotypic characterisation of microbiota isolated from the fermented FVW was conducted, and its effectiveness toward wastewater treatment was investigated. Presumptive identification suggested that yeast is dominant in this study, accounting for 85% of total isolates. At the genus level, the enriched medium's microbial community consists of Saccharomyces, Bacillus and Candida. Ectoine (1,4,5,6-tetrahydro-2-methyl-4-pyrimidinecarboxylic acid) is a revolutionizing substance with vast applications in the cosmetic and food industries. Ectoine is often sourced from halobacteria. The increasing market demand for ectoine has urged the development of cost-effective and sustainable large-scale production of ectoine from microbial sources. This review describes the existing and potential microbial sources of ectoine and its derivatives, as well as microbial production and fermentation approaches for ectoine recovery. In addition, conventional methods and emerging technologies for enhanced production and recovery of ectoine from microbial fermentation with a focus on the aqueous biphasic system (ABS) are discussed. The ABS is a practically feasible approach for the integration of fermentation, cell disruption, bioconversion, and clarification of various biomolecules in a single-step operation. Nonetheless, the implementation of the ABS on an industrial-scale basis for the enhanced production and recovery of ectoine is yet to be exploited. Therefore, the feasibility of the ABS to integrate the production and direct recovery of ectoine from microbial sources is also highlighted in this review.

Global demand for plastic materials has severely harm the environment and marine sea life. Therefore, bioplastics have emerged as an environmentally friendly alternative due to sustainability, minimal carbon footprint, less toxicity and high degradability. This review highlights the sustainable and environmentally friendly approach towards bioplastic production by utilizing microalgae as a feed source in several ways. First, the microalgae biomass obtained through the biorefinery approach can be processed into PHA under certain nutrient limitations. Additionally, microalgae biomass can act as potential filler and reinforcement towards the enhancement of bioplastic either blending with conventional bioplastic or synthetic polymer. The downstream processing of microalgae via suitable extraction and pre-treatment of bioactive compounds such as lipids and cellulose are found to be promising for the production of bioplastics. Moving on, the intermediate processing of bioplastic via lactic acid synthesized from microalgae has favoured the microwave-assisted synthesis of polylactic acid due to cost efficiency, minimum solvent usage, low energy consumption, and fast rate of reaction. Moreover, the reliability and effectiveness of microalgae-based bioplastics are further evaluated in terms of techno-economic analysis and degradation mechanism. Future improvement and recommendations are listed towards proper genetic modification of algae strains, large-scale biofilm technology, low-cost cultivation medium, and novel avocado seed-microalgae bioplastic blend.

Microalgae have emerged as an effective dual strategy for bio-valorisation of food processing wastewater and food waste hydrolysate which favours microalgae cultivation into producing value-added by products mainly lipids, carbohydrates, and proteins to the advantages of bioplastic production. Moreover, various microalgae have successfully removed high amount of organic pollutants from food processing wastewater prior discharging into the environment. Innovation of microalgae cultivating in food processing wastewater greatly reduced the cost of wastewater treatment compared to conventional approach in terms of lower carbon emissions, energy consumption, and chemical usage while producing microalgae biomass which can benefit low-cost fertilizer and bioplastic applications. The study on several microalgae species has all successfully grown on food waste hydrolysates showing high exponential growth rate and biomass production rich in proteins, lipids, carbohydrates, and fatty acids. Multiple techniques have been implemented for the extraction of food wastes to be incorporate into the bioplastic production.

