1. Investigation of H-mode density limit in mixed protium–deuterium plasmas at JET with ITER-like wall

Abstract: Analysis of comparable discharges fuelled by either deuterium or protium reveals a clear relationship between the isotope mass and the H-mode density limit. Notably, the density limit is significantly lower in protium, showing a reduction of up to 35 % compared to identical deuterium plasma conditions. Within mixed H-mode density limit (HDL) plasmas, the maximum achievable density, or H-mode density limit, decreases with increasing protium concentration, denoted as cH. For instance, the highest corresponding maximum Greenwald fraction (fGW) of about 1.02 was observed in the pulse with the lowest cH value of 4.4 %. This fGW decreases to 0.96 at cH = 48 %. The average atomic mass, A¯, of the plasma species decreases in these pulses from the value of 1.96 (cH = 4.4 %) down to 1.52 (cH = 48 %). Interestingly, the maximum achievable density appears to be largely unaffected by the applied power value, regardless of whether deuterium or protium is used, as well as under mixed H/D fuelling conditions.Additionally, the measured Greenwald fractions are agreed with a heuristic model based on the SOL pressure threshold of an MHD instability, as proposed by Goldston. This comparison, especially concerning the model’s dependence on isotopic mass, shows full consistency between the measured and predicted Greenwald fractions.

2. A systematic investigation of radiation collapse for disruption avoidance and prevention on JET tokamak

Abstract: To produce fusion reactions efficiently, thermonuclear plasmas have to reach extremely high temperatures, which is incompatible with their coming into contact with material surfaces. Confinement of plasmas using magnetic fields has progressed significantly in the last years, particularly in the tokamak configuration. Unfortunately, all tokamak devices, and particularly metallic ones, are plagued by catastrophic events called disruptions. Many disruptions are preceded by anomalies in the radiation patterns, particularly in ITER-relevant scenarios. These specific forms of radiation emission either directly cause or reveal the approaching collapse of the configuration. Detecting the localization of these radiation anomalies in real time requires an innovative and specific elaboration of bolometric measurements, confirmed by visible cameras and the inversion of sophisticated tomographic algorithms. The information derived from these measurements can be interpreted in terms of local power balances, which suggest a new quantity, the radiated power divided by the plasma internal energy, to determine the criticality of the plasma state. Combined with robust indicators of the temperature profile shape, the identified anomalous radiation patterns allow determination of the sequence of macroscopic events leading to disruptions. A systematic analysis of JET campaigns at high power in deuterium, full tritium, and DT, for a total of almost 2000 discharges, proves the effectiveness of the approach. The warning times are such that, depending on the radiation anomaly and the available actuators, the control system of future devices is expected to provide enough notice to enable deployment of effective prevention and avoidance strategies.

Abstract: Experiments on ASDEX Upgrade (AUG) in 2021 and 2022 have addressed a number of critical issues for ITER and EU DEMO. A major objective of the AUG programme is to shed light on the underlying physics of confinement, stability, and plasma exhaust in order to allow reliable extrapolation of results obtained on present day machines to these reactor-grade devices. Concerning pedestal physics, the mitigation of edge localised modes (ELMs) using resonant magnetic perturbations (RMPs) was found to be consistent with a reduction of the linear peeling-ballooning stability threshold due to the helical deformation of the plasma. Conversely, ELM suppression by RMPs is ascribed to an increased pedestal transport that keeps the plasma away from this boundary. Candidates for this increased transport are locally enhanced turbulence and a locked magnetic island in the pedestal. The enhanced D-alpha (EDA) and quasi-continuous exhaust (QCE) regimes have been established as promising ELM-free scenarios. Here, the pressure gradient at the foot of the H-mode pedestal is reduced by a quasi-coherent mode, consistent with violation of the high-n ballooning mode stability limit there. This is suggestive that the EDA and QCE regimes have a common underlying physics origin. In the area of transport physics, full radius models for both L- and H-modes have been developed. These models predict energy confinement in AUG better than the commonly used global scaling laws, representing a large step towards the goal of predictive capability. A new momentum transport analysis framework has been developed that provides access to the intrinsic torque in the plasma core. In the field of exhaust, the X-Point Radiator (XPR), a cold and dense plasma region on closed flux surfaces close to the X-point, was described by an analytical model that provides an understanding of its formation as well as its stability, i.e., the conditions under which it transitions into a deleterious MARFE with the potential to result in a disruptive termination. With the XPR close to the divertor target, a new detached divertor concept, the compact radiative divertor, was developed. Here, the exhaust power is radiated before reaching the target, allowing close proximity of the X-point to the target. No limitations by the shallow field line angle due to the large flux expansion were observed, and sufficient compression of neutral density was demonstrated. With respect to the pumping of non-recycling impurities, the divertor enrichment was found to mainly depend on the ionisation energy of the impurity under consideration. In the area of MHD physics, analysis of the hot plasma core motion in sawtooth crashes showed good agreement with nonlinear 2-fluid simulations. This indicates that the fast reconnection observed in these events is adequately described including the pressure gradient and the electron inertia in the parallel Ohm’s law. Concerning disruption physics, a shattered pellet injection system was installed in collaboration with the ITER International Organisation. Thanks to the ability to vary the shard size distribution independently of the injection velocity, as well as its impurity admixture, it was possible to tailor the current quench rate, which is an important requirement for future large devices such as ITER. Progress was also made modelling the force reduction of VDEs induced by massive gas injection on AUG. The H-mode density limit was characterised in terms of safe operational space with a newly developed active feedback control method that allowed the stability boundary to be probed several times within a single discharge without inducing a disruptive termination. Regarding integrated operation scenarios, the role of density peaking in the confinement of the ITER baseline scenario (high plasma current) was clarified. The usual energy confinement scaling ITER98( p,y ) does not capture this effect, but the more recent H20 scaling does, highlighting again the importance of developing adequate physics based models. Advanced tokamak scenarios, aiming at large non-inductive current fraction due to non-standard profiles of the safety factor in combination with high normalised plasma pressure were studied with a focus on their access conditions. A method to guide the approach of the targeted safety factor profiles was developed, and the conditions for achieving good confinement were clarified. Based on this, two types of advanced scenarios (‘hybrid’ and ‘elevated’ q -profile) were established on AUG and characterised concerning their plasma performance.

Abstract: Within the 9th European Framework programme, since 2021 EUROfusion is operating five tokamaks under the auspices of a single Task Force called ‘Tokamak Exploitation’. The goal is to benefit from the complementary capabilities of each machine in a coordinated way and help in developing a scientific output scalable to future largre machines. The programme of this Task Force ensures that ASDEX Upgrade, MAST-U, TCV, WEST and JET (since 2022) work together to achieve the objectives of Missions 1 and 2 of the EUROfusion Roadmap: i) demonstrate plasma scenarios that increase the success margin of ITER and satisfy the requirements of DEMO and, ii) demonstrate an integrated approach that can handle the large power leaving ITER and DEMO plasmas. The Tokamak Exploitation task force has therefore organized experiments on these two missions with the goal to strengthen the physics and operational basis for the ITER baseline scenario and for exploiting the recent plasma exhaust enhancements in all four devices (PEX: Plasma EXhaust) for exploring the solution for handling heat and particle exhaust in ITER and develop the conceptual solutions for DEMO. The ITER Baseline scenario has been developed in a similar way in ASDEX Upgrade, TCV and JET. Key risks for ITER such as disruptions and run-aways have been also investigated in TCV, ASDEX Upgrade and JET. Experiments have explored successfully different divertor configurations (standard, super-X, snowflakes) in MAST-U and TCV and studied tungsten melting in WEST and ASDEX Upgrade. The input from the smaller devices to JET has also been proven successful to set-up novel control schemes on disruption avoidance and detachment.

