Abstract Background The intricacies of nucleotide metabolism within tumor cells specific to colorectal cancer (CRC) remain insufficiently characterized. A nuanced examination of particular tumor clusters and their dynamic interplay with the tumor microenvironment (TME) may yield profound insights into these therapeutically auspicious communicative networks. Methods By integrating ten types of single-cell enrichment scoring methods, we carried out enrichment analysis on CRC cell types, which was validated through four additional single-cell cohorts. Groups of tumor cells were determined using the average values of the scores. Using cellphonedb, monocle, inferCNV, SCENIC, and Cytotrace, functional analyses were performed. Utilizing the RCTD approach, single-cell groupings were mapped onto spatial transcriptomics, analyzing cell dependency and pathway activity to distinguish between tumor cell subtypes. Differential expression analysis identified core genes in nucleotide metabolism, with single-cell and spatial transcriptomics analyses elucidating the function of these genes in tumor cells and the immune microenvironment. Prognostic models were developed from bulk transcriptome cohorts to forecast responses to immune therapy. Laboratory experiments were conducted to verify the biological function of the core gene. Results Nucleotide metabolism is significantly elevated in tumor cells, dividing them into two groups: NUhighepi and NUlowepi. The phenotype NUhighepi was discerned to exhibit pronounced malignant attributes. Utilizing the analytical tool stlearn for cell-to-cell communication assessment, it was ascertained that NUhighepi engages in intimate interactions with fibroblasts. Corroborating this observation, spatial transcriptome cell interaction assessment through MISTy unveiled a particular reliance of NUhighepi on fibroblasts. Subsequently, we pinpointed NME1, a key gene in nucleotide metabolism, affirming its role in thwarting metastasis via in vitro examination. Utilizing multiple machine learning algorithms, a stable prognostic model (NRS) has been developed, capable of predicting survival and responses to immune therapy. In addition, targeted drugs have been identified for both high and low scoring groups. Laboratory experiments have revealed that NME1 can inhibit the proliferation and invasion of CRC tumor cells. Conclusion Our study elucidates the potential pro-tumor mechanism of NUhighepi and the role of NME1 in inhibiting metastasis, further deepening the understanding of the role of nucleotide metabolism in colorectal cancer, and providing valuable targets for disrupting its properties.

Relational governance has showed great potential in addressing many of the construction industry’s problems, such as opportunism and agency problems. While multiple research efforts have been directed at examining different roles of relational governance elements, few studies paid attention to show the link between different relational governance elements and project management performance. So, this study aims to address such gap by fully considering relational governance elements affecting project management performance. To this end, this paper (1) divided relational governance elements into relational situation, relational norm, and relational behavior for the first time, deepening our understanding of relational governance elements in the construction industry; (2) determined the impact of these relational governance elements on project management performance. The findings contribute to the understanding of relational governance elements in the construction industry, how relational governance and project management performance are really linked, and what the fundamental paths are in the chain by specifying the elements of relational governance in a more detailed way. These findings also give owners and contractors insight into improving project management performance.

This study aimed to select Lactobacillus with high bile salt hydrolase (BSH) and to explore its protective role against neonatal jaundice. The LBS selective medium supplemented with 25 U/mL nystatin was used to screen and identify Lactobacillus strains isolated from the feces of healthy newborns and breast milk. L. rhamnosus (LGG) was used as a positive control to evaluate the in vitro probiotic properties and bile salt hydrolase activity of the Lactobacillus strains. Thereafter, the protective effect and mechanism of the selected isolates with high bile salt hydrolase activity against phenylhydrazine hydrochloride (PHZ)-induced neonatal jaundice in rats were investigated by analyzing serum bilirubin levels, hepatotoxicity, and the expression of inflammatory cytokines and nuclear transcription factors in the liver. The results showed that compared with the LGG, L. gasseri FWJL-5, which was isolated from infant feces, exhibited outstanding in vitro probiotic properties and higher bile salt hydrolase activity than LGG. L. gasseri FWJL-5 significantly alleviated the increase in serum bilirubin levels, liver tissue swelling and hemolysis in PHZ-treated neonatal rats, reduced the release of hepatic enzymes, inhibited the secretion of proinflammatory cytokines, and promoted the expression of UGT1A1, pregnane X receptor (PXR), and farnesol X receptor (FXR). Taken together, L. gasseri FWJL-5 could regulate bilirubin metabolism in the liver by up-regulating the expression of FXR/PXR and UGT1A1, thereby alleviating neonatal jaundice in rats. Our findings may provide new ideas for the prevention and treatment of neonatal jaundice by L. gasseri.

Abstract Background Delphinium L. represents a taxonomically intricate genus of significant phylogenetic and economic importance in Ranunculaceae. Despite the existence of few chloroplast genome datasets, a comprehensive understanding of genome structures and selective pressures within the genus remains unknown. Furthermore, several taxa in this genus are exclusively found in Xinjiang, China, a region renowned for its distribution and diversity of Chinese and Central Asian Delphinium species. Therefore, investigating the features of chloroplast genomes in this area will provide valuable insights into the evolutionary processes and phylogenetic relationships of the genus. Results In this study, the eight newly completed chloroplast genomes are examined, ranging in length from 153,979 bp to 154,284 bp. Alongside these, analysing six previously reported taxa re-annotated in Delphinium, 111 unique genes are identified across all samples. Genome structure, distributions of simple sequence repeats and short dispersed repeats, as well as gene content are similar among these Delphinium taxa. Nine hypervariable intergenic spacers and protein coding regions, including ndhF-trnL(TAG), rpl16-intron, rpl33, rps15, rps18, trnK(TTT)-trnQ(TTG), trnP(TGG)-psaJ, trnT(GGT)-psbD and ycf1, are identified among 13 perennial Delphinium. Selective pressure and codon usage bias of all the plastid genes are performed within 14 Delphinium taxa. Phylogenetic analysis based on 14 Delphinium plastomes, alongside two Aconitum (Ranunculaceae) species serving as outgroup taxa, reveals the monophyletic nature of Delphinium. Our findings further discern Delphinium into two distinct clades: perennial species (clade I) and annual species (clade II). In addition, compared with the nrDNA ITS topology, cytological data and morphological characters, D. mollifolium and D. maackianum showed potential involvement in hybridization or polyploidization processes. Excluding these two species, the perennial Delphinium (clade I) exhibits a stronger consistency with the morphology-based system that utilized seed morphology. Conclusion This study represents the first comprehensive analysis of plastomic variations among Delphinium taxa, based on the examination of 14 complete plastomes. The chloroplast genome structure of Delphinium is similar to other angiosperms and possesses the typical quadripartite structure with the conserved genome arrangement and gene features. In addition, the variation of non-coding regions is larger than coding regions of the chloroplast genome. Through DNA sequence divergence across Delphinium plastomes and subsequent phylogenomic analyses ndhF-trnL(TAG) and ycf1 are identified as promising molecular markers. These highly variable loci held significant potential for future phylogenetic and phylogeographic studies on Delphinium. Our phylogenomic analyses based on the whole plastomes, concatenation of 132 unique intergenic spacer regions, concatenation of 77 unique protein-coding genes and nrDNA ITS, all support the monophyly of Delphinium and perennial taxa clusters together into one clade within this genus. These findings provide crucial data for systematic, phylogenomic and evolutionary research in the genus for future studies.

