Pathogen interactions with cultured fish populations are well studied, but their effects on native fishes have not been characterized. In Chile, the disease caused by bacterial species Piscirickettsia salmonis represents one of the main issues and is considered to be one of the important pathogens in the field of aquaculture. They have been found to infect native fish. Therefore, it is necessary to understand the impact of P. salmonis on native species of local commercial value, as well as the potential impact associated with the emergence of antibiotic-resistant strains of P. salmonis. Due to this purpose, the native fish Eleginops maclovinus was used in our study. Fish were randomly distributed in tanks and intraperitoneally inoculated with two strains of P. salmonis. No mortality was recorded during the experiment. Cortisol, glucose and total α-amino acid levels increased in fish injected with AUSTRAL-005 strain compared to sham-injected and LF-89-inoculated fish. Moreover, results showed an increase in the activity of carbohydrates and lipids metabolism in liver; and an increase in the carbohydrates, lipids and total α-amino acid metabolism in muscle after injection with AUSTRAL-005. Our results suggest that P. salmonis modulates the physiology of E. maclovinus and the physiological impact increase in the presence of the antibiotic-resistant strain AUSTRAL-005.

Francisella noatunensis subsp. noatunensis, the etiological agent of Francisellosis, affects a large number of farmed species such as Salmo salar. This species coexists with several native species in the same ecosystem, including Eleginops maclovinus. Our objective was to evaluate the susceptibility, presence of clinical symptoms, and the ability of Eleginops maclovinus to respond to Francisella infection. For this, healthy individuals were inoculated with 1.5 × 101, 1.5 × 105, and 1.5 × 1010 bact/μL of Francisella by intraperitoneal injection, subsequently the fish were sampled on days 1, 3, 7, 14, 21, and 28 post injection (dpi). At the end of the experiment, no mortality, nor internal and external clinical signs were observed, although in the high dose anaemia was detected. Additionally, bacteria were detected in all three doses, however there was replication at day 28 only in the liver in the high dose. Analysis of gene expression by qPCR showed that the spleen generated an immune response against infection from day 1 dpi, however at day 7 dpi most of the genes suffered repressed expression; observing over expression of the genes C3, NLRC3, NLRC5, MHCI, IgM. In contrast, expression in the anterior kidney did not vary significantly during the challenge. IgM quantification showed the production of antibodies in the medium and high doses. This study provides new knowledge about Francisella infection and the long-lasting and specific immune response generated by Eleginops maclovinus. It also demonstrates its susceptibility to Francisellosis where there is a difference in the immune response according to the tissue.

The aquaculture industry has many problems associated with bacteria, viruses, and parasites, and the stress caused by these infections can modulate the physiological response in fish. In Chile, the sea lice Caligus rogercresseyi constitutes a major problem affecting the Chilean salmonid industry, having a strong negative effect on salmon production. For this reason, the aim of this study was to investigate the neuroendocrine and stress response of the most commonly cultured salmonid species in Chile, the Atlantic salmon (Salmo salar) and the Coho salmon (Oncorynchus kisutch), which were experimentally infested with the sea lice Caligus rogercresseyi. We analyzed the monoamine response in the telencephalon, optic tectum, and hypothalamus, as well as cortisol levels in plasma. In the liver and muscle we analyzed the cellular response by measuring heat shock proteins (60, 70, 90) and glucocorticoid mRNA transcription. After 14 days post infection C. rogercresseyi infestation modified the neuroendocrine and stress response in Atlantic and Coho salmon, with Coho salmon presenting a faster and higher response than the Atlantic salmon.

