Nlrp3 is a vital integration point of diverse extracellular stimuli and cellular stress. However, inappropriate activation of Nlrp3 results in progression of autoinflammatory and metabolic disorders. Atrazine, using widely in agricultural sector, is a toxic environmental factor for human. Here, this study found that atrazine could induce oxidative stress and the expression of Nfkb and IRF1 in spleen, promoting the ox-mtDNA formation. And production and release of ox-mtDNA stimulated the Nlrp3 inflammasome. Lastly, atrazine induced the pyroptosis in spleen mediating the activation of Nlrp3 inflammasome. Additionally, lycopene, a kind of carotenoid, is natural bioactive component in fruits and vegetables, which is applied towards reducing oxidative stress. It was found that lycopene could ameliorate the pyroptosis induced by atrazine via inhibition of the ox-mtDNA production. The results also provided evidence that lycopene had the potential role in prevention Nlrp3 inflammasome activation by depleting the ox-mtDNA.

One of the most intriguing concepts in animal ecology is the reproductive advantages offered by larger body size, and the females prefer to mate with larger males to gain reproductive advantage. Currently, it is not clear how females recognize signs of male 'quality' and what mechanisms are involved in producing offspring with direct or indirect benefits. Our study aims to assess the preferences of females for males in Ophraella communa, determine the reproductive benefits and reveal the underlying mechanism behind this advantage. We demonstrate that male body size is an important determinant in the evolutionary process of O. communa, affecting female mate choice. Moreover, our study establishes that females prefer males with a larger body size, and this could further improve the developmental and reproductive fitness of their offspring. Finally, we focus on the seminal fluid proteins (SFPs) in O. communa, determine differentially expressed genes (i.e. OcACE, OcCBP and OcSFP) by analysing their proteomes and transcriptomes, and define the role of these SFPs-related genes through RNAi. Our study proved that the reproductive benefit of large males may be regulated by biased expression of crucial SFPs genes. The present study advances our understanding of the biological significance of preferential mating.

A 19-year-old male patient who suffered from sudden and repeated multiple organ dysfunction syndrome one month after the bar removal procedure of Nuss surgery for pectus excavatum was admitted to our department. With organ function supportive treatment, the etiology was finally identified to be a bone spur located at the inner border of the left costa due to repeated friction between the implanted steel bar and the rib, which damaged the heart repeatedly and induced the consequent acute cardiac tamponade. After operation, the patient was successfully managed and discharged. Follow-ups till three years indicated a good recovery.

Lycopene, a natural bioactive component, has potential to reduce the risk of environmental factors inducing chronic diseases. It is important to explore lycopene's health benefits and its mechanism. The uncontrolled use of atrazine in agriculture causes critical environmental pollution issues worldwide. Exposure to atrazine through water and food chains is a risk to humans. In this study, mice were orally treated with lycopene and/or different concentrations of atrazine for 21 days to explore the influence of atrazine on the spleen and the role of lycopene's protection in atrazine exposure. The work found that atrazine exerted its toxic role in the B cell zone of the spleen by inducing Foxo1 deficiency. Atrazine caused ROS generation and Pink1/Parkin dysfunction via inducing Foxo1 deficiency, which led to apoptosis in the B cell zone. Additionally, the work revealed that lycopene ameliorates atrazine-induced apoptosis in the B cell zone of the spleen via regulating the miR-27a-3p/Foxo1 pathway. The finding also underscored a novel target of lycopene in maintaining homeostasis during B cell maturation.

Agasicles hygrophila , a leaf-feeding Chrysomelid beetle, was introduced into China in 1986 as a biological control for Alternanthera philoxeroides (alligatorweed), a noxious perennial invasive weed. However, Hunan’s Ag. hygrophila populations decrease suddenly during summers (at high temperatures), leading to the inefficient control of alligatorweed. To investigate this phenomenon, herein, the effects of four different temperatures (26, 30, 36, or 39°C) on the reproductive capacity and egg-hatch, pupation, eclosion, and adult survival rates of Ag. hygrophila were explored. The effects of four different durations of each temperature treatment (4, 8, 12, or 16 days) were investigated. All the studied parameters were significantly affected following each of the four temperature and time treatments. At 30°C, the egg-hatch, pupation, and eclosion rates were considerably lower than those in the control group at 26°C. However, there were no significant differences in the number of eggs laid and the adult survival rates. Heat-shock at 36°C and 39°C led to significant decreases in the number of eggs laid, and the egg-hatch, pupation, eclosion, and adult survival rates compared to those at 26°C. Furthermore, high temperature had the greatest effect on Ag. hygrophila pupae, and the eclosion rate was close to zero; this may have caused the sharp decrease in the Ag. hygrophila population. The results will help better understanding the impact of biological control on Al. philoxeroides during summer temperatures and will also support optimization of biocontrol strategies, notably for the release of biocontrol agents across different geographical regions with different climates.

