It has been reported that binding of STAT3 protein to the 5'-flanking region of the relaxin gene may result in downregulation of the relaxin expression. There is a Guanine(G)-rich segment located in about 3.8 Kb upstream of the relaxin gene and very close to the STAT3's binding site. In our study, NMR spectroscopy revealed the formation of G-quadruplex by this G-rich strand, and the result was confirmed by ESI mass spectrometry and CD spectroscopy. The theoretical structure of RLX G-quadruplex was constructed and refined by molecular modeling. When this relaxin G-quadruplex was stabilized by berberine(ΔTm = 10°C), a natural alkaloid from a Chinese herb, the gene expression could be up-regulated in a dose-dependent manner which was proved by luciferase assay. This result is different from the general G-quadruplex function that inhibiting the telomere replication or down-regulating many oncogenes expression. Therefore, our study reported a novel G-quadruplex in the relaxin gene and complemented the regulation mechanism about gene expression by G-quadruplexes.

Abstract The enzyme glutamine synthetase (GLN) is mainly responsible for the assimilation and reassimilation of nitrogen (N) in higher plants. Although the GLN gene has been identified in various plants, there is little information about the GLN family in cotton (Gossypium spp.). To elucidate the roles of GLN genes in cotton, we systematically investigated and characterized the GLN gene family across four cotton species (G. raimondii, G. arboreum, G. hirsutum, and G. barbadense). Our analysis encompassed analysis of members, gene structure, cis-element, intragenomic duplication, and exploration of collinear relationships. Gene duplication analysis indicated that segmental duplication was the primary driving force for the expansion of the GhGLN gene family. Transcriptomic and quantitative real-time reverse-transcription PCR (qRT-PCR) analyses indicated that the GhGLN1.1a gene is responsive to N induction treatment and several abiotic stresses. The results of virus-induced gene silencing revealed that the accumulation and N use efficiency (NUE) of cotton were affected by the inactivation of GhGLN1.1a. This study comprehensively analyzed the GhGLN genes in Gossypium spp., and provides a new perspective on the functional roles of GhGLN1.1a in regulating NUE in cotton.

Introduction: Defoliation by applying defoliants before machine picking is an important agricultural practice that enhances harvesting efficiency and leads to increased raw cotton purity. However, the fundamental characteristics of leaf abscission and the underlying genetic basis in cotton are not clearly understood. Objectives: In this study, we aimed to (1) reveal the phenotypic variations in cotton leaf abscission, (2) discover the whole-genome differentiation sweeps and genetic loci related to defoliation, (3) identify and verify the functions of key candidate genes associated with defoliation, and (4) explore the relationship between haplotype frequency of loci and environmental adaptability. Methods: Four defoliation-related traits of 383 re-sequenced Gossypium hirsutum accessions were investigated in four environments. The genome-wide association study (GWAS), linkage disequilibrium (LD) interval genotyping and functional identification were conducted. Finally, the haplotype variation related to environmental adaptability and defoliation traits was revealed. Results: Our findings revealed the fundamental phenotypic variations of defoliation traits in cotton. We showed that defoliant significantly increased the defoliation rate without incurring yield and fiber quality penalties. The strong correlations between defoliation traits and growth period traits were observed. A genome-wide association study of defoliation traits identified 174 significant SNPs. Two loci (RDR7 on A02 and RDR13 on A13) that significantly associated with the relative defoliation rate were described, and key candidate genes GhLRR and GhCYCD3;1, encoding a leucine-rich repeat (LRR) family protein and D3-type cell cyclin 1 protein respectively, were functional verified by expression pattern analysis and gene silencing. We found that combining of two favorable haplotypes (HapRDR7 and HapRDR13) improved sensitivity to defoliant. The favorable haplotype frequency generally increased in high latitudes in China, enabling adaptation to the local environment. Conclusion: Our findings lay an important foundation for the potentially broad application of leveraging key genetic loci in breeding machine-pickable cotton.

The application of autonomous navigation technology of electric crawler tractors is an important link in the development of intelligent greenhouses. Aiming at the characteristics of enclosed and narrow space and uneven ground potholes in greenhouse planting, to improve the intelligence level of greenhouse electric crawler tractors, this paper develops a navigation system of electric crawler tractors for the greenhouse planting environment based on LiDAR technology. The navigation hardware system consists of five modules: the information perception module, the control module, the communication module, the motion module, and the power module. The software system is composed of three layers: the application layer, the data processing layer, and the execution layer. The developed navigation system uses LiDAR, Inertial Measurement Unit (IMU) and wheel speed sensor to sense the greenhouse environment and the crawler tractor’s information, employs the Gmapping algorithm to build the greenhouse environment map, and utilizes the adaptive Monte Carlo positioning algorithm for positioning. The simulation test of different global path planning algorithms in Matlab shows that the A* algorithm obtains the optimal overall global path. In the scene of map 5, the path planned by the A* algorithm is the most significant, and the number of inflection points is reduced by 40.00% and 87.50%, respectively; meanwhile, the path length is the same as that of the Dijkstra algorithm, but the runtime is reduced by 68.87% and 81.49%, respectively; compared with the RRT algorithm, the path length is reduced by 7.27%. Therefore, the A* algorithm and the Dynamic Window Approach (DWA) method are used for tractor navigation and obstacle avoidance, which ensures global path optimality while also achieving effective local path planning for obstacle avoidance. The test results suggest that the maximum lateral deviation of the built map is 6 cm, and the maximum longitudinal deviation is 16 cm, which meets the requirement of map accuracy. Additionally, the results of the navigation accuracy test indicate that the maximum lateral deviation of navigation is less than 13 cm, the average lateral deviation is less than 7 cm, and the standard lateral deviation is less than 8 cm. The maximum heading deviation is less than 14°, the average heading deviation is less than 7°, and the standard deviation is less than 8°. These results show that the developed navigation system meets the navigation accuracy requirements of electric crawler tractors in the greenhouse environment.

