Simple Summary: For the extraction of equine chorionic gonadotropin (eCG), mares in the first 2–4 months of pregnancy are harvested for blood. The extracted eCG has been used in pharmaceutical products for the control of reproduction in other animal species. Blood harvesting from mares has raised concerns around their welfare, such as the possibility of them developing anaemia. This study was carried out to investigate the effect on the haematological variables of repeated blood harvesting from pregnant mares in two free-range herds. Our results show that up to 14.3% of mares in one herd had transient moderate to marked anaemia, but that no mares in the other herd developed more than mild anaemia. No mares in either herd had anaemia as defined by Hct three weeks after the last blood harvest. This difference between the herds indicates that external factors such as nutrients ingested by the mares are vitally important in the response to blood loss. Studies have been carried out on the effect of large-volume blood harvesting from horses, but they were performed on nonpregnant horses of various breeds other than Icelandic horses. This study aimed to investigate the effect on the haematological variables indicating erythropoiesis of repeated blood harvesting from pregnant mares. To account for regional variation, two herds of mares were chosen, both kept under free-range conditions. Sequential EDTA blood samples were collected weekly from 160 mares and analysed for haematological variables in an automated analyser. Serum samples from 115 mares were analysed for total protein. In both herds, after three harvests, mares began to measure below the minimum value for erythrocyte numbers, and mild anaemia was present in up to 37% at one time. Mares in only one herd had moderate or marked anaemia, 14.3% of the herd. Both herds showed evidence of increased erythropoiesis, but there was a difference between the herds in the intensity of the response. In both herds, however, all mares had reached the minimum normal haematocrit value three weeks after the last harvest. Abstract Background The back plays a vital role in horse locomotion, where the spine functions as a spring during the stride cycle. A complex interaction between the spine and the muscles of the back contribute to locomotion soundness, gait ability, and performance of riding and racehorses. Conformation is commonly used to select horses for breeding and performance in multiple horse breeds, where the back and croup conformation plays a significant role. The conformation of back and croup plays an important role on riding ability in Icelandic horses. However, the genes behind this trait are still unknown. Therefore, the aim of this study was to identify genomic regions associated with conformation of back and croup in Icelandic horses and to investigate their effects on riding ability. One hundred seventy-seven assessed Icelandic horses were included in the study. A genome-wide association analysis was performed using the 670 K+ Axiom Equine Genotyping Array, and the effects of different haplotypes in the top associated region were estimated for riding ability and additional conformation traits assessed during breeding field tests. Results A suggestive quantitative trait loci (QTL) for the score of back and croup was detected on Equus caballus (ECA) 22 (p-value = 2.67 × 10− 7). Purpose: To describe the most common ocular abnormalities in the Icelandic horse with focus on equine recurrent uveitis (ERU) and association between ocular abnormalities and summer eczema and coat colors. Methods: A descriptive cross‐sectional study. A complete physical and ophthalmic examination as well as measurement of serum titers for Leptospira serovariants was performed on Icelandic horses from Denmark (DK) and the United States (USA). Results: One hundred and twelve Icelandic horses living in DK and 26 Icelandic horses living in the United States were included in this study (total of 138 horses, 274 eyes). The three most common ocular abnormalities were follicular conjunctivitis (55.8%, 153/274 eyes), cataracts (27.4%, 75/274 eyes), and multifocal chorioretinopathy ("bullet holes"; 19%, 52/274 eyes). The prevalence for ERU among Icelandic horses aged "eight years and older" was 8% (6/75 horses). The "insidious ERU" type was found in 83% of the ERU cohort (5/6 horses), while "classic ERU" was found in 17% (1/6 horses). A correlation between ERU and cataracts was found in the population (p <.01). Leptospiral serology results were not associated with ERU in either DK or US‐Icelandic horses (p = 1.00, p = 1.00, respectively). ERU was not associated with summer eczema (p =.49), and no coat colors were associated with the ocular abnormalities found in the Icelandic horse (all p‐values >.05). Conclusion: Follicular conjunctivitis, cataracts, and multifocal chorioretinopathy were the three most common ocular abnormalities findings. This genomic region harbors three genes: C20orf85, ANKRD60 and LOC100056167. ANKRD60 is associated with body height in humans. C20orf85 and ANKRD60 are potentially linked to adolescent idiopathic scoliosis in humans. Conclusions: Our results show that the detected QTL for conformation of back and croup is of importance for quality of lateral gaits in Icelandic horses. These findings could result in a genetic test to