Atrazine (ATR), a ubiquitous environmental contaminant in water and soil, causes environmental nephrosis. To reveal the toxic effect of ATR on the kidney and the potential chemical nephroprotective effect of lycopene (LYC), Kun-Ming mice of specific pathogen-free (SPF) grade were treated with LYC (5 mg kg-1) and/or ATR (50 mg kg-1 or 200 mg kg-1) for 21 days. The degree of renal injury was evaluated by measuring the ion concentration, ATPase activities and the mRNA expressions/levels of associated ATPase subunits. In addition, the expression of renal aquaporins (AQPs) was analyzed. The results showed that the renal tubular epithelial cells of ATR-exposed mice were swollen, the glomeruli were significantly atrophied, and the ion concentrations were obviously changed. The activity of Na+-K+-ATPase and the transcription of its subunits were downregulated. The activity of Ca2+-Mg2+-ATPase and the transcription of its subunits were upregulated. The expression of AQPs, especially the critical AQP2, was affected. Notably, ATR-induced nephrotoxicity was significantly improved by LYC supplementation. Therefore, LYC could protect the kidney against ATR-induced nephrotoxicity via maintaining ionic homeostasis, reversing the changes in ATPase activity and controlling the expression of AQPs on the cell membrane. These results suggested that AQP2 was a target of LYC and protected against ATR-induced renal ionic homeostasis disturbance.

Here, during a routine surveillance, an unexpectedly high prevalence of Escherichia coli with reduced susceptibility to colistin (69.9%) was observed in a Chinese broiler farm. Fifty-three (63.9%) E. coli isolates were positive for mcr-1. All identified mcr-1-positive E. coli (MCREC) were multidrug resistant and carried other clinically significant resistance genes. Furthermore, the mcr-1 genes were mainly located on the IncI2 and IncHI2 plasmids. Conjugation experiments unraveled the co-transfer of mcr-1 with other antibiotic resistance genes (blaCTX-M-55, blaCTX-M-14, floR, and fosA3) via the IncI2 (n=3) and IncHI2 (n=4) plasmids. The stable genetic context mcr-1-pap2 was common in the IncI2 plasmids, whereas ISApl1-mcr-1-pap2-ISApl1 was mainly found in the IncHI2 plasmids. The dominance of mcr-1-bearing IncI2 and IncHI2 plasmids and co-selection of mcr-1 with other antimicrobial resistance genes might contribute to the exceptionally high prevalence of mcr-1 in this broiler farm. Our results emphasized the importance of appropriate antibiotic use in animal production.

Atrazine (ATR) is a widely used herbicide with biologically toxic effects that can lead to neurotoxicity. Lycopene (LYC) is an antioxidant with chemoprotective properties. However, little know about the mechanisms of preventative interventions about LYC alleviated ATR-induced neurotoxicity. Objective: This study aimed to detect whether optic nerve sheath diameter (ONSD) can be used to dynamically monitor intracranial pressure (ICP). Methods: Adult patients being monitored for invasive ICP on day one after hospital admission were included. For each patient, ONSD was first measured in the supine position, then at the 30° head-up position. The dynamic test was conducted in 16 patients. The ONSD was measured in the supine position once a day for three continuous days (day one to day three after admission).Results: The ONSD and ICP values obtained in the supine position on admission were strongly correlated (r of 0.799). When patients changed their position from supine to the 30° head-up position, the ICP and ONSD values decreased correspondingly. However, the change in ICP did not correlate well with the change in ONSD (r of 0.358). In the dynamic test, a good agreement between ICP and ONSD only existed in three (18.75%) patients. In addition, three patients had completely different profiles for ICP and ONSD.Conclusions: The results suggest that the changes in ONSD and ICP values were not correlated well after dynamic observation. Measurement of ONSD may not be a suitable tool to dynamically monitor ICP.