5. COREDIV simulations of D and D–T high current–high power Baseline pulses in JET-ITER like wall

Abstract: The two best performing pulses of the so called ITER-Baseline scenario ( I _p = 3.5 MA and P _in ≈ 35 MW) of JET-ITER like wall, one in deuterium (D) the other in deuterium–tritium (D–T) plasma are examined and compared in this study. Generally, the D–T Baseline pulses exhibit an electron density level higher than the D pulses and the plasma energy is higher than in the comparable D pulses by up to 20%, reaching about 12 MJ in the pulse studied here. In contrast with the D pulses, the D–T pulses are often characterised by the increase in time of the radiated power in the mantle region (0.70 < ρ < 0.95), which may lead to the loss of the edge localised mode activity when the threshold H–L transition power is approached and to the subsequent plasma disruption due to excessive radiation. In this study we try to identify the physical mechanisms responsible for this behaviour using the available experimental data (principally the total radiated power from the bolometry) and the results of the fluid COREDIV model (1D in the core, 2D in the scrape-off-layer (SOL)), self-consistent with respect to core-SOL and also to main plasma-impurities. In fact, the loss of power caused by impurity radiation affects the temperature profile and finally the power to the divertor plate. The electron density and temperature profiles are numerically reconstructed as well as the radiated power density profiles, indicating no major difference in impurity transport in D and D–T. Indeed, the impurity transport coefficients used in COREDIV to match the experimental radiated power profiles are similar in the two pulses. The computed tungsten sources and densities are lower in the D–T pulse and the divertor impurity retention capability is a little better in the D–T pulse, indicatinga stronger collisional drag force in the SOL. The higher electron density and the broadening of its profile are the main cause of the observed increase of the radiated power in the D–T pulse.

Abstract: Superconducting (SC) tokamak JT-60SA plays an essential role in fusion research and development by supporting and complementing the ITER project, providing directions to the DEMO design activity and fostering next generation scientists and engineers. Since the short circuit incident at the terminal joints of equilibrium field coil #1 during the integrated commissioning (IC) in March 2021, both EU and JA implementing agencies (IAs) have examined how to ensure safe operation of JT-60SA by mitigating the risk of possible discharge occurrence inside the cryostat. Based on the experience of the global Paschen tests, the IAs have established a strategy of risk mitigation measures, which is a combination of (i) reinforcement of insulation, (ii) avoiding unnecessary voltage application to the coil systems and (iii) immediate de-energization of the coils when deteriorated vacuum conditions are detected. Thanks to the considerable efforts of the Integrated Project Team members, the IC restarted in May 2023. After confirmation of the SC state of the coil systems (TF, EF and CS), the coil energization test and the plasma operation phase 1 (OP-1) started. The first plasma was successfully achieved on 23 October 2023 with a limited value of voltage and current applied to the coils. The plasma configuration control was also confirmed with low plasma current and low auxiliary heating power conditions. Based on the IO–F4E–QST collaboration, activities of JT-60SA have been shared with the IO and provided an important lesson for ITER assembly and commissioning, and will provide an outstanding contribution to fusion research at large. After OP-1, maintenance & enhancement phase 1 (M/E-1) starts from January 2024, in which in-vessel components are installed, and heating and diagnostic systems are extensively upgraded to allow a high power heating experiment planned in OP-2. In order to make the best use of JT-60SA, a newly organized JT-60SA experiment team will refine the research plan for the future high heating power operation phase.

Abstract: In 2021 JET exploited its unique capabilities to operate with T and D–T fuel with an ITER-like Be/W wall (JET-ILW). This second major JET D–T campaign (DTE2), after DTE1 in 1997, represented the culmination of a series of JET enhancements—new fusion diagnostics, new T injection capabilities, refurbishment of the T plant, increased auxiliary heating, in-vessel calibration of 14 MeV neutron yield monitors—as well as significant advances in plasma theory and modelling in the fusion community. DTE2 was complemented by a sequence of isotope physics campaigns encompassing operation in pure tritium at high T-NBI power. Carefully conducted for safe operation with tritium, the new T and D–T experiments used 1 kg of T (vs 100 g in DTE1), yielding the most fusion reactor relevant D–T plasmas to date and expanding our understanding of isotopes and D–T mixture physics. Furthermore, since the JET T and DTE2 campaigns occurred almost 25 years after the last major D–T tokamak experiment, it was also a strategic goal of the European fusion programme to refresh operational experience of a nuclear tokamak to prepare staff for ITER operation. The key physics results of the JET T and DTE2 experiments, carried out within the EUROfusion JET1 work package, are reported in this paper. Progress in the technological exploitation of JET D–T operations, development and validation of nuclear codes, neutronic tools and techniques for ITER operations carried out by EUROfusion (started within the Horizon 2020 Framework Programme and continuing under the Horizon Europe FP) are reported in (Litaudon et al Nucl. Fusion accepted), while JET experience on T and D–T operations is presented in (King et al Nucl. Fusion submitted).

Abstract: This work studies the influence of radio frequency (RF) waves in the ion cyclotron resonance heating (ICRH) range of frequencies on fusion alphas during the recent JET D-T campaign. Fusion alphas from D-T reactions are created with energies of about 3.5 MeV and therefore have significant Doppler shifts enabling synergistic interactions between them and RF waves at a broad range of frequencies, including the ones foreseen for future fusion machines in ITER (Schneider et al 2021 Nucl. Fusion 61 126058) and SPARC (Creely et al 2020 J. Plasma Phys. 86 865860502). Resonant interactions between RF waves and alphas, also called synergistic effects, will modify the alpha distribution and ultimately will have an impact on alpha orbit losses and heating. Data from JET 3.43 T/2.3 MA pulses based on the hybrid scenario (Hobirk et al 2023 Nucl. Fusion ; Hobirk et al 29th IAEA FEC23 Conf. ( 16–21 October 2023 ); Challis et al 48th EPS Conf. on Plasma Physics ( 27 June–1 July 2022 ) during the DTE2 campaign (Maggi et al 2023 Nucl. Fusion )) were used for the analysis in this study. The impact of synergistic effects on alpha orbit losses and alpha heating are assessed. The conclusions are based on the analysis of experimental data for fast alpha losses, i.e. measurements from neutral particle analyser (NPA), fast ion losses scintillator detector, Faraday cups (FCs), and TRANSP (Hawryluk et al 1980 Physics of Plasmas Close to Thermonuclear Conditions vol 1 (CEC) pp 19–46) simulations. Experimental data and TRANSP analysis indicates that there are indeed changes in the alpha distribution function (DF) due to interaction with RF waves. Data from the NPA show increased ^4 He flux in the range from a few hundred keV up to 800 keV for pulses with RF power, while TRANSP clearly shows modifications in the fast alpha DF for these energies. Data from the scintillator detector and the FCs were compared for pulses with and without ICRH power and versus cases with enhanced alpha losses due to MHD activity. The trends from these diagnostics consistently show no additional alpha losses due to interaction with RF waves. TRANSP predictions for the impact of ynergistic effects on alpha heating show up to a 42% increase in alpha electron heating and up to a 25% increase in alpha ion heating. These effects, however, become negligibly small, less than 1%, when alpha heating is compared to the total auxiliary heating power in the investigated JET pulses.

