Near-infrared (NIR) spectroscopy is a powerful non-invasive technique for assessing the optical properties of human tissues, capturing spectral signatures that reflect their biochemical and structural characteristics. In this study, we investigated the use of NIR reflectance spectroscopy combined with chemometric analysis to distinguish between patients with Essential Tremor (ET) and healthy individuals. ET is a common movement disorder characterized by involuntary tremors, often making it difficult to clinically differentiate from other neurological conditions. We hypothesized that NIR spectroscopy could reveal unique optical fingerprints that differentiate ET patients from healthy controls, potentially providing an additional diagnostic tool for ET. We collected NIR reflectance spectra from both extracranial (biceps and triceps) and cranial (cerebral cortex and brainstem) sites in ET patients and healthy subjects. Using Partial Least Squares Discriminant Analysis (PLS-DA) and Partial Least Squares (PLS) regression models, we analyzed the optical properties of the tissues and identified significant wavelength peaks associated with spectral differences between the two groups. The chemometric analysis successfully classified subjects based on their spectral profiles, revealing distinct differences in optical properties between cranial and extracranial sites in ET patients compared to healthy controls. Our results suggest that NIR spectroscopy, combined with machine learning algorithms, offers a promising non-invasive method for the in vivo characterization and differentiation of tissues in ET patients. Studying the interaction between pesticide contamination in the plant system and the dissolved organic matter (DOM) composition is important to understand the impact of pesticides and plants on the ecological function of DOM. The present study investigated the effects of DOM on the bioaccumulation and biotransformation of carbamates in plants, carbamate exposure on DOM composition, and plant root secretion on the interaction between DOM and carbamates. The concentrations of carbamates and their metabolites in living cabbage plants were continuously tracked through an in vivo analytical method. The presence of DOM was found to reduce the highest bioconcentrations and shorten the time it took to reach the highest bioaccumulated amounts of isoprocarb and carbofuran in plants, while it showed no significant effect on the uptake behavior of carbaryl. DOM profiling results indicated that carbamate exposure substantially decreased the number and molecular diversity of DOM. Notably, plant root secretion alleviated carbamate-induced DOM molecular alterations by inducing a higher turnover rate of DOM compared to that in the uncontaminated group, highlighting the role of plants in mitigating the effects of exogenous pesticide exposure on DOM composition and maintaining DOM molecular homeostasis. Abstract Caffeic acid phenethyl ester (CAPE) and its derivatives exhibit considerable effects against hepatocellular carcinoma (HCC), with unquestioned safety. Here we investigated CAPE derivative 1ʹ (CAPE 1ʹ) monotherapy to HCC, compared with sorafenib. HCC Bel-7402 cells were treated with CAPE 1ʹ, the IC50 was detected using CCK-8 analysis, and acute toxicity testing (5 g/kg) was performed to evaluate safety. In vivo, tumor growth after CAPE 1ʹ treatment was evaluated using an subcutaneous tumor xenograft model. Five groups were examined, with group 1 given vehicle solution, groups 2, 3, and 4 given CAPE 1ʹ (20, 50, and 100 mg/kg/day, respectively), and group 5 given sorafenib (30 mg/kg/day). Tumor volume growth and tumor volume-to-weight ratio were calculated and statistically analyzed. An estimated IC50 was 5.6 µM. Acute toxicity tests revealed no animal death or visible adverse effects with dosage up to 5 g/kg. Compared to negative controls, CAPE 1ʹ treatment led to significantly slower increases of tumor volume and tumor volume-to-weight. CAPE 1ʹ and sorafenib exerted similar inhibitory effects on HCC tumors. CAPE 1ʹ was non-inferior to sorafenib for HCC treatment, both in vitro and in vivo. It has great potential as a promising drug for HCC, based on effectiveness and safety profile.

The current synthesis of benorilate typically involves the Schotten-Baumann esterification, which generates harmful gases HCl and SO2 from the reagent SOCl2, posing environmental concerns. Moreover, the process entails multiple steps. In this study, we have optimized a one-step method using EDCI/4-DMAP esterification for the synthesis of benorilate. The product’s structure and purity were characterized through melting point determination, NMR, and HPLC-MS analysis, followed by in vivo dynamic evaluation in rats based on plasma and bile samples. Implantable electrode-based in vivo electrochemical sensing research has consistently played a pivotal role in driving advancements in analytical methods within the field of neurochemical research. The intricacies of brain structures and the real-time nature of various physiological activities impose rigorous demands on analytical approaches. High specificity, elevated sensitivity, robust interference resistance, and favorable biocompatibility are essential prerequisites for in situ analysis in living systems. With the development of novel functional materials and advancements in electrochemical research methodologies, there has been significant progress in the field of in vivo electrochemical analysis employing implantable electrodes. Nonetheless, this field continues to encounter a series of challenges. This article reviews the current applications of implantable electrodes, such as enzymemodified electrodes, aptamer-modified electrodes, ion-selective electrodes, liquid-liquid interface-based electrodes, and ion current rectifying electrodes in the field of in vivo electrochemical sensing, as well as the major progress in the research of their key properties such as biocompatibility and sensitivity. The study aimed to synthesis biocomposite material that was more bioactive and could be employed as a graft during bone formation. Cordierite was fabricated via sintering a mixture of 51.4% silicon oxide, 34.8% aluminum oxide, and 13.8% magnesium oxide at 1400 °C. The produced substance was identified using X-ray Diffraction (XRD), the patterns showed that just indialite was identified as a single phase. To formulate sample with supreme biological behavior, cordierite was added to hydroxyapatite in 30 wt% and then sintered at 1100 °C. Following, XRD reveals the emergence of the Mg-whitlockite phase. Scanning Electron Microscope (SEM) had been employed to investigate their morphology and the effect of treatment temperatures. Fourier-transform infrared spectroscopy (FTIR) employed to assess if there is chemical reaction between cordierite and hydroxyapatite was tack placed. Prepared