The aim of this study was to find and use a heterogeneous catalyst to catalyze the transesterification reaction of biodiesel. The purpose of this is to have a more sustainable production of biodiesel which uses fewer chemicals and can reuse the catalyst easily. Our team chose a base catalyst of calcium oxide (CaO) based on early research but with a change of doping the base catalyst with sodium, thus making sodium doped calcium oxide (Na-CaO). The catalyst was synthesized using calcium hydroxide and sodium hydroxide as precursors. The sodium concentration was chosen to be at 3 weight%. The main part of the study was on testing the efficiency of the catalyst and determining at which conditions the reaction performs best. The best conditions for the catalyst were found at 60 °C and the ratio of rapeseed oil to methanol was 1:9. The mass concentration of the catalyst was 2 weight% and the reaction time was 2 hours. These conditions resulted in a biodiesel yield of 97,6 %. Studien som gjordes gick ut på att tillverka en heterogen katalysator för att katalysera transesterifiering reaktionen för biodieseltillverkning. Syftet med detta är att ha en mer hållbar produktion som använder sig utav färre kemikalier och samtidigt kan återanvända katalysatorn. Valet av katalysator skedde efter värdering av äldre studier och basen för katalysatorn valdes till kalciumoxid (CaO), med en ändring av att dopa denna med natrium, det vill säga natriumdopad kalciumoxid (Na-CaO). Katalysatorn syntetiserades utav kalciumhydroxid och natriumhydroxid. Koncentrationen av natriumet låg vid 3 vikt%. Nästa del utav studien gick ut på att testa den syntetiserade katalysatorn och hitta de bästa betingelserna för bästa utbyte av biodiesel. De bästa betingelserna var vid temperaturen 60 °C och ett förhållande mellan rapsolja och metanol på 1:9, där masshalten av katalysator var 2 vikts% gentemot oljan och reaktionstiden var 2 timmar. Detta gav ett utbyte på 97,6 %.

Liberalized electricity market conditions and concentrating solar power technologies call for increased power plant operational flexibility. Concerning the steam turbine component, one key aspect of its flexibility is the capability for fast starts. In current practice, turbine start-up limitations are set by consideration of thermal stress and low cycle fatigue. However, the pursuit of faster starts raises the question whether other thermal phenomena can become a limiting factor to the start-up process. Differential expansion is one of such thermal properties, especially since the design of axial clearances is not included as part of start-up schedule design and because its measurement during operation is often limited or not a possibility at all. The aim of this work is to understand differential expansion behavior with respect to transient operation and to quantify the effect that such operation would have in the design and operation of axial clearances. This was accomplished through the use of a validated thermo-mechanical model that was used to compare differential expansion behavior for different operating conditions of the machine. These comparisons showed that faster starts do not necessarily imply that wider axial clearances are needed, which means that the thermal flexibility of the studied turbine is not limited by differential expansion. However, for particular locations it was also obtained that axial rubbing can indeed become a limiting factor in direct relation to start-up operation. The resulting approach presented in this work serves to avoid over-conservative limitations in both design and operation concerning axial clearances.

Chantier qualité GA; International audience; Legumes (Fabaceae or Leguminosae) are unique among cultivated plants for their ability to carry out endosymbiotic nitrogen fixation with rhizobial bacteria, a process that takes place in a specialized structure known as the nodule. Legumes belong to one of the two main groups of eurosids, the Fabidae, which includes most species capable of endosymbiotic nitrogen fixation1. Legumes comprise several evolutionary lineages derived from a common ancestor 60 million years ago (Myr ago). Papilionoids are the largest clade, dating nearly to the origin of legumes and containing most cultivated species2. Medicago truncatula is a long-established model for the study of legume biology. Here we describe the draft sequence of the M. truncatula euchromatin based on a recently completed BAC assembly supplemented with Illumina shotgun sequence, together capturing ~94% of all M. truncatula genes. A whole-genome duplication (WGD) approximately 58 Myr ago had a major role in shaping the M. truncatula genome and thereby contributed to the evolution of endosymbiotic nitrogen fixation. Subsequent to the WGD, the M. truncatula genome experienced higher levels of rearrangement than two other sequenced legumes, Glycine max and Lotus japonicus. M. truncatula is a close relative of alfalfa (Medicago sativa), a widely cultivated crop with limited genomics tools and complex autotetraploid genetics. As such, the M. truncatula genome sequence provides significant opportunities to expand alfalfa’s genomic toolbox.

