The Cosmic Ray Energetics And Mass for the International Space Station (ISS-CREAM) experiment successfully recorded the data for about 539 days from August 2017 to February 2019. In this talk, we report the measurement of the cosmic-ray proton energy spectrum from the ISS-CREAM experiment in the energy range of 2.5 TeV - 650 TeV. For the analysis, we used the silicon charge detector (SCD) placed at the top of the ISS-CREAM payload to identify the incoming cosmic-ray charge. The SCD is finely segmented to minimize charge misidentification due to backscatter effects. The four-layer SCD consists of 10,752 silicon pixels, each of which is 1.37×1.57×0.05 cm^3 in size. The calorimeter (CAL) consists of 20 layers of tungsten/scintillating fibers preceded by carbon targets. It provided cosmic-ray tracking, energy determination, and the high-energy trigger. The Top and Bottom Counting detectors (T/BCD) are above and below the CAL, respectively, and provided the low energy trigger. Each T/BCD is composed of an array of 20×20 photodiodes on plastic scintillators. The measured proton spectral index of 2.67±0.02 between 2.5 and 12.5 TeV is consistent with prior CREAM measurements. The spectrum softens above∼10 TeV consistent with the bump-like structure as reported by CREAM-I+III, DAMPE, and NUCLEON, but ISS-CREAM extends measurements to higher energies than those prior measurement

Background Boron nitride (BN) nanomaterials are promising in biomedical research owing to their large surface area, graphene-like structure, and chemical and thermal properties. However, the toxicological effects of BN on erythrocytes and blood coagulation remain uninvestigated. Objective The aims of our study were to synthesize glycol chitosan (GC)- and hyaluronic acid (HA)-coated BNs, and to investigate the effects of these BNs on human cancer cells, erythrocytes, and whole blood. Methods We prepared hemocompatible forms of BN coated with GC and HA, and evaluated them using cell uptake/viability tests, hemolysis analysis and FE-SEM, as well as through hemorheological evaluation methods such as RBC deformability and aggregation, and blood coagulation. Results GC/BN and HA/BN were both ∼200 nm, were successfully taken into cells, and emitted blue fluorescence. Both BNs were less toxic than bare BN, even at higher concentrations. The aggregation index of human red blood cells (RBCs) after 2 h incubation with BN, GC/BN, and HA/BN was greatly influenced, whereas RBC deformability did not dramatically change. Conclusions We found that GC/BN affected the intrinsic coagulation pathway, whereas both GC/BN and HA/BN affected the extrinsic pathway. Therefore, HA/BN is less detrimental to RBCs and blood coagulation dynamics than bare BN and GC/BN.

Poly(lactide-co-glycolide) (PLGA) is a biocompatible and biodegradable polymer that has been widely used in devices for tissue engineering and drug delivery applications. Gold nanoparticles (GNPs) have also been used as biomaterials and have been found to have a positive effect on bone formation. In this study, we synthesized thiol end-capped PLGA (PLGA-SH) and used it for binding GNPs. This PLGA was processed into a sheet form via electrospinning. GNPs with an approximate size of 30 nm were attached onto the PLGA-SH sheet surfaces (PLGA-GNPs). This membrane was characterized by thermogravimetric analysis, ultraviolet/visible spectrophotometry, field emission scanning electron microscopy, energy dispersive X-ray spectroscopy, X-ray photoelectron spectroscopy, and confocal laser scanning microscopy. Characterization results show that the GNPs are well attached on the PLGA-SH sheet and it is possible to control the GNPs load. Additionally, in-vitro results showed that PLGA-GNPs have good biocompatibility. They were also found to enhance osteogenic differentiation of human adipose derived stem cells. From these results, we have determined that the PLGA-GNP fibers can be useful as materials for bone regeneration and can also potentially serve as drug carriers.