The construction phase of nuclear construction projects presents various risks and hazards, putting workers in potentially dangerous situations. Safety guidelines provided by Occupational Health and Safety Administration (OSHA) aim to address construction safety concerns, but limitations persist, especially in the context of workers safety and the transportation of Small Modular Reactors (SMRs) and Micro Reactors (MRs) in the nuclear construction domain. This research focuses on identifying issues related to safety guidelines in nuclear construction and aims to develop new guidelines and strengthen existing ones. Through a critical literature review, key factors influencing safety in nuclear construction were identified. A questionnaire survey was prepared in line with those identified key safety factors and distributed among 40 professionals from the United States to gather feedback on these factors in countering safety issues and developing new guidelines in the context of nuclear construction. The findings reveal a lack of specific guidelines concerning the safe transportation of SMRs and MRs, as well as a gap in understanding accidents during the construction and transportation phases of nuclear projects. The results of this research contribute to enhancing safety protocols and addressing the unique challenges faced in nuclear construction. The research has also highlighted the importance of digital technology tools such as Artificial Intelligence (AI), Augmented Reality (AR), Virtual Reality (VR), and Unmanned Aerial Vehicles (UAVs) as emerging digital tools, which could be helpful to counter safety concerns linked to SMRs and MRs in future.

Abstract Background While de novo cholesterol biosynthesis plays a crucial role in chemotherapy resistance of colorectal cancer (CRC), the underlying molecular mechanism remains poorly understood. Methods We conducted cell proliferation assays on CRC cells with or without depletion of squalene epoxidase (SQLE), with or without 5-fluorouracil (5-FU) treatment. Additionally, a xenograft mouse model was utilized to explore the impact of SQLE on the chemosensitivity of CRC to 5-FU. RNA-sequencing analysis and immunoblotting analysis were performed to clarify the mechanism. We further explore the effect of SQLE depletion on the ubiquitin of NF-κB inhibitor alpha (IκBα) and (S)-2,3-epoxysqualene on the binding of IκBα to beta-transducin repeat containing E3 ubiquitin protein ligase (BTRC) by using immunoprecipitation assay. In addition, a cohort of 272 CRC patients were selected for our clinical analyses. Results Mechanistically, (S)-2,3-epoxysqualene promotes IκBα degradation and subsequent NF-κB activation by enhancing the interaction between BTRC and IκBα. Activated NF-κB upregulates the expression of baculoviral IAP repeat containing 3 (BIRC3), sustains tumor cell survival after 5-FU treatment and promotes 5-FU resistance of CRC in vivo. Notably, the treatment of terbinafine, an inhibitor of SQLE commonly used as antifungal drug in clinic, enhances the sensitivity of CRC to 5-FU in vivo. Additionally, the expression of SQLE is associated with the prognosis of human CRC patients with 5-FU-based chemotherapy. Conclusions Thus, our finding not only demonstrates a new role of SQLE in chemoresistance of CRC, but also reveals a novel mechanism of (S)-2,3-epoxysqualene-dependent NF-κB activation, implicating the combined potential of terbinafine for 5-FU-based CRC treatment.

Abstract Triple-negative breast cancer (TNBC) is highly malignant and lacks effective biotherapeutic targets. The development of efficient anticancer drugs with low toxicity and few side effects is a hotspot in TNBC treatment research. Although erianin is known to have potent antitumor activity, its regulatory mechanism and target in TNBC have not been fully elucidated, hampering further drug development. This study showed that erianin can significantly inhibit TNBC cell proliferation and migration, promote cell apoptosis, and inhibit the growth of transplanted tumors in mice. Mechanistically, through network pharmacology analysis, molecular docking and cellular thermal shift assays, we preliminarily identified SRC as the cellular target of erianin. Erianin potently inhibited the expression of SRC, which mediated the anticancer effect of erianin in TNBC. Moreover, erianin can downregulate the expression of genes related to cholesterol synthesis and uptake by targeting SRC, interfering with cholesterol levels in TNBC, thereby inhibiting the progression of TNBC in vivo and in vitro. Taken together, our results suggest that erianin may inhibit the progression of TNBC by suppressing SRC-mediated cholesterol metabolism, and erianin has the great potential to be an effective treatment for TNBC patients.

Abstract Intestinal stem cells (ISCs) play a crucial role in the continuous self-renewal and recovery of the intestinal epithelium. In previous studies, we have revealed that the specific absence of Claudin-7 (Cldn-7) in intestinal epithelial cells (IECs) can lead to the development of spontaneous colitis. However, the mechanisms by which Cldn-7 maintains homeostasis in the colonic epithelium remain unclear. Therefore, in the present study, we used IEC- and ISC-specific Cldn-7 knockout mice to investigate the regulatory effects of Cldn-7 on colonic Lgr5+ stem cells in the mediation of colonic epithelial injury and repair under physiological and inflammatory conditions. Notably, our findings reveal that Cldn-7 deletion disrupts the self-renewal and differentiation of colonic stem cells alongside the formation of colonic organoids in vitro. Additionally, these Cldn-7 knockout models exhibited heightened susceptibility to experimental colitis, limited epithelial repair and regeneration, and increased differentiation toward the secretory lineage. Mechanistically, we also established that Cldn-7 facilitates the proliferation, differentiation, and organoid formation of Lgr5+ stem cells through the maintenance of Wnt and Notch signalling pathways in the colonic epithelium. Overall, our study provides new insights into the maintenance of ISC function and colonic epithelial homoeostasis.

In the context of “Industry 4.0,” the construction industry is undergoing a continuous process of digital transformation. In recent years, with the rapid development of artificial intelligence (AI) technology, AI has been successfully applied across various domains within the construction sector. This paper conducts a comprehensive review based on scientific bibliometric analysis and systematic evaluation to provide an overview of AI applications and developments in the architecture, engineering, and construction (AEC) industry, as well as to discuss future research trends. Firstly, a bibliometric analysis was carried out on 3,410 journal articles published between 2003 and 2022. This analysis examined characteristics such as annual publication trends, key researchers and their affiliations, countries of origin, collaborative relationships, prominent journals, and keyword clustering. Subsequently, through systematic review, three critical themes in the field were identified and analyzed: performance prediction, intelligent optimization, and engineering safety and risk. To gain a deeper understanding of cutting-edge AI technologies and their potential applications in the AEC sector, potential future research directions were discussed, including digital twins, blockchain, augmented reality, and virtual reality, all aimed at driving the intelligent and digital transformation of the AEC industry.

Abstract Background Numerous previous reports have demonstrated the efficacy of Lactic acid bacteria (LAB) in promoting growth and preventing disease in animals. In this study, Enterococcus faecium ZJUIDS-R1 and Ligilactobaciiius animalis ZJUIDS-R2 were isolated from the feces of healthy rabbits, and both strains showed good probiotic properties in vitro. Two strains (108CFU/ml/kg/day) were fed to weaned rabbits for 21 days, after which specific bacterial infection was induced to investigate the effects of the strains on bacterial diarrhea in the rabbits. Results Our data showed that Enterococcus faecium ZJUIDS-R1 and Ligilactobaciiius animalis ZJUIDS-R2 interventions reduced the incidence of diarrhea and systemic inflammatory response, alleviated intestinal damage and increased antibody levels in animals. In addition, Enterococcus faecium ZJUIDS-R1 restored the flora abundance of Ruminococcaceae1. Ligilactobaciiius animalis ZJUIDS-R2 up-regulated the flora abundance of Adlercreutzia and Candidatus Saccharimonas. Both down-regulated the flora abundance of Shuttleworthia and Barnesiella to restore intestinal flora balance, thereby increasing intestinal short-chain fatty acid content. Conclusions These findings suggest that Enterococcus faecium ZJUIDS-R1 and Ligilactobaciiius animalis ZJUIDS-R2 were able to improve intestinal immunity, produce organic acids and regulate the balance of intestinal flora to enhance disease resistance and alleviate diarrhea-related diseases in weanling rabbits.

