Abstract

Abstract Adopting the Standard Halo Model (SHM) of an isotropic Maxwellian velocity distribution for dark matter (DM) particles in the Galaxy, the most stringent current constraints on their spin-dependent scattering cross-section with nucleons come from the IceCube neutrino observatory and the PICO-60 $$\hbox {C}_3\hbox {F}_8$$ C 3 F 8 superheated bubble chamber experiments. The former is sensitive to high energy neutrinos from the self-annihilation of DM particles captured in the Sun, while the latter looks for nuclear recoil events from DM scattering off nucleons. Although slower DM particles are more likely to be captured by the Sun, the faster ones are more likely to be detected by PICO. Recent N-body simulations suggest significant deviations from the SHM for the smooth halo component of the DM, while observations hint at a dominant fraction of the local DM being in substructures. We use the method of Ferrer et al. (JCAP 1509: 052, 2015) to exploit the complementarity between the two approaches and derive conservative constraints on DM-nucleon scattering. Our results constrain $$\sigma _{\mathrm{SD}} \lesssim 3 \times 10^{-39} \mathrm {cm}^2$$ σ SD ≲ 3 × 10 - 39 cm 2 ( $$6 \times 10^{-38} \mathrm {cm}^2$$ 6 × 10 - 38 cm 2 ) at $$\gtrsim 90\%$$ ≳ 90 % C.L. for a DM particle of mass 1 TeV annihilating into $$\tau ^+ \tau ^-$$ τ + τ - ( $$b\bar{b}$$ b b ¯ ) with a local density of $$\rho _{\mathrm{DM}} = 0.3~\mathrm {GeV/cm}^3$$ ρ DM = 0.3 GeV / cm 3 . The constraints scale inversely with $$\rho _{\mathrm{DM}}$$ ρ DM and are independent of the DM velocity distribution.

Abstract

Abstract Background Neuronal uptake and subsequent spread of proteopathic seeds, such as αS (alpha-synuclein), Tau, and TDP-43, contribute to neurodegeneration. The cellular machinery participating in this process is poorly understood. One proteinopathy called multisystem proteinopathy (MSP) is associated with dominant mutations in Valosin Containing Protein (VCP). MSP patients have muscle and neuronal degeneration characterized by aggregate pathology that can include αS, Tau and TDP-43. Methods We performed a fluorescent cell sorting based genome-wide CRISPR-Cas9 screen in αS biosensors. αS and TDP-43 seeding activity under varied conditions was assessed using FRET/Flow biosensor cells or immunofluorescence for phosphorylated αS or TDP-43 in primary cultured neurons. We analyzed in vivo seeding activity by immunostaining for phosphorylated αS following intrastriatal injection of αS seeds in control or VCP disease mutation carrying mice. Results One hundred fifty-four genes were identified as suppressors of αS seeding. One suppressor, VCP when chemically or genetically inhibited increased αS seeding in cells and neurons. This was not due to an increase in αS uptake or αS protein levels. MSP-VCP mutation expression increased αS seeding in cells and neurons. Intrastriatal injection of αS preformed fibrils (PFF) into VCP-MSP mutation carrying mice increased phospho αS expression as compared to control mice. Cells stably expressing fluorescently tagged TDP-43 C-terminal fragment FRET pairs (TDP-43 biosensors) generate FRET when seeded with TDP-43 PFF but not monomeric TDP-43. VCP inhibition or MSP-VCP mutant expression increases TDP-43 seeding in TDP-43 biosensors. Similarly, treatment of neurons with TDP-43 PFFs generates high molecular weight insoluble phosphorylated TDP-43 after 5 days. This TDP-43 seed dependent increase in phosphorlyated TDP-43 is further augmented in MSP-VCP mutant expressing neurons. Conclusion Using an unbiased screen, we identified the multifunctional AAA ATPase VCP as a suppressor of αS and TDP-43 aggregate seeding in cells and neurons. VCP facilitates the clearance of damaged lysosomes via lysophagy. We propose that VCP’s surveillance of permeabilized endosomes may protect against the proteopathic spread of pathogenic protein aggregates. The spread of distinct aggregate species may dictate the pleiotropic phenotypes and pathologies in VCP associated MSP.

