Abstract Background A new species of smut fungus, Urocystis heteropogonis, was discovered infecting Heteropogon contortus in Shawar Valley, Swat district, Khyber Pakhtunkhwa, Pakistan. The study aimed to characterize this fungus based on its morpho-anatomical and molecular features and clarify its phylogenetic position within the genus Urocystis. Results Urocystis heteropogonis was identified as a novel species, distinct from other Urocystis species. Morphologically, it is characterized by larger spore balls (14–69 × 11–45 μm) and central spores that are 14–28 × 11–20 μm in size, with each spore containing1–8 central spores. The spore walls measure 0.9–2.5 μm in thickness and the species differs in infection patterns compared to other Urocystis species. Phylogenetic analysis based on the ITS and LSU regions of nuclear ribosomal DNA (nrDNA) further confirmed the novelty of the species, placing it within a distinct clade alongside U. agropyri, U. occulta, U. piptatheri, and U. tritici. Conclusions The discovery of Urocystis heteropogonis adds to the diversity of smut fungi infecting grasses and highlights the need for further research into its ecological and agricultural implications. Future studies should focus on the disease’s spread, management, and potential impact on host populations.

Abstract This study explored the drought resistance mechanisms of different populations of Sporobolus ioclados (Poaceae), locally known as “Sawri,” “Drabhri” and “Dhrbholi” native to Africa and the Indian Subcontinent. These populations were grown in conventional nursery practices at Khawaja Fareed Government College in Rahim Yar Khan, Pakistan, and subsequently subjected to four distinct levels of drought within carefully monitored experimental settings. The experiment was conducted in a two-factorial design involving populations and drought treatments and was repeated three times. The physiological and morphological responses of S. ioclados, including plant height, number of roots, root length, flag leaf area, stomatal features, proline concentration and nitrogen content, displayed significant variability in response to the imposed drought stress. Drought resulted in increases in proline concentration and nitrogen content. The number of roots decreased, while the length and width of the stomata increased in various populations. A combination of advanced statistical techniques, such as ANOVA, PCA, HCA, and DFA, provided a comprehensive understanding of the mechanism of plant adaptation and the extent of population diversity within the species. The Yazman and Nwab Wala populations exhibited the highest rates of photosynthesis and stomatal conductance, while S. ioclados demonstrated notable drought tolerance at the T4 level of drought stress. A negative correlation was found between proline levels, nitrogen contents, and photosynthesis, suggesting that proline has a protective role in drought. The diverse adaptation strategies indicated by S. ioclados populations have revealed the potential of this species for afforestation and climate change mitigation in dry environments.

Abstract Bio-transformations refer to the chemical modifications made by an organism on a chemical compound that often involves the interaction of plants with microbes to alter the chemical composition of soil or plant. Integrating bio-transformations and entomopathogenic fungi into litchi cultivation can enhance symbiotic relationships, microbial enzymatic activity in rhizosphere, disease suppression and promote overall plant health. The integration of biological formulations and entomopathogenic fungi can significantly influence growth, nutrient dynamics, physiology, and rhizosphere microbiome of air-layered litchi (Litchi chinensis Sonn.) saplings. Biological modifications included, K-mobilizers, AM fungi, Pseudomonas florescence and Azotobacter chroococcum along with Metarhizium, entomopathogenic fungi have been used. The treatments included, T1-Litchi orchard soil + sand (1:1); T2-Sand + AM fungi + Azotobacter chroococcum (1:2:1); T3-Sand + Pseudomonas florecence + K-mobilizer (1:1:1); T4- AM fungi + K-mobilizers (1:1); T5, P. Florecence + A. chroococcum + K-mobilizer (1:1:1); T6-Sand + P. florecence (1:2) and T7-Uninoculated control for field performance. Treatments T4-T6 were further uniformly amended with drenching of Metarrhizium in rhizosphere. T2 application significantly increased resident microbe survival, total chlorophyll content and root soil ratio in seedlings. A. chroococcum, Pseudomonas, K-mobilizers and AM fungi increased in microbial biomass of 2.59, 3.39, 2.42 and 2.77 times, respectively. Acidic phosphatases, dehydrogenases and alkaline phosphatases were increased in rhizosphere. Leaf nutrients reflected through DOP were considerably altered by T2 treatment. Based on Eigen value, PCA-induced changes at biological modifications showed maximum total variance. The study inferred that the bio-transformations through microbial inoculants and entomopathogenic fungi could be an encouraging strategy to enhance the growth of plants, health and productivity. Such practices align well with the goals of sustainable agriculture through biological means by reducing dependency on chemical inputs. By delving into these aspects, the research gaps including microbial processes, competitive and symbiotic relationships, resistance in microbes and how complex interactions among bio-transformations, entomopathogenic fungi and microbes can significantly impact the health and productivity of litchi. Understanding and harnessing these interactions can lead to more effective and sustainable farming practices.

