In this article, we present several organic synthetic way to synthesize a family of five polyaromatic molecules based on a cyclophane core. Our strategies revolves around palado-catalyzed substitution on a [2.2]paracyclophane (pCp) building block. Direct formation of a cyclophane was also employed for two molecules. The polyaromatic nature of the cyclophane library we synthetized made them good fluorophores candidate, we hence performed full photophysical characterization (Absorption, Emission, TCSPC) in different solvent as well as embed in polystyrene films. We evaluate how the cyclophane moiety influence their photo physical properties compared to their corresponding homologues without pCp core, demonstrating greater stoke shift and intramolecular exciplex behavior. The general behavior among cyclophanes was also compared and show solvent dependent properties as well as consistency of the photophysics between toluene and polystyrene matrix., (© 2024 The Authors. A new class of photoswitches and the corresponding elementary photoinduced reaction, the so-called Excited-State Cation Transfer (ESCT), are investigated. This reaction relies on an intramolecular photo-release/photo-complexation of cation: after irradiation, the cation is translocated from a complexation site 1 to a site 2 during the excited state lifetime. Our purpose is thus to develop a computational strategy based on Density Functional theory (DFT) and its time-dependent counterpart (TD-DFT) to improve the different properties of the ESCT photoswitches, namely (i) the ground state complexation constant K , (ii) the excited state complexation constant K *, (iii) the photoejection properties and (iv) the population of the triplet states from a singlet state via intersystem crossing to increase the lifetime of the excited state. In this work, we are interested in optimizing the ESCT properties of a betaine pyridinium chromophore substituted by a 15-aza-5-crown, that was previously shown to efficiently photoeject a Ca 2+ cation from the site 1 but no photo-recapture was observed in the site 2 [Aloïse et al. , Phys. Chem. Chem. Phys ., 2016, 22 , 15384]. To this purpose, we have investigated the impact of the modification of the site 1 on the ESCT properties by introducing different substituents (EDG groups, halogen atoms) at different positions. So far, promising systems have been identified but a simultaneous improvement of all the ESCT photoswitches properties has yet not been achieved.

Objectives: We aimed to evaluate (1) fathers' perceptions and care involvement for their very premature infants and their views of the hospitalization period based on parental reports and (2) their evolution over time., Methods: We used an online parental survey to assess answers from parents of very preterm infants who were successfully discharged from French neonatal units. We analysed answers from February 2014 to January 2019 to an anonymous internet-based survey from the GREEN committee of the French Neonatal Society. Responses were compared for period 1 (P1, 1998 to 2013) and period 2 (P2, 2014 to 2019)., Results: We analyzed 2,483 surveys, 124 (5%) from fathers and 2,359 (95%) from mothers. At birth, 1,845 (80%) fathers were present in the hospital, but only 879 (38%) were near the mother. The presence of fathers in the NICU increased from P1 to P2 (34.5% vs. 43.1%, p = 0.03). Nearly two thirds of fathers accompanied their infants during transfer to the NICU (1,204 fathers, 60.6%). Fathers and mothers had similar perceptions regarding relationships with caregivers and skin-to-skin contact with their infants. However, more fathers than mothers felt welcome in the NICU and in care involvement regarding requests for their wishes when they met their infant (79% vs. 60%, p = 0.02) and in the presentation of the NICU (91% vs. 76%; p = 0.03). Mothers and fathers significantly differed in the caring procedures they performed ( p = 0.01), procedures they did not perform but wanted to perform ( p < 0.001), and procedures they did not perform and did not want to perform ( p < 0.01)., Conclusion: Most fathers were present at the births of their very preterm infants, but fewer fathers were near the mother at this time. Less than two thirds of fathers accompanied their infants to the NICU. There should be further changes to better meet the specific needs of the fathers of infants requiring care in the NICU. In this study, we have undertaken the development of two fluorescent sensors based on calixarene compounds for the purpose of detecting cesium in water. By introducing the sulfonate functional groups, we have considerably improved the water solubility of sensors, enabling complete dissolution of products in aqueous media and direct analysis of polluted water samples. Through rigorous experiments, we have demonstrated that the complexation of Cs + ions with sensors 1 and 2 in water leads to a remarkable enhancement of fluorescence. This fluorescence enhancement serves as a reliable indication of cesium presence and allows for sensitive detection. To further advance the practical application of our sensors, we have successfully integrated calixarene sensors 1 and 2 into a microfluidic sensor chip. This integration has enabled real-time, on-line measurements and has resulted in the development of a portable detection device capable of detecting cesium ions in water samples at parts per billion (ppb) levels. This device holds great promise for environmental monitoring and assessment, providing a convenient and efficient solution for cesium detection. Our work represents a significant advancement in the field of cesium detection, displaying the efficacy of calixarene-based fluorescent sensors and their integration into microfluidic systems. The enhanced water solubility, fluorescence response, and portability of our detection device offers tremendous potential for applications in environmental monitoring, water quality assessment, and emergency response scenarios where rapid and accurate cesium detection is crucial.