Low winter temperatures are a severe environmental pressure for newly arriving insect species. Adaptive evolutionary changes in cold tolerance facilitate establishment of these species in new environments. Ambrosia artemisiifolia, a noxious invasive plant, is distributed from southern to northern China. As an effective biological control agent of A. artemisiifolia, Ophraella communa is mainly spread in southern China. Previously, O. communa successfully established stable populations in Beijing (39.98°N, 115.97°E) following introduction from Laibin (23.62°N, 109.37°E) in 2012. This means that O. communa has adapted the cold winter temperatures in Beijing. However, the mechanisms underlying its rapid evolution of cold tolerance remain unknown. We investigated the levels of cryoprotectants and energy reserves in adult O. communa from two latitudes. We found that higher levels of trehalose, proline, glycerol, total sugar, and lipid were accumulated in O. communa from high latitudes. We also identified five potential genes (Tret1a, Tret1b, Tret1-2, P5CS, and GST) responsible for regulating cold tolerance that were highly expressed in high latitudes. These genes were involved in trehalose transport, proline biosynthesis, and glutathione S-transferase activation. These changes in hybridization indicated facilitating adaptation to cold temperatures. Our results demonstrate the genetic basis underlying the rapid adaptation of cold tolerance in O. communa, which indicates its extension in regions of A. artemisiifolia distribution at higher latitudes. This implies that the specialist herbivores can often adapt to a new low temperature environment where their host plant exist by rapid genetic evolution.

Di(2-ethylhexyl) phthalate (DEHP), as a widely used plasticizer, is reported to have widespread environmental and global health hazards. Trace amounts of phthalates in the environment are sufficient to disrupt ecological balance and affect human health. However, DEHP-induced splenic toxicity remains in an unknown state. Therefore, to explore the mechanism of DEHP-induced splenic toxicity, male quail were employed with 0, 250, 500 and 750 mg/kg body weight DEHP by daily gastric perfusion for 45 days. Notably, splenic corpuscular border and cell gap enlargement were observed in the spleen tissue of DEHP-exposed quail under the histopathological analysis. Furthermore, DEHP induced dysregulation of oxidative stress markers by increasing malondialdehyde (MDA) content and decreasing superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activities. Low concentration of DEHP (≤250 mg/kg) exposure suppressed nuclear factor-E2-related factor 2 (Nrf2) signaling pathway, while high concentration of DEHP (≥500 mg/kg) exposure activated Nrf2-mediated defense response. DEHP induced splenic oxidative stress via interfering Nrf2 signal pathway and altering the transcription of its downstream genes. In conclusion, this study suggested that DEHP induced splenic toxicity.

Ophraella communa (Coleoptera: Chrysomelidae), the ragweed leaf beetle, is a biological control agent of the invasive common ragweed, Ambrosia artemisiifolia (Asterales: Asteraceae). Adults can survive cold conditions that occur during winter. The adults mate before entering overwintering. Understanding the connection between copulation and overwintering will be useful for determining O. communa seasonality. Determining the relationship between overwintering and copulation required comparison of mated and unmated beetles at mean lethal temperature (LTemp50) exposures for 2 h. Cold-related physiological indices, including the water ratio, super cooling point (SCP), cryoprotectant levels, and energy reserve levels, were also measured. Mating treatment decreased the LTemp50 of both sexes by reducing their mean SCP and water ratios. Although the changes of cryoprotectant levels in mated adults were not precisely consistent in between the genders, they increased greatly in both males and females. Body sugar may play a role in copulation and may also elevate cold hardiness in O. communa.

