Abstract

The importance of site selection for road transportation management and emergency services has been underscored, particularly in the wake of the global Covid-19 pandemic in addition to road accident injuries, which emphasized the need for swift patient, injured and wounded peoples transfer to hospitals. As a result, comprehensive road transportation management is essential during crises to ensure the safety and convenience of road users. To achieve effective road network management, a robust database is imperative. The aim of this study is to employ multi-criteria decision analysis with the aid of Geographical Information System (GIS) technology and remote sensing for the optimization of site selection for traffic signals, fire stations, and emergency services in Kafr El-Sheikh city, Egypt. The methodology incorporates two different methods which are Fuzzy Analytical Hierarchy Process (F-AHP) and the spline transformation method. The integration of Fuzzy set theory with AHP principles has given rise to a variant of AHP known as F-AHP. F-AHP combines qualitative and quantitative approaches. Fuzzy logic handles situations that are vague or poorly defined by assigning numerical values. Through the F-AHP approach, criteria that decision-makers unanimously deem “absolutely not important” in comparison to others are eliminated, allowing them to focus on the most significant criteria. As a case study, an Egyptian city was selected to apply these techniques. The results revealed multiple sites that were identified and selected through the applied analytical methods. These selected sites were also marked on the city map and verified in reality.

8. The effect of PTEN silencing on motor neuron survival and neuromuscular degeneration in experimental models of amyotrophic lateral sclerosis

Abstract

Amyotrophic Lateral Sclerosis (ALS) is a fatal neurodegenerative disease characterised by progressive loss of upper and lower motor neurons, which ultimately leads to death within 2-3 years of diagnosis. While multiple mechanisms have been proposed to cause motor neuron death, altered signalling of the pro-survival phosphatydyl-inositol-3-kinase (PI3K/Akt) signalling pathway has been shown in spinal cords of human cases and rodent models of ALS. The depletion of the phosphatase and tensin homologue (PTEN), an essential negative regulator of PI3K, promotes neuronal survival and axonal regeneration. The main aim of this study was to investigate the effect of PTEN silencing using viral vectors, on motor neuron survival and neuromuscular degeneration (NMJ) in experimental models of ALS. Lentiviral vectors expressing small interfering RNA (siRNA) against mouse PTEN achieved approximately 60% PTEN knockdown in purified motor neurons from transgenic SOD1G93A embryos and their non-transgenic littermates. PTEN silencing was associated with increased expression of phosphorylated Akt, and enhanced motor neuron survival. To validate the potential neuroprotective effect of PTEN silencing in vivo, wild-type (WT) mice were used to determine the efficiency of adeno-associated virus serotypes 6 (AAV6) and 9 (AAV9), to deliver PTEN siRNA to spinal cord motor neurons after intramuscular (i.m) administration. Despite effective transduction and PTEN knockdown in the injected muscles, low levels of spinal cord motor neuron transduction were observed for both vectors suggesting poor retrograde transport efficiency. Similar findings were seen in transgenic SOD1G93A mice treated with AAV9 vectors expressing PTEN siRNA, which may explain the lack of significant effect seen on spinal cord motor neuron survival. In addition, no significant effect was seen on neuromuscular junction degeneration in the muscles of the injected hind limbs. In conclusion, these results suggest that viral-mediated PTEN silencing may enhance the survival of transgenic SOD1G93A motor neurons by activating the PI3K/Akt pathway. However, the i.m route for AAV-mediated siRNA delivery to the spinal cord is insufficient for effective PTEN silencing. Alternative CNS delivery routes are needed for proof-of principle studies to determine the therapeutic efficacy of PTEN silencing ALS.

29. Challenging inflammatory process at molecular, cellular and in vivo levels via some new pyrazolyl thiazolones

Abstract

The work reported herein describes the synthesis of a new series of anti-inflammatory pyrazolyl thiazolones. In addition to COX-2/15-LOX inhibition, these hybrids exerted their anti-inflammatory actions through novel mechanisms. The most active compounds possessed COX-2 inhibitory activities comparable to celecoxib (IC50 values of 0.09–0.14 µM) with significant 15-LOX inhibitory activities (IC50s 1.96 to 3.52 µM). Upon investigation of their in vivo anti-inflammatory activities and ulcerogenic profiles, these compounds showed activity patterns equivalent or more superior to diclofenac and/or celecoxib. Intriguingly, the most active compounds were more effective than diclofenac in suppressing monocyte-to-macrophage differentiation and inflammatory cytokine production by activated macrophages, as well as their ability to induce macrophage apoptosis. The latter finding potentially adds a new dimension to the previously reported anti-inflammatory mechanisms of similar compounds. These compounds were effectively docked into COX-2 and 15-LOX active sites. Also, in silico predictions confirmed the appropriateness of these compounds as drug-like candidates.

Abstract

Psychological stressors have been reported to precede the onset of psoriasis in 44% of patients, initiate recurrent skin flares in 88% of psoriatic and may worsen psoriasis severity and may even lengthen the time to disease clearance. This study aimed to: 1. Identifying the types of stressors, which are related to the forms and severity of clinical profile of psoriasis, 2. Identifying the types of psychosocial factors which are related to the forms and severity of clinical profile of psoriasis in Egyptian sample. Methods: One hundred and fifty patients randamised samples were collected from dermatological out-patient clinic affiliated to ministry of health and universities hospitals. All subjects were exposed to the followings: A full dermatological history including exacerbating factors. General examination to exclude chronic diseases. Clinical examination, Socioeconomic data will be collected for all patients. Holms and Rahe for assessment of stressors. Psoriasis area severity index. Results: The socio-demographic characteristics show that the mean age of patients with psoriasis of both sexes was 40.91 (standard deviation, SD 14.26) years, while the men:women ration was 2.66:1. Additionally, 68% of psoriatic patient were occupationally skilled person; 18% were un-employed; 10% were professional and 4% were students. A significant relation between weather condition and type of psoriasis severity was noticed. It appears that psoriatic patients were improved in summer and worsened in winter. During autumn and springs, patients didn't notice changes in psoriasis signs. A significant relation between presence of psychological stress and degree of psoriasis severity. It appears that moderate degree of psoriasis was the most common among psoriatic patient with stress (45.5%) or without stress (65.3%). Conclusion: The study showed that environmental stressors have relevant effects on severity of psoriasis. In addition, there is a specific relation between both climate and psychological stresses on the severity of psoriasis.