CRISPR-Cas9 cleavage efficacy and accuracy are the main challenges gene editing faces, and they are particularly affected by the optimal formation of the ribonucleoprotein (RNP) complex. We used nano differential scanning fluorimetry, a label and immobilization-free assay, to demonstrate that an equimolar ratio of Cas9 and guide RNA (gRNA) is optimal for RNP complex formation. We almost achieved 50% of green fluorescent protein (GFP) to blue fluorescent protein (BFP) conversion using a biallelic homozygous GFP human induced pluripotent stem cell line, when 0.4 μM of Cas9, equimolar Cas9/gRNA ratio and 2 μM of single-stranded oligonucleotide, were used and showed that increasing Cas9/gRNA ratio did not further improve KI efficiency. Additionally, excess gRNA decreased point mutation KI efficiency in rat embryos and drastically increased the occurrence of on-target large deletions. These findings highlight the importance of CRISPR/Cas9 stoichiometric optimization to ensure efficient and accurate KI generation, which will be applicable to other in vitro as well as in vivo models., Competing Interests: YC and VC were genOway employees. Patients carrying autosomal dominant mutations in the histone/lysine acetyl transferases CBP or EP300 develop a neurodevelopmental disorder: Rubinstein-Taybi syndrome (RSTS). The biological pathways underlying these neurodevelopmental defects remain elusive. Here, we unravel the contribution of a stress-responsive pathway to RSTS. We characterize the structural and functional interaction between CBP/EP300 and heat-shock factor 2 (HSF2), a tuner of brain cortical development and major player in prenatal stress responses in the neocortex: CBP/EP300 acetylates HSF2, leading to the stabilization of the HSF2 protein. Consequently, RSTS patient-derived primary cells show decreased levels of HSF2 and HSF2-dependent alteration in their repertoire of molecular chaperones and stress response. Moreover, we unravel a CBP/EP300-HSF2-N-cadherin cascade that is also active in neurodevelopmental contexts, and show that its deregulation disturbs neuroepithelial integrity in 2D and 3D organoid models of cerebral development, generated from RSTS patient-derived iPSC cells, providing a molecular reading key for this complex pathology., (© 2022. Presynaptic glutamate replenishment is fundamental to brain function. In high activity regimes, such as epileptic episodes, this process is thought to rely on the glutamate-glutamine cycle between neurons and astrocytes. However the presence of an astroglial glutamine supply, as well as its functional relevance in vivo in the healthy brain remain controversial, partly due to a lack of tools that can directly examine glutamine transfer. Here, we generated a fluorescent probe that tracks glutamine in live cells, which provides direct visual evidence of an activity-dependent glutamine supply from astroglial networks to presynaptic structures under physiological conditions. This mobilization is mediated by connexin43, an astroglial protein with both gap-junction and hemichannel functions, and is essential for synaptic transmission and object recognition memory. Our findings uncover an indispensable recruitment of astroglial glutamine in physiological synaptic activity and memory via an unconventional pathway, thus providing an astrocyte basis for cognitive processes., (© 2022. Highly pure millimeter-sized MOF-5 single crystals were synthesized and characterized. Photoluminescence (PL) spectroscopy and time-correlated single photon counting (TCSPC) demonstrate a solvent-guest dependency of MOF-5 emission and its ligand-centred nature. These results allow measuring the true MOF-5 luminescence free of solvent at a wavelength of 355 nm, a significantly lower wavelength than previously published. MOF-5 emission was also evaluated with different solvents and various degrees of water intake, explaining previously published observations. Comparison between lifetimes shows the fluorophore stabilization within the frameworks and demonstrates the progressive influence of the Zn 4 O subunits on the fluorescence during hydration. Overall, this work highlights the necessity to obtain phase-pure material, especially when moisture sensitivity can play a role, before ascribing electronic transitions. Interleukin (IL)-15 plays an important role in the communication between immune cells. It delivers its signal through different modes involving three receptor chains: IL-15Rα, IL-2Rβ and IL-2Rγc. The combination of the different chains result in the formation of IL-15Rα/IL-2Rβ/γc trimeric or IL-2Rβ/γc dimeric receptors. In this study, we have investigated the role of the IL-15Rα chain in stabilizing the cytokine in the IL-2Rβ/γc dimeric receptor. By analyzing the key amino acid residues of IL-15 facing IL-2Rβ, we provide evidence of differential interfaces in the presence or in the absence of membrane-anchored IL-15Rα. Moreover, we found that the anchorage of IL-15Rα to the cell surface regardless its mode of presentation - i.e. cis or trans - is crucial for complete signaling. In this study the evolution in the efficiency of electrochemotherapy (reversible electroporation) with pulse number was assessed in vitro. Experiments were performed using 100 μs pulses at different electric field intensities and the chemotherapeutic agent bleomycin. Additionally, electrical impedance spectroscopy measurements were used as a different method to study in real time the changes produced on cells with pulse number during trains of consecutive pulses. Our results show that the relation between pulse number and the observed outcome is complex and difficult to fully characterize. This relation can display a highly linear behaviour up to a certain number of pulses and/or field intensity applied. However, the relation between the number of pulses and the observed outcome always evolves to a saturation or at least a reduction in the electric field effects that is displayed when either electric field intensity or pulse number are increased. An exponential model was found to best describe this relation within the range of experimental conditions considered. Electrical impedance measurements confirmed the results and gave a more precise quantification of this dependence. IL-15 is a cytokine playing a crucial role in the function of immune cells, including NK and CD8 T cells. In this study, we demonstrated that in vivo, in mice, IL-15-prestimulated NK cells were no longer able to respond to a second cycle of IL-15 stimulation. This was illustrated by defects in cell maturation, proliferation, and activation, seemingly linked to the environment surrounding NK cells but not related to the presence of CD4 regulatory T cells, TGF-β, or IL-10. Moreover, NK cells from immunodeficient mice could respond to two cycles of IL-15 stimulation, whereas an adoptive transfer of CD44 + CD8 + cells impaired their responsiveness to the second cycle. Conversely, in immunocompetent mice, NK cell responsiveness to a second IL-15 stimulation was restored by the depletion of CD8 + cells. The effects of pyroelectricity on cell membrane permeability had never been explored. Pyroelectricity consists in the generation of an electric field in the surface of some materials when a change in temperature is produced. In the present study, tourmaline microparticles, which are known to display pyroelectrical properties, were subjected to different changes in temperature upon exposure to cells in order to induce an electric field at their surface. Then, the changes in the permeability of the cell membrane to a cytotoxic agent (bleomycin) were assessed by a cloning efficacy test. An increase in the permeability of the cell membrane was only detected when tourmaline was subjected to a change in temperature. This suggests that the apparition of an induced pyroelectrical electric field on the material could actually be involved in the observed enhancement of the cell membrane permeability as a result of cell electropermeabilization., (Copyright © 2017 Elsevier B.V. Objective: We previously reported that the olfactory receptor OR51E2, overexpressed in LNCaP prostate cancer cells, promotes cell invasiveness upon stimulation of its agonist β-ionone, and this phenomenon increases metastatic spread. Furthermore, we showed that the induced cell invasiveness involves a PI3 kinase dependent signalling pathway. We report here the results of a new investigation to address whether gallein, a small inhibitor of G protein βγ subunit interaction with PI3 kinase, can inhibit β-ionone effects both in vitro and in vivo., Results: We demonstrate that gallein can inhibit the β-ionone-induced cell invasiveness in vitro, as well as the spread of metastases in vivo. LNCaP cell invasiveness, assessed using spheroid cultures in collagen gels in vitro, was increased by β-ionone and the effect was reversed by co-administration of gallein. LNCaP tumour cells, subcutaneously inoculated to immunodeficient mice, generated more metastases in vivo when β-ionone was applied through the skin. Furthermore, the intraperitoneal injection of gallein inhibited this increased metastasis spread. Our results thus support the role of OR51E2 in the β-ionone observed effects, and suggest that gallein could be a potential new agent in personalized medicine of the tumours expressing OR51E2.

