The current study was done to determine the molecular status and hormone changes in polycystic ovary syndrome and polycystic ovary disease. The study included 155 clinical specimens that included age (20- 45) years, which were collected from November 2022 to June 2023. The specimens included blood specimens for patients visiting Imam Al-Sadiq Hospital and Babylon Teaching Hospital for Maternity and Children in Babylon Governorate. The results of hormones included elevation in levels of prolactin and Free Testosterone were obtained in PCOS, PCOD patients group compared with healthy women. Coronary heart disease (CHD) is a well-established multifactorial disease. Inflammation and atherosclerotic lesions play a fundamental role in CHD progression. Interleukin-10 (IL-10), an anti-inflammatory cytokine has the potential to influence the development of CHD. Thus, the identification of variations in IL-10 gene might be beneficial for the early detection of CHD. The current study aimed to examine the role of IL-10 gene polymorphisms (rs1 800 896, rs1 800 871 and rs1 800 872) in association with CHD. After taking blood samples, DNA extraction was done followed by genotyping through tetra primer amplification refractory mutation system-polymerase chain reaction (tetra primer ARMS-PCR). Obtained results showed a significant difference in the genotypic distributions of rs1 800 896 (p < 0.001) and rs1 800 871 (p < 0.001). The obtained odds ratio (OR) with 95% confidence interval (CI) for rs1 800 896 revealed significant association with CHD (OR = 1.90, 95% CI 1.3–2.7, p < 0.001). Furthermore, dominant, co-dominant and recessive models of rs1 800 896 indicated a significant correlation with the elevated risk of disease. Whereas, for rs1 800 871, a decreased risk was detected (OR = 0.55, 95% CI 0.4–0.8, p < 0.01). Moreover, it has been observed that rs1 800 871 might play a protective role for CHD under co-dominant and recessive models. However, rs1 800 872 was not observed to be significantly associated with the genetic risk to CHD among the studied population. The acquired results suggested that rs1 800 896 and rs1 800 871 have a substantial relationship with CHD and rs1 800 896 may be used as a diagnostic biomarker for early detection of the disease. Abstract Background The involvement of cytokines in pathogenesis of pseudoexfoliation syndrome and glaucoma has been demonstrated in several studies. The aim of the present study was to explore the association between three promoter polymorphisms −592C/A (rs1800872), − 819C/T (rs1800871) and -1082A/G (rs1800896) of interleukin 10 (IL-10) gene with susceptibility to pseudoexfoliation syndrome (PEX), pseudoexfoliative glaucoma (PEXG), and primary open-angle glaucoma (POAG). Methods In this study, 114 PEX, 118 PEXG, 114 POAG patients and 126 healthy individuals from Iranian population were participated. Detailed ophthalmic examinations by an ophthalmologist including slit-lamp bio-microscopic examination, dilated examination of the lens, gonioscopy, and funduscopy were carried out on patients and controls. Genomic DNA was extracted from the blood samples and ARMS–PCR was performed to detect promoter polymorphisms of IL-10. Results In all three SNPs studied, there was a significant difference in the genotype distribution between patients and control subjects. Results revealed that the AA genotype of IL-10 -592C/A SNP is associated with PEX. However, TT genotype of −819C/T and AA genotype of -1082A/G SNP are significantly associated with susceptibility to either PEX or PEXG and POAG disorders. Furthermore, the ACC haplotype containing the IL-10 -1082A allele was associated with PEX (P = 0.02, OR = 5.76, 95% CI = 5.17–24.49), PEXG (P = 0.006, OR = 7.54, 95% CI = 6.62–30.76) and POAG (P = 0.003, OR = 8.11, 95% CI = 7.13–33.15). Conclusions Our results demonstrated that IL-10 gene promoter polymorphisms are associated with susceptibility to PEX, PEXG and POAG in Iranian population. Considering the fact that IL-10 polymorphisms are associated with various IL-10 expressions, further research is needed to explain its involvement in these disorders and the formation of extracellular fibrillar amyloid deposits in PEX and PEXG.