Abstract This research purpose was focused to document the traditional ethnobotanical knowledge (TEK) associated with important wild plants and indigenous communities residing in mountainous zone of District Sudhnoti of Azad Jammu and Kashmir, Pakistan and to determine their conservation status with suggestions and recommendations to protect and propagate the rare and endangered wild flora of the area for sustainable use. The data regarding traditional ethnomedicines (TEMs) and phytogeography with conservation analysis of wild plants were collected via structured and semi structured interviews of 150 participants belonging to various professions and of both genders. Prior consent and permission were obtained from family heads and from all the participants, and the data were shared with all the informants through the local female translator guide. To validate the collected data, various microstatistical tools, such as the informant consensus factor (ICF), fidelity level (FL) and data matrix ranking (DMR) were used. The relative frequency of citations (RFC) and rank order of popularity (ROP) were also calculated to determine and authenticate the relative importance of TEMs which may be further studied in future research and used for drug discovery. In the present study, 150 locally important plants belonging to 69 families were documented, and the Rosaceae was the most dominant and prevalent family. The results indicated that many locally important plants have multiple uses such as food, medicine, fodder, shelter, ornamental and fuel. The TEMs obtained from these plants have been known to be useful for curing various infirmities such as flu, renal disorders, fever, malaria, cough, migraine, whooping cough, influenza, skin rashes, allergies, stomach aches, wounds and bruises, diabetes, tumours and joint pains. The ICF analysis revealed that renal calculus, malaria, fever, whooping cough, rheumatoid arthritis and arthritis were prevalent infirmities of the mountainous area. The FL analysis indicated the popularity of these plants used in various TEMs and among these Artemisia maritima and Berberis lycium were the most common. The DMR indicated that Pinus roxburghii was the most common species being used for multipurpose by the indigenous communities. The highest use value index was found for Indigofera linifolia. The relative frequency of citations was calculated to determine the importance of plants in traditional ethnomedicine, highlighting Morus nigra, Pinus wallichiana, and Rosa indica as significant species. The Jaccard index indicated a high level of novelty in the research which can be exploited for neo-drug discovery and drug development. In conclusion, this study has successfully documented the ethnobotanically and ethnomedicinally important plants in District Sudhnoti, Azad Jammu and Kashmir, Pakistan. The findings of this research contribute to the preservation and documentation of cultural heritage, as well as provide a foundation for further studies in ethnobotany, ethnopharmacology and biodiversity conservation efforts for sustainable provision of wild flora to the indigenous communities which will also assist in combating drasting climatic changes.

Fiber trait is multigenic and each parent of the population carrying different genes aid in arriving at varied blend of association between yield and fiber quality traits. Eleven parental lines were crossed in half-diallel fashion to arrive at 55 hybrid combinations, which were evaluated under seven environments. The results manifested highly significant differences among the traits studied, indicating wide range of genetic diversity in the parental genotypes. Combining ability studies revealed parental genotypes SCS-PL-01, SCS-PL-04 as best general combiners for seed cotton yield; SCS-PL-04, SCS-PL-12, SCS-PL-51, SCS-PL-59 for fiber length; SCS-PL-51 for fiber strength and SCS-PL-02 for micronaire value. Genetic diversity of parental genotypes estimated using microsatellites was compared with SCA effects to understand the role of genetic distance (GD) in realizing the heterosis. Comparative characterization of parental genotypes indicated efficiency of capillary gel electrophoresis over metaphor agarose gel electrophoresis and could identify 55 unique bands. The study indicated predicting heterosis using GD estimates is competent when markers linked to the QTLs are used. Also, marker-assisted parental selection in combining ability studies would be a better approach in realizing potential crosses with high yield and fiber quality traits.

Rice False Smut (RFS) disease (Ustilaginoidea virens) has become a severe problem in many parts of the world. Managing this disease is a major priority as the disease causes a significant yield loss. In the present study, we have investigated the effectiveness of bioformulations, biocontrol agents, and fungicides for managing RFS disease at the lab and field levels. The study evaluated bioformulations LBD 1 and 6, biocontrol agents Trichoderma viride, Bacillus velezensis (P-42 and A-6), and Pseudomonas fluorescens strain Pf-1, and various fungicides such as trifloxystrobin 25% + tebuconazole 50% WG, propiconazole 25% EC, difenconazole 25% EC, flusilazole 12.5% + carbendazim 25% EC, and azoxystrobin 23% SC against RFS. All fungicides tested in the in vitro assay exhibited complete mycelial inhibition against U. virens. On the other hand, biocontrol and bioformulations showed 41.03%–93.53% mycelial inhibition. Pooled data from two seasons (Kharif 2019 & 2020) indicated that all the tested biocontrol agents and fungicides effectively reduced the severity of the RFS disease in field conditions. The fungicide trifloxystrobin 25% + tebuconazole 50% WG at 0.4 gL-1 was found to be the most effective, with the lowest disease severity of 18.23%, followed by 20.24% severity by flusilazole 12.5% + carbendazim 25% EC at 1.0 mLL-1. Fungicides were found to be the best for the management of the disease among different treatments. This is the first comprehensive record of successfully managing the RFS disease using seaweed extracts, fungicides, and microbial bioagents and to standardize the spray schedule for better disease control.