Abstract: The experimental and theoretical analysis were focused on experiments conducted to assess the effect of plasma isotopes, protium (H), deuterium (D), and tritium (T) on ion cyclotron resonance heating (ICRH) related plasma wall interactions. Comparison of L-mode discharges with N = 1 ^3 He and N = 1 H minority ICRH heating scenarios were done for different isotopes. For the selected pulses, the behaviour of high-Z, mid-Z and low-Z intrinsic impurity and radiated power behaviour was investigated based on data from VUV, visible spectroscopy, and bolometry diagnostic at Joint European Torus. It was found that for N = 1 ^3 He scenario during radiofrequency antennas operation, core W, Ni content, Be source and the radiated power are higher for π /2 in comparison to dipole antenna phasing. Lowest core Ni, W content and radiated power is clearly observed for H plasmas in comparison to D and T, where for this ICRH scenario behaviour was similar. However, lower Be photon flux is observed for T in comparison to D plasmas. Be sputtering by He particles is responsible for such an effect. Additionally, several computer simulations were conducted using the COREDIV code. The difference in the electron temperature was due to the difference in the isotope masses. Increased temperature in the central plasma in the case of T plasmas leads to higher radiation in the central plasma in comparison to H plasmas. As a result, the power across separatrix is lower and the temperature on the divertor plate decreases with the increase of the isotope mass. At these temperatures on the divertor plate, W is not sputtered by the main plasma ions H, D and T and by He. For the N = 1 H ICRH scenario clear difference between D and T plasma was observed with higher metallic impurity content for T plasma in comparison to D. Impurity content in the plasmas is found to be sensitive to the power balance between the antenna straps. Its minimum is observed for the maximum of P _cen / P _tot .

Abstract: The fusion reaction between deuterium and tritium, D ( T,n ) ^4 He is the main source of energy in future thermonuclear reactors. Alpha-particles ( ^4 He -ions) born with an average energy of 3.5 MeV transferring energy to the thermal plasma during their slowing down, should provide the self-sustained D–T plasma burn. The adequate confinement of α -particles is essential to provide efficient heating of the bulk plasma and steady burning of a reactor plasma. That is why the fusion-born α -particle studies have been a priority task in the second D–T experiments (DTE2) on the Joint European Torus (JET) to understand the main mechanisms of their slowing down, redistribution and losses and to develop optimal plasma scenarios. JET with Be -wall and W -divertor, enhanced auxiliary heating systems and improved energetic-particle diagnostic capabilities, producing significant population of α -particles, provided the possibility for comprehensive studying of the α -particle behaviour. Selected results of the confined and lost α -particle measurements, evidence of α -particle self-heating and assessments of the fusion performance are presented in this paper giving an opportunity for further modelling and extrapolation to the International Thermonuclear Experimental Reactor and burning plasma reactors.

Abstract: JET, the world’s largest operating tokamak with unique Be/W wall and tritium handling capability, completed a Deuterium-Tritium (D-T) campaign in 2021 (Maggi et al 29th Fusion Energy Conf. ) following a decade of preparatory experiments, dedicated enhancements, technical rehearsals and training (Horton et al 2016 Fusion Eng. Des. 109–111 925). Operation with tritium raises significant technical, safety and scientific challenges not encountered in standard protium or deuterium operation. This contribution describes the tritium operational requirements, pulses and technical preparations, new operating procedures, lessons learned and details on the achieved operational availability and performance. The preparation and execution of the recent JET tritium experiments benefitted from the previous experience in 1991 (Preliminary Tritium Experiment), 1997 (DTE1 campaign) and 2003 (Trace Tritium Campaigns) and consisted of the following five phases: technical rehearsals and scenario preparation, tritium commissioning, 100% tritium campaign, D-T campaign (DTE2), tritium clean-up. Following the clean-up JET resumed normal operation and is currently undertaking a further D-T campaign (DTE3).

Abstract: This paper reports the first experiment carried out in deuterium–tritium addressing the integration of a radiative divertor for heat-load control with good confinement. Neon seeding was carried out for the first time in a D–T plasma as part of the second D–T campaign of JET with its Be/W wall environment. The technical difficulties linked to the re-ionisation heat load are reported in T and D–T. This paper compares the impact of neon seeding on D–T plasmas and their D counterpart on the divertor detachment, localisation of the radiation, scrape-off profiles, pedestal structure, edge localised modes and global confinement.

Abstract: The JET hybrid scenario has been developed from low plasma current carbon wall discharges to the record-breaking Deuterium-Tritium plasmas obtained in 2021 with the ITER-like Be/W wall. The development started in pure Deuterium with refinement of the plasma current, and toroidal magnetic field choices and succeeded in solving the heat load challenges arising from 37 MW of injected power in the ITER like wall environment, keeping the radiation in the edge and core controlled, avoiding MHD instabilities and reaching high neutron rates. The Deuterium hybrid plasmas have been re-run in Tritium and methods have been found to keep the radiation controlled but not at high fusion performance probably due to time constraints. For the first time this scenario has been run in Deuterium-Tritium (50:50). These plasmas were re-optimised to have a radiation-stable H-mode entry phase, good impurity control through edge T _i gradient screening and optimised performance with fusion power exceeding 10 MW for longer than three alpha particle slow down times, 8.3 MW averaged over 5 s and fusion energy of 45.8 MJ.

Abstract: In JET deuterium-tritium (D-T) plasmas, the fusion power is produced through thermonuclear reactions and reactions between thermal ions and fast particles generated by neutral beam injection (NBI) heating or accelerated by electromagnetic wave heating in the ion cyclotron range of frequencies (ICRFs). To complement the experiments with 50/50 D/T mixtures maximizing thermonuclear reactivity, a scenario with dominant non-thermal reactivity has been developed and successfully demonstrated during the second JET deuterium-tritium campaign DTE2, as it was predicted to generate the highest fusion power in JET with a Be/W wall. It was performed in a 15/85 D/T mixture with pure D-NBI heating combined with ICRF heating at the fundamental deuterium resonance. In steady plasma conditions, a record 59 MJ of fusion energy has been achieved in a single pulse, of which 50.5 MJ were produced in a 5 s time window ( P _fus = 10.1 MW) with average Q = 0.33, confirming predictive modelling in preparation of the experiment. The highest fusion power in these experiments, P _fus = 12.5 MW with average Q = 0.38, was achieved over a shorter 2 s time window, with the period of sustainment limited by high-Z impurity accumulation. This scenario provides unique data for the validation of physics-based models used to predict D-T fusion power.

Abstract: A toroidal Alfvén eigenmode (TAE) has been observed to be driven by alpha particles in a JET deuterium-tritium internal transport barrier plasma. The observation occurred 50 ms after the removal of neutral beam heating (NBI). The mode is observed on magnetics, soft-xray, interferometry and reflectometry measurements. We present detailed stability calculations using a similar tool set validated during deuterium only discharges. These calculations strongly support the conclusion that the observed mode is a TAE, and that this mode was destabilized by alpha particles. Non-ideal effects from the bulk plasma are interpreted as responsible for suppressing the majority of TAEs which were also driven by alpha particles, but the modes that match the observations are predicted to be particularly weak for these non-ideal effects. This mode located far from the core on the outboard midplane is found to be driven by both trapped and passing particles despite alpha particles originating in the core.