We synthesized and characterized blue-color-emitting graphene quantum dots (BGQDs) which is better than that of other GQDs synthesized from organic molecules. We investigated the biocompatibility of BGQDs with cells and erythrocytes. Evaluation of epithelial carcinoma cancer cells indicated that most cells successfully took up BGQDs and toxicity testing showed no effect on cell viability. We also examined alterations in deformability and aggregation of human erythrocytes exposed to BGQDs at different concentrations and incubation times. These results suggest that BGQDs are biocompatible and do not cause cell membrane damage, and thus, are promising nanomaterials for various medical diagnoses and treatments.

We introduce a label-free biosensing cellulose strip sensor with surface-enhanced Raman spectroscopy (SERS)-encoded bimetallic core@shell nanoparticles. Bimetallic nanoparticles consisting of a synthesis of core Ag nanoparticles (AgNP) and a synthesis of shell gold nanoparticles (AuNPs) were fabricated on a cellulose substrate by two-stage successive ionic layer absorption and reaction (SILAR) techniques. The bimetallic nanoparticle-enhanced localized surface plasmon resonance (LSPR) effects were theoretically verified by computational calculations with finite element models of optimized bimetallic nanoparticles interacting with an incident laser source. Well-dispersed raspberry-like bimetallic nanoparticles with highly polycrystalline structure were confirmed through X-ray and electron analyses despite ionic reaction synthesis. Background Exenatide exerts cardioprotective effects by attenuating ischaemic reperfusion (IR) injury, possibly through activating the opening of mitochondrial ATP-sensitive potassium channels. We used atomic force microscopy (AFM) to investigate changes in mitochondrial morphology and properties in order to assess exenatide-mediated cardioprotection in IR injury. Methods We used an in vivo Sprague-Dawley rat IR model and ex vivo Langendorff injury model. In the left anterior descending artery (LAD) occlusion model, animals were randomly divided into three groups: sham-operated rats (Sham, n = 5), IR-injured rats treated with placebo (IR, n = 6), and IR-injured treated with exenatide (IR + EXE, n = 6). For the Langendorff model, rats were randomly divided into two groups: IR injury with placebo (IR, n = 4) and IR injury with exenatide (IR+EXE, n = 4). Morphological and mechanical changes of mitochondria were analysed by AFM. Results Exenatide pre-treatment improved cardiac function as evidenced by improvement in echocardiographic results. The ratio of infarct area (IA) to risk area (RA) was significantly reduced in exenatide-treated rats. According to AFM, IR significantly increased the area of isolated mitochondria, indicative of mitochondrial swelling. Treatment with exenatide reduced the mitochondrial area and ameliorated the adhesion force of mitochondrial surfaces. Conclusions Exenatide pre-treatment improves morphological and mechanical characteristics of mitochondria in response to IR injury in a rat model. These alterations in mitochondrial characteristics appear to play a cardioprotective role against IR injury.

International audience; Cosmic Ray Energetics And Mass for the International Space Station (ISS-CREAM) experi-ment is designed to study the composition and energy spectra of cosmic-ray particles from 10^12 to 10^15 eV. ISS-CREAM was launched and deployed to the ISS in August 2017. The ISS-CREAM payload employs a Silicon Charge Detector for charge measurements, Top and Bot-tom Counting Detector for electron-hadron separation and a low-energy trigger, a Boronated Scintillator Detector for additional electron-hadron separation, and a Calorimeter (CAL) for en-ergy measurements and a high-energy trigger. The CAL is constructed of 20 layers of tungsten plates interleaved with scintillating fiber ribbons read out by hybrid-photodiodes (HPDs) and densified carbon targets. Each CAL layer is made of 3.5 mm (1 X_0) thick tungsten plates alter-nating with fifty 0.5 mm thick and 1 cm wide scintillating fiber ribbons. Consecutive layers of fiber ribbons are installed orthogonal to each other. Energy deposition in the CAL determines the particle energy and provides tracking information to determine which segment(s) of the charge detectors to use for the charge measurement. Tracking for showers is accomplished by extrapolating each shower axis back to the charge detectors. The performance of the ISS-CREAM CAL during flight is presented.