Variations in the mRNA expression of hepatic and muscle genes that are related to calcium signaling were analyzed by real-time qPCR in farmed Atlantic salmon (Salmo salar L. 1758) to determine changes in expression between parr and smolt stages. These organs were selected due to their close relationship with calcium signaling and metabolism (e.g., glycolysis, oxidative phosphorylation, muscle contraction). Differential expression between smolt and parr specimens and between organs was observed. Compared to parr specimens, smolts exhibited upregulated expression of the calcitonin receptor precursor, calcitonin receptor, calcitonin isoform, parathyroid hormone, and calmodulin in the liver. This pattern was inverse in muscle, with the exception of calmodulin, which was significantly upregulated in smolts compared to parr. Additionally, plasma calcium was decreased in the smolt condition. This study is the first to characterize the expression pattern of calcium signaling-related genes in the liver and muscle of parr and smolt S. salar. However, further functional studies are required to obtain a wider understanding about the physiological changes that accompany the productive conditions during smoltification.

Piscirickettsiosis caused by Piscirickettsia salmonis constitutes one of the main problems in farmed salmonid and marine fishes. The objective of this study was to evaluate the modulation of genes involved in the oxidative stress in the liver and muscle of Salmo salar challenge with low dosage of P. salmonis. The treatment (in duplicate) were as follows: Control injection (culture medium) and P. salmonis injection (1 × 102 PFU/mL) with sampling (liver and muscle) at several time-points during the 42-days experimental period (dpi). In liver, the gene expression of superoxide dismutase (SOD) and acetylcholinesterase (AChE) had differences with the control group only at 7 dpi, compared with glutathione-S-transferase (GST) and heat shock protein 70 (HSP70) that presented increases at 7 and 21 dpi. The glutathione peroxidase (GPx) and catalase (CAT) mRNAs were elevated at 13 and 21 dpi, respectively. While glutathione reductase (GR) and cytochrome P450 (P450) did not show variations in their expression during the experimental course. In muscle, the expression of CAT and AChE was higher than in the control condition at 2 and 42 dpi, respectively. While the number of transcripts SOD, GPx, GR, GST, P450 and HSP70 showed increases at 7- and 42-days post injection. The results suggest a transcriptional activation of genes involved in oxidative stress in both liver and muscle, with expression profiles that were tissue-specific and dependent on the time. This is the first study that reveals the transcriptional participation of all these genes associated with oxidative stress in response to the injection of P. salmonis.

Eleginops maclovinus is a eurythermic fish that under natural conditions lives in environments with temperatures ranging from 4 to 18 °C and can be usually captured near salmon farming areas. The aim of this study was to evaluate the effect of temperature over the innate and adaptive immune response of E. maclovinus challenged with Piscirickettsia salmonis following different treatments: C (control injection with culture medium at 12 °C), C+ (bacterial injection at 12 °C), 18 °C c/A + B (injection with culture medium in acclimation at 18 °C), 18 °C c/A + B (bacterial injection in acclimation at 18 °C), 18 °C s/A + M (injection with culture medium without acclimation at 18 °C) and 18 °C s/A + B (bacterial injection without acclimation at 18 °C). Each injection had 100 μL of culture medium or with 100 μL at a concentration 1 × 108 of live bacteria, sampling six fish per group at 4, 8, 12, 16 and 20 days post-injection (dpi). Expression of the mRNA related with the innate immune response gene (TLR1, TLR5, TLR8, NLRC3, NLRC5, MyD88 and IL-1β) as well as the adaptive immune response gene (MHCI, MHCII, IgMs and IgD) were measured in spleen and head kidney. Gene expression profiles were treatment-type and time dependent. Levels of Immunoglobulin M (IgM) increased in challenged groups with P. salmonis from day 8–20 post challenge, which suggest activation of B cells IgM + through P. salmonis epitope detection. Additionally, a rise in temperature from 12 °C (C+) to 18 °C (with/without acclimation) also resulted in antibody increment detected in serum with significant differences between “18 °C c/A + B” and “18 °C s/A + B” groups. This is the first study that evaluates the effect of temperature changes and mRNA expression related with immune system gene over time on E. maclovinus, a native wild life fish that cohabits in the salmon farming environment.