Interleukin-10 (IL-10) gene polymorphisms have been associated with severity and outcomes in patients with respiratory and nonrespiratory viral infections. The aim of this study was to assess whether rs1800871 and rs1800872 polymorphisms of IL-10 gene are associated with the clinical outcomes of COVID-19 in a Mexican population. Study subjects were 193 COVID-19 patients. The genotyping was carried out with real-time PCR and serum IL-10 levels were measured with enzyme-linked immunosorbent assay. Logistic regression analysis was used for analysis association with clinical outcomes. There was no evidence of an association between alleles, genotypes, or haplotypes frequencies between patient groups according to severity and outcomes. The rs1800871 and rs1800872 polymorphisms might not be genetic risk factors for severity and mortality for COVID-19 in Mexican mestizos patients from northwest Mexico. [ABSTRACT FROM AUTHOR]

Background: Tuberculosis (TB) remains a universal health problem with significant morbidity and mortality. Understanding the genetic factors affecting TB susceptibility is crucial for effective prevention and treatment. Interleukin-10 (IL-10), a regulatory cytokine, may influence TB pathogenesis through genetic variations., Methods: The PubMed, Embase, and Google Scholar databases were searched to find studies on the relationship between IL-10 gene variants and tuberculosis. Relevant studies from 2016 to 2024 were identified through database searches. The selected case-control studies met the inclusion criteria. Software such as Review Manager was used to analyze quantitative data, with statistical significance set at p< 0.05. We calculated odds ratios and their respective confidence intervals to evaluate the associations., Results: Nine studies examined IL-10 gene polymorphisms (rs1800871 and rs1800872) in TB susceptibility. The present study did not show a notable association between IL-10 gene polymorphisms and TB among all genetic models (allelic, homozygote, heterozygote, dominant, and recessive). The obtained p-value > 0.05 indicates an insignificant association between both gene polymorphisms of IL-10. An OR-1.13; 95% CI-0.85, 1.50 was obtained for the SNP rs1800871, whereas an OR-1.02; 95% CI-0.75, 1.40 was obtained for the SNP rs1800872., Conclusion: Our meta-analysis revealed no significant association between IL-10 gene polymorphisms and TB susceptibility, suggesting that these variations may not significantly contribute to TB susceptibility. Further research with a larger sample size and diverse ethnicities is needed to explore additional genetic variations and their implications in TB pathogenesis.

Aim. To study the association of the polymorphic markers (PMs) G(-238)A of the TNF gene, G(–174)C of the IL-6 gene, and G(–1082)A of the IL-10 gene with the clinical characteristics of chronic glomerulonephritis (CGN) and a response to immunosuppressive therapy (IST). Subjects and methods. Clinical syndromes at the time of diagnosis, the morphological types of nephritis, and a response to IST were analyzed in relation to the carriage of the examined PMs of the TNF, IL-6, and IL-10 genes in 102 patients with CGN. Results. No association was found between the PM G(-238)A of the TNF gene and the clinical features of CGN. The carriers of the C allele of the PM G(-174) C of the IL-6 gene versus the homozygous individuals were observed to have more frequently kidney dysfunction at the time of diagnosis (р=0.014). Hypertension was more common in the carriers of the AA genotype of the PM G(-1082)A of the IL-10 gene (p=0.023); moreover, they tended to have a more frequent concurrence of nephrotic syndrome and hypertension (p=0.082). Analysis of the distribution of the morphological types of CGN disclosed that the proliferative variants were more common in the patients with the GG genotype (the TNF gene) as compared to the A allele carriers (p=0.067); and the nonproliferative forms were in the individuals homozygous for GG (the IL-6 gene) as compared to the C allele carriers (p=0.067). Examination of an IST response showed that a complete response at 12 months of treatment occurred more frequently in the carriers of the C allele of the IL-6 gene (p=0.045) and in those of the GG genotypes of the IL-10 gene (p=0.030). Conclusion. There was an association of the PMs G(–174)C of the IL-6 gene and G(–1082)A of the IL-10 gene with the clinical features of CGN and a response to IST.