Blockchain-based cryptocurrencies, such as Bitcoins, are increasingly popular. However, the decentralized and anonymous nature of these currencies can also be (ab)used for nefarious activities such as money laundering, thus reinforcing the importance of designing tools to effectively detect malicious transaction misbehaviors. In this paper, we propose TMAS, a transaction misbehavior analysis scheme for blockchain-based cryptocurrencies. Specifically, the proposed system includes ten features in the transaction graph, two heuristic money laundering models, and an analysis method for account linkage, which identifies accounts that are distinct but controlled by an identical entity. To evaluate the effectiveness of our proposed indicators and models, we analyze 100 million transactions and compute transaction features, and are able to identify a number of suspicious accounts. Moreover, the proposed methods can be applied to other cryptocurrencies, such as token-based cryptocurrencies (e.g., Bitcoins) and account-based cryptocurrencies (e.g., Ethereum).

BackgroundImmunoglobulin A nephropathy (IgAN) is the most prevalent form of chronic kidney disease (CKD), marked by diverse pathological patterns and variable prognostic outcomes. Nutritional indexes are crucial for disease assessment and prognosis prediction. This study investigates associations between nutritional indexes and renal function in patients with IgAN.MethodsA cohort of 736 adults diagnosed with IgAN, who underwent renal biopsy at the First Hospital of Jilin University between January 2010 and October 2022, was examined. Clinical and laboratory data were reviewed, and four nutritional indexes were calculated: controlling nutritional status (CONUT) score, geriatric nutritional risk index (GNRI), body mass index (BMI), and prognostic nutritional index (PNI). Summary: Background: Whether hepatitis B virus (HBV) infection of women prior to pregnancy can influence risk of congenital malformations in offspring remains controversial. We assessed the association between them by considering congenital malformations in the aggregate as well as risk of organs systems using a large national sample of Chinese women. Methods: We performed a record-linkage cohort study of women who participated in National Free Preconception Health Examination Project, between January 1, 2010, and December 31, 2019 for whom data on congenital malformations in their offspring were available from the National Population-Based Birth Defects Surveillance Network. A total of 498,968 linked records were obtained, of which 127,371 were excluded because HBV status before pregnancy was unknown, the records involved multiple pregnancies, or pre-pregnancy examinations were conducted after conception. Based on pre-pregnancy status, mothers were assigned to two categories of HBsAg− or HBsAg+ and, in certain analyses, to three categories of HBsAg−, HBsAg+/HBeAg− or HBsAg+/HBeAg+. Potential associations of serological status with risk of congenital malformations, considered separately or in aggregate, were explored using multilevel logistic regression. Factors that might influence such associations were also explored. Findings: Among the 371,597 women analyzed, 21,482 (5.78%) were HBsAg+ before pregnancy, and 8333 (2.24%) had a fetus or child diagnosed with congenital malformations, composed of 7744 HBsAg− women and 589 HBsAg+ women. HBsAg+ status was associated with increased risk of congenital malformations in the aggregate (OR 1.14, 95% CI 1.03–1.25) and of cardiovascular malformations specifically (OR 1.18, 95% CI 1.03–1.35). HBsAg+/HBeAg− status was associated with significantly higher risk of cardiovascular malformations (OR 1.19, 95% CI 1.01–1.39) as well as reproductive malformations (OR 1.51, 95% CI 1.02–2.23). Associations between HBsAg+ status before pregnancy and risk of congenital malformations was modified by alanine aminotransferase activity (Pinteraction < 0.05). Interpretation: Prepregnancy HBV infection might be associated with fetal malformations. This association needs further investigation to confirm whether it is a causal association, and assess whether antiviral therapy of women with HBsAg+ planning to conceive might reduce the risk of fetal malformations. Abstract Ozone (O3) pollution is a severe air quality issue in China, posing a threat to human health and ecosystems. The climate change will affect O3 levels by directly changing physical and chemical processes of O3 and indirectly changing natural emissions of O3 precursors. In this study, near‐surface O3 concentrations in China in 2030 and 2060 are predicted using the process‐based interpretable Extreme Gradient Boosting (XGBoost) model integrated with multi‐source data. The results show that the climate‐driven O3 levels over eastern China are projected to decrease by more than 0.4 ppb in 2060 under the carbon neutral scenario (SSP1‐1.9) compared with the high emission scenario (SSP5‐8.5). Among this reduction, 80% is attributed to the changes in physical and chemical processes of O3 related to a cooler climate, while the remaining 20% is attributed to the reduced biogenic isoprene emissions.

National parks are rare natural resources that provide refuges for wildlife, harmonious ecology, and leisure activities. These protected areas, which include a variety of ecosystems, habitats for wildlife, and geological wonders, have been set aside to preserve and highlight the beauty of the natural world. Under the goals of carbon neutrality of all countries in the world, it is urgently need to pay attention to the relationship between climate change and protected natural resources. This study uses the Xianju pilot national park as an example to identify key activities that increase carbon absorptions and reduce carbon emissions, calculate the carbon sink of national park construction, and discuss whether tourism industry can achieve low carbon or even negative emissions based on the analysis of low-carbon measures of national parks. Using the Mento-Carlo method, we find that the total carbon sink in Xianju National Park is around 70832–97930 tons CO2, which accounts for about 10 % of Xianju County's total carbon emissions. This illustrates that the development of tourism has indeed brought benefits to the low carbon development of Xianju National Park. From the perspective of stakeholders, this paper also summarizes the stakeholders, relevant actions and policy tools for Xianju National Park to carry out low-carbon tourism. Finally, compared with the development experience of national parks in France and the United States, this paper gives policy suggestions on low-carbon development of Xianju National Park, and proposes to mobilize stakeholders to participate in low-carbon tourism and low-carbon governance.

In this study, microcapsules were prepared using conjugated linoleic acid (CLA) from Tan sheep tail fat as the core material and whey protein isolate (WPI) and maltodextrin (MD) as the wall material, and their physicochemical properties were characterized using scanning electron microscopy (SEM), laser scanning confocal microscopy (LSCM), thermogravimetric analysis (TGA), and X-ray diffraction (XRD) analysis. Besides, the oxidative stability and release behavior of the microcapsules were analyzed through accelerated storage experiments and in vitro simulated digestion experiments, and their antioxidant and cholesterol lowering activities were evaluated as well. The results showed that the particles of the CLA microcapsules were intact, with a relatively smooth surface, and had good thermal stability, oxidative stability, and sustained-release performance. After gastrointestinal digestion, the antioxidant stability and cholesterol lowering activity of the microcapsules were significantly higher than those of free CLA. At the end of in vitro simulated digestion, the scavenging rates of the microcapsules against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation radical, and superoxide anion radical were (70.84 ± 3.97)%, (78.53 ± 4.28)%, and (59.88 ± 2.58)%, respectively. The inhibition rate against cholesterol micelles was (73.48 ± 2.53)%, and the binding rates to sodium taurocholate, sodium glycine cholate, and sodium cholate were (40.62 ± 3.14)%, (66.79 ± 3.01)%, and (55.43 ± 2.39)%, respectively. This study indicates that microencapsulation not only significantly enhances the oxidative stability of Tan sheep tail fat CLA, but also improves its antioxidant and cholesterol lowering activity. This study provides a theoretical basis for the deep processing and comprehensive utilization of Tan sheep tail fat CLA.