Salinity stress can significantly impact productivity in agricultural area with limited water re-sources. Our study focused on how plants under salt stress respond to phosphorus availability in terms of growth and biochemical reactions in cotton genotypes. Two cotton genotypes with different P efficiencies (SK39 and JM21) were used in a hydroponic experiment with 300 mM NaCl and three P treatments (10, 20, and 30 mM). Salinity stress decreases root growth, shoot growth, biomass production, and chlorophyll content, according to the experimental findings. In treated plants, it also increased the levels of oxidative stress. However, this effect was alleviated by phosphorus therapy, which controlled the production of proline, total soluble sugars, and hydrogen peroxide (H2O2). Interestingly, salt-sensitive JM21 responded to phosphorus supplementation more favorably than salt-tolerant SK39. Our research emphasizes the critical role that phosphorus especially P20 plays increasing the salinity stress sensitivity of cotton plants and offers insightful in-formation on the mechanisms underlying the role of phosphorus in reducing salinity stress effects. This study also revealed interspecific variability in cotton genotypes and characteristics, primarily represented by attributes related to cotton growth and morphological indicators such as dry matter biomass.

Abstract Many different types of stem cells utilize asymmetric cell division (ACD) to produce two daughter cells with distinct fates. Haspin-catalyzed phosphorylation of histone H3 at Thr3 (H3T3ph) plays important roles during mitosis, including ACD in stem cells. However, whether and how Haspin functions in ACD regulation remains unclear. Here, we report that Haspin knockout (Haspin-KO) mouse embryonic stem cells (mESCs) had increased ratio of ACD, which cumulatively regulates cell fate decisions. Furthermore, Wnt5a is significantly downregulated due to decreased Pax2 in Haspin-KO mESCs. Wnt5a knockdown mESCs phenocopied Haspin-KO cells while overexpression of Wnt5a in Haspin-KO cells rescued disproportionated ACD. Collectively, Haspin is indispensable for mESCs to maintain a balanced ratio of ACD, which is essential for normal development and homeostasis.

Objective The present study was conducted to examine the gas production, fermentation characteristics, nutrient degradation, and methanogenic community composition of a rumen fluid culture with Broussonetia papyrifera (B. papyrifera) subjected to ensiling or steam explosion (SE) pretreatment. Methods Fresh B. papyrifera was collected and pretreated by ensiling or SE, which was then fermented with ruminal fluids as ensiled B. papyrifera group, steam-exploded B. papyrifera group, and untreated B. papyrifera group. The gas and methane production, fermentation characteristics, nutrient degradation, and methanogenic community were determined during the fermentation. Results Cumulative methane production was significantly improved with SE pretreatment compared with ensiled or untreated biomass accompanied with more volatile fatty acids production. After 72 h incubation, SE and ensiling pretreatments decreased the acid detergent fiber contents by 39.4% and 22.9%, and neutral detergent fiber contents by 10.6% and 47.2%, respectively. Changes of methanogenic diversity and abundance of methanogenic archaea corresponded to the variations in fermentation pattern and methane production. Conclusion Compared with ensiling pretreatment, SE can be a promising technique for the efficient utilization of B. papyrifera, which would contribute to sustainable livestock production systems.

Abstract CRISPR/RfxCas13d (CasRx) editing system can specifically and precisely cleave single-strand RNAs, which is a promising treatment for various disorders by downregulation of related gene expression. Here, we tested this RNA-editing approach on Beethoven (Bth) mice, an animal model for human DFNA36 due to a point mutation in Tmc1. We first screened 30 sgRNAs in cell cultures and found that CasRx with sgRNA3 reduced the Tmc1 Bth transcript by 90.8%, and the Tmc1 wild type transcript (Tmc1 + ) by 44.3%. We then injected a newly developed AAV vector (AAV-PHP.eB) based CasRx into the inner ears of neonatal Bth mice, and we found that Tmc1 Bth was reduced by 70.2% in 2 weeks with few off-target effects in the whole transcriptome. Consistently, we found improved hair cell survival, rescued hair bundle degeneration, and reduced mechanoelectrical transduction current. Importantly, the hearing performance, measured in both ABR and DPOAE thresholds, was improved significantly in all ages over 8 weeks. We, therefore, have validated the CRISPR/CasRx-based RNA editing strategy in treating autosomal-dominant hearing loss, paving way for its further application in many other hereditary diseases in hearing and beyond.