3. Faculty Experiences as Members of Assessment Committees: A Case Study

Abstract

The use of student learning assessment is an important practice for institutions of higher education. External stakeholders expect colleges and universities to understand the extent to which students learn and use data to improve their learning. Student learning assessment includes identifying learning outcomes, developing assessment methods, collecting data, and using the data to improve teaching and learning. Faculty engagement is an essential aspect of having effective assessment practices. One way that faculty can engage with assessment and be involved in the leadership and decision-making process is by serving as members of assessment committees, a common practice of most institutions. This qualitative case study explored faculty experiences on assessment committees and the impact that those experiences can have on their perceptions of and engagement in assessment. Data analysis of interviews and committee documents from three Midwestern universities revealed many different practices among their assessment committees and identified some standard practices related to their committee purpose, gains of service on the committee, conditions of shared leadership, role as opinion leaders, and the similar views of assessment held by members. Additionally, one of the cases emerged as an exemplar model of an effective assessment committee, which resulted in recommendations for best practices. Abstract

Background: Neuronal uptake and subsequent spread of proteopathic seeds, such as αS (alpha-synuclein), tau, and TDP-43, contribute to neurodegeneration and disease progression. The cellular machinery necessary for this process is poorly understood. Methods: Cas9 expressing αS FRET biosensors were transduced with a whole-genome guide RNA (gRNA) library, seeded with αS fibrils, and flow-sorted. Candidate genes protective against αS seeding were identified following gRNA sequencing of FRET+ and FRET- cell populations. Secondary validation of the high probability candidate suppressor VCP, utilized VCP inhibitors or gene knockdown in αS biosensors and primary neurons. In vivo validation was performed in VCP disease mutation mice following intrastriatal injection of αS seeds. TDP-43 seeding was performed in primary neurons from control or VCP mutant mice.Results: We devised a genome-wide CRISPR-Cas9 screen to identify suppressors of αS seeding. This approach identified Valosin Containing Protein (VCP) as a suppressor of αS seeding. Dominant mutations in VCP cause multisystem proteinopathy (MSP) a phenotypically and pathologically variable neurodegeneraive disease characterized by myopathy, motor neuron disease and dementia with TDP-43, αS and tau inclusions. VCP inhibition or MSP disease mutations increased αS seeding in cells and primary cultured neurons. This was similar to treatment with the lysosomal damaging agent, LLoMe or knockdown of the endolysosomal damage response associated VCP cofactor, UBXD1. Intrastriatal injection of αS seeds into VCP disease mice demonstrated enhanced seeding efficiency as compared with controls. Finally, this phenomenon was not specific to αS since VCP disease mutant expression increased TDP-43 seeding in neurons.Conclusion: VCP surveillance of permeabilized late endosomes protects neurons against the proteopathic spread of pathogenic protein aggregates. The spread of distinct aggregate species may dictate the pleiotropic phenotypes and pathologies in VCP associated MSP.

Abstract

Physical review letters 125(14), 141801 (1-11) (2020). doi:10.1103/PhysRevLett.125.141801, The results of a 3+1 sterile neutrino search using eight years of data from the IceCube Neutrino Observatory are presented. A total of 305 735 muon neutrino events are analyzed in reconstructed energy-zenith space to test for signatures of a matter-enhanced oscillation that would occur given a sterile neutrino state with a mass-squared differences between 0.01 and 100 eV$^2$. The best-fit point is found to be at sin$^2$($2θ_{24}$)=0.10 and $Δm_{41}^2$=4.5 eV$^2$, which is consistent with the no sterile neutrino hypothesis with a $p$ value of 8.0%., Published by APS, College Park, Md.

Abstract

The observation of electromagnetic radiation from radio to $\gamma$-ray wavelengths has provided a wealth of information about the universe. However, at PeV (10$^{15}$ eV) energies and above, most of the universe is impenetrable to photons. New messengers, namely cosmic neutrinos, are needed to explore the most extreme environments of the universe where black holes, neutron stars, and stellar explosions transform gravitational energy into non-thermal cosmic rays. The discovery of cosmic neutrinos with IceCube has opened this new window on the universe. In this white paper, we present an overview of a next-generation instrument, IceCube-Gen2, which will sharpen our understanding of the processes and environments that govern the universe at the highest energies. IceCube-Gen2 is designed to: 1) Resolve the high-energy neutrino sky from TeV to EeV energies; 2) Investigate cosmic particle acceleration through multi-messenger observations; 3) Reveal the sources and propagation of the highest energy particles in the universe; 4) Probe fundamental physics with high-energy neutrinos. IceCube-Gen2 will increase the annual rate of observed cosmic neutrinos by a factor of ten compared to IceCube, and will be able to detect sources five times fainter than its predecessor. Furthermore, through the addition of a radio array, IceCube-Gen2 will extend the energy range by several orders of magnitude compared to IceCube. Construction will take 8 years and cost about \$350M. The goal is to have IceCube-Gen2 fully operational by 2033. IceCube-Gen2 will play an essential role in shaping the new era of multi-messenger astronomy, fundamentally advancing our knowledge of the high-energy universe. This challenging mission can be fully addressed only in concert with the new survey instruments across the electromagnetic spectrum and gravitational wave detectors which will be available in the coming years., Comment: 56 pages, 29 figures