Abstract Background To determine the prognostic value of cumulative calcification score of coronary artery calcification (CAC), thoracic aortic calcification (TAC) and aortic valve calcification (AVC) in acute ST segment elevation myocardial infarction (STEMI) patients. Methods This was a retrospective, single-center cohort study. A total of 332 STEMI patients who received primary percutaneous coronary intervention (PPCI) were enrolled in this study between January 2010 to October 2018. We assessed the calcification in the left anterior descending branch (LAD), left circumflex branch (LCX), right coronary artery (RCA), thoracic aorta, and aortic valve. Calcification of each part was counted as 1 point, and the cumulative calcification score was calculated as the sum of all points. The primary endpoint was all-cause mortality. Multivariate Cox proportional hazards models were used to determine association of cumulative calcification score with end points. The performance of the score was evaluated by receiver operating characteristic (ROC) curve analysis and absolute net reclassification improvement (NRI), compared with the Global Registry of Acute Coronary Events (GRACE) risk score. Results The overall population’s calcification score was 2.0 ± 1.6. During a mean follow-up time of 69.8 ± 29.3 months, the all-cause mortality rate was 12.1%. Abstract With the exponential growth of various data interactions on network systems, network intrusions are also increasing. The emergence of edge computing technology brings a new solution to network security. However, due to the difficulty of processing massive and unbalanced data at the edge, higher accuracy requirements are necessary for deployed detection models. This paper proposes a multi-classification model for network intrusion detection based on reconstruction and feature matching. This model can be deployed on small-scale edge nodes, effectively identifying various attack behaviors through the utilization of reconstruction errors and adaptive scaling. Furthermore, we proposed a model transfer method based on feature matching to enhance the training and detection efficiency of multi-classification models under different data distribution conditions. The proposed model has been evaluated on the CICIDS2017 dataset in terms of accuracy, recall, precision and F1 score. The model demonstrates high accuracy for normal flows in the network, majority class attacks, and minority class attacks, achieving an overall multi-class accuracy of 99.81%, outperforming similar models. Furthermore, this model demonstrates faster convergence and training speed after feature matching, exhibiting better robustness and outstanding performance at the edge.

Infectious bursal disease (IBD) is a widespread problem in the poultry industry, and vaccination is the primary preventive method. However, moderately virulent vaccines may damage the bursa, necessitating the development of a safe and effective vaccine. The Newcastle disease virus (NDV) has been explored as a vector for vaccine development. In this study, reverse genetic technology was used to obtain three recombinant viruses, namely, rClone30-VP2L (P/M)-chGM-CSF (NP), rClone30-chGM-CSF (P/M)-VP2L (NP), and rClone30-VP2L-chGM-CSF (P/M). Animal experiments showed that the three biological adjuvant bivalent vaccines effectively increased anti-NDV and anti-infectious bursal disease virus (IBDV) titres, enhancing both humoral and cellular immune responses in chickens without leading to any harm. Amongst the three biological adjuvant bivalent vaccines, the rClone30-chGM-CSF (P/M)-VP2L (NP) group had higher levels of anti-NDV antibodies at 14 days after the first immunization and stimulated a greater humoral immune response in 7–10 days. While, the rClone30-VP2L (P/M)-chGM-CSF (NP) group was the most effective in producing a higher level of IBDV antibody response. In conclusion, these three vaccines can induce immune responses more rapidly and effectively, streamline production processes, be cost-effective, and provide a new avenue for the development of Newcastle disease (ND) and IBD bivalent vaccines

Recently, CAR-T cell therapy in hematological malignancies has received extensive attention. The objective of this study is to gain a comprehensive understanding of the current research status, development trends, research hotspots, and emerging topics pertaining to CAR-T cells in the treatment of hematological malignancies. Articles pertaining to CAR-T cell therapy for hematological malignancies from the years 2012 to 2023 were obtained and assessed from the Web of Science Core Collection (WoSCC). A bibliometric approach was employed to conduct a scientific, comprehensive, and objective quantitative analysis, as well as a visual analysis, of this particular research domain. A comprehensive analysis was conducted on a corpus of 3643 articles, which were collaboratively authored by 72 countries and various research institutions. CAR-T cell research in treating hematological malignancies shows an increasing trend each year. Notably, the study identified the countries and institutions displaying the highest level of activity, the journals with the most citations and output, as well as the authors who garnered the highest frequency of citations and co-citations. Furthermore, the analysis successfully identified the research hotspots and highlighted six emerging topics within this domain. This study conducted a comprehensive exploration and analysis of the research status, development trends, research hotspots, and emerging topics about CAR-T cells in the treatment of hematological malignancies from 2012 to 2023. The findings of this study will serve as a valuable reference and guide for researchers seeking to delve deeper into this field and determine the future direction of their research.

Appropriate risk sharing is crucial for the successful implementation of Public-Private Partnership projects. However, under different social systems, the international community lacks a clear risk-sharing framework in Public-Private Partnership research. This study employs a multi-case study method to examine the practice of risk sharing in infrastructure Public-Private Partnership projects within China and the US, aiming to provide a clear risk management framework for Public-Private Partnership projects. The research findings reveal common elements and similar characteristics in risk-sharing practices. Contrary to the goal of transferring all risks to the private sector, both countries actually allocate risks to the party with stronger risk control capabilities or share risks between the government and the private sector. The US benefits from more mature policies and has a comprehensive Public-Private Partnership model implementation framework; in contrast, as an emerging market for Public-Private Partnership models, China's current laws and regulations need further improvement. The results of this study not only contribute to optimizing risk-sharing arrangements and improving investment efficiency of the project, but also promote the development of cross-border infrastructure projects, and provide valuable experience and insights for the sustainable development of global infrastructure.

Background Inflammation plays a crucial role in occurrence of kidney injury, and specific dietary patterns can influence systemic inflammation levels. However, the relationship between dietary inflammatory potential and early-stage kidney damage remains unclear.Method 2,108 participants was recruited from 2001–2002 National Health and Nutrition Examination Survey (NHANES). Dietary Inflammatory Index (DII) is utilized to assess dietary inflammatory potential, calculated through a 24-h dietary recall questionnaire. Early renal injury was evaluated using urinary albumin to creatinine (UACR), cystatin C (CysC), β-2 microglobulin (β2M), and estimated glomerular filtration rate (eGFR) based on serum creatinine (eGFRs), cystatin C (eGFRc), and both Scr and CysC (eGFRs&c). Participant characteristics were analyzed, and association between DII, hypertension, and early renal injury markers was explored using multiple linear and logistic regression models.Results The average age of participants was 53.9 years. DII exhibited a positive correlation with UACR (β = −0.048[0.017,0.078]), β2M (β = 0.019[0.010,0.027]), CysC (β = 0.012 [0.004,0.021]). Conversely, a negative correlation was observed between DII and eGFRc (β = −1.126[−1.554, −0.699]), eGFRs&c (β=-1.101[−1.653, −0.549]). A significant association was observed between hypertension and abnormality of early kidney damage markers. Subgroup analysis reveals that the positive correlation between DII and the occurrence of abnormal markers of early kidney damage is only observed in individuals with hypertension. Furthermore, an interaction between DII and hypertension was detected in eGFRs&c (OR:1.250[1.042, 1.499], p for interaction = 0.03).Conclusion Higher levels of DII may be associated with occurrence of early kidney damage. For individuals with hypertension, avoiding excessive consumption of pro-inflammatory foods may reduce the risk of renal injury.

