Abstract

A series of novel arylpyrid-3-ylmethanones (7a-aa) were designed as modulators of α7 nicotinic acetylcholine receptors (nAChRs). The methanones were found to be type I positive allosteric modulators (PAMs) of human α7 nAChRs expressed in Xenopus ooctyes. Structure-activity relationship (SAR) studies resulted in the identification of compound 7v as a potent and efficacious type I PAM with maximum modulation of a nicotine EC5 response of 1200% and EC50 = 0.18 μM. Compound 7z was active in reversing the effect of scopolamine in the novel object recognition (NOR) paradigm with a minimum effective ip dose of 1.0 mg/kg (2.7 μmol/kg). This effect was blocked by the selective α7 nAChR antagonist methyllycaconitine (MLA). These compounds are potent type I positive allosteric modulators of α7 nAChRs that may have therapeutic value in restoring impaired sensory gating and cognitive deficits in schizophrenia and Alzheimer's disease.

24. The effects of environmental contaminants on the immune functions of marine flatfish

Abstract

Given the widespread concern that chemical contaminants may be associated with infectious disease outbreaks in marine fish populations, work has been undertaken with the aim of developing a suite of non-specific and specific assays of marine fish immune function and the application of these techniques in a variety of field and laboratory investigations. Most of the work focused on dab, Limanda limanda (L.) in view of the importance of this species in several North Sea fish disease monitoring programmes, and was also supplemented with investigations of specific immune function in turbot, Scopthalmus maximus (L.). Initial field studies examined non-specific immune function in terms of lymphoid organ morphology in dab sampled along a North Sea gradient of chemical contamination during the March 1990 ICES/IOC Bremerhaven workshop. Significant differences were observed in the kidney and spleen cell populations from dab, and these observations were considered in view of the various other physico-chemical and biological results generated during the Bremerhaven workshop. Following the valuable experience gained of the practical aspects of the field monitoring approach, laboratory investigations were initiated with the aim of developing a suite of immune function assays for deployment in either laboratory or field studies of marine fish health. Assays for non-specific immune functions were considered, including serum protein and lysozyme levels, methods of phagocyte collection, phagocyte chemiluminescence, calorimetric detection of individual reactive oxygen species and in vitro cell migration assays. Additional field work was undertaken, with the monitoring of serum total protein levels and lysozyme activity in dab sampled from Lyme Bay, UK This study provided evidence of a marked seasonal variation in non-specific immune function, which appeared to be associated with environmental factors (e.g. water temperature) and the reproductive cycle. Selected non-specific assays were applied to dab and turbot exposed under controlled laboratory conditions to a variety of important marine contaminants, including cadmium, polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs), and the biological data integrated with appropriate chemical characterisation of the exposure matrix. Using cultured turbot as a test species, the non-specific assays were also supplemented with the assay of specific immune function in fish exposed to PCB contaminated sediments. In brief, there was evidence of significant impairment of immune function in fish exposed to either individual contaminants (viz. cadmium and PAHs) or contaminant mixtures (viz. PAHs and PCBs) under the laboratory conditions described. In summary, the project was successful in its primary aims of developing a suite of techniques for evaluating both cellular and humoral immune functions in marine flatfish, and applying these techniques in the laboratory to assess the impact of important classes of environmental contaminants on fish health. Selected techniques were also used in field monitoring studies of marine fish immune function, illustrating the potential of such techniques for use in future laboratory and field studies of fish health.

26. Case Study – When A Roof is More Than a Roof

Abstract

The repurposing of existing buildings is becoming more prevalent. But changing a buildings use is not without concern. A European Company purchased an existing warehouse located north of Chicago, Illinois, USA as the space filled their needs for the layout of their manufacturing equipment. Producing medical grade synthetic mesh requires interior temperatures of 90°F (32.2C) with a relative humidity of 90%. The roof of the facility was low-sloped trapezoidal standing seam metal roof system that was watertight in all types of precipitation: Rain, snow, ice. The insulation, below the roof panels and above the steel purlins, was vinyl face fiberglass batt laid over the structural steel bar joist and pinned by the metal roof panels. Shortly after manufacturing commenced, water build-up in the batt insulation during winter started and was so severe that the vinyl facing would belly downward and split the lap seams, which would release the water, soiling the 2m wide rolls of mesh that then did not meet medical standards and had to be discarded. This case study will review the issues that arose out of repurposing a warehouse to a manufacturing facility and what the solution to correct moisture related issues. Hutchinson Design Group, Ltd. of Barrington, Illinois, was retained to investigate the conditions, determine the causes and develop a corrective solution.

38. Methodological Developments to Monitor Process or Some Sources of Error in the Efficiency of Evaluations.

Abstract

Specific evaluation design is a non trivial problem which must be developed through a set of procedures involving considerable study of the enterprise to be evaluated. Three criteria to be considered when an evaluator provides data for a decision maker are: (1) the percentage of the evaluation data that are actually used by the decision maker; (2) the percentage of the decision maker's decisions that are made, in part, with the use of the evaluation data; and (3) the degree to which the data is utilized in the most important decisions, even where all decisions are not made using the evaluation data. These criteria have been named efficiency, completeness, and focus, respectively. Evaluators should avoid errors, and prefer operational to non operational techniques. Evaluator/decision maker interaction patterns should be incorporated into the evaluation design. (Author/BJG)

43. The Relationship Between Academic Disadvantagement and Socioeconomic Disadvantagement as Reported by Teachers on a National Sample Basis.

Abstract

The assumption is often made that economically and academically disadvantaged students constitute the same basic population within disadvantaged schools. This paper seeks to ascertain the relationship between these two target-group sectors and to identify the overlap of academically disadvantaged pupils with the other five prevalent target groups: (1) potential dropouts; (2) emotionally or mentally handicapped students; (3) migrant pupils; (4) neglected and delinquent pupils; and, (5) physically handicapped pupils. The paper proposes to use a data bank from the national 1970 Evaluative Survey of Compensatory Education to entertain the following four questions concerning the overlap between academic and economic disadvantagement: (1) in what variables do children targeted as academically disadvantaged differ from the overall population in the eyes of their teachers? (2) in what variables do children targeted as socioeconomically disadvantaged differ from the overall population in the eyes of their teachers? (3) what is the overlap of these two categories? Are academic disadvantagement and socioeconomic disadvantagement highly related variables? and, (4) in relationship to the target groups--academically disadvantaged pupils, socioeconomically disadvantaged--what variables interact differently for crosstabulation built upon these three categories and on the estimated national population? 44. The Contrast Between Title I and Non-Title I Schools.

Abstract

The problem addressed in this paper is to compare the schools identified as participating in Title I, 1965 Elementary Secondary Education Act programs, with those identified as not participating in Title I, with regard to percentage of pupils on welfare, number of target group pupils, percentage of pupils reading below grade level, percentage of pupils receiving free lunches, type and quality of a library and special classroom facilities, conditions of physical plant, and ratings of instructional materials. Since degrees of urbanism had previously been shown to be related to some of the variables in question, the schools were grouped across the urban location variable for each comparison. Data was derived from the 1970 Survey of Compensatory Education, which included over 58,000 schools from 800 districts chosen at random from the states participating in the Belmont system. Of those schools over 33,000 have been identified as having Title I programs and nearly 25,000 have been identified as non-Title I schools. The survey had been so administrated that these schools were weighted to project the national population of elementary schools in districts having enrollments greater than 300 pupils. The comparisons between the Title I schools and the non-Title I schools utilized crosstabulations built on each of the subgroups. (Author/JM)