Abstract

Pulsar wind nebulae (PWNe) are the main gamma-ray emitters in the Galactic plane. They are diffuse nebulae that emit nonthermal radiation. Pulsar winds, relativistic magnetized outflows from the central star, shocked in the ambient medium produce a multiwavelength emission from the radio through gamma-rays. Although the leptonic scenario is able to explain most PWNe emission, a hadronic contribution cannot be excluded. A possible hadronic contribution to the high-energy gamma-ray emission inevitably leads to the production of neutrinos. Using 9.5 yr of all-sky IceCube data, we report results from a stacking analysis to search for neutrino emission from 35 PWNe that are high-energy gamma-ray emitters. Abstract

Journal of Instrumentation 15(06), P06032 (2020). doi:10.1088/1748-0221/15/06/P06032, We describe an improved in-situ calibration of the single-photoelectron charge distributions for each of the in-ice Hamamatsu Photonics R7081-02[MOD] photomultiplier tubes in the IceCube Neutrino Observatory. The characterization of the individual PMT charge distributions is important for PMT calibration, data and Monte Carlo simulation agreement, and understanding the effect of hardware differences within the detector. We discuss the single photoelectron identification procedure and how we extract the single-photoelectron charge distribution using a deconvolution of the multiple-photoelectron charge distribution., Published by Inst. of Physics, London

Abstract

Physical review letters 124(5), 051103 (1-9) (2020). doi:10.1103/PhysRevLett.124.051103, This paper presents the results from point-like neutrino source searches using ten years of IceCube data collected between April 6, 2008 and July 10, 2018. We evaluate the significance of an astrophysical signal from a point-like source looking for an excess of clustered neutrino events with energies typically above $\sim1\,$TeV among the background of atmospheric muons and neutrinos. We perform a full-sky scan, a search within a selected source catalog, a catalog population study, and three stacked Galactic catalog searches. The most significant point in the Northern hemisphere from scanning the sky is coincident with the Seyfert II galaxy NGC 1068, which was included in the source catalog search. The excess at the coordinates of NGC 1068 is inconsistent with background expectations at the level of $2.9\,\sigma$ after accounting for statistical trials. The combination of this result along with excesses observed at the coordinates of three other sources, including TXS 0506+056, suggests that, collectively, correlations with sources in the Northern catalog are inconsistent with background at 3.3$\,\sigma$ significance. These results, all based on searches for a cumulative neutrino signal integrated over the ten years of available data, motivate further study of these and similar sources, including time-dependent analyses, multimessenger correlations, and the possibility of stronger evidence with coming upgrades to the detector., Published by APS, College Park, Md.

Abstract

The Command Support At-Sea Experiment (CS@SE) provides experimental advanced graphics display systems consisting of large screen color displays and operator console color displays in the combat information center (CIC) of an Aegis cruiser and in the tactical flag command center (TFCC) of an aircraft carrier. CS@SE systems are designed to prototype potential command support capabilities in an at-sea environment to validate and refine requirements for planned production system upgrades. These systems use sophisticated color graphics techniques to provide real-time tactical displays that improve the availability of information to an operator by reducing clutter through the use of color, area fill, transparen overlays and intensity coding of track symbols. Interfaces wen developed with the Aegis Display System (ADS), Shipboarc Gridlock System with Auto-correlation (SGS/AC), Flag Dats Display System (FDDS) and Tomahawk Engagement Planning and Exercise Evaluation System (TEPEE) that provided the data for the presentation of a tactical display. Display elements included both real-time and over-the-horizon (OTH) surface track data, velocity leaders, tags, uncertainty ellipses, and history trails. The display also included filled land masses, country boundaries, commercial airways, cities, graphics overlays (i.e., operational areas), weapon system missile performance contours, and engagement plans. This paper describes the experiment, its installation and integration into the shipboard environments of an Aegis cruiser (USS Leyte Gulf) and an aircraft carrier (USS America, its usage by the ships companies and embarked staffs, and the experiment result! and findings. Key conclusions of the experiment are: 1 Advanced graphics color displays can significantly enhance the ability of the warfighter to assimilate a complex tactical display. 2 Both ships reported a requirement for a correlated OTH and real-time track display with the ability to clearly differentiate the two types of tracks on the display. 3 On-line data bases such as detailed maps, airways, political boundaries, and cities provide important information that enhances the tactical track picture and the decision making process. 4 Classical rapid prototyping permits the validation of proposed command support concepts in an operational environment and permits some engineering evaluation of possible future capabilities.