The association of host-guest and coordination chemistry was used to develop a fluorescent molecular sensor. A calix[6]arene bearing three imidazole arms at the small rim and three quinoline fluorophores at the large rim was synthesized and characterized. A two-step coordination sequence was observed upon addition of Zn II . The first Zn II center binds the tris-imidazole small rim site, leading only to a small perturbation of the fluorescence. In contrast, a large bathochromic shift is observed upon binding of the second Zn II center at the large rim as a result of the direct interaction of Zn II with the quinoline fluorophores. The system acts as a selective receptor for primary amines. Host-guest adduct formation could be identified by a shift and enhancement of the fluorescence emission that is dependent on the length and shape of the primary amine. Interleukin 2 and IL-15 are two closely related cytokines, displaying important functions in the immune system. They share the heterodimeric CD122/CD132 receptor to deliver their signals within target cells. Their specificity of action is conferred by their α receptor chains, IL-2Rα and IL-15Rα. By combining an increased affinity for CD122 and an impaired recruitment of CD132, we have generated an original molecule named IL-2Rβ/γ (CD122/CD132) inhibitor (BiG), targeting the CD122/CD132 receptor. BiG efficiently inhibited IL-15- and IL-2-dependent functions of primary cells, including CD8 T and NK cells, in vitro and in vivo. We also report a differential dynamic of action of these cytokines by highlighting a major role played by the IL-2Rα receptor. Interestingly, due to the presence of IL-2Rα, BiG had no impact on IL-2-dependent regulatory T cell proliferation. A series of π conjugated naphthalimide derivatives having an imide group as an acceptor conjugated with a methoxy arylethynyl or a methoxyphenyl triazole as a donor were prepared by Sonogashira coupling or "click" chemistry. Their photophysical properties were investigated by steady state and time resolved fluorescence spectroscopy and modelled by TD-DFT calculations. Compound Naphth-AlkyneOMe has a high fluorescence quantum yield and displays efficient photoinduced charge transfer in solution as well as in the powder state. Compound Naphth-TriazoleOMe exhibits a very high Stokes shift and its fluorescence quantum yield is low, which can be rationalized by theoretical calculations.

Olfactory receptors are G protein-coupled receptors. Some of them are expressed in tumor cells, such as the OR51E2 receptor overexpressed in LNCaP prostate cancer cells. It is considered a prostate tumor marker. We previously demonstrated that this receptor is able to promote LNCaP cell invasiveness in vitro upon stimulation with its odorant agonist β-ionone, leading to increased generation of metastases in vivo. In the present study, we show that even a relatively short exposure to β-ionone is sufficient to promote metastasis emergence. Moreover, α-ionone, considered an OR51E2 antagonist, in fact promotes prostate tumor growth in vivo. The combination of α-ionone with β-ionone triggers a higher increase in the total tumor burden than each molecule alone. To support the in vivo results, we demonstrate in vitro that α-ionone is a real agonist of OR51E2, mainly sustaining LNCaP cell growth, while β-ionone mainly promotes cell invasiveness. So, while structurally close, α-ionone and β-ionone appear to induce different cellular effects, both leading to increased tumor aggressiveness. This behaviour could be explained by a different coupling to downstream effectors, as it has been reported for the so-called biased ligands of other G protein-coupled receptors.

