Most traditional research on snake venoms has focused on front-fanged snake families (Viperidae, Elapidae, and Atractaspididae). However, venom is now generally accepted as being a much more broadly possessed trait within snakes, including species traditionally considered harmless. Unfortunately, due to historical inertia and methodological challenges, the toxin repertoires of non-front-fanged snake families (e.g., Colubridae, Dipsadidae, and Natricidae) have been heavily neglected despite the knowledge of numerous species capable of inflicting medically relevant envenomations. Integrating proteomic data for validation, we perform a de novo assembly and analysis of the Duvernoy's venom gland transcriptome of the Central American Road Guarder (Dipsadidae: Xenodontinae: Conophis lineatus), a species known for its potent bite. We identified 28 putative toxin transcripts from 13 toxin families in the Duvernoy's venom gland transcriptome, comprising 63.7% of total transcriptome expression. In addition to ubiquitous snake toxin families, we proteomically confirmed several atypical venom components. The most highly expressed toxins (55.6% of total toxin expression) were recently described snake venom matrix metalloproteases (svMMPs), with 48.0% of svMMP expression contributable to a novel svMMP isoform. We investigate the evolution of the new svMMP isoform in the context of rear-fanged snakes using phylogenetics. Finally, we examine the morphology of the venom apparatus using μCT and explore how the venom relates to autecology and the highly hemorrhagic effects seen in human envenomations. Importantly, we provide the most complete venom characterization of this medically relevant snake species to date, producing insights into the effects and evolution of its venom, and point to future research directions to better understand the venoms of 'harmless' non-front-fanged snakes.

Among the centipede of the chilopode Myriapoda class, the Cryptops genus is one of the most associated with accidents in humans in the metropolitan region of the state of Sao Paulo. To date, there is no study in the literature about Cryptops iheringi toxins. Thus, in this work a transcriptomic analysis of the C. iheringi venom gland was performed to obtain a profile of the toxins of this species. In addition, the crude venom was subjected to mass spectrometry analysis to establish an association between unknown sequences. These approaches for the construction of a general profile of the venom gland expression of this species led to the he identification of a Hemocyanin (Hc) subunit. Hemocyanins are copper-containing, respiratory proteins that occur in the haemolymph of many arthropod species. Here, we report the presence of Hc in the chilopode Myriapoda C. iheringi. Such respiratory proteins have long been considered unnecessary in Myriapoda, due to its tracheal systems. These respiratory proteins are potent immunogens, which induce the synthesis of large amounts of specific antibodies. Studies pointed out its interaction with polymorphonuclear monocytes and lymphocytes and in vitro tests have shown a potential anticancer activit, with in vitro significant inhibition of the growth of cancerous strains of the breast, pancreas and prostate. Currently scientific data is mostly limited to the study of native Hc of M. crenulata molluscs, therefore, the biotechnological potential of Hcs isolated from centipedes is still unexplored. Herein, the Hc sequence that was present in both proteome and transcriptome analysis have signal peptide and 76 kDa range. The Hc subunit sequence was synthesized with codon optimization for bacteria expression and the protein expressed as inclusion bodies. Refolding attempts provided soluble forms of Hc. At the moment efforts to access your biological activities are being carried out.

Objectives The presence of minority transmitted drug resistance mutations was assessed using ultra-deep sequencing and correlated with disease progression among recently HIV-1-infected individuals from Brazil. Methods Samples at baseline during recent infection and 1 year after the establishment of the infection were analysed. Viral RNA and proviral DNA from 25 individuals were subjected to ultra-deep sequencing of the reverse transcriptase and protease regions of HIV-1. Results Viral strains carrying transmitted drug resistance mutations were detected in 9 out of the 25 patients, for all major antiretroviral classes, ranging from one to five mutations per patient. Ultra-deep sequencing detected strains with frequencies as low as 1.6% and only strains with frequencies >20% were detected by population plasma sequencing (three patients). Venom proteins are added to reptile venoms through duplication of a body protein gene, with the duplicate tissue-specifically expressed in the venom gland. Molecular scaffolds are recruited from a wide range of tissues and with a similar level of diversity of ancestral activity. Transcriptome studies have proven an effective and efficient tool for the discovery of novel toxin scaffolds. In this study, we applied venom gland transcriptomics to a wide taxonomical diversity of advanced snakes and recovered transcripts encoding three novel protein scaffold types lacking sequence homology to any previously characterised snake toxin type: lipocalin, phospholipase A2 (type IIE) and vitelline membrane outer layer protein. In addition, the first snake maxillary venom gland isoforms were sequenced of ribonuclease, which was only recently sequenced from lizard mandibular venom glands. Further, novel isoforms were also recovered for the only recently characterised veficolin toxin class also shared between lizard and snake venoms. The additional complexity of snake venoms has important implications not only for understanding their molecular evolution, but also reinforces the tremendous importance of venoms as a diverse bio-resource.

