O transtorno do espectro autista (TEA) é uma desordem multifatorial complexa cuja fisiopatologia ainda não é completamente compreendida. Entretanto, algumas características, como as alterações nos componentes inibitórios encefálicos, se evidenciam nessa desordem. Dentre os principais fatores de risco ambientais para o TEA se destaca a exposição ao ácido valproico (VPA) durante a gestação, de forma que se utilizou o modelo animal baseado nessa abordagem para a obtenção dos dados experimentais. No Capítulo I, realizou-se um compilado de dados da literatura sobre o papel de fatores de transcrição (FTs) em diferentes desordens do neurodesenvolvimento; além disso, foram feitas análises, utilizando ferramentas de bioinformática, dos principais locais de expressão, bem como dos momentos de pico de expressão e das possíveis rotas de interação entre os FTs, incluindo os possíveis prejuízos induzidos pelo VPA. Essa análise contribuiu para a identificação de diversas hipóteses que poderiam estar associadas com danos observados na vida pós-natal. Sabendo-se que o VPA possui características pró-inflamatórias e pró-oxidantes, o resveratrol (RSV), polifenol com características anti-inflamatórias, antioxidantes e neuroprotetoras, surge como uma estratégia interessante de contraposição dessas características, viabilizando um estudo importante de vias envolvidas no TEA. No Capítulo II, foi possível observar que o VPA induziu vastas alterações na composição neuronal do córtex pré-frontal medial (CPFm) e, em menor escala, no hipocampo (HC), além de reduzir a expressão de receptor GABAA e das proteínas sinápticas neuroliguina-2 e gefirina. O tratamento com RSV foi capaz de prevenir, de forma geral, as alterações em neurônios totais e interneurônios GABAérgicos (IGs) no CPFm, porém não no HC. Além disso, o RSV também teve um efeito similar ao VPA na expressão de proteínas sinápticas no CPFm. Análises de bancos de dados de modelos animais em idade embrionária ressaltam alterações na maquinaria transcricional, metabolismo de carboidratos, metabolismo mitocondrial e ciclo celular, via da WNT, via da NOTCH e outros, sugerindo, junto ao Capítulo I, hipóteses referentes às ações do VPA e do RSV. No Capítulo III, foi observada uma expansão do dano hipocampal induzida pela exposição ao VPA, com descontinuidade do giro denteado e descompactação de CA1, além de alterações disseminadas na composição neuronal em todas as sub-regiões do HC. O RSV preveniu a alteração morfológica, além de equilibrar diversos parâmetros associados a IGs e neurônios totais. A exposição ao VPA induziu alterações na expressão de PTEN, AKT e CK2, não prevenidas por RSV. A partir destes dados, conclui-se que a exposição ao VPA pode desencadear alterações em etapas iniciais do desenvolvimento embrionário, com consequentes alterações pós-natais. Em animais jovens, foram observadas alterações em IGs, sinapses e receptor GABAA. Em animais adultos, o HC se destaca como uma região fortemente alterada, indicando um provável agravamento ao longo da vida. É provável que a ação preventiva do RSV ocorra por oposição ao VPA em vias de regulação da transcrição, metabolismo e outras, conforme apontam os Capítulos I e II. Dessa forma, observa-se que uma intervenção precoce é capaz de mudar o panorama de evolução da desordem, abrindo horizontes para estudos de novas abordagens no TEA. Autism spectrum disorder (ASD) is a complex multifactorial disorder whose pathophysiology is not yet fully understood. However, some characteristics, such as changes in brain inhibitory components, emerge in this disorder. Among the main environmental risk factors for ASD, exposure to valproic acid (VPA) during pregnancy stands out, so the animal model based on this approach was used to obtain the experimental data. In Chapter I, a compilation of data from the literature on the role of transcription factors (TFs) in different neurodevelopmental disorders was carried out, in addition, analyzes were conducted, using bioinformatics