Abstract

La criopreservación de espermatozoides y su utilización en inseminación artificial ha causado gran impacto sobre la reproducción de especies comerciales y en humanos. Sin embargo, a pesar del gran avance en el tema, constantemente se modifican los procedimientos de congelamiento, a fin de obtener mejores resultados de viabilidad y movilidad espermática, para lograr de esta forma mayores porcentajes de fecundaciones exitosas. No obstante, el proceso de criopreservación induce perdida de la función espermática que se refleja en alteraciones de la membrana plasmática, daño a nivel mitocondrial, deterioro en la integridad del ADN, aumento en la producción de las especies reactivas de oxígeno (ROS) y alteraciones del citoesqueleto, lo que finalmente tiene efectos negativos en la viabilidad y motilidad espermática, elementos esenciales para la fecundación. En la búsqueda de métodos que induzcan el menor daño celular, es que ha desarrollado en los últimos años los métodos de vitrificación espermática. Estos han permitido no solo superar en cuanto a la mantención de la función espermática a los métodos convencionales de congelación, sino también que lo hace más rápido, seguro y de menor costo. Esto último al eliminar los equipos necesarios para la congelación lenta y también en algunos casos la no utilización de nitrógeno líquido para el almacenamiento, bastando un congelador de - 80°C. Su aplicación en mamíferos, ha sido reportada exitosamente en humanos con nacimientos de niños sanos, tanto en técnicas de fecundación in vitro como inseminación intrauterina y recientemente con el nacimiento de los primeros felinos (gatos) con técnicas de inseminación intrauterina.

Abstract

Background: This study aims to evaluate the effectiveness of Tumor Dissociation Enzyme (TDE) for the isolation of cryopreserved human ovarian cortex tissues, proposing an optimized applicable for artificial ovary.Methods: This is a prospective experimental study. We present a comparative analysis among the outputs of follicles isolated from cryopreserved ovarian biopsies of ten young women undergoing laparoscopy. Follicles were isolated by Tumor Dissociation Enzyme (TDE) or Liberase Dispase High (DH) enzymatic digestion. Follicles were assessed by evaluating the number, viability, morphology, oxidative stress. Moreover, the follicle growth and viability were analyzed after eight days of in vitro culture (IVC) between each protocol.Results: The recovery rate of follicles in Group 1 was significantly higher by 78 follicles compare to Group 2 (p < 0.05), while the difference in weight and volume of biopsies in both groups was no significant (p > 0.05). Group 1 allowed to isolate more primordial follicles (p < 0.05) and smaller diameter of follicles (p < 0.01) than Group 2. Group 1 allowed to isolate 7% more of bright red follicles than Group 2 (p < 0.01). We also found that Group 1 had isolated a significantly higher percent of viable follicles (p < 0.05), more morphologically normal follicles (p < 0.05), and lower oxidative stress levels compared with Group 2 (p < 0.01). The velocity of follicles growth from Day 0 to Day 8 in Group 1 was significantly higher (p < 0.05) than in Group 2. The viability of follicles on Day 8 of in vitro culture in Group 1 was significantly higher than Group 2 (p < 0.05).Conclusions: TDE treatment can be an alternative for Liberase DH, allows the isolation of highly viable follicles from the cryopreserved human ovarian cortex, with an intact morphology and low oxidative stress, and with high proliferation potential after culture in vitro.

Abstract

Short-term storage of semen is a useful strategy for preservation of fish spermatozoa. However, there is a significantly decrease on sperm function mainly due to oxidative stress. In this way, sodium alginate plays an important role as free radical scavenger compound. Accordingly, the aim of our study was to analyse the effect of a sodium alginate-based extender on sperm function in the short-term storage of salmonids semen. Samples of Salmo salar, Oncorhynchus kisutch, and Oncorhynchus mykiss were stored in Storfish® (Ext-C) and Storfish® supplemented with sodium alginate (Ext-A) during 10 days at 4°C. After storage, motility, viability, mitochondrial membrane potential (ΔΨmit), superoxide anion (O2−) level and DNA fragmentation ( DNA Frag) were assessed. Ext-A had positive effect in preservation of sperm motility, viability, ΔΨmit, O2− level and DNA integrity in the three species analysed compared to control samples. In Ext-A, the spermatozoa of S. salar and O. mykiss showed significantly higher motility, viability and ΔΨmit than O. kisutch. However, O. kisutch and O. mykiss had significantly lower O2− level than S. salar, and DNA fragmentation in O. kisutch and S. salar was significantly lower than in samples of O. mykiss ( p < 0.05). Dilution of salmonids semen in a sodium alginate-based extender is effective for protecting sperm quality during 10 days of short-term storage. [ABSTRACT FROM AUTHOR]