The aim of this study was the isolation of the LAAO from Lachesis muta venom (LmLAAO) and its biochemical, functional and structural characterization. Two different purification protocols were developed and both provided highly homogeneous and active LmLAAO. It is a homodimeric enzyme with molar mass around 120 kDa under non-reducing conditions, 60 kDa under reducing conditions in SDS-PAGE and 60852 Da by mass spectrometry. Forty amino acid residues were directly sequenced from LmLAAO and its complete cDNA was identified and characterized from an Expressed Sequence Tags data bank obtained from a venom gland. A model based on sequence homology was manually built in order to predict its three-dimensional structure. LmLAAO showed a catalytic preference for hydrophobic amino acids (Km of 0.97 mmol/L with Leu). A mild myonecrosis was observed histologically in mice after injection of 100 μg of LmLAAO and confirmed by a 15-fold increase in CK activity. LmLAAO induced cytotoxicity on AGS cell line (gastric adenocarcinoma, IC50: 22.7 μg/mL) and on MCF-7 cell line (breast adenocarcinoma, IC50:1.41 μg/mL). It presents antiparasitic activity on Leishmania brasiliensis (IC50: 2.22 μg/mL), but Trypanosoma cruzi was resistant to LmLAAO. In conclusion, LmLAAO showed low systemic toxicity but important in vitro pharmacological actions. Elementos móveis influenciam expressivamente a evolução do grupo Bacteria, uma vez que são integrantes importantes do processo de geração da variabilidade genética e promovem a diversidade da população frente à estocasticidade do ambiente. Investigamos a influência de elementos genéticos móveis em linhagens de X. oryzae sob três diferentes aspectos nesta tese. Primeiramente, investigamos a transferência horizontal de um operon adquirido exclusivamente por Xoc, porém ausente em Xoo e linhagens filogenéticamente próximas. A importância da produção do fitormônio GA4 por Xoc é indicada pela alta prevalência do operon em diferentes populações, e pela ampla distrubuição geográfica em um amplo perído de tempo. Também abordamos a influência de elementos genéticos móveis mais amplamente no genoma de X. oryzae. Descrevemos como elementos de transposição, representados por elementos do tipo IS, são capazes de interferir em um sistema central do metabolismo bacteriano, como o sistema de reparo de DNA. Por fim, descrevemos um novo elemento encontrado em X. oryzae e analisamos sua organização, distribuição, filogenia, o nível transcricional e o potencial funcional na interação com seu hospedeiro Nosso conjunto de resultados reitera a importância dos elementos de transposição no aumento de variabilidade genética de populações, e dessa forma, infere sobre seu papel relevante na diversificação em X. oryzae. Descrevemos a variabilidade, exploramos as consequências da amplificação de elementos de transposição nesta espécie, evidenciamos a complexidade desta variação em nível populacional. Mobile elements are genomic components that trigger genetic variability and promote the diversity of the population against the stochasticity of the environment and therefore, significantly influence the evolution of the Bacteria group. We investigate the influence of mobile genetic elements on X. oryzae lineages under three different aspects in this thesis. First, we investigated the horizontal transfer event of a operon, acquired exclusively by Xoc, but absent in Xoo and phylogenetically close lineages. The importance of the production of the phytohormone GA4 by Xoc is indicated by its high prevalence in different populations, in a wide geographic distribution and in a large time period.We also address the influence of mobile genetic elements more comprehensively in the X. oryzae genome. We describe how transposable elements, represented by IS elements, are capable of interfering in a central system of bacterial metabolism, such as the DNA repair system. Finally, we describe a new element found in X. oryzae and we analyze its organization, distribution, phylogeny and its transcriptional level. We also speculate its function in the X. oryzae-host interaction. Our results reiterate the importance of transposable elements enriching genetic variability of populations and infers on their relevant role in the diversification of X. oryzae lineage. We describe the variability, explore the consequences resulting from the amplification of transposition elements in the X. oryzae species and we show the complexity of this variation at the population level.