The development of highly selective sensors for potassium is of great interest in biology. Two new hydrosoluble potassium sensors (Calix-COU-Alkyne and Calix-COU-Am) based on a calix[4]arene bis(crown-6) and an extended coumarin were synthesized and characterized. The photophysical properties and complexation studies of these compounds have been investigated and show high molar extinction coefficients and high fluorescence quantum yields. Upon complexation with potassium in the millimolar concentration range, an increase of one- and two-photon fluorescence emission is detected. A twofold fluorescence enhancement is observed upon excitation at λ=405 nm. The ligands present excellent selectivity for potassium in the presence of various competitive cations in water and in a physiological medium. The photophysical properties are not affected by the presence of a large amount of competing cations (Na + , Ca 2+ , Mg 2+ , etc.). Ex vivo measurements on mouse hippocampal slices show that Calix-COU-Alkyne accumulates extracellularly and does not alter the neuronal activity. Using a combination of advanced DFT/TDDFT calculations together with ultrafast and stationary spectroscopies we have investigated the photochemistry and cationic complexation ability of 1-pyridinio-benzimidazolate (PyB) and analogs substituted by 15-aza-5-crown (PyB-Aza) or dimethyl-amino groups (PyB-DiMe). Focusing on PyB-Aza, the first aim was to assess the competitive complexation of the imidazole bridge vs. the macrocycle. In acetonitrile, it was found by absorption and emission that the imidazole moiety binds efficiently through lateral electrostatic interaction of high charge density cations and especially Ca(2+) to form a 1 : 1, metal : ligand (M : L) complex. Modulation of the complexation toward para substitution of the phenyl ring with a donor group is reported with values ranging from log K = 3.4 to 6.8. Complexation values are properly predicted by DFT calculations. From a photochemical point of view, for the same series, the trend is parallel to the rate of the photo-release process, found to be less than 200 femtosecond (fs), the fastest photorelease characteristic time reported so far. Unlike photoinduced charge transfer molecules linked with an aza-crown group, the mechanism appears simpler with no participation of loose complexes due to the macrocavity effect. Relaxation mechanisms after cation ejection are discussed as well. Finally, even if any photoinduced translocation of cation is reported for the PyB-Aza molecule between two complexation sites, a discussion about the use of betaine pyridinium as a molecular tool for the smart manipulation of cation systems is initiated.

The impact of external medium conductivity on the efficiency of the reversible permeabilisation caused by pulsed electric fields was investigated. Pulses of 12 ns, 102 ns or 100 μs were investigated. Whenever permeabilisation could be detected after the delivery of one single pulse, media of lower conductivity induced more efficient reversible permeabilisation and thus independently of the medium composition. Effect of medium conductivity can however be hidden by some saturation effects, for example when pulses are cumulated (use of trains of 8 pulses) or when the detection method is not sensitive enough. This explains the contradicting results that can be found in the literature. The new data are complementary to those of one of our previous study in which an opposite effect of the conductivity was highlighted. It stresses that the conductivity of the medium influences the reversible permeabilization by several ways. Moreover, these results clearly indicate that electropermeabilisation does not linearly depend on the energy delivered to the cells.

Electric pulses of a few nanoseconds in duration can induce reversible permeabilization of cell membrane and cell death. Whether these effects are caused by ionic or purely dielectric phenomena is still discussed. We address this question by studying the impact of conductivity of the pulsing buffer on the effect of pulses of 12 ns and 3.2 MV/m on the DC-3F mammalian cell line. When pulses were applied in a high-conductivity medium (1.5 S/m), cells experienced both reversible electropermeabilization and cell death. On the contrary, no effect was observed in the low-conductivity medium (0.1 S/m). Possible artifacts due to differences in viscosity, temperature increase or electrochemical reactions were excluded. The influence of conductivity reported here suggests that charges still play a role, even for 12-ns pulses. All theoretical models agree with this experimental observation, since all suggest that only high-conductivity medium can induce a transmembrane voltage high enough to induce pore creation, in turn. However, most models fail to describe why pulse accumulation is experimentally required to observe biological effects. They mostly show no increase of permeabilization with accumulation of pulses. In this study, electrical impedance spectroscopy measurements are performed during electroporation of monolayers of differentiated myotubes. The time resolution of the system (1 spectrum/ms) enable 860 full spectra (21 frequencies from 5 kHz to 1.3 MHz) to be acquired during the time gap between consecutive pulses (interpulse) of a classical electroporation treatment (8 pulses, 100 μs, 1 Hz). Additionally, the characteristics of the custom microelectrode assembly used allow the experiments to be performed directly in situ in standard 24 multi-well plates. The impedance response dynamics are studied for three different electric field intensities (400, 800 and 1200 V/cm). The multifrequency information, analysed with the Cole model, reveals a short-term impedance recovery after each pulse in accordance with the fast resealing of the cell membrane, and a long-term impedance decay over the complete treatment in accordance with an accumulated effect pulse after pulse. The analysis shows differences between the lowest electric field condition and the other two, suggesting that different mechanisms that may be related with the reversibility of the process are activated. As a result of the multifrequency information, the system is able to measure simultaneously the conductivity variations due to ion diffusion during electroporation. New fluorescent molecular sensors based on a calix[4]arene biscrown-6 ether as coordination site and BODIPY derivative as signaling unit were synthesized, and their photophysical properties were characterized. The complexation properties of these sensors with potassium and cesium cations were investigated using both steady-state and time-resolved fluorescence methods. The studies show that the sensitivity with cations depends upon the position of substituted coordination site on the BODIPY core. The complexation with cations does not have much effect on the absorption and emission wavelength when the coordination site (calix[4]arene biscrown-6 ether) is introduced at the meso position of the BODIPY core. In contrast, the same calix[4]arene biscrown-6 ether attached via a styryl linker to the α-position of BODIPY core leads to a sensitive sensor for alkali cations thanks to the better conjugation between the coordination site and the BODIPY core. The complexation of cations induces a hypsochromic shift of the absorption and emission maximums due to the diminution of donor character of the oxygen atoms in the coordination site. The stability constants of complexes with potassium and cesium ion were measured.

