1. Ultrastructural changes in esophageal tissue undergoing stretch tests with possible impact on tissue engineering and long gap esophageal repairs performed under tension

Abstract

Abstract Esophageal biomechanical studies are being performed to understand structural changes resulting from stretches during repair of esophageal atresias as well as to obtain biomechanical values for tissue-engineered esophagus. The present study offers insights into ultrastructural changes after stretching of the ovine esophagus using uniaxial stretch tests. In vitro uniaxial stretching was performed on esophagi (n = 16) obtained from the abattoir within 4–6 h of 1-month-old lambs. Esophagi were divided into 4 groups (4 esophagi/group): control, Group1 (G1), Group2 (G2), Group3 (G3) stretched to 20%, 30% and 40% of their original length respectively. Force and lengthening were measured with 5 cycles performed on every specimen. Transmission electron microscopic (TEM) studies were performed on the 4 groups. During observational TEM study of the control group there were no significant differences in muscle cell structure or extracellular matrix. In all stretched groups varying degrees of alterations were identified. The degree of damage correlated linearly with the increasing level of stretch. Distance between the cells showed significant difference between the groups (control (μ = 0.41 μm, SD = 0.26), G1 (μ = 1.36 μm, SD = 1.21), G2 (μ = 2.8 μm, SD = 1.83), and G3 (μ = 3.01 μm, SD = 2.06). The diameter of the cells (control μ = 19.87 μm, SD = 3.81; G1 μ = 20.38 μm, SD = 4.45; G2 μ = 21.7 μm, SD = 6.58; G3 μ = 24.48 μm, SD = 6.69) and the distance between myofibrils (control μ = 0.23 μm, SD = 0.08; G1 μ = 0.27 μm, SD = 0.08; G2 μ = 0.4 μm, SD = 0.15; G3 μ = 0.61 μm, SD = 0.2) were significantly different as well ( p 30% alters the regular intracellular and extracellular structure of the esophageal muscle and leads to disruption of intra- and extracellular bonds. These findings could provide valuable insights into alterations in the microscopic structure of the esophagus in esophageal atresias repaired under tension as well as the basis for mechanical characterization for tissue engineering of the esophagus.

Abstract

Introduction: We present the first case of appendiceal intussusception associated with myeloid sarcoma in a young patient. Minimally invasive techniques used along the clinical course are highlighted. Case description: A 2.5-year-old boy was admitted after three weeks of COVID-19 infection with ongoing symptoms of MIS-C. Due to constipation, distended belly and vomiting, US was done which showed ileocolic intussusception. After unsuccessful hydrostatic reduction laparoscopic exploration was performed, where the vermiform appendix was found to be thickened and partially intussuscepted into the coecum. The ileocecal region was exteriorized transumbilically. After manual reduction of the intussusception, a long, thickened, fragile appendix was removed. Histopathology revealed myeloid sarcoma. Bone marrow investigation identified acute myeloid leukemia. During the oncological treatment, laparoscopic cholecystectomy was necessary due to cholecystitis and cholelithiasis. The child recovered uneventfully in terms of surgical complications, with good cosmetic result. Conclusion: No similar case in childhood was found in the English literature. Unusual symptoms and radiological findings of intussusception can conceal unexpected disorders. Minimally invasive technique offered advantages in the treatment of the presented patient and can be recommended to treat intussusception or cholelithiasis, if applicable, during an ongoing oncological treatment as well.

Abstract

Background and aims: One of the objectives of the ESC-EORP EUROASPIRE V survey is to determine how well European guidelines on the management of dyslipidaemias are implemented in coronary patients. Methods: Standardized methods were used by trained technicians to collect information on 7824 patients from 130 centers in 27 countries, from the medical records and at a visit at least 6 months after hospitalization for a coronary event. All lipid measurements were performed in one central laboratory. Patients were divided into three groups: on high-intensity LDL-C-lowering-drug therapy (LLT), on low or moderate-intensity LLT and on no LLT. Results: At the time of the visit, almost half of the patients were on a high-intensity LLT. Between hospital discharge and the visit, LLT had been reduced in intensity or interrupted in 20.8% of the patients and had been started or increased in intensity in 11.7%. In those who had interrupted LLT or had reduced the intensity, intolerance to LLT and the advice of their physician were reported as the reason why in 15.8 and 36.8% of the cases, respectively. LDL-C control was better in those on a high-intensity LLT compared to those on low or moderate intensity LLT. LDL-C control was better in men than women and in patients with self-reported diabetes. Conclusions: The results of the EUROASPIRE V survey show that most coronary patients have a less than optimal management of LDL-C. More professional strategies are needed, aiming at lifestyle changes and LLT adapted to the need of the individual patient.