A Bothrops jararaca é a serpente de maior importância médica no Brasil. Vários estudos foram realizados com o objetivo de caracterizar os componentes do veneno de serpentes, entretanto, a base molecular dos genomas das serpentes é pouco conhecida. Assim, foi realizado o sequenciamento e montagem do genoma da serpente Bothrops jararaca. Foram construídas bibliotecas tipo shotgun e mate-pair para realização de corridas de sequenciamento usando a tecnologia Illumina e sequências complementares foram obtidas em equipamento PACBIO RS II. Uma biblioteca de BACs também foi construída e 768 pools de 12 BACs foram sequenciados. Um grande conjunto de segmentos genômicos foi obtido e foi possível identificar genes de várias toxinas, entre elas SVMPs, SVSPs, BPPs, CRISPs e VEGF. Ainda foi possível depreender o contexto genômico de muitos destes genes e identificamos os principais elementos repetitivos genômicos. Estes achados são relevantes para o entendimento da função e evolução do sistema venenífero e podem servir de base para outros estudos futuramente. The pit viper Bothrops jararaca is the most medically important snake in Brazil. Several studies were conducted in order to characterize the components of snake venom. However, the molecular basis of snake genomes is poorly known. Hence, it was carried out the sequencing and assembly of the Bothrops jararaca snake genome. Shotgun and mate-pair libraries were constructed to perform sequencing runs using Illumina technology and complementary sequences were obtained in PACBIO RS II equipment. A BAC library was also constructed and 768 pools of 12 BACs were sequenced. A large number of genomic segments was obtained. It was possible to identify genes of several toxins, including SVMPs, SVSPs, BPPs, CRISPs and VEGF. In addition, it was possible to infer the genomic context related to most of these genes and identify the main genomic repetitive elements. These findings are relevant for understanding the function and evolution of the venom system and it provides the basis for further studies.

O escorpião Tityus serrulatus é o responsável pelos acidentes mais graves no Brasil. Dentre os componentes já estudados de sua peçonha estão neurotoxinas capazes de interagir com canais iônicos, além de peptídeos biologicamente ativos também estarem presentes. Neste estudo foram isolados peptídeos da fração de baixa massa molecular da peçonha que interagiram com as oligopeptidases timet oligopeptidase (EP24.15) e neurolisina (EP24.16) através do emprego de substratos fluorescentes específicos como ferramentas. Usando espectrometria de massas, as sequências KEILG, FTR, YLPT e do análogo KELLG foram determinadas e posteriormente sintetizadas. In vitro, os peptídeos não foram substratos para enzimas já citadas, além de testes com a neprelisina e ECA. Em relação à inibição, os destaques ficam por conta de KELLG e KEILG, capazes de inibirem a EP 24.15 e de não inibirem a EP 24.16. Outro destaque foi o YLPT, apresentando um Ki de 0,94 mM perante a neprilisina. In vivo os peptídeos foram testados em relação à nocicepção, rolamento de leucócitos e reatividade vascular, onde se destacou o FTR, apresentando efeito antinociceptivo e o KEILG, capaz de aumentar o número de leucócitos, ressaltando a importância de pequenas moléculas na composição da peçonha. The scorpion Tityus serrulatus is responsible for the most serious accidents in Brazil. Among the components already studied in its venom, neurotoxins are able to interact with ion channels, and peptides with biological activity are also present. In this study, peptides from low molecular weight fraction of the venom that interacted with the oligopeptidases thimet oligopeptidase (EP24.15) and neurolysin (EP24.16) were isolated using specific fluorescent substrates as a tool. Sequences from the peptides KEILG, FTR, YLPT and the analogue KELLG were determined by mass spectrometry and subsequently synthesized. In vitro, the peptides were not substrates for those enzymes, neither for neprilysin and ACE enzymes. Concerning inhibition, the highlights are the KELLG and KEILG, which were able to inhibit EP 24.15 but not EP 24.16. Another peptide thats worth to mention is YLPT, with a Ki of 0.94 mM for neprilysin. In vivo, the peptides were tested for nociception, rolling of leukocytes and vascular reactivity, being the FTR distinguished to be able to cause antinociceptive effect and KEILG to increase the number of leukocytes. These results emphasize the importance of small molecules in the scorpion venom constitution.