aid in the selection of breeding horses, thus they are of major interest for horse breeders. Equine insect bite hypersensitivity (IBH) is a pruritic skin allergy caused primarily by biting midges, Culicoides spp. IBH susceptibility has polygenic inheritance and occurs at high frequencies in several horse breeds worldwide, causing increased costs and reduced welfare of affected horses. The aim of this study was to identify and validate single nucleotide polymorphisms (SNPs) associated with equine IBH susceptibility. After quality control, 33,523 SNPs were included in a Bayesian genome‐wide association study on 177 affected and 178 unaffected Icelandic horses. We report associated regions in E. caballus (ECA) 1, 3, 15 and 18, overlapping with known IBH QTLs in horses, and novel regions containing several genes, together explaining 11.46% of the total genetic variance. For validation, three SNPs on ECA 1 and ECA X (explaining the largest percentage of genetic variance) within 1‐mb genomic windows for IBH were genotyped in an independent population of 280 Exmoor ponies. The associated genomic region (152–153 mb) on ECA 1 was confirmed in Exmoor ponies and contains the AQR gene involved in splicing processes and a long non‐coding RNA. This study confirms the polygenic nature of IBH susceptibility and suggests a role of transcriptional regulatory mechanisms (e.g., alternative splicing) for IBH predisposition in these horse breeds. This study investigated the effects of rider weight in the BW ratio (BWR) range common for Icelandic horses (20% to 35%), on stride parameters in tölt in Icelandic horses. The kinematics of eight experienced Icelandic school horses were measured during an incremental exercise test using a high-speed camera (300 frames/s). Each horse performed five phases (642 m each) in tölt at a BWR between rider (including saddle) and horse starting at 20% (BWR20) and increasing to 25% (BWR25), 30% (BWR30), 35% (BWR35) and finally 20% (BWR20b) was repeated. One professional rider rode all horses and weight (lead) was added to saddle and rider as needed. For each phase, eight strides at speed of 5.5 m/s were analyzed for stride duration, stride frequency, stride length, duty factor (DF), lateral advanced placement, lateral advanced liftoff, unipedal support (UPS), bipedal support (BPS) and height of front leg action. Stride length became shorter (Y=2.73−0.004x; P0.05). In conclusion, increased BWR decreased stride length and increased DF proportionally to the same extent in all limbs, whereas BPS increased at the expense of decreased UPS. These changes can be expected to decrease tölt quality when subjectively evaluated according to the breeding goals for the Icelandic horse. However, beat, symmetry and height of front leg lifting were not affected by BWR.

In total, eight experienced school-horses were used in an incremental exercise test performed outdoors on an oval riding track and one rider rode all horses. ABSTRACT Iceland is free of the major infectious diseases of horses. However, in 2010 an epidemic of respiratory disease of unknown cause spread through the country’s native horse population of 77,000. Microbiological investigations ruled out known viral agents but identified the opportunistic pathogen Streptococcus equi subsp. zooepidemicus (S. zooepidemicus) in diseased animals. We sequenced the genomes of 257 isolates of S. zooepidemicus to differentiate epidemic from endemic strains. We found that although multiple endemic clones of S. zooepidemicus were present, one particular clone, sequence type 209 (ST209), was likely to have been responsible for the epidemic. Concurrent with the epidemic, ST209 was also recovered from a human case of septicemia, highlighting the pathogenic potential of this strain. Epidemiological investigation revealed that the incursion of this strain into one training yard during February 2010 provided a nidus for the infection of multiple horses that then transmitted the strain to farms throughout Iceland. This study represents the first time that whole-genome sequencing has been used to investigate an epidemic on a national scale to identify the likely causative agent and the link to an associated zoonotic infection. Our data highlight the importance of national biosecurity to protect vulnerable populations of animals and also demonstrate the potential impact of S. zooepidemicus transmission to other animals, including humans. IMPORTANCE An epidemic of respiratory disease affected almost the entire native Icelandic horse population of 77,000 animals in 2010, resulting in a self-imposed ban on the export of horses and significant economic costs to associated industries. Although the speed of transmission suggested that a viral pathogen was responsible, only the presence of the opportunistic pathogen Streptococcus zooepidemicus was consistent with the observed clinical signs. We applied genomic sequencing to differentiate epidemic from endemic strains and to shed light on the rapid transmission of the epidemic strain throughout Iceland. We further highlight the ability of epidemic and endemic strains of S. zooepidemicus to infect other animals, including humans. This study represents the first time that whole-genome sequencing has been used to elucidate an outbreak on a national scale and identify the likely causative agent.

