In addition to the monographs which have been published in the past 4 years by the working group 'Drug Monitoring' of the Swiss Society of Clinical Chemistry (SSCC) [1-4], new monographs have been written. The data presented in these monographs provide an overview of important information for the request and interpretation of results. Therefore, laboratory health professionals and the receivers of the reports are the targeted readers. In this series, several antiepileptic drugs are presented. Monographs on carbamazepine [1], lamotrigine [2], phenobarbital [2], and valproic acid [2] have been published previously. First, information about pharmacology and pharmacokinetics of these drugs (protein binding, metabolic pathways and enzymes involved, elimination half-life time and elimination route(s) of the parent drug and therapeutic as well as toxic concentrations) is given. Second, the indications for therapeutic drug monitoring are listed. Last but not least, important pre-analytical information is provided, including time points of blood sampling and time interval after which steady-state concentrations are reached after changing the dose. Furthermore, the stability of the drug and its metabolite(s) after blood sampling is described. For readers with a specific interest, references to important publications are given. The number of the monographs will be further enlarged. The updated files are presented on the homepage of the SSCC (www.sscc.ch). We hope that these monographs are helpful for the better handling of therapeutic drug monitoring and we are looking forward to comments from the readers. [ABSTRACT FROM AUTHOR]

Red foxes (Vulpes vulpes) are a major pest species in Europe and Australia. Traditional methods of control such as hunting or poisoning are no longer sufficient or feasible. As with domestic dogs and cats, prolactin (PRL) in the vixen is an essential luteotropin during the second half of gestation. Hence, PRL inhibitors such as cabergoline have been used to induce abortions. Eighteen mated silver fox vixens (three groups of six foxes each) were treated orally with a placebo of paraffin oil (I), or with 15 μg/kg cabergoline in feed once (II) or twice (III), on day 30 (I and II) or days 30 and 32 (III) post-coitum. Blood samples were taken prior to and after treatments and concentrations of PRL and progesterone (P4) were determined. Normal parturitions were observed in five of six, five of six and two of six vixens in groups I, II and III, respectively. In group III plasma concentrations of PRL and P4 decreased significantly but only temporarily. This drop in hormone concentrations was more pronounced in the vixens that did not carry to term. In conclusion, doses in excess of 15 μg/kg of cabergoline are likely to prevent the development of fetuses to term in pregnant vixens. [ABSTRACT FROM AUTHOR]

In two trials, effects of adding fully acetylated monoglycerides (DFG) (0 to 4%) to ground meat (beef) were evaluated. In both trials, ground beef was subsequently vacuum packaged and stored at 3°, 5° or 10°C for up to 18 days. Samples were taken in 3 or 4-day intervals and examined microbiologically and organoleptically. In trial 1, development of the spontaneous contamination, and in trial 2, growth of added pathogens were determined. In trial 1, refrigerated ground beef without treatment had become unacceptable by day 6. Treatment with 2% DFG provided a prolongation of shelf life for 10 to 16 days at 3°C or 5°C. Higher concentrations of DFG (3 or 4%) resulted in unpleasant high levels of acidity in odor and taste after 10 days, without additional benefits. DFG treatment left the development of the desirable lactobacilli undisturbed, but inhibited the growth of enterobacteriaceae. In trial 2, ground beef was spiked prior to DFG treatment and vacuum packaging