A glândula de veneno da serpente Bothrops jararaca é uma glândula exócrina relacionada a glândula salivar dos mamíferos. Diferentemente de outras glândulas exócrinas, esta possui um lúmen central no qual o veneno produzido fica estocado. Os mecanismos envolvidos na regulação da síntese e secreção de toxinas pela glândula de veneno são pouco conhecidos. Sabe-se que a inervação noradrenérgica possui um papel essencial no ciclo de produção de veneno, pois serpentes Bothrops jararaca tratadas com reserpina, um potente bloqueador da atividade simpática, não acumulam veneno no lúmen. Porém a ativação direta dos adrenoceptores α e β, através da ação de agonistas, tem a capacidade de reverter a ação da reserpina. No presente trabalho utilizamos métodos combinados de análise de expressão gênica em larga escala a fim de identificar os processos celulares sob controle do sistema simpático durante o ciclo de produção de veneno da glândula de veneno de Bothrops jararaca. Foi construído um array de cDNA em membrana da náilon contendo 4608 clones provenientes da biblioteca de cDNA construída a partir das glândulas de veneno de um macho e uma fêmea, adultos, de Bothrops jararaca. Para a análise temporal da expressão gênica foram utilizados machos adultos de B. jararaca. As glândulas de veneno foram extraídas em diferentes dias do ciclo de produção de veneno (0, 1, 2, 4 e 15 dias). Através da análise do perfil de expressão gênica identificamos que os transcritos de toxinas e de não toxinas (celulares) possuem perfil semelhante de expressão ao longo do ciclo, sendo que no 2° dia do ciclo ocorre o pico de expressão desses transcritos. Para identificar os processos celulares sob controle da inervação noradrenérgica machos adultos de B. jararaca foram submetidos a tratamento farmacológico com reserpina (glândula 4dR) e com reserpina e agonistas dos adrenoceptores α e β (glândula 4dA). Na análise da expressão gênica utilizando macroarranjos, entre os clones com expressão aumentada na glândula 4dR, aproximadamente 51% eram de toxinas, indicando que a inibição da atividade simpática não interfere na transcrição das toxinas. A análise dos transcritos celulares confirmou que os processos de transcrição e tradução não são afetados pelo tratamento com reserpina. A análise da expressão por PCR quantitativo em tempo real, confirmou que as toxinas são expressas normalmente na glândula 4dR. Além disso, a análise da expressão de genes envolvidos nos processos de enovelamento protéico e secreção revelou que genes responsivos a estresse de retículo endoplasmático apresentam aumento na expressão na glândula 4dR. Também realizamos a análise transcriptômica por sequenciamento em larga escala (RNA-seq) da glândula 4d e da glândula 4dR. Entre os contigs identificados como toxinas não houve diferenças quantitativas nem qualitativas significativas entre as glândulas 4d e 4dR, confirmando que o processo de transcrição de toxinas ocorre independentemente da ativação dos adrenoceptores α e β. A análise de enriquecimento de termos do gene ontology revelou predominância de processos biológicos relacionados a resposta a estresse de retículo endoplasmático entre os transcritos mais expressos na glândula 4dR e de processos envolvendo a formação de vesículas de transporte entre os transcritos menos expressos na glândula 4dR. Na análise dos transcritos exclusivos da glândula 4d e exclusivos da glândula 4dR identificamos diversas isoformas de small GTPases da família Ras (Rab) que possuem papel fundamental na regulação da formação de vesículas. Assim, nesse trabalho mostramos que o processo de transcrição de toxinas ocorre independentemente da ativação dos adrenoceptores α e β e que a ativação dos adrenoceptores parece ser necessária para que ocorra a formação de vesículas secretoras. Já a inibição do processo pela ação da reserpina possivelmente provoca a ativação da resposta UPR (unfolded protein response), o que pode estar associado com o acúmulo de proteínas no lúmen do retículo endoplasmático. The venom gland of the Brazilian venomous snake Bothrops jararaca (Crotalinae, Viperidae) is an exocrine tissue related to the salivary gland. The venom gland has a central lumen where the venom is stored. When the venom is released, the production of new venom is triggered by the activation of noradrenaline on both α1- and β-adrenoceptors. But the genes involved and the regulation of venom production cycle are poorly known. When the Bothrops jararaca is treated with reserpine, a depletor of catecholamine, the venom production is inhibited. At present work we used combined methods of high throughput analysis of gene expression to identify cellular process controlled by the sympathetic system during the cycle of venom production in the venom glands of Bothrops jararaca. Was constructed a cDNA array with 4608 clones from a cDNA library of the venom glands of one male and one female of B.jararaca. In order to get a time series analysis adult males of B.jararaca were used. The venom gland was extracted at different time points of the venom production cycle (0, 1, 2, 4 and 15 days). The resulting profile of the gene expression of toxins and non-toxins during the cycle was shown to be similar, and the higher level of gene expression was found at the 2nd day of the venom production cycle. A differential gene expression analysis was performed also with venom glands of B.jararaca treated with reserpine. Although previous results reported that venom glands under effect of reserpine are not able to produce venom, we found that 51% of upregulated clones of the venom gland treated with reserpine (4dR) are toxins. Moreover, most of the non-toxins clones were involved with transcription and translation processes, showing that these processes are not affected by the inhibition of the sympathetic system. The analysis of gene expression by quantitative real-time PCR confirmed that in the venom gland treated with reserpine (4dR) the toxins are normally produced, and the genes responsive for unfolded protein response (UPR) are upregulated. We also performed the next generation sequencing to produce a transcriptomic profile (RNA-seq) of normal venom gland (4d) and venom gland treated with reserpine (4dR). The comparison between the transcripts of toxins found at 4d and 4dR transcriptomes revealed no qualitative or quantitative differences, confirming that the toxins transcription are independent of the activation of α1- and β-adrenoceptors. The enrichment analysis of Gene Ontology terms revealed that the unfolded protein response are activated at 4dR venom gland and the membrane trafficking are possibly inhibited. We also identify many isoforms of Ras family small GTPases (Rab proteins) that has a key role ate regulation of membrane trafficking. At present work we showed that the transcriptional control of the toxins in the venom gland are independent of the activation by α1- and β-adrenoceptors, however, the adrenoceptors seems to be necessary to activate the secretory pathway. The inhibition of the activation of α1- and β-adrenoceptor by reserpine seems to be recruiting an unfolded protein response, probably due to the accumulation of proteins at the lumen of the endoplasmic reticulum.