Microbial carotenoids are gaining attention because of the unsustainable supplies of natural plant sources of carotenoids. The downstream processes of the microbial carotenoids are often limited by high cost of production and multi-steps unit operation. Aqueous biphasic system (ABS) comprised of alcohol and salt is suggested as a potential approach to overcome these limitations for recovery of Gordonia terrae carotenoids in this study. Aliphatic alcohols inclusive of 1-propanol and 2-propanol and sulphate, citrate and phosphate salts were used to develop the alcohol/salt ABS to recover the microbial carotenoids. The optimum ABS for the integration of G. terrae cells disruption and simultaneous recovery of carotenoids was developed by evaluating the effects of concentration of phase-forming components, pH, amount of crude loads, concentration of additives, sonication time on the recovery yield of carotenoids. Repetitive batch-extraction of carotenoids using the optimum alcohol/salt ABS was performed to investigate the feasibility of ABS as the sustainable and consistent method for recovery of microbial carotenoids. High recovery yield of carotenoids (90.86 ± 2.97%) was obtained with 20% (w/w) 1-propanol and 20% (w/w) trisodium citrate salt of pH 9. A total of 10% (w/w) of G. terrae cells was loaded into the optimized system with the addition of 4% (w/w) of NaCl. The optimized system is suggested to perform without sonication because degradation of the carotenoids was observed with sonication applied. A total of 4 batches of ABS extraction were conducted until all carotenoids were completely extracted from the G. terrae cells. The high-water content and cost-effective one-step ABS allows the process integration of extraction and separation of carotenoids from G. terrae fermentation.

Microbial astaxanthin with strong antioxidant activity is greatly demanded for diverse applications. Extractive disruption in aqueous biphasic system (ABS) integrates the cells disruption and biomolecules recovery processes in one-step operation, allowing the direct recovery of intracellular biomolecules with biphasic system upon released from cells. In this study, astaxanthin was recovered from recombinant Kluyveromyces marxianus yeast cells via extractive disruption using alcohol/salt ABS. Recombinant K. marxianus yeast is engineered to produce high concentration of free form astaxanthin. Highest partition coefficient (K = 90.02 ± 2.25) and yield (Y = 96.80% ± 0.05) of astaxanthin were obtained with ABS composed of 20% (w/w) 1-propanol and 20% (w/w) sodium citrate of pH 5, 0.5% (w/w) yeast cells loading and additional of 1% (w/w) 1-butyl-3-methylimidazolium tetrafluoroborate (Bmim)BF4 to improve the migration of astaxanthin to alcohol-rich top phase. The incorporation of 2.5 h of ultrasonication to the biphasic system further enhanced the astaxanthin recovery in ABS. The direct recovery of astaxanthin from recombinant K. marxianus cells was demonstrated with the ultrasonication-assisted alcohol/salt ABS which integrates the extraction and concentration of astaxanthin in a single-step operation.

Microalgae is a sustainable protein source that has been widely applied in animal feeds, functional foods, pharmaceutical, and cosmeceutical industries. Waste products could be a potential cost-saving and nutrient-rich substrate in the cultivation of microalgae for protein production. This study aims to investigate the cultivation condition of Haematococcus pluvialis for protein synthesis using synthetic brewery wastewater (BW). H. pluvialis was cultivated in the Bold's Basal Medium (BBM) mixed with synthetic BW at different concentrations. Various cultivation conditions including brewer's spent grain hydrolysate (BSGH) concentrations, pH, and light sources were studied. The molecular weight, amino acids profile and antioxidant activity of synthesized protein were determined. Carotenoids are a diverse group of lipid-soluble pigments that exhibit potent biological activities such as antioxidant, anti-inflammatory, and provitamin A activities. The potent health benefits of carotenoids result in the surge in the market demands for carotenoids, especially natural carotenoids from sustainable sources. Microbial carotenoids have attracted considerable interests for many industrial applications because of the low costs and ease of scaling-up with shorter production time. There is a growing interest in the search of new and sustainable microbial sources and cost-efficient production strategies following the high economical values and vast commercial applications of carotenoids. This article presents a review on the industrial production strategies of microbial carotenoids from microalgae, fungi, and bacteria sources. The industrial significance of the mass production of microbial carotenoids is also discussed. The structure, classification, and biosynthesis pathway of the carotenoids are also presented in this review.

