目的 :分析跆拳道青少年選手完成 12週阻力訓練後，執行停訓 (detraining, DT) 或減量訓練 (tapering, TP)對力量表現之影響。方法 : 22名完成 12週阻力訓練之現役男性青少年跆拳道選手為受試者，以最大肌力水準分層隨機分配為 11位停訓組 (detraining group. DTG) 及 11位減量組 (tapering group, TPG)。TPG維持前一期訓練計畫中「速度與爆發力訓練期」之訓練強度與頻率，但訓練量減半的阻力訓練。前一期阻力訓練結束 48小時進行前測，前測結束後第 7天及第 14天分別進行中測與後測。依變項包括最大肌力、50毫秒發力率、100毫秒發力率及反向跳高度。以單因子相依變異數分析及獨立樣本t檢定比較組內與組間差異，組內差異若達顯著水準以 Bonferroni 法進行事後比較。結果 : DTG 中測與後測之最大肌力顯著優於前測 (p < .01)，中測之50毫秒發力率顯著優於前測 (p < .05)。TPG中測的最大肌力、50毫秒發力率、100毫秒發力率及反向跳高度顯著優於前測 (p < .01)，後測的50毫秒發力率亦優於前測及中測 (p < .01)。組間比較發現 DTG 中測的最大肌力進步率顯著優於TPG(p < .01)，但 TPG 後測的 50毫秒發力率進步率顯著優於 DTG (p < .01)。結論 : 7天與14天的DT與TP均能維持前一期阻力訓練之成果。7天 TP 比7天的 DT有更多力量發展的優勢且 14天TP的 50毫秒發力率達最高進步率。故TP策略比DT策略更有利跆拳道選手專項所需的力量表現。 Purpose: The study was to analyze the effect of detraining (DT) and tapering (TP) after 12-week resistant training on the strength performance of adolescent taekwondo athletes. Methods: 22 active male teenage taekwondo athletes who completed 12 weeks of resistance training were selected as subjects, and were randomly assigned to 11 detraining group (DTG) and 11 tapering group. TPG maintains the training intensity and frequency of the "speed and explosive power training period" in the previous training plan, but the training volume was halved. The pre-test was conducted 48 hours after the end of the previous resistance training cycle, and the mid-test and post-test were conducted on the 7th and 14th days after the pre-test. The dependent variables included maximal strength, 50-ms rate of force development (RFD), 100-ms RFD, and the height of countermovement jump. Repeated measured ANOVA and independent sample t-test were used to compare the differences within and between groups. Bonferroni’s method was used for post-hoc comparison if the differences within groups reached a significant level. Results: In DTG, the maximal strength at the mid-test and post-test was significantly greater than that of the pre-test (p< .01), and the 50-ms RFD at the mid-test was significantly better than that of the pre-test (p < .05). In TPG, the maximal strength, 50-ms RFD, 100-ms RFD, and the height of countermovement jump of the post-test were significantly better than those in the pre-test (p< .01), and the 50-ms RFD was significantly greater than the pre-test and mid-test. Between-group comparisons found that DTG was significantly better than TPG in the improvement of maximal strength (p < .01), but the 50-ms RFD at TPG post-test was significantly better than DTG post-test (p < .01). Conclusions: Both 7-day and 14-day DT and TP can maintain the training effect of pervious resistant training program. The 7-day TP could develop more strength than the 7-day DT, and the 50-ms RFD improved the most in 14-day TP. Therefore, the TP strategy is more beneficial to the strength performance required by taekwondo athletes than the DT strategy. &nbsp