Abstract: The reference ion cyclotron resonance frequency (ICRF) heating schemes for ITER deuterium–tritium (D-T) plasmas at the full magnetic field of 5.3 T are second harmonic heating of T and ^3 He minority heating. Experiments have been carried out in the second major D-T campaign (DTE2) at JET, and in its prior D campaigns, to integrate these ICRF scenarios in JET high-performance plasmas and to compare their performance with the commonly used hydrogen (H) minority heating. In 50:50 D:T plasmas, up to 35% and 5% larger fusion power and diamagnetic energy content, respectively, were obtained with second harmonic heating of T as compared to H minority heating at comparable total input powers and gas injection rates. The core ion temperature was up to 30% and 20% higher with second harmonic T and ^3 He minority heating, respectively, with respect to H minority heating. These are favourable results for the use of these scenarios in ITER and future fusion reactors. According to modelling, adding ICRF heating to neutral beam injection using D and T beams resulted in a 10%–20% increase of on-axis bulk ion heating in the D-T plasmas due to its localisation in the plasma core. Central power deposition was confirmed with the break-in-slope and fast Fourier transform analysis of ion and electron temperature in response to ICRF modulation. The tail temperature of fast ICRF-accelerated tritons, their enhancement of the fusion yield and time behaviour as measured by an upgraded magnetic proton recoil spectrometer and neutral particle analyser were found in agreement with theoretical predictions. No losses of ICRF-accelerated ions were observed by fast ion detectors, which was as expected given the high plasma density of n _e ≈ 7–8 × 10 ^19 m ^−3 in the main heating phase that limited the formation of ICRF-accelerated fast ion tails. ^3 He was introduced in the machine by ^3 He gas injection, and the ^3 He concentration was measured by a high-resolution optical penning gauge in the sub-divertor region. The DTE2 experiments with ^3 He minority heating were carried with a low ^3 He concentration in the range of 2%–4% given the fact that the highest neutron rates with ^3 He minority heating in D plasmas were obtained at low ^3 He concentrations of ∼2%, which also coincided with the highest plasma diamagnetic energy content. In addition to ^3 He introduced by ^3 He gas injection, an intrinsic concentration of ^3 He of the order of 0.2%–0.4% was measured in D-T plasmas before ^3 He was introduced in the device, which is attributed to the radioactive decay of tritium to ^3 He. According to modelling, even such low intrinsic concentrations of ^3 He lead to significant changes in ICRF power partitioning during second harmonic heating of T due to absorption of up to 30% of the wave power by ^3 He.

Abstract: Neutral beam injection (NBI) is a flexible auxiliary heating method for tokamak plasmas, capable of being efficiently coupled to the various plasma configurations required in the Tritium and mixed deuterium-tritium experimental campaign on the Joint European Torus (JET) device. High NBI power was required for high fusion yield and alpha particle studies and to provide mixed deuterium-tritium (D-T) fuelling in the plasma core, it was necessary to operate the JET NBI systems in both deuterium and tritium. Further, the pure tritium experiments performed required T NBI for high isotopic purity and reduced 14 MeV neutron yield. Accurate power calibrations are also essential to machine safety. Previously on JET there have been a number of questions raised on the NBI power calibration, in particular following the Trace Tritium Experiments (TTEs). Operator activities on the tokamak NBI system, including calibrations, were performed in 2020. Following these activities, a series of plasma experiments were devised to further corroborate the T NBI power by comparing the plasma response to the D NBI power. A series of stationary, L-mode plasmas were performed on JET with different beam combinations used in different phases of the same pulse. By comparing the plasma response for D and T NBI it was possible to corroborate the T NBI power calibration using the D NBI power calibration. The stored energy as measured by magnetic diagnostics, corrected for fast particle stored energy, show that the uncertainty in NBI power calibration in T is comparable to that in D.

Abstract: The recent deuterium–tritium campaign in JET-ILW (DTE2) has provided a unique opportunity to study the isotope dependence of the L-H power threshold in an ITER-like wall environment (Be wall and W divertor). Here we present results from dedicated L-H transition experiments at JET-ILW, documenting the power threshold in tritium and deuterium–tritium plasmas, comparing them with the matching deuterium and hydrogen datasets. From earlier experiments in JET-ILW it is known that as plasma isotopic composition changes from deuterium, through varying deuterium/hydrogen concentrations, to pure hydrogen, the value of the line averaged density at which the threshold is minimum, ${\bar n_{{\text{e}},{\text{min}}}}$ , increases, leading us to expect that ${\bar n_{{\text{e}},{\text{min}}}}$ (T) < ${\bar n_{{\text{e}},{\text{min}}}}$ (DT) < ${\bar n_{{\text{e}},{\text{min}}}}$ (D) < ${\bar n_{{\text{e}},{\text{min}}}}$ (H). The new power threshold data confirms these expectations in most cases, with the corresponding ordering of the minimum power thresholds. We present a comparison of this data to power threshold scalings, used for extrapolation to future devices such as ITER and DEMO.

19. Impact of interaction between RF waves and fast NBI ions on the fusion performance in JET DTE2 campaign

Abstract: This work presents a study of the interaction between radio frequency (RF) waves used for ion cyclotron resonance heating and the fast deuterium (D) and tritium (T) neutral Beam injected (NBI) ions in DT plasma. The focus is on the effects of this interaction, also referred to as synergistic effects, on the fusion performance in the recent JET DTE2 campaign. Experimental data from dedicated pulses at 3.43 T/2.3 MA heated at (i) 51.4 MHz, giving the central minority H and n = 2 D, and at (ii) 32.2 MHz for the central minority ^3 He and n = 2 T. Resonances are analysed and conclusions are drawn and supported by modelling of the synergistic effects. Modelling with transport code TRANSP runs with and without the RF kick operator predict a moderate increase, of about 10%, in DT rates for the case of the RF wave—fast D NBI ion interactions at the n = 2 harmonic of ion cyclotron resonance, and a negligible impact due to synergistic interaction between fast T NBI ions and RF waves. JETTO modelling gives a 29% enhancement in fusion rates due to the interction between RF waves and fast D NBI ions, and an 18% enhancement in fast T NBI ions. Analysis of experimental neutron rates compared to TRANSP predictions without synergistic effects and magnetic proton recoil neutron spectrometer indicate an enhancement of approximately 25%–28% in fusion rates due to RF interaction with fast D ions, and an enhancement of approximately 5%–8% when RF waves and fast T NBI ions are interacting. The contributions of various heating and fast ion sources are assessed and discussed.

Abstract: This work applies the coupled JINTRAC and QuaLiKiz-neural-network (QLKNN) model on the ohmic current ramp-up phase of a JET D discharge. The chosen scenario exhibits a hollow $T_\mathrm{e}$ profile attributed to core impurity accumulation, which is observed to worsen with the increasing fuel ion mass from D to T. A dynamic D simulation was validated, evolving j , $n_\mathrm{e}$ , $T_\mathrm{e}$ , $T_\mathrm{i}$ , n _Be , n _Ni , and n _W for 7.25 s along with self-consistent equilibrium calculations, and was consequently extended to simulate a pure T plasma in a predict-first exercise. The light impurity (Be) accounted for Z _eff while the heavy impurities (Ni, W) accounted for P _rad . This study reveals the role of transport on the $T_\mathrm{e}$ hollowing, which originates from the isotope effect on the electron-ion energy exchange affecting $T_\mathrm{i}$ . This exercise successfully affirmed isotopic trends from previous H experiments and provided engineering targets used to recreate the D q -profile in T experiments, demonstrating the potential of neural network surrogates for fast routine analysis and discharge design. However, discrepancies were found between the impurity transport behaviour of QuaLiKiz and QLKNN, which lead to notable $T_\mathrm{e}$ hollowing differences. Further investigation into the turbulent component of heavy impurity transport is recommended.