Ambrosia artemisiifolia is a noxious invasive alien weed, that is harmful to the environment and human health. Ophraella communa is a biocontrol agent for A. artemisiifolia, that was accidentally introduced to the Chinese mainland and has now spread throughout southern China. Recently, we found that upon artificial introduction, O. communa can survive in northern China as well. Therefore, it is necessary to study the cold hardiness of O. communa. Many genes have been identified to play a role in cold-tolerance regulation in insects, but the function of the gene encoding non-muscle myosin regulatory light chain (MRLC-sqh) remains unknown. To evaluate the role played by MRLC-sqh in the cold-tolerance response, we cloned and characterized MRLC-sqh from O. communa. Quantitative real-time PCR revealed that MRLC-sqh was expressed at high levels in the gut and pupae of O. communa. The expression of MRLC-sqh was shown to decrease after cold shock between 10 and 0 &deg, C and ascend between 0 and &minus, 10 &deg, C, but these did not show a positive association between MRLC-sqh expression and cold stress. Silencing of MRLC-sqh using dsMRLC-sqh increased the chill-coma recovery time of these beetles, suggesting that cold hardiness was reduced in its absence. These results suggest that the cold hardiness of O. communa may be partly regulated by MRLC-sqh. Our findings highlight the importance of motor proteins in mediating the cold response in insects.

The rate of occurrence of fosA3 in ESBL-producing Enterobacterales from retail vegetables is high, and fosA3 was found to be carried by diverse plasmids. Some novel genetic contexts of fosA3 and novel fosA3-carrying plasmids, including several plasmid types common in K. pneumoniae, were identified, increasing the number of known transfer vectors for the fosA3 gene and reflecting the complexity of fosA3 transmission in Enterobacterales. The capture of fosA3 by the resident plasmid of K. pneumoniae will accelerate the spread of fosA3 in K. pneumoniae, one of the most pathogenic species in clinical medicine. Considering the clinical importance of fosfomycin, and the fact that vegetables are directly consumed, the fosfomycin resistance genes present a risk of transmission to the human body through the food chain and thus pose a threat to public health., The plasmid-mediated fosfomycin resistance gene fosA3 has been detected in Enterobacterales from various sources but has rarely been reported in vegetables. In this study, the aim was to investigate the prevalence of and, subsequently, to characterize fosA3-positive Enterobacterales isolates from retail vegetables. Seventeen (7.3%) fosA3-carrying strains were identified from 233 extended-spectrum-β-lactamase-producing Enterobacterales isolates from vegetables. All 17 isolates, including six Escherichia coli, seven Klebsiella pneumoniae, two Raoultella ornithinolytica, and two Citrobacter freundii isolates, carried blaCTX-M. S1-nuclease pulsed-field gel electrophoresis (S1-PFGE) and hybridization confirmed that the fosA3 genes in 16 isolates were located on plasmids ranging in size from ∼40 kb to ∼250 kb, except one located on chromosome of C. freundii. All the fosA3-carrying plasmids from 16 fosA3-positive isolates were successfully transferred into the recipient bacteria by transformation or conjugation. In agreement with data determined with isolates from food animals, the IncHI2/ST3 and IncN-F33:A-:B-/F33:A-:B plasmids were the main vectors of fosA3 in E. coli. Additionally, F24:A-:B1, IncFIIK-IncR, IncFIIS, IncR, and two untypeable plasmids were found for the first time to be vectors for fosA3 in Enterobacterales. The genetic contexts of fosA3 in 15 Enterobacterales isolates differed due to insertion and/or loss of molecular modules mediated by mobile elements. However, all fosA3 genes were flanked by IS26, as commonly observed in other fosA3-carrying plasmids. Here, we report a high rate of detection of fosA3 genes, mediated by multiple plasmid vectors, in ESBL-producing Enterobacterales from retail vegetables. FosA3-producing Enterobacterales could be transmitted to the human body by direct contact or consumption of vegetables, which might pose a potential threat to public health. Objectives This study aimed to determine whether the serial changes of the electrocardiogram is associated with regression of left ventricular hypertrophy (LVH) after Liwen procedure. Background Clinical application of the echocardiography-guided percutaneous intramyocardial septal radiofrequency ablation (PIMSRA, Liwen procedure) is an innovative approach to treat hypertrophic obstructive cardiomyopathy (HOCM). Methods We enrolled 30 consecutive patients with HOCM who had undergone Liwen procedure in our Hypertrophic Cardiomyopathy Center, from June 2016 to January 2018. Electrocardiography (ECG) and echocardiogram were performed before and after Liwen procedure, and at each follow-up (1-week, 1, 3, 6 months and 1 year). The Sokolow–Lyon index (SLi), Q wave, R wave, S wave amplitude of 12-lead ECG and interventricular septal (IVS) thickness, left ventricular mass index (LVMI) by echocardiograms were measured in each patient. The sum of the ECG QRS amplitude on each lead was calculated. The reduction of SLi and QRS amplitude were used as improvement index. Results The ECG leads with most improvement rate of the QRS wave amplitude of all cases were V1 and V2, both at 90%. The QRS wave amplitude in V1 leads and SLi were positively correlated with IVS thickness and LVMI at baseline and 1 year after Liwen procedure, respectively. The reduction of IVS thickness, LVMI and QRS wave amplitude in leads V1 and V2 were significant at one month after ablation and the follow-up period. SLi was significantly decreased at 3 months during the observation period. Similarly, the improvement of ECG QRS wave amplitude after the Liwen procedure tracked the gradual thinning of the IVS and the changes of SLi reflected the regression of LVH. Conclusion The QRS wave amplitude reductions in lead V1 and SLi may be good indicators for evaluating the postoperative interventricular septal remodeling of the Liwen procedure.