International audience; The Cosmic Ray Energetics And Mass experiment for the International Space Station (ISS-CREAM) is designed and built to measure the elemental energy spectra of cosmic-ray particles (1 ≤ Z ≤ 26) and electrons. It measures the energy of incident cosmic rays from 10^12 to 10^15 eV. ISS-CREAM was launched and deployed to the ISS in August 2017. The Science Operations Center (SOC) at the University of Maryland has been operating the payload on the Interna-tional Space Station (ISS) in coordination with the Payload Operations Integration Center (POIC) at NASA’s Marshall Space Flight Center. The SOC has been responsible for sending commands to and receiving data from the Science Flight Computer (SFC) on board ISS-CREAM. The ISS-CREAM data taking program interfaces with the POIC using the Telescience Resources Kit through the Software Toolkit for Ethernet Lab-Like Architecture developed by the Boeing Company. The command uplink and data downlink have been through the Track-ing and Data Relay Satellite System. We present the ISS-CREAM flight operations including ISS communications, SFC performance, etc.

International audience; Cosmic Ray Energetics and Mass for the International Space Station (ISS-CREAM) is designed to directly measure the energy spectra of high-energy cosmic rays, encompassing proton to iron nuclei, over the energy range from 1012 to 1015 eV [1]. The capability to measure an extended energy range enables us to probe the origin and acceleration mechanisms of cosmic rays. The ISS-CREAM instrument is configured with the balloon-borne CREAM calorimeter (CAL) for energy measurements and four layers of a finely segmented Silicon Charge Detector (SCD) for charge measurements. In addition, two new compact detectors have been developed for electron/proton separation: Top and Bottom scintillator-based counting detectors (TCD/BCD) and a boronated scintillator detector (BSD). Simulations use the GEANT3 package [2] with the FLUKA hadronic model [3]. An isotropic event generator was developed for the ISS-CREAM geometry with particles incident from the upper hemisphere. We will present simulation results regarding ISS-CREAM performance, including trigger rates, energy resolution, energy response, tracking resolution, charge efficiency, etc.

To analyze and compare several commercially available acrylic intraocular lenses (IOLs) with particular regard to their clinical significance, we examined the physicochemical and surface properties of four currently available acrylic IOLs using static water contact angle, atomic force microscopy (AFM), Raman spectroscopy, and differential scanning calorimetry (DSC) measurements. The hydrophobic acrylic IOLs, ZA9003, and MA60BM, had contact angles ranging from 77.9° ± 0.65° to 84.4° ± 0.09°. The contact angles in the hydrophilic acrylic (970C) and heparin-surface-modified (HSM) hydrophilic acrylic IOLs (BioVue) were 61.8° ± 0.45° and 69.7° ± 0.76°, respectively. This study investigates why a silver nanoparticle (SNP)-induced surface-enhanced Raman scattering (SERS) paper chip fabricated at low successive ionic layer absorption and reaction (SILAR) cycles leads to a high SERS enhancement factor (7 × 10 8 ) with an inferior nanostructure and without generating a hot spot effect. The multi-layered structure of SNPs on cellulose fibers, verified by magnified scanning electron microscopy (SEM) and analyzed by a computational simulation method, was hypothesized as the reason. The pattern of simulated local electric field distribution with respect to the number of SILAR cycles showed good agreement with the experimental Raman intensity, regardless of the wavelength of the excitation laser sources. The simulated enhancement factor at the 785-nm excitation laser source (2.8 × 10 9 ) was 2.5 times greater than the experimental enhancement factor (1.1 × 10 9 ). A 532-nm excitation laser source exhibited the highest maximum local electric field intensity (1.9 × 10 11 ), particularly at the interparticle gap called a hot spot. The short wavelength led to a strong electric field intensity caused by strong electromagnetic coupling arising from the SNP-induced local surface plasmon resonance (LSPR) effects through high excitation energy. These findings suggest that our paper-based SILAR-fabricated SNP-induced LSPR model is valid for understanding SNP-induced LSPR effects.