Interleukin-10 (IL-10) is a anti-inflammatory cytokine, which controls inflammation by inhibiting the synthesis of several cytokines produced by Th1 cells and macrophages. The association between Interleukin-10 promoter polymorphisms with the risk of multiple sclerosis (MS) remains inconclusive. In this study, a meta-analysis has been performed to assess the relationship between IL-10 gene polymorphisms rs1800896, rs1800871 and rs1800872 with the risk of MS. Nine case-control studies were selected involving 2755 participants. The association between the polymorphisms and MS was examined by the pooled odds ratios (ORs) with 95% confidence intervals (CIs) in allelic, homozygote, heterozygote, dominant and recessive genetic models. Of analyzed genetic models, the pooled ORs and CIs of each SNPs calculated based on random (I 2 > 50) or fixed effects (I 2 < 50) methods, which showed no significant association (p-value > 0.05) of genetic predisposition with MS susceptibility across Asian and Caucasian populations. In addition, assessment based on funnel plot and Egger's linear regression test suggests no publication bias in all analyzed genetic models. Overall, our results demonstrated that rs1800896, rs1800871 and rs1800872 polymorphisms may not be the risk factor for the development of MS in both the populations. [ABSTRACT FROM AUTHOR]

Objectives The purpose of this study was to explore whether interleukin-10 ( IL-10 ) gene promoter polymorphisms and their haplotypes contribute to the incidence of non-Hodgkin lymphoma (NHL). Methods A meta-analysis was conducted on the associations of the IL-10 -3575T/A, -1082 G/A, -819 C/T, -592C/A polymorphisms and the haplotypes with NHL. Results A total of 23 studies involving 21,563 NHL patients and 23,837 controls were included in the meta-analysis. Pooled results indicated that IL-10 -3575T/A was associated with an increased risk of NHL based on the dominant model (OR: 1.095, 95% CI: 1.020–1.178), similar results were found for -1082A/G in the heterozygous and recessive models (OR: 1.042, 95% CI: 1.012–1.074; and OR: 1.034, 95% CI: 1.011–1.057, respectively). In the ethnic subgroup analysis, -3575T/A had an increased risk of NHL in Caucasians based on the homozygous model (OR: 1.071, 95% CI: 1.001–1.146), and similar results were found for -1082A/G in the heterozygous and recessive models (OR: 1.041, 95% CI: 1.009–1.075; and OR: 1.031, 95% CI: 1.008–1.055, respectively). When stratified by subtypes, -3575T/A and -1082A/G polymorphisms were found significant association with an increased risk of B cell lymphoma, specifically diffuse large B-cell lymphoma (DLBCL). Moreover, -3575T/A was associated with an increased risk of follicular lymphoma (FL) in the homozygous and recessive models. Furthermore, we observed that significantly increased risk of NHL and DLBCL were associated with the A-G-C-C haplotype (IL-10-3575T/A, -1082A/G, -819C/T and -592C/A), and a decreased risk of DLBCL subtype was associated with the T-A-C-C haplotype. Conclusions IL-10 -3575T/A and -1082A/G polymorphisms were associated with altered NHL susceptibility, especially for Caucasians and B cell lymphoma. Background and Aims: Hepatitis C virus (HCV) commonly causes a chronic infection but portion of hemodialysis patients are able to resistant to HCV infection, even clear the virus naturally. Interleukin-10 (IL-10) and interleukin-12 (IL-12) are crucial regulators of the immune response to T-helper 1 (Th1) or T-helper 2 (Th2) categories and play a role in autoimmune and infectious diseases. This study was undertaken to investigate the possible association of genetic polymorphisms of 3′ untranslated region (3′UTR) of the IL-12B (−1188) and IL-10 (−592 and −819) and hepatitis C in Chinese Han hemodialysis (HD) patients. Methods: The genotyping of IL-12B 3′UTR and IL-10 −592 and −819 were performed by polymerase chain reaction (PCR) restriction fragment length polymorphism (RFLP) method. Results: Compared with the IL-12B-AA genotype, CC and combined CC/AC genotypes were associated with a significant decreased risk of HCV infection in Chinese hemodialysis patients ( p < 