The experiment consisted of 12 treatments (weed interference until 15, 30, 45, 60, and 75 days after emergence (DAE), weedy until crop maturity, and weed-free until 15, 30, 45, 60, 75, and a weed-free until crop maturity) and the treatments were arranged in a randomized complete block design (RCBD) with three replications. This study indicated increased weed interference period reduced the pod yield by 6.5–36 and 0.7–32%, and haulm yield by 6.4 – 34 and 6.5 – 34% during rainy and winter season respectively. Further, reduction in total biomass production by crop due to weed interference declined the total nutrient uptake by crop, i.e. nitrogen by 7.8–52.1 and 9.6–54.5%, phosphorus by 4.8–45.4 and 6–47.5%, potassium by 6.9–43.7 and 6.3–44.4%, sulphur by 18.4–53.3 and 18.8–53.9%, during rainy and winter season respectively. However, weed-free until crop harvest increased RUE by 293% and 314%, chlorophyll content by 8.7 and 10.7%, respectively, during rainy and winter seasons. This study indicated critical duration for crop-weed interference under 5% relative yield loss (RYL) was 16 to 66 DAE in rainy and 15 to 63 DAE in winter. Similarly, at 10%, RYL were 23 to 62 and 21 to 61 DAE, respectively, during the rainy and winter seasons. As a result, farmers should keep their groundnut crop weed-free between 15 and 66 DAE during the rainy and winter seasons.

Abstract High temperature stress influences plant growth, seed yield, and fatty acid contents by causing oxidative damage. This study investigated the potential of thiourea (TU) to mitigate oxidative stress and restoring seed oil content and quality in canola. The study thoroughly examined three main factors: (i) growth conditions—control and high temperature stress (35 °C); (ii) TU supplementation (1000 mg/L)—including variations like having no TU, water application at the seedling stage, TU application at seedling stage (BBCH Scale-39), water spray at anthesis stage, and TU application at anthesis stage (BBCH Scale-60); (iii) and two canola genotypes, 45S42 and Hiola-401, were studied separately. High temperature stress reduced growth and tissue water content, as plant height and relative water contents were decreased by 26 and 36% in 45S42 and 27 and 42% Hiola-401, respectively, resulting in a substantial decrease in seed yield per plant by 36 and 38% in 45S42 and Hiola-401. Seed oil content and quality parameters were also negatively affected by high temperature stress as seed oil content was reduced by 32 and 35% in 45S42 and Hiola-401. High-temperature stress increased the plant stress indicators like malondialdehyde, H2O2 content, and electrolyte leakage; these indicators were increased in both canola genotypes as compared to control. Interestingly, TU supplementation restored plant performance, enhancing height, relative water content, foliar chlorophyll (SPAD value), and seed yield per plant by 21, 15, 30, and 28% in 45S42; 19, 13, 26, and 21% in Hiola-401, respectively, under high temperature stress as compared to control. In addition, seed quality, seed oil content, linoleic acid, and linolenic acid were improved by 16, 14, and 22% in 45S42, and 16, 11, and 23% in Hiola-401, as compared to control. The most significant improvements in canola seed yield per plant were observed when TU was applied at the anthesis stage. Additionally, the research highlighted that canola genotype 45S42 responded better to TU applications and exhibited greater resilience against high temperature stress compared to genotype Hiola-401. This interesting study revealed that TU supplementation, particularly at the anthesis stage, improved high temperature stress tolerance, seed oil content, and fatty acid profile in two canola genotypes.

Abstract Pakistan’s economy greatly benefits from citrus production since these fruits are sold and consumed all over the world. Although citrus fruits are easy to cultivate, they are susceptible to diseases caused by bacteria, viruses, and fungi. These challenges, as well as difficulties in obtaining the proper nutrients, might negatively impact fruit yields and quality. Citrus canker is another complicated problem caused by the germ Xanthomonas axonopodis. This germ affects many types of citrus fruits all over the world. This study looked closely at how citrus canker affects the leaves and the quality of the fruit in places like Sargodha, Bhalwal, Kotmomin, and Silanwali, which are big areas for growing citrus in the Sargodha district. What we found was that plants without the disease had more chlorophyll in their leaves compared to the sick plants. Also, the healthy plants had better amounts of important minerals like calcium, magnesium, potassium, and phosphorus in their fruits. But the fruits with the disease had too much sodium, and the iron levels were a bit different. The fruits with the disease also didn’t have as much of something that protects them called antioxidants, which made them more likely to get sick. This study helps us understand how citrus canker affects plants and fruit, so we can think of ways to deal with it.