Abstract: Intra-ELM tungsten sources, which dominate the total W source, are quantified in the inner and outer divertor of JET-ILW. The amount of the sputtered W atoms for individual ELMs demonstrates a clear dependence on the ELM frequency. It decreases when the pedestal temperature is lower and, correspondingly, the ELM frequency is higher. Nevertheless, the entire gross erosion W source (the number of eroded W atoms per second due to ELMs) increases initially with ELM frequency and reaches its maximum at fELM ≈ 50–55 Hz followed by its reduction in the high frequency range.The in/out asymmetry of the intra-ELM W sources during ELMs is a critical issue and is investigated in this contribution. At a lower ELM frequency of about 35 Hz the outer divertor gross W source is larger by a factor of 1.5. However the in/out asymmetry of the W erosion decreases strongly with ELM frequency demonstrating a nearly symmetric W source in both divertor legs at frequencies above 70 Hz. The screening of tungsten in the open magnetic configuration with strike points on tile 5 is more efficient by a factor of about 1.7 than in the corner configuration with the outer strike point at the pumping duct entrance.

Abstract: Unusually high power loads on the beryllium limiter caused by neutral beam re-ionisation, and much cooler divertor target surfaces were observed during the recent JET tokamak tritium campaign. As both phenomena are driven by scrape-off layer (SOL) physics, the SOL features of 72 tritium H-mode discharges and their deuterium references have been studied. The majority (70) of tritium H-mode discharges had exponentially decaying SOL profiles. The tritium plasmas are observed to have increased separatrix density and collisionality compared to their deuterium references. This is associated with $ \approx 2 - 3$ times broader SOL width for both density and temperature profiles. This is consistent with previous observations in highly collisional deuterium H-mode plasma on the ASDEX Upgrade tokamak (Sun et al 2015 Plasma Phys. Control. Fusion 57 125011) and interpreted as high collisionality enhancing cross-field transport across the separatrix and resulting in the broadening of near SOL above a critical value. The other two tritium H-mode discharges had near flat SOL density profiles, similar to the so-called ‘ density shoulder formation ’ observed in L-mode plasma. The SOL collisionality of these two pulses lies within the range of T pulses without density shoulder formation. This supports the conclusion of previous studies (Vianello et al 2017 Nucl. Fusion 57 116014; Wynn et al 2018 Nucl. Fusion 58 056001) that increased collisionality is not sufficient for the formation of a ‘ density shoulder ’ and additional factors, likely divertor condition or interaction with neutrals, are required. JET tritium plasma provides evidence of favourable and unfavourable effects of enhanced cross-field SOL transport on machine operation. The larger limiter power loads due to re-ionisation of neutral beam injection observed in the T pulses relative to their D references has been shown to be consistent with the combined effects of the broadening of the SOL profile and larger beam ion Larmor radius. The enhanced cross-field particle transport and the resulting broader SOL width provides more particles to ionize the fast Beam neutrals, causing the unfavourable power load issue on the beryllium limiter. The broader near SOL profiles of the T plasma spreads the heat load over a larger area and, together with the increased separatrix density, results in a favourably cooler divertor target surface.

23. Peripheral temperature gradient screening of high-Z impurities in optimised ‘hybrid’ scenario H-mode plasmas in JET-ILW

Abstract: Screening of high-Z (W) impurities from the confined plasma by the temperature gradient at the plasma periphery of fusion-grade H-mode plasmas has been demonstrated in the JET-ILW (ITER-like wall) tokamak. Through careful optimisation of the hybrid-scenario, deuterium plasmas with sufficient heating power ( $\gtrsim$ 32 MW), high enough ion temperature gradients at the H-mode pedestal top can be achieved for the collisional, neo-classical convection of the W impurities to be directed outwards, expelling them from the confined plasma. Measurements of the W impurity fluxes between and during edge-localised modes (ELMs) based on fast bolometry measurements show that in such plasmas there is a net efflux (loss) between ELMs but that ELMs often allow some W back into the confined plasma. Provided steady, high-power heating is maintained, this mechanism allows such plasmas to sustain high performance, with an average D–D neutron rate of ${\sim} 3.2 \times 10^{16}$ s ^−1 over a period of ∼3 s, after an initial overshoot (equivalent to a D–T fusion power of ∼9.4 MW), without an uncontrolled rise in W impurity radiation, giving added confidence that impurity screening by the pedestal may also occur in ITER, as has previously been predicted (Dux et al 2017 Nucl. Mater. Energy 12 28–35).

Abstract: Research over the past seventy years has established that mitochondrial-l-lactate dehydrogenase (m-L-LDH) is vital for mitochondrial bioenergetics. However, in recent report, Fulghum et al. concluded that lactate is a poor fuel for mitochondrial respiration [1]. In the present study, we have followed up on these findings and conducted an independent investigation to determine if lactate can support mitochondrial bioenergetics. We demonstrate herein that lactate can fuel the bioenergetics of heart, muscle, and liver mitochondria. Lactate was just as effective as pyruvate at stimulating mitochondrial coupling efficiency. Inclusion of LDH (sodium oxamate or GSK 2837808A) and pyruvate dehydrogenase (PDH; CPI-613) inhibitors abolished respiration in mitochondria energized with lactate. Lactate also fueled mitochondrial ROS generation and was just as effective as pyruvate at stimulating H2O2 production. Additionally, lactate-induced ROS production was inhibited by both LDH and PDH inhibitors. Enzyme activity measurements conducted on permeabilized mitochondria revealed that LDH is localized in mitochondria. In aggregate, we can conclude that mitochondrial LDH fuels bioenergetics in several tissues by oxidizing lactate.

28. Characterization of the impact of glutaredoxin-2 (GRX2) deficiency on superoxide/hydrogen peroxide release from cardiac and liver mitochondria

Abstract: Mitochondria are critical sources of hydrogen peroxide (H2O2), an important secondary messenger in mammalian cells. Recent work has shown that O2•-/H2O2 emission from individual sites of production in mitochondria is regulated by protein S-glutathionylation. Here, we conducted the first examination of O2•-/H2O2 release rates from cardiac and liver mitochondria isolated from mice deficient for glutaredoxin-2 (GRX2), a matrix-associated thiol oxidoreductase that facilitates the S-glutathionylation and deglutathionylation of proteins. Liver mitochondria isolated from mice heterozygous (GRX2+/-) and homozygous (GRX2-/-) for glutaredoxin-2 displayed a significant decrease in O2•-/H2O2 release when oxidizing pyruvate or 2-oxoglutarate. The genetic deletion of the Grx2 gene was associated with increased protein expression of pyruvate dehydrogenase (PDH) but not 2-oxoglutarate dehydrogenase (OGDH). By contrast, O2•-/H2O2 production was augmented in cardiac mitochondria from GRX2+/- and GRX2-/- mice metabolizing pyruvate or 2-oxoglutarate which was associated with decreased PDH and OGDH protein levels. ROS production was augmented in liver and cardiac mitochondria metabolizing succinate. Inhibitor studies revealed that OGDH and Complex III served as high capacity ROS release sites in liver mitochondria. By contrast, Complex I and Complex III were found to be the chief O2•-/H2O2 emitters in cardiac mitochondria. These findings identify an essential role for GRX2 in regulating O2•-/H2O2 release from mitochondria in liver and cardiac tissue. Our results demonstrate that the GRX2-mediated regulation of O2•-/H2O2 release through the S-glutathionylation of mitochondrial proteins may play an integral role in controlling cellular ROS signaling.