The alligator weed flea beetle, Agasicles hygrophila is a monophagous natural enemy of the invasive alligator weed Alternanthera philoxeroides. Oogenesis plays a vital role in the process of individual development and population continuation of oviparous insects. Sox is an ancient and ubiquitous metazoan gene family that plays a key regulatory role in various physiological processes, including oogenesis, which is closely related to fecundity. In this study, two Sox genes AhDichaete and AhSox3 were cloned and characterized, and then the expression profiles of AhDichaete and AhSox3 were qualified by a quantitative reverse transcription-polymerase chain reaction. The result showed that these two Sox genes were expressed significantly higher in ovary, especially in the adult developmental stage. Furthermore, the functions of AhDichaete and AhSox3 in A. hygrophila females were studied using RNA interference (RNAi). Fewer offsprings were produced when AhDichaete and AhSox3 RNAi females mated with wild-type males. Moreover, dsAhSox3 injection reduced the hatching rate of eggs but injection with dsAhDichaete did not. Further study of the reproductive system of AhDichaete and AhSox3 RNAi females showed that yolk protein deposition reduction in the ovarioles, then the expression of vitellogenin gene AhVg2 in ovaries was decreased. These results indicate that AhDichaete and AhSox3 play an important regulatory role in the process of ovarian development and oogenesis by affecting yolk synthesis in the ovary of A. hygrophila.

The spread and oviposition of Ophraella communa LeSage (Coleoptera Chrysomelidae) adults on Ambrosia artemisiifolia L. (Asterales: Asteraceae) were affected by wind direction. The densities of adults and eggs were the highest on plants located in the direction from which the wind blew. Our results suggest that oviposition preference of O. communa adults may be more dependent on the odors emitted by host plants than on their color. BackgroundWe previously reported four heterozygous missense mutations of MYH7, KCNQ1, MYLK2, and TMEM70 in a single three-generation Chinese family with dual Long QT and hypertrophic cardiomyopathy phenotypes for the first time. However, the clinical course among the family members was various, and the potential myocardial dysfunction has not been investigated.ObjectivesThe objective of this study was to investigate the echocardiographic and electrocardiographic characteristics in a genetic positive Chinese family with hypertrophic cardiomyopathy and further to explore the association between myocardial dysfunction and electric activity, and the identified mutations.MethodsA comprehensive echocardiogram – standard two-dimensional Doppler echocardiography and three-dimensional speckle tracking echocardiography – and electrocardiogram were obtained for members in this family.ResultsAs previously reported, four missense mutations – MYH7-H1717Q, KCNQ1-R190W, MYLK2-K324E, and TMEM70-I147T – were identified in this family. The MYH7-H1717Q mutation carriers had significantly increased left ventricular mass indices, elevated E/e’ ratio, deteriorated global longitudinal stain, but enhanced global circumferential and radial strain compared with those in non-mutation patients (all pKCNQ1-R190W carriers showed significantly prolonged QTc intervals, and the MYLK2-K324E mutation carriers showed inverted T-waves (both pTMEM70-I147T mutation carriers had similar echocardiography and electrocardiographic data as non-mutation patients.ConclusionsThree of the identified four mutations had potential pathogenic effects in this family: MYH7-H1717Q was associated with increased left ventricular thickness, elevated left ventricular filling pressure, and altered myocardial deformation; KCNQ1-R190W and MYLK2-K324E mutations were correlated with electrocardiographic abnormalities reflected in long QT phenotype and inverted T-waves, respectively.