Curcumin is one of the most valuable natural products due to its pharmacological activities. However, the low bioavailability of curcumin has long been a problem for its medicinal use. Large studies have been conducted to improve the use of curcumin; among these studies, curcumin metabolites have become a relatively new research focus over the past few years. Additionally, accumulating evidence suggests that curcumin or curcuminoid metabolites have similar or better biological activity than the precursor of curcumin. Recent studies focus on the protective role of plasma tetrahydrocurcumin (THC), a main metabolite of curcumin, against tumors and chronic inflammatory diseases. Nevertheless, studies of THC in eye diseases have not yet been conducted. Since ophthalmic conditions play a crucial role in worldwide public health, the prevention and treatment of ophthalmic diseases are of great concern. Therefore, the present study investigated the antioxidative, anti-inflammatory, antiangiogenic, and neuroprotective effects of THC on four major ocular diseases: age-related cataracts, glaucoma, age-related macular degeneration (AMD), and diabetic retinopathy (DR). While this study aimed to show curcumin as a promising potential solution for eye conditions and discusses the involved mechanistic pathways, further work is required for the clinical application of curcumin.

Process integration can be achieved using ABS by incorporating multiple-steps of purification, concentration and purification of biomolecules in a single-step operation which often results in high product recovery yield and purity. Conventional ABS is usually formed by aqueous solutions of two polymers or a polymer and a salt above a critical concentration. The high viscosity of polymer-based ABS causes slow phase separation and hinders the mass transfer of biomolecules, whereas polymer/salt ABS is characterized by high ionic strength resulting in the loss of bioactivity of recovered biomolecules. These limitations have encouraged the development of novel ABS which is more cost-effective for various biotechnological applications. This review discusses the characteristics and mechanisms of several types of emerging unconventional ABS using phase-forming components such as hyperbranched polymers, special salts, surfactants, magnetic fields, the addition of nanoparticles and incorporation of various solvent. Moreover, several novel applications of ABS for different separation purposes such as microfluidic-based ABS, ABS bioreactors, application of ABS as an analytical tool, and ABS micropatterning are discussed in this review. In the last section of this review, a comprehensive summary of process integration using ABS for extractive fermentations, bioconversion, crystallization and precipitation is also supplemented for the comprehensive review of various types and applications of ABS in recent years.

The paper evaluates the routes towards the fabrication and characterization of membranes using ZIF-62 metal organic framework (MOF) nano-hybrid dots for environmental remediation. Optimization of interaction of operating parameters over the rooted membrane is challenging issue. Subsequently, the interaction of operating parameters including temperature, pressure and CO2 gas concentration over the resultant rooted membranes are evaluated and optimized using response surface methodology for environmental remediation. In addition, the stability and effect of hydrocarbons on the performance of the resulting membrane during the gas mixture separation are evaluated at optimum conditions to meet the industrial requirements. The characterization results verified the fabrication of the ZIF-62 MOF rooted composite membrane. The permeation results demonstrated that the CO2 permeability and CO2/CH4 selectivity of the composite membrane was increased from 15.8 to 84.8 Barrer and 12.2 to 35.3 upon integration of ZIF-62 nano-glass into cellulose acetate (CA) polymer, respectively. Subsequently, the optimum conditions have been found at a temperature of 30 °C, the pressure of 12.6 bar and CO2 feed concentration of 53.3 vol%. These optimum conditions revealed the highest CO2 permeability, CH4 permeability and CO2/CH4 separation factor of 47.9 Barrer, 0.2 Barrer and 26.8, respectively. The presence of hydrocarbons in gas mixture dropped the CO2 permeability of 56.5% and separation factor of 46.4% during 206 h of testing. The separation performance of the composite membrane remained stable without the presence of hydrocarbons for 206 h.