The Er3+/Yb3+-codoped Na0.5Gd0.5MoO4 phosphors were prepared via a facile sol-gel method. Under 980 nm excitation, the characteristic emissions of Er3+ ions were observed in all the samples and the upconversion (UC) emission intensity increased gradually with the increment of Yb3+ ion concentration, achieving its optimum value when the doping concentration was 0.11 mol. Meanwhile, the UC mechanism for both green and red emissions in Er3+/Yb3+-codoped Na0.5Gd0.5MoO4 phosphors was a two-photon process. Furthermore, the fluorescence intensity ratio of two green UC emissions from the thermally-coupled energy levels (2H11/2, 4S3/2) of Er3+ ions was studied as a function of temperature in the range of 298–778 K. The maximum sensitivity was determined to be around 0.00856 K−1 at 590 K, which was independent of the UC emission intensity. The heating effect which was induced by the laser excitation was also investigated. It was found that the temperature of sample varied from 265.143 to 328.094 K with elevating the pump power from 334 to 1032 mW. These results demonstrated that the Er3+/Yb3+-codoped Na0.5Gd0.5MoO4 phosphors may have potential applications in optical temperature sensors and optical heaters.

This paper presents a low-power, high-performance current-feedback instrumentation amplifier (CFIA) for portable bio-potential sensing applications. Noise analysis is performed to assign an optimized current for the input stage of the amplifier. Analysis on selecting nested chopping frequencies is performed, further reducing 1/f noise and the residual offset. Enhanced power efficiency is achieved by sharing cascode branches and using a Class-AB output stage. Through these methods, a good balance between noise performance and other parameters such as output ripples and power consumption of the ripple reduction feedback loop (RRFL) is achieved. The amplifier is developed using a 1-poly 6-metal 0.18 μm CMOS process. Three gain stages with a gain-boosting input stage provide a low-frequency, open-loop gain >250 dB. When configured to a closed-loop gain of 60 dB, the amplifier achieves a noise voltage density of 18 $${\text{nV}}/\sqrt {{\text{H}}z}$$nV/Hz and a 1/f noise corner of 3 Hz. With a current of 75 μA and a supply voltage of 3.3 V, a CMRR of 110 dB and a PSRR of 120 dB are achieved, with an average input offset of about 6.5 μV. The amplifier achieves a state-of-art noise efficiency factor of 4.2. Practical application of the CFIA is demonstrated with an in vivo electrocardiogram detection.

The Pap smear is the primary screening tool for invasive cervical cancer resulting from a persistent infection with oncogenic human papillomavirus (HPV); however, there are the problems such as the inability to distinguish between HPV infection and cervical dysplasia and a low sensitivity remain. We present preliminary findings of a label-free method to detect and classify HPV infection and cervical dysplasia using human cervical fluids. Three experimental groups, defined as normal, HPV-positive, and cervical dysplasia, were evaluated through their Raman spectral patterns for noise-independence, high reproducibility, and uniformity. Clinical diagnosis was performed through liquid-based cervical cytology, HPV test, and cervical histologic examination. Healthy cervical fluids showed a strong Raman intensity at 877 cm-1 (symmetric C-C stretching), and at 963 cm-1 (phosphate), compared to a reference Raman peak at 1003 cm-1 (phenylalanine symmetric ring breath). The HPV-positive cervical fluids showed a strong intensity of a Raman peak at 1448 cm-1 corresponding to C-H deformation vibration mode and the highest similarity between the central and ring zones among the three groups. The cervical dysplasia fluids showed the presence of strong peaks compared to the control and HPV-positive groups. In addition, different Raman spectra were acquired according to HPV type. Therefore, all ranges of cervical fluid-induced Raman spectra could be used to detect the presence of cervical pre-cancer. Raman peak-gated assessment provides a label-free and nondestructive tool for the clinical diagnosis of HPV infection and cervical precancerous changes.