The ongoing monkeypox virus outbreak includes at least 7553 confirmed cases in previously non-endemic countries worldwide as of July 2022. Clinical presentation has been reported as highly variable, sometimes lacking classically described systemic symptoms, and only small numbers of cutaneous lesions in most patients. The aim of this study was to compare clinical data with longitudinal qPCR results from lesion swabs, oropharyngeal swabs and blood in a well characterized patient cohort.16 male patients (5 hospitalized, 11 outpatients) were included in the study cohort and serial testing for monkeypox virus-DNA carried out in various materials throughout the course of disease. Laboratory analysis included quantitative PCR, next-generation sequencing, immunofluorescence tests and virus isolation in cell culture.All patients were male, between age 20 and 60, and self-identified as men having sex with men. Two had a known HIV infection, coinciding with an increased number of lesions and viral DNA detectable in blood. In initial- and serial testing, lesion swabs yielded viral DNA-loads at, or above 10The data presented underscore the reliability of lesion swabs for monkeypox virus-detection, even in later stages of the disease. Oropharyngeal swabs and blood samples alone carry the risk of false negative results, but may hold value in pre-/asymptomatic cases or viral load monitoring, respectively.

BackgroundSince May 2022, a rising number of monkeypox-cases has been reported in non-endemic countries of the northern hemisphere. In contrast to previous clusters, infections seem predominantly driven by human-to-human transmission, rather than animal sources. In this study, we adapted two published qPCR assays (non-variola orthopoxvirus and monkeypoxvirus specific) for use as a lab-developed dual-target monkeypoxvirus-test on widely used automated high-throughput PCR-systems (cobas5800/6800/8800).MethodsSelected assays were checked for in-silico inclusivity and exclusivity in current orthopoxvirus sequences, as well as for multiplex compatibility. Analytic performance was determined by serial dilution of monkeypoxvirus reference material, quantified by digital PCR. Cross reactivity was ruled out through a clinical exclusivity set containing various bloodborne and respiratory pathogens. Clinical performance was compared to a commercial manual RUO-kit using clinical remnant samples.ResultsAnalytic lower limit of detection (LoD) was determined as 4.795 dcp/ml (CI95%: 3.598 - 8.633 dcp/ml) for both assays combined, with a dynamic range of at least 5 log-steps. The assay showed 100% positive and negative agreement with the manual RUO orthopoxvirus PCR test kit in clinical swab samples.DiscussionWhile the full extend of the ongoing monkeypox outbreak remains to be established, the WHO and local health authorities are calling for increased awareness and efforts to limit further spread. For this, timely and scalable PCR tests are an important prerequisite. The assay presented here allows streamlined high-throughput molecular testing for monkeypoxvirus on existing hardware, broadly established previously for SARS-CoV-2 diagnostics.

Background: The recent emergence of distinct and highly successful SARS-CoV-2 lineages has substantial implications for individual patients and public health measures. While next-generation-sequencing is routinely performed for surveillance purposes, RT-qPCR can be used to rapidly rule-in or rule-out relevant variants, e.g., in outbreak scenarios. The objective of this study was to create an adaptable and comprehensive toolset for multiplexed Spike-gene SNP detection, which was applied to screen for SARS-CoV-2 B.1.617 lineage variants. Methods: We created a broad set of single nucleotide polymorphism (SNP)-assays including del-Y144/145, E484K, E484Q, P681H, P681R, L452R, and V1176F based on a highly specific multi-LNA (locked nucleic acid)-probe design to maximize mismatch discrimination. As proof-of-concept, a multiplex-test was compiled and validated (SCOV2-617VOC-UCT) including SNP-detection for L452R, P681R, E484K, and E484Q to provide rapid screening capabilities for the novel B.1.617 lineages. Results: For the multiplex-test (SCOV2-617VOC-UCT), the analytic lower limit of detection was determined as 182 IU/mL for L452R, 144 IU/mL for P681R, and 79 IU/mL for E484Q. A total of 233 clinical samples were tested with the assay, including various on-target and off-target sequences. All SNPs (179/179 positive) were correctly identified as determined by SARS-CoV-2 whole genome sequencing. Conclusion: The recurrence of SNP locations and flexibility of methodology presented in this study allows for rapid adaptation to current and future variants. Furthermore, the ability to multiplex various SNP-assays into screening panels improves speed and efficiency for variant testing. We show 100% concordance with whole genome sequencing for a B.1.617.2 screening assay on the cobas6800 high-throughput system.