with known amount of E. coli, Enterobacter aerogenes and cloacae, Yersinia enterocolitica and Salmonella species. Samples were stored at 5° and 10°C. DFG (2%) showed significant inhibitory effects on the growth of E. coli (p <.05), little effects on Enterobacter aerogenes at 5°C, but clearly at 10°C (p <.05 -.005), no effects on E. cloacae at 5° and 10°C (p <.01 to.0005) and Salmonella spec., especially at 10°C (p <.005). Zusammenfassung Lagerung von vakuumverpacktem Hackfleisch nach Behandlung mit acetylierten Monoglyzeriden: Einfluß von Temperatur auf natürlich vorkommende oder künstlich mikrobielle Kontamination und Lagerfähigkeit In zwei Studien wurde die Wirkung von 0-4% vollständig azetyliertem Monoglyzerid (DFG) auf Hackfleisch vom Rind untersucht, das nach Einmischung des DFG vakuumverpackt und bei 3, 5 oder 10°C bis zu 18 Tagen gelagert wurde. Proben wurden in 3- bis 4tägigem Abstand organoleptisch und mikrobiologisch untersucht. Dabei wurde in Studie 1 die Entwicklung spontan vorkommender Flora, in Studie 2 von zugesetzten pathogenen Keimen untersucht. In Studie 1 erwies sich unbehandeltes Hackfleisch nach 6 Tagen als nicht mehr akzeptabel. Mit 2% DFG behandeltes Hackfleisch erwies sich über 10 bis 16 Tage haltbar. DFG in höheren Konzentrationen (3 und 4%) verursachte einen starken essigsäure-artigen Geruch und Geschmack, insbesondere nach 10 Tagen, ohne deswegen besondere Vorteile zu bieten. Während die Entwicklung der Laktobazillen-Flora ungestört verlief, war die von Enterobakterien gehemmt. Deswegen wurde in Studie 2 Hackfleisch mit bekannten Mengen von pathogenen Keimen versetzt, mit 1 bzw. 2% DFG vermischt und bei 5 oder 10°C für 10 bis 13 Tage gelagert. Die 2%ige DFG-Behandlung hemmte die Entwicklung von E. coli bei 5 und 10°C (p < 0.05-0.001), die von Enterobacter aerogenes bei 10°C (p < 0.005), die von Enterobacter cloacae nicht, dafür aber die von Yersinia enterocolitica bei beiden Temperaturen (p < 0.005 und 0.0005) und die von Salmonella spec. bei 10°C (p < 0.005). Histological evidence is presented from biopsies taken from burn wounds and surgical lesions in horses, treated double blind with Solcoseryl cream 5%, the Solcoseryl Fraction 3 (4 x) or (8 x) in a cream base, and Placebo cream. Solcoseryl treatment speeded up and enhanced tissue repair, i. e. wound healing. Effects of Solcoseryl were characterized by an initially enhanced basophilic infiltration of the wound bed, but also an earlier decline in perivascular infiltration. Collagen fiber formation and arrangement occurred earlier, almost two weeks ahead of placebo treatment. Epithelization was well synchronized with advanced stages of tissue repair. This synchronization was less good and tissue repair in general less well organized in placebo treated animals. Effects of Fraction 3 (8 x) differed from Solcoseryl by showing less infiltration and less fiber forming activities, but showed earlier signs of fiber arrangement (Scaron formation) and earlier epithelization. These results are in good agreement with highly significant clinical observations on wound size reduction over time and the clinical impression on epithelization (wound closure) in the same animals. Zusammenfassung Histologische Untersuchung der Wundheilung bei Pferden nach Behandlung mit dem proteinfreien Hämodialysat Solcoseryl und dessen Hexosylceramid-Fraktion In einer Doppelblind-Studie wurden Biopsien von chirurgischen Läsionen und Brandwunden bei Pferden histologisch untersucht, die mit einer 5%igen Solcoseryl Creme, einer Creme mit der Solcoseryl Fraktion 3 (4 x) oder (8 x), oder mit einer Plazebo Creme behandelt worden waren. Die Behandlung mit Solcoseryl und mit der Fraktion 3 begünstigte die Gewebs-Reparaturvorgänge und verkürzte damit die Zeit für die Wundheilung. Solcoseryl zeigte seine Wirkungen zuerst