Iron and zinc are two trace elements that are essential for rice. But they are toxic at higher concentrations, leading to severe rice yield losses especially in acid soils and inland valleys. In this study, two reciprocal introgression line (IL) populations sharing the same parents were used with high-density SNP bin markers to identify QTL tolerant to iron and zinc toxicities. The results indicated that the japonica variety 02,428 had stronger tolerance to iron and zinc toxicities than the indica variety Minghui 63. Nine and ten QTL contributing to iron and zinc toxicity tolerances, respectively, were identified in the two IL populations. The favorable alleles of most QTL came from 02,428. Among them, qFRRDW2, qZRRDW3, and qFRSDW11 appeared to be independent of genetic background. The region C11S49–C11S60 on chromosome 11 harbored QTL affecting multiple iron and zinc toxicity tolerance-related traits, indicating partial genetic overlap between the two toxicity tolerances. Our results provide essential information and materials for developing excellent rice cultivars with iron and/or zinc tolerance by marker-assisted selection (MAS).

Desde o século quinze que é tradicionalmente cultivado trigo na Ilha da Madeira. Em sequeiro, sob condições microclimáticas diversificadas, em terraços isolados e dispostos em encostas íngremes, o trigo ocupa solos ácidos, sobretudo da classe dos Andossolos, com elevada concentração de ferro. Nestes solos, os óxidos de ferro constituem o principal agente inorgânico de ligação das partículas do solo, contribuindo para a estabilidade dos agregados. Foi realizada uma experiência com sessenta cultivares tradicionais de trigo (landracesdeTriticumspp.) do Banco de Germoplasma, da Universidade da Madeira, em soluções nutritivas com 10 (controlo) ou 600 mM Fe, durante 5 dias, após 2 dias de crescimento inicial das plântulas. O alongamento da principal raiz primária foi medido para cada planta sob stress e calculada a média do incremento relativo ao controlo (como percentagem de Incremento Radicular Relativo ou IRR; n = 28). O IRR evidenciou ser um indicador sensível da toxicidade do ferro em trigo. Mais de 85% do germoplasma estudado evidenciou IRR superior a 50%, enquanto o IRR correspondente a setelandracesultrapassou o valor de 70%. Isto foi considerado como indicador da maior tolerância ao excesso de ferro destaslandracesde trigo, as quais poderão ter especial interesse na ocupação de solos ricos em compostos inorgânicos de ferro, nas zonas tropicais e subtropicais., Since the fifteen century, the rainfed-cultivation of wheat for grain is traditionally performed on the Island of Madeira. Under several microclimatic conditions and along very sloppy mountains, the landraces are grown on isolated terraces of Andosols with high amounts of iron. Iron oxides are the main inorganic binding agent contributing to the stability of aggregates and to soil fertility in long-term sustainable agriculture in acid and iron-rich soils. After a two day period of seedling initial growth, a screening test of sixty traditional wheat (Triticumspp.) landraces from the ISOPlexis Genebank at the University of Madeira, Funchal, was performed using nutrient solutions containing 10 or 600 mM Fe, during five days, under controlled laboratory conditions. The elongation of the longest primary root was measured for each genotype and the mean root increment relative to control (as, % relative root increment or RRI; n=28) calculated. This parameter appeared to be a sensitive indicator of Fe tolerance in wheat. Over 85% of wheat germplasm showed the RRI higher than 50%, while the RRI of seven accessions exceeded 70%. This indicates that those landraces are Fe tolerant and might be of particular interest for cultivation under acid rich iron soils of tropical and subtropical areas.

Foi estudado o comportamento diferencial de dez cultivares de trigo em soluções nutritivas com e sem arejamento, contendo cinco concentrações de ferro (0 56; 5,0; 10,0; 20,0 e 40,0mg/litro), mantendo-se constante a temperatura, 27 ± 1ºC, e o pH das soluções, 4,0. A tolerância foi avaliada pelo comprimento médio da raiz primária central e pelo peso da matéria seca da parte aérea (folhas) de vinte plântulas cultivadas dez dias em soluções nutritivas contendo diferentes concentrações de ferro. A medida que foram aumentadas as concentrações de ferro de 0,56 para 40,0mg/litro nas soluções tanto arejadas como sem arejamento, ocorreram reduções nos comprimentos das raízes e nos pesos da matéria seca da parte aérea, segundo o grau de tolerância de cada cultivar. 'Siete Cerros' 'CNT-8' e 'Alondra-4546' foram considerados tolerantes a níveis crescentes de Fe nas duas soluções, e, 'IRN-204-63', 'IAC-13', 'BH-1146', 'IAC-5', 'IAC-17 ', 'IAC-24' e 'IAC-18', sensíveis. Os teores de fósforo, potássio, magnésio, cálcio e manganês na matéria seca da parte aérea dos cultivares BH-1146 e Siete Cerros tenderam a diminuir com o aumento das concentrações de ferro em ambas as soluções nutritivas. Os teores de ferro na matéria seca da parte área dos cultivares BH-1146 e Siete Cerros elevaram-se com o aumento das concentrações de ferro nas soluções, sendo maiores nas soluções arejadas. A metodologia desenvolvida foi eficiente, possibilitando a separação de cultivares tolerantes e sensíveis à toxicidade de ferro em curto período de tempo.Ten wheat cultivars were studied with relation to iron toxicity using five different levels of this element (0.56; 5.0; 10.0; 20.0 and 40.0 mg/l) in nutrient solution with and without aeration, under constant temperature (27ºC ± 1ºC) and pH 4.0. The tolerance of the plants was measured taking into account the mean of the primary root length and the total leaf dry weight of 20 seedlings per treatment solution. The presence of high levels of iron (10.0; 20.0 and 40.0 mg/l) in the solution, with or without aeration, was harmfull to all wheat cultivars under study, but there were differences in growth among the plants, depending on the degree of tolerance of each cultivar. The cultivars Siete Cerros, Alondra 4546 and CNT-8 were tolerant to the increating levels of iron in the aerated and non-aerated nutrient solution and the cultivars IRN-204-63, IAC-5, IAC-18, IAC-17, IAC-13, BH-1146 and IAC-24 were susceptible. The phosphorus, potassium, calcium, magnesium and manganese concentrations decreased in the top part of the plants as the levels of iron increased in the nutrient solution for the cultivars BH-1146 and Siete Cerros. The iron concentration in the leaves of BH-1146 and Siete Cerros increased as the levels of iron increased in the nutrient solution. The aeration in the nutrient solution caused an increase in the leaf iron concentration.