Abstract Background Nitrogen (N) is important for improving various morphological and physiological processes of cotton but their contribution to fiber quality is still lacking. Aims The current study aimed to explore the relationship between root morphology, subtending leaf physiology, and fiber quality of contrasting N-efficient cotton genotypes in response to N. Methods We analyzed the above parameters of CCRI 69 (N-efficient) and Xinluzao-30 (XLZ-30, N-inefficient) under control (2.5 mmol·L−1) and high N (5 mmol·L−1) conditions. Results The results showed that root morphological traits were increased in CCRI-69 under control conditions than high N. Subtending leaf morphology, chlorophyll and carotenoid contents, free amino acids, and soluble proteins were higher under high N as compared with the control. However, soluble sugars, fructose, sucrose contents, and sucrose phosphate synthase were higher under control conditions than high N across the growth stages. Irrespective of the N conditions, all morphological and physiological traits of cotton subtending leaf were higher in CCRI-69 than XLZ-30. Except for fiber uniformity, fiber quality traits like fiber length, strength, micronaire, and elongation were improved under control conditions than high N. Between the genotypes, CCRI-69 had significantly higher fiber length, strength, micronaire, and elongation as compared with XLZ-30. Strong positive correlations were found between root morphology, soluble sugars, sucrose content, and sucrose phosphate synthase activity with fiber quality traits, respectively. Conclusions These findings suggest that CCRI-69 performed better in terms of growth and fiber quality under relatively low N condition, which will help to reduce fertilizer use, the cost of production, and environmental pollution.

Acetylation and tri-methylation of histone H3 lysine 9 (H3K9ac and H3K9me3) play an interactive regulatory role in the epigenetic regulation of gene expression during heart development and cardiovascular disease, but little is known about their possible role in heart regeneration. Here we utilized genome-wide high-throughput RNA sequencing (RNA-seq) and chromatin immunoprecipitation with high-throughput sequencing (ChIP-seq) for H3K9ac and H3K9me3, carried out on regenerative cardiac tissues at different days post amputation in zebrafish (Danio rerio) to investigate dynamic changes in gene expression and the epigenetic landscape of H3K9ac and H3K9me3. The STAR, Bowtie2, MACS2, and deepTools2 were mainly used for RNA-Seq or ChIP-seq data analysis. In this article, we present detailed information on experiment design, data generation, quality assessment and processing pipeline. Raw reads of the RNA-seq and ChIP-seq data have been deposited at the NCBI GEO repository with the accession number GSE158104. Our data will be a valuable resource for the elucidation of H3K9ac and H3K9me3 involvement in the regulation of gene transcription during cardiac regeneration.

Phosphorus (P) is an essential macronutrient required for fundamental processes in plants. Trait plasticity is crucial for plant adaptation to environmental change. Variations in traits underlie diverse phosphorus (P) acquisition strategies among plants. Nevertheless, how the intraspecific plasticity and integration of morphological traits contribute to Phosphorus-Use-Efficiency (PUE) in cotton is unknown. In this study, 25 morphological traits were evaluated in 384 cotton genotypes grown with low P (LP, 10μmol. L−1) and normal nutrition (CK, 500μmol. L−1) to assess the genetic variability of morphological traits and their relationship to phosphorus use efficiency. Results revealed a large genetic variation in mostly morphological traits under low P. Significant enhancement in root traits and phosphorus efficiency-related traits like PUE was observed at LP as compared to CK conditions. In response to low P availability, cotton genotypes showed large plasticity in shoot and total dry biomass, phosphorus, and nitrogen efficiency-related traits (i.e., phosphorus/nitrogen use efficiency, phosphorus/nitrogen uptake efficiency), and most root traits, but a limited response in root dry biomass, taproot length, root surface area, root volume, and SPAD value. In addition, significant correlations were observed between PUtE (phosphorus uptake efficiency), NUE (nitrogen use efficiency), TDB (total dry biomass), and RTD (root tissue density) with PUE under both P supply level and phosphorus stress index, which may be a key indicator for improving PUE under LP conditions. Most root traits are most affected by genotypes than nutrition level. Conserved PUE is more affected by the nutrition level than the genotype effect. Principal component analysis depicted the comprehensive indicators under two P supply conditions were mainly reflected in root-related traits and morphological indicators such as dry matter biomass. These results indicate that interspecific variations exist within these cotton genotypes and traits. Our study provides suggestions for future research to enhance the ability of the earth system model to predict how crops respond to environmental interference and provide target quality for cotton breeding in phosphorus-deficient areas.

