Abstract Imbalance of proinflammatory and anti-inflammatory responses plays a crucial role in the progression of abdominal aortic aneurysms. ILF3, a known modulator of the innate immune response, is involved in cardiovascular diseases. This study aims to investigate the role of ILF3 in abdominal aortic aneurysm formation. Here, we use multi-omics analyzes, transgenic male mice, and multiplex immunohistochemistry to unravel the underlying involvement of ILF3 in abdominal aortic aneurysms. The results show that macrophage ILF3 deficiency attenuates abdominal aortic aneurysm progression, while elevated macrophage ILF3 exacerbates abdominal aortic aneurysm lesions. Mechanistically, we reveal that macrophagic ILF3 increases NF-κB activity by hastening the decay of p105 mRNA, leading to amplified inflammation in macrophages. Meanwhile, ILF3 represses the anti-inflammatory action by inhibiting the Keap1-Nrf2 signaling pathway through facilitating the ILF3/eIF4A1 complex-mediated enhancement of Keap1 translational efficiency. Moreover, Bardoxolone Methyl treatment alleviates the severity of abdominal aortic aneurysm lesions in the context of elevated ILF3 expression. Together, our findings underscore the significance of macrophage ILF3 in abdominal aortic aneurysm development and suggest its potential as a promising therapeutic target for abdominal aortic aneurysms.

Abstract Background Due to its unique anatomical characteristics, supracondylar fractures of the humerus are often difficult to achieve firm fixation with internal fixation equipment, resulting in delayed functional exercise, often leaving cubitus varus deformity, elbow stiffness, contractures, and other complications. Here, we report an adult patient with a supracondylar fracture of the humerus who underwent internal fixation through an anterior median incision in the humerus with our self-developed anterior anatomical locking plate of the distal humerus. Case presentation A 29-year-old male patient of Chinese ethnicity with trauma-induced right supracondylar fracture of the humerus and multiple soft tissue contusions, without nerve damage, blood vessel damage, or other injuries, underwent an internal incision in our hospital using a new anatomical locking plate for the anterior distal humerus fixed treatment. During the 16-month follow-up period, the patient’s elbow range of motion was almost completely restored, functional scores were excellent, and there were no minor or major postoperative complications. Conclusion In this study, we propose a surgical reconstruction strategy for adult patients with supracondylar humeral fractures. Through the anterior median incision of the humerus, open reduction and internal fixation were performed with an anatomic locking plate on the anterior side of the distal humerus to restore and fix the structure of the distal humerus, and satisfactory clinical results were achieved in our case.

Abstract Hyperglycemia accelerates calcification of atherosclerotic plaques in diabetic patients, and the accumulation of advanced glycation end products (AGEs) is closely related to the atherosclerotic calcification. Here, we show that hyperglycemia-mediated AGEs markedly increase vascular smooth muscle cells (VSMCs) NF90/110 activation in male diabetic patients with atherosclerotic calcified samples. VSMC-specific NF90/110 knockout in male mice decreases obviously AGEs-induced atherosclerotic calcification, along with the inhibitions of VSMC phenotypic changes to osteoblast-like cells, apoptosis, and matrix vesicle release. Mechanistically, AGEs increase the activity of NF90, which then enhances ubiquitination and degradation of AGE receptor 1 (AGER1) by stabilizing the mRNA of E3 ubiquitin ligase FBXW7, thus causing the accumulation of more AGEs and atherosclerotic calcification. Collectively, our study demonstrates the effects of VSMC NF90 in mediating the metabolic imbalance of AGEs to accelerate diabetic atherosclerotic calcification. Therefore, inhibition of VSMC NF90 may be a potential therapeutic target for diabetic atherosclerotic calcification.

Abstract Water eutrophication has emerged as a pressing concern for massive algal blooms, and these harmful blooms can potentially generate harmful toxins, which can detrimentally impact the aquatic environment and human health. Consequently, it is imperative to identify a safe and efficient approach to combat algal blooms to safeguard the ecological safety of water. This study aimed to investigate the procedure for extracting total flavonoids from Z. bungeanum residue and assess its antioxidant properties. The most favorable parameters for extracting total flavonoids from Z. bungeanum residue were a liquid–solid ratio (LSR) of 20 mL/g, a solvent concentration of 60%, an extraction period of 55 min, and an ultrasonic temperature of 80 °C. Meanwhile, the photosynthetic inhibitory mechanism of Z. bungeanum residue extracts against M. aeruginosa was assessed with a particular focus on the concentration-dependent toxicity effect. Z. bungeanum residue extracts damaged the oxygen-evolving complex structure, influenced energy capture and distribution, and inhibited the electron transport of PSII in M. aeruginosa. Furthermore, the enhanced capacity for ROS detoxification enables treated cells to sustain their photosynthetic activity. The findings of this study hold considerable relevance for the ecological management community and offer potential avenues for the practical utilization of resources in controlling algal blooms.

Abstract Pulse shape discrimination (PSD) is widely used in particle and nuclear physics. Specifically in liquid scintillator detectors, PSD facilitates the classification of different particle types based on their energy deposition patterns. This technique is particularly valuable for studies of the diffuse supernova neutrino background (DSNB), nucleon decay, and dark matter searches. This paper presents a detailed investigation of the PSD technique, applied in the DSNB search performed with the Jiangmen Underground Neutrino Observatory (JUNO). Instead of using conventional cut-and-count methods, we employ methods based on boosted decision trees and neural networks and compare their capability to distinguish the DSNB signals from the atmospheric neutrino neutral-current background events. The two methods demonstrate comparable performance, resulting in a 50–80% improvement in signal efficiency compared to a previous study performed for JUNO (An et al. [JUNO] in J Phys G 43(3):030401, 2016). Moreover, we study the dependence of the PSD performance on the visible energy and final state composition of the events and find a significant dependence on the presence/absence of $$^{11}$$ 11 C. Finally, we evaluate the impact of the detector effects (photon propagation, PMT dark noise, and waveform reconstruction) on the PSD performance.

Summary: Gestational diabetes mellitus (GDM) is a distinct form of diabetes that poses a significant threat to the health of both pregnant women and fetuses. The objective of this study was to investigate the impact of behenic acid (BA) on glucose metabolism, inflammation, and insulin resistance in GDM mice, and to elucidate the underlying molecular mechanism. Here, we demonstrated that daily administration of 10 mg/mL BA during pregnancy effectively ameliorated abnormal glucose metabolism in GDM mice and their offspring and improved birth outcomes in the offspring. Moreover, BA promoted the proliferation of islet β cells, restored their normal function, and augmented glucose uptake by skeletal muscle cells. Mechanistically, BA mitigated inflammation and insulin resistance in GDM mice by inhibiting activation of the TLR4/NF-κB signaling pathway. Our study provides compelling evidence supporting the efficacy of BA in improving GDM, suggesting its potential use as a dietary supplement for preventing and treating GDM.