Diverse intrinsic and extrinsic mechanical factors have a strong influence on the regulation of stem cell fate. In this work, we examined recent literature on the effects of mechanical environments on stem cells, especially on differentiation of mesenchymal stem cells (MSCs). We provide a brief review of intrinsic mechanical properties of single MSC and examined the correlation between the intrinsic mechanical property of MSC and the differentiation ability. The effects of extrinsic mechanical factors relevant to the differentiation of MSCs were considered separately. The effect of nanostructure and elasticity of the matrix on the differentiation of MSCs were summarized. Finally, we consider how the extrinsic mechanical properties transfer to MSCs and then how the effects on the intrinsic mechanical properties affect stem cell differentiation.

Hydrogels are physically and chemically cross-linked, 3D, porous, hydrated molecular structures that mimic the native tissue microenvironment. Hydrogels fabricated with environmentally sensitive polymers can possess additional properties such as thermoresponsiveness. This property originates from its phase transition from gel to solution and vice versa with temperature variation. Phase transition mechanisms also depend on the interaction between the polymers and surrounding environment. Their thermoresponsiveness makes hydrogels attractive for drug delivery, gene therapy, cell culture, and tissue engineering approaches. The resemblance to living tissue of such thermoresponsive hydrogels produced by crosslinking of natural polymers opens up many biomedical application opportunities for human use. The most intensively studied natural thermoresponsive polymer is poly(N-isopropyl-acrylamide) (PNIPAM). This review focuses on the phase transition mechanism in thermoresponsive PNIPAM, the factors affecting the behavior of the gel, its specific properties, and, in particular, the role of PNIPAM’s phase transition in drug delivery, cell culture, and tissue engineering.

We demonstrated the apoptotic effect of bee venom (BV) on human MDA-MB-231 breast cancer cells using Raman spectroscopy and principal component analysis (PCA). Biochemical changes in cancer cells were monitored following BV treatment; the results for different concentrations and treatment durations differed markedly. Significantly decreased Raman vibrations for DNA and proteins were observed for cells treated with 3.0 µg/mL BV for 48 h compared with those of control cells. These results suggest denaturation and degradation of proteins and DNA fragmentation (all cell death-related processes). The Raman spectroscopy results agreed with those of atomic force microscopy and conventional biological tests such as viability, TUNEL, and western blot assays. Therefore, Raman spectroscopy, with PCA, provides a noninvasive, label-free tool for assessment of cellular changes on the anti-cancer effect of BV.

We demonstrate the fabrication and spot-on bioassay application of a low-cost, monometallic, surface-enhanced Raman scattering (SERS) paper platform using gold nanoparticle (AuNP)-loaded screen printing inks. The AuNPs were used to enhance the Raman intensity through a surface plasmon resonance (SPR)-driven optical property and sodium carboxymethylcellulose (CMC) was used as a viscous ink to create uniform distribution of the AuNPs on the paper substrate. To minimize the coffee ring effect and uniformly disperse the nanoparticles, the size of citrate-capped AuNPs, the CMC concentration, the volume ratio of CMC solution and AuNPs, and the printing cycles were optimized. Two printing cycles of optimized ink with a 7:1 mixture of 2-wt% CMC and AuNPs produced the strongest SERS effect and an enhancement factor of 1.8 × 104 with a SERS paper platform based on a Rhodamine B probe molecule. The platform exhibited high reproducibility, with less than 5% spot-to-spot variation in Raman intensity. Furthermore, the SERS spectra of normal and two virus-infected tear biofluids were comparable to the SERS spectral findings of gold-deposited SERS substrates. The enhanced SERS activity and high reproducibility of a low-cost, flexible, portable, and lightweight paper platform suggest the potential of point-of-care applications of biofluids in fields ranging from clinical analysis to industry.