Life-history strategies of animals are governed by fitness-related trait trade-offs. In particular, the age at which either one of the two sexes copulate has been shown to affect insect reproductive output and longevity. This suggests that trade-offs between longevity and reproduction might drive the choice of a mating partner based on their age and ultimately such choices might select for different life-histories. Although several studies indicate that female age at mating dictates subsequent insect longevity, fecundity, and egg hatch rate, it is unclear how male age at copulation affects these life-history traits. Common ragweed (Ambrosia artemisiifolia) is a notorious invasive weed that has spread across most temperate regions of the world. The beetle (Ophraella communa) is considered to be an effective control agent against A. artemisiifolia. As an oligophagous insect, its olfactory system is extremely important for host seeking in the wild. To the best of our knowledge, there is no report on the molecular mechanisms underlying olfaction recognition in this beetle. Hence, in this study, we characterized the odorant receptor co-receptor of O. communa and named it as 'OcomORco'. Real-time quantitative PCR (qRT-PCR) showed that, compared to the control treatment, RNA interference (RNAi) strongly reduced the expression of OcomORco by 89% in male and 90% in female beetles. Electroantennogram assay showed that the antennal response of both male and female beetles to four volatiles of A. artemisiifolia was significantly reduced. The injected male or female beetles lost their preference for plant leaves as observed in the behavioural tests. In addition, disruption of the expression of OcomORco resulted in a reduction of oviposition, while there was no difference in larval hatching rate between control and knockdown females. We demonstrated that OcomORco plays a vital role in olfactory perception and host search in O. communa, and it is involved in oviposition in an indirect way.

Agasicles hygrophila is an effective biological control agent for the invasive weed Alternanthera philoxeroides, and because of this it has been introduced to many parts of the world where A. philoxeroides is a problem. Despite this, there are no reports at present about the reproduction of this important insect. Vitellogenin receptors (VgRs) belong to the superfamily of low-density lipoprotein receptors (LDLRs). One of the roles of VgRs is to regulate the absorption of yolk protein in insects. In this study, the full length vitellogenin receptor gene (AhVgR) from A. hygrophila was sequenced and found to encode a predicted protein of 1,642 amino acids. Sequence analysis of AhVgR revealed that it contains conserved structural motifs common to LDLR family members, and a phylogenetic analysis placed AhVgR as a separate group among the order Coleoptera. AhVgR was found to be specifically expressed in ovarian tissues, and it is first transcribed in the newly-emerged females. The expression patterns are consistent with VgR genes in other insects. RNA interference (RNAi)-mediated suppression of AhVgR gene expression in adult A. hygrophila females inhibited yolk protein deposition in the ovaries, shortened the ovariole, drastically reduced egg production, and ultimately led to a decrease in fecundity. In summary, our work shows that AhVgR is critical for transporting Vg into the oocytes and plays an important role in A. hygrophila reproduction.

The alligatorweed flea beetle Agasicles hygrophila is an insect used for biological control of the aquatic weed Alternanthera philoxeroides (alligatorweed). Because these insects are oviparous, synthesis, and transportation of yolk proteins is integral to reproduction. Vitellin, the chief protein constituent in egg yolk, is mainly synthesized in the fat body and its synthesis is regulated by the transcript levels of Vitellogenin (Vg). In our study, we first cloned and characterized three Vg genes from A. hygrophila and quantified the expression levels of these Vgs in different tissues and developmental stages by RT-qPCR. Analysis of the full-length cDNA sequences of the three A. hygrophila Vg genes revealed that the open reading frames of AhVg1, AhVg2, and AhVg3 were 5175, 5346, and 5385 bp, encoding 1724, 1781, and 1794 amino acids, respectively. RT-qPCR analysis revealed that these three AhVgs have similar expression patterns; expression in the fat body was significantly higher than that in other tissues, and the highest expression was observed in the adult developmental stage. RNA interference was used to explore the functions of the AhVgs. A. hygrophila female adults injected with dsRNA targeting the AhVg genes showed decreased AhVg gene expression. Down regulation of all three AhVgs significantly affected ovary development, reduced egg laying capacity, and reduced the egg hatch rate compared with the control groups. Our findings provide the basis for further study of the functions of Vg genes in other insect species.