Mepiquat chloride (MC) priming alleviates the effects of salt stress during seed germination in cotton (Gossypium hirsutum L.), but the mechanisms underlying its effects are unknown. We found that MC priming increases salt tolerance, as evidenced by marked increases in seed vigor and germination rates, and alleviated salt toxicity by reducing Cl− accumulation in germinating seeds. Consistently, electrophysiological experiments revealed that the seeds with MC priming displayed superior Cl− exclusion ability in the root apex. These beneficial effects of MC priming were abolished by the abscisic acid (ABA)-synthesis blocker fluridone under salt stress. MC priming induced an early response to acclimatization and stress, as indicated by rapidly increasing ABA content during initial exposure to salt stress. Transcriptome analyses revealed that MC priming induced an array of differentially expressed genes (DEGs) in germinating seeds. The most noticeable changes in germinating seeds were MC priming-induced increases in the expression of DEGs encoding components of ABA biosynthesis, ABA catabolism, and ABA signaling pathways under salt stress. MC priming also increased the expression of some DEGs encoding Cl− ion transporters (e.g. CCC, SLAC1/SLAH1/SLAH3, CLC, and ALMT9) in germinating seeds. These results indicate that MC priming-induced ABA contributes to Cl− homeostasis in tissues and acts as a positive regulator of salt tolerance via regulation of Cl− transporters (particularly CCC and SLAC1/SLAH1/SLAH3). Taken together, these findings shed light on the molecular mechanism underlying MC-mediated tolerance to salt stress during seed germination.

Phosphorus (P) is an essential macronutrient, and an important component of plant metabolism. However, little is known about the effects of low P availability on P absorption, the photosynthetic electron transport chain, and the antioxidant system in cotton. This study used cotton genotypes (sensitive FJA and DLNTDH and tolerant BX014 and LuYuan343) with contrasting low-P tolerance in a hydroponic experiment under 15 µM, 50 µM, and 500 μM P concentrations. The results showed that low P availability reduced plant development and leaf area, shoot length, and dry weight in FJA and DLNADH, compared to BX014 and LuYuan343. The low P availability decreased the gas-exchange parameters such as the net photosynthetic rate, transpiration rate, and stomatal conductance, and increased the intercellular CO2 concentration. Chlorophyll a fluorescence demonstrated that the leaves’ absorption and trapped-energy flux were largely steady. In contrast, considerable gains in absorption and trapped-energy flux per reaction center resulted from decreases in the electron transport per reaction center under low-P conditions. In addition, low P availability reduced the activities of antioxidant enzymes and increased the content of malondialdehyde in the cotton genotypes, especially in FJA and DLNTDH. Moreover, low P availability reduced the activity of PEPC and generated a decline in the content of ATP and NADPH. Our research can provide a theoretical physiological basis for the growth and tolerance of cotton under low-P conditions.

In a high-power microwave device, the intense electron beam causes a high-heat-flux on the collector’s inner surface which results in an instant temperature increase which eventually causes the output power to drop. The vapor chamber is one of the passive cooling devices effective for high-heat-flux heat dissipation. To study the heat dissipation capacity and the surface temperature control effect of the vapor chamber, a numerical heat transfer model of the vapor chamber is proposed based on the effective thermal conductivity method. The accuracy of the steady-state heat transfer analysis and the applicability of the transient simulation are verified by comparing with experimental data, and the heat transfer are calculated under the condition of periodic pulsed heat sources. Lastly, the transient performance of a vapor chamber relative to a copper heat spreader of the same external dimensions is investigated as a function of the wick effective thermal conductivity, pulsed heat flux, and pulse duration. The transient behavior of the vapor chambers are examined to find a performance threshold to determine if the performance is superior to that of a copper heat spreader. The present work provides a basis to understand the advantages and limitations of metal heat spreader in pulse mode operation of vapor chamber and plays a vital role in the design of improving transient performance.

Abstract Background Salt stress significantly inhibits the growth, development, and productivity of cotton because of osmotic, ionic, and oxidative stresses. Therefore, the screening and development of salt tolerant cotton cultivars is a key issue towards sustainable agriculture. This study subjected 11 upland cotton genotypes at the seedling growth stage to five different salt concentrations and evaluated their salt tolerance and reliable traits. Results Several morpho-physiological traits were measured after 10 days of salinity treatment and the salt tolerance performance varied significantly among the tested cotton genotypes. The optimal NaCl concentration for the evaluation of salt tolerance was 200 mmol·L− 1. Membership function value and salt tolerance index were used to identify the most consistent salt tolerance traits. Leaf relative water content and photosynthesis were identified as reliable indicators for salt tolerance at the seedling stage. All considered traits related to salt tolerance indices were significantly and positively correlated with each other except for malondialdehyde. Cluster heat map analysis based on the morpho-physiological salt tolerance-indices clearly discriminated the 11 cotton genotypes into three different salt tolerance clusters. Cluster I represented the salt-tolerant genotypes (Z9807, Z0228, and Z7526) whereas clusters II (Z0710, Z7514, Z1910, and Z7516) and III (Z0102, Z7780, Z9648, and Z9612) represented moderately salt-tolerant and salt-sensitive genotypes, respectively. Conclusions A hydroponic screening system was established. Leaf relative water content and photosynthesis were identified as two reliable traits that adequately represented the salt tolerance of cotton genotypes at the seedling growth stage. Furthermore, three salt-tolerant genotypes were identified, which might be used as genetic resources for the salt-tolerance breeding of cotton.

