Abstract

Objective: Urine is an ideal source of biomarkers, but data on cat urine proteome is still very limited. The aim of our study is to apply proteomic techniques to identify putative biomarkers of nephropathy in cats affected by chronic kidney disease (CKD). Design and Methods: Urine samples were collected by cystocentesis from 4 healthy and 4 cats affected by CKD and analyzed by two-dimensional electrophoresis (2DE). The first dimension was performed by isoelectric focusing on 17 cm long IPG strips (pH 3-10); the second dimension was performed on 10% SDS-PAGE and stained with colloidal Coomassie. Spots were excised from the gel, reduced, alkylated and digested with trypsin and identified using ESI-Q-TOF mass spectrometry. Results: 2DE allowed the separation of 66 spots in the urine proteome of healthy and CKD cats. Twenty-one spots were successfully identified. In CKD samples, Uromodulin and Carboxylesterase-5A showed a significant reduction, supporting the tubular involvement in the pathology; Perlecan, a negatively charged proteoglycan showed no visible expression, suggesting its role in maintaining the integrity of the glomerular filtration barrier. On the other hand, 3 low molecular weight proteins, Retinol binding protein, Immunoglobulin K light chain and Complement factor D, increased significantly in CKD cats, confirming tubulo-interstitial involvement in cats affected by this disease. Conclusions: 2DE was successfully applied to the separation of cat urine proteome leading to the identification of 6 differentially expressed proteins; our data suggest their use as putative biomarker of nephropathy in cats and possibly in other veterinary species.

Abstract

The European eel Anguilla anguilla has declined significantly over recent decades, exceeding its safe biological limits and probably reaching a historical minimum (1% of the 1960 recruitment level). Twenty-three migrating females at the IV stage of silvering were collected from the Comacchio lagoon (Northern Italy); they showed high levels of cholesterol and triglycerides, probably due to lipid accumulation before migration across the Atlantic Ocean. Electrophoretic patterns showed high amounts of putative α and β globulins, probably due to high concentrations of lipoproteins. Genetic results obtained from a total of 49 specimens (20 from the Comacchio lagoon and 29 from Val Dogà) agree with the panmixia hypothesis, lacking any genetic differentiation over time and space at neutral loci. In conclusion, despite the small sampling and the population decline in recent years, high levels of variability were recorded. [ABSTRACT FROM AUTHOR]

1. 1. The origin of carbon incorporated in succinate in the first 12 hr of environmental anoxia was studied in two bivalve species of the Northern Adriatic Sea. 2. 2. Posterior adductor muscles were excised after various periods of exposure to anoxic seawater and incubated under nitrogen in the presence of metabolic inhibitors (aminooxyacetate, iodoacetate, 3-mercaptopicolinate) and UL-14C substrates, in order to examine the redirection of glycolytic flux from pyruvate kinase to phosphoenolpyruvate carboxykinase. 3. 3. Changes in the levels of key metabolites and the distribution of radiolabeled glucose and aspartate indicate that PEP-carboxykinase did not participate in anaerobic metabolism of these species for at least the initial 12 hr of anaerobiosis., 1. 1. Abstract

Lead concentrations in blood of 45 specimens of mute swan from the molting area of the Isonzo River Mouth Nature Reserve (Italy) were determined in two consecutive years (2006-2007), some birds were neck ringed to identify their homing behavior. The second sampling included whole body X-ray radiography and Cu and Zn plasma analyses to investigate the health impact of putative Pb exposure. X-ray images of all investigated specimens did not show any radiopacity due to the ingestion of metal bodies. Lead levels (0.08-0.44 µg/ml) were in the range of those reported for swans living in unpolluted or slightly polluted environments and excluded acute intoxication, as confirmed by clinical investigation. Zinc concentrations ranged between 2.93 and 7.59 µg/ml and were one order of magnitude higher than Cu concentrations (0.21-0.42 µg/ml). The negative correlation between Pb and Zn concentrations could be indicative of adverse health effects caused by chronic lead exposure. To our knowledge this is the first study reporting Pb, Zn and Cu blood levels, X-ray radiographies and data on the origin of swan populations. [ABSTRACT FROM AUTHOR]

43. Selective enrichment in human megakaryocyte progenitors by the B203.13 surface differentiation antigen

Abstract

The B203.13 monoclonal antibody specifically recognizes a subset (8–18.5%) of human CD34+ haemopoietic progenitors, which are significantly (P < 0.05) enriched in megakaryocyte progenitors (CFU-meg). Moreover, B203.13 expression was progressively lost as maturation proceeded along the megakaryocytic lineage. In fact: (i) CFU-meg derived from CD34+/B203.13+ showed a greater number of cells/colony with respect to CD34+/B203.13−; (ii) after 10 d of liquid cultures with 100 ng/ml of thrombopoietin, only a minority of CD34-derived cells positive for CD41 coexpressed B203.13; (iii) in situ immunocytochemical staining revealed that B203.13 was expressed exclusively on immature megakaryoblasts; (iv) circulating platelets did not express B203.13.
