Abstract

The spin polarization measurements of particles emitted in heavy-ion collisions have opened the possibility for new phenomenological investigations of spin physics in relativistic fluids. The theoretical predictions of global polarization are in agreement with the data, but consistent discrepancies stand out for the local polarization. We show that the covariant theory of relativistic quantum fluids at local equilibrium implies an additional, non-dissipative contribution to the spin polarization vector, which is proportional to the thermal shear, which has been previously overlooked. This additional contribution, together with an improved approximation in the expansion of the local equilibrium density operator, restores the quantitative agreement between the theoretical predictions and the experimental data., Comment: 6 pages, 2 figures, presented at the XXIXth International Conference on Ultra-relativistic Nucleus-Nucleus Collisions (Quark Matter 2022). v2: Added ancillary files for the figures

2. Local polarization and isothermal local equilibrium in relativistic heavy ion collisions

Abstract

We show that the inclusion of a recently found additional term of the spin polarization vector at local equilibrium which is linear in the symmetrized gradients of the velocity field, and the assumption of hadron production at constant temperature restore the quantitative agreement between hydrodynamic model predictions and local polarization measurements in relativistic heavy ion collisions at $\sqrt{s_{\rm NN}} = 200$ GeV. The longitudinal component of the spin polarization vector turns out to be very sensitive to the temperature value, with a good fit around 155 MeV. We report on broadly based systematic investigations of the modeling components for open heavy-flavor diffusion and energy loss in strongly interacting matter in their application to heavy-flavor observables in high-energy heavy-ion collisions, conducted within an EMMI Rapid Reaction Task Force framework. Initial spectra including cold-nuclear-matter effects, a wide variety of space-time evolution models, heavy-flavor transport coefficients, and hadronization mechanisms are scrutinized in an effort to quantify pertinent uncertainties in the calculations of nuclear modification factors and elliptic flow of open heavy-flavor particles in nuclear collisions. Abstract

We present a quantitative study of vorticity formation in peripheral ultrarelativistic heavy ion collisions at sqrt(s)NN = 200 GeV by using the ECHO-QGP numerical code, implementing relativistic dissipative hydrodynamics in the causal Israel-Stewart framework in 3+1 dimensions with an initial Bjorken flow profile. We consider and discuss different definitions of vorticity which are relevant in relativistic hydrodynamics. After demonstrating the excellent capabilities of our code, which proves to be able to reproduce Gubser flow up to 8 fm/c, we show that, with the initial conditions needed to reproduce the measured directed flow in peripheral collisions corresponding to an average impact parameter b=11.6 fm and with the Bjorken flow profile for a viscous Quark Gluon Plasma with \eta/s=0.1 fixed, a vorticity of the order of some 10^{-2} c/fm can develop at freezeout. The ensuing polarization of Lambda baryons does not exceed 1.4% at midrapidity. We show that the amount of developed directed flow is sensitive to both the initial angular momentum of the plasma and its viscosity., Comment: 13 pages, 22 figures. Abstract

We present ECHO-QGP, a numerical code for $(3+1)$-dimensional relativistic viscous hydrodynamics designed for the modeling of the space-time evolution of the matter created in high energy nuclear collisions. The code has been built on top of the \emph{Eulerian Conservative High-Order} astrophysical code for general relativistic magneto-hydrodynamics [\emph{Del Zanna et al., Astron. Astrophys. 473, 11, 2007}] and here it has been upgraded to handle the physics of the Quark-Gluon Plasma. ECHO-QGP features second-order treatment of causal relativistic viscosity effects in both Minkowskian or Bjorken coordinates; partial or complete chemical equilibrium of hadronic species before kinetic freeze-out; initial conditions based on the optical Glauber model, including a Monte-Carlo routine for event-by-event fluctuating initial conditions; a freeze-out procedure based on the Cooper-Frye prescription. The code is extensively validated against several test problems and results always appear accurate, as guaranteed by the combination of the conservative (shock-capturing) approach and the high-order methods employed. Abstract

Patient-derived xenografts (PDXs) are resected human tumors engrafted into mice for preclinical studies and therapeutic testing. It has been proposed that the mouse host affects tumor evolution during PDX engraftment and propagation, affecting the accuracy of PDX modeling of human cancer. Here, we exhaustively analyze copy number alterations (CNAs) in 1,451 PDX and matched patient tumor (PT) samples from 509 PDX models. CNA inferences based on DNA sequencing and microarray data displayed substantially higher resolution and dynamic range than gene expression-based inferences, and they also showed strong CNA conservation from PTs through late-passage PDXs. CNA recurrence analysis of 130 colorectal and breast PT/PDX-early/PDX-late trios confirmed high-resolution CNA retention. We observed no significant enrichment of cancer-related genes in PDX-specific CNAs across models. Moreover, CNA differences between patient and PDX tumors were comparable to variations in multiregion samples within patients. Our study demonstrates the lack of systematic copy number evolution driven by the PDX mouse host., Analysis of copy number alterations in 1,451 patient-derived xenografts (PDXs) and matched patient tumor samples shows strong conservation from patient tumors through late-passage PDXs and a lack of systematic copy number evolution driven by the mouse host.

