Background We reported in a previous study that photodynamic therapy (PDT) of Helicobacter pylori(H. pylori) could potentiate bactericidal effect by adding chitosan. As a next step, we compared the bactericidal effects of low molecular weight (LMW) combined with Photodynamic Therapy to high molecular weight (HMW) chitosan. Method To perform PDT to kill H. pylori, we used endoscopic light as light source, methylene blue (MB) as a photosensitizer and chitosan (310–375, 50–190 kDa). We evaluated bacterial removal rate and its membrane damage by ethidium bromide monoazide PCR method (EMA q-PCR). 8-oxo-2′-dexoyguanosine by ELISA was measured for oxidative stress. Results At a chitosan concentration of ≤0.05%, the killing effect did not differ between the two molecular weights, and 100% bacterial removal rate was observed at a light energy ≥ 6.23 mJ/cm2 powers under 0.02% MB. After 15 min irradiation, LMW chitosan with high concentration of MB (0.004%) showed highest killing effects, which were consistent with the results of EMA q-PCR but not with the level of 8-OHdG. Bactericidal effects of LMW chitosan plus PDT using 0.002 and 0.004% MB for 15 min irradiation were significantly higher than those using HMW chitosan plus PDT. Conclusion We found that PDT using methylene blue with LMW chitosan to kill H. pylori exerted greater bactericidal effects through bacterial membrane damage than PDT with HMW chitosan. These results suggest that it would be better to choose LMW chitosan to enhance the effect of PDT for clinical application, even at a very low concentration of PS.

The purposes of this study were to identify physicochemical properties and evaluate bioactive compound levels and antioxidant characteristics at 30 day intervals during the 90 days of fermentation of gochujang fortified with five different varieties of red pepper: Juktoma pepper (RP1), facing heaven pepper (RP2), Thai chili pepper (RP3), bird’s eye pepper (RP4), and red bell pepper (RP5). Physicochemicals properties, including reducing sugar, capsaicin, pH, β-carotene, and color parameters, of gochujang were evaluated. Antioxidant compounds of total polyphenols and total flavonoids were analyzed with antioxidant activities of DPPH and FRAP assays. The results showed that gochujangs (GRP1, GRP5) fortified with RP1, and RP5, had consistently higher values of reducing sugars, total polyphenols, and total flavonoids with antioxidant activities, but lower values of capsaicin, pH, β-carotene, and color parameters as compared to GRP2, GRP3, GRP4 during 90 days of fermentation. GRP5 especially had the highest reducing sugar, amino acid contents, total polyphenols, and total flavonoids with antioxidant activities and the lowest value of capsaicin during the 90 days of fermentation.

In this study we explore patterns of same-turn self-repair within the word, across ten typologically and areally diverse languages. We find universal processes emerging through language-specific resources, namely: recycling is used to delay a next item due, while replacement is used to replace an inappropriate item. For example, most of our languages with prefixes or proclitics recycle those elements to delay production of the root/host, while languages with suffixes tend not to recycle just suffixes without their roots/hosts, since that would not serve to delay the production of the root/host; rather, the whole word is recycled. Replacement of affixes and clitics is rare, regardless of position. We provide several possible explanations for these facts, all based on the nature of replacement.

Plant breeders have focused on improving plant architecture as an effective means to increase crop yield. Here, we identify the main-effect quantitative trait loci (QTLs) for plant shape-related traits in rice (Oryza sativa) and find candidate genes by applying whole genome re-sequencing of two parental cultivars using next-generation sequencing. To identify QTLs influencing plant shape, we analyzed six traits: plant height, tiller number, panicle diameter, panicle length, flag leaf length, and flag leaf width. We performed QTL analysis with 178 F7 recombinant in-bred lines (RILs) from a cross of japonica rice line ‘SNUSG1’ and indica rice line ‘Milyang23’. Using 131 molecular markers, including 28 insertion/deletion markers, we identified 11 main- and 16 minor-effect QTLs for the six traits with a threshold LOD value > 2.8. Our sequence analysis identified fifty-four candidate genes for the main-effect QTLs. By further comparison of coding sequences and meta-expression profiles between japonica and indica rice varieties, we finally chose 15 strong candidate genes for the 11 main-effect QTLs. Our study shows that the whole-genome sequence data substantially enhanced the efficiency of polymorphic marker development for QTL fine-mapping and the identification of possible candidate genes. This yields useful genetic resources for breeding high-yielding rice cultivars with improved plant architecture.