Smoltification under intensive aquaculture conditions involves important physiological changes to intestinal structures that facilitate acclimation to hyperosmotic environments. This study evaluated if genic expression of primary contributors during the smoltification process in the intestine are in synchrony with gill Na+/K+-ATPase activity. Discordance was found between gill Na+/K+-ATPase activity levels and intestinal genic expressions. In particular, the intestine showed increased mRNA expression for prolactin, a hormone that participates in hypoosmotic environments, and in mRNA expression of the growth hormone, which mediates the hydromineral balance in hyperosmotic environments. These findings indicate a lack of sufficient intestinal maturation in the fish at the moment when they were transferred to seawater environments in the normal aquaculture practices. This intestinal immaturity is a possible explanation for the high rates of desadapted individuals in the seawater phase in Chilean salmon aquaculture.

Although Caligus rogercresseyi negatively impacts Chilean salmon farming, the metabolic effects of infection by this sea louse have never been completely characterized. Therefore, this study analyzed lactate responses in the plasma, as well as the liver/muscle lactate dehydrogenase (LDH) activity and gene expression, in Salmo salar and Oncorhynchus kisutch infested by C. rogercresseyi. The lactate responses of Atlantic and Coho salmon were modified by the ectoparasite. Both salmon species showed increasing in plasma levels, whereas enzymatic activity increased in the muscle but decreased in the liver. Gene expression was overexpressed in both Coho salmon tissues but only in the liver for Atlantic salmon. These results suggest that salmonids need more energy to adapt to infection, resulting in increased gene expression, plasma levels, and enzyme activity in the muscles. The responses differed between both salmon species and over the course of infection, suggesting potential species-specific responses to sea-lice infection.

Piscirickettsia salmonis is the etiological agent of piscirickettsiosis, which, as the main systemic disease in the Chilean salmon industry, causes significant economic losses. This bacterium can produce biofilm as a persistence and survival strategy in adverse conditions. In other bacteria, cheA is a key gene for modulating the onset of bacterial chemotaxis, as well as having a secondary role in biofilm production. Notwithstanding this association, the potential relationships between biofilm formation and genes involved in P. salmonis chemotaxis are poorly understood. This study aimed to determine P. salmonis cheA gene expression when grown in different culture media known to induce biofilm production. Piscirickettsia salmonis AUSTRAL-005 produced moderate/high biofilm levels after 144 h of incubation in the AUSTRAL-SRS and marine broths. In contrast, LF-89 biofilm production was weak/nonexistent in the aforementioned broths. Both assessed P. salmonis strains contained the cheYZA operon. Additionally, AUSTRAL-005 cheA transcripts increased in both culture media. In conclusion, these results suggest potential relationships between biofilm formation and genes related to chemotaxis in the fish pathogen P. salmonis.

To clarify the role of growth hormone (GH) in the immune system of fish, we examine the comparative effect of GH and Growth Hormone Release Factor (GRF) on leukocytes culture of the head kidney of Atlantic salmon and the SHK-1 cell line. There are studies that associate the growth hormone (GH) / insulin-like growth factor (IGF) axis with the immune regulation of fish. However, there is no evidence that GH and GRF stimulate Atlantic salmon leukocyte cell lines, where there arent reports on expression changes in different immune response markers. Thus, we investigated the effect of GH and GRF in Atlantic salmon leukocytes extracted from head kidney and the SHK-1 cell line on the different immune response markers such as: NLRC5, NLRC3, IL-1β, TNF-α, and IL-8 through qPCR. Our data suggest that GH increases the expression of NLRC5, NLRC3, and IL-1β mainly at 16 h post-stimulation in Atlantic salmon leukocytes. This indicates differential regulation between the two models used, helping us to better understand the independent action of GH on the immune system and the GH / IGF axis for future research.