6. Strength, durability, and environmental properties of concrete utilizing recycled tire particles for pavement applications

Abstract

The Rubber Manufacturers Association reported that approximately 230.7 million tires were disposed in the United States in 2011. Over 197.5 million of these disposed tires are recycled as tire derived fuel, lightweight fill, retaining walls, erosion protection as well as many other marketable uses. The primary objective of this study was to examine an alternative use for the 33.2 million remaining waste tires land disposed throughout the United States. This study used recycled tires in the form of crumb rubber as a fine aggregate replacement in concrete mixtures. Sustainable concrete mixtures for pavement applications that incorporated waste-stream materials such as fly ash, crumb rubber, and recycled concrete aggregate were evaluated for their performance. Fresh and hardened concrete properties were measured on mixtures containing 15% cement replacement with fly ash and sand volume replacements with crumb rubber of 10%, 20%, 30%, 40%, and 50%. The effects of the crumb rubber inclusion were determined by comparing mixtures containing the crumb rubber to a control mixture with only sand as the fine aggregate. Recycled concrete aggregate was included as a 50% coarse aggregate replacement by volume in two mixtures containing 20% and 30% crumb rubber content. The mixtures were tested for fresh concrete properties including slump, air content, unit weight and temperature, and hardened concrete properties including compressive strength, split-tensile strength, modulus of rupture, modulus of elasticity, permeability, and freeze–thaw durability. In addition, tests were conducted to determine whether any volatile organics leached from the crumb rubber concrete. As the crumb rubber content increased, the compressive strength, split-tensile, modulus of rupture, and the modulus of elasticity decreased. The crumb rubber concrete’s permeability increased within acceptable levels up to a 40% replacement of sand. The results of this study determined that a 30% replacement of sand with crumb rubber (approximately 5.5% of the total mixture volume) was optimum and produced the necessary fresh and hardened properties for concrete pavement.

Abstract

Electroporation, an attractive process for delivering DNA and other molecules into target cells in vivo and in vitro is limited by the necessity of electrodes that need to be in contact with the subject or object to be electroporated. We have used magnetic fields, which do not require material contact with the subject, to temporarily permeabilize cells in guinea pig skin in vivo to enhance uptake and expression of GFP plasmid DNA. The results show for the first time that magnetic fields can trigger a process likely similar to electroporation. In designing the magnetic pulses, our most important criterion was a high rate of change of the magnetic field, based on the principle described by Michael Faraday which is expressed by the formula: E = -dB/dt, (E, electric field, B, magnetic field, t, time). Magnetic fields were generated by a flat electromagnet in a hand-held applicator positioned above the target tissue. The magnetic pulses had a peak magnetic flux density of 4 tesla; 50 pulses were applied in 5 sec. Biphasic magnetic pulses were twice as effective as monophasic pulses and about equally effective as traditional electroporation pulses . Advantages of magnetopermeabilization over electoporation include: No contact between applicator and subject (“contact-less”); no need for invasive, disposable, sterile electrodes (“needle-less”); no pain from needles and reduced overall pain; no known side effects; easier and faster to administer than electroporation; less expensive due to absence of disposables; and, importantly, greater tissue penetration of the magnetic field allowing treatment of anatomical areas inaccessible by electroporation.