This study examined the response in terms of heart rate (HR), respiratory rate (RR), haematocrit (Htc), rectal temperature (RT), and some plasma variables in Icelandic horses of different sexes and ages performing the riding assessment in a breed evaluation field test (BEFT). The study was conducted in Iceland on 266 horses (180 mares and 86 stallions, divided into four age groups; 4, 5, 6 and ⩾7 years old). RT and RR were recorded and blood samples were taken before the warm-up and after the riding assessment. Horse HR, velocity and distance were recorded during the warm-up, the riding assessment and a 5-min recovery period. The distance covered in the BEFT was 2.9±0.4 km (range: 1.8 to 3.8 km, n=248), the duration was 9:37±1:22 min:s (range: 5:07 to 15:32 min:s, n=260) and the average speed was 17.8±1.4 km/h (range: 13.2 to 21.3 km/h, n=248). Average HR was 184±13 b.p.m. (range: 138 to 210 b.p.m., n=102) and peak HR 224±9 b.p.m. (range: 195 to 238 b.p.m., n=102), and 36% of the BEFT was performed at HR ⩾200 b.p.m. Post-exercise plasma lactate concentration (Lac) was 18.0±6.5 mmol/l (range: 2.1 to 34.4 mmol/l, n=266), and there was an increase in total plasma protein, plasma creatine kinase and aspartate amino transferase concentration, as well as RR, RT and Htc. While HR, Hct and BF recovered to values at rest within 30 min, Lac and RT did not. All horses had no clinical remarks on palpation and at walk 1 and 2 days after the test. In conclusion, increasing BWR from 20% to 35% resulted in increased HR, Lac, RT and BF responses in the test group of experienced adult Icelandic riding horses. The horses mainly worked aerobically until BWR reached 22.7%, but considerable individual differences (17.0% to 27.5%) existed that were not linked to horse size, but to back BCS. Horses are often stabled in individual boxes, a method that does not meet their natural needs and may cause psychical and musculoskeletal diseases. This problem is particularly evident in Iceland, where horses often spend the long winter periods in cramped boxes. The aim of this study was to analyze the suitability of a group housing system in Iceland, but the results are also applicable to horses of other regions. Eight Icelandic horses were observed in an active stable system, and their behavior and time budget were recorded. Movement and lying behavior were studied with ALT (Activity, Lying, Temperature detection) pedometers. The effect of an automatic concentrate feeding station (CFS) on the horses’ behavior was examined. In the first period of investigation, the horses were fed concentrates manually, and in the second period, they were fed with the CFS. Additional behavioral observations and a determination of social hierarchy occurred directly or by video surveillance. The physical condition of the horses was recorded by body weight (BW) measurement and body condition scoring (BCS). The results showed a significant increase between the first and second trial periods in both the activity (P < 0.001) and the lying time (P = 0.003) of the horses with use of the CFS. However, there was no significant change in BW during the first period without the CFS (P = 0.884) or during the second period with the CFS (P = 0.540). The BCS of the horses was constant at a very good level during both trial periods, and the horses showed a low level of aggression, a firm social hierarchy and behavioral synchronization. This study concludes that group housing according to the active stable principle is a welfare-friendly option for keeping horses and is a suitable alternative to conventional individual boxes.

We explore how herd composition and management factors correlate with frequencies of social interactions in horse groups. Since the welfare of horses correlates with low aggression levels and social contact opportunities, information of this kind is important. The data are a collection of records of social interactions of 426 Icelandic horses in 20 groups of at least eight horses. The complexities and limitations of the data prohibit useful statistical modelling so the results are presented descriptively. Interesting and informative patterns emerge which can be of use both in management and in future studies. Of special interest are the low levels of agonistic behaviours in breeding groups where one stallion was present. The horses were less agonistic when in groups with young foals and where group membership was stable. Unfamiliar yearlings in peer groups were especially aggressive. Allogrooming was most frequent in groups with relatively more young horses and in unstable and small groups. Interestingly, the horses allogroomed more if they had few preferred allogrooming partners. The findings show that composition (age/sex) and stability of groups are of great importance with respect to aggression levels and opportunities for establishing bonds.