O veneno da serpente Crotalus durissus terrificus e seus componentes desencadeiam importantes efeitos biológicos que envolvem direta e/ou indiretamente, componentes do sistema circulatório. Contudo, não há estudos específicos na literatura sobre os efeitos do veneno crotálico ou de suas toxinas, em células endoteliais. As células endoteliais constituem a camada de revestimento interna dos vasos sanguíneos, denominada endotélio. Este tecido é metabolicamente ativo, com função protetora do sistema cardiovascular e desempenha papel central na regulação da função circulatória, através do controle da coagulação, permeabilidade e do tônus vascular. Neste contexto, este estudo teve como objetivo avaliar os efeitos do veneno de Crotalus durissus terrificus (VCdt), do seu componente majoritário, a crotoxina (CTX) e de suas subunidades, fosfolipase A2 (CB) e crotapotina (CA), sobre células endoteliais, em cultura, quanto à: i) viabilidade e proliferação celular; ii) integridade das monocamadas; iii) produção de óxido nítrico, de prostaciclina e mecanismos envolvidos neste efeito. Os resultados obtidos demonstram que o veneno de Crotalus durissus terrificus afetou a viabilidade e a integridade de células endoteliais em cultura, de modo tempo-dependente e apenas na maior concentração, sugerindo sua baixa toxicidade sobre as células endoteliais. A subunidade CB, mas não a CTX nem a crotapotina, reproduziu os efeitos causados pelo VCdt. Em concentrações não citotóxicas, tanto o veneno quanto as toxinas não alteraram a proliferação celular nem a produção basal de óxido nítrico pelas células endoteliais. Por outro lado, o veneno e a subunidade CB, mas não a CTX nem a CA causaram aumento significativo da produção de prostaciclina, via COX-1 e COX-2, sendo que a expressão protéica da isoforma COX-2 foi induzida por estes agentes. Além disso, foi demonstrado que a fosfolipase citosólica é relevante para o aumento da produção de prostaciclina, induzido pela CB. Adicionalmente, foi demonstrado que a atividade catalítica da subunidade CB é essencial para os efeitos descritos. Isto reforça a sugestão de que a subunidade fosfolipásica, isoladamente, possa contribuir para os efeitos do veneno total no endotélio. Nesse sentido, se houver alguma fração desta enzima na sua forma livre, no veneno total, sugere-se que ela contribua, de modo significativo, para os efeitos do veneno de Crotalus durissus terrificus no endotélio. Crotalus durissus terrificus snake venom (CdtV) and their components induces systemic effects, which interfere with blood vessel system. Endothelial cells (EC) are central elements for haemostasis, regulating blood vessel-wall permeability, blood fluidity and adhesion properties of circulating leukocytes. However, there is no available data on the effects of this venom and its components on endothelial cells. In this study, the effects of CdtV, crotoxin (CTX) which is formed by two distinct subunits named crotapotin (CA) and phospholipase A2 (CB), on endothelial cells in vitro were investigated, analyzing EC viability and proliferation, EC monolayers integrity, release of both nitric oxide and prostacyclin (PGI2). CdtV, at the highest concentration, time-dependently decreased the viability of EC and the integrity of cell monolayers. The CB subunit, but not CTX nor CA, reproduced the effects caused by crude venom. In contrast, neither EC proliferation nor release of oxide nitric were affected by non-cytotoxic concentrations of CdtV or isolated toxins. However, at the same experimental condition, both CdtV and CB increased the prostacyclin release by endothelium through activation of COX-1 and -2 enzyme systems. Moreover, these toxins upregulated protein expression of COX-2 isoform, but did not alter constitutive expression of COX-1. On the other hand, neither CTX nor CA affected basal production of PGI2. Inhibition of cytosolic PLA2 (cPLA2) by AACOCF3 significantly reduced PGI2 increments caused by both CdtV and CB implying that cPLA2 cooperates for the synthesis of PGI2 induced by them. Inhibition of the catalytic activity of CB abrogated its ability to induce the release of PGI2, thus suggesting the importance of the phospholipase A2 enzyme activity for this effect. These findings provide evidence that CdtV and CB can directly activate EC and up-regulate cyclooxygenase pathways for production of prostacyclin, an important mediator of vasodilation and inflammation. Moreover, CB through its catalytic activity may significantly contribute for the stimulatory effect of CdtV in EC. Therefore, these findings indicate novel regulatory mechanisms for both CdtV and venom secretory PLA2 in endothelial cells.