ceramic powder used as reinforcing phase with 30, 40 and 50 wt% in PCL matrix to formulate biocomposite system. All samples were inspected via SEM to determine their microstructure. FTIR showed there is no chemical reaction occurred between PCL and ceramic powder and it was just a physical blending. Energy Dispersive X-ray (EDX) showed that Z40 exhibits the better composition, as compared with other samples, in terms of Ca/P ratio. Z40 hybrid composite promotes the better results in terms of microstructure and chemical composition. Also, employed better in vitro behavior represented by degradation rate that equal to 39.86%. Abstract Background Excessive range of tibial rotation (rTR) may be a reason why athletes cannot return to sports after ACL reconstruction (ACLR). After ACLR, rTR is smaller in reconstructed knees compared to contralateral knees when measured during low-to-moderate-demand tasks. This may not be representative of the amount of rotational laxity during sports activities. The purpose of this study is to determine whether rTR is increased after ACL injury compared to the contralateral knee and whether it returns to normal after ACLR when assessed during high-demand hoptests, with the contralateral knee as a reference. Methods Ten ACL injured subjects were tested within three months after injury and one year after reconstruction. Kinematic motion analysis was conducted, analysing both knees. Subjects performed a level-walking task, a single-leg hop for distance and a side jump. A paired t-test was used to detect a difference between mean kinematic variables before and after ACL reconstruction, and between the ACL-affected knees and contralateral knees before and after reconstruction. Results RTR was greater during high-demand tasks compared to low-demand tasks. Pre-operative, rTR was smaller in the ACL-deficient knees compared to the contralateral knees during all tests. After ACLR, a greater rTR was seen in ACL-reconstructed knees compared to pre-operative, but a smaller rTR compared to the contralateral knees, even during high-demand tasks. Conclusion The smaller rTR, compared to the contralateral knee, seen after a subacute ACL tear may be attributed to altered landing technique, neuromuscular adaptation and fear of re-injury. The continued reduction in rTR one year after ACLR may be a combination of this neuromuscular adaptation and the biomechanical impact of the reconstruction. Occult HBV infection (OBI) is a special infection status during Hepatitis B virus (HBV) infection. The underlying mechanism of its occurrence remains unclear. This study conducted sequencing analysis on 104 OBI plasma samples and 524 HBsAg positive samples from 29 blood centres, and searched for high-frequency mutations in transmembrane domain (TMD) of S protein in the OBI population. Plasmids with TMD high-frequency mutations were constructed, in vivo and in vitro functional experiments were performed to investigate possible molecular mechanisms of OBI occurrence. We found 22 high-frequency TMD mutations in genotype B OBI strains. Among them, five mutations can lead to impairment of HBsAg secretion; seven mutations had accumulated intracellular HBsAg while extracellular HBsAg didn’t decrease compared to wildtype. This study chose C85R from TMD2, F220C, and F220Y from TMD4 for further exploration. Protein structure predication showed these three mutant HBsAg displayed changed hydrophilic properties and tended to accumulate in the phospholipid bilayer of cell membrane. Mutant HBsAg’s secretion disorder may induce OBI. On the other hand, V168A + V177A from TMD3 expressed increased HBsAg both in intracellular and extracellular levels. This mutation had most unstable natural conformation and may be inclined to transition into V177A or V168A + S174N + V177A. These three mutations were more prone to mixed infection, presenting a state of coexistence, thus approaching the impaired secretion pattern of OBI. This study demonstrated TMD mutations could contribute to the occurrence of OBI and provided a theoretical basis for OBI study and the functional cure of chronic hepatitis B virus infection.

Muscular dystrophies (MDs) are a heterogeneous group of myopathies characterized by progressive muscle weakness leading to death from heart or respiratory failure. MDs are caused by mutations in genes involved in both the development and organization of muscle fibers. Several animal models harboring mutations in MD-associated genes have been developed so far. Together with rodents, the zebrafish is one of the most popular animal models used to reproduce MDs because of the high level of sequence homology with the human genome and its genetic manipulability. This review describes the most important zebrafish mutant models of MD and the most advanced tools used to generate and characterize all these valuable transgenic lines. Zebrafish models of MDs have been generated by introducing mutations to muscle-specific genes with different genetic techniques, such as (i) N-ethyl-N-nitrosourea (ENU) treatment, (ii) the injection of specific morpholino, (iii) tol2-based transgenesis, (iv) TALEN, (v) and CRISPR/Cas9 technology. All these models are extensively used either to study muscle development and function or understand the pathogenetic mechanisms of MDs. Several tools have also been developed to characterize these zebrafish models by checking (i) motor behavior, (ii) muscle fiber structure, (iii) oxidative stress, and (iv) mitochondrial function and dynamics. Further, living biosensor models, based on the expression of fluorescent reporter proteins under the control of muscle-specific promoters or responsive elements, have been revealed to be powerful tools to follow molecular dynamics at the level of a single muscle fiber. Neurochemical events involving biosignals of different time and space dimensionalities constitute the complex basis of neurological functions and diseases. In view of this fact, electrochemical measurements enabling real‐time quantification of neurochemicals at multiple levels of spatiotemporal resolution can provide informative clues to decode the molecular networks bridging vesicles and brains. This Minireview focuses on how scientific questions regarding the properties of single vesicles, neurotransmitter release kinetics, interstitial neurochemical dynamics, and multisignal interconnections in vivo have driven the design of electrochemical nano/microsensors, sensing interface engineering, and signal/data processing. Purpose: To use a novel in vivo method to simulate a moving hip model. Then, measure the dynamic bone-to-bone distance, and analyze the ischiofemoral space (IFS) of patients diagnosed with ischiofemoral impingement syndrome (IFI) during dynamic activities.Methods: Nine healthy subjects and 9 patients with IFI were recruited to collect MRI images and motion capture data. The motion