Fluctuations in ambient temperature along with the presence of pathogenic microorganisms can induce important cellular changes that alter the homeostasis of ectothermic fish. The aim of this study was to evaluate how sudden or gradual changes in environmental temperature together with the administration of Piscirickettsia salmonis modulate the transcription of genes involved in cellular stress response in the liver of Eleginops maclovinus. Fish were subjected to the following experimental conditions in duplicate: C− 12 °C: Injection only with culture medium, C+ 12 °C: Injection with P. salmonis, AM 18 °C: Injection only with culture medium under acclimation at 18 °C, AB 18 °C: Injection with P. salmonis under acclimation at 18 °C, SM 18 °C: Injection only with culture medium and thermal shock at 18 °C and SB 18 °C: Injection with P. salmonis and thermal shock at 18 °C and sampling at 4-, 8-, 12-, 16- and 20-day post injection (dpi). The genes implied in the heat shock response (HSP70, HSC70, HSP90, and GRP78), apoptosis pathway (BAX and SMAC/Diablo), ubiquitination (E2, E3, ubiquitin, and CHIP), and 26 proteasome complex (PSMB7, PSMC1, and PSMA2) showed expression profiles dependent on time and type of injection applied. All the genes greatly increased their expression levels at day 16 and showed moderate increases at day 20, except for PSMA2 which showed a higher increase between 4- and 12-day post challenges. Our results suggest that the changes observed at the final days of the experiment are due to temperature more than P. salmonis.

The nucleotide binding oligomerization domain like receptors, or NOD like receptors (NLRs), are intracellular receptors responsible for recognizing pathogens in vertebrates. Several NLR mammalian models have been characterized and analyzed but few studies have been performed with teleost species. In this study, we analyzed the nucleotide sequence of six mRNA variants of NLRC3 in Atlantic salmon (SsNLRC3), and we deduced the amino acid sequence coding for two different isoforms with a total length of 1135 amino acids and 1093 amino acids. We analyzed the phylogeny of all variants, including a Piscirickettsia salmonis infection in Atlantic salmon. All variants and their expression pattern during infection were analyzed using real-time qPCR. One of the analyzed variants was over-expressed during the early stages of Piscirickettsia salmonis infection, and we were able to identify two different SsNLRC3 isoforms. Lastly, we observed that an alteration in the amino acid sequence of one of the isoforms can directly affect the pathogen recognition function.

Eleginops maclovinus is a subantartic notothenic with eurythermal characteristics and potential for Chilean aquaculture. However, the information regarding their physiological response to long-term temperatures and optimum temperature range for physiological process is unknown. In the present study, we evaluated under experimental conditions, the long-term effects (60 days) of temperatures of 15 °C and high temperatures of 20 and 25 °C, (10 °C as control) on the physiology of E. maclovinus juveniles. At the end of the experimental period, survival was 36% at 25 °C and mortality started after 30 days, while at the other temperatures the survival was 100%. The growth and food intake did not present significant differences at 10, 15 and 20 °C, while at 25 °C growth decreased at day 15, to subsequently present a compensatory growth at day 30 and finally a decrease in growth and food intake at day 60. In general, the energy substrates in liver, gills, serum, and spleen showed a significant decrease at 25 °C, with a marked decrease in triglycerides, while white muscle presented an opposite pattern with an increase in total lipids and glycogen at 25 °C. Serum cortisol and ammonium increased at the higher temperature suggesting an increased metabolism. In the brain, high temperatures had a marked effect on monoamine neurotransmitters, with increased levels of serotonin (5-HT) and norepinephrine (NAd). Fatty acids in white muscle were modulated by high temperatures. In general, saturated fatty acids were significantly decreased at higher temperatures, while for monounsaturated fatty acids only C20:1n9 showed a decrease at 25 °C. The most abundant polyunsaturated fatty acids in white muscle were C22:6n3 (DHA) and C20:5n3 (EPA), which did not change at a higher temperature, while C20:4n6 (ARA) and C18:3n3 (ALA) showed an increase and a decrease at 25 °C, respectively. In conclusion, we suggest that E. maclovinus juveniles tolerate up to 20 °C, increasing their metabolism without negatively affecting their physiology, while a temperature of 25 °C or higher could be lethal in a long-term scenario.

