Abstract

Phytolacca dodecandra (L' Herit), or 'Endod', is one of the widely known medicinal plants in Ethiopia. Berries of the endod have been used as a detergent for centuries. The present study was aimed to test the hepatoprotective effects of the plant against acetaminophen (APAP)-induced liver injury in rats. Mice of either sex were used for oral acute toxicity tests and APAP-induced lethality tests. Hepatoprotective experiments were done on male rats using 2 g/kg of APAP to induce liver damage. Liver enzymes, total bilirubin (TB), and lipid profile were determined. Liver tissues were also examined histopathologically to see a morphologic change in the control and experiment groups. The protective effect of the plant extract was also tested through sodium pentobarbital (SPB)-induced sleeping time. A significant increase in serum levels of liver enzymes, TB, low-density lipoprotein (LDL), and triglycerides (TGs) was seen from oral administration of 2 g/kg APAP. Total cholesterol (TC) and high-density lipoprotein (HDL) levels were decreased. Serum levels of all parameters were reversed to normal after administration of silymarin 100 mg/kg and, 100, 200, and 400 mg/kg doses of the extract. A significant dose-dependent hepatoprotective effect of Phytolacca dodecandra Methanol Root Extract (PDME) was seen in terms of LDL. Histopathological investigations and SPB-induced sleeping time confirmed the findings of biochemical analysis. Abstract

Çalışmamızda, sıçanlarda değişik heparin türevlerinin (heparin, enoksaparin ve bemiparin) akut ve kronik anti-inflamatuvar ve analjezik etkilerinin var olup olmadığını araştırdık.Çalışmamızda akut inflamasyon karragenin modeli, kronik inflamasyon koton-pellet granüloma testi, analjezik etki hot-plate ağrı modeli ile değerlendirildi. Akut inflamasyon ve ağrı çalışmalarında heparin (100-500-1000 Ü/kg), enoksaparin (100-200-400 Ü/kg) ve bemiparin (125-250-500 Ü/kg) dozlarında kullanıldı.Karragenin pençe ödemi modelinde, pençe hacimleri, iki ayak arasındaki ağırlık farkları ve histopatolojik olarak değerlendirildiğinde heparin (100-1000 Ü/kg) kontrol grubuna göre belirgin antiinflamatuvar etki göstermiştir (p0.05). Bemiparin ise sadece düşük dozda (125 Ü/kg) 2. ve 3.saatte inflamasyonu azaltmış (p0.05) ancak iki ayak arasındaki ağırlık farkı ve histopatolojik sonuçlar bunu desteklememiştir. Enoksaparin ise anlamlı antiinflamatuvar etki göstermemiştir. Koton-pellet granüloma testinde heparin (1000 Ü/kg) ve bemiparin (125 Ü/kg) belirgin antiproliferatif etkiler göstermiş. Hot-plate testinde, enoksaparin (100, 200 ve 400 Ü/kg) genel olarak tüm ölçümlerde anlamlı antinosiseptif etki gösterirken, heparin(tüm dozlarda) ve bemiparin (125 Ü/kg) ise sadece 30.dakikada belirgin antinosiseptif etki göstermişlerdir.Sonuç olarak, heparin ve bemiparin akut ve kronik antiinflamatuvar ve erken dönemde antinosiseptif etkiler göstermişlerdir. Akut antiinflamatuvar etkilerinin bazı nöromediyatörlerin (histamin, serotonin, kinin, prostaglandin vb, salınımı) ve aktive pıhtılaşma faktörlerinin inhibisyonuna bağlı olabileceğini, sergiledikleri kronik antiinflamatuvar etkilerinin ise bazı kronik inflamatuvar hastalıklar için olumlu bir özellik olduğunu düşünmekteyiz. Çalışmamızda, enoksaparinin sadece antinosiseptif etkiler göstermesi ise, bu etkilerinin farklı mekanizmalar üzerinden olabileceğini düşündürmüştür. In our study, we investigated whether different heparin derivatives (heparin, enoxaparin and bemiparin) have acute and chronic anti-inflammatory and analgesic effects in rats.In our study, acute inflammation in carrageenan model, chronic inflammation in cotton-pellet granuloma test and analgesic effect by hot-plate pain model were evaluated. In acute inflammation and pain studies, heparin (100-500-1000 U/kg), enoxaparin (100-200-400 U/kg) and bemiparin (125-250-500 U/kg) doses were used. In carrageenan induced paw edema model in rats, heparin (100-1000 Ü/kg) showed significant anti-inflammatory effect compared to the control group based on paw volumes, weight differences between the two feet and histopathologic evaluations. Bemiparin reduced inflammation at 2nd and 3rd h only at low doses (125 U / kg), but the weight difference between the two feet and histopathological results did not support this. Enoxaparin did not show significant anti-inflammatory effect. Heparin (1000 U/kg) and bemiparin (125 U / kg) showed significant antiproliferative effects in the coton-pellet granuloma test. In the hot-plate test, enoxaparin (100, 200 and 400 U / kg) showed significant antinociceptive effects in all measurements, whereas heparin (at all doses) and bemiparin (125 U / kg) showed significant antinociceptive effect only at the 30th minute.In conclusion, heparin and bemiparin showed acute and chronic antiinflammatory and early phase antinociceptive effects. We think that acute antiinflammatory effects may be due to the inhibition of some neuromediators (histamine, serotonin, prostaglandin, etc) and active coagulation factors, while their chronic antiinflammatory effects may be a favorable feature for some chronic inflammatory diseases. In our study, enoxaparin showed only antinociceptive effect and this may be through a different mechanisms. 88

14. The Investigation of Therapeutic Implications of Mast Cell Stabilizer Cromolyn Sodium on Cholestasis and Cholestatic Pruritus in Experimental Cholestasis.

Abstract

Background: Relevant studies have indicated that hepatic mast cells may have potential roles in the progression of cholestasis and cholestasis-induced itch. We aimed to compare the effects of cromolyn sodium and other medications on cholestatic pruritus, serum biochemistry, histamine, total bile acids, autotaxin, liver histopathology, and mast cell distribution in tissues in an experimental cholestasis model conducted by bile duct ligation., Methods: Rats received the determined treatment consecutively for 10 days in addition to bile duct ligation. On the 5th and 10th days of the experiment, the rats' itching behaviors were observed for 5 minutes. After 10 days, blood and tissue samples were taken., Results: Significant decreases in serum histamine and autotaxin levels, plasma total bile acids, total bilirubin, and biliary enzymes were reported only in cromolyn sodium-treated rats compared to the control group. In immunohistochemistry of the liver samples, the peribiliary mast cells stained positive for autotaxin. Except for bile duct infarctus, all histopathological findings of cholestasis significantly improved only in cromolyn sodium-treated and sertraline-treated rats. The liver and peritoneal mast cells significantly decreased only in cromolyn sodium-treated rats compared to the control group. On the 10th day of the experiment, the mean duration of itching was significantly lower in all groups, except for naloxone- and ondansetron-treated rats., Conclusion: Cromolyn sodium has promising antipruritic efficacy and provides biochemical and histopathological recovery of the relevant parameters of cholestasis induced by bile duct ligation. For the first time in the literature, we showed that peribiliary mast cells can produce autotaxin, which is a very important pruritogenic signal in the setting of cholestasis.