trail of the hip during motion capture was matched to a personalized 3D hip model reconstructed from MRI images to get a dynamic bone model. This personalized dynamic in vivo method was then used to simulate the bone motion in dynamic activities. Validation was conducted on a 3D-printed sphere by comparing the calculated data using this novel method with the actual measured moving data using motion capture. Moreover, the novel method was used to analyze the in vivo dynamic IFS between healthy subjects and IFI patients during normal and long stride walking.Results: The validation results show that the root mean square error (RMSE) of slide and rotation was 1.42 mm/1.84° and 1.58 mm/2.19°, respectively. During normal walking, the in vivo dynamic IFS was significantly larger in healthy hips (ranged between 15.09 and 50.24 mm) compared with affected hips (between 10.16 and 39.74 mm) in 40.27%–83.81% of the gait cycle (p = 0.027). During long stride walking, the in vivo dynamic IFS was also significantly larger in healthy hips (ranged between 13.02 and 51.99 mm) than affected hips (between 9.63 and 44.22 mm) in 0%–5.85% of the gait cycle (p = 0.049). Additionally, the IFS of normal walking was significantly smaller than long stride walking during 0%–14.05% and 85.07%–100% of the gait cycle (p = 0.033, 0.033) in healthy hips. However, there was no difference between the two methods of walking among the patients.Conclusions: This study established a novel in vivo method to measure the dynamic bone-to-bone distance and was well validated. This method was used to measure the IFS of patients diagnosed with IFI, and the results showed that the IFS of patients is smaller compared with healthy subjects, whether in normal or long stride walking. Meanwhile, IFI eliminated the difference between normal and long stride walking.

Developing techniques for the highly sensitive assay of neurotransmitters is essential for understanding physiological and pathological processes. Here, we demonstrate a fast‐scanning potential (FSP)‐gated organic electrochemical transistor (OECT): for the highly sensitive sensing of dopamine (DA) in a living rat brain. The configuration combines the selectivity of fast‐scan cyclic voltammetry (FSCV) with the high sensitivity of an OECT. The combined use of FSP as a gating mode and transconductance (gm) as a sensing parameter further improve the sensing performance in terms of sensitivity, limit of detection, reproducibility, and stability. The FSP‐OECT exhibits a sensitivity of 0.899 S M−1 and a low limit of detection down to 5 nM and was validated for in vivo monitoring of the basal level and electrically stimulated release of DA. Chemical pollution is a threat to coral reefs. To preserve them, it is crucial to monitor novel contaminants and assess the related risks. The occurrence of active pharmaceutical ingredients (APIs) in coral reefs has been poorly investigated until now. Under this light, we tested the use of the marine sponge Cf. Hyrtios as bio-monitors and conducted a pilot study in the Faafu Atoll (Maldives). Analyses were carried out by in vivo solid-phase microextraction (SPME) and liquid chromatography (LC) electrospray ionization (ESI) tandem mass spectrometry (MS/MS). Twelve APIs were selected for method optimization. Limits of quantitation (LOQs) were in the 0.6 and 2.5 ng/g range, accuracy between 86.5 % and 104.7 %, and precision between 3.0 % and 14.9 %. All the sponges located in the inner reefs resulted contaminated with at least one API. Gabapentin and Carbamazepine displayed the highest detection rates, while Ketoprofen had the highest concentration (up to 15.7 ng/g). Abstract Background Rice quality research attracts attention worldwide. Rice chalkiness is one of the key indexes determining rice kernel quality. The traditional rice chalkiness measurement methods only use milled rice as materials and are mainly based on naked-eye observation or area-based two-dimensional (2D) image analysis and the results could not represent the three-dimensional (3D) characteristics of chalkiness in the rice kernel. These methods are neither in vivo thus are unable to analyze living rice seeds for high throughput screening of rice chalkiness phenotype. Results Here, we introduced a novel method for 3D visualization and accurate volume-based quantification of rice chalkiness in vivo by using X-ray microcomputed tomography (micro-CT). This approach not only develops a novel volume-based method to measure the 3D rice chalkiness index, but also provides a high throughput solution for rice chalkiness phenotype analysis by using living rice seeds. Conclusions Our method could be a new powerful tool for rice chalkiness measurement, especially for high throughput chalkiness phenotype screening using living rice seeds. This method could be used in chalkiness phenotype identification and screening, and would greatly promote the basic research in rice chalkiness regulation as well as the quality evaluation in rice production practice.

Abstract Background Myelin sheaths surrounding axons are critical for electrical signal transmission in the central nervous system (CNS). Diseases with myelin defects such as multiple sclerosis (MS) are devastating neurological conditions for which few effective treatments are available. Dysfunction of the dopaminergic system has been observed in multiple neurological disorders. Its role in myelin pathogenesis, however, is unclear. Methods This work used a combination of literature curation, bioinformatics, pharmacological and genetic manipulation, as well as confocal imaging techniques. Literature search was used to establish a complete set of genes which is associated with MS in humans. Bioinformatics analyses include pathway enrichment and crosstalk analyses with human genetic association studies as well as gene set enrichment and causal relationship analyses with transcriptome data. Pharmacological and CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9) genetic manipulation were applied to inhibit the dopaminergic signaling in zebrafish. Imaging techniques were used to visualize myelin formation in vivo. Results Systematic analysis of human genetic association studies revealed that the dopaminergic synapse signaling pathway is enriched in candidate gene sets. Transcriptome analysis confirmed that expression of multiple dopaminergic gene sets was significantly altered in patients with MS. Pathway crosstalk analysis and gene set causal relationship analysis reveal that the dopaminergic synapse signaling pathway interacts with or is associated with other critical pathways involved in MS. We also found that disruption of the dopaminergic system leads to myelin deficiency in zebrafish. Conclusions Dopaminergic signaling may be involved in myelin pathogenesis. This study may offer a novel molecular mechanism of demyelination in the nervous system.