O sistema complemento parece ter surgido com o aparecimento de C3 e fator B (FB), os únicos componentes encontrados nos organismos mais primitivos, o que sugere que a via alternativa seja a mais antiga. Fragmentos de cDNA codificantes para FB (Lox-FB) e C3 (Lox-C3) foram sintetizados a partir do RNA total isolado da glândula de veneno de Loxosceles laeta e amplificados por técnicas de RACE-PCR. Lox-FB apresenta uma organização de domínios clássica, composta por domínios CCP, vWFA e de serino protease. Os aminoácidos envolvidos na ligação ao C3b são conservados, no entanto, a tríade catalítica clássica não foi encontrada. Lox-C3 apresenta uma configuração de domínios similar a do C3 humano, contendo dois sítios putativos de processamento: um entre as cadeias α e γ, e outro entre as cadeias α e β, indicando que Lox-C3 seja composto por três cadeias. As análises filogenéticas indicaram que Lox-C3 e Lox-FB são mais próximos evolutivamente aos componentes equivalentes da aranha Hasarius adansoni, com valores de identidade de 53% e 43%, respectivamente. The complement system seems to have arisen with the appearance of C3 and factor B (FB), the only components found in the most primitive organisms, suggesting that the alternative pathway is the oldest. cDNA fragments coding for the complement factor B (Lox-FB) and C3 (Lox-C3) were synthesized from total RNA Loxosceles laeta venom gland and amplified using RACE-PCR techniques. Lox-FB has a classical domain organization in mosaic, composed by CCPs, vWFA and serine protease domains. The amino acids involved in binding to C3b are conserved, however, the classical calatytic triad was not found. Lox-C3 presents a similar configuration of domains to C3 human and has two putative processing sites: the first one is located between α and γ chains and other between α and β chains, indicating that Lox-C3 is composed by three chains. The phylogenetic analyses indicated that Lox-C3 and Lox-FB are evolutionary closer to the equivalent components of Hasarius adansoni, with identity values of 53% and 43%, respectively.

Spiders and scorpions are efficient predators arachnid (PA) consuming preys larger than themselves. Few studies reported, molecularly, the digestion in PA. This work describes a biochemical, transcriptomic and proteomic analysis of the midgut and midgut glands (MMG) and digestive juice (DJ) from Nephilengys cruentata and Tityus serrulatus MMG. Cathepsin L, B, D and F, legumain, trypsin, astacin, carbohydrases and lipases were identified by these approaches. Peptide isomerase and ctenitoxins, which are venom proteins were identified, showing a correlation among digestive and venom enzymes. Summarily, PA relies in multi peptidase system mainly constituted of astacins for extracellular prey liquefaction and cathepsin L for intracellular digestion, describing a molecular model for digestion. Probably, during evolution, gene duplication led a diversification of astacins in the derived groups of Arachnida, like spiders, distinctly from what is observed in basal groups like scorpions. These data on Arachnida digestion will allow detailed multi disciplinary studies. Aranhas e escorpiões são aracnídeos predadores eficientes (AP) consumindo presas maiores que eles mesmos. Poucos estudos descrevem molecularmente a digestão em AP. Neste trabalho caracterizamos bioquimicamente, por transcriptoma e proteoma o intestino e glândulas digestivas (IGD) e suco digestivo (SD) de Nephilengys cruentata e o IGD de Tityus serrulatus. Catepsinas L, B, D e F, legumaína, tripsinas, astacinas, carboidrases e lipases foram identificadas. Peptídeo isomerase e ctenitoxina foram identificadas no IGD. Estas proteínas podem indicar uma correlação entre enzimas digestivas e do veneno. Portanto, AP apresentam várias peptidases principalmente astacinas para liquefazer a presa extraoralmente e catepsinas L para digestão intracelular, descrevendo um modelo molecular para a digestão. Provavelmente, durante a evolução, eventos duplicação gênica levaram à diversificação das astacinas aracnídeos derivados, como as aranhas, diferentemente dos grupos basais, como os escorpiões. Estes dados sobre a digestão em Arachnida permitirão estudos multidisciplinares.