0.001). However, the IL-10−592 and −819 genotypes were not found significant difference both between the anti-HCV (+) and anti-HCV (−) patients ( p > 0.05) and the persistent infection and viral clearance HD patients ( p > 0.05). Recently, it has been shown that the capacity of the innate immune system to produce cytokines relates to skeletal muscle mass and strength in older persons. The interleukin-10 ( IL-10) gene regulates the production capacities of IL-10 and tumour necrosis factor-α (TNF-α). In rural Ghana, IL-10 gene variants associated with different production capacities of IL-10 and TNF-α are enriched compared with Caucasian populations. In this setting, we explored the association between these gene variants and muscle strength. Among 554 Ghanaians aged 50 years and older, we determined 20 single nucleotide polymorphisms in the IL-10 gene, production capacities of IL-10 and TNF-α in whole blood upon stimulation with lipopolysaccharide (LPS) and handgrip strength as a proxy for skeletal muscle strength. We distinguished pro-inflammatory haplotypes associated with low IL-10 production capacity and anti-inflammatory haplotypes with high IL-10 production capacity. We found that distinct haplotypes of the IL-10 gene associated with handgrip strength. A pro-inflammatory haplotype with a population frequency of 43.2% was associated with higher handgrip strength ( P = 0.015). An anti-inflammatory haplotype with a population frequency of 7.9% was associated with lower handgrip strength ( P = 0.006). In conclusion, variants of the IL-10 gene contributing to a pro-inflammatory cytokine response associate with higher muscle strength, whereas those with anti-inflammatory response associate with lower muscle strength. Future research needs to elucidate whether these effects of variation in the IL-10 gene are exerted directly through its role in the repair of muscle tissue or indirectly through its role in the defence against infectious diseases. Summary: Objectives: To evaluate the correlation between IL-10 gene polymorphisms and hepatitis B infection. Methods: Tag single nucleotide polymorphisms (SNPs) were used to investigate the relationship between IL-10 gene polymorphisms and susceptibility to chronic hepatitis B virus (HBV) infection by comparing 996 chronic HBV infection cases to 301 acute infection controls. Results: This study found that rs3024490 G/T allele, located in the intron 1 region and highly prevalent in Chinese populations, was significantly different between the chronic HBV infection cases and the acute infection controls in single allele analysis, genetic models analysis, and haplotypes analysis. Conclusions: This suggested that the rs3024490 within IL-10 was associated with susceptibility to chronic hepatitis B in a Chinese Han population. [Copyright &y& Elsevier]

Increasing evidence suggests that interleukin-10 ( IL-10) gene promoter polymorphisms may be associated with chronic hepatitis C virus ( HCV) infection and HCV clearance. To more precisely estimate the association between these variants and the risk of HCV infection, we performed a meta-analysis of 26 studies describing the IL-10-1082A/G, -819C/T, -592C/A genotypes, including 4039 chronic HCV infection cases and 2902 controls. When compared with a healthy population, the -1082 GG allele had a 43% increased risk of chronic HCV infection in combined populations ( GG vs GA + AA: odds ratio ( OR) = 1.433, 95% confidence interval ( CI) = 1.052-1.952, P = 0.023). In subgroup analysis by ethnicity, a significant increased risk was associated with the −1082 GG genotype in the Caucasian population ( GG vs AA: OR = 1.390, 95% CI: 1.108-1.744, P = 0.004; GG vs GA + AA: OR = 1.621, 95% CI: 1.267-2.075, P = 0.000). However, no significant association was found in Asian, African or Chinese populations. Moreover, a higher distribution of −592A was found in the spontaneously recovered population ( AA vs CC: OR = 0.585, 95% CI = 0.387-0.884, P = 0.011; AA + AC vs CC: OR = 0.738, 95% CI = 0.551-0.988, P = 0.041; AA vs AC + CC: OR = 0.788, 95% CI = 0.664-0.935, P = 0.006) than that in the chronic HCV infection population. In