Francisella noatunensis subsp. noatunensis is the responsible agent of Francisellosis, a bacterial disease that affects an important amount of aquatic farmed species. Eleginops maclovinus is a fish that cohabits with salmonids cages in Chile and can also act as a vector of this bacterial disease. In the present study, we evaluated calcium metabolism in the liver of E. maclovinus injected intraperitoneally with different doses of F. noatunensis subsp. noatunensis (low 1.5 × 101, medium 1.5 × 105 and high doses 1.5 × 1010 cells/μL). Fish were sampled at 1, 3, 7, 14, 21 and 28 days post injection (dpi). No mortalities nor clinical signs were observed. Plasma calcium levels were higher in the high doses group of F. noatunensis subsp. noatunensis at day 7 and 14 compared to the control group (fish injected with bacterial medium alone). Hypercalcemic factors increased at day 14 and 21 for the medium and low dose (parathyroid hormone-related protein precursor), while vitamin D3 receptor increased its expression at times 1, 3 and 7 for the low dose. On the other hand, hypocalcemic factors such as calcitonin receptor and stanniocalcin increased its expression at time 7 and 14, respectively. Calmodulin involved in calcium storage decreased its expression during all experimental days in fish subjected to high bacterial dose. Proteins involved in calcium transport, such as L-type voltage-gated calcium channel and trpv5 increased their transcription at day 1 and 14, compared to calcium sensing-receptor and plasma membrane Ca2 +- ATPase that showed peak expression at times 14 and 28. The results suggest a clear alteration of calcium metabolism, mainly in high bacterial doses. This study provides new knowledge about the calcium metabolism in fish infected with bacteria.

Voltage-gated ion channels are the molecular determinants of cellular excitability. This group of ion channels is one of the most important pharmacological targets in excitable tissues such as nervous system, cardiac and skeletal muscle. Moreover, voltage-gated ion channels are expressed in non-excitable cells, where they mediate key cellular functions through intracellular biochemical mechanisms rather than rapid electrical signaling. This review aims at illustrating the pharmacological impact of these ion channels, highlighting in particular the structural details and physiological functions of two of them - the high conductance voltage- and Ca(2+)-gated K(+) (BK) channels and voltage-gated proton (Hv1) channels- in non-excitable cells. BK channels have been implicated in a variety of physiological processes ranging from regulation of smooth muscle tone to modulation of hormone and neurotransmitter release. Interestingly, BK channels are also involved in modulating K(+) transport in the mammalian kidney and colon epithelium with a potential role in the hyperkalemic phenotype observed in patients with familial hyperkalemic hypertension type 2, and in the pathophysiology of hypertension. In addition, BK channels are responsible for resting and stimulated Ca(2+)-activated K(+) secretion in the distal colon. Hv1 channels have been detected in many cell types, including macrophages, blood cells, lung epithelia, skeletal muscle and microglia. These channels have a central role in the phagocytic system. In macrophages, Hv1 channels participate in the generation of reactive oxygen species in the respiratory burst during the process of phagocytosis.

In this study we assessed the influence of three different environmental salinities (5, 15 and 31 psu during 90 days) on growth, osmoregulation, energy metabolism and digestive capacity in juveniles of the Notothenioid fish Eleginops maclovinus. At the end of experimental time samples of plasma, liver, gill, intestine, kidney, skeletal muscle, stomach and pyloric caeca were obtained. Growth, weight gain, hepatosomatic index and specific growth rate increased at 15 and 31 psu and were lower at 5 psu salinity. Gill Na(+), K(+)-ATPase (NKA) activity presented a "U-shaped" relationship respect to salinity, with its minimum rates at 15 psu, while this activity correlated negatively with salinity at both anterior and posterior intestinal portions. No significant changes in NKA activity were observed in kidney or mid intestine. Large changes in plasma, metabolite levels and enzymatic activities related to energy metabolism in liver, gill, intestine, kidney and muscle were generally found in the groups exposed to 5 and 31 psu compared to the 15 psu group. Only the pepsin activity (digestive enzymes) assessed enhanced with environmental salinity, while pyloric caeca trypsin/chymotrypsin ratio decreased. This study suggests that juvenile of E. maclovinus presents greater growth near its iso-osmotic point (15 psu) and hyperosmotic environment (31 psu). Acclimation to low salinity increased the osmoregulatory expenditure as seen by the gill and anterior intestine results, while at high salinity, branchial osmoregulatory activity was also enhanced. This requires the mobilization of lipid stores and amino acids, thereby holding the growth of fish back. The subsequent reallocation of energy sources was not sufficient to maintain the growth rate of fish exposed to 5 psu. Thus, E. maclovinus juveniles present better growth efficiencies in salinities above the iso-osmotic point and hyperosmotic environment of this species, showing their best performance at 15 psu as seen by the main osmoregulatory and energy metabolism enzymatic activities.