To address the fungal wilt of pigeon pea caused by Fusarium oxysporium f. sp. udum, farmers currently rely on chemical fungicides, despite their harmful effects. However, there is a growing need for safer alternatives like green pesticides. Bacterial biocontrol agents and their derivatives serve as potential green pesticides in the management of plant pathogens. In the present study, we aimed to identify indigenous Bacillus subtilis strains effective against F. oxysporium f. sp. udum. We used PCR and MALDI-TOF analysis to identify the active components responsible for the efficiency of efficient strain. Biochemical studies of cell-free extracts extracted from B. subtilis strains demonstrated the highest biosurfactant activity in NBAIR BSWG1, with an oil displacement of 2 cm and an emulsification index of 60 %. Molecular characterization confirmed the presence of surfactin, fengycin, and iturin coding genes in the B. subtilis strains, among them, NBAIR BSWG1 showed the highest number of lipopeptide-producing genes. Meanwhile, NBAIR BSWG1 showed inhibition of 79.84 % against F. oxysporium f. sp. udum using cell-free extract. Further metabolite profiling of NBAIR BSWG1 using MALDI-TOF analysis further confirmed surfactin, fengycin, and iturin in the purified cell-free extract of NBAIR BSWG1. Two peaks with m/z of 923.77 and 1149.92 were identified as novel lipopeptide compounds which need further characterization. The present study identified NBAIR BSWG1 as an efficient bacterial strain for the inhibition of F. oxysporium f. sp. udum and its antifungal properties are mainly due to the production of cyclic lipopeptides.

The Fabaceae is renowned for its diverse range of chemical compounds with significant biological activities, making it a valuable subject for pharmacological studies. The chemical composition and biological activities of three Fabaceae species were investigated using methanol separately and in combination with dimethyl sulfoxide (DMSO) and glycerol for extraction. The results revealed the highest phenolic (49.59 ± 0.38 mg gallic acid equivalent/g), flavonoid (29.16 ± 0.39 mg rutin equivalent/g), and alkaloid (14.23 ± 0.54 mg atropine equivalent/g) contents in the Caesalpinia decapetala methanol extracts. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and DNA protection activity were the highest (0.88 ± 0.43 μg/mL IC50 and 2149.26 band intensity) in Albizia julibrissin methanol extracts. The α-amylase activity was highest in all methanol extracts ( C. decapetala (70.86 %) > Indigofera atropurpurea (65.34 %). Gas chromatography-mass spectrometry revealed 33 volatile compounds and, aamong these (Z)-9-octadecenamide was detected in the highest concentration ranging from 21.85 to 38.61 %. Only the methanol extracts of the examined species could be assessed for in vivo studies for immediate applications.

Multi-environment trials (MET) are crucial for selecting genotypes that are well-suited to different environmental conditions. Incorporating multiple traits in the analysis can provide more reliable recommendations for selecting genotypes with desirable traits, including resistance to the Mungbean Yellow Mosaic Virus (MYMV) and high yield potential. The use of a Multi-Trait Stability Index (MTSI) is a good approach for analyzing the stability of genotypes across multiple traits under MYMV stress. In the present investigation, the performance of thirteen green gram genotypes were evaluated for traits such as yield, plant height, number of branches per plant, and resistance to MYMV. The main objective of the study is to identify highly productive and stable mung bean genotypes resistant to MYMV. MTSI can be calculated by combining information on the performance of genotypes across multiple traits and environmental conditions to provide a single index that indicates the overall stability of genotypes across traits and environments. The results helped to identify two green gram genotypes (Yadadri and JNG-18) that were high-yielding with stable resistance to MYMV stress across multiple environmental conditions. This can provide useful information to breeders for the development of suitable genotypes against MYMV in the affected areas.

The designing of acceptors materials for the organic solar cells is a hot topic. The normal experimental methods are tedious and expensive for large screening. Machine learning guided exploration is more suitable solution. Bagging regression, random forest regression, gradient boosting regression, and linear regression are trained to predict exciton binding energy. Breaking Retrosynthetically Interesting Chemical Substructures (BRICS) methodology has utilized for designing of new non-fullerene acceptors (NFAs). The predicted values were used to select the designed NFAs. On the selected NFAs, clustering and chemical similarity analyses are also performed. Chemical fingerprints are used for this purpose, and the synthetic accessibility score of the new NFAs is also investigated.30 NFAs have selected with low exciton binding energy values. This approach will allow for the rapid screening of NFAs for organic solar cells. Our proposed framework stands out as a valuable tool for strategically selecting the most effective NFAs for organic solar cells and offers a streamlined approach for material discovery.