The main focus of this study is the application of image processing in the field of identification and classification of fruits. It is an application that has received much less attention than other applications of image processing. The proposed systems presented in this article, are software solutions based on image processing techniques, including histogram matching techniques, for detection, Sable edge detection algorithms, Private edge and Kenny edge, Otsu threshold limit, and clustering method It is an automatic mean and classification of different degrees of fruit. In addition, it has been mentioned more about the examination and description of product grading and clustering methods, that by using the proposed application hardware and its connection with the software, a big step can be taken in product quality grading. This method can be used in product classification and packaging. The accuracy rate for peaches, lemons, apples, and tomatoes is 94.58%, 88.23%, 70%, and 93.33%, respectively. The best accuracy for all 20 sample levels is for peach fruit.

Acute hepatopancreatic necrosis disease (AHPND), caused by certain strains of Vibrio, has resulted in substantial economic losses in global shrimp industries. Hence, developing effective and sustainable alternatives to antibiotics to control this disease is of profound importance. In this study, we isolated a strain of antagonistic bacterium, JSHY-K3, from shrimp pond sediment, evaluated its efficacy in preventing AHPND in shrimp and tentatively investigated its mechanism of defence and control against AHPND. The isolated strain exhibited strong antagonistic activity against Vibrio parahaemolyticus, the etiological agent of AHPND (VpAHPND) harboring toxin genes pirA and pirB. JSHY-K3 was identified as B. subtilis based on its morphological, physiological, biochemical characteristics and 16 S rDNA sequence analysis. In vivo challenge assays demonstrated that JSHY-K3 could significantly mitigate cumulative mortality of shrimps infected with VpAHPND, suppress the expression of VpAHPND virulence genes and modulate shrimp immune responses. To elucidate the mechanistic basis of JSHY-K3 in preventing and controlling AHPND, whole-genome sequencing and untargeted metabolomics-LCMS-EMDB analyses were conducted. Whole-genome analysis revealed 12 gene clusters involved in synthesizing of secondary metabolites including the bacteriostatic agents surfactin, aurantinin B/C/D, fengycin, sublancin 168, bacillibactin, subtilosin A and bacilysin. Untargeted metabolomics uncovered 3691 known metabolites, with caffeic acid, fosfomycin, minocycline and surfactin A documented as V. parahaemolyticus growth inhibitors. We hypothesize that the production of these bacteriostatic metabolites by JSHY-K3 underlies its antagonistic effects against VpAHPND. Collectively, these findings suggest B. subtilis JSHY-K3 could be applied as a probiotic to prevent AHPND in shrimp aquaculture.

Objective: The objective of this study was to evaluate the efficacy and safety of Tongxin Formula in the treatment of coronary microvascular disease. Methods: We conducted a randomized, double-blind, placebo-controlled study simultaneously in two hospitals, consisting of 80 participants. Using a random number table, we assigned patients to the treatment and control groups. Patients in both groups received conventional Western medicine for coronary microvascular disease. In addition, those in the treatment group received Tongxin formula granules, while those in the control group received a placebo. The treatment course for both groups was three months, and the follow-up duration was six months. The primary efficacy indicators were coronary blood flow reserve and cardiovascular adverse events; the secondary efficacy indicators were the traditional Chinese medicine (TCM) syndrome score, the angina symptom score, the Seattle Angina Questionnaire (SAQ) score, left ventricular function, and adverse reactions. Results: After treatment, patients in the treatment group showed significantly higher variation in the coronary flow reserve (CFR) levels (CFR >2) and improvement of diastolic function (peak filling rate, or PFR >2.5) than those in the control group (P 0.05). Conclusion: We found that Tongxin Formula combined with conventional Western medicine can significantly improved the level of coronary blood flow reserve, reduced the occurrence of cardiovascular adverse events, improved the clinical symptoms of patients, and enhanced the quality of life of patients with coronary microvascular disease with favorable safety.

BackgroundToxic epidermal necrolysis (TEN) and Stevens-Johnson syndrome (SJS) are rare, life-threatening immunologic reactions. Previous relevant literature has provided limited information regarding this disease’s genetic susceptibility and management principles.ObjectivesThis study aimed to describe a phenobarbital-induced TEN case report with HLA-B*15:02 and HLA-B*58:01 negative, CYP2C19*1/*2. In addition, we revised the existing literature on phenobarbital-induced SJS/TEN to explore its clinical characteristics.MethodsWe describe a woman undergoing treatment with Phenobarbital for status epilepticus who developed classic cutaneous findings of TEN. A systematic search was conducted in the PubMed, Medline, WanFang, and CNKI databases from 1995 to 2023. The search terms used were “Stevens-Johnson Syndrome,” “Toxic Epidermal Necrolysis,” and “Phenobarbital.”ResultsWe report a case of TEN resulting from phenobarbital; it tested negative for the HLA-B*15:02 and HLA-B*58:01 allele and CYP2C19*1/*2 intermediate metabolism. Supportive treatment with steroids and antihistamines resulted in complete resolution of the skin lesions and improvement in clinical symptoms after 14 days. Physicians and clinical pharmacists should be aware of these potential phenobarbital-related adverse events and closely monitor patients with first-time use of phenobarbital. Among 19 cases were identified in the literature, with 11 (57.9%) cases of SJS, 6 (31.6%) cases of TEN, and 2 (7.2%) cases of SJS-TEN/DRESS overlap. A total of 5 (26.3%) did not survive, of which 4 (21.1%) were under 12 years old and 1 (5.3%) was over 12 years old.ConclusionPhenobarbital-induced SJS/TEN may still occur in patients who test negative for HLA-B*15:02 and HLA-B*58:01, CYP2C19*1/*2. Most cutaneous adverse events occur early in the course of Phenobarbital therapy and should be closely monitored early in the course of treatment. In addition, Phenobarbital should be used with caution in patients with a history of asthma and allergy to antipyretics and analgesics.

As pivotal structural component, steels operating at high temperatures play a vital role in promoting the development of advanced equipment technology. Enduring and efficient lubricants are critical bottleneck for the construction of high-temperature frictional subsystems. Polydiethylsiloxane oil (PDES) significantly reduces the coefficient of friction (COF) (9.80–16.74 %) and wear rate (28.95–54.59 %) in the Si3N4–8Cr4Mo4V system without sulfur and phosphorus anti-wear additives at 250 °C. With only 1 wt% PDES, there is a remarkable 28.95 % wear rate reduction in the Si3N4–8Cr4Mo4V frictional system. The enhanced lubrication performance come from the Si–O tribofilm at lower PDES concentrations (1–15 wt%). Once exceeding a PDES concentration of 30 wt%, the as-formed Si–O tribofilm transitions to a Fe–Mo compound film, exhibiting superior friction reduction and wear resistance properties.