Análise proteômica das glândulas de veneno e acessória durante o ciclo de produção de veneno e a influência da inervação noradrenérgica na síntese de proteínas foi verificada. Nossos dados mostram que a síntese de proteínas envolvidas no processo de síntese e secreção de proteínas está aumentada nos estágios ativados da glândula de veneno e que a estimulação da inervação noradrenérgica regula a síntese de algumas proteínas importantes para estes processos. Mostramos pela primeira vez, a presença de várias toxinas no estágio quiescente da glândula e que a produção e secreção de toxinas ocorrem de maneira não sincronizada. Na glândula acessória verificamos uma modulação na síntese de algumas proteínas da glândula após a extração de veneno. Mostramos também a presença de várias toxinas e de inibidores de enzimas do veneno nesta glândula. A composição proteômica das glândulas do aparelho glandular de veneno de serpente contribuirá para um melhor conhecimento dos mecanismos de produção e secreção de veneno e para os estudos das toxinas e suas diversidades. Proteomic analysis of venom gland and accessory gland during venom production cycle and the influence of noradrenergic innervation on protein synthesis of venom gland were verified. Our data show that the synthesis of proteins involved in the process of synthesis and secretion of proteins is increased in activated stages of venom gland and the sympathetic outflow regulates the synthesis of some proteins that is important to this process. For the first time we showed that many toxins are present in quiescent stage of venom gland and the production and secretion of toxins is not synchronized. Regarding the accessory gland, we verify that the syntheses of some protein of gland were regulated after venom extraction. We also showed the presence of some toxins of the venom and enzyme inhibitors in this gland. The proteomic composition of snake venom gland apparatus will contribute to better understand the mechanisms involved in venom gland activation and consequently, venom production. These data will also contribute to the studies on snake toxins and their diversities.

A peçonha das anêmonas do mar é uma fonte de compostos bioativos, incluindo toxinas peptídicas que são ferramentas para o estudo da estrutura e função dos canais voltagem dependentes de K+ (KV). Neste trabalho, quatro neurotoxinas foram purificadas da peçonha das anêmonas do mar Actinia bermudenesis e Bunodosoma caissarum. AbeTx1 e BcsTx4 possuem um motivo estrutural semelhante à das \"kappa-toxinas\" e análises funcionais e estruturais permitiram concluir que são os primeiros membros de um novo (tipo 5) de neurotoxinas de anêmonas do mar que atuam em canais KV. Por sua vez, a similaridade estrutural das toxinas BcsTx 1 e BcsTx2 nos permitiu inferir que estas são membros do já descrito tipo 1 (subtipo 1b) de neurotoxinas de anêmona que também atuam em canais KV. A caracterização funcional foi realizada utilizando-se diferentes subtipos de canais KV, expressos em ovócitos de Xenopus laevis e as medidas eletrofisiológicas foram feitas empregando-se a técnica de \"voltage-clamp\" com dois microelétrodos. AbeTx1, BcsTx1 e BcsTx2 (3 μM) apresentaram uma seletividade de atividade para os subtipos de KV1.1-KV1.3, KV1.6 e Shaker IR, ao passo que a BcsTx4 (3 μM) é somente capaz de bloquear a corrente dos subtipos de KV1.1, KV1.2 e KV1.6. Os mecanismos de ação envolvidos na seletividade da atividade e na potência com que estas se ligam aos seus alvos biológicos foram discutidos com base nos resultados obtidos e análises fisiológicas permitiram propor que estas toxinas atuam como \"armas\" para defesa contra predadores e/ou para captura de presas The sea anemones venom is a rich source of bioactive compounds, including peptide toxins which are tools for studying the structure and function of voltage-dependent channels K+ (KV). In this work, four neurotoxins were purified from the venom of the sea anemones Actinia bermudenesis and Bunodosoma caissarum. AbeTx1 and BcsTx4 have a structural motif similar to that of kappa-toxins and functional and structural analysis showed that they are the first members of a new type (type 5) of sea anemone neurotoxins acting on KV channels. Moreover, the structural analysis of BcsTx1 and BcsTx2 toxins allowed us to conclude that they are members of the previously described type 1 (subtype 1b) of sea anemone neurotoxins. Functional characterization was performed by means of a wide electrophysiological screening on different KV channels using oocytes of Xenopus laevis and electrophysiological measurements were performed employing the voltage-clamp technique. AbeTx1, BcsTx1 and BcsTx2 (μM) showed a selective activity for KV1.1-KV1.3, KV1.6 and Shaker IR, while BcsTx4 (3 μM) only blocks KV1.1, KV1.2 and KV1.6. The mechanisms involved in potency and selectivity were discussed based on the results obtained and physiological analyses have provided new insights on the role of these toxins in the physiology of the sea anemones