Chorea-acanthocytosis (ChAc) is a rare autosomal recessive inherited syndrome with heterogeneous symptoms, which makes it a challenge for early diagnosis. The mutation of VPS13A is considered intimately related to the pathogenesis of ChAc. To date, diverse mutation patterns of VPS13A, consisting of missense, nonsense, and frameshift mutations, have been reported. In this study, we first report a clinical case that was misdiagnosed as epilepsy due to recurrent seizures accompanied by tongue bite for 9 months, which was not rectified until seizures were controlled and involuntary orolingual movements with awareness became prominent and were confirmed to be orolingual dyskinesia. The patient was eventually diagnosed as ChAc based on whole-exome sequencing revealing novel homozygous c.2061dup (frameshift mutation) and c.6796A > T dual mutations in VPS13A. The patient from a family with consanguineous marriage manifested epileptic seizures at onset, including both generalized tonic–clonic seizures and absence but normal long-term electroencephalography, and gradually developed orofacial dyskinesia, including involuntary tongue protrusion, tongue biting and ulcers, involuntary open jaws, occasionally frequent eye blinks, and head swings. The first test of the peripheral blood smear was negative, and repeated checks confirmed an elevated percentage of acanthocytes by 15–21.3%. Structural brain MRI indicated a mildly swollen left hippocampus and parahippocampal gyrus and a progressively decreased volume of the bilateral hippocampus 1 year later, along with atrophy of the head of the caudate nucleus but no progression in 1 year. We deeply analyzed the reasons for long-term misdiagnosis in an effort to achieve a more comprehensive understanding of ChAc, thus facilitating early diagnosis and treatment in future clinical practice.

Purpose: This study aims to identify the key factors influencing health-related quality of life (HRQoL) of pediatric acute myeloid leukemia (AML) patients following their initial diagnosis and examine their impact on the five-year survival prognosis. Methods: A chart review and follow-up were conducted for children with AML who participated in a prospective cohort study between 2017 and 2020. We identified factors influencing HRQoL through Pediatric Quality of Life Inventory™ (PedsQL™ 4.0), PedsQL™ Cancer Module 3.0 (CM 3.0) and PedsQL™ Family Impact Module 2.0 (FIM 2.0), as well as assessed the impact of impaired HRQoL on the overall outcomes of patients. Results: Sixty-four subjects enrolled in the study had complete HRQoL outcome data, and 61 of them completed the 5-year follow-up. In CM 3.0, age was positively associated with parental proxy reports (p = 0.040), whereas divorced families were negatively associated with child self-reports (p = 0.045). A positive medical history correlates with FIM 2.0 (p = 0.025). Residence (p = 0.046), the occupation of caregivers (p = 0.014), disease severity (p = 0.024), and the only child (p = 0.029) exhibited statistically significant associations with the impairment of HRQoL. Impaired HRQoL scores shown by the PedsQL™4.0 parent proxy report (p = 0.013) and FIM 2.0 (p = 0.011) were associated with a reduced 5-year survival rate. Conclusions: This study demonstrated that early impairment of HRQoL in pediatric acute myeloid leukemia patients has predictive value for long-term prognosis. Once validated, these findings may provide some guidance to clinicians treating children with AML.

Neurodevelopmental cell communication plays a crucial role in neuroblastoma prognosis. However, determining the impact of these communication pathways on prognosis is challenging due to limited sample sizes and patchy clinical survival information of single cell RNA-seq data. To address this, we have developed the cell communication pathway prognostic model (CCPPM) in this study. CCPPM involves the identification of communication pathways through single-cell RNA-seq data, screening of prognosis-significant pathways using bulk RNA-seq data, conducting functional and attribute analysis of these pathways, and analyzing the post-effects of communication within these pathways. By employing the CCPPM, we have identified ten communication pathways significantly influencing neuroblastoma, all related to axongenesis and neural projection development, especially the BMP7-(BMPR1B-ACVR2B) communication pathway was found to promote tumor cell migration by activating the transcription factor SMAD1 and regulating UNK and MYCBP2. Notably, BMP7 expression was higher in neuroblastoma samples with distant metastases. In summary, CCPPM offers a novel approach to studying the influence of cell communication pathways on disease prognosis and identified detrimental communication pathways related to neurodevelopment.

Abstract Combining π‐conjugated and non‐π‐conjugated groups is an important strategy for synthesizing new nonlinear optical (NLO) crystals. However, the second harmonic generation (SHG) response and optical anisotropy can be limited by improper spatial alignment of these functional groups in the crystal structure. In this work, it is revealed that non‐π‐conjugated [NH2SO3] group acts as both hydrogen bond donor and acceptor, effectively regulating the 2D planar structure formed by π‐conjugated [C4N3H6] groups. The resulting organic–inorganic hybrid crystal C4N3H6SO3NH2 exhibits a strong SHG response (2.5 × KDP), large optical anisotropy (0.233@546 nm), and blue‐violet and green fluorescence near 360 and 520 nm, respectively. This work expands the methodology for creating new NLO crystals through organic–inorganic hybridization, while also showcasing the potential of C4N3H6SO3NH2 as a multifunctional optical material.

BackgroundEarly neurological deterioration (END) is a frequent complication in patients with perforating artery territory infarction (PAI), leading to poorer outcomes. Therefore, we aimed to apply machine learning (ML) algorithms to predict the occurrence of END in PAI and investigate related risk factors.MethodsThis retrospective study analyzed a cohort of PAI patients, excluding those with severe stenosis of the parent artery. We included demographic characteristics, clinical features, laboratory data, and imaging variables. Recursive feature elimination with cross-validation (RFECV) was performed to identify critical features. Seven ML algorithms, namely logistic regression, random forest, adaptive boosting, gradient boosting decision tree, histogram-based gradient boosting, extreme gradient boosting, and category boosting, were developed to predict END in PAI patients using these critical features. We compared the accuracy of these models in predicting outcomes. Additionally, SHapley Additive exPlanations (SHAP) values were introduced to interpret the optimal model and assess the significance of input features.ResultsThe study enrolled 1,020 PAI patients with a mean age of 60.46 (range 49.11–71.81) years. Of these, 30.39% were women, and 129 (12.65%) experienced END. RFECV selected 13 critical features, including blood urea nitrogen (BUN), total cholesterol (TC), low-density-lipoprotein cholesterol (LDL-C), apolipoprotein B (apoB), atrial fibrillation, loading dual antiplatelet therapy (DAPT), single antiplatelet therapy (SAPT), argatroban, the basal ganglia, the thalamus, the posterior choroidal arteries, maximal axial infarct diameter (measured at < 15 mm), and stroke subtype. The gradient-boosting decision tree had the highest area under the curve (0.914) among the seven ML algorithms. The SHAP analysis identified apoB as the most significant variable for END.ConclusionOur results suggest that ML algorithms, especially the gradient-boosting decision tree, are effective in predicting the occurrence of END in PAI patients.