Isomers are widely distributed in Chinese herbal medicines,and can be discriminated by energy-resolved mass spectrometry( ER-MS). However,ER-MS was performed through direct injection of reference compounds with syringe pump,which encountered a significant technical barrier for high-throughput and automated measurements. Herein,online ER-MS was conducted using LC-MS platform,and a pair of isomers,kaempferol vs luteolin,were employed as a case study to illustrate and assess the utility of online ER-MS for isomeric discrimination. High-resolution tandem mass spectrometry data of both flavonoids were acquired on LC-QE-Orbitrap-MS,and the fragmentation pathways responsible for the primary fragment ions were proposed. The primary signal in MS1 occurred at m/z 285( [M-H]-),and the primary signals of either compound generated by retro-Diels-Alder fragmentation were observed at m/z 151 and 133. The spectral information was subsequently transferred onto LC-Qtrap-MS platform to carry out online ER-MS. Two precursor-to-product ion transition candidates were constructed as m/z 285gt;151 and 285gt;133,and either afterward derived a set of pseudo-ion transitions( PITs) and so forth,exactly corresponding to a series of progressive collision energies( eg-5,-8,-11 e V,and so on). All PITs were typed into the monitoring list of multiple reaction monitoring program to generate the peak area datasets. Either dataset was normalized using the highest values in the set and imported into Graph Pad Prism software to plot the Gaus-sian-shaped curve that was termed as the break-down graph. The apex of the regressive curve was termed as optimal collision energy( OCE). The OCE values corresponding to m/z 285gt;151 were calculated as-29. 06 e V and-35. 71 e V for kaempferol and luteolin,respectively. In the case of m/z 285gt;133,the OCEs were yielded as-44. 15 e V for kaempferol and-49. 01 e V for luteolin. With re-ference to their chemical structures,the location of hydroxyl group was regarded to be responsible for the differences of either m/z 285gt;151 or 285gt;133 between the isomers,attributing to their different bond properties. Above all,online ER-MS offers an eligible tool for isomeric discrimination,and provides meaningful information for the accurate chemical composition characterization based on LC-MS,which is not limited to Chinese herbal medicines.

Human development continues to degrade ecosystems on a global scale. Such challenge calls for more informed decision-making in all fields of human activities so that each of our undertakings can be planned and designed for a more sustainable and equitable future. Sustainability assessment (SA), in its simplest form, is a tool that bridges evidence and decision-making. This study proposes to investigate the wholeness of SA from the perspective of behavioral decision-making theories, which can be classified into quantitative and qualitative approaches. Assessment and predictions in quantitative terms have their own merits; yet, in a world of emerging technologies and changing paradigms, we can no longer rely purely on mathematical representations of the decision to be made. SA currently in place often fails to give fair consideration to qualitative factors of more inherent nature, namely, the ethical issues relevant for sustainability. This study will elaborate the logics and the need of incorporating the ethical dimension into decision-making toward sustainability.

Taiwan, located in subtropical areas, relies heavily on air-conditioning to adjust the environment to maintain occupant comfort and work efficiency. Among numerous indicators that measure living comfort, the one corresponding to the change of temperature and relative humidity is the most felt by people. This study will investigate data on changes in the atmospheric environment in Taiwan over the years, and will correlate them to temperature and relative humidity within the comfort range for human beings. The Heat Index will also be examined and applied in this research in order to help air-conditioning systems better adapt to more volatile weather patterns and variable human needs. A simultaneous and precise control of temperature and relative humidity with reference to the Heat Index can reduce the load for air-conditioning without compromising environmental comfort.

Using the FDTD method, we investigate the electromagnetic propagation in two-dimensional photonic crystals, formed by parallel air cylinders in a dielectric medium. The corresponding frequency band structure is computed using the standard plane-wave expansion method. It is shown that within partial bandgaps, waves tend to bend away from the forbidden directions. This phenomenon perhaps need not be explained in terms of negative refraction or `superlensing' behavior, contrast to what has been conjectured., Comment: 3 pages, 4 figures