A partir de uma biblioteca de cDNA de glândulas de veneno de Micrurus corallinus (cobra-coral), uma serpente da Família Elapidae bastante representada no Brasil e muito comum em áreas florestais tropicais, foram gerados 1.438 Expressed Sequences Tags (ESTs), agrupados em 611 clusters. O banco representa os genes mais expressos na glândula de veneno de M. corallinus. Os transcritos relacionados às toxinas apresentaram ao redor de 46% de representação nesse banco de seqüências. A composição geral das toxinas inclui: toxinas de três dígitos (3FTx) (24%), fosfolipases A2 (PLA2) (16%), lectinas do tipo C (5%), entre outros. O banco permitiu não somente a identificação de possíveis toxinas, mas também de transcritos celulares, sendo a maioria envolvida nas funções fisiológicas de células da glândula de veneno. A maior parte dessas moléculas apresenta um envolvimento na expressão gênica e protéica, o que reflete uma alta especialização do tecido para a síntese de toxinas. A análise do transcriptoma de glândulas de veneno de M. corallinus possibilitou a identificação de alguns candidatos antigênicos para um anti-soro antielapídico alternativo. Cinco candidatos antigênicos foram selecionados por meio da análise do transcriptoma obtido: Atg1 (Grupo das neurotoxinas Homolog 8), Atg2 (Grupo das neurotoxinas Homolog 7/3/1), Atg3 (Outras neurotoxinas 1), Atg4 (Outras neurotoxinas 2) e Atg5 (fosfolipase do tipo A2). Avaliamos a viabilidade de imunização com o DNA desses candidatos. Para isso, os cinco grupos de antígenos foram clonados, primeiramente em pGEM-T e, posteriormente, em pSecTag2A, que é um vetor de expressão em células de mamíferos. As clonagens foram inicialmente testadas em células do tipo COS (transfecção transiente), entretanto não ficou clara a capacidade dessas células em expressar os antígenos. Para a análise da resposta imunológica da vacina de DNA, proteínas recombinantes produzidas em E. coli foram utilizadas para o coating de ELISA para detectar anticorpos presentes no soro primário proveniente da imunização com DNA. Os resultados mostraram que o soro dos animais imunizados foi capaz de reconhecer os antígenos recombinantes. Isso indica que a imunização por DNA em camundongos poderia ser uma boa alternativa em relação à imunização do veneno puro de serpente, que é custosa e muito depende da disponibilidade do veneno. Apesar da necessidade de testes complementares, esse é um resultado promissor, já que a produção de anticorpos pode ser alcançada por via de imunização intramuscular, mais prática para objetivos de produção. Micrurus corallinus(coral snake) is a tropical forest snake belonging to the Elapidae Family, and is very common in Brazil. From the cDNA library of its venom glands, 1.438 Expressed Sequences Tags (ESTs) were generated and grouped into 611 clusters. This database contains the most expressed genes in the M. corallinus venom glands. The transcripts related to toxins represent approximately 46% of the total genes in this database. The toxin compound consists of: three finger toxins (24%), phospholipases A2 (PLA2s) (16%), type-C lectins (5%), among others. This database allowed not only the identification of possible toxins, but also the identification of cellular transcripts, most of which seems to be involved in physiological functions of venom gland cells. The majority of these molecules are involved in gene and protein expression, revealing the high level of specialization of the tissue for toxin synthesis. The analysis of the M. corallinus venom gland transcriptome allowed the identification of some antigenic candidates for an alternative antielapidic antiserum. Five antigenic candidates were selected after analysing the transcriptome: Atg1 (Homolog group 8), Atg2 (Homolog group 7/3/1), Atg3 (Other neurotoxins 1), Atg5 (A2-type phospholipase). These five antigenic groups were used for DNA immunization. Then they were first cloned in pGEM-T and, after, in pSecTag2A, which is an expression vector in mammal cells. The cloning was tested in COS-type cells (transient transfection), without signs of expression. To analyze the immunological response, recombinant proteins were produced in E. coli and used for ELISA coating to react with the primary serum deriving from the DNA immunization. The results showed that the serum from the immunized animals was able to recognize the recombinant antigens, indicating that the DNA immunization in mice could be a feasible alternative regarding the traditional immunization with crude snake venom, which is costly and heavily dependent on the availability of the venom. Regardless the need for additional tests, this is a promising result, because the antibody production can be achieved by intramuscular immunization, a more effective method when aiming for downstream production.