Schistosoma species are the causative agent of schistosomiasis and shows worldwide distribution. There is a great need to develop a sensitive diagnostic approach for controlling the disease. Previously, we identified large numbers of Extracellular Vesicle (EV) proteins from Schistosoma japonicum (S. japonicum), but rarely these proteins have been evaluated for their diagnostic potential. In the present study, we performed bioinformatic analyses of S. japonicum identified EV-associated proteins from the previous study and then identified Schistosoma-specific proteins with potentially secreted capability. Among them, we selected SJCHGC02838 protein, SJCHGC05593 protein, SJCHGC05668 protein and a hypothetical protein (SJHYP) to evaluate their diagnostic potential for detecting S. japonicum infection. First, we determined the expression of these four proteins at the transcript levels using qRT-PCR and revealed that all these genes showed higher expression in adult stage. Then, we cloned the full-length cDNA for each protein into a prokaryotic expression vector and successfully generated the recombinant proteins. Upon the purification of recombinant proteins, we developed an indirect ELISA method to evaluate the diagnostic potential of these purified recombinant proteins. The results showed high sensitivity for detecting Schistosoma infection. Additionally, these proteins also displayed a good potential for detecting Schistosoma infection, especially SJCHGC05668 protein at an early stage. The diagnostic potentials of these recombinant proteins were further evaluated by Western blot and comparatively analyzed by our previously developed cfDNA methods.

IntroductionBiliary Infection in patients is a common and important phenomenon resulting in severe complications and high morbidity, while the distributions and drug resistance profiles of biliary bacteria and related risk factors are dynamic. This study explored the characteristics of and risk factors for biliary infection to promote the rational use of antibiotics in clinically.MethodsBacterial identification and drug susceptibility testing were completed using the Vitek 2 Compact analysis system. The distribution and antibiotic-resistant characteristics of 3,490 strains of biliary bacteria in patients at Nankai Hospital from 2019 to 2021 were analyzed using Whonet 5.6 and SPSS 26.0 software. We then retrospectively analyzed the clinical data and risk factors associated with 2,340 strains of Gram-negative bacilli, which were divided into multidrug-resistant bacteria (1,508 cases) and non-multidrug-resistant bacteria (832 cases) by a multivariate Cox regression model.Results and discussionA total of 3,490 pathogenic bacterial strains were isolated from bile samples, including 2,340 (67.05%) Gram-negative strains, 1,029 (29.48%) Gram-positive strains, and 109 (4.56%) fungal strains. The top five pathogenic bacteria were Escherichia coli, Klebsiella pneumoniae, Enterococcus faecium, Enterococcus faecalis, and Pseudomonas aeruginosa. Performance-based payment mechanism is one of the key issues to ensure all stakeholders’ benefits in infrastructure Public-Private Partnership (PPP) projects. However, most existing research on performance-based payment with a fixed incentive coefficient can’t play a good incentive role. This study aims to the intrinsic mechanism between the performance appraisal score and the performance-based payment structure, so as to design the dynamic performance-based payment mechanism for infrastructure PPP projects. Firstly, the multi-objective optimal method is used to calculate the unit-payment. Second, principal-agent theory is used to construct the performance-based payment model with a changeable incentive coefficient. The findings of this study show that, the performance-based payment mechanism can effectively motivate participants to provide high-quality and efficient services, because their remuneration directly depends on their performance. When the outcome does not meet expectations, the amount paid can be adjusted accordingly, thus ensuring the maximum protection of public resources as well as the private sector’s profits. They serve a dual purpose, on one hand, they offer insights to rectify the shortcomings in the current unsatisfactory payment structure. On the other hand, the study provides a theoretical reference for the public sector to effectively incentivize the private sector in enhancing project performance.

Benzotriazoles (BTRs) are a class of benzoheterocyclic chemicals that are frequently used as metal-corrosive inhibitors, both in industry and daily use. However, the exposure effect information on BTRs remains relatively limited. In this study, an integrated metabolomic and transcriptomic approach was utilized to evaluate the effect of three BTRs, benzotriazole, 6-chloro-1-hydroxi-benzotriazole, and 1-hydroxy-benzotriazole, in the mouse liver with results showing disrupted basal metabolic processes and vitamin and cofactor metabolism after 28 days. The expression of several genes that are related to the inflammatory response and aryl hydrocarbon receptor pathways, such as Gstt2 and Arntl, was altered by the exposure to BTRs. Exposure to BTRs also affected metabolites and genes that are involved in the immune system and xenobiotic responses. The altered expression of several cytochrome P450 family genes reveal a potential detoxification mechanism in the mouse liver. Taken together, our findings provide new insights into the multilayer response of the mouse liver to BTRs exposure as well as a resource for further exploration of the molecular mechanisms by which the response occurs.

Cone beam computed tomography (CBCT) is widely employed in modern dentistry, and tooth segmentation constitutes an integral part of the digital workflow based on these imaging data. Previous methodologies rely heavily on manual segmentation and are time-consuming and labor-intensive in clinical practice. Recently, with advancements in computer vision technology, scholars have conducted in-depth research, proposing various fast and accurate tooth segmentation methods. In this review, we review 55 articles in this field and discuss the effectiveness, advantages, and disadvantages of each approach. In addition to simple classification and discussion, this review aims to reveal how tooth segmentation methods can be improved by the application and refinement of existing image segmentation algorithms to solve problems such as irregular morphology and fuzzy boundaries of teeth. It is assumed that with the optimization of these methods, manual operation will be reduced, and greater accuracy and robustness in tooth segmentation will be achieved. Finally, we highlight the challenges that still exist in this field and provide prospects for future directions.

Abstract Fast screening strategies that enable high‐throughput evaluation and identification of desired variants from diversified enzyme libraries are crucial to tailoring biocatalysts for the synthesis of D‐allulose, which is currently limited by the poor catalytic performance of ketose 3‐epimerases (KEases). Here, the study designs a minimally equipment‐dependent, high‐throughput, and growth‐coupled in vivo screening platform founded on a redesigned D‐allulose‐dependent biosensor system. The genetic elements modulating regulator PsiR expression levels undergo systematic optimization to improve the growth‐responsive dynamic range of the biosensor, which presents ≈30‐fold facilitated growth optical density with a high signal‐to‐noise ratio (1.52 to 0.05) toward D‐allulose concentrations from 0 to 100 mm. Structural analysis and evolutionary conservation analysis of Agrobacterium sp. SUL3 D‐allulose 3‐epimerase (ADAE) reveal a highly conserved catalytic active site and variable hydrophobic pocket, which together regulate substrate recognition. Structure‐guided rational design and directed evolution are implemented using the growth‐coupled in vivo screening platform to reprogram ADAE, in which a mutant M42 (P38N/V102A/Y201L/S207N/I251R) is identified with a 6.28‐fold enhancement of catalytic activity and significantly improved thermostability with a 2.5‐fold increase of the half‐life at 60 °C. The research demonstrates that biosensor‐assisted growth‐coupled evolutionary pressure combined with structure‐guided rational design provides a universal route for engineering KEases.