Observations of soothing strategies that daycare teachers used with infants and toddlers in 10 centres, revealed that distress episodes lasted the longest for the youngest babies (0–12 months). The youngest babies received more positive caregiver responses when distressed compared with babies 13–24 months or toddlers 25–36 months old. The frequency of distress cues did not differ by child gender. Recorded levels of distress were significantly lower for babies cared for by teachers who had high school education or less, and those teachers responded more rapidly to infant/toddler distress signals than did teachers with college-level education.

In Arabidopsis, EARLY FLOWERING3 (ELF3) has pivotal roles in controlling circadian rhythm and photoperiodic flowering. In addition, ELF3 negatively regulates leaf senescence by repressing the transcription of PHYTOCHROME-INTERACTING FACTOR4 (PIF4) and PHYTOCHROME-INTERACTING FACTOR5 (PIF5); elf3 mutants senesce earlier and ELF3-overexpressing (ELF3-OX) plants senesce later than wild type (WT). Here, we show that in contrast to Arabidopsis ELF3, which represses senescence, the rice homolog OsELF3.1 promotes leaf senescence; oself3.1 mutants showed delayed senescence and OsELF3.1-OX plants senesced earlier under both dark-induced and natural senescence conditions. Microarray analysis revealed that in the senescing leaves, a number of senescence-associated genes, phytohormone-related genes, and NAC and WRKY family genes (OsNAP, ONAC106, and OsWRKY42) were differentially expressed in oself3.1 mutants compared with WT. Interestingly, we found that Arabidopsis plants overexpressing OsELF3.1 show delayed leaf senescence, produce short petioles, and flower late in long days, just like Arabidopsis ELF3-OX plants. This demonstrates that the regulatory functions of ELF3 and OsELF3.1 are conserved between Arabidopsis and rice, but the downstream regulatory cascades have opposite effects.

Polysaccharide microspheres (PAMs) from acetylated pullulan were designed for the long-term delivery of peptide/protein drugs, as an alternative to a PLGA depot system. Three kinds of samples were obtained according to their different degrees of acetylation (0.8(PA1), 1.5(PA2), 2.3(PA3) acetyl groups in one glucose unit in pullulan), and then utilized to prepare a microsphere via a water-in-oil-in-water (W 1 /O/W 2 ) emulsion method. The mean particle size of PAMs was shown to be in a range between 35 and 110 μm, as determined by a particle size analyzer. In order to evaluate their potential as a depot for protein/peptide delivery, exenatide, a drug used for the treatment of type II diabetes, was employed. The encapsulation efficiency of exenatide in PAMs was 69.1%, 80.4%, and 90.3% in PAM 1, PAM 2, and PAM 3, respectively. Although the release of exenatide from the PLGA microspheres evidenced a fast and high-burst behavior, PAMs evidenced a sustained release profile for 21 days. After 16 days, the released peptide was found to have a molecular weight almost identical to that of native exenatide, indicating that the stability of the peptide in the PAMs was maintained. The tissue reaction evidenced by the PAM was characterized by minimal foreign body reaction and minimal configurations of immune cells such as neutrophils and macrophages, but that of the PLGA microspheres was characterized by relatively elevated inflammation. On the basis of these results, we have concluded that the PAM may provide new insights into the development of new protein/peptide depots in long-term delivery.

Silver nanoparticles were prepared by using polyvinyl pyrrolidone (PVP) as a stabilizer and gamma-irradiation. Transmission electron microscopy (TEM) results showed that both the amount and the molecular weight of PVP in the irradiated solution considerably affect the average size of the silver nanoparticles. The average size of the silver nanoparticles decreases with increasing the amount of PVP in the solution, but increases with increasing its molecular weight. Further, TEM showed that the silver nanoparticles become disassembled into smaller nanoparticles after dilution with distilled water and sonication. Since the processes of dilution and sonication are not expected to result in chemical reactions or to split the silver nanoparticles, we conclude that each silver nanoparticle prepared by [Formula: see text] -irradiation consists of several smaller nanoparticles surrounded by PVP. Thus, based on these observations, we propose a three-step mechanism for the growth of the silver nanoparticles under the conditions considered here. In the first step, the silver ions interact with PVP, then in the second step the silver ions that are exposed to gamma-irradiation are reduced to silver atoms; nearby silver atoms then aggregate at close range. These aggregates are the primary nanoparticles. Finally, these primary nanoparticles coalesce with other nearby primary nanoparticles or interact with PVP to form larger aggregates which are the secondary (final) nanoparticles.