Bacillus thuringiensis (Bt) is a ubiquitous bacterium that produces several proteins that are toxic to different invertebrates such as insects, nematodes, mites, and also some protozoans. Among these, Cry and Cyt proteins are most explored as biopesticides for their action against agricultural pests and vectors of human diseases. In 2000, a group of researchers from Japan isolated parasporal inclusion proteins from B. thuringiensis , and reported their cytotoxic action against human leukemia. Later, other proteins with similar antitumor properties were also isolated from this bacterium and these cytotoxic proteins with specific activity against human cancer cells were named parasporins. At present, nineteen different parasporins are registered and classified in six families. These parasporins have been described to have specific in vitro antitumor activity against several cancer cell lines. The antitumor activity makes parasporins possible candidates as anticancer agents. Various research groups around the world are involved in isolating and characterizing in vitro antitumor activity of these proteins and many articles reporting such activities in detail have been published. However, there are virtually no data regarding the antitumor activity of parasporins in vivo. This review summarizes the properties of these potentially useful antitumor agents of natural origin, focusing on their in vivo activity thus also highlighting the importance of testing these proteins in animal models for a possible application in clinical oncology. Numerous neurochemicals have been implicated in the modulation of brain function, making them appealing analytes for sensors and diagnostics. However, it is a grand challenge to selectively measure multiple neurochemicals simultaneously in vivo because of their great variations in concentrations, dynamic nature, and composition. Herein, we present a deep learning‐based voltammetric sensing platform for the highly selective and simultaneous analysis of three neurochemicals in a living animal brain. The system features a carbon fiber electrode capable of capturing the mixed dynamics of a neurotransmitter, neuromodulator, and ions. Then a powerful deep neural network is employed to resolve individual chemical and spatial‐temporal information. With this, a single electrochemical measurement reveals an interplaying concentration changes of dopamine, ascorbate, and ions in living rat brain, which is unobtainable with existing analytical methodologies. High operational costs of modern medical devices and the required specialized, skilled personnel with certifications to operate most medical instrumentation remains a challenge and an impediment to rapid medical diagnosis and clinical analysis. The simplicity, portability, and ease of operation makes Raman spectrometers appealing for rapid medical diagnosis and clinical analysis at an affordable cost. Besides, the combined use of Raman spectroscopy and multivariate analyses has further facilitated effective pattern recognition providing accurate classification, and/or differentiation of biological and clinical samples. This review article highlights recent advances in Raman spectroscopy in medical diagnosis and clinical analysis between January 2018 and December 2020. Recent innovations in the use of Raman spectroscopy for chemical analysis in human specimens are discussed. Applications of Raman spectroscopy in cancer immunotherapy, cancer imaging, and detecting disease biomarkers in clinical samples are further highlighted. The review article highlights recent innovations in the use of Raman spectroscopy for the detection of various pathogens in human specimens. Moreover, recent innovations of combined uses of Raman spectroscopy and multivariate regression analyses for pattern recognition, and/or classification of clinical samples are discussed. Monitoring extracellular calcium ion (Ca2+) chemical signals in neurons is crucial for tracking physiological and pathological changes associated with brain diseases in live animals. Potentiometry based solid-state ion-selective electrodes (ISEs) with the assist of functional carbon nanomaterials as ideal solid-contact layer could realize the potential response for in vitro and in vivo analysis. Herein, we employ a kind of biomass derived porous carbon as a transducing layer to prompt efficient ion to electron transduction while stabilizes the potential drift. The eco-friendly porous carbon after activation (APB) displays a high specific area with inherit macropores, micropores, and large specific capacitance. When employed as transducer in ISEs, a stable potential response, minimized potential drift can be obtained. Benefiting from these excellent properties, a solid-state Ca2+ selective carbon fiber electrodes (CFEs) with a sandwich structure is constructed and employed for real time sensing of Ca2+ under electrical stimulation. This study presents a new approach to develop sustainable and versatile transducers in solid-state ISEs, a crucial way for in vivo sensing. A potentiometric ion selective electrode-based approach for in vivo analysis of extracellular Ca2+ in rat brain by using pine cone derived porous carbon as transducer have been successfully demonstrated. At present the information regarding the occurrence of human pharmaceuticals (PhaCs) in coral reefs and their potential impacts on the associated fauna is limited. To optimize the collection of data in these delicate environments, we employed a solid-phase microextraction (bioSPME) and liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) procedure that enabled in vivo determinations in soft corals. Specifically, we researched the antibiotics Ofloxacin Sulfamethoxazole and Clarithromycin, the anti-inflammatory Diclofenac Propyphenazone Ketoprofen and Amisulpride, the neuroactive compounds Gabapentin-lactam, the beta-blocker Metoprolol and the antiepileptic Carbamazepine. Reproducibility was between 2.1% and 9.9% and method detection limits LODs) were between 0.2 and 1.6 ng/g and LOQs between 0.8 and 5.4 mg/g. The method was then applied to establish a baseline for the occurrence of these compounds in the Maldivian archipelago. Colonies of Sarcophyton sp. and Sinularia sp. were sampled along an inner-outer reef transect. Five of the ten targeted PhaCs were identified, and 40% of the surveyed coral colonies showed the occurrence of at least one of the selected compounds. The highest concentrations were found inside the atoll rim. Oxoflacin (9.5 ± 3.9 ng/g) and Ketoprofen (4.5 ± 2.3 ng/g) were the compounds with the highest average concentrations. Outside the atoll rim, only one sample showed contamination levels above the detection limit. No significant differences were