Xiong′an，as an important carrier of Beijing′s non-capital functions, is of great significance in optimizing and reconstructing the layout of innovative cyberspace of the Beijing-Tianjin-Hebei region. At present, Xiong′an is still in the construction stage, and the evolution of the regional innovation network is more objective and feasible to use cooperative patents as an index. Firstly, the Xiong′an—Beijing-Tianjin-Hebei innovation cooperation network was constructed based on patent cooperation data and the characteristics of Xiong′an nodes was analyzed. Secondly, the QAP method was used to explore the influencing factors of the Xiong′an—Beijing-Tianjin-Hebei innovation cooperation network. Finally, the extended PMC index model was used to analyze the science and technology innovation policy of Xiong′an. The results show that: 1) Xiong′an expands the cooperative city at a faster rate but less frequently, and gradually evolves to the central position, but the control ability of resources is not significantly improved. 2) Institutional proximity has a positive impact on the Xiong′an—Beijing-Tianjin-Hebei innovation cooperation network. 3) Xiong′an′s existing science and technology policies are mainly based on investment subsidies, and the cultivation of innovation and entrepreneurship platforms and the promotion of innovation output policies are insufficient.

Abstract The mechanism underlying acute kidney injury (AKI) and AKI-to-Chronic kidney disease (CKD) transition remains unclear, but mitochondrial dysfunction may be a key driving factor. Literature reports suggest that dual-specificity phosphatase 1 (DUSP1) plays a critical role in maintaining mitochondrial function and structural integrity. In this study, ischemic Acute Kidney Injury (AKI) and post-ischemic fibrosis models were established by clamping the renal pedicle with different reperfusion times. To investigate the role of DUSP1, constitutional Dusp1 knockout mice and tubular-specific Sting knockout mice were used. Mitochondrial damage was assessed through electron microscopy observation, measurements of mitochondrial membrane potential, mtDNA release, and BAX translocation. We found that Dusp1 expression was significantly upregulated in human transplant kidney tissue and mouse AKI tissue. Dusp1 gene deletion exacerbated acute ischemic injury, post-ischemic renal fibrosis, and tubular mitochondrial dysfunction in mice. Mechanistically, DUSP1 could directly bind to JNK, and DUSP1 deficiency could lead to aberrant phosphorylation of JNK and BAX mitochondria translocation. BAX translocation promoted mitochondrial DNA (mtDNA) leakage and activated the cGAS-STING pathway. Inhibition of JNK or BAX could inhibit mtDNA leakage. Furthermore, STING knockout or JNK inhibition could significantly mitigate the adverse effects of DUSP1 deficiency in ischemic AKI model. Collectively, our findings suggest that DUSP1 is a regulator for the protective response during AKI. DUSP1 protects against AKI by preventing BAX-induced mtDNA leakage and blocking excessive activation of the cGAS-STING signaling axis through JNK dephosphorylation.

Abstract Objectives Innate immunity plays a significant role in the pathogenesis of temporomandibular joint osteoarthritis (TMJOA), which is characterized by synovial inflammation and condylar cartilage degradation. We are urged to investigate the impact of Resatorvid, a preventative drug that inhibits Toll-like receptor 4 (TLR4), on experimental inflammatory TMJOA pathology. Methods An intra-articular injection of complete Freund’s adjuvant (CFA) was used to induce an experimental inflammatory mouse TMJOA model, and TLR4 expression was identified by immunofluorescent labeling. Intraperitoneal injections of Resatorvid were administered to CFA-induced TMJOA mice, and the pathology of TMJOA animals with and without Resatorvid treatment was examined by H&E, Safranin-O/Fast Green, and TRAP staining, as well as micro-CT, immunohistochemistry, and immunofluorescence. The impact of Resatorvid on chondrocyte pyroptosis and macrophage inflammation was further investigated using ATDC5 chondrocytes and RAW264.7 macrophages pretreated with relevant antagonists. Results CFA-induced TMJOA mice revealed remarkable synovial inflammation, together with a time course of cartilage degradation and bone destruction, with TLR4 elevated in the synovium and condylar cartilage. Prophylactic treatment with Resatorvid mitigated synovial inflammation, cartilage degeneration, and bone destruction in CFA-induced TMJOA mice and downregulated MyD88/NF-κB expression. Ex vivo studies demonstrated that Resatorvid treatment alleviated NOD-like receptor protein 3 (NLRP3)-mediated chondrocyte pyroptosis and degeneration and relieved macrophage inflammation by preventing reactive oxygen species (ROS) production through NLRP3 signaling. Conclusion Prophylactic treatment with Resatorvid alleviates TMJOA pathology by inhibiting chondrocyte pyroptosis and degeneration, as well as ROS-induced macrophage inflammation, through TLR4/MyD88/NF-κB/NLRP3.

This study focuses on the ecological and environmental safety of Ta-pieh Mountain. Drought episodes can lead to ecological problems such as vegetation damage. Therefore, quantifying the response of vegetation to drought is essential for ecological management. The study utilized normalized difference vegetation index (NDVI) and precipitation datasets from 1999 to 2019 to derive seasonal NDVI and standardized precipitation index (SPI) data. Using Theil-Sen median trend analysis and Mann-Kendall significance test analysis, we initially examined the characteristics of vegetation and drought for the 21-year time series. SPI is used to investigate and assess the occurrence and severity of drought in the research region. Then, the strength and variability of cropland, woodland, and grassland drought resistance in the Ta-pieh Mountains were discussed using the ratio of coefficient of variation (RCV). Finally, the cross-spectrum was used to calculate the vegetation lag time to drought. The study found that NDVI increased across all seasons, while SPI increased in spring and autumn and decreased in summer and winter. The spring drought had the most significant impact on vegetation. Cropland showed the highest improvement in drought tolerance and woodland showed the highest drought tolerance. The lagged response periods of cropland, woodlands, and grassland to drought were 1.62 months, 8.94 months, and 2.49 months, respectively. These findings provide a scientific basis for the management and preservation of the ecology of the Ta-pieh Mountains. Abstract In order to effectively use of phages as antimicrobial agents for controlling multidrug-resistant E. coli, it is important to understand phage biology. The isolation and research of novel bacteriophages are urgently needed for food safety and animal health. Phage is considered as a novel alternative antibacterial agents and a new way of prevent, control and treat pathogens. In this paper, we isolated and characterized a novel lytic phage from chicken samples, named vB_EscP_vE20 (in brief vE20). We identified vE20 as belonging to the Podoviridae family through morphological and phylogenetic analysis. It had lytic activities against 24 out of 51 different serotypes of clinical E. coli strains, interestingly it also can lyse Salmonella species, such as Salmonella Enteritidis (S. Enteritidis) and Samonella Typhimurium (S. Typhimurium). One step growth curve showed that the latent period and lysis period was 10 min and 60 min, respectively. The burst size of vE20 was about 60 PFU/cell. The phage vE20 survived in a wide range of temperatures (30–60 °C) and pH (3–11). The whole genome size of phage vE20 is 77,938 bps, which is double stranded DNA. There are 121 Open reading frame (ORFs) in total, and the GC-content is 42.17%, without virulence-associated, antibiotic and lysogeny related genes. Phage vE20 has high bactericidal activity in killing E. coli EXG20-1 in cultures containing 4 × 109 cfu/mL in several MOIs culture. The results revealed the promising potential of phage vE20 as attractive candidates for the control of E. coli infections. Graphical Abstract