Background: Obesity is related to the development of several diseases like insulin resistance and laminitis in horses. The prevalence of obesity among mature Icelandic horses in Denmark has not been investigated previously. This study aimed to find the prevalence of obesity, to compare body condition score (BCS) based on owner perception with that of an experienced person and to correlate the BCS to body weight (BW) and morphometric measures in a group of mature Icelandic horses in Denmark. A total of 254 Icelandic horses (=4 years; 140 geldings, 105 mares, 9 stallions) from 46 different farms were included. All horses were assigned a BCS on a scale from 1 to 9 (1 is poor, 5 is moderate and 9 is extremely fat) by their owner and by an experienced person. Two weight tapes were used to assess BW. Girth circumference (GC), neck circumference (NC) and height at withers (HW) were measured, and the GC:HW and NC:HW ratios were calculated. Results: Categorising the horses into four groups, 5.9% were underweight (BCS 3-4), 70.1% were optimal (BCS 5-6), 13.8% were overweight (BCS 7) and 10.2% were obese (BCS 8-9). The GC:HW and NC:HW ratios increased with increasing BCS, as did the BW estimated with the weight tapes. A GC:HW ratio >1.21 might indicate overweight or obesity in Icelandic horses. Horse owners underestimated the BCS of their horses compared to an experienced person. Conclusions: The results from this study show that 24.0% of mature Icelandic horses in Denmark are overweight or obese, and that owners tend to underestimate the BCS of their Icelandic horses. The GC:HW ratio might indicate overweight or obesity, however, the ratio for Icelandic horses is different than reported for horses and ponies of other breeds. [ABSTRACT FROM AUTHOR]

Cryptorchidism is a defect in males where one or both testes fail to descend normally into the scrotum. If both testes are retained, the stallion will be infertile. There are few studies of the prevalence of the condition in horses based on field data, and heritability estimates for equine cryptorchidism are lacking. The objective of this study was to provide data on prevalence and heritability of cryptorchidism in Icelandic horses born in Sweden. A questionnaire was sent to breeders, asking whether or not stallions born at the farm had both testes present in the scrotum at the age of 1, 6 and 12 months. Fixed effects were analysed using logistic regression and genetic parameters were estimated using linear animal models. Information about 595 yearling stallions born 1997–2011 was used in the analyses. Close to 9% of the yearlings did not have both testes in the scrotum. Probability of cryptorchidism in yearlings was significantly influenced by farm and time period of birth. Heritability estimates for cryptorchidism ranged from 0.12 to 0.32 (SE 0.08–0.12) on the observable scale, and from 0.35 to 0.96 (SE 0.24–0.40) when transformed to the underlying continuous scale. The results support that equine cryptorchidism is heritable. Further studies with more individuals included would be needed to confirm this. [ABSTRACT FROM AUTHOR]

This study compared the effect of ridden tölt and trot at 3 speeds on physiological responses in trained adult (15.3 ± 1.6 yr) Icelandic horses. The experiment had a crossover design with 8 horses, 2 treatments (incremental exercise test in tölt and trot), and 2 riders. Each horse performed 2 tests per day (1 gait with 2 riders, minimum 4.5 h between) on 2 separate days, with 1 d of rest in between. The exercise test consisted of three 642-m phases at 3.0 m/s (Speed3), 4.0 m/s (Speed4), and 5.0 m/s (Speed5) and was performed outdoors on a 300- m oval gravel riding track in northern Iceland in May 2012. Heart rate (HR) was measured during warm-up, the exercise test, and after 5, 15, and 30 min of recovery. Blood samples were taken at rest, after warm-up, after each phase of the exercise test, and after 5, 15, and 30 min of recovery. Respiratory rate was counted for at least 15 s at rest, at the end of the exercise test, and at the end of the 30-min recovery, and rectal temperature was measured on these occasions. There were no differences in HR between tölt and trot at any time point (P > 0.05). At Speed3, hematocrit and plasma lactate concentration were greater (P < 0.05) in tölt (40% ± 1%, 1.1 ± 0.06 mmol/L) than in trot (39% ± 1%; 0.9 ± 0.06 mmol/L). There was a prolonged recovery of hematocrit and respiratory rate, a slower decrease in rectal temperature, and a tendency of a prolonged recovery of plasma lactate concentration (P = 0.0675) after tölt. In conclusion, there were only minor differences in physiological responses to tölt and trot in this selected group of experienced adult Icelandic horses and the biological and practical significance of the slightly elevated physiological responses to tölt and the slower recovery remains to be determined. Previous studies have shown that a single base-pair mutation, a change from cytosine (C) to adenine (A), in the DMRT3 gene affects both the ability