Abstract Background Clear cell renal cell carcinoma is a prototypical tumor characterized by metabolic reprogramming, which extends beyond tumor cells to encompass diverse cell types within the tumor microenvironment. Nonetheless, current research on metabolic reprogramming in renal cell carcinoma mostly focuses on either tumor cells alone or conducts analyses of all cells within the tumor microenvironment as a mixture, thereby failing to precisely identify metabolic changes in different cell types within the tumor microenvironment. Methods Gathering 9 major single-cell RNA sequencing databases of clear cell renal cell carcinoma, encompassing 195 samples. Spatial transcriptomics data were selected to conduct metabolic activity analysis with spatial localization. Developing scMet program to convert RNA-seq data into scRNA-seq data for downstream analysis. Results Diverse cellular entities within the tumor microenvironment exhibit distinct infiltration preferences across varying histological grades and tissue origins. Higher-grade tumors manifest pronounced immunosuppressive traits. The identification of tumor cells in the RNA splicing state reveals an association between the enrichment of this particular cellular population and an unfavorable prognostic outcome. The energy metabolism of CD8+ T cells is pivotal not only for their cytotoxic effector functions but also as a marker of impending cellular exhaustion. Sphingolipid metabolism evinces a correlation with diverse macrophage-specific traits, particularly M2 polarization. The tumor epicenter is characterized by heightened metabolic activity, prominently marked by elevated tricarboxylic acid cycle and glycolysis while the pericapsular milieu showcases a conspicuous enrichment of attributes associated with vasculogenesis, inflammatory responses, and epithelial–mesenchymal transition. The scMet facilitates the transformation of RNA sequencing datasets sourced from TCGA into scRNA sequencing data, maintaining a substantial degree of correlation. Conclusions The tumor microenvironment of clear cell renal cell carcinoma demonstrates significant metabolic heterogeneity across various cell types and spatial dimensions. scMet exhibits a notable capability to transform RNA sequencing data into scRNA sequencing data with a high degree of correlation.

Infectious myonecrosis virus (IMNV), first identified in 2003, predominantly infects the shrimp species Litopenaeus vannamei and has caused significant economic losses in major shrimp farming regions in Brazil and Southeast Asia. However, the infection mechanisms of IMNV has not been elucidated. In this study we have investigated the molecular mechanisms of IMNV-shrimp interaction and identified potential antiviral key genes to provide a theoretical reference for developing new control technologies. Transcriptome sequencing of control and infected shrimp groups at 30 (early), 60 (mid), and 90 days (late) post-infection identified 695, 2411, and 401 differentially expressed genes (DEGs), respectively. Early infection analysis revealed IMNV promotes colonization and spread by inhibiting hemagglutination and apoptosis-related genes while activating antiviral pathways like JAK-STAT, indicating it evades host defenses by interfering with the innate immune response. During mid-stage infection, glucose and amino acid metabolism pathways were enriched, suggesting IMNV induces host metabolic reprogramming to support viral replication. Late-stage analysis showed downregulated apoptosis and autophagy pathways, and upregulated cytoskeleton and extracellular matrix genes, indicating the host mitigates viral tissue damage by regulating cell fate and repair mechanisms. This transcriptomics study uncovers dynamic shrimp gene expression changes induced by IMNV infection, identifying multiple immune and metabolic pathways involved in the antiviral response. It elucidates IMNV's pathogenic mechanism and identifies key regulatory genes in pathways like PI3K-Akt-mTOR and JAK-STAT. These findings enhance our understanding of IMNV-shrimp host interaction and could inform novel antiviral strategy development.

Background: Cardiac rehabilitation is a beneficial multidisciplinary treatment of exercise promotion, patient education, risk factor management, and psychosocial counseling for people with coronary heart disease (CHD) that is underutilized due to substantial disparities in access, referral, and participation. Empirical studies suggest that cardiac telerehabilitation (CTR) have safety and efficacy comparable to traditional in-person cardiac rehabilitation, however, older adults are under-reported with effectiveness, feasibility, and usability remains unclear. Methods: The study randomized 43 older adults (84 % males) to the 12-week CTR intervention or standard of care. Guided by Social Cognitive Theory, participants received individualized in-person assessment and e-coaching sessions, followed by CTR usage at home. Data were collected at baseline (T0), six-week (T1), and 12-week (T2). For many applications, raw satellite observations need to be converted to high-level products of various essential environmental variables. While numerous products are available at kilometer spatial resolutions, there are few global products at high spatial resolutions (10–30 m), which are also referred to fine or medium resolutions in the literature. To facilitate the development of more high spatial resolution products, this paper systematically reviews the state-of-the-art progress on inversion algorithms and publicly available regional and global products. We begin with an inventory of available high-resolution satellite data, and then present different algorithms for determining cloud masks, estimating aerosol optical depth, and performing atmospheric correction and topographic correction for land surface reflectance retrieval. The majority of this paper reviews the inversion algorithms and existing regional to global products of 18 variables in four major categories: 1) Land surface radiation, including broadband albedo, land surface temperature, and all-wave net radiation; 2) Terrestrial ecosystem variables, including leaf area index, fraction of absorbed photosynthetically active radiation, fractional vegetation cover, fractional forest cover, tree height, forest above-ground biomass gross primary production, net primary production, and agricultural crop yield; 3) Water cycle and cryosphere, including soil moisture, evapotranspiration, and snow cover; and 4) Land surface types, such as global land cover, impervious surface, inland water, crop type, and fire. Since the existing products over large regions are usually spatially discontinuous due to cloud contamination, different data fusion and data assimilation algorithms and some products for producing spatially seamless and temporally continuous products are presented. In the end, we discuss a variety of challenges in generating global high spatial resolution satellite products.

With the change of seasons, the shear strength of saline soil subgrade filler will change with the change of external temperature, which will aggravate the adverse effects of seismic on the subgrade. To explore the influence of seismic action on the stability of saline soil subgrade under the influence of temperature on the strength of saline soil subgrade filler, this paper first carried out saline soil shear tests at different temperatures to obtain the influence of temperature on the shear strength of saline soil. Then, the temperature field of the saline soil subgrade was simulated, and then based on the subgrade isothermal stratification model and FLAC3D, the displacement and acceleration amplification effects of seismic action on the shady slope, sunny slope and subgrade of saline soil subgrade in different months were analyzed. The following conclusions were finally drawn: under the action of seismic, In the process of the change of subgrade temperature of Qarhan - Golmud Expressway between −7.7 ​°C and 27 ​°C, the change of saline soil cohesion is the main factor affecting the stability of subgrade slope, and the maximum and minimum values of subgrade surface settlement appear in September and June of each year, respectively. In August, the differences of settlement between the shady slope and the sunny slope shoulder of the subgrade were the largest, and the acceleration of the shady slope and the sunny slope and the inside of the subgrade changed most significantly in the vertical direction. Special attention should be paid to the seismic early warning in the above key months; In the range from both sides of the shoulder to the centerline of the roadbed, the acceleration amplification effect starts to increase significantly from about 3m from the centerline of the roadbed to the centerline, so it is necessary to pay attention to the seismic design of this range.