We introduce a novel, facile, rapid, low-cost, highly reproducible, and power-free synthesizable fabrication method of paper-based silver nanoparticle (AgNP) immersed surface-enhanced Raman scattering (SERS) platform, known as the successive ionic layer absorption and reaction (SILAR) method. The rough and porous properties of the paper led to direct synthesis of AgNPs on the surface as well as in the paper due to capillary effects, resulting in improved plasmon coupling with interparticles and interlayers. The proposed SERS platform showed an enhancement factor of 1.1 × 10(9), high reproducibility (relative standard deviation of 4.2%), and 10(-12) M rhodamine B highly sensitive detection limit by optimizing the SILAR conditions including the concentration of the reactive solution (20/20 mM/mM AgNO3/NaBH4) and the number of SILAR cycles (six). The applicability of the SERS platform was evaluated using two samples including human cervical fluid for clinical diagnosis of human papillomavirus (HPV) infection, associated with cervical cancer, and a malachite green (MG) solution for fungicide and parasiticide in aquaculture, associated with human carcinogenesis. The AgNP-immersed SERS-functionalized platform using the SILAR technique allowed for high chemical structure sensitivity without additional tagging or chemical modification, making it a good alternative for early clinical diagnosis of HPV infection and detection of MG-activated human carcinogenesis.

Carbon nanotube (CNT) is a type of carbon allotrope with excellent physical and electrical properties, including high thermal conductivity, mechanical strength, and thermal stability. Therefore, applications of CNT have been considered for a variety of fields, including biosensors, molecular electronics, X-ray, and fuel cells. However, the application of CNT to biomedicine is limited because this material is cytotoxic and inhomogeneous. In particular, the irregularity in the structural properties of paste or bundle-type CNTs causes an uncontrolled modification in biomolecules. Therefore, using CNT as biosensors to obtain quantitative analyses is difficult. In this study, we developed a new method to perform end-selective functionalization of CNT in order to enable quantitative analysis for biomedical applications. The process was as follows: (1) etching the tip of vertically-aligned CNTs under optimum conditions, (2) oxidation of exposed CNTs, and (3) end-selective linkage of functionalized CNTs with biomolecules (dsDNA).

This study investigates a new paper-based 3D microfluidic analytical device for analyzing multiple biological fluids. A wax-printed and -impregnated device was operated using tip-pinch manipulation of the thumb and index fingers and applied the chemical reaction of a preloaded colorimetric indicator and biological solutions. Chemical sensing of protein and glucose concentrations was quantitatively analyzed by changes in the color intensity of the image taken from three image readout devices including scanner (Epson Perfection V700), microscope (USB-embedded handheld digital microscope), and smartphone (LG Optimus Vu). Paper-based 3D microfluidic analytic device with three image analyzers successfully quantified 1.5–75 μM protein concentrations and 0–900 mg/dL glucose concentrations. Paper-based 3D microfluidic device combined with the smartphone showed the performance in protein bioassay (1.5–75 μM) and glucose bioassay (0–50 mM) including clinically relevant ranges comparable to other devices. An origami-driven paper-based 3D microfluidic analytic is a useful platform with great potential for application in point-of-care diagnostics.