In order to improve the startup flexibility of steam turbines, it becomes relevant to analyze their dynamic thermal behavior. In this work, the relative expansion between rotor and casing was studied during cold-start conditions. This is an important property to monitor during startup given that clearances between rotating and stationary components must be controlled in order to avoid rubbing. The investigation was performed using a turbine thermal simplified model from previous work by the authors. The first step during the investigation was to extend and refine the modeling tool in order to include thermomechanical properties. Then, the range of applicability of the model was validated by a twofold comparison with a higher order finite element (FE) numerical model and measured data of a cold start from an installed turbine. Finally, sensitivity studies were conducted with the aim of identifying the modeling assumptions that have the largest influence in capturing the correct thermal behavior of the turbine. It was found that the assumptions for the bearing oil and intercasing cavity temperatures have a large influence ranging between ±25% from the measured values. In addition, the sensitivity studies also involved increasing the initial temperature of the casing in order to reduce the peak of differential expansion. Improvements of up to 30% were accounted to this measure. The studies performed serve as a base toward further understanding the differential expansion during start and establishing future clearance control strategies during turbine transient operation.

In order to increase the flexibility of the turbines while preserving lifting requirements, several operational modifications for maintaining turbine temperatures during offline periods are proposed and investigated. The modifications were implemented in a dynamic thermal turbine model and the potential improvements were quantified. The modifications studied included: increasing the gland steam pressure injected to the end-seals, increasing the back pressure and increasing the barring speed. These last two take advantage of the ventilation and friction work. The effects of the modifications were studied both individually as well as in different combinations. The temperatures obtained when applying the combined modifications were compared to regular turbine cool-down (CD) temperatures and showed significant improvements on the startup times of the turbine.

This paper presents the detailed numerical analysis including parametric studies on the aerodynamic excitation mechanisms in a turbine stage due to the unsteady stator-rotor interaction. The work is part of the pre-design study of a high pressure subsonic turbine for a rocket engine turbopump. The pressure level in such turbines can be remarkably high (in this case 54 MPa inlet total pressure). Hence, large unsteady rotor blade loads can be expected, which impose difficult design requirements. The parameter studies are performed at midspan with the numerical flow solver UNSFLO, a 2D/Q3D unsteady hybrid Euler/Navier-Stokes solver. Comparisons to 2D and steady 3D results obtained with a fully viscous solver, VOLSOL, are made. The investigated design parameters are the axial gap (∼8%–29% of rotor axial chord length) and the stator vane size and count (stator-rotor pitch ratio ∼1–2.75). For the nominal case the numerical solution is analyzed regarding the contributions of potential and vortical flow disturbances at the rotor inlet using rotor gust computations. It was found that gust calculations were not capable to capture the complexity of the detected excitation mechanisms, but the possibility to reduce excitations by enforcing cancellation of the vortical and potential effects has been elaborated. The potential excitation mechanism in the present turbine stage is found dominant compared to relatively small and local wake excitation effects. The parameter studies indicate design recommendations for the axial gap and the stator size regarding the unsteady rotor load.

The genome of tomato (Solanum lycopersicum L.) is being sequenced by an international consortium of 10 countries (Korea, China, the United Kingdom, India, the Netherlands, France, Japan, Spain, Italy, and the United States) as part of the larger “International Solanaceae Genome Project (SOL): Systems Approach to Diversity and Adaptation” initiative. The tomato genome sequencing project uses an ordered bacterial artificial chromosome (BAC) approach to generate a high-quality tomato euchromatic genome sequence for use as a reference genome for the Solanaceae and euasterids. Sequence is deposited at GenBank and at the SOL Genomics Network (SGN). Currently, there are around 1000 BACs finished or in progress, representing more than a third of the projected euchromatic portion of the genome. An annotation effort is also underway by the International Tomato Annotation Group. The expected number of genes in the euchromatin is ∼40,000, based on an estimate from a preliminary annotation of 11% of finished sequence. Here, we present this first snapshot of the emerging tomato genome and its annotation, a short comparison with potato (Solanum tuberosum L.) sequence data, and the tools available for the researchers to exploit this new resource are also presented. In the future, whole-genome shotgun techniques will be combined with the BAC-by-BAC approach to cover the entire tomato genome. The high-quality reference euchromatic tomato sequence is expected to be near completion by 2010.