tools, of the main sites of expression, as well as the moments of peak expression and possible routes of interaction among TFs, including possible damage induced by VPA. This analysis contributed to the identification of several hypotheses that could be associated with the damage observed in postnatal life. Knowing that VPA has pro-inflammatory and pro-oxidant characteristics, resveratrol (RSV), a polyphenol with anti-inflammatory, antioxidant, and neuroprotective characteristics, appears as an interesting strategy to counteract these characteristics, enabling an important study of the pathways involved in ASD. In Chapter II, it was possible to observe that VPA induced vast changes in the neuronal composition of the medial prefrontal cortex (mPFC) and, to a lesser extent, in the hippocampus (HC), in addition to reducing the expression of GABAA receptor and synaptic proteins neuroligin-2 and gephyrin. Treatment with RSV was able to generally prevent changes in total neurons and GABAergic interneurons (GIs) in mPFC, but not in the HC. Furthermore, RSV also had a similar effect to VPA on the expression of synaptic proteins in the mPFC. Database analyzes of animal models at embryonic age highlight changes in transcriptional machinery, carbohydrate metabolism, mitochondrial metabolism, cell cycle, WNT pathway, NOTCH pathway, and others, suggesting, together with Chapter I, hypotheses regarding the actions of VPA and the RSV. In Chapter III, an expansion of the hippocampal damage induced by exposure to VPA was observed, with discontinuity of the dentate gyrus and decompaction of the CA1, in addition to widespread changes in neuronal composition in all HC subregions. RSV prevented the morphological change, in addition to balancing several parameters associated with IGs and total neurons. Exposure to VPA induced changes in the expression of PTEN, AKT and CK2, not prevented by RSV. From these data, it is concluded that exposure to VPA can trigger changes in early stages of embryonic development, with consequent postnatal changes. In young animals, changes were observed in GIs, synapses, and GABAA receptor. In adult animals, the HC stands out as a strongly altered region, indicating a probable worsening throughout life. It is likely that the preventive action of RSV occurs in opposition to VPA in transcriptional, metabolic, and other regulatory pathways, as indicated in Chapters I and II. In this way, it is observed that an early intervention can change the course of the disorder, opening horizons for studies of new approaches in ASD.

Autism spectrum disorder (ASD) is a prevalent neurodevelopmental disorder characterized by several alterations, including disorganized brain cytoarchitecture and excitatory/inhibitory (E/I) imbalance. We aimed to analyze aspects associated with the inhibitory components in ASD, using bioinformatics to develop notions about embryonic life and tissue analysis for postnatal life. We analyzed microarray and RNAseq datasets of embryos from different ASD models, demonstrating that regions involved in neuronal development are affected. We evaluated the effect of prenatal treatment with resveratrol (RSV) on the neuronal organization and quantity of parvalbumin-positive (PV+), somatostatin-positive (SOM+), and calbindin-positive (CB+) GABAergic interneurons, besides the levels of synaptic proteins and GABA receptors in the medial prefrontal cortex (mPFC) and hippocampus (HC) of the ASD model induced by valproic acid (VPA). VPA increased the total number of neurons in the mPFC, while it reduced the number of SOM+ neurons, as well as the proportion of SOM+, PV+, and CB+ neurons (subregion-specific manner), with preventive effects of RSV. In summary, metabolic alterations or gene expression impairments could be induced by VPA, leading to extensive damage in the late developmental stages. By contrast, due to its antioxidant, neuroprotective, and opposite action on histone properties, RSV may avoid damages induced by VPA.