to show ambling and lateral gaits in a wide range of horse breeds, as well as racing performance and trotting technique in Standardbred and Nordic trotters. The variant allele is present in gaited breeds but is absent, or found at a very low frequency, in breeds used for Western-European style riding and flat racing, like the Swedish Warmblood and Thoroughbreds as well as in draught horses. This indicates that the variant allele might have a negative effect on certain riding performance traits in horses. Therefore, one aim of this study was to investigate whether the DMRT3 variant affects canter in Standardbred trotters, and to test if heterozygous horses ( CA ) were better suited for Western-European style riding than homozygous horses ( AA ). Riding traits were studied in 115 Standardbred horses, and a similar study was also performed with data from 55 Nordic trotters. The results showed that CA Standardbreds had significantly better balance in canter, both collected and extended canter, than AA horses. The CA horses also got significantly higher scores for transitions in collected canter. For the rhythm we found no significant differences between the genotypes. In the Nordic trotters we were unable to establish any significant difference for canter ability. Another aim of this study was to investigate the effect of the variant allele on riding abilities and gaits in the Icelandic horse ( n =446). Practically all horse breeds considered to be three-gaited have a CC genotype, in contrast Icelandic CC horses can show tölt. We therefore tested whether the variant influenced how difficult it was to initiate tölt training for these horses. It was also investigated whether the variant affects which gaits Icelandic horses choose, both at liberty and during initial training. Icelandic CC horses were significantly more difficult to train to tölt compared to CA and AA horses. The AA Icelandic horses showed the lateral gaits tölt and pace significantly more frequent, both at liberty and during initial training, than CA or CC horses. This study examined the response in terms of heart rate (HR), respiratory rate (RR), haematocrit (Htc), rectal temperature (RT), and some plasma variables in Icelandic horses of different sexes and ages performing the riding assessment in a breed evaluation field test (BEFT). The study was conducted in Iceland on 266 horses (180 mares and 86 stallions, divided into four age groups; 4, 5, 6 and ⩾7 years old). RT and RR were recorded and blood samples were taken before the warm-up and after the riding assessment. Horse HR, velocity and distance were recorded during the warm-up, the riding assessment and a 5-min recovery period. The distance covered in the BEFT was 2.9±0.4 km (range: 1.8 to 3.8 km, n=248), the duration was 9:37±1:22 min:s (range: 5:07 to 15:32 min:s, n=260) and the average speed was 17.8±1.4 km/h (range: 13.2 to 21.3 km/h, n=248). Average HR was 184±13 b.p.m. (range: 138 to 210 b.p.m., n=102) and peak HR 224±9 b.p.m. (range: 195 to 238 b.p.m., n=102), and 36% of the BEFT was performed at HR ⩾200 b.p.m. Post-exercise plasma lactate concentration (Lac) was 18.0±6.5 mmol/l (range: 2.1 to 34.4 mmol/l, n=266), and there was an increase in total plasma protein, plasma creatine kinase and aspartate amino transferase concentration, as well as RR, RT and Htc. Stallions covered a longer total distance (in the warm-up and BEFT) (P<0.05), at a faster speed during BEFT (P<0.001) than mares and had higher Htc and lower HR and post-exercise Lac values. There were few effects of age, but the 4- and 5-year-old horses had lower Htc than older horses and 4-year-old horses had higher post-exercise RR than older horses, although they were ridden for a shorter distance, shorter duration and at lower peak velocity (P<0.1). The results showed that the riding assessment in the BEFT is a high-intensity exercise. The results also showed that aerobic fitness was higher in stallions and that age had a limited effect on the physiological response. It is suggested that these results should be used as a guide for the development of training programmes and fitness tests in Icelandic horses that would improve both performance and welfare of the horse. Abstract: Earlier research indicates that stallions may supress interactions of their harem members, leading to less stable hierarchies and friendship bonds in harems compared to non-stallion groups. In this paper, the effect of the presense of a stallion on the social behaviour of mares was studied by comparing six harems containing stallions to four mixed sex groups not containing stallions. Both temporary and permanent harems were studied, giving the possibility to investigate the effect of group stability on social interactions. A significant linear hierarchy was found in all non-stallion groups that were used for comparison, while the hierarchies were only found to be linear in three of the six harems containing stallions (Landaus h′, p <0.05). Aggression rate was lower (t-test, p <0.05) and fewer friendship