Background: Bevacizumab resistance poses barriers to targeted therapy in clear cell renal cell carcinoma (ccRCC). Whether there exist epigenetic targets that modulate bevacizumab sensitivity in ccRCC remains indefinite. The focus of this study is to explore the role of UCHL1 in ccRCC. Methods: Both in vitro and in vivo experiments were utilized to investigate the roles of UCHL1 in ccRCC. In vivo ubiquitination assays were performed to validate the posttranslational modification of KDM4B by UCHL1. Luciferase reporter and chromatin immunoprecipitation (ChIP) assays were utilized to explore KDM4B/VEGFA epigenetic regulations. Results: UCHL1 was increased in ccRCC and associated with unfavorable survival outcomes in patients. UCHL1 was required for ccRCC growth and migration. Mechanistically, the wild-type UCHL1, but not C90A mutant, mediated the deubiquitination of KDM4B and thereby stabilized its proteins. KDM4B was up-regulated in ccRCC and potentiated cell growth. UCHL1 depended on KDM4B to augment ccRCC malignancies. Targeting UCHL1 suppressed tumor growth, colony formation, and migration abilities, which could be rescued by KDM4B. Furthermore, KDM4B was directly bound to the promoter region of VEGFA, abolishing repressive H3K9me3 modifications. KDM4B coordinated with HIF2α to activate VEGFA transcriptional levels. UCHL1-KDM4B axis governs VEGFA levels to sustain the angiogenesis phenotypes. Finally, a specific small-molecule inhibitor (6RK73) targeting UCHL1 remarkably inhibited ccRCC progression and further sensitized ccRCC to bevacizumab treatment. Conclusion: Overall, this study defined an epigenetic mechanism of UCHL1/KDM4B in activating VEGF signaling. The UCHL1-KDM4B axis represents a novel target for treating ccRCC and improving the efficacy of anti-angiogenesis therapy.

ObjectiveEphedra, widely used in clinical practice as a medicinal herb, belongs to the genus Ephedra in the family Ephedraceae. However, the presence of numerous Ephedra varieties and variants requires differentiation for accurate identification.MethodsIn this study, we employed headspace gas chromatography mass spectrometry (HS-GC-MS), ultra-high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS), and global natural products social molecular networking (GNPS) for chemical component identification. Chemometric analysis was used to analyze the differential components. Metabolic analysis and Kyoto encyclopedia of genes and genomes (KEGG) enrichment were utilized to explore the synthesis pathways of different components.ResultA total of 83 volatile and 79 non-volatile components were identified in Ephedra species. Differential analysis revealed that among the eight Ephedra stems, 18 volatile and 19 non-volatile differential compounds were discovered, whereas Ephedra roots exhibited 21 volatile and 17 non-volatile markers. Volatile compounds were enriched in four synthetic pathways, while non-volatile components were enriched in five pathways among the differentiated components.ConclusionThis study is the first to conduct a comparative analysis of chemical components in different Ephedra species and parts. It provides a foundational reference for authenticating Ephedra herbs, evaluating medicinal resources, and comparing quality in future studies.

In the rapidly evolving landscape of smart transportation, the passenger volume in urban rail transit consistently demonstrates an upward trajectory. In this context, precise and scientifically grounded short-term passenger flow prediction methods are essential for optimizing operational scheduling and ensuring safety in urban rail transit. Consequently, this paper introduces Temporal Graph Attention Long Short-Term Memory (TGALSTM), a spatiotemporal integrated prediction network model that incorporates the surrounding environment of the station. Initially, the paper enhanced the Temporal Convolutional Network (TCN) model to capture temporal features more accurately. Subsequently, the paper utilizes the Graph Attention Network (GAT) network module specifically to extract the topological structure and surrounding environmental features of the station. Lastly, the prediction task is accomplished by weighted fusion of various features, inputting them into the Attention Long Short-Term Memory (LSTM) network. Experiments were conducted on two authentic datasets, revealing that the TGALSTM model outperforms the baseline model in both single-step and double-step predictions, showcasing the model’s exceptional performance and robustness. This research offers a robust method and support to enhance the operational efficiency and passenger flow management of urban rail transit systems.

Abstract Background Autophagic defects are involved in Methamphetamine (Meth)-induced neurotoxicity. Syntaxin 17 (Stx17), a member of the SNARE protein family, participating in several stages of autophagy, including autophagosome-late endosome/lysosome fusion. However, the role of Stx17 and potential mechanisms in autophagic defects induced by Meth remain poorly understood. Methods To address the mechanism of Meth-induced cognitive impairment, the adenovirus (AV) and adeno-associated virus (AAV) were injected into the hippocampus for stereotaxis to overexpress Stx17 in vivo to examine the cognitive ability via morris water maze and novel object recognition. In molecular level, the synaptic injury and autophagic defects were evaluated. To address the Meth induced neuronal damage, the epidermal growth factor receptor (EGFR) degradation assay was performed to evaluate the degradability of the “cargos” mediated by Meth, and mechanistically, the maturation of the vesicles, including autophagosomes and endosomes, were validated by the Co-IP and the GTP-agarose affinity isolation assays. Results Overexpression of Stx17 in the hippocampus markedly rescued the Meth-induced cognitive impairment and synaptic loss. For endosomes, Meth exposure upregulated Rab5 expression and its guanine-nucleotide exchange factor (GEF) (immature endosome), with a commensurate decreased active form of Rab7 (Rab7-GTP) and impeded the binding of Rab7 to CCZ1 (mature endosome); for autophagosomes, Meth treatment elicited a dramatic reduction in the overlap between Stx17 and autophagosomes but increased the colocalization of ATG5 and autophagosomes (immature autophagosomes). After Stx17 overexpression, the Rab7-GTP levels in purified late endosomes were substantially increased in parallel with the elevated mature autophagosomes, facilitating cargo (Aβ42, p-tau, and EGFR) degradation in the vesicles, which finally ameliorated Meth-induced synaptic loss and memory deficits in mice. Conclusion Stx17 decrease mediated by Meth contributes to vesicle fusion defects which may ascribe to the immature autophagosomes and endosomes, leading to autophagic dysfunction and finalizes neuronal damage and cognitive impairments. Therefore, targeting Stx17 may be a novel therapeutic strategy for Meth-induced neuronal injury.