The present study describes an easy and clean method that makes the pattern of silver colloids by means of microcontact printing. Hydrophilic silver colloids adsorbed on a hydrophobic poly(dimethyl siloxane) (PDMS) stamp are transferred directly to the self-assembled monolayer (SAM) of 4-mercaptopyridine or aminoethanethiol (AET) on an Au-coated silicon substrate. To adsorb silver colloids on the PDMS stamp successfully, adhesion layers such as poly(vinyl pyrrolidone) and stearic acid were pre-treated onto the PDMS stamp. Finally, we obtained a surface-enhanced Raman scattering (SERS) spectrum and image of rhodamine 6G (R6G) on the patterned silver colloids.

Amyloid precursor protein binding protein-1 (APP-BP1) binds to the carboxyl terminus of amyloid precursor protein and serves as a bipartite activation enzyme for the ubiquitin-like protein, NEDD8. Previously, it has been reported that APP-BP1 rescues the cell cycle S-M checkpoint defect in Ts41 hamster cells, that this rescue is dependent on the interaction of APP-BP1 with hUba3. The exogenous expression of APP-BP1 in neurons has been reported to cause DNA synthesis and apoptosis via a signaling pathway that is dependent on APP-BP1 binding to APP. These results suggest that APP-BP1 overexpression contributes to neurodegeneration. In the present study, we explored whether APP-BP1 expression was altered in the brains of Tg2576 mice, which is an animal model of Alzheimer's disease. APP-BP1 was found to be up-regulated in the hippocampus and cortex of 12 month-old Tg2576 mice compared to age-matched wild-type mice. In addition, APP-BP1 knockdown by siRNA treatment reduced cullin-1 neddylation in fetal neural stem cells, suggesting that APP-BP1 plays a role in cell cycle progression in the cells. Collectively, these results suggest that increased expression of APP-BP1, which has a role in cell cycle progression in neuronal cells, contributes to the pathogenesis of Alzheimer's disease.

In this study, mesenchymal stem cells (MSCs) embedded in biodegradable and water-swollen, elastic block copolymer scaffolds were assessed for MSC chondrogenesis. To determine the optimal conditions for chondrogenesis of the embedded rMSCs, transforming growth factor-beta 3 (TGF-beta 3) was physically conjugated with chondroitin sulfate (CS) and mixed into scaffolds, which were subsequently evaluated for the differentiation of transplanted rMSCs. In determination of CS-bound growth factors for chondrogenesis, scaffold mixed with rMSCs and TGF-beta 3 was then tested by growth factor release profiles, confocal laser microscopy, RT-PCR analysis, real time-QPCR, and histology. The results of several different analyses of the transplanted rMSCs embedded in the scaffolds showed that rMSCs coupled with a CS-bound TGF-beta 3 encapsulated scaffold evidenced superior cartilage tissue formation as measured by an assay of specific gene and protein expression. Moreover, the scaffold exhibited more rapid and more distinct morphology of differentiated rMSCs than was observed with other scaffolds, as determined by histology and immunochemical histology analysis. These results indicate that the elastic block copolymer scaffolds combined with a CS-bound TGF-beta 3 should prove very suitable matrix for cell-based cartilage tissue engineering.