Abstract

Histone deacetylase inhibitors (HDACi) are active agents for peripheral T-cell lymphoma (PTCL). Anecdotally angioimmunoblastic T-cell lymphoma (AITL) appears to respond better than PTCL-not otherwise specified (NOS) to HDACi. The new World Health Organization classification shows that a subgroup of PTCL carries similarities in phenotype and gene expression profiling to AITL, comparable to T follicular helper (TFH) cells. The disease might behave similarly to AITL when treated with HDACi. We analyzed 127 patients with AITL or PTCL-NOS treated with HDACi at relapse as a single agent or in combination. We re-reviewed the pathology of all PTCL-NOS to identify the TFH phenotype. Patients received HDACi at relapse as a single agent in 97 cases (76%, 59 TFH, 38 non-TFH) or in combination in 30 cases (24%, 18 TFH, 12 non-TFH) including duvelisib, lenalidomide, lenalidomide plus carfilzomib, and pralatrexate. Seven PTCL-NOS had TFH phenotype; 2 PTCL-NOS were reclassified as AITL. Overall response rate (ORR) was 56.5% (28.9% complete response [CR]) in TFH and 29.4% (19.6% CR) in non-TFH phenotype patients (P = .0035), with TFH phenotype being an independent predictor of ORR (P = .009). Sixteen patients sufficiently responded to HDACi or HDACi in combination with another agent to proceed directly to allogeneic transplantation; 1 of 16 responded to donor lymphocyte infusion (12 TFH, 4 non-TFH). Our results, although retrospective, support that HDACi, as a single agent or in combination, may have superior activity in TFH-PTCL compared with non-TFH PTCL. This differential efficacy could help inform subtype-specific therapy and guide interpretation of HDACi trials.

Abstract

Anaplastic large-cell lymphomas (ALCLs) are a group of clinically and biologically heterogeneous diseases including the ALK(+) and ALK(-) systemic forms. Whereas ALK(+) ALCLs are molecularly characterized and can be readily diagnosed, specific immunophenotypic or genetic features to define ALK(-) ALCL are missing, and their distinction from other T-cell non-Hodgkin lymphomas (T-NHLs) remains controversial. In the present study, we undertook a transcriptional profiling meta-analysis of 309 cases, including ALCL and other primary T-NHL samples. Pathway discovery and prediction analyses defined a minimum set of genes capable of recognizing ALK(-) ALCL. Application of quantitative RT-PCR in independent datasets from cryopreserved and formalin-fixed paraffin-embedded samples validated a 3-gene model (TNFRSF8, BATF3, and TMOD1) able to successfully separate ALK(-) ALCL from peripheral T-cell lymphoma not otherwise specified, with overall accuracy near 97%. In conclusion, our data justify the possibility of translating quantitative RT-PCR protocols to routine clinical settings as a new approach to objectively dissect T-NHL and to select more appropriate therapeutic protocols. Abstract

Here we present an overview of why and how ideal relativistic magneto-hydrodynamics has been implemented in the ECHO-QGP code, with the aim to describe in a self-consistent way the dynamical evolution of the Quark Gluon Plasma in the presence of strong magnetic fields, like those generated by the fast moving charges of the nuclei in heavy ion collisions. After explaining the motivations, we will briefly illustrate the adopted formalism and we will present some preliminary results. In the conclusions, we will focus on the future developments.

Abstract

The receptor tyrosine kinases (RTKs) play a critical role, controlling cell proliferation, survival, and differentiation of normal cells. Their pivotal function has been firmly established in the pathogenesis of many cancers as well. The anaplastic lymphoma kinase (ALK), a transmembrane RTK, originally identified in the nucleophosmin (NPM)–ALK chimera of anaplastic large cell lymphoma, has emerged as a novel tumorigenic player in several human cancers. In this review, we describe the expression of the ALK–RTK, its related fusion proteins, and their molecular mechanisms of activation. Novel tailored strategies are briefly illustrated for the treatment of ALK-positive neoplasms.

Abstract

Background: Deletions at 13q14.3 are common in chronic lymphocytic leukemia and are also present in diffuse large B-cell lymphomas (DLBCL) but never in immunodeficiency-related DLBCL. To characterize DLBCL with 13q14.3 deletions, we combined genome-wide DNA profiling, gene expression and clinical data in a large DLBCL series treated with rituximab, cyclophosphamide, doxorubicine, vincristine and prednisone repeated every 21 days (R-CHOP21). Patients and methods: Affymetrix GeneChip Human Mapping 250K NspI and U133 plus 2.0 gene were used. MicroRNA (miRNA) expression was studied were by real-time PCR. Median follow-up of patients was 4.9 years. Results: Deletions at 13q14.3, comprising DLEU2/MIR15A/MIR16, occurred in 22/166 (13%) cases. The deletion was wider, including also RB1, in 19/22 cases. Samples with del(13q14.3) had concomitant specific aberrations. No reduced MIR15A/MIR16 expression was observed, but 172 transcripts were significantly differential expressed. Among the deregulated genes, there were RB1 and FAS, both commonly deleted at genomic level. No differences in outcome were observed in patients treated with R-CHOP21. Conclusions: Cases with 13q14.3 deletions appear as group of DLBCL characterized by common genetic and biologic features. Deletions at 13q14.3 might contribute to DLBCL pathogenesis by two mechanisms: deregulating the cell cycle control mainly due RB1 loss and contributing to immune escape, due to FAS down-regulation