Abstract The electrochemical conversion of nitrate to ammonia is a way to eliminate nitrate pollutant in water. Cu-Co synergistic effect was found to produce excellent performance in ammonia generation. However, few studies have focused on this effect in high-entropy oxides. Here, we report the spin-related Cu-Co synergistic effect on electrochemical nitrate-to-ammonia conversion using high-entropy oxide Mg0.2Co0.2Ni0.2Cu0.2Zn0.2O. In contrast, the Li-incorporated MgCoNiCuZnO exhibits inferior performance. By correlating the electronic structure, we found that the Co spin states are crucial for the Cu-Co synergistic effect for ammonia generation. The Cu-Co pair with a high spin Co in Mg0.2Co0.2Ni0.2Cu0.2Zn0.2O can facilitate ammonia generation, while a low spin Co in Li-incorporated MgCoNiCuZnO decreases the Cu-Co synergistic effect on ammonia generation. These findings offer important insights in employing the synergistic effect and spin states inside for selective catalysis. It also indicates the generality of the magnetic effect in ammonia synthesis between electrocatalysis and thermal catalysis.

Abstract Many types of tumors feature aerobic glycolysis for meeting their increased energetic and biosynthetic demands. However, it remains still unclear how this glycolytic phenomenon is achieved and coordinated with other metabolic pathways in tumor cells in response to growth stimuli. Here we report that activation of AKT1 induces a metabolic switch to glycolysis from the mitochondrial metabolism via phosphorylation of cytoplasmic malic enzyme 2 (ME2), named ME2fl (fl means full length), favoring an enhanced glycolytic phenotype. Mechanistically, in the cytoplasm, AKT1 phosphorylates ME2fl at serine 9 in the mitochondrial localization signal peptide at the N-terminus, preventing its mitochondrial translocation. Unlike mitochondrial ME2, which accounts for adjusting the tricarboxylic acid (TCA) cycle, ME2fl functions as a scaffold that brings together the key glycolytic enzymes phosphofructokinase (PFKL), glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and pyruvate kinase M2 (PKM2), as well as Lactate dehydrogenase A (LDHA), to promote glycolysis in the cytosol. Thus, through phosphorylation of ME2fl, AKT1 enhances the glycolytic capacity of tumor cells in vitro and in vivo, revealing an unexpected role for subcellular translocation switching of ME2 mediated by AKT1 in the metabolic adaptation of tumor cells to growth stimuli.

Eukaryotic genomes are hierarchically packaged into cell nucleus, affecting gene regulation. The genome is organized into multiscale structural units, including chromosome territories, compartments, topologically associating domains (TADs), and DNA loops. The identification of these hierarchical structures has benefited from the development of experimental approaches, such as 3C-based methods (Hi-C, ChIA-PET, etc.), imaging tools (2D-FISH, 3D-FISH, Cryo-FISH, etc.) and ligation-free methods (GAM, SPRITE, etc.). In recent two decades, numerous studies have shown that the 3D organization of genome plays essential roles in multiple cellular processes via various mechanisms, such as regulating enhancer activity and promoter-enhancer interactions. However, there are relatively few studies about the 3D genome in livestock species. Therefore, studies for exploring the function of 3D genomes in livestock are urgently needed to provide a more comprehensive understanding of potential relationships between the genome and production traits. In this review, we summarize the recent advances of 3D genomics and its biological functions in human and mouse studies, drawing inspiration to explore the 3D genomics of livestock species. We then mainly focus on the biological functions of 3D genome organization in muscle development and its implications in animal breeding.

Abstract Superhydrophobic coatings show great application potential in many fields, but they have not been widely used due to poor stability. A Ni-PTFE-SiC composite coating was proposed in this paper. Firstly, the Ni-PTFE-SiC coating structure was designed by molecular dynamics simulations. The feasibility of superhydrophobic effect of this structure and the adsorption stability of the composite coatings were proved. This approach can also be used to avoid trial and errors when selecting other similar candidate substances. Then, the Ni-PTFE-SiC superhydrophobic coatings were successfully prepared by composite electrodeposition, with the water contact angle of more than 155° and the sliding angle of close to 0°. The effects of current density, deposition time and nanoparticle concentration on the wettability and morphology of the coatings were also investigated. Through comparative experiments, the best preparation parameters were: PTFE 14 g L−1, SiC 6 g L−1, primary current density 7 A dm−² (10 min) and secondary current density 15 A dm−² (5 min). This preparation method doesn’t need additional modification with low surface energy substances like the traditional preparation process, and greatly improves the corrosion resistance, wear resistance and durability of the coatings.

A high resolution, acceptable accuracy and low power consumption time-domain temperature sensor is proposed and simulated in this paper based on a 180 nm standard CMOS technology. A diode stacking structure is introduced to enhance the accuracy of the temperature sensing core. To improve the resolution of the sensor, a dual-input capacitor multiplexing voltage-to-time converter (VTC) is implemented. Additionally, a low-temperature drift voltage-mode relaxation oscillator (ROSC) is proposed, effectively reducing the large oscillation frequency drift caused by significant temperature impacts on delay errors. The simulated results show that the resolution is as high as 0.0071 °C over 0∼120 °C with +0.43 °C/−0.38 °C inaccuracy and 1.9 pJ · K2 resolution FoM, consuming only 1.48 μW at a 1.2 V supply voltage.

Microglia respond to stressors by secreting cytokines or growth factors, playing a crucial role in maintaining brain homeostasis. While the antidepressant-like effects of Polygonatum sibiricum polysaccharides (PSPs) have been observed in mice, their potential effectiveness involving microglial regulation remains unknown. This study investigates the antidepressant-like mechanism of PSP by regulating microglial phenotype and signaling pathways in the prefrontal cortex of chronic restraint stress (CRS)-induced mice. PSP was extracted, purified, characterized, and orally administered to CRS mice. High-performance gel permeation chromatography (HPGPC) revealed that PSP has a molecular weight of 5.6 kDa. Scanning electron microscopy (SEM) and atomic force microscopy (AFM) showed that PSP exhibited a layered structure with densely packed, irregular surfaces. PSP treatment significantly increased sucrose preference (low: 71%, p < 0.01; medium: 69%, p < 0.05; high: 75%, p < 0.001 vs. CRS: 58%) and reduced immobility time (low: 74 s, p < 0.01; medium: 68 s, p < 0.01; high: 79 s, p < 0.05 vs. CRS: 129 s), indicating the alleviation of depressive-like behaviors. PSP inhibited microglial activation (PSP, 131/mm2 vs. CRS, 173/mm2, p = 0.057), reversing CRS-induced microglial hypertrophy and hyper-ramification. Furthermore, PSP inactivated microglial activation by inhibiting NLRP3/ASC/caspase-1/IL-1β signaling pathways, increasing BDNF synthesis and activating brain-derived neurotrophic factor (BDNF)-mediated neurogenesis (PSP, 80/per DG vs. CRS, 49/per DG, p < 0.01). In conclusion, PSP exerts antidepressant-like effects through the regulation of microglial activity and neuroinflammatory pathways, indicating it as a potential natural compound for depression treatment.