A tese é composta por dois trabalhos complementares. No primeiro trabalho, estudamos o aprendizado do odor materno em ratos neonatos, com foco no final do período sensível para o aprendizado precoce de preferência de odor. No segundo trabalho, estudamos as características das correntes GABAérgicas dos interneurônios do córtex piriforme anterior (CPa) e seu papel no aprendizado precoce de preferência de odor. Para ambos os estudos, foram usados dados eletrofisiológicos experimentais e modelos computacionais foram desenhados usando modelagem baseada em agentes. Os resultados são descritos como o número acumulado de spikes e número de neurônios ativos, antes e depois do condicionamento. No primeiro artigo, mostramos que as mudanças nas propriedades intrínsecas das células piramidais no CPa reduzem a disponibilidade de células piramidais que respondem à exposição do odor materno. No segundo artigo, experimentos computacionais mostraram que a entrada GABAérgica no CPa melhora a habilidade do circuito olfatório para o aprendizado do odor materno. A discussão geral apresenta uma integração dos níveis da neuroetologia à biofísica como uma perspectiva de trabalho. The thesis consists of two complementary studies. In the first study, we investigated the maternal odor learning in neonatal rats, focusing on the end of the sensitive period for early odor preference learning. In the second study, we examined the characteristics of the GABAergic currents of the interneurons of the anterior piriform cortex (aPC) and their role in early odor preference learning. For both studies, experimental electrophysiological data were used, and computational models were designed using agent-based modeling. The results are described as the cumulative number of spikes and the number of active neurons, before and after conditioning. In the first article, we showed that changes in the intrinsic properties of aPC pyramidal cells reduced the availability of the responsive pyramidal cells during maternal odor exposure. In the second article, computational experiments showed that GABAergic entry into the aPC improves the olfactory circuit's ability to learn maternal odor. The general discussion presents an integration of the levels from neuroethology to biophysics as a work perspective.

O transtorno do espectro autista é uma desordem do neurodesen- volvimento caracterizada por prejuízos de interação e comunicação social, bem como comportamentos repetitivos e/ou estereotipados. Apesar da crescente prevalência, a etiologia permanece desconhecida, embora se saiba que existam interações entre fatores genéticos e ambientais. O modelo animal de autismo por exposição pré-natal ao ácido valproico (VPA) é consolidado por sua transla- cionalidade em relação aos indivíduos com TEA. Em todos os capítulos de dados, utilizou-se o modelo animal por injeção intraperitoneal de VPA (600 mg/kg) no dia embrionário (E) E12,5 nas ratas Wistar prenhes e avaliou-se o efeito de um tratamento pré-natal com resveratrol (RSV), através de injeções subcutâneas (3,6 mg/Kg) administradas nas ratas prenhes nos dias E6,5-E18,5. O RSV, por ser um polifenol com características anti-inflamatórias e neuro- protetoras, surge como uma importante ferramente para estudo de vias biológicas envolvidas na fisiopatologia desse transtorno. No presente trabalho, avaliamos: questões relativas a alterações sensoriais em diferentes idades, bem como aspectos histológicos e eletrofisiológicos possivelmente envolvidos nos comportamentos alterados; prejuízos em diferentes parâmentros relacionados ao comportamento do tipo empático; e, por fim, revisão da literatura em forma de capítulo de livro sobre possíveis mecanismos neuroimunes desencadeados por ativação imune materna, os quais trazem pistas importantes sobre a fisiopatologia do TEA. Nesse contexto, foi possível observar: I) alterações em comportamentos sensoriais olfatórios e táteis nas idades pós-natais (P) 10 e P30, respectivamente; alteração na disposição de neurônios parvalbumina- positivos (PV+) nas camadas da área somatossensorial primária (ASSp); redução na proporção de PV+ na amígdala; redução na expressão da gefirina na ASSp; II) alterações em componentes associados ao comportamento do tipo empático como visto pelo atraso no dia de abertura e redução na porcentagem da abertura do aparato nos dias iniciais de teste; III) alterações