highlighted among the two surveyed soft coral species, both in terms of average concentrations and bioconcentration factors (BCFs). Abstract Background The increasing use of gold nanoparticles (AuNPs) in the field of neuroscience instilled hope for their rapid translation to the clinical practice. AuNPs can be engineered to carry therapeutics or diagnostics in the diseased brain, possibly providing greater cell specificity and low toxicity. Although there is a general enthusiasm for these tools, we are in early stages of their development. Overall, their brain penetrance, stability and cell specificity are critical issues that must be addressed to drive AuNPs to the clinic. Results We studied the kinetic, distribution and stability of PEG-coated AuNPs in mice receiving a single injection into the cisterna magna of the 4th ventricle. AuNPs were conjugated with the fluorescent tag Cy5.5 (Cy5.5-AuNPs) to track their in vivo distribution. Fluorescence levels from such particles were detected in mice for weeks. In situ analysis of brains by immunofluorescence and electron microscopy revealed that Cy5.5-AuNPs penetrated the brain parenchyma, spreading in the CNS parenchyma beneath the 4th ventricle. Cy5.5-AuNPs were preferentially found in neurons, although a subset of resting microglia also entrapped these particles. Conclusions Our results suggest that the ICM route for delivering gold particles allows the targeting of neurons. This approach might be pursued to carry therapeutics or diagnostics inside a diseased brain with a surgical procedure that is largely used in gene therapy approaches. Furthermore, this approach could be used for radiotherapy, enhancing the agent’s efficacy to kill brain cancer cells.

Background: Nature has always been considered as the primary source of pharmaceutical ingredients. A variety of hemicelluloses, as well as their modified forms, have been under investigation. Herein, a study was designed to explore the biocompatibility of hemicellulose and its modified form (thiolated hemicellulose) as well as its potential as a pharmaceutical excipient.Method: For thiol modification thiourea was used as the thiol donor, HCl as the catalytic reagent, and methanol was used for washing purposes. Modified polymers were characterized for physicochemical characteristics, including surface morphology, the amorphous or crystalline nature of the particles, modification of polymer by FTIR, and biocompatibilities. For acute oral toxicity study, a single dose of 2 g/kg was administered to albino rats of 200 g average weight (n = 3). Polymers were evaluated as pharmaceutical excipients by preparing compressed tablets of antiplatelet drug (Ticagrelor), followed by various quality control tests, such as swelling index, thickness and diameter, disintegration, and in-vitro drug release.Results: From the results, it was observed that thiol modification has been successfully accomplished as characteristic peaks belonging to –SH group appeared at 2667.7691 cm−1 in FTIR scan. The modified polymer was found safe in the use concentration range, confirming their safe use for in vivo analysis. No significant effect has been observed in the behavior, biological fluid (blood), or on vital organs. Thiolated hemicellulose was found to be an excellent drug retarding polymer as 8 h of dissolution studies showed that 67.08% of the drug has been released.Conclusion: Conclusively, incorporation of thiol moiety made the polymer more mucoadhesive with, and a worthy carrier of, the drug with good biocompatibilities.

In this study, we developed contactless electrospray ionization mass spectrometry (ESI‐MS) for in vivo analysis of living organisms in different applications. The in vivo sampling and direct analysis processess of living organisms were integrated into an operation that only requires the organism close to MS inlet that was applied to a high voltage. Living plants and animals were directly induced to generate spray ionization. Direct detection and in vivo monitoring of metabolites and chemical residues in various living organisms were successfully demonstrated. Analysis of a single sample could be completed within 30 s. Overall, contactless ESI‐MS provides an attractive in vivo method to straightforward investigation of living organisms. [ABSTRACT FROM AUTHOR]

Objectives: Rheumatoid arthritis (RA) is characterized by inflammation in multiple joints. In addition to causing joint destruction, the persistent systemic inflammation with RA increases the risk of cardiovascular disease. Although there are in vitro studies showing the prothrombotic effect of inflammatory cytokines, especially TNF, in vivo experimental evidence is lacking due to the complexity of in vivo modeling and observation. In this study, we aimed to model in vivo thrombus formation in arthritic mice and to determine whether the arthritic condition would further promote thrombotic formation. Methods: Human TNF-transgenic mice were used as the arthritis model. Thrombus formation was observed on the testicular arterioles. Thrombus formation was induced by reactive oxygen species generated from hematoporphyrin under laser irradiation. Results: Platelet thrombus formation was observed in real-time using a laser confocal microscopy in both wild-type and arthritic mice. Quantitative analyses revealed that no significant differences were observed in thrombus formation, represented by platelet attachment time and vascular obstruction time, in our experimental setting. Conclusion: Although we confirmed the usefulness of this novel technique for in vivo studies, further investigation is required to conclude the possible mechanism of prothrombotic phenotypes under inflammatory conditions. Selective and nondisruptive in vivo neurochemical monitoring within the central nervous system has long been a challenging endeavor. We introduce a new sensing approach that integrates neurocompatible galvanic redox potentiometry (GRP) with customizable phosphorothioate aptamers to specifically probe dopamine (DA) dynamics in live rat brains. The aptamer-functionalized GRP (aptGRP) sensor demonstrates nanomolar sensitivity and over a 10-fold selectivity for DA, even amidst physiological levels of major interfering species. Notably, conventional sensors without the aptamer modification exhibit negligible reactivity to DA concentrations exceeding 20 μM. Critically, the aptGRP sensor operates without altering neuronal activity, thereby permitting real-time, concurrent recordings of both DA flux and electrical signaling in vivo. This breakthrough establishes aptGRP as a viable and promising framework for the development of high-fidelity sensors, offering novel insights into neurotransmission dynamics in a live setting.