Abstract: The required heating power, P L H , to access the high confinement regime (H-mode) in tritium containing plasmas is investigated in JET with ITER-like wall at a toroidal magnetic field of B t = 1.8 T and a plasma current of I p = 1.7 MA. P L H , also referred to as the L-H power threshold, is determined in plasmas of pure tritium as well as mixtures of hydrogen with tritium (H-T) and mixtures of deuterium with tritium (D-T), and is compared to the L-H power threshold in plasmas of pure hydrogen and pure deuterium. It is found that, for otherwise constant parameters, P L H is not the same in plasmas with the same effective isotope mass, A e f f , when they differ in their isotope composition. Thus, A e f f is not sufficient to describe the isotope effect of P L H in a consistent manner for all considered isotopes and isotope mixtures. The electron temperature profiles measured at the L-H transition in the outer half of the radius are very similar for all isotopes and isotope mixtures, despite the fact that the L-H power threshold varies by a factor of about six. This finding, together with the observation of an offset linear relation between the L-H power threshold, P L H , and an effective heat diffusivity, χ e f f , indicates that the composition-dependent heat transport in the low confinement mode (L-mode) determines, how much power is needed to reach the necessary electron temperatures at the edge, and hence P L H .

Abstract: 2-Oxoglutarate dehydrogenase (Ogdh) is an important mitochondria redox sensor that can undergo S-glutathionylation following an increase in H2O2 levels. Although S-glutathionylation is required to protect Ogdh from irreversible oxidation while simultaneously modulating its activity it remains unknown if glutathione can also modulate reactive oxygen species (ROS) production by the complex. We report that reduced (GSH) and oxidized (GSSG) glutathione control O2∙-/H2O2 formation by Ogdh through protein S-glutathionylation reactions. GSSG (1 mM) induced a modest decrease in Ogdh activity which was associated with a significant decrease in O2∙-/H2O2 formation. GSH had the opposite effect, amplifying O2∙-/H2O2 formation by Ogdh. Incubation of purified Ogdh in 2.5 mM GSH led to significant increase in O2∙-/H2O2 formation which also lowered NADH production. Inclusion of enzymatically active glutaredoxin-2 (Grx2) in reaction mixtures reversed the GSH-mediated amplification of O2∙-/H2O2 formation. Similarly pre-incubation of permeabilized liver mitochondria from mouse depleted of GSH showed an approximately ~3.5-fold increase in Ogdh-mediated O2∙-/H2O2 production that was matched by a significant decrease in NADH formation which could be reversed by Grx2. Taken together, our results demonstrate GSH and GSSG modulate ROS production by Ogdh through S-glutathionylation of different subunits. This is also the first demonstration that GSH can work in the opposite direction in mitochondria-amplifying ROS formation instead of quenching it. We propose that this regulatory mechanism is required to modulate ROS emission from Ogdh in response to variations in glutathione redox buffering capacity.

Abstract: At its core mitochondrial function relies on redox reactions. Electrons stripped from nutrients are used to form NADH and NADPH, electron carriers that are similar in structure but support different functions. NADH supports ATP production but also generates reactive oxygen species (ROS), superoxide (O2·-) and hydrogen peroxide (H2O2). NADH-driven ROS production is counterbalanced by NADPH which maintains antioxidants in an active state. Mitochondria rely on a redox buffering network composed of reduced glutathione (GSH) and peroxiredoxins (Prx) to quench ROS generated by nutrient metabolism. As H2O2 is quenched, NADPH is expended to reactivate antioxidant networks and reset the redox environment. Thus, the mitochondrial redox environment is in a constant state of flux reflecting changes in nutrient and ROS metabolism. Changes in redox environment can modulate protein function through oxidation of protein cysteine thiols. Typically cysteine oxidation is considered to be mediated by H2O2 which oxidizes protein thiols (SH) forming sulfenic acid (SOH). However, problems begin to emerge when one critically evaluates the regulatory function of SOH. Indeed SOH formation is slow, non-specific, and once formed SOH reacts rapidly with a variety of molecules. By contrast, protein S-glutathionylation (PGlu) reactions involve the conjugation and removal of glutathione moieties from modifiable cysteine residues. PGlu reactions are driven by fluctuations in the availability of GSH and oxidized glutathione (GSSG) and thus should be exquisitely sensitive to changes ROS flux due to shifts in the glutathione pool in response to varying H2O2 availability. Here, we propose that energy metabolism-linked redox signals originating from mitochondria are mediated indirectly by H2O2 through the GSH redox buffering network in and outside mitochondria. This proposal is based on several observations that have shown that unlike other redox modifications PGlu reactions fulfill the requisite criteria to serve as an effective posttranslational modification that controls protein function.

Abstract: Mitochondria are quantifiably the most important sources of superoxide (O2●−) and hydrogen peroxide (H2O2) in mammalian cells. The overproduction of these molecules has been studied mostly in the contexts of the pathogenesis of human diseases and aging. However, controlled bursts in mitochondrial ROS production, most notably H2O2, also plays a vital role in the transmission of cellular information. Striking a balance between utilizing H2O2 in second messaging whilst avoiding its deleterious effects requires the use of sophisticated feedback control and H2O2 degrading mechanisms. Mitochondria are enriched with H2O2 degrading enzymes to desensitize redox signals. These organelles also use a series of negative feedback loops, such as proton leaks or protein S-glutathionylation, to inhibit H2O2 production. Understanding how mitochondria produce ROS is also important for comprehending how these organelles use H2O2 in eustress signaling. Indeed, twelve different enzymes associated with nutrient metabolism and oxidative phosphorylation (OXPHOS) can serve as important ROS sources. This includes several flavoproteins and respiratory complexes I-III. Progress in understanding how mitochondria generate H2O2 for signaling must also account for critical physiological factors that strongly influence ROS production, such as sex differences and genetic variances in genes encoding antioxidants and proteins involved in mitochondrial bioenergetics. In the present review, I provide an updated view on how mitochondria budget cellular H2O2 production. These discussions will focus on the potential addition of two acyl-CoA dehydrogenases to the list of ROS generators and the impact of important phenotypic and physiological factors such as tissue type, mouse strain, and sex on production by these individual sites.