Abstract

Hintergrund Durch hohe Sicherheitsvorkehrungen bei Herstellung, Transport und Lagerung von Heizol kommt es kaum noch vor, dass Heizol in die Umwelt freigesetzt wird. Eine Ausnahme von dieser Erfahrung bildeten die mit dem „Jahrhunderthochwasser“ 2002 in Deutschland verbundenen Heizolaustritte aus Heizoltanks vor allem im privaten Bereich. In einer im Auftrag von IWO (Institut fur wirtschaftliche Oelheizung e. V.) erstellten Studie haben die Autoren das Umweltverhalten von Heizol nach Hochwasserereignissen untersucht. Ziel Da sich Heizol nach seiner Freisetzung rasch auf Wasseroberflachen ausbreitet und grose Flachen kontaminiert, wird ein groser Umweltschaden erwartet. Zur Bewertung muss das Verhalten von Heizol in Wasser und Boden ebenso wie die Wirkung von Heizol auf hohere und niedere Tiere und Pflanzen, auf Wasserorganismen sowie auf die Flora und Fauna des Bodens differenziert betrachtet werden. Aus der Bewertung der Umweltgefahren konnen erforderliche behordliche Masnahmen sowie Vorsorge und Sicherungsmasnahmen gegen die Gefahren durch Hersteller und Nutzer von privaten Heizolanlagen abgeleitet werden. Methodisches Vorgehen Zur Beantwortung der unterschiedlichen Fragestellungen stutzte sich das Autorenteam auf die umfangreichen Analysen des Sondermessprogramms Sachsen-Anhalt, auf Befragungen von Betroffenen (Bevolkerung sowie Behorden), auf Luftbildaufnahmen, auf umfangreiche Literaturbefunde einschlieslich okotoxikologischer Untersuchungen, auf Erfahrungen bei der uber 70-jahrigen Verwendung von Heizol im Pflanzenschutz und auf praktische Erfahrungen bei der grosraumigen Sanierung von Olschaden nach Havarien bzw. Unfallen. Die Autoren nahmen Bilanzierungen auf der Basis der vorhandenen Analysenergebnisse und eigener Berechnungen vor. Ergebnisse Die Freisetzung von Heizol in die Luft stellt kein besonderes Problem dar, da es zu ca. 40 % schnell verdunstet und vollstandig zu Kohlendioxid und Wasser abgebaut wird. Charakteristisch ist neben der Verdunstung die rasche Ausbreitung des Heizols auf der Wasseroberflache zu sehr dunnen Schichten. Bei den typischen, bunt schillernden Ollachen hat sich z. B. ein Kubikmeter Ol auf einer Wasserflache von ca. 3 km2 ausgebreitet, das entspricht einer Olmenge von 3 ml/m2 Flache, die nach dem Abtrocknen im Boden zu einer Olkonzentration von ca. 3 µg/kg Boden fuhrt und damit um Grosenordnungen unter dem naturlichen Gehalt an Kohlenwasserstoffen in Boden liegt. Aufgrund des Absorptionsvermogens der Boden und des mikrobiellen Abbaus im Boden durch ubiquitar verbreitete Kohlenwasserstoff abbauende Mikroorganismen dringt das Ol nur wenige Zentimeter in den Boden ein und wird in wenigen Monaten abgebaut, sodass Grundwasserschaden durch Heizolaustritte nach Hochwasserereignissen praktisch ausgeschlossen sind. Auch beim Hochwasser 2002 wurden bei Nachmessungen wenige Monate nach den Heizolaustritten keine Olkomponenten mehr im Boden nachgewiesen. Diskussion Akute Schaden durch Heizol an der Mikroflora und -fauna des Bodens sowie des Wassers konnen nicht von vornherein ausgeschlossen werden. So liegen die Schwellenwerte der Schadwirkung fur einige Wasserorganismen unterhalb von 1 mg/l. Jedoch wurden diese Werte, wie das Sondermessprogramm zum Hochwasser 2002 in Sachsen-Anhalt zeigte, meist weit unterschritten. Detailliert werden in der Studie (Deininger et al. 2006a) Ausbreitung und Verdunstung von Heizol auf Wasseroberflachen, die Verteilung, die Ausbreitung und der Abbau von Heizol im Boden beschrieben. Eine quantitative Auswertung des Heizolverbleibs erfolgt anhand eines konkreten Hochwasserereignisses. Schlussfolgerungen Besondere Masnahmen an Boden nach Heizolfreisetzungen bei Hochwasserereignissen sind nicht erforderlich. Selbst bei der Sanierung von Mineralolschaden nach Transportunfallen bzw. an Altstandorten mit wesentlich hoheren Konzentrationen an Mineralolkohlenwasserstoffen als bei Heizolfreisetzungen nach Hochwasserereignissen wurden mit bodenpflegerischen Masnahmen durch die naturlichen Mikroorganismen hohe Abbauleistungen erreicht und in wenigen Monaten die Konzentrationen an Kohlenwasserstoffen um 70–90 % reduziert. Ausblick Mit den vorliegenden Ergebnissen erfolgt eine realistische Bewertung der Umweltgefahren von Heizolaustritten nach Hochwasserereignissen. Daraus konnen adaquate Masnahmen fur behordliches Handeln und vorsorgende sowie sichernde Masnahmen fur Heizolanlagen in hochwassergefahrdeten Gebieten abgeleitet werden.

Abstract

Parasitemia levels of Calomys callosus inoculated with a high dose (HBT) of 4 × 10 3 Trypanosoma cruzi strain M226 bloodstream trypomastigotes (BT) exceeded those with the same inoculum of metacyclic trypomastigotes (MT) while a similar parasitemia was obtained with a low dose (LBT) of 5 × 10 2 of BT. Serum IFN-γ levels during the acute phase of infection were higher in the LBT inoculated group when compared with the group inoculated with HBT, while the IFN-γ levels in MT inoculated animals were close to uninfected controls. Spontaneous liberation of H 2 O 2 of peritoneal macrophages explanted from animals on days 21 and 28 after infection was comparable to that of controls for HBT and LBT groups while that of the MT inoculated group was significantly higher. Phorbol Myristate Acetate (PMA) stimulation resulted in high H 2 O 2 liberation specially in the infected groups. In vitro challenge with BT suppressed the small amount of spontaneous H 2 O 2 release, while MT challenge stimulated this release to a limited degree in infected groups. In this animal model, interacting with a parasite strain isolated from the same host, macrophage activation as measured by H 2 O 2 release was low, while the same strain had been previously observed to result in hyperactivation of mouse macrophages. We suggest that this distinctive behavior may be due to a host-parasite adaptation.