Myofibrillar proteins are an important component of proteins. Flavor characteristics are the key attributes of food quality. The ability of proteins to bind flavor is one of their most fundamental functional properties. The dynamic balance of release and retention of volatile flavor compounds in protein-containing systems largely affects the sensory quality and consumer acceptability of foods. At present, research on flavor mainly focuses on the formation mechanism of flavor components, while there are few reports on the release and perception of flavor components. This review introduces the composition and structure of myofibrillar proteins, the classification of flavor substances, the physical binding and chemical adsorption of myofibrillar proteins and volatile flavor substances, as well as clarifies the regulation law of flavor substances from the viewpoint of endogenous flavor characteristics and exogenous environment factors, to provide a theoretical reference for the flavor regulation of meat products.

Background: The cellular mechanism of the formation of abdominal aortic aneurysm (AAA) is very complicated. A series of sophisticated events eventually led to significant pathological changes in the anatomical structure and function of the arterial wall and they are still not clear nowadays. Methods: We pooled publicly available GEO datasets (GSE57691 and GSE47472) to get a comprehensive comparisons between normal tissues and AAA tissues to try to reveal molecular mechanism underlying the disease. Litopenaeus vannamei represents the most cultivated crustacean species, accounting for approximately 80% of global shrimp aquaculture output. The intestinal microbiota structure and functionality are known to critically influence L. vannamei growth. While previous investigations of shrimp gut microbes emphasized the whole intestine, limited research has examined the composition and functional differences of microbiota across specific intestinal sections in L. vannamei. In this study, healthy shrimp samples were collected and divided into four sections after removing the contents: S(stomach), H (from posterior stomach to the distal end of the hepatopancreas.), G (the section from hepatopancreas’s end to the sixth abdominal segment), and R(rectum). The microbial composition and function were analyzed across locations via 16 S rRNA high-throughput sequencing. The results revealed that significant α- and β-diversity distinctions existed between sections (p 10%) across sections. The rectum pathways including carbon, nitrogen, sulfur metabolisms were significantly elevated. Quorum sensing and two-component systems were also overrepresented in rectum. One-way network analysis showed that the dominant microbiota of group S, H and G were Bacillus, Mycoplasmataceae, and Flavobacterium, which are promoters of beneficial bacteria in the gut, while the dominant flora of rectumthe most competitive genus of bacteria in the intestinal tract was Vibrio,and Pseudofulvibacter, Alcaligenaceae, and Pseudomonas represent potential probiotic genera that could be screened as antagonists against Vibrio in the shrimp gut. In summary, we hypothesize the stomach and H sections degraded organic matter, G section absorbed and digested macronutrients, and rectum section recycled materials and participated in circular metabolism. These segment-specific analyses provide novel perspectives into section-specific microbial interactions and functions along L. vannamei intestine.

Abstract In optic neuropathies, including glaucoma, retinal ganglion cells (RGCs) die. Cell transplantation and endogenous regeneration offer strategies for retinal repair, however, developmental programs required for this to succeed are incompletely understood. To address this, we explored cellular reprogramming with transcription factor (TF) regulators of RGC development which were integrated into human pluripotent stem cells (PSCs) as inducible gene cassettes. When the pioneer factor NEUROG2 was combined with RGC-expressed TFs (ATOH7, ISL1, and POU4F2) some conversion was observed and when pre-patterned by BMP inhibition, RGC-like induced neurons (RGC-iNs) were generated with high efficiency in just under a week. These exhibited transcriptional profiles that were reminiscent of RGCs and exhibited electrophysiological properties, including AMPA-mediated synaptic transmission. Additionally, we demonstrated that small molecule inhibitors of DLK/LZK and GCK-IV can block neuronal death in two pharmacological axon injury models. Combining developmental patterning with RGC-specific TFs thus provided valuable insight into strategies for cell replacement and neuroprotection.

Abstract This study aimed to evaluate the pharmacokinetic (PK) similarity of the proposed biosimilar HS628 compared with the reference tocilizumab (Actemra®) and also to demonstrate similar safety and immunogenicity profiles in healthy Chinese male subjects. Eighty eligible subjects were randomized into two treatment groups in a 1:1 ratio to receive a single intravenous infusion of HS628 or tocilizumab at 4 mg/kg over 60 min. Blood samples were collected at the scheduled time points for PK and immunogenicity analysis. PK biosimilarity was determined using the standard bioequivalence criteria 80%–125%. A total of 77 subjects received the study drug and completed the study. The main PK parameters were similar for the test and reference groups. The ratio of geometric least‐squares means (GMR) and its 90% CIs for AUC0–t, AUC0–∞, and Cmax between the test group and reference group were 1.06 (1.00–1.12), 1.07 (1.00–1.14), and 1.04 (0.99–1.10), respectively, which were fully within the predefined bioequivalent range of 80%–125%. The incidence of treatment‐emergent adverse events (TEAEs) was similar for HS628 and tocilizumab (p > 0.05). The most common TEAEs were decreased fibrinogen, decreased neutrophils, pharyngalgia, oral ulcer, decreased leukocytes, and increased erythrocyte sedimentation rate. The results of the present study provide strong evidence to support the PK similarity and bioequivalence of HS628 and tocilizumab. The safety and immunogenicity profiles of HS628 were also shown to be similar to those of the reference tocilizumab.

Abstract Background Aberrant interplay between epigenetic reprogramming and hypoxia signaling contributes to renal cell carcinoma progression and drug resistance, which is an essential hallmark. How the chromatin remodelers enhance RCC malignancy remains to be poorly understood. We aimed to elucidate the roles of CHD1L in determining hypoxia signaling activation and sunitinib resistance. Methods The qRT-PCR, western blotting, and immunohistochemistry technologies were used to detect CHD1L expressions. Lentivirus transfection was used to generate stable CHD1L-KD cells. The roles of SIRT7/CHD1L were evaluated by CCK-8, wound healing, transwell assays, xenograft models, and tail-vein metastasis models. Co-immunoprecipitation, Chromatin Immunoprecipitation (ChIP), and luciferase reporter assays were conducted to explore epigenetic regulations. Results We screened and validated that CHD1L is up-regulated in RCC and correlates with poorer prognosis of patients. CHD1L overexpression notably enhances cell proliferation, migration, and self-renewal capacities in vitro and in vivo. Mechanistically, SIRT7 physically interacts with CHDL1 and mediates the deacetylation of CHD1L. Wild-type SIRT7, but not H187Y dead mutant, stabilizes CHD1L protein levels via attenuating its ubiquitination levels. SIRT7 is increased in RCC and correlates with hazardous RCC clinical characteristics. SIRT7 depends on CHD1L to exert its tumor-promoting functions. Accumulated CHD1L amplifies HIF-2α-driven transcriptional programs via interacting with HIF-2α. CHD1L recruits BRD4 and increases the RNA polymerase II S2P loading. CHD1L ablation notably abolishes HIF-2α binding and subsequent transcriptional activation. CHD1L overexpression mediates the sunitinib resistance via sustaining VEGFA and targeting CHD1L reverses this effect. Specific CHD1L inhibitor (CHD1Li) shows a synergistic effect with sunitinib and strengthens its pharmaceutical effect. Conclusions These results uncover a CHD1L-mediated epigenetic mechanism of HIF-2α activation and downstream sunitinib resistance. The SIRT7–CHD1L–HIF-2α axis is highlighted to predict RCC prognosis and endows potential targets.