Abstract Background Drought is one of the limiting factors for quality and quantity of cotton lint in tropical and sub-tropical regions. Therefore, development of drought tolerant cotton genotypes have become indispensable. The identification of drought tolerant genotypes is pre-requisite to develop high yielding cultivars suitable for drought affected areas. Methods Forty upland cotton accessions were selected on the basis of their adaptability and yield. The collected germplasm accessions were evaluated at seedling stage on the basis of morphological, physiological and biochemical parameters. The experiment was conducted under controlled conditions in greenhouse where these genotypes were sown under different levels of drought stress by following factorial under completely randomized design. The data were collected at seedling stages for root and shoot lengths, relative leaf water content, excised leaf water losses, peroxidase content and hydrogen peroxide concentrations in leaf tissues. Results The biometrical analysis revealed that germplasm is significantly varied for recorded parameters, likewise interaction of genotypes and water stress was also significantly varied. The cotton germplasm was categorized in eight clusters based on response to water stress. The genotype Cyto-124 exhibited lowest H2O2 content under drought conditions, minimum excised leaf water loss under stress environment was exhibited by genotypes Ali Akber-802 and CEMB-33. Overall, on the basis of morphological and biochemical traits, SL-516 and Cyto-305 were found to be drought tolerant. Genotypes 1852 − 511, Stoneville 15–17 and Delta Pine-55 showed low values for root length, peroxidase activity and higher value for H2O2 contents. On the basis of these finding, these genotypes were declared as drought susceptible. Conclusion The categorization of cotton germplasm indicating the differential response of various parameters under the control and drought stress conditions. The recorded parameters particularly relative leaf water contents and biochemical assays could be utilized to screen large number of germplasm of cotton for water deficit conditions. Besides, the drought tolerant genotypes identified in this research can be utilized in cotton breeding programs for the development of improved cultivars.

The Spodoptera frugiperda is a notorious pest with a broad host range. It severely damages crops, mainly in areas of the globewhere maize and sorghum are grown. The pest is difficult to control due to its adaptive nature and resistance to several insecticides available in the market. So, an identification of the alternative strategy is the prime important in the present context. Insecticidal activities of cyanobacterial extracts were evaluated in the laboratory as a biocomponent against S. frugiperda. The crude extracts of Nostoc muscorum and Spirulina sp. were prepared by using ethanol, methanol and petroleum ether solvents. Soxhlet apparatus was used for extraction. S. frugiperda larvae in their second instar were given access to fragments of maize leaf that had been treated with various cyanobacterial extracts. The findings displayed that the petroleum ether extract of N. muscorum had the lowest LC50 value of 155.22 ppm, followed by petroleum ether extracts of Spirulina, ethanol extract of N. Muscorum, methanol extract of N. muscorum, ethanol and methanol extract of Spirulina with an LC50 values of 456.02, 710, 780, 1050 and 1070 ppm respectively. Later, the effect of LC50 values on many biological parameters like the larval duration and pupal stages, the percentage of pupation, the weight of the pupal stage, the malformation of the pupal and adult stages, adult emergence percentage, fertility and the longevity of the male and female adult stages of S. frugiperda was examined. The gas chromatography-mass spectrometry (GC-MS) was used to analyse the crude extract to identify the bioactive components that were responsible for the insecticidal properties. The major compounds detected were diethyl phthalate (19.87 %), tetradecane (5.03%), hexadecanoic acid, ethyl ester (4.10 %), dodecane (4.03%), octadecane (3.72%), octadecanoic acid, methyl ester (3.40 %), ethyl oleate (3.11 %), methyl ester. octadecenoic acid (3.04 %), heptadecane (3.04 %) and phytol (3.02 %). The presence of several bioactive chemicals in the cyanobacterial extracts may be the reason for their insecticidal actions, thus it can be used as an alternative and new source to combat fall armyworm and other crop pests.

Mountainous ecosystems are experiencing rapid shifts in distribution patterns and community structure due to the impacts of climate change and vegetation dynamics. This study offers a comprehensive insight into the adaptive mechanisms of Panicum antidotale to varying elevations within the Pothohar Plateau, shedding light on its survival strategies in climate-affected regions. The findings reveal that populations at lower elevations exhibit maximum height, leaf area, shoot length, and fresh weight, whereas those at higher elevations display significant decrease in these parameters. Moreover, a significant decline in species density, frequency and cover is evident at high elevation. High-elevation populations demonstrate decreased photosynthetic traits, including chlorophyll, carotenoid, and Na and Ca content. Furthermore, there are notable variations in the antioxidant enzyme activity across elevation gradients. Populations at lower elevations exhibit higher activities of superoxide dismutase and peroxidase. Across the gradient, there were significant reductions in root and stem radius, sclerenchyma thickness, bulliform cell size, metaxylem vessels, and phloem area. Populations at moderate elevations displayed thicker lamina and epidermis, larger cortical cells, wider xylem vessels, and increased phloem area; however, these traits declined at higher elevations. High-elevation populations showed thicker lamina and epidermis, extensive sclerenchyma, greater cortical cell area, and higher stomatal density. Photosynthetic traits and enzymatic activity were major contributors at moderate elevations while stem and leaf traits were significantly changed at high elevations. Overall, this research contributes significantly to the broader knowledge of plant adaptation in mountainous ecosystems and stresses the importance of considering elevation as a critical factor in ecological studies.