Abstract

Ambient level of gamma -aminobutyric acid (GABA), the major inhibitory neurotransmitter of the brain is mediated by neuronal and glial GABA transporters (GATs), members of the sodium and chloride ion-dependent solute carrier family. The neuronal GABA transporter subtype (GAT-1) has already been proven to be the target for the antiepileptic drug Tiagabine. However, druggability of glial GAT-2 and GAT-3 is yet to be established. Recent advances in structure elucidation of a bacterial orthologue leucine transporter in complex with different substrates substantiate homology modeling of human GATs (hGATs). These modeling studies can provide mechanistic clues for structure-based prediction of the potential of medicinal chemistry campaigns. A recently identified characteristic structural feature of the occluded conformation of hGATs is that similar extra- and intracellular gates are formed by middle-broken transmembrane helices TM1 and TM6. Binding crevice formed by unwound segments of broken helices facilitates symport of GABA with Na+ ion via fitting of GABA to TM1-bound Na+ closely inside. Favored accommodation of substrate inhibitors with high docking score predicts efficient inhibition of the neuronal hGAT-1 if the TM1-TM8 binding prerequisite for GABA was used. Docking, molecular dynamics and transport data indicate, that amino acids participating in substrate binding of the neuronal hGAT-1 and the glial hGAT-2 and hGAT-3 subtypes are different. By contrast, substrate binding crevices of hGAT-2 and hGAT-3 cannot be distinguished, avoiding sensible prediction of efficient selective substrate inhibitors. Glial subtypes might be specifically distinguished by interfering Zn2+ binding in the second extracellular loop of hGAT-3. Formation of the unique ring-like Na+-GABA complex in the occluded binding crevices anticipates family member symporters exploring chemiosmotic energy via reversible chemical coupling of Na+ ion.

Publisher Summary This chapter focuses on the principles that have applicability to a great variety of biological membranes. The study of the movement of solutes across biological membranes has progressed from the collection of phenomenological observations on cellular transport and electrical behavior to the identification, purification, characterization, and reconstitution of some of the integral membrane proteins responsible for the phenomena. To overcome the time-consuming separation of bound from free ligands, that cannot be automated, and that often can lead to increased non-specific binding, a scintillation proximity assay has been developed. The principle of this assay is that only radiolabeled ligands, which are bound to the receptor molecule generate a signal on the fluomicrosphere coated with receptors. The technique that is carried out in aqueous media (buffer) eliminates the use of liquid scintillants required for tritium-based assays. The scintillation proximity assay requires radiolabels that emit low-energy radiation, which is dissipated easily into an aqueous media.

Abstract

3-isopropylmalate dehydrogenase (IPMDH) from the psychrotrophic bacterium Vibrio sp. I5 has been expressed in Escherichia coli and purified. This cold-adapted enzyme is highly homologous with IPMDHs from other organisms, including mesophilic E. coli and thermophilic Thermus thermophilus bacteria. Its molecular properties are similar to these counterparts. Whereas the E. coli and T. thermophilus enzymes are hardly active at room temperature, the Vibrio IPMDH has reasonable activity below room temperature. The thermal stabilities, conformational flexibilities (hydrogen-deuterium exchange), and kinetic parameters of these enzymes were compared. The temperature dependence of the catalytic parameters of the three enzymes show similar but shifted profiles. The Vibrio IPMDH is a much better enzyme at 25 degrees C than its counterparts. With decreasing temperature i.e. with decreasing conformational flexibility, the specific activity reduces, as well; however, in the case of the Vibrio enzyme, the residual activity is still high enough for normal physiological operation of the organism. The cold-adaptation strategy in this case is achieved by creation of an extremely efficient enzyme, which has reduced but still sufficient activity at low temperature.