in vitro em correntes inibitórias pós sinápticas espontâneas (redução na frequência e tempo de decaimento), além da presença in vivo de períodos de descargas de ondas lentas durante o ciclo de vigília em 40% dos animais VPA. Nesse contexto, o resveratrol foi capaz de estabelecer prevenção na maioria das alterações (exceto no imunoconteúdo de gefirina e no comportamento do tipo empático), demonstrando que a restauração da organização de componentes inibitórios, como os interneurônios PV+, possivelmente proporciona a norma-lização do componente eletrofisiólogico que, por fim, restabelece o prejuízo sensorial. No caso dos parâmetros comportamentais associados à empatia, possivelmente outros substratos neurais, além dos sensoriais primários, estejam envolvidos (tais como mecanismos de tomada de decisão, processamento cognitivo e integração em regiões associativas), os quais possivelmente não são substratos da prevenção demonstrada pelo RSV. Finalmente, considerando as características intrínsecas do RSV (anti-inflamatória e neuroprotetora) e que esse tratamento preveniu diversos parâmetros analisados, hipotetiza-se que o VPA deixe sua impressão na prole por meio de mecanismos neuroimunes. Autism spectrum disorder is a neurodevelopmental disorder characterized by impairments of interaction and social communication, as well as repetitive and/or stereotyped behaviors. Although the increasing prevalence, the etiology remains unclear, despite it is known that there are interactions between genetic and environmental factors. The animal model of autism by prenatal exposure to valproic acid (VPA) is consolidated by its translationality in relation to individuals with ASD. In all data chapters, the animal model used was performed by intraperitoneal injection of VPA (600 mg kg) at embryonic (E) day E12.5 in pregnant Wistar rats and the effect of prenatal treatment with resveratrol (RSV) was evaluated, (3.6 mg / kg) given in pregnant rats on 6.5-E18.5 days. RSV, a polyphenol with anti-inflammatory and neuroprotective properties, appears as an important tool for the study of biological pathways involved in the pathophysiology of this disorder. In the present work, we evaluated: questions related to sensory alterations at different ages, as well as histological and electrophysiological aspects possibly involved in the altered behaviors; impairments in different parameters related to empathic behavior; and, finally, a review of the literature in a book chapter format about possible neuroimmune mechanisms triggered by maternal immune activation, which provide important clues about the pathophysiology of ASD. In this context, it was possible to observe: I) alterations in olfactory and tactile sensorial behaviors at the postnatal ages (P) 10 and P30, respectively; alteration in the arrangement of parvalbumin-positive neurons (PV +) in the primary somatosensory area (ASSp) layers; reduction in the proportion of PV + in the amygdala; reduction in the expression of gephyrin in ASSp; II) changes in components associated with the empathy-like behavior as seen by the opening day delay and reduction in percentage of apparatus opening on the initial test days; III) in vitro changes in spontaneous post synaptic inhibitory currents (reduction in frequency and decay time), in addition to in vivo presence of slow wave discharges during the wake cycle in 40% of the VPA animals. In this context, resveratrol was able to establish prevention in most of the alterations (except in the gephyrin immunocontent and in the parameters of empathy-like behavior), demonstrating that the restoration of the organization of inhibitory components, such as PV+ interneurons, possibly provides the normalization of the component electrophysiology, which finally restores sensory impairment. In the case of the behavioral parameters associated with empathy, other neural substrates, besides the primary sensory ones, may be involved (such as decision-making mechanisms, cognitive processing and integration in associative regions), which possibly are not substrates of the prevention demonstrated by RSV. Finally, considering the intrinsic characteristics of RSV (anti-inflammatory and neuroprotective) and that this treatment prevented several parameters analyzed, it is hypothesized that the VPA leaves its impression in the offspring through neuroimmune mechanisms.