The cotton mealybug, Phenacoccus solenopsis Tinsley and papaya mealybug, Paracoccus marginatus Williams and Granara de Willink (Hemiptera: Pseudococcidae) are becoming major threats to the production of Gymnema sylvestre R. Br. (Asclepiadaceae) in India. Management mainly depends on chemical insecticides which cause a serious problem of pesticide residue and insecticide resistance. The use of biorational insecticides such as biopesticides, botanicals, insect growth regulators, and microbial insecticides is important components of an Integrated Pest Management (IPM) program for successful management. We evaluated the bio-efficacy of twelve biorational insecticides, including entomopathogenic fungi (EPF), using the leaf spray method in laboratory conditions at 25 ± 1 °C, 70 % ± 5 % RH. The results revealed that the highest percent mortality was recorded by acetamiprid 20 % SP (100.00 %), followed by azadirachtin (98.27 %), Lecanicillium muscarium (2 × 109 spores/mL) (85.70 %) and Ocimum sanctum leaf extract (76.87 %) at 120 h after treatment (HAT) in P. solenopsis. In P. marginatus, 100.00 %, 96.39 % and 85.67 % and 74.90 % mortalities were achieved by acetamiprid 20 % SP, azadirachtin, L. muscarium (2 × 109 spores/mL) and O. sanctum leaf extract, respectively, at 120 HAT during the first spray. Various biorational insecticides showed a more or less similar trend of percent mortality in both species during the second spray. In both species, the lowest percent mortality was recorded by Andrographis paniculata leaf extract (46.29, 44.54) and (41.03, 46.39) at 120 Hours after treatment in the first and second spray, respectively. It was concluded that all the prescribed treatments are more effective than the control. Overall, azadirachtin recorded the highest percent mortality after acetamiprid and had the shortest LT50 (12.52 h) and (13.87 h) values in P. solenopsis and P. marginatus, respectively. Our study emphasizes that biopesticides like Azadirachtin 1 % EC (10000 ppm), L. muscarium (2 × 109 spores/mL) (5 mL/L) and O. sanctum leaf extract (5 %) may be recommended as alternatives to synthetic insecticides. Botanicals and EPF would be the most effective approach for sustainable integrated management of P. solenopsis and P. marginatus in the G. sylvestre ecosystem.

AbstractLow nutrient availability in sandy soils is a serious threat to crop productivity. The present study evaluated the effectiveness of cotton-sticks biochar, sugarcane press-mud, and vegetable compost on soil chemical properties and chickpea yield in sandy soils. Two chickpea varieties, Bittal-2016 and Noor-2019, were grown in soil amended with cotton-sticks biochar at 10 and 15 Mg ha−1; sugarcane press-mud at 10 and 15 Mg ha−1; vegetable compost at 10 and 15 Mg ha−1; cotton-sticks biochar at 10 and 15 Mg ha−1 plus loaded with Fe/Zn including no amendment. Cotton-sticks biochar at 15 Mg ha−1 loaded with Fe/Zn significantly increased chlorophyll (48.40), N (3.1, 2.5, and 3.9%), P (1.13%, 1.10, and 1.4%), and K (1.44, 2.26, and 1.27%) in root, shoot, and grains of var. Noor-2019. Soil amended with vegetable compost at 10 Mg ha−1 and Fe/Zn-loaded biochar at 15 Mg ha−1 exhibited a marked increase in shoot length (43.2 cm), root length (25.8 cm), root fresh weight (8.0 g), pods per plant (34), 100-grain weight (32.7 g), and grain yield (25.9 g) for var. Noor-2019. Amended soil with Fe/Zn-loaded biochar at 10 Mg ha−1 depicted a remarkable increase in soil EC, pH, OM, and N by 2.12 dS m−1, 8.4, 0.67% and 0.20%, respectively. Conclusively, Fe/Zn-loaded cotton-sticks biochar at 15 Mg ha−1 and vegetable compost at 10 Mg ha−1 effectively improved the soil chemical properties, nutrient uptake, and yield of var. Noor-2019. Hence, cotton-sticks biochar could be used as eco-friendly approach for improving chickpea production in coarse-textured soils.

Staphylococcus aureus is part of normal human flora and is widely associated with hospital-acquired bacteremia. S. aureus has shown a diverse array of resistance to environmental stresses and antibiotics. Methicillin-resistant S. aureus (MRSA) is on the high priority list of new antibiotics discovery and glycopeptides are considered the last drug of choice against MRSA. S. aureus has developed resistance against glycopeptides and the emergence of vancomycin-intermediate-resistant, vancomycin-resistant, and teicoplanin-resistant strains is globally reported. Teicoplanin-associated genes tcaR-tcaA-tcaB (tcaRAB) is known as the S. aureus glycopeptide resistance operon that is associated with glycopeptide resistance. Here, for the first time, the role of tcaRAB in S. aureus persister cells formation, and ΔtcaA dependent persisters’ ability to resuscitate the bacterial population was explored. We recovered a clinical strain of MRSA from a COVID-19 patient which showed a high level of resistance to teicoplanin, vancomycin, and methicillin. Whole genome RNA sequencing revealed that the tcaRAB operon expression was altered followed by high expression of glyS and sgtB. The RNA-seq data revealed a significant decrease in tcaA (p = 0.008) and tcaB (p = 0.04) expression while tcaR was not significantly altered. We knocked down tcaA, tcaB, and tcaR using CRISPR-dCas9 and the results showed that when tcaA was suppressed by dCas9, a significant increase was witnessed in persister cells while tcaB suppression did not induce persistence. The results were further evaluated by creating a tcaA mutant that showed ΔtcaA formed a significant increase in persisters in comparison to the wild type. Based on our findings, we concluded that tcaA is the gene that increases persister cells and glycopeptide resistance and could be a potential therapeutic target in S. aureus.