Estrogens and estrogen receptors (ESRs) have been implicated in the stimulation of aberrant prostate growth and the development of prostate diseases. The aim of the present study was to investigate four single nucleotide polymorphisms (SNPs) of the ESR2 gene in order to examine whether ESR2 is a susceptibility gene for benign prostatic hyperplasia (BPH). In order to evaluate whether an association exists between ESR2 and BPH risk, four polymorphisms [rs4986938 (intron), rs17766755 (intron), rs12435857 (intron) and rs1256049 (Val328Val)] of the ESR2 gene were genotyped by direct sequencing. A total of 94 patients with BPH and 79 control subjects were examined. SNPStats and Haploview version 4.2 we used for the genetic analysis. Multiple logistic regression models (codominant1, codominant2, dominant, recessive and log-additive) were produced in order to obtain the odds ratio, 95% confidence interval and P-value. Three SNPs (rs4986938, rs17766755 and rs12435857) showed significant associations with BPH (rs4986938, P=0.015 in log-additive model; rs17766755, P=0.033 in codominant1 model, P=0.019 in dominant model and P=0.020 in log-additive model; rs12435857, P=0.023 in dominant model and P=0.011 in log-additive model). The minor alleles of these SNPs increased the risk of BPH, and the AAC haplotype showed significant association with BPH (χ2=6.34, P=0.0118). These data suggest that the ESR2 gene may be associated with susceptibility to BPH.

Objectives This study investigated the effects of sliding on the ultrastructure of three representative esthetic superelastic 0.014 inch nickel-titanium (NiTi) archwires. Methods: Atomic force microscopy, scanning electron microscopy, and light microscopy were used to estimate the surface roughness of archwires and bracket systems. Energy-dispersive X-ray spectroscopy was used to estimate the molecular differences between coated and uncoated areas. A combination of four different types of 0.014 inch metallic wires and two different types of 0.022 inch × 0.028 inch conventional brackets were evaluated by in vitro sliding tests using a novel self-made tensile-strength tester with a miniature load cell and syringe pump. The NiTi wires included an uncoated NiTi archwire (CO group), epoxy resin-coated NiTi archwire (ER group), Teflon®-coated NiTi archwire (TF group), and Ag/biopolymer-coated NiTi archwire (AG group). The brackets included contained stainless steel (SS) and ceramic (CE) brackets. Results: Both ER and TF wire groups exhibited less surface roughness than CO wire groups. The aim of this study was to investigate and compare the relationship between the surface roughness and frictional resistance of four archwire and bracket combinations consisting of the 0.016-inch NiTi and 0.019 × 0.025-inch stainless steel archwires interacting clinically with two representative self-ligating brackets, active-type Clippy-C(®) ceramic self-ligating brackets, and passive-type Damon(®) stainless steel self-ligating brackets, using the lateral force microscopy technique. A 0.016-inch NiTi archwire interacting with passive-type Damon(®) stainless steel self-ligating brackets showed the smoothest surface roughness and the lowest frictional resistance compared to other combinations. The archwires interacting with passive-type Damon(®) stainless steel self-ligating brackets showed significantly lower surface roughness and frictional resistance than those interacting with active-type Clippy-C(®) ceramic self-ligating brackets. The frictional force in the in vivo archwire and bracket system increased with increasing surface roughness of the archwire. This positive correlation suggests that surface roughness can be used as an evaluating marker for estimating the efficiency of orthodontic treatment, rather than the direct measurement of frictional force.