Abstract

Several lines of evidence indicate that augmented neuronal activity is associated with increased mitochondrial function, however, the mechanisms of coupling are still unclear. In this study we used a low extracellular Mg2+ concentration and short stimulus trains to evoke neuronal hyperactivity in the form of seizure-like events (SLE) in hippocampal slice cultures. Simultaneous microfluorimetric and electrophysiological techniques were applied to gain insight into changes of Ca2+ concentration in different compartments and into mitochondrial function. SLEs were associated with a large decrease of the extracellular Ca2+ concentration ([Ca2+]e), a spiking increase of the cytoplasmic and a smoothed elevation of the mitochondrial Ca2+ concentration (cytoplasmic concentration [Ca2+]i; intramitrochondrial concentration [Ca2+]m). Following an initial apparent decline in the mitochondrial membrane potential (DeltaPsi) and NAD(P)H autofluorescence, mitochondria depolarized and NADH production was augmented. Furthermore, SLEs were associated with increased oxidation of dihydroethidine (HEt). Our data suggest that intramitochondrial Ca2+ accumulation stimulates NADH production and production of radical oxygen species (ROS). Interestingly, mitochondrial depolarization followed [Ca2+]i and [Ca2+]m changes with a delay implying that electrogenic extrusion of Ca2+ from the mitochondrial matrix might be responsible for the depolarization of the mitochondrial membrane.

Abstract

We considered the evolution of Ca2+ oscillation dynamics in recurrent seizure-like events. Dynamic system behaviour was characterized in the state space reconstructed from intra- and extracellular [Ca2+] fluctuations simultaneously measured in cultured rat hippocampal slices under low-[Mg2+] conditions. When associated in the seizure-like event, these fluctuations occurred on a restricted set, the attractor, embedded in the full state space with less than five degrees of freedom. Instantaneous relative phase differences indicated field potential-driven phase jumps locked onto seizure-like events. To account for recurrent dynamics, calculations were performed on different extensions of a model for Ca2+ oscillation. These identified bidirectional, asymmetrical coupling of extracellular with intracellular (cytosolic, Ca2+ store, mitochondrial) Ca2+ dynamics as critical in its development.

This thesis examines the initial effort to formulate principles for information-based operations. Although it is impossible to explore each aspect of this transformation, it is worthwhile to examine current efforts by the US military to develop a doctrinal foundation for Information Operations (10). It explores the ongoing struggle to capture within the confines of Joint military doctrine those critical features of this "new age driven by information". The world community is increasingly dependent on reliable information traffic. Information has become a commodity and source of power unto itself. Alvin Toffler describes this period as the transformation of societies from second-wave (industrial/mechanical) to third-wave' (information-based) means. The growing dependence of the US military on these infrastructures reveals potentially vulnerable elements of the National Information Infrastructure (NII). This monograph examines the need for a comprehensive 10 doctrine. It yields a critical analysis of existing doctrine, illuminates several flaws within the current construct, and concludes with a suggested model for 10 development. Doctrinal models are developed for the Army, Air Force, and Navy respectively. These models explain those aspects which most essentially describe the doctrinal culture' of each service component. These factors include: service organization; employment of forces (both in peace and during crisis); and methods of control. In turn, each component model is compared to the revised 10 model.

Abstract

A spectroscopic method, using fluorescent Ca2+, K+ and Na+ ion indicators in combination with the use of fast-kinetic techniques on the time scale of 0.00004-10 s has been applied to study mechanisms of P2 pyrimidoceptor-mediated signal transduction in brain homogenates. Effects of the known P2 receptor ligands (ATP, alpha, beta-methylene-ATP, UTP, UDP and uridine) and the P1 receptor ligand, adenosine, were compared by measuring the rates of transmembrane Ca2+, K+ and Na+ ion fluxes in resealed plasmalemma fragments and nerve endings from the rat cerebral cortex. In homogenates containing resealed plasmalemma fragments, uridine (0.03-30 microM), but not adenosine, activated two phases of Ca2+ ion influx with onsets of a few ms and hundred ms in a concentration-dependent manner. Also, the activation of the fast-phase Ca2+ ion response by ATP, UDP and alpha, beta-methylene-ATP whereas that of the slow-phase by UTP and UDP were observed with 3 microM concentration of these P2 receptor ligands. In homogenates containing resealed nerve endings, the fast-phase Ca2+ ion response to uridine was absent. UTP, but not uridine and UDP (3 microM), activated a fast K+ ion influx with onset of1 ms. Adenosine (3 microM) evoked a slow Na+ ion influx with onset of0.1 s whereas the influx of Na+ ion to uridine was detectable below 0.01 s. Both nucleotides, ATP and UTP (3 microM), activated fluctuations of transmembrane Na+ ion influx and efflux. By contrast, UDP caused efflux of Na+ ion in the subsecond range of time. Collectively these results suggest that transmembrane cation fluxes mediated by kinetically distinguishable P2U pyrimidoceptor subtypes are different.