To expedite the development of personalized medicine, new and reliable biomarkers are required to facilitate early diagnosis, to determine prognosis, predict response or resistance to different therapies, and to monitor disease progression or recurrence. Human body fluids, such as blood, present a promising resource for biomarker discovery, in every sense. Microspot immunoassays allow the simultaneous quantification of multiple analytes from a minute amount of samples in a single measurement. The experimental design of microspot immunoassays is based on antibody pairs recognizing different epitopes of the analyte. The first antibody is used to capture the analyte from the complex sample, and the second antibody is used for detection. As with traditional enzyme-linked immunosorbent assays, highly reliable and reproducible results are obtained.

Protein microarrays are an ideal technology platform which allow for a robust and standardized profiling of the cellular proteome. Many cellular functions are not simply controlled by the presence of certain proteins, especially the propagation of external stimuli, which depend on transient post-translational modifications that determine whether a protein is in its active or inactive state. Thus, complex biological processes require the availability of a sound set of quantitative and time-resolved measurements to be understood. For this reason, new assay platforms which allow for the investigation of several proteins in parallel are necessary. The current best understood mode of cellular regulation occurs via phosphorylation and dephosphorylation processes, which are mediated via a large panel of kinases and phosphatases. The microspot immunoassay technique described here allows for an exact determination of several different phosphorylated proteins in parallel, as well as from small sample amounts, and is therefore an appropriate system to deepen the understanding of the complex regulatory networks implicated in health and disease.

Direct steam generation (DSG) is one alternative to the current oil-based parabolic trough solar thermal power plants. Within the German research project ITES, the dynamic behavior of a DSG collector field and the interactions with the conventional power block are assessed in detail. A transient solar field model developed by DLR is used to simulate the steam temperature behavior. Artificial irradiance disturbances as well as real irradiance data are used as input to the system. The resulting main steam temperature gradients are then analyzed by Siemens considering the standards for steam turbines. This paper presents the transient simulation results of the steam temperature as well as the corresponding results of the steam turbine analysis. It is found that the occurring temperature gradients are challenging for a safe turbine operation, if a conservative control system is used. Therefore, the use of an additional thermal inertia to stabilize the steam temperature is suggested. Its impact is also analyzed and discussed in this paper.

Steam turbines in solar thermal power plants experience a much greater number of starts than those operating in base-load plants. In order to preserve the lifetime of the turbine whilst still allowing fast starts, it is of great interest to find ways to maintain the turbine temperature during idle periods. A dynamic model of a solar steam turbine has been elaborated, simulating both the heat conduction within the body and the heat exchange with the gland steam, main steam and the environment, allowing prediction of the temperatures within the turbine during off-design operation and standby. The model has been validated against 96h of measured data from the Andasol 1 power plant, giving an average error of 1.2% for key temperature measurements. The validated model was then used to evaluate a number of modifications that can be made to maintain the turbine temperature during idle periods. Heat blankets were shown to be the most effective measure for keeping the turbine casing warm, whereas increasing the gland steam temperature was most effective in maintaining the temperature of the rotor. By applying a combination of these measures the dispatchability of the turbine can be improved significantly: electrical output can be increased by up to 9.5% after a long cool-down and up to 9.8% after a short cool-down.

Summary: Protein microarrays are well-established as sensitive tools for proteomics. Particularly, the microspot immunoassay (MIA) platform enables a quantitative analysis of (phospho-) proteins in complex solutions (e.g. cell lysates or blood plasma) and with low consumption of samples and reagents. Despite numerous biological and clinical applications of MIAs there is currently no user-friendly open source data analysis software available with versatile options for data analysis and data visualization. Here, we introduce the open source software QuantProReloaded that is specifically designed for the analysis of data from MIA experiments.