The NPF (NITRATE TRANSPORTER 1/PEPTIDE TRANSPORTER FAMILY) transports various substrates, including nitrogen (N), which is essential for plant growth and development. Although many NPF homologs have been identified in various plants, limited studies on these proteins have been reported in cotton. This study identified 75, 71, and 150 NPF genes in Gossypium arboreum, G. raimondii, and G. hirsutum, respectively, via genome-wide analyses. The phylogenetic tree indicated that cotton NPF genes are subdivided into eight subgroups, closely clustered with Arabidopsis orthologues. The chromosomal location, gene structure, motif compositions, and cis-elements have been displayed. Moreover, the collinearity analysis showed that whole-genome duplication event has played an important role in the expansion and diversification of the NPF gene family in cotton. According to the transcriptome and qRT-PCR analyses, several GhNPFs were induced by the nitrogen deficiency treatment. Additional functional experiments revealed that virus-induced silencing (VIGS) of the GhNPF6.14 gene affects the growth and N absorption and accumulation in cotton. Thus, this study lays the foundation for further functional characterization of NPF genes in cotton.

Silicon (Si) could alleviate the adverse effect of salinity in many crops, but the effect in cotton remains unclear. In this study, we evaluated the role of Si in regulating the salt stress tolerance of cotton by analyzing the induced morpho-physiological changes. A hydroponic experiment was conducted by using contrasting salt-tolerant cotton genotypes (sensitive Z0102; tolerant Z9807) and four treatments (CK, control; CKSi, 0.4 mM Si; NaCl, 150 mM NaCl; NaClSi, 150 mM NaCl+0.4 mM Si). The results showed that Si significantly enhanced the net photosynthesis rate and improved the growth of cotton seedling under salt stress in both salt-sensitive and salt-tolerant genotypes. Exogenous Si significantly reduced the accumulation of reactive oxygen species (ROS) and decreased the malondialdehyde (MDA) content in salt-stressed cotton. In addition, the application of Si up-regulated the expression of CAT1, SODCC and POD, and significantly enhanced the antioxidant enzymatic activities, such as catalase (CAT) and peroxidase (POD), of the salt-stressed cotton seedlings. Further, Si addition protected the integrity of the chloroplast ultrastructure, including key enzymes in photosynthesis such as ferredoxin-NADP reeducates (FNR), ATP synthase (Mg2+Ca2+-ATPase) and ribulose-1, 5-bisphosphate carboxylase/oxygenase (RubisCO), and the structure and function of the photosynthetic apparatus PSII from salt stress. Moreover, Si significantly increased the effective stomatal density and stomatal aperture in the salt-stressed cotton seedlings. Taken together, Si could likely ameliorate adverse effects of salt stress on cotton by improving the ROS scavenging ability and photosynthetic capacity.

Abstract Background CRISPR-Cas9 gene-editing technology has facilitated the generation of knockout mice, providing an alternative to cumbersome and time-consuming traditional embryonic stem cell-based methods. An earlier study reported up to 16% efficiency in generating conditional knockout (cKO or floxed) alleles by microinjection of 2 single guide RNAs (sgRNA) and 2 single-stranded oligonucleotides as donors (referred herein as “two-donor floxing” method). Results We re-evaluate the two-donor method from a consortium of 20 laboratories across the world. The dataset constitutes 56 genetic loci, 17,887 zygotes, and 1718 live-born mice, of which only 15 (0.87%) mice contain cKO alleles. We subject the dataset to statistical analyses and a machine learning algorithm, which reveals that none of the factors analyzed was predictive for the success of this method. We test some of the newer methods that use one-donor DNA on 18 loci for which the two-donor approach failed to produce cKO alleles. We find that the one-donor methods are 10- to 20-fold more efficient than the two-donor approach. Conclusion We propose that the two-donor method lacks efficiency because it relies on two simultaneous recombination events in cis, an outcome that is dwarfed by pervasive accompanying undesired editing events. The methods that use one-donor DNA are fairly efficient as they rely on only one recombination event, and the probability of correct insertion of the donor cassette without unanticipated mutational events is much higher. Therefore, one-donor methods offer higher efficiencies for the routine generation of cKO animal models.

The lack of fresh water in the world has become a growing concern. As an open-source incremental technology for water resources, desalination has become an important method to solve the global water crisis. Based on the inherent safety, versatility, modularity, and advantages of high-temperature gas-cooled reactors, the Saudi Arabia desalination project is the relying background. This paper proposes a complete solution for the high-temperature gas-cooled reactor power and water coproduction project by selecting a combination of process-proven multi-effect distillation (MED) and reverse osmosis (RO). In the scheme, a tertiary circuit is designed for the isolation of radioactive entities. An innovative comparative analysis of the engineering investment and production costs of different desalination technologies, such as MED and RO, and a comparison of the investment estimates of the “thermal” and “membrane” methods for the production of 10,000 tonnes of fresh water per day are performed. The feasibility and energy efficiency of the multi-effect distillation–reverse osmosis (MED-RO) scheme are presented, demonstrating the feasibility and practicality of the above approach.