ABSTRACT Methylglyoxal (MG) is a detrimental metabolic by-product that threatens most organisms (in humans MG causes diabetes). MG is predominantly detoxified by the glyoxalase pathway. This process begins with the conjugation of MG with glutathione (GSH), yielding a hemithioacetal product that is subsequently transformed by the glyoxalase enzymes into d-lactate and GSH. MG has been overlooked in photosynthetic organisms, although they inevitably produce it not only by the catabolism of sugars, lipids, and amino acids, as do heterotrophic organisms, but also by their active photoautotrophic metabolism. This is especially true for cyanobacteria that are regarded as having developed photosynthesis and GSH-dependent enzymes to detoxify the reactive oxygen species produced by their photosynthesis (CO2 assimilation) and respiration (glucose catabolism), which they perform in the same cell compartment. In this study, we used a combination of in vivo and in vitro approaches to characterize a logical, but as yet never described, link between MG detoxification and a (prokaryotic) representative of the evolutionarily conserved glutathione transferase (GST) detoxification enzymes. We show that the Sll0067 GST of the model cyanobacterium Synechocystis sp. strain PCC 6803 plays a prominent role in MG tolerance and detoxification, unlike the other five GSTs of this organism. Sll0067 catalyzes the conjugation of MG with GSH to initiate its elimination driven by glyoxalases. These results are novel because the conjugation of MG with GSH is always described as nonenzymatic. They will certainly stimulate the analysis of Sll0067 orthologs from other organisms with possible impacts on human health (development of biomarkers or drugs) and/or agriculture. IMPORTANCE In most organisms, methylglyoxal (MG), a toxic metabolite by-product that causes diabetes in humans, is predominantly detoxified by the glyoxalase enzymes. This process begins with the so-called “spontaneous” conjugation of MG with the cytoprotectant metabolite glutathione (GSH). In this study, we unravel a logical, but as yet unsuspected, link between MG detoxification and a (prokaryotic) representative of the ubiquitous glutathione transferase (GST) enzymes. We show that a GST of a model cyanobacterium plays a prominent role in the detoxification of MG in catalyzing its conjugation with GSH. This finding is important because this reaction, always regarded as nonenzymatic, could exist in plants and/or human and thus have an impact on agriculture and/or human health.

The chronic inflammatory conditions like psoriasis has an increased prevalence and is linked with various associated life threatening disease conditions. The main objective of this project was to developed a methotrexate-olive loaded nano emulsion. The formulation was assessed for various parameters including Thermodynamic Stability, physico-chemically characterization, drug release kinetics and entrapment efficiency and in vitro/in vivo skin permeation analysis. Final optimized formulation had a particle size 18.27±5.78 nm with a PDI of 0.25±0.01, whereas the average entrapment efficiency of formulation was 74.68±2.1%. The release kinetics suggested 97.72% drug release at pH 5 after 20 hrs. The FTIR data confirmed that the chemical structure of drug is retained with efficient loading into the formulation. Permeation data showed that an average of 79.23±3.6μg/cm2 of methotrexate was permeated from the nano emulsion with an average flux of 2.326±0.45μg/cm2/h after 24 hrs. Finally in vivo studies on rabbit skin confirmed that the structural changes of intercellular lipid layers in the stratum corneum are not responsible for enhanced skin permeation of methotrexate loaded nano emulsion. Electrochemical sensing performance is often compromised by electrode biofouling (e.g. proteins nonspecific binding) in complex biological fluids; however, the design and construction of a robust biointerface remains a great challenge. Herein, inspired by nature, we demonstrate a robust polydopamine‐engineered biointerfacing, to tailing zwitterionic molecules (i.e. sulfobetaine methacrylate, SBMA) through Michael Addition. The SBMA‐PDA biointerface can resist proteins nonspecific binding in complex biological fluids while enhancing interfacial electron transfer and electrochemical stability of the electrode. In addition, this sensing interface can be integrated with tissue‐implantable electrode for in vivo analysis with improved sensing performance, preserving ca. 92.0% of the initial sensitivity after 2 h of implantation in brain tissue, showing low acute neuroinflammatory responses and good stability both in normal and in Parkinson′s disease (PD) rat brain tissue. The development of new principles and techniques with high neuronal compatibility for quantitatively monitoring the dynamics of neurochemicals is essential for deciphering brain chemistry and function but remains a great challenge. We herein report a neuron‐compatible method for in vivo neurochemical sensing by powering a single carbon fiber through spontaneous bipolar electrochemistry as a new sensing platform. By using ascorbic acid as a model target to prove the concept, we found that the single‐carbon‐fiber‐powered microsensor exhibited a good response, high stability and, more importantly, excellent neuronal compatibility. The microsensor was also highly compatible with electrophysiological recording, thus enabling the synchronous recording of both chemical and electrical signals. The sensing principle could be developed for in vivo monitoring of various neurochemicals in the future by rationally designing and tuning the electrochemical reactions at the two poles of the carbon fiber. Background: Myelin sheaths surrounding axons are critical for electrical signal transmission in the central nervous system (CNS). Diseases with myelin defects such as multiple sclerosis (MS) are devastating neurological conditions for which few effective treatments are available. Dysfunction of the dopaminergic system has been observed in multiple neurological disorders. Its role in myelin pathogenesis, however, is unclear.Methods: This work used a combination of literature curation, bioinformatics, pharmacological and genetic manipulation, as well as confocal imaging techniques. Literature search was used to establish a complete set of genes which is associated with MS in humans. Bioinformatics analyses include pathway enrichment and crosstalk analyses with human genetic association studies as well as gene set enrichment and causal relationship