Citrobacter freundii is characterized by AmpC β-lactamases that develop resistance to β-lactam antibiotics. The production of extended-spectrum β-lactamase (ESBL) is substantially high in Escherichia coli, C. freundii, Enterobacter cloacae, and Serratia marcescens, but infrequently explored in C. freundii. The present investigation characterized the ESBL C. freundii and delineated the genes involved in decrease in antibiotics resistance. We used the VITEK-2 system and Analytical Profile Index (API) kit to characterize and identify the Citrobacter isolates. The mRNA level of AmpC and AmpR was determined by RT-qPCR, and gel-shift assay was performed to evaluate protein-DNA binding. Here, a total of 26 Citrobacter strains were isolated from COVID-19 patients that showed varying degrees of antibiotic resistance. We examined and characterized the multidrug resistant C. freundii that showed ESBL production. The RT-qPCR analysis revealed that the AmpC mRNA expression is significantly high followed by a high level of AmpR. We sequenced the AmpC and AmpR genes that revealed the AmpR has four novel mutations in comparison to the reference genome namely; Thr64Ile, Arg86Ser, Asp135Val, and Ile183Leu while AmpC remained intact. The ΔAmpR mutant analysis revealed that the AmpR positively regulates oxidative stress response and decreases β-lactam and aminoglycosides resistance. The AmpC and AmpR high expression was associated with resistance to tazobactam, ampicillin, gentamicin, nitrofurantoin, and cephalosporins whereas AmpR deletion reduced β-lactam and aminoglycosides resistance. We conclude that AmpR is a positive regulator of AmpC that stimulates β-lactamases which inactivate multiple antibiotics.

This research pursued the effect of various calcination temperatures (300–1000 °C) on developing structural phases and the optical, dielectric, and photodegradation characteristics of cobalt-doped titanium dioxide nanoparticles (TiO2 NPs). The prepared NPs were characterized using X-ray diffraction (XRD), scanning electron microscopy coupled with energy-dispersive X-ray spectroscopy (SEM-EDX), Fourier transform infrared (FTIR) spectroscopy analysis, ultraviolet–visible light (UV–VIS) spectroscopy, and photodegradation of methylene blue in the presence of visible light. XRD analysis revealed the formation of a tetragonal anatase phase for the samples calcined at 300–600 °C, while the Co-doped samples calcined at 800 °C and 1000 °C displayed a tetragonal rutile phase. The optical band gap analysis indicated that doping in the host matrix produced lower band gap energy for all the prepared samples. Dielectric measurements showed that the rutile phase heated at 800 °C had a larger dielectric constant and dielectric loss than the un-doped TiO2 and the other cobalt-doped samples. Finally, the anatase Co-doped TiO2 exhibited a maximum MB degradation of 93 % in 90 min compared to un-doped TiO2, which only degraded 14 % and rutile-TiO2 NPs (62 % degradation). The underlying mechanism responsible for the diverse photodegradation performance displayed by the anatase and rutile phases of TiO2 NPs is discussed. Overall, these results demonstrate that cobalt doping and the crystalline phase of TiO2 NPs are vital parameters involved in optimizing the photocatalytic activity of TiO2.

Inadequate hygienic conditions and poor handling are the primary causes of contamination in abattoirs. This study aimed to identify and molecularly detect pathogenic bacteria in sheep meat at slaughterhouses in Riyadh City, Saudi Arabia. Additionally, the study evaluated the sensitivity of these bacteria to various antimicrobials. In total, 150 samples were collected three times every two months from November 2021 to March 2022 from four abattoirs located in the south, west, east, and north of Riyadh. Pathogenic bacteria were separated using selective media, and the Vitek 2 system was utilized to identify all species and test their response to antibiotics. PCR was employed to detect virulence genes. The four pathogenic bacteria identified in all samples were Escherichia coli (12%), Klebsiella pneumoniae (9.3%), Salmonella enterica (7.3%), and Pseudomonas aeruginosa (6.6%). Abattoir D had a high number of bacteria isolated in January. K. pneumoniae and S. enterica exhibited resistance to ampicillin. S. enterica also demonstrated resistance to gentamicin, ciprofloxacin, and trimethoprim/sulfamethoxazole. P. aeruginosa was resistant to tigecycline. PCR results indicated positive tests for the E. coli gene FimH, the P. aeruginosa genes plcH and toxA, and the K. pneumoniae gene mrkD. Pathogenic bacteria with positive results for virulence genes have the potential to cause contamination and human diseases. To improve quality and reduce contamination, the government must address the issue of providing adequate and safe water for activities in all abattoirs in Riyadh City.