Aims To produce and characterize egg yolk immunoglobulin (IgY) against the fish intracellular pathogen Piscirickettsia salmonis as well as to evaluate the antibacterial activity of IgY in vitro and the availability in the serum of fish immunized orally. Methods and Results Specific IgY was produced by immunizing hens with P. salmonis proteins. The IgY was obtained from egg yolks using the ammonium sulphate precipitation method and it was characterized by SDS-PAGE, Western-blot and ELISA, demonstrating that anti-P. salmonis IgY strongly reacted specifically against P. salmonis proteins. In an in vitro neutralization assay, IgY inhibited the growth of P. salmonis in liquid medium at concentrations ranging from 128 to 256 μg ml−1 in a dose-dependent manner. Interestingly, IgY against P. salmonis also generates a strong protective effect on the infection of P. salmonis in salmon head kidney-1 cells. In addition, the bacteriostatic function of IgY appears to result possibly from agglutination by the interaction of IgY with surface components of the pathogen. Finally, to confirm this IgY as an alternative for salmonid treatment, Atlantic salmon (Salmo salar) specimens were orally inoculated with IgY. The analysis of the sera demonstrates that IgY was effectively transported by fish intestine and that this immunoglobulins maintains its properties and recognizes several proteins of P. salmonis up to 12 h after inoculation of IgY against P. salmonis. Conclusions Specific IgY effectively inhibited the growth of P. salmonis and this immunoglobulin can be released in the Atlantic salmon sera when administered orally to fish. Significance and Impact of the Study We propose that this specific IgY against this fastidious micro-organism could be a useful strategy for the treatment of piscirickettsiosis.

Eleginops maclovinus is a sub-Antarctic fish endemic to the South American coast and is a monotypic species of the Eleginopidae family. A scarce amount of scientific information exists regarding the effects of temperature on E. maclovinus physiology. In this study, E. maclovinus specimens were acclimated for 2 weeks at 10 °C (control), 14, and 18 °C. Posterior assessments were performed for the intermediary metabolism of carbohydrates, lipids, and amino acids (in the plasma, liver, gills, and kidney) and on the osmoregulatory capacity [plasma osmolality and gill, kidney, and foregut Na+, K+-ATPase (NKA) activities]. In the plasma, only lactate and total amino acid levels were affected by the temperature. NKA activity presented a linear relationship with the increased temperatures. The liver exhibited no change in the carbohydrates metabolism, but the glycogen/glucose metabolite levels did differ. Amino acid metabolism showed a direct relationship between temperature and GDH and Asp-AT activities, and an inverse relationship between temperature and Ala-AT activity. Lipid metabolism was similar to Ala-AT. In the gills, carbohydrate metabolism (G6PDH and HK activities) presented a direct relationship with the increased temperatures. In the kidney, carbohydrate metabolism (only G6PDH activity) was greater at higher temperatures, and amino acid metabolism (GDH activity) showed a direct relationship with temperature. Lipid metabolism (G3PDH activity) was greatest at 14 °C, presenting significant differences compared to 10 °C. The obtained results suggest that E. maclovinus can acclimate to the projected thermal increase of climate change, with a reorganization of the intermediary metabolism components being tissue-dependent and osmoregulatory effects being affected by NKA activity and greater rates of nitrogenous waste extrusion.