analyses with transcriptome data. Pharmacological and CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9) genetic manipulation were applied to inhibit the dopaminergic signaling in zebrafish. Imaging techniques were used to visualize myelin formation in vivo.Results: Systematic analysis of human genetic association studies revealed that the dopaminergic synapse signaling pathway is enriched in candidate gene sets. Transcriptome analysis confirmed that expression of multiple dopaminergic gene sets was significantly altered in patients with MS. Pathway crosstalk analysis and gene set causal relationship analysis reveal that the dopaminergic synapse signaling pathway interacts with or is associated with other critical pathways involved in MS. We also found that disruption of the dopaminergic system leads to myelin deficiency in zebrafish.Conclusions: Dopaminergic signaling may be involved in myelin pathogenesis. The flagellated Gram-negative bacterium Escherichia coli is one of the most studied microorganisms. Despite extensive studies as a model prokaryotic cell, the ultrastructure of the cell envelope at the nanometre scale has not been fully elucidated. Here, a detailed structural analysis of the bacterium using a combination of small-angle X-ray and neutron scattering (SAXS and SANS, respectively) and ultra-SAXS (USAXS) methods is presented. A multiscale structural model has been derived by incorporating well established concepts in soft-matter science such as a core-shell colloid for the cell body, a multilayer membrane for the cell wall and self-avoiding polymer chains for the flagella. The structure of the cell envelope was resolved by constraining the model by five different contrasts from SAXS, and SANS at three contrast match points and full contrast. This allowed the determination of the membrane electron-density profile and the inter-membrane distances on a quantitative scale. The combination of USAXS and SAXS covers size scales from micrometres down to nanometres, enabling the structural elucidation of cells from the overall geometry down to organelles, thereby providing a powerful method for a non-invasive investigation of the ultrastructure. This approach may be applied for probing in vivo the effect of detergents, antibiotics and antimicrobial peptides on the bacterial cell wall.

The current study was designed to convert the poloxamer (PLX) into thiolated poloxamer (TPLX), followed by its physicochemical, biocompatibilities studies, and applications as a pharmaceutical excipient in the development of tacrolimus (TCM)-containing compressed tablets. Thiolation was accomplished by using thiourea as a thiol donor and hydrochloric acid (HCl) as a catalyst in the reaction. Both PLX and TPLX were evaluated for surface morphology based on SEM, the crystalline or amorphous nature of the particles, thiol contents, micromeritics, FTIR, and biocompatibility studies in albino rats. Furthermore, the polymers were used in the development of compressed tablets. Later, they were also characterized for thickness, diameter, hardness, weight variation, swelling index, disintegration time, mucoadhesion, and in vitro drug release. The outcomes of the study showed that the thiolation process was accomplished successfully, which was confirmed by FTIR, where a characteristic peak was noticed at 2695.9968 cm−1 in the FTIR scan of TPLX. Furthermore, the considerable concentration of the thiol constituents (20.625 µg/g of the polymer), which was present on the polymeric backbone, also strengthened the claim of successful thiolation. A mucoadhesion test illustrated the comparatively better mucoadhesion strength of TPLX compared to PLX. The in vitro drug release study exhibited that the TPLX-based formulation showed a more rapid (p < 0.05) release of the drug in 1 h compared to the PLX-based formulation. The in vivo toxicity studies confirmed that both PLX and TPLX were safe when they were administered to the albino rats. Conclusively, the thiolation of PLX made not only the polymer more mucoadhesive but also capable of improving the dissolution profile of TCM.

Modulation of interfacial electron transfer has been proven to pave a new approach to in vivo electrochemical monitoring of brain chemistry; however, designing and establishing highly efficient electrocatalytic scheme towards neurochemicals remain a long-standing challenge. Here, we find that recently established single-atom catalyst (SAC) can be used for catalyzing the electro-chemical process of physiologically relevant chemicals and thus offers a new avenue to in vivo electrochemical biosensing. To prove this new concept, we used Co single-atom catalyst (Co-SAC), in which the atomic active sites are dispersed in ordered porous N-doping carbon matrix at atomic level, as an example of SACs for analyzing glucose as the physiologically relevant model chemicals. We found that Co-SAC catalyzes the electrochemical oxidation of hydrogen peroxide (H2O2) at a low potential of ca. +0.05 V (vs. Ag/AgCl). This property was further used for developing an oxidase-based glucose biosensor that was used subsequently as a selective detector of an online electrochemical system (OECS) for continuous monitoring of microdialysate glucose in rat brain. The OECS with Co-SAC-based glucose biosensor as the online detector was well responsive to glucose without interference from other electroactive species in brain microdialysate. Quantitively and stably tracking ion dynamics in the living brain of animals is essential to understanding many physiological and pathological processes. Solid-state ion-selective electrodes (ISEs) are powerful tools for monitoring the dynamic change of ions at physiological concentration range; however, the unintentional accumulation of an aqueous layer at the ion selective membrane/ solid contact interface compromises the electrode potential stability, limiting its in vivo application. Here, using manganese dioxide (MnO2) and potassium ISE (K+-ISE) as model solid contact and ISEs, we demonstrate for the first time that graphdiyne oxide (GDYO) can enhance the potential stability of solid contact-based ISEs. Our results suggest that the intrinsic structural and hydrophobic features of GDYO, plays a key role in impeding and stabilizing the formation of water layer. With GDYO-MnO2 acting as the solid contact, the K+-ISE displays an excellent short-term potential stability and maintains great selectivity, achieving reliable K+ sensing at the animal level. The GDYO-based strategy is generalizable to different ISEs and does not