Pomegranate production in Afghanistan is increasing remarkably, but nursery services are lacking, leading to inconsistent growth and low rooting percentages in cuttings. Hence, this research was conducted at the greenhouse of Nangarhar University, Faculty of Agriculture, to investigate the efficacy of indole-3-butyric acid (IBA) and organic materials, namely banana, garlic, aloe vera, and panchagavya, as rooting promoters on cuttings of the ideal seedless pomegranate cultivar (Bedana). The cuttings were derived from one-year-old dormant branches in mid-February of 2023. The findings indicated that panchagavya and aloe vera significantly (p < 0.05) accelerated the sprouting process of pomegranate cuttings, resulting in 100% rooting, while IBA showed a delayed initiation of sprouting. In contrast, banana had a detrimental effect on the sprouting of the cuttings. IBA and organic materials, except banana and garlic, significantly influenced all the shoot parameters. Among the treatments, panchagavya (21.1%), IBA (23.4%), and aloe vera (26.8%) demonstrated the highest number of shoots per cutting compared to the control. A consistent soil plant analysis development (SPAD) pattern was observed across treatments, positively correlating with shoot and root parameters (range, r = 0.30–0.97). The application of organic materials significantly (p < 0.05) impacted root length, acrobasal roots, root diameter, total root length, and root fresh weight. However, banana and garlic yielded contrasting results in most of these parameters. In conclusion, aloe vera and panchagavya are found to be more suitable for propagating pomegranates through cuttings, making them excellent alternatives to IBA for nurserymen and companies.

Oxidative stress and chronic inflammation interplay with the pathogenesis of cancer. Breast cancer in women is the burning issue of this century, despite chemotherapy and magnetic therapy. The management of secondary complications triggered by post-chemotherapy poses a great challenge. Thus, identifying target-specific drugs with anticancer potential without secondary complications is a challenging task for the scientific community. It is possible that green technology has been employed in a greater way in order to fabricate nanoparticles by amalgamating plants with medicinal potential with metal oxide nanoparticles that impart high therapeutic properties with the least toxicity. Thus, the present study describes the synthesis of Titanium dioxide nanoparticles (TiO2 NPs) using aqueous Terenna asiatica fruit extract, with its antioxidant, anti-inflammatory and anticancer properties. The characterisation of TiO2 NPs was carried out using a powdered X-ray diffractometer (XRD), Fourier transform infrared (FTIR), scanning electron microscopy (SEM), energy-dispersive X-ray diffraction (EDX), high-resolution transmission electron microscopy (HR-TEM), dynamic light scattering (DLS), and zeta-potential. TiO2 NPs showed their antioxidant property by scavenging 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals in a dose-dependent manner with an IC50 value of 80.21 µg/µL. To ascertain the observed antioxidant potential of TiO2 NPs, red blood cells (RBC) were used as an in vitro model system. Interestingly, TiO2 NPs significantly ameliorated all the stress parameters, such as lipid peroxidation (LPO), protein carbonyl content (PCC), total thiol (TT), superoxide dismutase (SOD), and catalase (CAT) in sodium nitrite (NaNO2)-induced oxidative stress, in RBC. Furthermore, TiO2 NPs inhibited RBC membrane lysis and the denaturation of both egg and bovine serum albumin, significantly in a dose-dependent manner, suggesting its anti-inflammatory property. Interestingly, TiO2 NPs were found to kill the MCF-7 cells as a significant decrease in cell viability of the MCF-7 cell lines was observed. The percentage of growth inhibition of the MCF-7 cells was compared to that of untreated cells at various doses (12.5, 25, 50, 100, and 200 µg/mL). The IC50 value of TiO2 NPs was found to be (120 µg/mL). Furthermore, the Annexin V/PI staining test was carried out to confirm apoptosis. The assay indicated apoptosis in cancer cells after 24 h of exposure to TiO2 NPs (120 µg/mL). The untreated cells showed no significant apoptosis in comparison with the standard drug doxorubicin. In conclusion, TiO2 NPs potentially ameliorate NaNO2-induced oxidative stress in RBC, inflammation and MCF-7 cells proliferation.

Salmonellosis is a considerable public health problem worldwide, with high economic importance in developed countries. The main purpose of this study was to determine the prevalence of Salmonella infection and antibiogram analysis of isolated strains in a cross-sectional study in Egypt 2016-2017. The study investigated twenty-eight Salmonella isolates from different areas in Egypt and different types of samples, such as human stool (9.3%), Egyptian cattle egrets and storks (28.5%) and grilled chicken from electric grills (36.6%). No isolates were detected from grilled chicken from charcoal grills or drinking water. The main Salmonella serotype detected in the isolates was S. typhimurium (86.5%). Molecular characterization of the invA gene by PCR was carried out and then confirmed by sequencing, and the results were submitted to GenBank. Antibiogram analysis of Egyptian isolates carried out on 9 antimicrobial discs reported that the routine regimes of treatment were not yet effective for recent new Salmonella generations in 2016-2017. The new isolates could be treated with levofloxacin, cefaperazone/sulbactam, chloramphenicol, imipenem or meropenem.