In the olfactory system, cholinergic modulation has been associated with contrast modulation and changes in receptive fields in the olfactory bulb, as well the learning of odor associations in olfactory cortex. Computational modeling and behavioral studies suggest that cholinergic modulation could improve sensory processing and learning while preventing pro-active interference when task demands are high. However, how sensory inputs and/or learning regulate incoming modulation has not yet been elucidated. We here use a computational model of the olfactory bulb, piriform cortex (PC) and horizontal limb of the diagonal band of Broca (HDB) to explore how olfactory learning could regulate cholinergic inputs to the system in a closed feedback loop. In our model, the novelty of an odor is reflected in firing rates and sparseness of cortical neurons in response to that odor and these firing rates can directly regulate learning in the system by modifying cholinergic inputs to the system. In the model, cholinergic neurons reduce their firing in response to familiar odors – reducing plasticity in the PC, but increase their firing in response to novel odor – increasing PC plasticity. Recordings from HDB neurons in awake behaving rats reflect predictions from the model by showing that a subset of neurons decrease their firing as an odor becomes familiar.

Fear conditioned analgesia (FCA) can be defined as a reduction of pain sensitivity resulting from the exposure to a stimulus previously associated with an aversive event. Indeed, among the classically studied fear reactions - like freezing or avoidance – analgesia has been observed after fear conditioning although the underlying neuronal circuits are largely unknown. The ventro-lateral part of the periaqueductal grey (vlPAG) has been shown to be necessary for fear expression but is also considered as a key structure in descending modulation of pain. Thus, the vlPAG, which is highly connected to brain regions involved in fear processing (such as the amygdala and the medial prefrontal cortex) but also to the spinal cord, could be an interface between fear and pain behaviours observable in FCA. In this context, the present project had two main objectives: the first aim was to investigate further the behavioural paradigm of FCA recently established in the laboratory. We observed that animals submitted to a hot-plate (HP) test show a delayed response when the trial is paired with a conditioned stimulus (CS+). This result suggests that CS+-evoked fear responses can alter the animals pain threshold. We verified that this effect could not be due to a specific stress effect, and observed that FCA can be replicated with the same strength when the HP test is repeated. The second goal of the study was to test whether the vlPAG is involved in FCA. To do so, we used optogenetic tools to manipulate two populations of inhibitory interneurons: the somatostatin-expressing (SOM+) and parvalbumin-expressing (PVa+) interneurons. We observed that the activation of these two subpopulations induced a pro-nociceptive behavior, while the inhibition of PVa+ interneurons induced an analgesic-like behaviour in the HP test. Importantly, these modulations of pain threshold were not any more visible when the animals were submitted to the FCA paradigm, suggesting that the fear-induced analgesia overcomes the optogenetic effects. Analgesia condicionada por medo (FCA) pode ser definida como uma redução da sensibilidade à dor resultante da exposição de um estímulo que foi previamente associado a um evento aversivo. De facto, entre as respostas clássicas de medo, como imobilidade ou fuga , analgesia tem também sido observada depois de medo condicionado. Contudo, os circuitos neuronais inerentes são ainda desconhecidos. Apesar da parte ventro-lateral da substancia cinzenta periaquedutal (vlPAG) ser necessária para a expressão de medo esta estrutura é também considerada essencial na modulação da via descendente da dor. Assim, a vlPAG é uma área que está extensivamente conectada a regiões neuronais envolvidas no processamento do medo (como por exemplo a amigdala e o cortex pre-frontal) mas também à medula espinal, podendo funcionar como uma interface entre as manifestações de medo e dor observados em FCA. Desse modo, o presente estudo teve dois objectivos principais: o primeiro foi estudar em detalhe o paradigma comportamental de FCA que foi recentemente estabelecido no laboratório. Nós observamos que os animais quando sujeitos ao teste de placa quente (HP) demonstram um atraso na resposta quando o teste é emparelhado com o estímulo condicionado (CS+). Estes resultados sugerem que as respostas de medo elicitadas pelo CS+ podem alterar os limiares de dor dos animais. Além disso, verificámos que estes resultados não foram causados devido a um efeito de stress especifico e que quando o HP teste é repetido a FCA pode ser replicada, mantendo a mesma intensidade de resposta. O segundo objectivo deste estudo foi testar se a vlPAG está envolvida em FCA. Para tal, usamos ferramentas de optogenética para manipular duas subpopulações de interneurónios inibitórios: interneurónios expressando somatostatina (SOM+) e interneurónios expressando parvalbumina (PVa+). No teste de HP, constatámos que a activação destas duas subpopulações induz um comportamento pró-nociceptivo, enquanto que a inibição dos interneurónios de PVa+ resultam num comportamento analgésico. É importante salientar que a modulação destes limiares de dor não foi visível quando os animais foram submetidos ao paradigma de FCA, sugerindo que o efeito analgésico induzido por medo supera os efeitos da manipulação optogenética.