Aggregation-induced emission and aggregation-induced emission enhancement have attracted much attention due to their great potential in real-world applications. Up to now most of the reports are based on the restriction of free rotation of the luminogens in the aggregates. In the present work, we found that the dansyl fluorophore with typical intramolecular charge transfer characteristic also exhibited aggregation-induced emission enhancement, which was based on the change of micro-environmental polarity of the fluorophore. In the light of the phenomenon, a new water-soluble ligand bearing a tetrasulfonated calix[4]arene was constructed for ratiometric detection of Al(3+) based on an aggregation-induced emission enhancement mechanism. It displayed a distinct selectivity to Al(3+) among the tested cations in lutidine buffer solution (pH 6-7) with a detection limit of 1.8 μM. A reversible response was also demonstrated by the addition of EDTA or F(-).

We report a novel nontoxic, high-yield, gene delivery system based on the synergistic use of nanosecond electric pulses (NPs) and nanomolar doses of the recently introduced CM18-Tat11 chimeric peptide (sequence of KWKLFKKIGAVLKVLTTGYGRKKRRQRRR, residues 1-7 of cecropin-A, 2-12 of melittin, and 47-57 of HIV-1 Tat protein). This combined use makes it possible to drastically reduce the required CM18-Tat11 concentration and confines stable nanopore formation to vesicle membranes followed by DNA release, while no detectable perturbation of the plasma membrane is observed. Two different experimental assays are exploited to quantitatively evaluate the details of NPs and CM18-Tat11 cooperation: (i) cytofluorimetric analysis of the integrity of synthetic 1,2-dioleoyl-sn-glycero-3-phosphocholine giant unilamellar vesicles exposed to CM18-Tat11 and NPs and (ii) the in vitro transfection efficiency of a green fluorescent protein-encoding plasmid conjugated to CM18-Tat11 in the presence of NPs. Data support a model in which NPs induce membrane perturbation in the form of transient pores on all cellular membranes, while the peptide stabilizes membrane defects selectively within endosomes. Interestingly, atomistic molecular dynamics simulations show that the latter activity can be specifically attributed to the CM18 module, while Tat11 remains essential for cargo binding and vector subcellular localization. We argue that this result represents a paradigmatic example that can open the way to other targeted delivery protocols.

Olfactory receptors (ORs) are expressed in the olfactory epithelium, where they detect odorants, but also in other tissues with additional functions. Some ORs are even overexpressed in tumor cells. In this study, we identified ORs expressed in enterochromaffin tumor cells by RT-PCR, showing that single cells can co-express several ORs. Some of the receptors identified were already reported in other tumors, but they are orphan (without known ligand), as it is the case for most of the hundreds of human ORs. Thus, genes coding for human ORs with known ligands were transfected into these cells, expressing functional heterologous ORs. The in vitro stimulation of these cells by the corresponding OR odorant agonists promoted cell invasion of collagen gels. Using LNCaP prostate cancer cells, the stimulation of the PSGR (Prostate Specific G protein-coupled Receptor), an endogenously overexpressed OR, by β-ionone, its odorant agonist, resulted in the same phenotypic change. We also showed the involvement of a PI3 kinase γ dependent signaling pathway in this promotion of tumor cell invasiveness triggered by OR stimulation. Finally, after subcutaneous inoculation of LNCaP cells into NSG immunodeficient mice, the in vivo stimulation of these cells by the PSGR agonist β-ionone significantly enhanced metastasis emergence and spreading.

A series of fluorescent sensor molecules based on a phosphane sulfide derivative that is soluble in an organoaqueous solvent were designed and synthesized. The structure of the fluorophore has been optimized in order to have the best compromise in terms of solubility and photophysical properties. The obtained properties are in full agreement with quantum chemical calculations. A fluorescent molecular sensor containing one polyoxoethylene group has been synthesized and an efficient quenching upon mercury complexation has been observed. Finally, this sensing molecule has been introduced in a microfluidic chip in which fluorescence detection has been integrated. An efficient fluorescence response was observed upon mercury addition.