Cytochrome P450 monooxygenases (P450s) are a large class of enzymes that play essential roles in metabolic processes such as hormone synthesis and the catabolism of toxins and other chemicals in insects. In the present study, we identified 82 P450 genes using comprehensive RNA sequencing in the flea beetle Agasicles hygrophila, and all of the sequences were validated by cloning and sequencing. Phylogenetic analysis showed that the P450 genes in A. hygrophila fell into the mitochondrial clan, CYP2 clan, CYP3 clan and CYP4 clan and were classified into 20 families and 48 subfamilies. Most A. hygrophila P450 genes had high sequence homology with those from other coleopteran insects. To understand the effects of high temperatures on the metabolic processes of female and male adults, we studied the effects of two temperature regimes (constant temperature of 28 °C for 20 h with a 4-h period of high temperatures of 30 °C and 39 °C) on the expression levels of P450 genes in A. hygrophila using RT-PCR and qRT-PCR. The results showed that there were no differences in expression in 30 P450 genes between the control and high-temperature-treated A. hygrophila adults, while 22 P450 genes showed up-regulated expression and 19 P450 genes were down-regulated in A. hygrophila female adults after high-temperature treatment. For A. hygrophila male adults exposed to high temperatures, we found that 8 P450 genes had higher expression levels and 12 P450 genes had lower expression levels under the same conditions. The P450 genes are candidates that showed significantly different expression levels after high-temperature treatments in A. hygrophila adults, and further studies are needed to determine their possible roles in metabolic processes during the response to elevated temperatures.

Atrazine (ATR), an environmental persistent and bioaccumulative herbicide, has been associated with environmental nephrosis. Lycopene (LYC) exhibits important properties of nephroprotection, but there are limited data on the specific underlying mechanism. The primary objective of this study was to explore the therapeutic effect of LYC on ATR-induced nephrotoxicity in mice. The mice were divided randomly into 6 groups and treated as follows: control group (C), 5 mg/kg LYC group (L), 50 mg/kg ATR group (A1), 200 mg/kg ATR group (A2), 50 mg/kg ATR plus 5 mg/kg LYC group (A1+L), and 200 mg/kg ATR plus 5 mg/kg LYC group (A2+L) by oral gavage administration for 21 days. We found that pretreatment with LYC significantly suppressed the ATR-induced renal tubular epithelial cell swelling. Furthermore, LYC mitigated ATR-induced dysregulation of oxidative stress markers by reducing MDA, H2O2 levels, and increasing SOD, GPx, CAT concentration, and Nrf2 activation. Moreover, LYC activated the autophagic flux by a detectable change in autophagy-related genes (Beclin-1 and ATGs) and proteins (p62/SQSTM) and by the formation of autophagic vacuole (AV) and LC3 aggregation, in parallel with AMPK activation (pAMPK/AMPK). Herein, ATR-up-regulated nuclear factor erythroid 2-related factor 2 (Nrf2) expression and Nrf2-regulated redox genes, including quinoneoxidoreductase-1 (NQO1) and heme oxidase-1 (HO1), whereas LYC down-regulated those of the above genes. In addition, LYC suppressed ATR-induced activation of autophagy (increased LC3II/LC3I, ATGs, Beclin1, and p62, in parallel with increased AMPK activation). Collectively, our findings identified a cross talk between AMPK-activated autophagy and the Nrf2 signaling pathway in LYC-mediated nephroprotection against ATR-induced toxicity in mice kidney.

Ophraella communa LeSage (Coleoptera: Chrysomelidae) was accidentally introduced in China, and it is a biological control agent of common ragweed, Ambrosia artemisiifolia L. (Asterales: Asteraceae). Life history and life table parameters of O. communa were studied in the open field of Langfang city, Hebei province, north China (39°30′42″N, 116°36′07″E) in early spring. Development durations for egg, larva, pupa and entire immature stage of O. communa were 13.6, 21.1, 7.9 and 42.8 d, respectively. Adults could survive in 37.41 d, no differences in the longevities between female and male, and fecundity was 713.1 eggs per female. The intrinsic rate of increase (r), finite rate of increase (λ), net reproductive rate (R0), and mean generation time (T) were 0.0660 d− 1, 1.0683 d− 1, 73.9 offspring/individual and 63.0 d, respectively. The results of our study can offer valuable insight on the population demography of O. communa in early spring in the field. In addition, the effects of temperature fluctuation on the development, survival and fecundity of O. communa and the potential of using the beetle for controlling A. artemisiifolia are also discussed in the early spring.