bonds (G-test, p <0.0001) were found within the harems, compared to the groups without stallions. Stallions seldom intervene directly in interactions between harem members. Thus, our results give support to the hypothesis that stallions may suppress interactions of harem members, but in a more indirect way than with direct interference. In addition, our results give support for earlier findings that aggression rate may be affected by group stability. We found a higher aggression rate in the temporary harems compared to the permanent harems (Kruskal–Wallis, p <0.05) and in the temporary non-stallion group compared to the permanent non-stallion group. Abstract: The aim of the present study was to evaluate the quality of raw and cooled semen in Icelandic stallions. Experiments were performed using seven stallions aged between 3 and 19 years. From each stallion, six ejaculates were collected, and semen quality was determined. Thereafter, the semen was split into eight equal parts and processed with and without centrifugation using the extenders INRA 82-egg yolk, INRA 96, GENT, and Equi-Pro to a final concentration of 30 × 106 sperm/mL. The extended semen was then cooled in an Equitainer, where it was stored for 24 hours, and subsequently refrigerated for another 24 hours at 5°C. Immediately after dilution as well as after 24 and 48 hours storage, sperm motility was analyzed using computer-assisted sperm analyzer, and viability was assessed after dual DNA staining with SYBR-14 in combination with propidium iodide. The results show that the stallion had a significant (P < .05) influence on all variables evaluated in raw semen, and mean (±SEM) values of 43.4 ± 4.3 mL for the volume, 193.0 ± 17.0 × 106 sperm/mL for the concentration, 6.7 ± 0.5 × 109 for total sperm and 73.5 ± 2.1% for total sperm motility, 48.7 ± 2.0% for progressive motility, and 65.3 ± 2.0% for rapid cells were measured. In the cold-stored semen, all variables were significantly (P < .05) influenced by the stallion, extender, and storage time (48 hours). Except for Equi-Pro, all extenders examined were suitable for cooled semen preservation. For storage of more than 24 hours, centrifugation and removal of the seminal plasma were advantageous for all extenders with the exception of Equi-Pro. [Copyright &y& Elsevier]

Reasons for performing study The role of equid γ-herpesviruses on ocular surface diseases has been disputed, because the diagnosis is usually based on clinical symptoms and detection of viral DNA from samples obtained from live animals. Objectives To describe the clinical course, results of polymerase chain reaction (PCR) analysis, in situ hybridisation, cell culture and pathohistological findings of select cases in a presumed outbreak of herpesvirus infection in a group of 15 Icelandic horses. Study design Case series. Methods Pooled ocular and nasal swabs and peripheral blood mononuclear cells of horses diagnosed clinically with herpesvirus-associated keratoconjunctivitis were analysed for presence of equine herpesviruses (EHV)-2 and EHV-5 nucleic acid using real-time PCR. Necropsy specimens from one horse, subjected to euthanasia due to deterioration of clinical symptoms were examined histopathologically, and analysed for presence of EHV-2 and EHV-5 nucleic acid using real-time PCR. In situ hybridisation and cell culture of select samples were performed. Results All horses with symptoms of severe keratoconjunctivitis were positive for presence of either EHV-2 and/or EHV-5 nucleic acid using real-time PCR. Assessment of necropsy specimens of the most severely affected case, revealed presence of EHV-2 and/or EHV-5 nucleic acid in several ocular and extraocular anatomical locations. The remaining horses responded favourably to symptomatic treatment. Conclusions This case series illustrates a severe outbreak of keratoconjunctivitis in a group of Icelandic horses, with suspected γ-herpesvirus involvement. The ingestive and post-digestion effect of a blend of special essential oil compounds (EO) on eating, chewing and faecal parameters were measured in horses. Ingestive effects appear after no adaptation. Post-digestion effects appear after adaptation. Six Icelandic horses were assigned to two groups in a Latin Square subplot design with EO treatments to four different roughage types and four different concentrates. The horses were fed four different roughage meals and two different concentrate meals on each of the four sampling days. Eating time and saliva were observed during meals. Jaw movements (JM) were recorded using a special chewing halter. Eating time was derived from JM and related to DM intake. The size characteristics of faecal particles were measured by using image analysis. All chewing characteristics measured were significantly affected by roughage (p < 0.001) and concentrate type (p < 0.01). EO had a significant ingestive effect on the frequency of observed saliva during concentrate meals. No significant (p < 0.05) post-digestive or ingestive effect of EO was found for