require complicated processing steps, paving an exciting opportunity for in vivo ion recognition and sensing. In this work, a vibrating tip spray ionization source was developed for direct mass spectrometric analysis of raw samples under voltage‐free condition. A solid tip was mounted on a vibrator, and the solid tip was placed on the front of MS inlet. Liquid, viscous, and bulk solid samples could be directly loaded on the tip‐end surface, and then a drop of solvent at microliter level was subsequently loaded on the tip for dissolution and extraction of analytes, and a vibrator was then started to atomize and ionize the analytes under ambient condition. We demonstrated vibrating tip spray mass spectrometry in various applications, including food safety, pharmaceutical analysis, and forensic science. Furthermore, in situ analysis of biological tissues and in vivo analysis of living plants were conveniently performed, due to voltage‐free. Different vibration frequencies and solvent compositions were investigated. The analytical performances, including sensitivity, reproducibility, and linear range, were investigated. Abstract Herein, a robust and homogeneous porous poly(3,4-ethylenedioxythiophene)/graphene (PEDOT/graphene) thin film surface-assisted laser desorption/ionization (SALDI) functional platform was prepared through a rapid and facile in-situ photopolymerization method. The graphene-embedded PEDOT skeleton well circumvented the aggregation-related problems in the traditional carbon-based SALDI method which combined with time-of-flight mass spectrometer (TOF MS). As a result, the reproducibility and quantitative capacity of the SALDI platform were significantly improved. Furthermore, the highly efficient adsorption performance of the PEDOT/graphene thin film was demonstrated in terms of in vitro and in vivo solid-phase microextraction (SPME) extraction. It showed that porous morphology with abundant graphene doping favored the adsorption and enrichment of target analytes. Owing to the excellent adsorption capability of the PEDOT/graphene thin film and the inherent strong laser absorption ability of graphene, expected SALDI effect (3–13 times higher than the commercial nanomaterial-assisted LDI plate) and quantitative analysis (linear range 0.5–100 μg L−1) of the PEDOT/graphene functional surfaces were achieved. As for the real-world applications, we deployed the PEDOT/graphene thin film SALDI platform for the analysis of five estrogens in biological samples at microliter-volume level, without tedious sample preparation procedures. Satisfactory recoveries ranging from 60.6% to 99.0% were obtained. The present study suggested that the graphene-embedded PEDOT skeleton with porous morphology would be developed as promising coating for the adsorption of analytes of interest. Additionally, the combination of PEDOT with graphene not only expanded the application fields of PEDOT, but also offered an efficient strategy for preparing homogeneous functional surfaces to realize the quantitative analysis in SALDI method. Microelectrode-based photoelectrochemical (PEC) sensing is a newly developed and promising analytical technique for in vivo analysis. However, the inadequate specificity in complex environment of living bodies restricted its further in vivo application. Herein, we utilized a small molecule probe as the energy acceptor to quench the photocurrent of CdTe quantum dots through energy transfer. The efficiency of energy transfer was modulated by the concentration of target SO 2 , resulting in changes in photocurrent. The chemical recognition reaction between small molecule probes and SO 2 enhanced the specificity of PEC sensing, thus guaranteeing its in vivo applications. Furthermore, with the use of light addressing strategy, simultaneous detection in the multiple brain regions was implemented. The energy transfer based light addressable PEC microsensor achieved monitoring fluctuations of SO 2 levels in multiple brain regions of rats with epilepsy. [Display omitted] [ABSTRACT FROM AUTHOR]

The present work outlines a detailed analysis of the treatment of scalp infection using silver nanomaterials (Ag NMs), and focuses on biocidal activity owing to manipulation of size, shape, and structure. Monodisperse silver spherical nanoparticles (NPs) and nanorods (NRs) were synthesized by chemical routes that were characterized using analytical and spectroscopic techniques. Ag NMs demonstrated enhanced biocidal tendencies compared to market available drugs, itracanozole and ketoconazole, showing greater zones of inhibition. The obtained 20 nm and 50 nm spherical-shaped NPs and 50 nm NRs showed concentration-, size-, and shape-dependent antifungal activity, with 20 nm spherical-shaped NPs exhibiting excellent potency. Minimum inhibitory concentration for 20 nm was lowest at 0.2 mg/mL in comparison to 0.3 mg/mL for NRs. Primary irritation index was 0.33 and 0.16 for 20 nm and 50 nm spherical-shaped NPs, respectively, while 50 nm rod-shaped NMs exhibited negligible redness. An in vivo model for M. furfur infection was generated by passing fungi subcutaneously in rats’ skin. Again, 20 nm particles showed best normalization of skin after 10 days on regular dosing, in comparison with bigger and rod-shaped particles. The statistical clinical score was highest for Ag nanorods, followed by 50 nm Ag NPs-treated animals. It was observed that 20 nm spherical particles exhibited the lowest score (0) compared with others as well as with antifungal drugs. Biochemical analysis performed by checking antioxidant enzymatic activities indicated tissue repair and normalization of enzymes and protein concentration by Ag NPs. The endoplasmic reticulum (ER) is the cellular site for the biosynthesis of proteins and lipids. The ER is highly dynamic, whose homeostasis is maintained by proper ER shaping, unfolded protein response (UPR), ER-associated degradation (ERAD), and selective autophagy of the ER (ER-phagy). In ERAD and ER-phagy, unfolded/misfolded proteins are degraded in the 26S proteasome and the vacuole, respectively. Both processes are vital for normal plant development and plant responses to environmental stresses. While it is known that ubiquitination of a protein initiates EARD, recent research indicated that ubiquitination of a protein also promotes the turnover of the protein through ER-phagy. In this chapter, we describe in detail two in vivo methods for investigating (1) the degradation efficiency and (2) ubiquitination level of an ER-associated protein in Arabidopsis thaliana., (© 2024.