In our study, we used bleomycin to evaluate the permeabilization caused by nanosecond duration electric pulses (nanopulses). Bleomycin is a non permeant molecule which can be used both as a sensitive and quantitative marker to evaluate cell electropermeabilization. Indeed, the penetration of as few as 500 molecules is sufficient to entail a major biological effect: cell death. We show that one single nanopulse with a duration of 10 ns and a field strength of 40 kV/cm is sufficient to allow the uptake of at least 500 molecules of bleomycin in 20% of the cells when the external bleomycin concentration is 3 μM. When the external bleomycin concentration is reduced by a 100 fold, the same levels of cytotoxicity require an increase of about 25 times in the number of pulses. These results are in favor of the fact that each nanopulse creates new pores or defects on the cell membrane even if most of these pores can reseal between two consecutive pulses. Results also suggest that the cell permeability observed with classical markers when a large number of pulses are delivered results from the large number of nanopores or defects of the cell membrane created by the train of nanopulses., (Copyright © 2011 Elsevier B.V. A series of rod-shaped and related three-branched push-pull derivatives containing phosphane oxide or phosphane sulfide (PO or PS)-as an electron-withdrawing group conjugated to electron-donating groups, such as amino or ether groups, with a conjugated rod consisting of arylene-vinylene or arylene-ethynylene building blocks-were prepared. These compounds were efficiently synthesized by a Grignard reaction followed by Sonogashira coupling. Their photophysical properties including absorption, emission, time-resolved fluorescence, and two-photon absorption (TPA) were investigated with special attention to structure-property relationships. These fluorophores show high fluorescence quantum yields and solvent-dependent experiments reveal that efficient intramolecular charge transfer occurs upon excitation, thereby leading to highly polar excited states, the polarity of which can be significantly enhanced by playing on the end groups and conjugated linker. Rod-shaped and related three-branched systems show similar fluorescence properties in agreement with excitation localization on one of the push-pull branches. By using stronger electron donors or replacing the arylene-ethynylene linkers with an arylene-vinylene one induces significant redshifts of both the low-energy one-photon absorption and TPA bands. Interestingly, a major enhancement in TPA responses is observed, whereas OPA intensities are only weakly affected. Similarly, phosphane oxide derivatives show similar OPA responses than the corresponding sulfides but their TPA responses are significantly larger. Finally, the electronic coupling between dipolar branches promoted by common PO or PS acceptor moieties induces either slight enhancement of the TPA responses or broadening of the TPA band in the near infrared (NIR) region. The synthesis and photophysical properties of a novel fluorescent sensor are described. The phosphorus-selenium moiety allowed a selective mercury salt complexation, followed by the formation of phosphane oxide, which leads to a turn-on of the fluorescence. The sensibility and selectivity toward mercury cations were evaluated (0.18 ppb) and found to be in complete adequation with the targeted level of the World Health Organization, which makes the dye an efficient dosimeter for mercury cations.

The early-response gene product IEX-1 (also known as IER3) was recently found to interact with the anti-apoptotic Bcl-2 family member, myeloid cell leukemia-1 (Mcl-1). In this study we show that this interaction specifically and timely controls the accumulation of Mcl-1 in the nucleus in response to DNA damage. The IEX-1 protein is rapidly induced by γ-irradiation, genotoxic agents or replication inhibitors, in a way dependent on ataxia telangiectasia mutated (ATM) activity and is necessary for Mcl-1 nuclear translocation. Conversely, IEX-1 protein proteasomal degradation triggers the return of Mcl-1 to the cytosol. IEX-1 and Mcl-1 are integral components of the DNA damage response. Loss of IEX-1 or Mcl-1 leads to genomic instability and increased sensitivity to genotoxic and replicative stresses. The two proteins cooperate to maintain Chk1 activation and G2 checkpoint arrest. Mcl-1 nuclear translocation may foster checkpoint and improve the tumor resistance to DNA damage-based cancer therapies. Deciphering the pathways involved in IEX-1 degradation should lead to the discovery of new therapeutic targets to increase sensitivity of tumor cells to chemotherapy.

The thermodynamics and kinetics of the complexation reaction between lead ions and the fluorescent sensor Calix-DANS4 are determined to optimize the geometry of the microreactor used for the flow-injection analysis of lead and to tune the working conditions of this microdevice. Under our experimental conditions (pH 3.2, low concentration of Calix-DANS4) the 1:1 Pb(2+)-Calix-DANS4 complex is predominantly formed with a high stability constant (log K(1:1)=6.82) and a slow second-order rate constant (k=9.4×10(4) L mol(-1) s(-1)). Due to this sluggish complexation reaction, the microchannel length must be longer than 130 mm and the flow rate lower than 0.25 mL h(-1) to have an almost complete reaction at the output of the microchannel and a high sensitivity for the heavy metal detection. After determination of the values of the reaction times in our different microdevices, it is possible to simulate the calibration curves for the fluorimetric detection of lead under different conditions. An original method is also presented to determine mixing times in microreactors.

Norbadione A (NbA) is a mushroom pigment, which is assumed to be involved in (137)Cs accumulation all over Europe during the Chernobyl nuclear accident. NbA bears seven acid-base functional groups, among which are two enolic and two carboxylic acid moieties. This work deals with complex formation of Cs(+) and NbA in ethanol, ethanol/water (9:1) (M1), and water with, when required, the support of two Cs(+) ionophore probes, calix[4]arene-bis(crown-6-ether)dioxycoumarine (A1) and its tetrasuslfonated form (A2). In ethanol, two Cs(+) complexes are formed, with the affinity constants K(1EtOH) = (1.1 ± 0.25) × 10(5) and K(2EtOH) = (2.1 ± 0.4) × 10(3). In M1, a single Cs(+) complex occurs when only the enols are deprotonated, whereas a bicomplex is formed when both enols and carboxylic acids are deprotonated: K(1M1) = (1.5 ± 0.3) × 10(5) and K(2M1) = (4 ± 2) × 10(3). These data are confirmed by stopped-flow and T-jump kinetics. In ethanol, a fast Cs(+) exchange occurs between NbA and A1: direct rate constant, k(1) = (3.1 ± 0.1) × 10(7) M(-1) s(-1); reverse rate constant k(-1) = (2.8 ± 1) × 10(5) M(-1) s(-1); and Cs(+) exchange constant, K(1Exchange) = (9 ± 4) × 10(-3). In M1, the quenching of A2 fluorescence by NbA is used to determine the kinetics of complex formation with Cs(+): k(2) = (1.8 ± 0.4) × 10(9) M(-1) s(-1); k(-2) = (1.80 ± 0.15) × 10(4) s(-1); and K(1M1) = (1.5 ± 0.5) × 10(5). The affinity of NbA for Cs(+) is probably the result of the particular structure in which the two pulvinic acid arms adopt a conformation that forms two complexation sites composed of the two enolates and/or the two carboxylates. This renders the efficiency in Cs(+) uptake comparable to that of some calixarenes or crown ethers.