durch eine verstärkte basophile Infiltration des Wundbettes, aber auch eine frühere Reduzierung perivaskulärer Infiltration. Die Kollagenfaser-Bildung und deren typische Organisation waren etwa zwei Wochen früher als mit dem Plazebo erkennbar. Epithelisierung war gut mit der rascher fortschreitenden Gewebsreparatur synchronisiert. Mit dem Plazebo war diese Synchronisation deutlich weniger gut ausgeprägt und die Gewebsreparatur-Vorgänge waren weniger gut organisiert. Wirkungen der Fraktion 3 (8 x) unterschieden sich vom Solcoseryl durch geringere Infiltration als auch geringere Kollagenfaser-Bildung, dafür waren Faser-Anordnung und Epithelisierung früher erkennbar. Diese Ergebnisse stimmen gut mit den hochsignifikanten klinischen Beobachtungen über den Ablauf des Wundverschlusses bei den gleichen Tieren überein. [ABSTRACT FROM AUTHOR]

Adding 3% acetylated monoglycerides* (DFG) to beef cuts during the process of packing into vacuum sealed Cryovac (CR) or laminated polyethylene (LP) film pouches prolonged freshness considerably when stored at + 2 °C, as tested by organoleptic and microbiological examinations and consumption. A fixation method for tissue surface samples was developed, using osmium tetroxide, which permitted study of the coating of beef cuts with DFG by scanning electron microscopy (SEM). SEM revealed patterns of the DFG coating's deterioration related to the quality of the oxygen barrier system used and also to hygenic conditions at processing. DFG forms an invisible coat which, where intact, prevents purge (meat juice release) and delays deterioration, but does not hinder maturation of meat. Film quality and hygienic conditions at processing and packing determine the duration of the DFG effect: in CR, and with minimum contamination, cuts can stay fresh for up to 10 weeks; and in LP up to 8 weeks. Under commercial conditions, DFG treated cuts retained freshness for 50 days in CR, and for 35 days in LP. Scores on the condition of the DFG coating over time, as provided by SEM, and from simultaneous microscopic examinations of the fluid retained in the barrier bags, were well correlated with each other and with the effectiveness of DFG under the differing conditions. Zusammenfassung Verlängerung der Haltbarkeit und Reduktion des Gewichtsverlustes durch Fleischsaftaustritt bei Fleischstücken nach Überzug mit azetylierten Monoglyceriden Der Zusatz von 3% azetylierten Monoglyceriden* (DFG) zu Fleischstücken wáhrend der Verpackung in vakuumdichten Cryovac- (CR) oder in geschichteten Polyäthylen- (P) Beuteln verlängert bei einer Lagertemperatur von + 2 °C die Haltbarkeit dieser Proben beträchtlich. Dieses Ergebnis wird anhand organoleptischer und mikrobiologischer Untersuchungsverfahren sowie durch rasterelektronenmikroskopische Strukturanalysen der Fleischoberflächen erhoben. Die mit Hilfe der Rasterelektronenmikroskopie dokumentierte Struktur von DFG während seines allmählichen Zerfalles als Schutzschicht weist einerseits auf die Bedeutung des Sauerstoff-Barriere-Systems, andererseits auf die hygienischen Bedingungen bei der Fleischverarbeitung. DFG bildet einen durchsichtigen Fleischüberzug, der - solange dieser geschlossen bleibt - den Austritt von Fleischsaft verhindert und das Verderben des Fleisches verzögert, ohne jedoch die Fleischreifung zu beeinträchtigen. Dabei bestimmen die Qualität der Verpackungsfolie und die fleischhygienischen Bedingungen bei der Vorbereitung und der Verpackung der Fleischstücke die Dauer der Wirksamkeit von DFG: in CR-Folie und bei minimaler Kontamination können Fleischstücke bis zu 10 Wochen frisch gehalten werden, in LP-Folie bis zu 8 Wochen. Unter kommerziell üblichen fleischhygienischen Bedingungen werden DFG vorbehandelte Fleischproben in CR-Folie bis zu 50 Tage, in LP-Folie' bis zu 35 Tage als genußtauglich angenommen.