conclusion, the IL-10-1082 GG allele may increase the risk of chronic HCV infection in Caucasian population, and people carrying the IL-10-592A allele are more likely to clear HCV spontaneously. Background: Up to now, many publications about the Chinese population have evaluated the correlation between interleukin-10 (IL-10) −1082 and −592 polymorphisms and persistent hepatitis B virus (HBV) infection. However, the results remain inconclusive. In order to resolve this conflict, a meta-analysis was performed. Methods: Seven studies were included and dichotomous data are presented as the odds ratio (OR) with a 95% confidence interval (CI). Results: The results of our study suggest that carriers of the IL-10 −592A allele were more likely to clear HBV spontaneously in the Chinese pooled population (A vs. C: OR = 0.799, 95% CI = 0.678-0.941, P = 0.007; AC vs. AA: OR = 1.343, 95% CI = 1.017-1.684, P = 0.011; AA vs. AC + CC: OR = 0.736, 95% CI = 0.594-0.912; AA + AC vs. CC: OR = 0.588, 95% CI = 0.408-0.848, P = 0.004) and the IL-10 −1082A allele was associated with significantly reduced persistent HBV infection risk in Chinese (A vs. G: OR = 0.701, 95% CI = 0.494-0.996, P = 0.047; AA vs. GG + GA: OR = 0.684, 95% CI = 0.476-0.982, P = 0.040). Conclusions: Persistent HBV infection susceptibility is associated with the gene polymorphism IL-10 −1082GA in the Chinese population and the clearance of HBV is associated with the gene polymorphism IL-10 −592CA in the Chinese population. Abstract The present study was carried out to examine the relationship of interleukin (IL)-10 gene polymorphism at position –819 for major depression and schizophrenia in the Korean population. DNA was extracted from 92 Korean patients with major depression, 141 Korean patients with schizophrenia, and 146 ethnically matched controls. DNA was amplified by a polymerase chain reaction-based method and digested by Mae III, and the restriction fragment length polymorphism of two alleles, IL-10*C and IL-10*T, were assessed. There were no significant differences in genotype frequencies of IL-10*T/T, IL-10*T/C, and IL-10*C/C as well as allelic frequencies of IL-10*T and IL-10*C between patients with major depression and controls in the Korean population. Comparison of genotype and allelic frequencies of IL-10 gene between patients with schizophrenia and controls were also not significant. The present study suggests that IL-10 gene polymorphism at position –819 does not confer susceptibility to major depression and schizophrenia, at least in the Korean population. Introduction: Type 2 diabetes mellitus (T2DM) is a chronic metabolic disease caused by insulin resistance. Insulin resistance leads to hyperglycaemia that causes complication such as microangiopathy and macroangiopathy. The immune system of T2DM will be produce IL-10 as an anti-inflammatory cytokine role immune-stimulator and immunosuppressant in the organ system. This present study investigated of IL-10 gene profile and protein expression in the rat organ ( Rattus norvegicus ) strain Wistar model T2DM., Material and Methods: This research was used three of male rats group T2DM and three of male of normal rat as a control. The DNA tissues were isolated, amplified and sequenced by using IL-10 gene primer. The IL-10 protein profile and expression of rat tissues was analyzed using Experion-Pro260 gel and dot blotting using IL-10 antibody., Results: This study showed the differential expression of IL-10 gene profile among tissues among normal and T2DM groups. The IL-10 gene sequences, we found eight mutations in brain and twenty-seven mutations on gastric of T2DM group compare with control group, meanwhile there are no mutation in other tissues of both groups. The protein profile of all tissues in both groups was completely diverse as proper. Moreover, the level expression of IL-10 of heart, lung, gastric and kidney of T2DM group was lower than other tissues of both groups., Conclusion: This study concludes that T2DM animal model triggering mutation of IL-10 gene sequences of brain and gastric and induced the increasing level expression of IL-10 of ileum, brain and liver.