The photophysics of a ditopic receptor of potassium ion consisting of a 1,3-alternate calix[4]biscrown with a merocyanine dye (DCM) inserted into each crown is reported. Thanks to the large difference between the binding affinity for one and two potassium ions, one can find relative total concentrations of ligand and potassium ion at which the 1:1 complex is most predominant with respect to the free ligand and the 2:1 complex whose amounts are a few percents. Investigation of the 1:1 complex by femtosecond transient absorption spectroscopy provides evidence for the ultrafast movement of a potassium ion through the calix[4]arene tube upon excitation at 400 nm of the dye. Phototranslocation occurs in the picosecond timescale with a non-exponential kinetics without competition with photoejection towards the bulk. The translocation time includes two main short components: 0.83 ps and 10 ps. A smaller-weighted third component of 101 ps might include a competition between phototranslocation and excitation energy transfer as shown by using Förster's theory. These findings open the way to new strategies for light-driven molecular shuttles with the aim of information storage and binary logic computing at a nanometric scale.

The photophysical properties of a complex based on diphenylphosphanoethane (DPPE) fluorescent ligands linked to a europium ion have been investigated by different spectroscopic methods. Upon complexation with europium, the interaction of the phosphane oxide group with europium leads to a red shift of the absorption spectrum and a strong quenching of the ligand emission. The typical sensitized emission of Eu(3+) is observed upon excitation of the ligand with a fluorescence quantum yield of 1%. Time-resolved absorption and emission experiments have been performed in order to investigate the photophysical mechanism involved in this complex. Photophysical studies show that an energy-transfer mechanism occurs from both the first excited singlet and triplet states of the ligand, and the population of the europium ion to the (5)D(1) state takes place, from which the (5)D(0) state is populated. Additionally, electron transfer from the excited singlet state of the ligand to the europium ion appears as a very efficient process.

Complex formations between calix[4]arene-bis(crown-6-ether) calix-COU2 (A1) and the tetrasulfonated species calix-COUSULF (A2) with Cs(+) are investigated in water and ethanol, and in 9:1 (M1) and 1:9 (M2) H(2)O/EtOH v:v mixtures, by chemical relaxation and molecular modeling. In ethanol and M2, two Cs(+) are included in A1 in two kinetic steps, whereas complex formation in M1 becomes controlled by a slow first-order kinetic process, which is accompanied by very fast Cs(+) inclusions, second-order rate constant: k'(1) = (3.4 +/- 0.8) x 10(7) M(-1) s(-1). In water and M1, A2 forms 1:1 and 1:2 cesium complexes in a single kinetic step, whereas in M2, two Cs(+) are included in two kinetic steps. The rate and thermodynamic constants involved are reported. They show that the second-order rate constants increase with the ethanol-to-water ratio, e.g., A2, second-order rate constant for the first Cs(+) in water: k(1A2water) = (9.7 +/- 0.3) x 10(4) M(-1) s(-1) and in M2: k(1A2M2) = (6.3 +/- 0.4) x 10(9) M(-1) s(-1). The affinities of both A1 and A2 for Cs(+) also increase with the ethanol-to-water ratio, e.g., first inclusion of A1 in M1: K(1A1M1) = (5 +/- 1.3) x 10(3) and in ethanol: K(1A1EtOH) = (7 +/- 3) x 10(6). The deviation from the expected mechanism of complex formation with alkali is attributed to the comparatively more difficult access of Cs(+) to the inclusion cavity of the capped calixarene. An analysis of calix-COU2 and calix-COUSULF and their Cs(+) complexes with only one rim capped by the crown ether confirms the thermodynamic and kinetic results, by showing that the inclusion cavity of calix-COUSULF is more adapted to Cs(+) than that of calix-COU2. This added to the presence of the shielding effect of the negative sulfonates can explain that the affinity of calix-COUSULF for Cs(+) is higher than that of calix-COU2. These results can be of interest in the search of an efficient Cs(+) decontaminant.

A microfabricated device has been developed for the selective detection of lead in water. It is based on the use of a selective and sensitive fluorescent molecular sensor for lead (Calix-DANS4) which contains a calix[4]arene bearing four dansyl groups. The microchip-based lead sensor contains a Y-shape microchannel equipped with a passive mixer and moulded on a glass substrate. An optimization of the microcircuit length has been performed in order to have a full complexation of the Calix-DANS4. The detection is performed by using a configuration in which the sensing molecules are excited by two optical fibres, each one connected to a 365 nm UV LED, and the light collection is made by another optical fibre with a photomultiplier. By using this configuration we have shown the possibility to detect lead with a detection limit of 5 ppb. The effect of interfering cations such as calcium has been evaluated. The obtained measurements have been validated by an alternative method (ASV).