In this study, out-of-plane flexural test was performed to analyze behavior properties for a beam specimen which imitated a structure with connection member between reinforced concrete and steel plate concrete part. Tie bars between a upper and a lower steel plate, and tie wide flange shapes between upper and lower ribs were designed to prevent the steel plate or th e ribs from breakaway in the connection of the specimen. As a result of the test, ductile failure behavior of the specimen and the functionality of the tie members were conformed as originally intended. Also, tension tests were performed to evaluate the desi gn appropriateness of two specimens produced to anchor and connect mechanically #14 bars. The two test results showed that the anchorage connection system behaves in elastic limit during the ma in bars yielded, and the integrity of the designed system was verified. Takju is a Korean alcoholic beverage made from rice, and is brewed with the yeast Saccharomyces cerevisiae. This study was conducted to evaluate the effects of exercise training and moderate Takju consumption on learning ability in 6-week old Sprague-Dawley male rats. The rats were treated with exercise and alcohol for 4 weeks in six separate groups as follows: non-exercised control (CC), exercised control (EC), non-exercised consuming ethanol (CA), exercised consuming ethanol (EA), non-exercised consuming Takju (CT), and exercised consuming Takju (ET). An AIN-93M diet was provided ad libitum. Exercise training was performed at a speed of 10 m/min for 15 minutes per day. Ethanol and Takju were administered daily for 6-7 hours to achieve an intake of about 10 ml after 12 hours of deprivation, and, thereafter, the animals were allowed free access to deionized water. A Y-shaped water maze was used from the third week to understand the effects of exercise and alcohol consumption on learning and memory. After sacrifice, brain acetylcholinesterase (AChE) activity was analyzed. Total caloric intake and body weight changes during the experiment were not significantly different among the groups. AChE activity was not significantly different among the groups. The number of errors for position reversal training in the maze was significantly smaller in the EA group than that in the CA and ET groups, and latency times were shorter in the EA group than those in the CC, EC, CT, and ET groups. The latency difference from the first to the fifth day was shortest in the ET group. The exercised groups showed more errors and latency than those of the non-exercised groups on the first day, but the data became equivalent from the second day. The results indicate that moderate exercise can increase memory and learning and that the combination of exercise and Takju ingestion may enhance learning ability.

Background: Methylmethacrylate monomer (MN) bone cement is commonly employed in orthopedic procedures, particularly total hip and knee replacement, to anchor prosthetic devices to bone. Numerous cardiopulmonary complications can occur just after injection of MN. And MN produces direct relaxation of vascular smooth muscle in vitro. The purpose of this study was to determine if MN could have relaxation effect in tracheal smooth muscle too. Methods: Each ring of rat trachea was suspended on wire supports in a bath with Tris Tyrode solution. Dose response curves of MN were recorded after contraction of tracheal ring with acethylcholine (Ach) 10－5 M or cabachol (Cch) 10－8 M. MN was administered in denuded tracheal rings and compared it`s effect with intact tracheal rings to see the effect of epithelium for contraction. And MN dose response curves were recorded after pretreatment of nitric oxide synthase inactivator (L-NAME), muscarinic receptor blocker (atropine), beta-adrenaline receptor blocker (propranolol), adenylyl cyclase inhibitor (SQ22536) respectively. The effects of MN on cellular Ca2+ and K+ migration in rat tracheal preparations were studied. Results: MN significantly inhibited acetylcholine or carbachol induced contractions of tracheal rings dose-dependently (P ＜ 0.05). This relaxation effect of MN was not recovered in denuded tracheal rings. And pretreatment with L-NAME, propranolol, atropine, SQ22536 or tetraethylammonium respectively did not recover the relaxation effect of MN. MN inhibited both intracellular calcium release and extracelluar calcium influx. Conclusions: The relaxation effects of MN on rat tracheal rings are not related with epithelium, nitric oxide, muscarinic, or beta-adrenergic receptor. Methylmethacrylate monomer inhibits both intracellular calcium release and extracelluar calcium influx. (Korean J Anesthesiol 2007; 52: 194~201)

Background: To reduce side effects (hyperlipidemia, pain on injection, etc.) of the present formation of propofol, many attempts to change the emulsifying agent for propofol have been tried. This study was designed to examine the poloxamer-407 as an emulsifying agent for propofol compared to soybean oil regarding histamine release and plasma lipid levels. Methods: Twelve Beagle dogs weighing 12-16 kg were randomly assigned to one of two groups according to the formulation of propofol. Group 1 received Diprivan propofol 1% (AstraZeneca Co. UK), and group 2 received poloxamer-407 formulated propofol by a continuous intravenous infusion at 30 mg/kg/h for 3 hours. Three, 6, 9 and 12 hours after discontinuing the propofol infusion, venous blood samples from the cranial tibial vein were analysed by an ELISA kit for the histamine level. Also, blood lipid levels were checked 3 hours after the infusion and blood propofol concentration were checked every hour during the infusion. Results: Group 2 showed significantly less histamine release than group 1 at 3, 6 and 9 hours after the infusion (P < 0.05). In the plasma lipid study, there was no difference in high-density lipoprotein (HDL) between the two groups, but triglyceride and cholesterol were significantly higher in group 2 (P < 0.05). There was no difference in propofol concentrations between the two groups.