There continues to be a paucity of data regarding the nanostructural changes of vocal fold (VF) collagen after injury. The aim of this study is to investigate the nanostructural and morphological changes in the rabbit VF lamina propria following acute injury using atomic force microscopy (AFM). Unilateral VF injury was performed on 9 New Zealand breeder rabbits. Sacrifice and laryngeal harvest were performed at three time points: 1 day, 3 days, and 7 days after injury. Histology and immunohistochemistry data were collected to confirm extracellular matrix (ECM) changes in rabbit VF. The progressive changes in thickness and D-spacing of VF collagen fibrils were investigated over a 7-day postinjury period using AFM. At post-injury day 1, a fibrin clot and inflammatory cell infiltration were observed at the injured VF. The inflammatory score at postinjury day 1 was highest in injured VF tissue, with a significant decrease at postinjury day 7. The immunoreactivity of inflammatory proteins (COX-2, TNF-α) was observed in VF up to day 7 after injury. AFM investigation showed clustered and disorganized collagen fibrils at the nanoscale resolution at post-injury day 7. Mitochondria play a central role in cell survival, and apoptotic cell death is associated with morphological changes in mitochondria. Quantification of the morphological and mechanical property changes in brain mitochondria is useful for evaluating the degree of ischemic injury and the neuroprotective effects of various drugs. This study was performed to investigate the changes in brain mitochondria in an 11-vessel occlusion ischemic model treated with magnesium sulfate (MgSO4), utilizing atomic force microscopy (AFM). Rats were randomly divided into three groups consisting of sham (n=6), global ischemia (GI, n=6), and MgSO4-treated global ischemia (MgSO4, n=6). The biophysical properties of brain mitochondria determined from AFM topographic images and adhesion force from force-distance measurements. The mean perimeter of ischemic mitochondria significantly increased to 2,396±541 nm (vs. 1,006±318 nm in control group, P0.001). The MgSO4 treatment during global ischemia reduced the perimeter of ischemic mitochondria (1,127±399 nm, P0.001). The other parameters including length, width and area were significantly different than the GI group. Besides, the adhesion force (23.2±3.9 nN) of isolated mitochondria from the MgSO4 group was close to normal levels (28.5±2.5 nN), compared with that of ischemic ones (17.7±3.3 nN, P0.001). To confirm the neuroprotective effects of MgSO4, we performed Nissl staining. This study suggested that quantitative analysis of mitochondrial changes utilizing AFM could be effective for evaluating neuronal injury and drug effects.

We report the design and fabrication of a carbon nanotube (CNT) based micro-resolution field emission mobile open type x-ray system for breast imaging. It can be used efficiently during the surgery of breast cancer removal to obtain accurate resection margin in partial resection of breast specimen. The obtained x-ray image of breast specimen with the proposed system shows the clear detection of micro-calcifications.

Objective. Angiotensin II (Ang II) plays a profibrotic role in the kidneys. Although many pathways of Ang II have been discovered, the morphological and mechanical aspects have not been well investigated. We observed the changes in tubular epithelial cells (TECs) after Ang II treatment with or without Ang II receptor blockers (ARBs) using atomic force microscopy (AFM). Methods. TECs were stimulated with Ang II with or without telmisartan, PD123319, and blebbistatin. AFM was performed to measure the cellular stiffness, cell volume, and cell surface roughness. Epithelial to mesenchymal transition markers were determined via immunocytochemistry. Results. After Ang II stimulation, cells transformed to a flattened and elongated mesenchymal morphology. Cell surface roughness and volume significantly increased in Ang II treated TECs. Ang II also induced an increase in phospho-myosin light chain and F-actin and a decrease in E-cadherin. Ang II coincubation with either telmisartan or blebbistatin attenuated these Ang II-induced changes. Conclusion. We report, for the first time, the use of AFM in directly observing the changes in TECs after Ang II treatment with or without ARBs. Simultaneously, we successfully measured the selective effect of PD123319 or blebbistatin. AFM could be a noninvasive evaluating strategy for cellular processes in TECs.

Skin aging is associated with changes in both the mechanical properties of the skin and extracellular matrix (ECM) components. In this study, we investigated the relationships between mechanical property and aging biomarkers in passaged human dermal fibroblasts (HDFs). The stiffness of HDFs from passages 5-20 was assessed by atomic force microscopy. The ECM components including collagen, elastin, and fibrillin-1 and that of signaling molecules (SIRTs) were determined from each passage of cells. The stiffness of HDFs increased linearly from passages 5-15 and then became saturated: the average stiffness was 0.356 N/m at passages 5 and 1.186 N/m at passages 15, respectively. Expression of all aging biomarkers, including pro-collagen I and VII, elastin, fibrillin-1, and SIRT1 and SIRT6, were down-regulated by passaging. In particular, a change in the level of procollagen Type I was significantly associated with early aging, while a change in the level of fibrillin-1 was associated with late aging. All biomarkers except elastin showed a strong correlation with the cellular stiffness of HDFs.