Background and Purpose: Emerging researches have regarded serum chloride as a capable predictor of mortality in liver cirrhosis. We aim to investigate the clinical role of admission chloride in cirrhotic patients with esophagogastric varices receiving transjugular intrahepatic portosystemic shunt (TIPS), which is unclear. Methods: We retrospectively analyzed data of cirrhotic patients with esophagogastric varices undergoing TIPS in Zhongnan Hospital of Wuhan University. Mortality outcome was obtained by following up for 1-year after TIPS. Univariate and multivariate Cox regression were used to identify independent predictors of 1-year mortality post-TIPS. The receiver operating characteristic (ROC) curves were adopted to assess the predictive ability of the predictors. In addition, log-rank test and Kaplan–Meier (KM) analyses were employed to evaluate the prognostic value of predictors in the survival probability. Results: A total of 182 patients were included ultimately. Age, fever symptom, platelet-to lymphocyte-ratio (PLR), lymphocyte-to-monocyte ratio (LMR), total bilirubin, serum sodium, chloride, and Child-Pugh score were related to 1-year follow-up mortality. Abstract Aim To evaluate the efficacy and safety of vonoprazan–amoxicillin (VA) dual therapy for radically eradicating Helicobacter pylori (H. pylori). Methods The PubMed, Cochrane Library, Embase, China National Knowledge Infrastructure (CNKI) and Wanfang databases were searched up to July 7, 2022, to identify clinical trials comparing the efficacy of VA dual therapy and triple therapy for H. pylori eradication. After evaluating the quality of the included studies, random effects models were conducted, and risk ratios (RRs) with 95% confidence intervals (CIs) were calculated to estimate the efficacy and safety of each approach. Results Six publications (including four randomized controlled trials) involving 2019 patients were included in this meta-analysis. Overall, the eradication rate for VA dual therapy was 89.9%, while it was 85.2% for triple therapy based on other acid inhibitors. With the development of technical requirements, the current challenges faced by bearing materials mainly revolve around high-temperature conditions and the trend towards material lightweighting. Full ceramic bearings are the new candidate due to their excellent properties. This article details the tribological and adsorption performance of chlorophenyl silicone oil (CPSO) as a high-temperature lubricant in ceramic tribological systems (ZrO2, Al2O3, and Si3N4). Among the three ceramic tribological systems, the lubrication performance can be ordered as Si3N4 > Al2O3 > ZrO2, as the wear rates of the ZrO2 and Al2O3 tribo-systems are almost 1135.67 and 283.33 times larger than that of the Si3N4 tribo-system, respectively. The observed results can be explained by the superior adsorption performance of CPSO on a Si3N4 ceramic surface, which was calculated by molecular dynamic simulation. The molecular dynamic simulation results show the adsorption energy of CPSO/Si3N4 is almost 54.09 and 61.18 times higher compared to that on ZrO2 and Al2O3 ceramics. These findings provide experimental and theoretical insights for understanding the lubrication performance of CPSO in a full ceramic tribo-system.

Viruses pose a significant threat to human health, causing widespread diseases and impacting the global economy. Perilla frutescens, a traditional medicine and food homologous plant, is well known for its antiviral properties. This systematic review examines the antiviral potential of Perilla frutescens, including its antiviral activity, chemical structure and pharmacological parameters. Utilizing bioinformatics analysis, we revealed the correlation between Perilla frutescens and antiviral activity, identified overlaps between Perilla frutescens target genes and virus-related genes, and explored related signaling pathways. Moreover, a classified summary of the active components of Perilla frutescens, focusing on compounds associated with antiviral activity, provides important clues for optimizing the antiviral drug development of Perilla frutescens. Our findings indicate that Perilla frutescens showed a strong antiviral effect, and its active ingredients can effectively inhibit the replication and spread of a variety of viruses in this review. The antiviral mechanisms of Perilla frutescens may involve several pathways, including enhanced immune function, modulation of inflammatory responses, and inhibition of key enzyme activities such as viral replicase. These results underscore the potential antiviral application of Perilla frutescens as a natural plant and provide important implications for the development of new antiviral drugs.

Glycosylation is one of the most common and important modifications in natural products (NPs), which can alter the biological activities and properties of NPs, effectively increase structural diversity, and improve pharmacological activities. The biosynthesis of glycosylation in natural products involves multiple complex biological processes, which are coordinated by many enzymes. UDP-glycosyltransferases (UGTs) play a crucial role in glycosylation modification, and have attracted long-term and widespread research attention. UGTs can catalyze the O-, C-, S-, and N-glycosylation of different substrates, producing a variety of glycosides with broad biological activity, while improving the solubility, stability, bioavailability, pharmacological activity, and other functions of NPs. In recent years, the rapid development of synthetic biology and advanced manufacturing technologies, especially the widespread application of artificial intelligence in the field of synthetic biology, has led to a series of new discoveries in the biosynthesis of NP glycosides by UGT. This work summarizes the latest progress and challenges in the field of NP glycosylation, covering the research results and potential applications of glycosylated derivatives of terpenes, flavonoids, polyphenols, aromatic compounds, and other compounds in terms of biogenesis. Looking to the future, research may leverage artificial intelligence-driven synthetic biology techniques to decipher genes related to the synthetic pathway, which is expected to further promote the large-scale synthesis and application of glycosylated NPs, and increase the diversity of NPs in the pharmaceutical, functional food, and cosmetic industries.

DNA methylation is an important way to regulate gene expression in eukaryotes. In order to reveal the role of DNA methylation in the regulation of germ cell-specific piwi gene expression during spermatogenesis of Japanese flounder (Paralichthys olivaceus), the expression profiles of piwil1 (piwi-like 1) and piwil2 (piwi-like 2) genes in the gonads of female, male, and sex-reversed pseudo-male P. olivaceus were analyzed, and the dynamic of DNA methylation was investigated. As a result, piwil1 and piwil2 genes were highly expressed in the testis of both male and pseudo-male P. olivaceus, with significant variation among male individuals. The DNA methylation levels in the promoter regions of both piwil1 and piwil2 were negatively correlated with their expression levels, which may contribute to the transcriptional regulation of piwi genes during spermatogenesis. There was also sperm quality variation among male P. olivaceus, and the sperm curvilinear velocity was positively correlated with the expression of both piwil1 and piwil2 genes. These results indicated that the DNA methylation in piwil1 and piwil2 promoter regions may affect the initiation of piwi gene transcription, thereby regulating gene expression and further affecting the spermatogenesis process and gamete quality in P. olivaceus.