Abstract Background 2-deoxy-2-[18F]fluoro-D-glucose positron emission tomography (18F-FDG PET) could help evaluate metabolic abnormalities by semi-quantitative measurement to identify autoimmune encephalitis (AE). Few studies have been conducted to analyze the prognostic factors of AE. The study aimed to explore the values of diagnosis and treatment evaluation by 18F-FDG PET and preliminarily discussed the potential value in predicting the prognosis of AE patients. Methods AE patients underwent 18F-FDG PET/CT and magnetic resonance imaging (MRI). There were two steps to analyse 18F-FDG PET imaging data. The first step was visual assessment. The second step was to analyse 18F-FDG PET parameters using Scenium software (Siemens Molecular Imaging Ltd). The mean standardized uptake value (SUVmean) and maximum standardized uptake value (SUVmax) of brain relative regional metabolism (BRRM) were quantified in the case and control groups according to the anatomical automatic labeling (AAL) partition. The main statistical method was the Kruskal–Wallis test. Finally, the simple linear regression method was used to analyse the relationships between 18F-FDG PET parameters and the modified Rankin Scale (mRS) scores before and after treatment. Results The results on 18F-FDG PET showed that visual assessment abnormalities were in the mesial temporal lobe (MTL) (70.8%), (mainly infringing on the hippocampus and amygdala), basal ganglia (62.5%), frontal lobes (37.5%), occipital lobes (29.2%), and parietal lobes (12.5%). The positive rate of abnormalities on 18F-FDG PET was more sensitive than that on MRI (95.5% vs 32.2%, p = 0.001). The number of lesions on PET was positively correlated with the mRS scores before and after treatment, and the correlation before treatment was more significant. Before treatment, the SUVmean of the left occipital lobe was the most remarkable (SUVmean, R2 = 0.082, p > 0.05) factor associated with the mRS score, and the correlation was negative. With regard to prognosis, the SUVmax of the MTL was the most notable (R2 = 0.1471, p > 0.05) factor associated with the mRS score after treatment, and the correlation was positive. Conclusions 18F-FDG PET could be more sensitive and informative than MRI in the early phases of AE. The common pattern of AE was high MTL metabolism on 18F-FDG PET, which was associated with hypometabolism of the occipital lobe, and the number of lesions on PET before treatment may be significant factors in assessing disease severity. The SUVmax of MTL hypermetabolism may serve as a prognostic biomarker in AE.

Public-private-partnership (PPP) model has been widely used in public services and infrastructure projects, and many relevant studies on PPP have emerged. Based on CiteSpace and VOS viewer, this study aims to comprehensively analyze the research status and development trend of the PPP model in China and US. Firstly, the essential statistical characteristics of authors, institutions, and publications are given based on the relevant data collection in Web of Science core collection database. Secondly, the research hotspots and evolution trends of PPP by scholars in China and US are discussed by carrying out keywords co-occurrence analysis and cluster analysis. Finally, from the literature analysis and keyword co-occurrence network over time, the research directions, such as payment risks for the government during the operation period, early termination mechanism and system compensation mechanism, government supervision, and related information, are summarized and forecast, to provide theoretical guidance for the future research on PPP.

In order to understand the radar climatology characteristics of convective activity over Anhui Province, the climatological distribution and influencing factors of convective activity during the warm season (May-September) were investigated using Doppler radar mosaic data from five Next Generation Weather Radars and ERA5 reanalysis data between 2015 and 2020. Results indicate that: (1) There are obvious inter-monthly variations and spatial differences of convective activity during the warm season. The convective activity peaks during the Meiyu period, with the highest frequency in June and July, followed by May and August, and the lowest in September. The region with the highest convective in July and July is mainly distributed in the southwest-northeast direction. (2) It is found that the diurnal variation of convective number is bimodally distributed in summer (June-August), with the main peak in the afternoon and a subpeak in the early morning. In June, the peak convective activity mainly occurs in the early morning, which is related to the strengthening of low-level southwest airflow from night to early morning, and thus enhancing the moisture flux convergence in Anhui. In July, the peak convective activity appears in the afternoon, with a subpeak in the early morning. Note that the main peak is stronger and the duration is longer, which is related to the enhancement of convective available potential energy in Anhui in July. The second peak in early morning is also caused by the strengthening of the low-level southwest airflow, which is similar to that in June, but the intensity is weaker. Convective activity in August exhibits a single peak in the afternoon with the most significant diurnal variation, which shows a typical afternoon thermal convection type.

This paper comprehensively analyses the first-ever monkeypox outbreak in Shenzhen, China, encompassing clinical symptomatology, therapeutic approaches, epidemiological research, and comprehensive laboratory tests, aiming to establish a robust reference for future monkeypox mitigation and management strategies. The investigation involved a thorough investigation of all identified positive cases, including extensive molecular analysis by nucleic acid detection and whole-genome sequencing of the monkeypox virus. Contact tracing and containment of the infected individuals were also undertaken. Three distinct monkeypox cases were identified in this unique outbreak, exhibiting mild and atypical clinical manifestations, primarily fever and rash. All cases were associated with a single transmission chain, primarily facilitated through close contact and homosexual behavior, indicative of a high-risk factor for monkeypox transmission.

Midknight Valencia Orange is one of the most important late maturing citrus varieties. In order to study the effect of different harvest maturity on the nutritional quality of this variety and to determine its optimal harvesting period, the quality indexes of Midknight Valencia Orange, including color difference, firmness, carotenoid, soluble sugars, organic acids, free amino acids, mineral elements, flavonoid and total phenol content were measured and a correlation analysis was performed. The result showed that different harvest maturity had significant effects on the quality of Midknight Valencia Orange. With the delay of harvesting time, the coloration process of the peel gradually changed from the stem end to the bottom end and the peel showed a bright yellow after 360 days of flowering. The contents of carotenoid, ascorbic acid, total phenol and flavonoid increased, while the content of soluble solid increased first and then decreased, and the titrable acid content decreased significantly. The main soluble sugars in Midknight Valencia Orange fruit were glucose, sucrose, fructose, talose and inositol, and the contents of glucose and sucrose showed an increasing trend during harvesting. The main organic acids were citric acid, malic acid and quinic acid, and the content of citric acid decreased during harvesting. The main free amino acids were proline, arginine and serine, which showed an increasing trend during harvesting. Based on the evaluation indexes prescribed above, 360~400 days (April~May) after flowering is the appropriate harvesting time for Midknight Valencia Orange. Besides, the correlation analysis results showed that, as the indicator often used to judge the maturity of fruit, the firmness and titratable acid content of Midknight Valencia Orange have highly correlation with the content of carotenoid, soluble sugars, organic acids, free amino acids, mineral elements, flavonoid and total phenol. The firmness, titratable acid content and TSSC/TA ratio of Midknight Valencia can be used for the determination of harvesting time, namely fruit with a firmness 1.33~1.03 kg, titratable acid content 1.77%~1.30% and 5.41~6.96 TSS/TA ratio showed a good quality. The results of this study can provide a scientific basis for the determination the suitable harvesting period of Midknight Valencia Orange.