Heavy metal pollution remains a global environmental challenge that poses a significant threat to human life. Various methods have been explored to eliminate heavy metal pollutants from the environment. However, most methods are constrained by high expenses, processing duration, geological problems, and political issues. The immobilization of metals, phytoextraction, and biological methods have proven practical in treating metal contaminants from the soil. This review focuses on the general status of heavy metal contamination of soils, including the excessive heavy metal concentrations in crops. The assessment of the recent advanced technologies and future challenges were reviewed. Molecular and genetic mechanisms that allow microbes and plants to collect and tolerate heavy metals were elaborated. Tremendous efforts to remediate contaminated soils have generated several challenges, including the need for remediation methodologies, degrees of soil contamination, site conditions, widespread adoptions and various possibilities occurring at different stages of remediation are discussed in detail.

The performance of tunable aqueous polymer phase impregnated resins (TAPPIR) which is the combination of the solvent impregnated resin principle and an aqueous two-phase system for the separation of esterase from Serratia marcescens was evaluated in this study. Different molecular weight of polyethylene glycol (PEG) (2000, 4000 and 6000) at concentration ranging from 5% to 20% (w/w) and potassium citrate were used to construct the aqueous phase in TAPPIR technology. Optimum composition of PEG and salt for esterase partitioning was determined using response surface methodology. The optimum condition for the purification of esterase was impregnation of 25% (w/w) of PEG 2000 into 4 mm porous glass beads and extraction of esterase using 15% (w/w) potassium citrate at pH 8 containing 12% (w/w) crude loading with the addition of 4% (w/w) NaCl. Esterase from S. marcescens was successfully purified by the TAPPIR technology up to 5.32 of purification factor with a yield of 75.98%.

Crude palm oil (CPO) and palm-pressed mesocarp fibre oil (PPMFO) consist of minor components such as Vitamin E, phytosterols, squalene, and carotene that play numerous health function in human. However, limitations are present in current separation processes to separate or enrich minor components from plant sources. In this study, interest is focusing on the enrichment efficiency of the minor components from CPO and PPMFO through proposed sequential adsorption-desorption strategy. The strategy started with the adsorption-desorption properties evaluation of six adsorbents. Both Diaion HP20 and Sepabeads SP850 showed better adsorption-desorption properties than silica gel, Florisil, Diaion HP2MG and Amberlite XAD-7HP. Diaion HP20 was selected as the suitable adsorbent to perform a series of selective desorption process using three different solvents: methanol, IPA, and n-hexane in Soxhlet extraction. Vitamin E, phytosterols, and squalene from CPO were obtained in the 1st fraction using methanol solution and their concentration increased from the initial concentration with the enrichment factor (EF) of 3.4, 3.9, and 1.8, respectively, which slightly higher than those minor components obtained from PPMFO, 1.2, 1.8, and 1.4, respectively. The carotene from both CPO and PPMFO was enriched in the 3rd fraction by using n-hexane solution with an enrichment factor of 1.1 and 1.5, respectively. In conclusion, the obtained result revealed the efficiency of the proposed sequential adsorption-desorption strategy to enrich the minor components from CPO and PPMFO.

Recent advances in genetic engineering technology have provided various methods in recombinant proteins production for pharmaceutical purposes. However, the development of the effective purification strategy remains the major challenge for large-scale production of therapeutic proteins because high purity of the proteins is often demanded. In this study, recombinant Oryza sativa L. cv Tainung 67 human serum albumin (OsrHSA) was expressed and purified with a single-step polyethylene glycol (PEG)/phosphate aqueous biphasic system (ABS). The optimum condition of PEG/phosphate ABS for the purification of OsrHSA was determined by investigating the effects of PEG molecular weight; pH; tie-line length (TLL); and volume ratio (VR) of ABS on the OsrHSA purification efficiency. The purification efficiency of OsrHSA was evaluated based on the partition coefficient (KHSA) and recovery yield (RB) of the OsrHSA in the PEG/phosphate ABS. An RB of 69.8% of OsrHSA was recovered in the optimum PEG/phosphate ABS consists of TLL of 26.0% (w/w) of pH 7.5. In addition, large-scale ABS were attempted and results showed that the recovery efficiency of 3000 g ABS was consistent with a 10 g ABS. This novel finding has demonstrated that the feasibility of ABS as a potential tool to purify HSA protein in one-step operation.