The fluorescence decays of a tetrachromophoric calix[4]arene and a heptachromophoric cyclodextrin with appended pyridin-2'-yl-1,2,3-triazole groups, and their complexes with the cadmium ion, are analyzed with a sum of compressed hyperbola. Such functions are shown to be well suited for the recovery of the underlying distributions of decay times. The heterogeneity of the micro-environments experienced by the chromophores, and/or their mutual interactions in the excited state can in this way be displayed graphically. A fit with a sum of four exponentials is also found to be satisfactory, but the respective parameters lack direct physical meaning.

A series of fluorescent sensor molecules based on a phosphane sulfide derivative were designed and synthesized. The effect of the distance between the complex entities as well as the number and the length of fluorophores was investigated using both steady-state and time-resolved fluorescence methods. The complexation behavior of these sensor molecules, which have a distinct affinity for Hg(2+), is reported. The coordination of Hg(2+) induces a photoinduced electron transfer from fluorophore to the complexed mercury and results in a significant decrease of the fluorescence. Theses sensors exhibit very low detection limits in CH(3)CN/H(2)O (80/20 v:v) and excellent sensitivity to Hg(2+) over other potentially interfering cations such as Na(+), K(+), Mg(2+), Ca(2+), Cu(2+), Ag(+), Zn(2+), Cd(2+) and Pb(2+).

G protein-coupled receptors (GPCRs) constitute the largest family of membrane receptors and are of major therapeutic importance. Structure determination of G protein-coupled receptors and other applications require milligram quantities of purified receptor proteins on a regular basis. Recombinant GPCRs fused to a heterologous biotinylation domain were produced in the yeast Pichia pastoris. We describe an efficient method for their rapid purification that relies on the capture of these receptors with streptavidin immobilized on agarose beads, and their subsequent release by enzymatic digestion with TEV protease. This method has been applied to several GPCRs belonging to the class A rhodopsin subfamily, leading to high yields of purified proteins; it represents a method of choice for biochemical and biophysical studies when large quantities of purified GPCRs are needed.

Photoinduced calcium release from the crown ether-linked merocyanine DCM-crown is reexamined by femtosecond transient absorption spectroscopy with sub-100 fs time resolution. Photodisruption of the bond linking the cation to the nitrogen atom shared by the crown and the chromophore is found to take place in 130 fs. Confirming our previous reports, the photoinduced intraligand charge transfer is observed in the picosecond regime but kinetics involving three-components (1 ps, 8 ps and 77 ps), together with a 56 ps stimulated-emission time-resolved red shift, are found in the present study. Both delayed intraligand charge transfer and cation release are assumed to occur in this time range due to repulsion effects between the positively charged nitrogen of the crown ether moiety and the cation. In the subnanosecond regime, a 670 ps time-resolved red shift of the stimulated-emission spectrum of the charge-transfer state, similar to the shift previously observed with Sr(2+), demonstrates the motion of the cation away from the crown to the bulk. A thorough examination of the present data allows us to conclude that calcium ion is photoejected to the bulk in a multistep process.

Introduction: In the present study, we investigated the ability of microparticles isolated from synovial fluids from patients with rheumatoid arthritis or osteoarthritis to induce the synthesis and release of key cytokines of B-lymphocyte modulation such as B cell-activating factor, thymic stroma lymphopoietin, and secretory leukocyte protease inhibitor by rheumatoid fibroblast-like synoviocytes., Methods: Microparticles were analyzed in synovial fluids from patients with rheumatoid arthritis, osteoarthritis, microcristalline arthritis, and reactive arthritis. In addition, microparticle release after activation from various cell lines (CEM lymphocyte and THP-1 cells) was assessed. Microparticles were isolated by differential centrifugation, and quantitative determinations were carried out by prothrombinase assay after capture on immobilized annexin V. B cell-activating factor, thymic stroma lymphopoietin, and secretory leukocyte protease inhibitor release was evaluated by enzyme-linked immunosorbent assay., Results: Microparticles isolated from synovial fluids obtained from rheumatoid arthritis and osteoarthritis patients or microparticles derived from activated THP-1 cells were able to induce B cell-activating factor, thymic stroma lymphopoietin, and secretory leukocyte protease inhibitor release by rheumatoid arthritis fibroblast-like synoviocytes. Conversely, CEM-lymphocytes-derived microparticles generated by treatment with a combination of PHA, PMA and Adt-D did not promote the release of B cell-activating factor but favored the secretion of thymic stroma lymphopoietin and secretory leukocyte protease inhibitor by rheumatoid arthritis fibrobast-like synoviocytes. However, microparticles isolated from actinomycin D-treated CEM lymphocytes were not able to induce B cell-activating factor, thymic stroma lymphopoietin, or secretory leukocyte protease inhibitor release, indicating that microparticles derived from apoptotic T cells do not function as effectors in B-cell activation., Conclusions: These results demonstrate that microparticles are signalling structures that may act as specific conveyors in the triggered induction and amplification of autoimmunity. This study also indicates that microparticles have differential effects in the crosstalk between B lymphocytes and target cells of autoimmunity regarding the parental cells from which they derive.

The photophysical properties of the new pyridin-2'-yl-1,2,3-triazole chromophore have been investigated. Spectroscopic experiments and molecular modelling have provided evidence for a photoinduced charge transfer occurring from the triazole group to the pyridine ring. Hepta- and tetrachromophoric systems have been synthesized by covalently linking seven or four chromophores of this kind, respectively, to a beta-cyclodextrin and a calix[4]arene. They exhibit different fluorescence spectra, decays and quantum yields. Special attention has been paid to the binding of cadmium and zinc ions and to the resulting photophysical effects which are various and very different for the grafted beta-cyclodextrin and calix[4]arene systems.