Abstract

The military's response to changes within the world political and technological environment has been termed a Revolution in Military Affairs (RMA). This monograph summarizes the effects that these changes will have on intelligence operations at the tactical level, or more specifically, the interaction between the tactical commander and the intelligence system which supports him. Though the definition of RMA varies from source to source, it can be distilled to the cumulative effects brought about by the progress of technology, doctrine, organization, and behavior. Recognition that the military is amidst the throes of a Revolution in Military Affairs is not sufficient to produce necessary and meaningful change. A strategy is needed in order to chart the Army's course for the near future and beyond. This concept is formulated in Army programs and has come to be known as "Force XXI". Concurrently, the Military Intelligence community is drafting its own, complementary program: "Intelligence XXI" - "INTEL XXI".

Abstract

Cerebellar granule cells in culture express receptors for GABA belonging to the GABA(A) and GABA(B) classes. In order to characterize the ability of the insecticide lindane to interact with these receptors cells were grown in either plain culture media or media containing 150 microM THIP as this is known to influence the properties of both GABA(A) and GABA(B) receptors. It was found that lindane regardless of the culture condition inhibited evoked (40 mM K+) release of neurotransmitter ([3H]D-aspartate as label for glutamate). In naive cells both GABA(A) and GABA(B) receptor active drugs prevented the inhibitory action of lindane but in THIP treated cultures none of the GABA(A) and GABA(B) receptor active drugs had any effect on the inhibitory action of lindane. This lack of effect was not due to inability of baclofen itself to inhibit transmitter release. It is concluded that lindane dependent on the state of the GABA(A) and GABA(B) receptors is able to indirectly interfere with both GABA(A) and GABA(B) receptors. In case of the latter receptors it was shown using [3H]baclofen to label the receptors that lindane could not displace the ligand confirming that lindane is likely to exert its action at a site different from the agonist binding site.

Abstract

The expression of GABAB receptors in cultured mouse cerebellar granule cells was investigated in binding experiments using [3H](S, R)-baclofen as well as in functional assessment of the ability of (R)-baclofen to interact with depolarization (15–40 mM KCI) coupled changes in intracellular Ca2+ homeostasis and neurotransmitter release. In the latter case a possible functional coupling between GABAA and GABAB receptors was investigated. The binding studies showed that the granule cells express specific binding sites for (R)-baclofen. The number of binding sites could be increased by exposure of the cells to the GABAA receptor agonist THIP (4,5,6,7-tetrahy-droisoxazolo[5,4-c]pyridin-3-ol) during the culture period. Pretreatment of the neurons with pertussis toxin showed that the GABAB receptors are coupled to G-proteins. This coupling was, however, less pronounced when the cells had been cultured in the presence of THIP. When 45Ca2+ uptake was measured or the intracellular Ca2+ concentration ([Ca2+]i) determined using the fluorescent Ca2+ chelator Fluo-3 it could be demonstrated that culturing the neurons in THIP influences intracellular Ca2+ homeostasis. Moreover, this homeostasis was found to be functionally coupled to the GABAB receptors as (R)-baclofen inhibited depolarization-induced increases in 45Ca2+ uptake and [Ca2+]i. (R)-Baclofen also inhibited K+-induced transmitter release from the neurons as monitored by the use of [3H]D-aspartate which labels the neurotransmitter pool of glutamate. Using the selective GABAA receptor agonist isoguvacine it could be demonstrated that the GABAB receptors are functionally coupled to GABAA receptors in the neurons leading to a disinhibitory action of GABAB receptor agonists. © 1994 Wiley-Liss, Inc.

Abstract

Excavations at the Celtic settlement of Sopron-Krautacker (W. Hungary) make it possible to form an overall view on the local practice of pottery manufacture. Study of the soil conditions has established that the source of raw material is the nearly calcium carbonate-free upper layer of the local soil resulting from a leaching process. By the various analytical methods used (XRD, XRF spectroscopy, and thermal analysis) composition of the pottery has been determined, and it has been shown that the vessels were fired at 600–700 °C.

Comparison of the kinetics of the inward Ca(2+) ion flux to (S)-alpha-Amino-3-hydroxy-5-methylisoxazole-4-propionic acid [(S)-AMPA] in cerebrocortical homogenates and that of the previously reported transmembrane Na(+) ion influx mediated by an AMPA receptor in hippocampal homogenates established that the agonist-induced opening of the AMPA receptor channels occurs in two kinetically distinguishable phases. Here we report that the 2-methyl-4-oxo-3H-quinazoline-3-acetic acid (Q1) inhibits the major slow-phase response specifically, whereas the acetyl piperidine derivative (Q5) is a more potent inhibitor of the fast-phase response. Both the quinazolone-3-propionic acid (Q2) and the quinazolone-3-acetic acid methyl ester (Q3) enhanced the slow-phase response to (S)-AMPA. The information provided by docking different Q1, Q2, and Q5 models at the ligand-binding core of iGluRs were used to define agonistic and antagonistic modes of interactions. Based on the effects of quinazolone-3-alkyl-carboxylic acid derivatives on specific [(3)H]Glu binding and kinetically distinguishable Ca(2+) ion permeability responses to (S)-AMPA and molecular modeling, the fast- and the slow-phase (S)-AMPA-elicited Ca(2+) ion fluxes were corresponded to different subunit compositions and degrees of S1S2 bridging interaction relative to substitution of kainate thereupon. Substitutions of agonists and antagonists into the iGluR2 S1S2 ligand binding core induced different modes of domain-domain bridging.