The method of hybrid classifying combining line classifying and surface classifying was adopted to create the classifying systems of food inspection items and food inspection methods. The methods of sequence coding, concatenation coding and combination coding were adopted to prepare the coding schemes of food inspection items and food inspection methods, and the coding structure of 6 code segments and 16 code bits was designed, Thus, the general system of food inspection classifying and coding with stable logical structure features and open data expansion functions was build. The general system showed a high degree of professional integration and a wide range of industry universality. It can assign a unique decimal Arabic numeral code to specific food inspection event, and achieve efficient query of food inspection. The prospect of using computer programming technology to develop the general information system of food inspection classifying and coding based on internet cloud, a public technical service product, was also put forward.

Ship propellers suffered from serious coupling damage of cavitation-erosion (CE) and corrosion when operated with high-speed in the marine environment. To improve the CE-corrosion resistance of the Ni–Al bronze (NAB) propeller alloy, a novel Cu-based medium-entropy alloy coating (Cu55Al20Ni12Ti8Si5, at.%) was prepared by high-velocity oxygen-fuel spraying technology in this study. The characteristics and CE behavior of coating and NAB were comparatively investigated, then the CE damage mechanism was discussed by analyzing CE damage morphologies. The influence of CE on corrosion behavior and the coupling effect between CE and corrosion were further elucidated by electrochemical corrosion experiments. The results indicated that the as-sprayed coating mainly comprised of BCC solid solution phase and NiTi-rich precipitates, which possessed higher hardness (6.73 ± 1.23 GPa) and superior resilience (27.0%) than the NAB. The cumulative mass loss rate for the coating after 18 h CE in distilled water and 3.5 wt.% NaCl solution was only 38.7% and 80.6% that for the NAB, revealing the better anti-CE performance of the coating. The preferential CE of the BCC matrix around the NiTi-rich precipitates, fatigue fracture and lamellar spalling were summarized as the main CE damage mechanism for the coating in 3.5 wt.% NaCl solution. CE had a significant influence on corrosion behavior of coating and NAB, which resulted in corrosion performance degradation of both two materials. The CE-corrosion coupling effect led to the remarkable mass loss, and mechanical effect rather than corrosion effect was ascribed to be the dominated factor for the damage of coating and NAB.

Abstract Background To evaluate the predictive effect of TCED-HFV grading and imaging biomarkers on anti-vascular endothelial growth factor (anti-VEGF) treatment in diabetic macular edema (DME). Methods 81 eyes of 81 DME patients who were treated with anti-VEGF were included in this retrospective cohort study. All patients underwent a comprehensive ophthalmic examination at baseline and follow-up, including best-corrected visual acuity (BCVA), fundus photography, and spectral domain–optical coherence tomography (SD-OCT). Baseline imaging biomarkers were qualitatively and quantitatively graded according to the TCED-HFV classification protocol, and DME was divided into early stage, advanced stage, severe stage, and atrophy stage. Abstract To create a functional neural circuit, neurons develop a molecular identity to discriminate self from non-self. The invertebrate Dscam family and vertebrate Pcdh family are implicated in determining synaptic specificity. Recently identified in Chelicerata, a shortened Dscam (sDscam) has been shown to resemble the isoform-generating characters of both Dscam and Pcdh and represent an evolutionary transition. Here we presented the molecular details of sDscam self-recognition via both trans and cis interactions using X-ray crystallographic data and functional assays. Based on our results, we proposed a molecular zipper model for the assemblies of sDscam to mediate cell-cell recognition. In this model, sDscam utilized FNIII domain to form side-by-side interactions with neighboring molecules in the same cell while established hand-in-hand interactions via Ig1 domain with molecules from another cell around. Together, our study provided a framework for understanding the assembly, recognition, and evolution of sDscam.

Introduction. Peripheral retinal refraction plays a crucial role in myopia, but the specific mechanism is not clear. We refined the retinal partitions to explore the characteristics of peripheral retinal refraction and its role in emmetropic, low, and moderate myopic children aged 6 to 12 years. Methods. A total of 814 subjects (814 eyes) were enrolled in the study. The participants were divided into three groups according to the central spherical equivalent refraction (SER), which were emmetropia group (E), low myopia group (LM) and moderate myopia group (MM). Multispectral refractive topography (MRT) was used to measure the retinal absolute and relative refractive difference value (RDV) in different regions. The range was divided into superior, inferior, temporal, and nasal RDV (SRDV, IRDV, TRDV, and NRDV) on the basis of several concentric circles extending outward from the macular fovea (RDV15, RDV30, RDV45, RDV30–15, RDV45–30, and RDV-45). Kruskal–Wallis test was used to analyze the differences of peripheral refraction for all the regions among the three groups. Spearman rank correlation was performed to explore correlations between SER and RDV, axial length (AL) and RDV. Results. High-power microwave (HPM) antenna, one of the key subsystems in the HPM system, is used for HPM radiation. Its performance is partly determined by the impulse responses of reflector materials under HPM. Therefore, it is essential to investigate the impulse response characteristics of reflector materials under HPM. Instead of the traditional waveguide cavity, this study was based on the double elliptical reflector system with an open boundary. By using this system, the impulse response of reflector materials was measured, and the response of reflector materials of multiple types under various environments with different electric field strengths and pulse widths was tested. In terms of breakdown, reflection efficiency, and pulse waveform, aluminum and carbon fiber test plates were affected by HPM and compared. Finally, the impulse response characteristics of carbon fiber test plates were summarized. The proposed method applied in this research is universal, matches the practical application environment, and can be extensively employed in the impulse response research and test of reflector materials.

This paper focuses on optimizing the management of delayed trains in operational scenarios by scientifically categorizing train delay levels. It employs static and dynamic models grounded in real-world train delay data from high-speed railways. This classification aids dispatchers in swiftly identifying and predicting delay extents, thus enhancing mitigation strategies' efficiency. Key indicators, encompassing initial delay duration, station impacts, average station delay, delayed trains' cascading effects, and average delay per affected train, inform the classification. Applying the K-means clustering algorithm to standardized delay indicators yields an optimized categorization of delayed trains into four levels, reflecting varying risk levels. This static classification offers a comprehensive overview of delay dynamics. Furthermore, utilizing Markov chains, the study delves into sequential dynamic analyses, accounting for China's railway context and specifically addressing fluctuations during the Spring Festival travel rush. This research, combining static and dynamic approaches, provides valuable insights for bolstering railway operational efficiency and resilience amidst diverse delay scenarios.