Abstract: Protein S-glutathionylation is a reversible redox modification that regulates mitochondrial metabolism and reactive oxygen species (ROS) production in liver and cardiac tissue. However, whether or not it controls ROS release from skeletal muscle mitochondria has not been explored. In the present study, we examined if chemically-induced protein S-glutathionylation could alter superoxide (O2●-)/hydrogen peroxide (H2O2) release from isolated muscle mitochondria. Disulfiram, a powerful chemical S-glutathionylation catalyst, was used to S-glutathionylate mitochondrial proteins and ascertain if it can alter ROS production. It was found that O2●-/H2O2 release rates from permeabilized muscle mitochondria decreased with increasing doses of disulfiram (100-500 μM). This effect was highest in mitochondria oxidizing succinate or palmitoyl-carnitine, where a ~80-90% decrease in the rate of ROS release was observed. Similar effects were detected in intact mitochondria respiring under state 4 conditions. Incubation of disulfiram-treated mitochondria with DTT (2 mM) restored ROS release confirming that these effects were associated with protein S-glutathionylation. Disulfiram treatment also inhibited phosphorylating and proton leak-dependent respiration. Radiolabelled substrate uptake experiments demonstrated that disulfiram inhibited pyruvate import but had no effect on carnitine uptake. Immunoblot analysis of complex I revealed that it contained several protein S-glutathionylation targets including NDUSF1, a subunit required for NADH oxidation. Taken together, these results demonstrate that O2●-/H2O2 release from muscle mitochondria can be altered by protein S-glutathionylation. We attribute these changes to the protein S-glutathionylation complex I and inhibition of mitochondrial pyruvate carrier.

Abstract: Mitochondria fulfill a number of biological functions which inherently depend on ATP and O2−•/H2O2 production. Both ATP and O2−•/H2O2 are generated by electron transfer reactions. ATP is the product of oxidative phosphorylation whereas O2−• is generated by singlet electron reduction of di-oxygen (O2). O2−• is then rapidly dismutated by superoxide dismutase (SOD) producing H2O2. O2−•/H2O2 were once viewed as unfortunately by-products of aerobic respiration. This characterization is fitting considering over production of O2−•/H2O2 by mitochondria is associated with range of pathological conditions and aging. However, O2−•/H2O2 are only dangerous in large quantities. If produced in a controlled fashion and maintained at a low concentration, cells can benefit greatly from the redox properties of O2−•/H2O2. Indeed, low rates of O2−•/H2O2 production are required for intrinsic mitochondrial signaling (e.g. modulation of mitochondrial processes) and communication with the rest of the cell. O2−•/H2O2 levels are kept in check by anti-oxidant defense systems that sequester O2−•/H2O2 with extreme efficiency. Given the importance of O2−•/H2O2 in cellular function, it is imperative to consider how mitochondria produce O2−•/H2O2 and how O2−•/H2O2 genesis is regulated in conjunction with fluctuations in nutritional and redox states. Here, I discuss the fundamentals of electron transfer reactions in mitochondria and emerging knowledge on the 11 potential sources of mitochondrial O2−•/H2O2 in tandem with their significance in contributing to overall O2−•/H2O2 emission in health and disease. The potential for classifying these different sites in isopotential groups, which is essentially defined by the redox properties of electron donator involved in O2−•/H2O2 production, as originally suggested by Brand and colleagues is also surveyed in detail. In addition, redox signaling mechanisms that control O2−•/H2O2 genesis from these sites are discussed. Finally, the current methodologies utilized for measuring O2−•/H2O2 in isolated mitochondria, cell culture and in vivo are reviewed.

Abstract: Background: It has been four decades since protein S-glutathionylation was proposed to serve as a regulator of cell metabolism. Since then, this redox-sensitive covalent modification has been identified as a cell-wide signaling platform required for embryonic development and regulation of many physiological functions. Scope of the Review: Mitochondria use hydrogen peroxide (H2O2) as a second messenger, but its availability must be controlled to prevent oxidative distress and promote changes in cell behavior in response to stimuli. Experimental data favor the function of protein S-glutathionylation as a feedback loop for the inhibition of mitochondrial H2O2 production. Major conclusions: The glutathione pool redox state is linked to the availability of H2O2, making glutathionylation an ideal mechanism for preventing oxidative distress whilst playing a part in desensitizing mitochondrial redox signals. General Significance: The biological significance of glutathionylation is rooted in redox status communication. The present review critically evaluates the experimental evidence supporting its role in negating mitochondrial H2O2 production for cell signaling and prevention of electrophilic stress.

Abstract: There is a dearth of evidence-based reports linking the generation of free radicals and associated redox modifications with other major physiological changes of the sleep-wake cycle. To address this shortcoming, we examine and hypothesize that circadian/ultradian interaction of the redoxome, bioenergetics, and thermal signaling strongly regulate the differential activities of the sleep-wake cycle. Post-translational modifications of proteins by reversible cysteine oxoforms, S-glutathionylation and S-nitrosylation, are shown to play a major role regulating mitochondrial reactive oxygen species production, protein synthesis, respiration, and metabolomics. Protein synthesis and nuclear DNA repair are maximized during the wake state, whereas the redoxome is restored and mitochondrial protection is maximized during sleep. Hence, our analysis of redox/bioenergetics/temperature cycling indicates that the wake phase is more restorative and protective to the nucleus, whereas sleep is more restorative and protective to mitochondria. The redox/bioenergetics/temperature regulatory hypothesis adds to the understanding of mitochondrial respiratory uncoupling, substrate or futile cycling control, sudden infant death syndrome, torpor and hibernation and space radiation effects. Similarly, the hypothesis clarifies how the oscillatory redox/bioenergetics/temperature-regulated sleep-wake states, when perturbed by mitochondrial interactome disturbances, contribute to aging and the pathogenesis of diseases of the metabolism and cerebral nervous system.

Abstract: Mitochondrial production of reactive oxygen species (ROS) affects many processes in health and disease. SPG7 assembles with AFG3L2 into the mAAA protease at the inner membrane of mitochondria, degrades damaged proteins, and regulates the synthesis of mitochondrial ribosomes. SPG7 is cleaved and activated by AFG3L2 upon assembly. A variant in SPG7 that replaces arginine 688 with glutamine (Q688) is associated with several phenotypes, including toxicity of chemotherapeutic agents, type 2 diabetes mellitus, and (as reported here) coronary artery disease. We demonstrate that SPG7 processing is regulated by tyrosine phosphorylation of AFG3L2. Carriers of Q688 bypass this regulation and constitutively process and activate SPG7 mAAA protease. Cells expressing Q688 produce higher ATP levels and ROS, promoting cell proliferation. Our results thus reveal an unexpected link between the phosphorylation-dependent regulation of the mitochondria mAAA protease affecting ROS production and several clinical phenotypes.

Abstract: Mitochondria have a myriad of essential functions including metabolism and apoptosis. These chief functions are reliant on electron transfer reactions and the production of ATP and reactive oxygen species (ROS). The production of ATP and ROS are intimately linked to the electron transport chain (ETC). Electrons from nutrients are passed through the ETC via a series of acceptor and donor molecules to the terminal electron acceptor molecular oxygen (O2) which ultimately drives the synthesis of ATP. Electron transfer through the respiratory chain and nutrient oxidation also produces ROS. At high enough concentrations ROS can activate mitochondrial apoptotic machinery which ultimately leads to cell death. However, if maintained at low enough concentrations ROS can serve as important signaling molecules. Various regulatory mechanisms converge upon mitochondria to modulate ATP synthesis and ROS production. Given that mitochondrial function depends on redox reactions, it is important to consider how redox signals modulate mitochondrial processes. Here, we provide the first comprehensive review on how redox signals mediated through cysteine oxidation, namely S-oxidation (sulfenylation, sulfinylation), S-glutathionylation, and S-nitrosylation, regulate key mitochondrial functions including nutrient oxidation, oxidative phosphorylation, ROS production, mitochondrial permeability transition (MPT), apoptosis, and mitochondrial fission and fusion. We also consider the chemistry behind these reactions and how they are modulated in mitochondria. In addition, we also discuss emerging knowledge on disorders and disease states that are associated with deregulated redox signaling in mitochondria and how mitochondria-targeted medicines can be utilized to restore mitochondrial redox signaling.