Foram estudados onze cultivares de arroz em soluções nutritivas, arejadas e não arejadas, contendo diferentes concentrações de ferro e alumínio, mantendo-se constante a temperatura de 30 ± 1ºC e o pH das soluções a 4,0. A tolerância foi medida pelo comprimento médio da raiz primária, peso seco da parte aérea e raízes de vinte plântulas cultivadas durante dez dias em solução nutritiva contendo diferentes concentrações de ferro e alumínio. Com as concentrações de 0; 20 e 40mg/litro de Al3+ nas soluções com ou sem arejamento, em presença de 0,56mg/litro de Fe, verificou-se, pelo, crescimento radicular, que os cultivares IAC-164, IAC-165, IAC-47, IAC-25, IAC-435, IAC-120, Blue Bonnet e Pérola foram tolerantes e, IR-8, IAC-899 e IR-841, sensíveis. Nas soluções arejadas sem Al onde se adicionaram 200mg/litro de Fe, o 'IAC-164' foi o que apresentou maior crescimento das raízes, diferindo dos cultivares IAC-435, IAC-899, Blue Bonnet, IAC-25, IR-8 e IR-841, mas não dos cultivares IAC-120, IAC-47, IAC-165 e Peróla. Nas soluções não arejadas, o mesmo resultado foi obtido, com exceção do 'IAC-164' que não diferiu do 'IAC-25'. Nas soluções arejadas sem Al, contendo 400mg/litro de Fe, o cultivar mais tolerante foi IAC-164, diferindo somente, porém, dos cultivares IAC-899, IR-841 e IR-8, considerando o crescimento das raízes. Nas soluções não arejadas sem Al empregando-se 400mg/litro de Fe, não ocorreram diferenças significativas para tolerância entre os cultivares estudados. O 'IAC-164' mostrou grande tolerância à toxicidade de Fe e Al, mesmo nos tratamentos onde foram empregados 20mg/litro de Al3+ combinados com 200mg/litro de Fe, e 40mg/litro de Al3+ combinados com 400mg/litro de Fe. Os cultivares tolerantes ao Al, quando plantados m soluções com 0,56mg/litro de Fe e doses crescentes de Al, mostraram que os teores de Al na matéria seca da parte aérea aumentaram, sobretudo nas soluções arejadas. Nessas condições, os teores de P, Ca, Mg, Fe, Cu, Zn e Mn tenderam a diminuir e, os de K, a aumentar, à medida que se aumentaram as concentrações de Al nas soluções, com ou sem arejamento. Os cultivares sensíveis ao Al mostraram aumento nos teores de P, Fe e Al na matéria seca da parte aérea e redução nos teores dos demais nutrientes, à medida que se elevaram as concentrações de Al nas soluções. Com o aumento nas concentrações de Fe nas soluções em ausência de Al, os cultivares estudados mostraram tendência de diminuição dos teores de Ca, Mg, Cu e Zn, na matéria seca da parte aérea e elevação dos de K e Fe, com pouca variação nos teores de P. Os resultados obtidos demonstraram que os cultivares de arroz estudados poderiam ser diferenciados em relação à tolerância à toxicidade de Fe e Al, desde que concentrações adequadas desses elementos fossem utilizadas nas soluções nutritivas, com ou sem arejamento.Eleven rice cultivars were studied as the tolerance to iron and aluminum toxicity using seven different levels of these elements in the nutrient solutions with and without aeration under constant temperature (30ºC ± 1ºC) and pH = 4.0. In aerated nutrient solutions containing 400mg/l of Fe the most tolerant cultivar was IAC-164 which differed significantly only from IAC-899, IR-841 and IR-8. When non-aerated solution was used, no differences were found among the studied cultivars. 'IAC-164' presented high tolerance to Al and Fe toxicities even when it was applied 20mg/l of Al plus 200mg/l of Fe and 40mg/l of Al plus 400mg/l of Fe in the solutions. The Al and K concentrations increased and the P, Ca, Mg, Fe, Cu, Zn and Mn concentrations decreased in the leaf dry matter for all aluminum tolerant cultivars, as the aluminum concentration in solution increased from 0 to 40mg/l. When the cultivars sensitive to Al toxicity were considered increasing levels of Al in solution caused an increase in Al, Fe and P concentrations, and a decrease in the other element concentrations in the leaf dry matter. The Fe and K concentrations increased, Ca, Mg, Mn, Cu and Zn concentrations decreased and no effect in P concentration was observed in the total leaf dry matter for all cultivars, when Fe concentration in nutrient solution increased from 0 to 400mg/l in absence of Al. The results showed that the studied rice cultivars could be separated in relation to Al and Fe toxicities in nutrient solution with and without aeration, since adequate quantities of Al and Fe are provided.