Abstract

Shallow traps that affect exciton dynamics in undoped and Ce doped yttrium aluminum garnet (CeYAG) single crystals were studied by low temperature thermo-luminescence spectroscopy. The nature of traps was identified and their energy levels in the band gap were measured. Three types of traps were found: two very shallow traps with activation energy of less than 0.2 eV associated with electronic defects, one trap with activation energy of 0.5 eV associated with oxygen vacancies, and one relatively deeper trap with activation energy of 0.7 eV associated with hydrogen impurities. The association of oxygen vacancies with shallow traps as evident from this work and with deep traps as reported by Varney et al. [J. Appl. Phys. 111, 063505 (2012)] suggest their presence in the lattice in more than one charge state.

Es werden die ersten Ergebnisse der Anwendung der qualitativen und quantitativen pc-Analyse auf die Untersuchung von Alkydharzen beschrieben. Verschiedene Alkydharze industrieller Herkunft werden auf dem Papier verseift und die Fettsauren papierchromatographisch getrennt. Die quantitative Auswertung erfolgte photometrisch nach Anfarbung mit Kupferacetat und alkoholischer Rubeanwasserstoff-Losung. Die Hydrierdifferenz-Methode zur Erkennung kritischer Fettsaurepartner hatte bei Anwendung von kolloidalem Palladium keinen Erfolg. Zum Vergleich wurden die zur Herstellung der Alkydharze verwendeten Ole und die aus den Harzen praparativ isolierten Fettsauren ebenfalls chromatographisch analysiert. A Contribution to the Analysis of Alkyd-Resins The authors describe the first results of the applications of qualitative and quantitative pc-analysis for the investigation of alkyd-resins. Different alkyd-resins from the industry are saponified on the paper and the fatty acids are separated by paperchromatography. The quantitative estimations were done photometrically after coloring with copper acetat and alcoholic solution of rubeanic acid. The hydrogenation-difference-method for the identification of critical fatty acid partners using colloidal palladium is not successful. For comparison the oils used for the preparation of alkyd-resins and the isolated fatty acids prepared from the resins are likewise analysed by paperchromatography.

Abstract

Software techniques used in an automated real-time fuze testing system are discussed. Most of the techniques are independent of the electrical circuit being tested and the computer controlling the system. Although the software described was initially designed for testing the XM596 proximity fuze, only the actual fuze testing programs need be specifically designed for a given testing system. The programs comprising SOSOFT are functionally organized into eight major subsystems--real-time priority scheduling system, interrupt servicing system, input/output control system, executive utility system, basic testing system, data-display system, conversational control system, and reliability monitoring system.

Abstract

Gamow-Teller (GT) transitions starting from unstable pf-shell nuclei are of interest not only in nuclear physics, but also in astrophysics, e.g. in violent neutrino induced reactions at the core-collapse stage of type II supernovae. In the b-decay study of these pf-shell nuclei, half-lives can be measured rather accurately. On the other hand, in high-resolution (3He, t) charge-exchange reactions at 0°, individual GT transitions up to high excitations can be studied. Assuming the isospin symmetry for the strengths of Tz = ±1 - 0 analogous GT transitions, we present a unique 'merged analysis' for the determination of absolute B(GT) values. [ABSTRACT FROM AUTHOR]

Abstract

We report on the first measurement of the astrophysical neutrino flux using particle showers (cascades) in IceCube data from 2010-2015. Assuming standard oscillations, the astrophysical neutrinos in this dedicated cascade sample are dominated (∼90%) by electron and tau flavors. The flux, observed in the sensitive energy range from 16 TeV to 2.6 PeV, is consistent with a single power-law model as expected from Fermi-type acceleration of high energy particles at astrophysical sources. We find the flux spectral index to be γ=2.53±0.07 and a flux normalization for each neutrino flavor of ϕ_{astro}=1.66_{-0.27}^{+0.25} at E_{0}=100  TeV, in agreement with IceCube's complementary muon neutrino results and with all-neutrino flavor fit results. In the measured energy range we reject spectral indices γ≤2.28 at ≥3σ significance level. Because of high neutrino energy resolution and low atmospheric neutrino backgrounds, this analysis provides the most detailed characterization of the neutrino flux at energies below ∼100  TeV compared to previous IceCube results. Results from fits assuming more complex neutrino flux models suggest a flux softening at high energies and a flux hardening at low energies (p value ≥0.06). The sizable and smooth flux measured below ∼100  TeV remains a puzzle. In order to not violate the isotropic diffuse gamma-ray background as measured by the Fermi Large Area Telescope, it suggests the existence of astrophysical neutrino sources characterized by dense environments which are opaque to gamma rays.