Particle track densities up to greater than 3 x 10(exp 9) per square centimeter have been measured in different samples. Rocks 17, 47, 57, and 58 have VH (Z greater than 22) galactic cosmic ray ages of 11, 14, 28, and 13 x 10(exp 6) years, respectively. Rock 57 has a calculated erosion rate of approximately less than 10(exp -7) centimeter per year. Near-surface track versus depth data in rock 17 can be fit with solar flare particles that have a differential energy spectrum alpha E(exp -s); lunar samples can be used to study the history of solar activity. The uranium in the crystalline rocks occurs principally in small regions less than 10 to approximately equal to 100 micrometers in size. The (low) thermoluminescence of the fines increases with depth in core 10004. With one possible exception, x-ray studies have not shown pronounced radiation damage effects. The total energy release upon heating is small up to 900 C and occurs in three broad regions.

Abstract

On the basis of 576 in vitro antibacterial positive results of 99 compounds containing gamma-pyridone-beta-carboxylic acid structure conclusions have been drawn on the mechanism of action of the derivatives by principal component analysis. It has been found that the compounds exert their effect essentially by influencing two types of physiological processes or series of processes. The more important of the two "mechanisms", responsible for 50-95% of the activity of the compounds, is common for the compounds investigated, and is associated with the gamma-pyridone-beta-carboxylic acid structure.

The enhancement of specific 3H-diazepam binding by increasing concentration of (+/-)-salutaridine follows a maximum curve indicating (+/-)-salutaridine to be a partial agonist at the GABA/benzodiazepine receptor complex.

Within the framework of a survey launched by the Istituto Superiore di Sanità to assess the average daily intake of Se with food in the Italian population, a preliminary study was carried out to develop an analytical procedure for the quantification of the element in food products. The method implies the use of a hydride generation system in combination with inductively-coupled plasma atomic emission spectrometry (ICP-AES) and exploits the inherent advantages of this last also for the analysis of non-metals. The overall approach was tested in some Hungarian and Italian foods, adequately lending itself to routine and reliable Se assay.

The technique of radiotracer 36Cl- influx in primary culture of rat cerebellar granule cells was applied to study the mechanism of inactivation of the GABAA receptor-activated chloride channel. During sustained application of GABA, muscimol and THIP the specific bicuculline-sensitive 36Cl- influx tends to decline with time. The sequence in decay half-time is GABA less than muscimol less than THIP. Diazepam accelerates the rate of decay of the peak response to GABA. (-)-Baclofen enhances the rate of decline of the response to muscimol in a dose-dependent manner. Treatment of the cells with pertussis toxin antagonized the effect of (-)-baclofen. It is concluded that rat neonatal cerebellar neurons maintained in tissue culture exhibit complex inactivation of the GABAA channel, indicating some interaction with the GABAB receptor system.

Six aliphatic esters of oxazepam were investigated for 3H-diazepam displacing activity and hydrolysis in rat in the brain P2-fraction treated with diisopropylfluorophosphate. The esters are prodrugs in relation to oxazepam. Increasing size of the 3-substituent and steric hindrance increased the value of IC50. Controversial reports on oxazepam hemisuccinate could be explained by its stereoselective hydrolysis in vivo.

Abstract

Sugammadex (SGM) is the first cyclodextrin (CD)-based selective relaxant binding agent. We investigated its ability to capture natural aminosteroid phytotoxins, and assessed its potential as an antidote for intoxication. Solasodine (SS), a toxic alkaloid from the Solanaceae family, was chosen as the model compound. Complexation was studied using nuclear magnetic resonance (NMR) spectroscopy, molecular modelling, and isothermal titration calorimetry (ITC). NMR in various D 2 O/DMSO‑d 6 media revealed a particularly stable inclusion-type complex, identifying a slow exchange process between the CD and the aminosteroid along with a less significant fast exchange between DMSO and SGM. Using various NMR techniques, the structure and kinetic/thermodynamic parameters of the inclusion complex were explored. Theoretical calculations showed the secondary amino head of SS near the carboxylate ends of the SGM sidechains, facilitating intermolecular ionic interactions. ITC experiments in an aqueous environment provided K a stability constants of 7.03 × 10 6  M -1 and 4.17 × 10 6  M -1 at 25 °C and 37 °C, respectively, similar to previously reported SGM complexes with aminosteroid neuromuscular blockers. Abstract