46. Sex-dependent Differences in the Bioenergetics of Liver and Muscle Mitochondria from Mice Containing a Deletion for glutaredoxin-2

Abstract: Our group recently published a study demonstrating that deleting the gene encoding the matrix thiol oxidoreductase, glutaredoxin-2 (GRX2), alters the bioenergetics of mitochondria isolated from male C57BL/6N mice. Here, we conducted a similar study, examining H2O2 production and respiration in mitochondria isolated from female mice heterozygous (GRX2+/−) or homozygous (GRX2−/−) for glutaredoxin-2. First, we observed that deleting the Grx2 gene does not alter the rate of H2O2 production in liver and muscle mitochondria oxidizing pyruvate, α-ketoglutarate, or succinate. Examination of the rates of H2O2 release from liver mitochondria isolated from male and female mice revealed that (1) sex has an impact on the rate of ROS production by liver and muscle mitochondria and (2) loss of GRX2 only altered ROS release in mitochondria collected from male mice. Assessment of the bioenergetics of these mitochondria revealed that loss of GRX2 increased proton leak-dependent and phosphorylating respiration in liver mitochondria isolated from female mice but did not alter rates of respiration in liver mitochondria from male mice. Furthermore, we found that deleting the Grx2 gene did not alter rates of respiration in muscle mitochondria collected from female mice. This contrasts with male mice where loss of GRX2 substantially augmented proton leaks and ADP-stimulated respiration. Our findings indicate that some fundamental sexual dimorphisms exist between GRX2-deficient male and female rodents.

Publisher Copyright: © 2022 IAEA, Vienna. DIII-D physics research addresses critical challenges for the operation of ITER and the next generation of fusion energy devices. This is done through a focus on innovations to provide solutions for high performance long pulse operation, coupled with fundamental plasma physics understanding and model validation, to drive scenario development by integrating high performance core and boundary plasmas. Substantial increases in off-axis current drive efficiency from an innovative top launch system for EC power, and in pressure broadening for Alfven eigenmode control from a co-/counter-I p steerable off-axis neutral beam, all improve the prospects for optimization of future long pulse/steady state high performance tokamak operation. Fundamental studies into the modes that drive the evolution of the pedestal pressure profile and electron vs ion heat flux validate predictive models of pedestal recovery after ELMs. Understanding the physics mechanisms of ELM control and density pumpout by 3D magnetic perturbation fields leads to confident predictions for ITER and future devices. Validated modeling of high-Z shattered pellet injection for disruption mitigation, runaway electron dissipation, and techniques for disruption prediction and avoidance including machine learning, give confidence in handling disruptivity for future devices. For the non-nuclear phase of ITER, two actuators are identified to lower the L-H threshold power in hydrogen plasmas. With this physics understanding and suite of capabilities, a high poloidal beta optimized-core scenario with an internal transport barrier that projects nearly to Q = 10 in ITER at ∼8 MA was coupled to a detached divertor, and a near super H-mode optimized-pedestal scenario with co-I p beam injection was coupled to a radiative divertor. The hybrid core scenario was achieved directly, without the need for anomalous current diffusion, using off-axis current drive actuators. Also, a controller to assess proximity to stability limits and regulate β N in the ITER baseline scenario, based on plasma response to probing 3D fields, was demonstrated. Finally, innovative tokamak operation using a negative triangularity shape showed many attractive features for future pilot plant operation.

48. Thinking with Christian Existentialism: Freedom in Burke’s Dramatism and Berdyaev’s Dostoevsky

Abstract: Kenneth Burke’s logology is a way of thinking about how to understand the use of language—what he calls “symbolic action”—and how to use language to make sense of various human practices, including interpretive acts. This is a dialectic in thought between rhetoric as language-use and interpretation as making-sense. In The Rhetoric of Religion Burke’s theotropic logology uses theology to interpret symbolic action and symbolic action to interpret theology. Burke extends to other interpretive projects this same rhetorical-hermeneutic strategy of analogically translating words from one domain into another, from one meaning into another. This strategy is one way Burke thinks with other authors and their texts. The present essay uses some of Burke’s published and unpublished work to show how he thinks with the Christian Existentialism of Nicholas Berdyaev and Fyodor Dostoevsky, especially on the topic of freedom. In his thinking with Berdyaev, Burke agrees with the Russian theo-philosopher about the importance of freedom. Indeed, the act of freedom, dramatized in Dostoevsky and described by Berdyaev, forms the very center of Burke’s theory of symbolic action, his Dramatism and ultimately his Logology. Freedom is the condition of possibility for human action as opposed to mere motion, and free will is the necessary product of the cycle of terms implicit in the idea of hierarchical order presented in Burke’s The Rhetoric of Religion.

Abstract: Iron oxides control the mobility of a host of contaminants in aquifer systems, and the microbial reduction of iron oxides in the subsurface is linked to high levels of arsenic in groundwater that affects greater than 150 million people globally. Paired observations of groundwater and solid-phase aquifer composition are critical to understand spatial and temporal trends in contamination and effectively manage changing water resources, yet field-representative mineralogical data are sparse across redox gradients relevant to arsenic contamination. We characterize iron mineralogy using X-ray absorption spectroscopy across a natural gradient of groundwater arsenic contamination in Vietnam. Hierarchical cluster analysis classifies sediments into meaningful groups delineating weathering and redox changes, diagnostic of depositional history, in this first direct characterization of redox transformations in the field. Notably, these groupings reveal a signature of iron minerals undergoing active reduction before the onset of arsenic contamination in groundwater. Pleistocene sediments undergoing postdepositional reduction may be more extensive than previously recognized due to previous misclassification. By upscaling to similar environments in South and Southeast Asia via multinomial logistic regression modeling, we show that active iron reduction, and therefore susceptibility to future arsenic contamination, is more widely distributed in presumably pristine aquifers than anticipated.

Abstract: Geogenic arsenic in drinking water is a worldwide problem. For private well owners, testing (e.g., private or government laboratory) is the main method to determine arsenic concentration. However, the temporal variability of arsenic concentrations is not well characterized and it is not clear how often private wells should be tested. To answer this question, three datasets, two new and one publicly available, with temporal arsenic data were utilized: 6370 private wells from New Jersey tested at least twice since 2002, 2174 wells from the USGS NAWQA database, and 391 private wells sampled 14 years apart from Bangladesh. Two arsenic drinking water standards are used for the analysis: 10 µg/L, the WHO guideline and EPA standard or maximum contaminant level (MCL) and 5 µg/L, the New Jersey MCL. A rate of change was determined for each well and these rates were used to predict the temporal change in arsenic for a range of initial arsenic concentrations below an MCL. For each MCL and initial concentration, the probability of exceeding an MCL over time was predicted. Results show that to limit a person to below a 5% chance of drinking water above an MCL, wells that are ½ an MCL and above should be tested every year and wells below ½ an MCL should be tested every 5 years. These results indicate that one test result below an MCL is inadequate to ensure long-term compliance. Future recommendations should account for temporal variability when creating drinking water standards and guidance for private well owners.