Background. Arrhythmia is one of the causes of death in severe coronary artery disease patients who also suffered from cancer. Our research aims to compare the incidence of arrhythmia between severe coronary artery disease patient with and without new-onset tumor. Methodology. We enrolled 79 patients (December 2019–December 2020) with severe coronary artery disease in this study, and 40 of them were complicated with new-onset tumor. The details of all subjects were thoroughly obtained; the laboratory tests were implemented including creatinine before coronary angiography. The appraisal of the severity of coronary artery disease was applied by Gensini score. The cardiac inspection includes UCG, 12-lead ECG, and Holter monitor. Results. Among them, there were 40 patients in the experimental group and 39 patients in the control group. According to the agronomic requirements of garlic sowing, the garlic morphology is studied and a garlic seed metering mechanism with excellent seeding performance is designed. Based on this design, a new garlic seeding machine with an adjustable-size seeding device is developed to realize efficient single-seed metering and seeding of different varieties of garlic. Further, the design scheme of the garlic seeder prototype is established, with the key components of the garlic seeding being designed on the basis of the garlic seeding mechanism. To achieve garlic single-seed metering for different varieties of garlic, the optimal adjustment size of the garlic seed metering device is determined through discrete element simulation analysis. A field experiment confirms the effectiveness of applying the proposed garlic planter to field sowing in terms of the metrics of missing seed and multiple seed rates. The results of the discrete element simulation test reveal that an adjustment size of 40 mm yields the best single-seed metering performance. At an operating speed of 15–35 rpm, the metering device can achieve more than an 80% qualification rate of single-seed metering, with a unit speed of 0.628–1.465 m/s. Thus, the developed garlic seeding device meets the requirements of precision sowing in China and can effectively realize the mechanized planting of garlic.

Nitrogen (N) fertilizer plays a vital role in increasing cotton yield, but its excessive application leads to lower yield, lower nitrogen use efficiency (NUE), and environmental pollution. The main objective of this study was to find an effective method to enhance the NUE in cotton production. A two-year field experiment was conducted by using a split plot design with N rates (N0, 0 kg N ha−1; N1, 112.5 kg N ha−1; N2, 225 kg N ha−1, N3, 337.5 kg N ha−1), and cotton cultivars (CRI 69; ZZM 1017; ZZM GD89 and XLZ 30) to evaluate both their individual effect and their interactions on cotton yield and NUE. The results showed that the biomass and N accumulation of four cultivars increased with an increase in N rate, whereas the NUE decreased. Cotton yield increased first and then decreased for CRI 69 and ZZM 1017, while kept increasing from N0 to N3 for ZZM GD89 and XLZ 30. Compared with ZZM GD89 and XLZ 30, CRI 69, and ZZM 1017 showed higher yield, resulted from higher biomass, bolls per plant, and boll weight, especially under low N level. In addition, the CRI 69 and ZZM 1017 had stronger N absorption and transformation capabilities, and showed higher NUE than those of ZZM GD89 and XLZ 30 under the same N rate. The results indicated that CRI 69 and ZZM 1017 show advantages over ZZM GD89 and XLZ 30 in yield, and NUE, especially under low N rate.

Chemical defoliation is an essential agricultural practice in cotton production for mechanic harvesting. Thidiazuron (TDZ) is the active ingredient of the chemical defoliant used on cotton. So far, few studies havefocused on the method of identifying the sensitivity of cotton cultivars to TDZ. Therefore, a greenhouse soil culture experiment was performed by using two widely cultivatedupland cotton cultivars CRI 49 and CRI 12 treated with seven different concentrations (0, 100, 200, 300, 400, 500, and 1000 mg L−1) of TDZ at the seedling stage to establish a screening system. Principal component analysis and the membership function value (MFV) method was used to analyze the physiological and phenotypic characters, including abscission rate, amino acids content, net photosynthetic rate (Pn), etc. Finally, we developed a mathematical evaluation model, selected 100 mg L−1 TDZ as the optimal concentration and identified reliable characters net photosynthetic rate (Pn), stomatal conductance (Gs), and transpiration rate (Tr) to evaluate cotton leaf abscission sensitivity. These results also confirmed that CRI 12 was more sensitive to TDZ than CRI 49. This is the first time using a mathematical evaluation method to evaluate the cotton leaf abscission sensitivity triggered by TDZ at the seedling stage and the results were also confirmed in the field experiment. Furthermore, it will be valuable that MFV method is applied to stress sensitivity evaluation in other crop species under stress environment.