1. Why animals mate multiple times, owing to the lack of immediate fitness benefits, presents an intriguing problem for evolutionary biologists. Yet, the profusion of this behaviour suggests it must be maintained by natural selection via increased performance. 2. The possible benefits of multiple mating using the leaf beetles Ophraella communa LeSage, the biological control agent of the invasive common ragweed Ambrosia artemisiifolia L., were studied and the fitness consequences of single, twice, three, four, and unrestricted mating events were assessed. 3. Overall, it was observed that the number of copulation events was positively associated with fitness parameters of the insects. Insects performed the best under unrestricted mating regimes, with average increases of 48% in longevity, 75% in fecundity, and 55% in egg hatch rate. In addition, females that experienced unrestricted access to mates maintained very high viability over their entire reproductive lives. Nevertheless, insects also performed better when allowed to mate four times compared with once or twice. 4. The present findings thus support the hypothesis that multiple-mating behaviour is maintained owing to increased fitness benefits in the current and the next generation. Selection for re-mating is, therefore, expected in field populations, which is likely to happen naturally owing to the aggregate lifestyle of O. communa.

Ophraella communa LeSage (Coleoptera: Chrysomelidae) is an important biological control agent of the common ragweed, Ambrosia artemisiifolia L., in China. Development and fecundity of O. communa, and hatch rate of progeny eggs were studied at five photoperiods (8:16, 10:14, 12:12, 14:10, and 16:8 [L:D] h). The highest survival rate of eggs was 92% at the photoperiod of 16:8 (L:D) h, and those of both larvae and pupae were observed at the photoperiod of 14:10 (L:D) h (85 and 96%). The shortest developmental durations of larvae and pupae were observed at photoperiods of 14:10 and 16:8 (L:D) h. Fecundity was 1,159-1,976 eggs per female from photoperiods of 8:16 to 16:8 (L:D) h. The hatch rates of progeny eggs were 67-92% from photoperiods of 8:16 to 16:8 (L:D) h, and photoperiods did not affect developmental duration of progeny eggs. The intrinsic rate for increase (r), the net reproductive rate (r0), and the finite rate of increase (λ) reached the maximum values at 16:8 (L:D) h (0.2219 d(-1), 721 hatched eggs and 1.2484 d(-1), respectively) and 14:10 (L:D) h (0.2133 d(-1), 605.6 hatched eggs and 1.2378 d(-1), respectively). Their minimum values were observed at the photoperiod of 8:16 (L:D) h, which were 0.1731 d(-1), 212.2 hatched eggs and 0.1890 d(-1), respectively. The shortest T value was 29.7 d at a photoperiod of 16:8 (L:D) h and the longest was 31.4 d at a photoperiod of 12:12 (L:D) h. Our study shows that O. communa could survive and reproduce successfully at different photoperiods, thus may expand its distribution to regions with different photoperiods.

Determining the best ratio of females to males of an insect's natural enemy is important for maximising population increase and promoting population establishment of a natural enemy. In this study, copulation behaviour, fecundity, progeny fitness and rate of population increase for the flea beetle, Agasicles hygrophila Selman & Vogt (Coleoptera: Chrysomelidae), were compared at different female percentage treatments (i.e., 80%, 66.7%, 50%, 33.3% and 20% females). The results showed that the copulation frequency and duration in males decreased, whereas those in females increased as the number of males increased. At 20%, 33.3% and 66.7% females, the rates of population increase were 3.4-, 2.17- and 0.79-fold higher than that at 50% females. Females at 20% and 33.3% were found to be optimal for mass rearing of the beetle.

Ophraella communa LeSage is native to North America and a biological control agent of the invasive weed Ambrosia artemisiifolia L. Since A. artemisiifolia plants grow old and die after September annually, O. communa suffers from food shortage. To understand the effect of food shortage or deprivation on population fitness of O. communa, the development and fecundity and hatchability of its progeny eggs were observed when larvae were offered A. artemisiifolia plants for either 3, 6, 12 or 24 hours daily. The results showed that larval food deprivation significantly influenced survival and developmental durations of larvae and pupae. Survival rates and developmental durations of larvae and pupae decreased and were prolonged significantly with decreasing time of larval daily food intake.