any measured chewing characteristic, which was reflected in the absence of effect on faecal particle dimensions. In this study we describe the generation of monoclonal antibodies (mAbs), which recognize different epitopes of the equine IgE constant heavy chain. Equi-murine recombinant IgE (rIgE), composed of the murine VH186.2 heavy chain variable region, linked to the equine IgE constant heavy chain and expressed together with the murine λ1 chain in J558L cells was used to immunize BALB/C mice. A total of 17 different mAbs were obtained, which recognized the rIgE heavy chain constant region. None of the mAbs reacted with monoclonal equine isotypes IgM, IgG1 (IgGa), IgG3 (IgG(T)), IgG4 (IgGb) or isolated equine light chains, IgGc and IgA from horse serum, or the native mAb B1-8δ, expressing the same heavy chain variable regions and light chains. One of the mAbs (αIgE-132) recognized the recombinant equine IgE, but did not recognize any protein in equine serum, i.e. native IgE. A total of 16 mAbs detected a serum protein of approximately 210,000 Da on Western blots, corresponding to the expected MW of native IgE. In addition, one of the mAbs (αIgE-176) detected a protein of 76,000 Da under reducing conditions, most likely the equine IgE heavy chain. According to binding inhibition studies, the equine IgE specific mAbs recognize at least two different epitopes of the equine IgE. In an ELISA using two anti-IgE mAbs which recognized different epitopes, no significant differences in the concentration of total serum IgE could be detected between adult Icelandic horses with IgE-mediated type I allergy (summer eczema) and healthy control animals. In Icelandic horse foals, no serum IgE could be measured 6 months post partum. All anti-IgE mAbs recognized a small population (1.3±0.5%) of leukocytes from adult Icelandic horses by surface immunofluorescence, but no cells could be detected in foal blood. The stained leukocytes from adult horses could be enriched by magnetic cell sorting and contained 32% basophils, 53% monocytes and/or large lymphocytes, 13% small lymphocytes and 2% eosinophils. Summary: The aim of this study was to evaluate potential risk factors associated with hindlimb lameness and radiographic signs of degenerative joint disease (DJD) in the distal tarsus in Icelandic horses. The material consisted of riding horses (n = 420) age 6–12 years sired by 17 selected sires representing all major breeding lines, and of riding horses (n = 194) in the same age range sired by other sires. The examination protocol included the following: interview with owners/trainers, assessment of conformation, motion evaluation and radiographic examination. At the interview, data related to factors outside the horse (environmental variables) and data associated directly with the horse (intrinsic variables) were obtained. Data analysis was performed in 2 steps: screening using bivariate analysis, followed by testing with a multivariate logistic regression model. In the multivariate analysis, the factors of sire, age when broken to saddle and stud show participation were strongly associated with the prevalence of lameness. Height at the croup and ability to perform different gaits were also associated with the prevalence of lameness, but to a lesser degree. The risk factors of age, tarsal angle and birthplace were significantly associated with radiographic signs of DJD in the distal tarsus. Neither the variation in applied training intensity, the use of a professional oramateurtrainernorthe temperament or front limb action of the individual horse was significantly associated with the prevalence of hindlimb lameness and/or radiographic signs of DJD in the distal tarsus in the Icelandic horse. Abstract: The body weight of horses is an interesting variable for horse owners, as well as for veterinarians. It is useful to have methods of estimation like weight tape or formula, rather than visual estimation, when a weight scale is not available. Different methods of weight estimation exist for adult horses, but to date, there has been no validation of these methods in Icelandic horses. Therefore, three different methods of weight determination (weight scale, weight tape, and weight formula) were compared in this study to investigate whether it is possible to obtain reliable weight measurements of Icelandic horses by using an estimation method. Both the weight pursuant to weight tape and pursuant to weight formula showed no significant differences (P = 1.00 and P = 0.37, respectively) from the actual weight. The correlation between estimated and actual weights was r = 0.94 for weight tape and r = 0.93 for weight formula. However, the differences between the estimated and actual weights were smallest when estimated using an adjusted weight formula: weight (kg) = girth² × length (both in cm)/11,400. In conclusion, the body weight of Icelandic horses can be estimated from the measurements of body girth and length, and weight tape also seems to be a suitable method. [Copyright &y& Elsevier]