Conselho Nacional de Desenvolvimento Científico e Tecnológico Cortical interneurons are characterized by their distinct morphological, physiological and biochemical properties, acting as modulators of the excitatory activity by pyramidal neurons, for example. Various studies have revealed differences in both distribution and density of this cell group throughout distinct cortical areas in several species. A particular class of interneuron closely related to cortical modulation is revealed by the immunohistochemistry for calcium binding proteins calbindin (CB), calretinina (CR) and parvalbumin (PV). Despite the growing amount of studies focusing on calcium binding proteins, the prefrontal cortex of primates remains relatively little explored, particularly in what concerns a better understanding of the organization of the inhibitory circuitry across its subdivisions. In the present study we characterized the morphology and distribution of neurons rich in calcium-binding proteins in the medial, orbital and dorsolateral areas of the prefrontal cortex of the marmoset (Callithrix jacchus). Using both morphometric and stereological techniques, we found that CR-reactive neurons (mainly double bouquet and bipolar cells) have a more complex dendritic arborization than CB-reactive (bitufted and basket cells) and PV-reactive neurons (chandelier cells). The neuronal densities of CR- and CB-reactive cells are higher in the supragranular layers (II/III) whilst PV-reactive neurons, conversely, are more concentrated in the infragranular layers (V/VI). CR-reactive neurons were the predominant group in the three regions evaluated, being most prevalent in dorsomedial region. Our findings point out to fundamental differences in the inhibitory circuitry of the different areas of the prefrontal cortex in marmoset Os interneurônios do córtex cerebral são caracterizados por suas diferentes propriedades morfológicas, fisiológicas e bioquímicas, atuando como moduladores da atividade excitatória cortical dos neurônios piramidais, por exemplo. Vários estudos revelaram diferenças na distribuição e densidade deste grupo celular ao longo de diferentes áreas corticais em diversas espécies. Uma classe particular de interneurônios intimamente relacionada à modulação cortical é revelada pela imunohistoquímica para as proteínas ligantes de cálcio calbindina (CB), calretinina (CR) e parvalbumina (PV). Em que pese a quantidade crescente de estudos focando nas proteínas ligantes de cálcio, o córtex préfrontal de primatas ainda permanece relativamente pouco explorado, especialmente no que se refere a um melhor entendimento da organização do circuito inibitório ao longo de suas subdivisões. No presente estudo caracterizamos a morfologia e a distribuição desse grupo neuronal nas regiões medial, orbital e dorso-lateral do córtex pré-frontal do sagui (Callithrix jacchus). Utilizando parâmetros morfométricos e técnicas estereológicas, evidenciamos que os neurônios reativos a CR (especialmente células em duplo-buquê e bipolares) possuem arborização dendrítica mais complexa quando comparados aos neurônios reativos a CB (neurônios de tufos duplos e células em cesto) e PV (células em candelabro). A densidade dos neurônios reativos a CB e CR é mais elevada nas camadas supragranulares (II/III), enquanto os neurônios reativos a PV se concentram predominantemente nas camadas infragranulares (V/VI). Os neurônios reativos a CR foram o grupo predominante nas três regiões avaliadas, sendo mais prevalente na região dorsolateral.