Current opinion varies in the roles of the IL-10 polymorphisms in the process of hepatitis B virus (HBV) infection. We have performed a systemic review and up-dated meta-analysis including 37 eligible case-control studies to summarize all the available data on the association between IL-10 polymorphisms and development of HBV infection. In the present study, we found that the IL-10-1082 G/A, -592 C/A polymorphisms were associated with a significantly decreased risk of chronic HBV infection (AA + GA vs. GG: P = 0.003, OR = 0.55, 95% CI = 0.37-0.82; AA vs. CA + CC: P = 0.03, OR = 0.83, 95% CI = 0.71-0.98). While the -819 C/T TT carriers were associated with a borderline significantly decreased risk of chronic HBV infection (TT vs. CT + CC: P = 0.05, OR = 0.82, 95% CI = 0.68-1.00). Significant result was observed in the association between IL-10-1082 G/A polymorphism and HBV clearance (AA vs. GG: P = 0.04, OR = 1.33, 95% CI = 1.01-1.75). In addition, significant association was found between the -1082 G/A, -819 C/T polymorphisms and an increased risk of progression of HBV infection from asymptomatic carrier to chronic hepatitis B (AA + GA vs. GG: P = 0.0003, OR = 2.13, 95% CI = 1.41-3.22; TT + CT vs. CC: P = 0.005, OR = 1.53, 95% CI = 1.13-2.07), whereas the -592 C/A polymorphism was associated with a significantly decreased risk of progression from asymptomatic carrier to hepatocellular carcinoma (AA vs. CC: P = 0.02, OR = 0.63, 95% CI = 0.43-0.92). Our meta-analysis suggested that the IL-10 polymorphisms might be associated with a decreased risk of chronic HBV infection, while the -1082 AA carriers might be more likely to clear HBV following acute infection. In addition, these three polymorphisms might cast in roles of the progression of HBV infection.

Lung cancer is one of the most common cancers in the world, especially in China. It is believed that genetic polymorphisms played a role in cancer susceptibility. Here we investigated the association of interleukin-6 (IL-6) and interleukin-10 (IL-10) gene polymorphisms with the susceptibility of lung cancer in never-smoking Chinese Han population. In this study, we performed a case-control study including 330 cases of never-smoking lung cancer patients and 336 cancer-free never-smoking controls in Chinese Han population. We used polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method to identify gene polymorphisms, and then verified by sequencing method. The results indicated that the four single nucleotide polymorphisms (IL-6 -1363T/G and -572G/C, IL-10 -819T/C and -592A/C) were genotyped by PCR-RFLP and confirmed by sequencing, and we found that the allelic frequencies of G in IL-6 -1363T/G, C in IL-10 -819T/C and C in IL-10 -592A/C were significantly increased in lung cancer patients, by comparing with the control group. However, there was no significant difference in the distribution of the IL-6 572G/C polymorphisms between patients and controls. In conclusion, the IL-6 -1363T/G, IL-10 -819T/C and IL-10 -592A/C polymorphisms are closely related to genetic susceptibility to lung cancer in never-smoking Chinese Han population, and these genetic variants might be used as molecular markers for detecting lung cancer susceptibility.