Indexación: Scopus. Piscirickettsia salmonis is the causative bacterial agent of piscirickettsiosis, a systemic fish disease that significantly impacts the Chilean salmon industry. This bacterium possesses a type IV secretion system (T4SS), several proteins of the type III secretion system (T3SS), and a single heat shock protein 60 (Hsp60/GroEL). It has been suggested that due to its high antigenicity, the P. salmonis Hsp60 could be surface-exposed, translocated across the membrane, and (or) secreted into the extracellular matrix. This study tests the hypothesis that P. salmonis Hsp60 could be located on the bacterial surface. Immunogold electron microscopy and proteomic analyses suggested that although P. salmonis Hsp60 was predominantly associated with the bacterial cell cytoplasm, Hsp60-positive spots also exist on the bacterial cell envelope. IgY antibodies against P. salmonis Hsp60 protected SHK-1 cells against infection. Several bioinformatics approaches were used to assess Hsp60 translocation by the T4SS, T3SS, and T6SS, with negative results. These data support the hypothesis that small amounts of Hsp60 must reach the bacterial cell surface in a manner probably not mediated by currently characterized secretion systems, and that they remain biologically active during P. salmonis infection, possibly mediating adherence and (or) invasion. https://www.mdpi.com/2076-2607/8/1/117

Caligus rogercresseyi, a marine ectoparasite, causes notable economic losses for the Chilean salmonid industry. Nevertheless, the immunological responses of infected fish remain poorly understood, including proinflammatory cytokine generation and the respective modulatory effects of various cytokine receptors. This study evaluated mRNA expression of the NLRC5, major histocompatibility complex (MHC) class II, I-kappa-B-alpha, a regulatory that inhibits NF-kappa-B, and proinflammatory cytokines (IL-1&beta, and IL-18) in the liver and muscle of Atlantic salmon (Salmo salar) and Coho salmon (Oncorhynchus kisutch) during a time-course C. rogercresseyi infestation trial. All assessed mRNA were strongly regulated during infestation, but S. salar showed up-regulated expression, possibly accounting for the high infestation vulnerability of this salmonid. In conclusion, this work helps to understand the modulation of the expression of different transcripts involved over short periods of C. rogercresseyi infestation in two salmonid species (S. salar and O. kisutch).

Ferritin is a major iron storage protein essential not only in the infectious process, but also in any circumstance generating oxidative stress. In this study, the cDNA coding sequence of ferritin-H was obtained from the sub-Antarctic Notothenioid fish Eleginops maclovinus through transcriptomic analysis of the head kidney. This sequence contained a 534 bp open reading frame that coded for a 177 amino acid protein with a molecular weight of 20,786.2 Da and a theoretical pI of 5.56. The protein displayed a region of iron putative response elements in the 5′UTR, two putative ferritin iron-binding region signatures, and seven characteristic amino acids with ferroxidase functions. Phylogenetic analysis related this sequence to ferritin-H sequences of other Antarctic Notothenioid fish, sharing 96.61% similarity. Constitutive gene expression analysis in different organs revealed increased ferritin-H gene expression in the gills, spleen, muscle, and liver. After infection with two bacterial strains of Piscirickettsia salmonis (LF-89 and Austral-005), ferritin-H was differentially expressed depending on bacterial strain and tissue. This study provides relevant information towards understanding the iron metabolism of a sub-Antarctic Notothenioid fish.

The NOD-like receptors (NLRs) were recently identified as an intracellular pathogen recognition receptor family in vertebrates. While the immune system participation of NLRs has been characterized and analyzed in various mammalian models, few studies have considered NLRs in teleost species. Therefore, this study analyzed the Atlantic salmon (Salmo salar) NLRC5. Structurally, Atlantic salmon NLRC5 presented leucine-rich repeat subfamily genes. Phylogenetically, NLRC5 was moderately conserved between S. salar and other species. Real-time quantitative PCR revealed NLRC5 expression in almost all analyzed organs, with greatest expressions in the head kidney, spleen, and hindgut. Furthermore, NLRC5 gene expression decreased during smolt stage. These data suggest that NLRC5 participates in the Atlantic salmon immune response and is regulated, at least partly, by the smoltification process, suggesting that there is a depression of immune system from parr at smolt stage. This is the first report on the NLRC5 gene in salmonid smolts.