Background: The brain extracellular fluid (ECF), composed of secreted neurotransmitters, metabolites, peptides, and proteins, may reflect brain processes. Analysis of brain ECF may provide new potential markers for synaptic activity or brain damage and reveal additional information on pathological alterations. Epileptic seizure induction is an acute and harsh intervention in brain functions, and it can activate extra- and intracellular proteases, which implies an altered brain secretome. Thus, we applied a 4-aminopyridine (4-AP) epilepsy model to study the hippocampal ECF peptidome alterations upon treatment in rats., Methods: We performed in vivo microdialysis in the hippocampus for 3-3 h of control and 4-AP treatment phase in parallel with electrophysiology measurement. Then, we analyzed the microdialysate peptidome of control and treated samples from the same subject by liquid chromatography-coupled tandem mass spectrometry. We analyzed electrophysiological and peptidomic alterations upon epileptic seizure induction by two-tailed, paired t-test., Results: We detected 2540 peptides in microdialysate samples by mass spectrometry analysis; and 866 peptides-derived from 229 proteins-were found in more than half of the samples. In addition, the abundance of 322 peptides significantly altered upon epileptic seizure induction. Several proteins of significantly altered peptides are neuropeptides (Chgb) or have synapse- or brain-related functions such as the regulation of synaptic vesicle cycle (Atp6v1a, Napa), astrocyte morphology (Vim), and glutamate homeostasis (Slc3a2)., Conclusions: We have detected several consequences of epileptic seizures at the peptidomic level, as altered peptide abundances of proteins that regulate epilepsy-related cellular processes. Thus, our results indicate that analyzing brain ECF by in vivo microdialysis and omics techniques is useful for monitoring brain processes, and it can be an alternative method in the discovery and analysis of CNS disease markers besides peripheral fluid analysis., (© 2024. For several histone lysine methyltransferases (HKMTs), RNA binding has been already shown to be a functionally relevant feature, but detailed information on the RNA interactome of these proteins is not always known. Of the six human KMT2 proteins responsible for the methylation of the H3K4 residue, two-SETD1A and SETD1B-contain RNA recognition domains (RRMs). Here we investigated the RNA binding capacity of SETD1A and identified a broad range of interacting RNAs within HEK293T cells. Our analysis revealed that similar to yeast Set1, SETD1A is also capable of binding several coding and non-coding RNAs, including RNA species related to RNA processing. We also show direct RNA binding activity of the individual RRM domain in vitro, which is in contrast with the RRM domain found in yeast Set1. Structural modeling revealed important details on the possible RNA recognition mode of SETD1A and highlighted some fundamental differences between SETD1A and Set1, explaining the differences in the RNA binding capacity of their respective RRMs.

Abstract

The aggregation and amyloid formation of α-synuclein is associated with Parkinson's disease and other synucleinopathies. In its native, monomeric form α-synuclein is an intrinsically disordered protein represented by highly dynamic conformational ensembles. Inhibition of α-synuclein aggregation using small molecules, peptides, or proteins has been at the center of interest in recent years. Our aim was to explore the effects of cross-linking on the structure and aggregation/amyloid formation properties of α-synuclein. Comparative analysis of available high-resolution amyloid structures and representative structural models and MD trajectory of monomeric α-synuclein revealed that potential cross-links in the monomeric protein are mostly incompatible with the amyloid forms and thus might inhibit fibrillation. Monomeric α-synuclein has been intramolecularly chemically cross-linked under various conditions using different cross-linkers. We determined the location of cross-links and their frequency using mass spectrometry and found that most of them cannot be realized in the amyloid structures. The inhibitory potential of cross-linked proteins has been experimentally investigated using various methods, including thioflavin-T fluorescence and transmission electron microscopy. We found that conformational constraints applied by cross-linking fully blocked α-synuclein amyloid formation. Moreover, DTSSP-cross-linked molecules exhibited an inhibitory effect on the aggregation of unmodified α-synuclein as well.

50. Designing multi-target-directed flavonoids: a strategic approach to Alzheimer's disease.

Abstract

The underlying causes of Alzheimer's disease (AD) remain a mystery, with multiple pathological components, including oxidative stress, acetylcholinesterase, amyloid-β, and metal ions, all playing a role. Here we report a strategic approach to designing flavonoids that can effectively tackle multiple pathological elements involved in AD. Our systematic investigations revealed key structural features for flavonoids to simultaneously target and regulate pathogenic targets. Our findings led to the development of a highly promising flavonoid that exhibits a range of functions, based on a complete structure-activity relationship analysis. Furthermore, our mechanistic studies confirmed that this flavonoid's versatile reactivities are driven by its redox potential and direct interactions with pathogenic factors.