Increasing soil salinity suppresses both productivity and fiber quality of cotton, thus, an appropriate management approach needs to be developed to lessen the detrimental effect of salinity stress. This study assessed two cotton genotypes with different salt sensitivities to investigate the possible role of nitrogen supplementation at the seedling stage. Salt stress induced by sodium chloride (NaCl, 200 mmol·L−1) decreased the growth traits and dry mass production of both genotypes. Nitrogen supplementation increased the plant water status, photosynthetic pigment synthesis, and gas exchange attributes. Addition of nitrogen to the saline media significantly decreased the generation of lethal oxidative stress biomarkers such as hydrogen peroxide, lipid peroxidation, and electrolyte leakage ratio. The activity of the antioxidant defense system was upregulated in both saline and non-saline growth media as a result of nitrogen application. Furthermore, nitrogen supplementation enhanced the accumulation of osmolytes, such as soluble sugars, soluble proteins, and free amino acids. This established the beneficial role of nitrogen by retaining additional osmolality to uphold the relative water content and protect the photosynthetic apparatus, particularly in the salt-sensitive genotype. In summary, nitrogen application may represent a potential strategy to overcome the salinity-mediated impairment of cotton to some extent.

Chemical defoliation is an important part of cotton mechanical harvesting, which can effectively reduce the impurity content. Thidiazuron (TDZ) is the most used chemical defoliant on cotton. To better clarify the mechanism of TDZ promoting cotton leaf abscission, a greenhouse experiment was conducted on two cotton cultivars (CRI 12 and CRI 49) by using 100 mg L−1 TDZ at the eight-true-leaf stage. Results showed that TDZ significantly promoted the formation of leaf abscission zone and leaf abscission. Although the antioxidant enzyme activities were improved, the reactive oxygen species and malondialdehyde (MDA) contents of TDZ increased significantly compared with CK (water). The photosynthesis system was destroyed as net photosynthesis (Pn), transpiration rate (Tr), and stomatal conductance (Gs) decreased dramatically by TDZ. Furthermore, comparative RNA-seq analysis of the leaves showed that all of the photosynthetic related genes were downregulated and the oxidation-reduction process participated in leaf shedding caused by TDZ. Consequently, a hypothesis involving possible cross-talk between ROS metabolism and photosynthesis jointly regulating cotton leaf abscission is proposed. Our findings not only provide important insights into leaf shedding-associated changes induced by TDZ in cotton, but also highlight the possibility that the ROS and photosynthesis may play a critical role in the organ shedding process in other crops.

Cotton production is highly sensitive to nitrogen (N) fertilization, whose excessive use is responsible for human and environmental problems. Lowering N supply together with the selection of N-efficient genotypes, more able to uptake, utilize, and remobilize the available N, could be a challenge to maintain high cotton production sustainably. The current study aimed to explore the intraspecific variation among four cotton genotypes in response to various N supplies, in order to identify the most distinct N-efficient genotypes and their nitrogen use efficiency (NUE)-related traits in hydroponic culture. On the basis of shoot dry matter, CCRI-69 and XLZ-30 were identified as N-efficient and N-inefficient genotypes, respectively, and these results were confirmed by their contrasting N metabolism, uptake (NUpE), and utilization efficiency (NUtE). Overall, our results indicated the key role of shoot glutamine synthetase (GS) and root total soluble protein in NUtE. Conversely, tissue N concentration and N-metabolizing enzymes were considered as the key traits in conferring high NUpE. The remobilization of N from the shoot to roots by high shoot GS activity may be a strategy to enhance root total soluble protein, which improves root growth for N uptake and NUE. In future, multi-omics studies will be employed to focus on the key genes and pathways involved in N metabolism and their role in improving NUE.

Drought is one of the most important abiotic stresses and hampers many plant physiological processes under suboptimal nitrogen (N) concentration. Seedling tolerance to drought stress is very important for optimum growth and development, however, the enhancement of plant stress tolerance through N application in cotton is not fully understood. Therefore, this study investigates the role of high N concentration in enhancing drought stress tolerance in cotton. A hydroponic experiment supplying low (0.25 mM) and high (5 mM) N concentrations, followed by 150 g L−1 polyethylene glycol (PEG)-induced stress was conducted in a growth chamber. PEG-induced drought stress inhibited seedling growth, led to oxidative stress from excessive malondialdehyde (MDA) generation, and reduced N metabolism. High N concentrations alleviated oxidative damage and stomatal limitation by increasing antioxidant enzymatic activities, leaf relative water content, and photosynthesis in cotton seedlings under drought stress. The results revealed that the ameliorative effects of high N concentration may be ascribed to the enhancement of N metabolizing enzymes and an increase in the amounts of osmoprotectants like free amino acids and total soluble protein. The present data suggest that relatively high N concentrations may contribute to drought stress tolerance in cotton through N metabolism, antioxidant capacity, and osmotic adjustment.