Using a streptozotocin-induced type 1 diabetic rat model, we analyzed and separated the effects of hyperglycemia and hyperinsulinemia over the in vivo expression and subcellular localization of hepatic fructose 1,6-bisphosphatase (FBPase) in the multicellular context of the liver. Our data showed that FBPase subcellular localization was modulated by the nutritional state in normal but not in diabetic rats. By contrast, the liver zonation was not affected in any condition. In healthy starved rats, FBPase was localized in the cytoplasm of hepatocytes, whereas in healthy re-fed rats it was concentrated in the nucleus and the cell periphery. Interestingly, despite the hyperglycemia, FBPase was unable to accumulate in the nucleus in hepatocytes from streptozotocin-induced diabetic rats, suggesting that insulin is a critical in vivo modulator. This idea was confirmed by exogenous insulin supplementation to diabetic rats, where insulin was able to induce the rapid accumulation of FBPase within the hepatocyte nucleus. Besides, hepatic FBPase was found phosphorylated only in the cytoplasm, suggesting that the phosphorylation state is involved in the nuclear translocation. In conclusion, insulin and not hyperglycemia plays a crucial role in the nuclear accumulation of FBPase in vivo and may be an important regulatory mechanism that could account for the increased endogenous glucose production of liver of diabetic rodents. Caligus rogercresseyi sea lice negatively impact Chilean salmonid farming, but no complete characterization for the metabolic effects of Caligus infestation currently exists. Therefore, the aim of this study was to analyze the effects of C. rogercresseyi infestation on the metabolic responses of Salmo salar and Oncorhynchus kisutch. Energy metabolism responses to C. rogercresseyi were examined over a time-course infestation of both salmonid species. Plasma metabolite levels and enzymatic activities related to intermediate metabolisms of carbohydrate, amino acid, and lipid were evaluated in the liver and muscle. Plasma glucose levels changed in both salmonid species, increasing at 1–3 days post-infestation. In turn, triglyceride levels increased at days 3 and 7 in O. kisutch and S. salar, respectively, while protein and total α-amino acids increased in O. kisutch but decreased in S. salar during infestation. Amino acid intermediate catabolic metabolism in the liver and muscle of O. kisutch increased during infestation, indicating a higher use of the gluconeogenic pathway than S. salar. Lipid intermediate anabolic metabolism increased in O. kisutch liver, remained unchanged in S. salar liver, and increased 1 day post-infestation in the muscle of both salmonids. Liver and muscle carbohydrate intermediate anabolic metabolism in O. kisutch increased during infestation, suggesting that this species preferentially uses the glycogenolytic-glucolytic pathway, in contrast with S. salar. In conclusion, amino acid and carbohydrate catabolism enzymes in O. kisutch activated soon after initial sea lice infestation, which would allow this species to dispose of energy substrates earlier than S. salar. This physiological data contributes towards the ability of O. kisutch to more adeptly cope with the increased energy demand imposed by C. rogercresseyi infestation.

This study describes de novo transcriptome sequencing and annotation analyses for the head kidney of the sub-Antarctic notothenioid fish Eleginops maclovinus, a sister group of the Antarctic notothenioid fish clade. Moreover, E. maclovinus is one of the most eurythermal and euryhaline representatives of the Notothenioidei suborder. RNA-seq data were generated by the 454 GS Junior system, resulting in 11,207 contigs that were then assembled by the Genomic Workbench CLC software. The transcriptome was annotated by BLASTing each sequence against the universal, non-redundant NCBI database (National Center for Biotechnology Information) using the AUSTRAL-omics computer cluster. A significant number of transcripts related to innate and adaptive immunity were found in the sequences, which could be used as references in future immunological studies in E. maclovinus.