Ectoine is a compatible cell protectant sourced from halophilic and halotolerant microorganisms. Various biotechnological applications of ectoine have urged the development of cost-effective downstream processing of ectoine. Aqueous biphasic system (ABS) composed of poly(propylene) glycol (PPG) 425 and salt was adopted for the recovery of intracellular ectoine from Halomonas salina DSM5928T fermentation. Phosphate, citrate and sulfate salts were evaluated for their effects on the recovery of intracellular ectoine using the PPG/salt ABS. In this study, ectoine exhibited a salt-rich bottom phase preference in the PPG/salt ABS because of the hydrophilic surface properties of ectoine. The effects of concentrations of PPG 425 and salts, crude load, pH and concentrations of neutral salts (NaCl and KCl) on the recovery of ectoine were investigated. Optimum recovery of intracellular ectoine was obtained with ABS (pH 5.5) composed of 30% (w/w) PPG 425 and 20% (w/w) sulfate; 15% (w/w) crude load; and 1.5% (w/w) of KCl. High yield of 94.7% of intracellular ectoine was successfully recovered from H. salina cells with an enrichment factor (EF) of 1.7 and a purity of 87.03%.

Microalgae metabolites include biologically active compounds with therapeutic effects such as anticancer, anti-inflammatory and immunomodulation effects. One of the most recent focuses is on utilizing microalgae lipid-based biologically active compounds in food applications. However, most microalgae biological active compounds in their natural forms have common drawbacks like low solubility, low physicochemical stability and strong susceptibility to degradation, which significantly limits their application in foods, therefore, it is important to find solutions to retain their functional properties. In the present work, a comprehensive review on multi-product biorefinery was carried out from upstream processing stage to downstream processing stage, and identify critical processes and factors that impact bioactive material acquisition and retention. Furthermore, since nanoencapsulation technology emerges as an effective solution for microalgae nutraceutical product's retention, this work also focus on the nanoparticle perspective and comprehensively reviews the current nanoencapsulation solutions of the microalgae bioactive extract products. The aim is to depict advances in the formulations of microalage bioactive nanoparticles and provide a critical analysis of the reported nanoparticle formation. Overall, through the investigation of microalgae from biomass to bioactive nanoparticles, we aim to facilitate microalgae nutraceuticals incorporation as high value-added ingredients in more functional food that can improve human health.

Aqueous biphasic electrophoresis system (ABES) incorporates electric fields into the biphasic system to separate the target biomolecules from crude feedstock. Ionic liquid (IL) is regarded as an excellent candidate as the phase-forming components for ABES because of the great electrical conductivity, which can promote the electromigration of biomolecules in ABES, and thereby enhances the separation efficiency of the target biomolecules from crude feedstock. The application of electric fields to the conventional biphasic system expedites the phase settling time of the biphasic system, which eases the subsequent scaling-up steps and reduces the overall processing time of the recovery process. Alkyl sulphate-based IL is a green and economical halide-free surfactant when compared to the other halide-containing IL. The feasibility of halide-free IL-based ABES to recover Kytococcus sedentarius TWHK01 keratinase was studied. Optimum partition coefficient (Ke = 7.53 ± 0.35) and yield (YT = 80.36% ± 0.71) were recorded with IL-ABES comprised of 15.0% (w/w) [EMIM][ESO4], 20.0% (w/w) sodium carbonate and 15% (w/w) crude feedstock. Selectivity (S) of 5.75 ± 0.27 was obtained with the IL-ABES operated at operation time of 5 min with 10 V voltage supplied. Halide-free IL is proven to be a potential phase-forming component of IL-ABES for large-scale recovery of keratinase.