Nitrogen (N) is the most important limiting factor for cotton production worldwide. Genotype-dependent ability to cope with N shortage has been only partially explored in cotton, and in this context, the comparison of molecular responses of cotton genotypes with different nitrogen use efficiency (NUE) is of particular interest to dissect the key molecular mechanisms underlying NUE. In this study, we employed Illumina RNA-Sequencing to determine the genotypic difference in transcriptome profile using two cotton genotypes differing in NUE (CCRI-69, N-efficient, and XLZ-30, N-inefficient) under N starvation and resupply treatments. The results showed that a large genetic variation existed in differentially expressed genes (DEGs) related to amino acid, carbon, and nitrogen metabolism between CCRI-69 and XLZ-30. Further analysis of metabolic changes in cotton genotypes under N resupply showed that nitrogen metabolism and aromatic amino acid metabolism pathways were mainly enriched in CCRI-69 by regulating carbon metabolism pathways such as starch and sucrose metabolism, glycolysis/gluconeogenesis, and pentose phosphate pathway. Additionally, we performed an expression network analysis of genes related to amino acid, carbon, and nitrogen metabolism. In total, 75 and 33 genes were identified as hub genes in shoots and roots of cotton genotypes, respectively. In summary, the identified hub genes may provide new insights into coordinating carbon and nitrogen metabolism and improving NUE in cotton.

A compact circularly polarized antenna operating at 915 MHz industrial, scientific, and medical (ISM) band for biomedical implantable applications is presented and experimentally measured. The proposed antenna can be miniaturized to a large extent with the compact size of 15 × 15 × 1.27 mm3 by means of loading patches to a ring-shaped microstrip patch antenna. An impedance bandwidth of 10.6% (865–962 MHz) for reflection coefficient less than −10 dB can be obtained. Meanwhile, the simulated 3 dB axial-ratio (AR) bandwidth reaches 14 MHz. Finally, the optimized design was fabricated and tested, and the measured results agree well with simulated results.

Low phosphorus (P) availability is a major constraint for cotton production. Consequently, P-efficient genotypes can improve productivity under conditions where the higher application of P is not economical. This study was conducted to characterize cotton genotypes for P-use efficiency under various P concentrations (0, 10, 20, 40, 80, and 500 μM KH2PO4). The results showed large genotypic variation in five selected traits, such as root dry weight, shoot dry weight, photosynthetic activity, P-utilization efficiency, and P-uptake efficiency. Based on these five selected traits, the genotypes were grouped into three main classes as efficient, moderate efficient, and inefficient genotypes as proposed by different researchers. Most of the genotypes behaved in a similar pattern under different P concentrations. Among the genotypes, Xinluzao-49 and Xinluzao-48 were considered as P efficient while CCRI-64 and Yumian-21 as inefficient genotypes. However, the rest of the genotypes were considered as moderately P efficient. The results prove that a large genetic potential exists in cotton genotypes for P-use efficiency, and the use of P-efficient genotypes for cultivation will reduce the application of phosphatic fertilizers. Furthermore, the use of P-efficient genotypes will improve cotton breeding activities and help in improving the environmental sustainability of cotton production.

In this study, we report the identification and functional characterization of three Brachypodium distachyon UEV genes. All three BdUev1s form heterodimers with BdUbc13s, which are capable of catalyzing Lys63-linked polyubiquitination in vitro. The three BdUEV1 genes are also able to functionally complement the budding yeast mms2 mutant defective in DNA-damage tolerance. BdUev1A differs from the other two BdUev1s in that it contains an 18-amino acid tail, which appears to compromise its function in yeast, as deletion of this tail restores full function. BdUev1A is located in the cytoplasm and plasma membrane but not in the nucleus, whereas BdUev1B, BdUev1C and the C-terminally-truncated BdUev1A are mainly found in the nucleus. These and the BdUEV1 gene expression analysis allow us to speculate that although all three BdUev1s function by promoting Lys63-linked polyubiquitination, BdUev1B and BdUev1C are involved in DNA-damage response and possibly other nuclear functions, while BdUev1A is required for non-nuclear function(s).

Bayesian network structure learning from data has been proved to be a NP-hard (Non-deterministic Polynomial-hard) problem. An effective method of improving the accuracy of Bayesian network structure is using experts’ knowledge instead of only using data. Some experts’ knowledge (named here explicit knowledge) can make the causal relationship between nodes in Bayesian Networks (BN) structure clear, while the others (named here vague knowledge) cannot. In the previous algorithms for BN structure learning, only the explicit knowledge was used, but the vague knowledge, which was ignored, is also valuable and often exists in the real world. Therefore we propose a new method of using more comprehensive experts’ knowledge based on hybrid structure learning algorithm, a kind of two-stage algorithm. Two types of experts’ knowledge are defined and incorporated into the hybrid algorithm. We formulate rules to generate better initial network structure and improve the scoring function. Furthermore, we take expert level difference and opinion conflict into account. Experimental results show that our proposed method can improve the structure learning performance.

This paper presents a novel high-gain planar dipole-array antenna for 2.45 GHz which consists of four planar dipole elements placed in two parallel lines. Phase retardation of each element is set by feeding network to form controllable radiation direction. The radiation pattern of the array is discussed according to Arrays Theorem. The measured −10 dB band is from 2.3 GHz to 2.57 GHz, and peak gain in this band is 7.5 dBi. The gain can even reach 10.5 dBi after installing additional ground. The proposed antenna has advantages of high gain, controllable direction, and planarity which are suitable for 2.45 GHz RFID base station.
