Abstract Epstein-Barr virus (EBV) related post-transplant lymphoproliferative disorder (EBV-PTLD) is a life-threatening complication after hematopoietic stem cell transplantation (HSCT) or solid organ transplantation (SOT), for which no standard therapeutic means have been developed. Significant increase expression of natural killer group 2 member D ligands (NKG2DLs) was observed on B-lymphoblastoid cells of EBV-PTLD, indicating NKG2DLs as potential therapeutic targets for treatment of EBV-PTLD. In this study, the recombinant constructs of NKG2D CAR and IL-15/IL-15Rα-NKG2D CAR were generated with a retroviral vector and then transduced to human T cells to produce NKG2D CAR-T and IL-15/IL-15Rα-NKG2D CAR-T cells, respectively. B-lymphoblastoid cell lines (B-LCLs) and the xenografted mouse models were established to evaluate the efficacy of these CAR-T cells. IL-15/IL-15Rα-NKG2D CAR-T cells exhibited superior proliferation and antigen-specific cytotoxic effect compared to NKG2D CAR-T, as IL-15/IL-15Rα signaling promoted the expansion of less differentiated central memory T cells (TCM) and increased expression of CD107a and IFN-γ. Moreover, EBV DNA load was dramatically reduced, and 80% B-LCL cells were eliminated by IL-15/IL-15Rα-NKG2D CAR-T cells after co-culturing. Abstract Purpose To build an Mult-Task Learning (MTL) based Artificial Intelligence(AI) model that can simultaneously predict clinical stage, histology, grade and LNM for cervical cancer before surgery. Methods This retrospective and prospective cohort study was conducted from January 2001 to March 2014 for the training set and from January 2018 to November 2021 for the validation set at Beijing Chaoyang Hospital, Capital Medical University. Preoperative clinical information of cervical cancer patients was used. An Artificial Neural Network (ANN) algorithm was used to build the MTL-based AI model. Accuracy and weighted F1 scores were calculated as evaluation indicators. The performance of the MTL model was compared with Single-Task Learning (STL) models. Additionally, a Turing test was performed by 20 gynecologists and compared with this AI model. Results A total of 223 cervical cancer cases were retrospectively enrolled into the training set, and 58 cases were prospectively collected as independent validation set. The accuracy of this cervical cancer AI model constructed with ANN algorithm in predicting stage, histology, grade and LNM were 75%, 95%, 86% and 76%, respectively. And the corresponding weighted F1 score were 70%, 94%, 86%, and 76%, respectively. The average time consumption of AI simultaneously predicting stage, histology, grade and LNM for cervical cancer was 0.01s (95%CI: 0.01–0.01) per 20 patients. The mean time consumption doctor and doctor with AI were 581.1s (95%CI: 300.0-900.0) per 20 patients and 534.8s (95%CI: 255.0-720.0) per 20 patients, respectively. Except for LNM, both the accuracy and F-score of the AI model were significantly better than STL AI, doctors and AI-assisted doctors in predicting stage, grade and histology. (P

Abstract Eunicellane diterpenoids, containing a typical 6,10-bicycle, are bioactive compounds widely present in marine corals, but rarely found in bacteria and plants. The intrinsic macrocycle exhibits innate structural flexibility resulting in dynamic conformational changes. However, the mechanisms controlling flexibility remain unknown. The discovery of a terpene synthase, MicA, that is responsible for the biosynthesis of a nearly non-flexible eunicellane skeleton, enable us to propose a feasible theory about the flexibility in eunicellane structures. Parallel studies of all eunicellane synthases in nature discovered to date, including 2Z-geranylgeranyl diphosphate incubations and density functional theory-based Boltzmann population computations, reveale that a trans-fused bicycle with a 2Z-configuration alkene restricts conformational flexibility resulting in a nearly non-flexible eunicellane skeleton. The catalytic route and the enzymatic mechanism of MicA are also elucidated by labeling experiments, density functional theory calculations, structural analysis of the artificial intelligence-based MicA model, and mutational studies.

Fructo-oligosaccharides (FOS) are well-known prebiotics that have the potential to improve sow reproductive performance and increase piglet growth. However, previous studies were observed in sole FOS-supplemented diets of sows or weaned piglets and did not consider the sow-to-piglet transfer effect on the performance and diarrhea rate of weaned piglets. This study explores the effects of dietary FOS supplementation on the reproductive performance of sows, and the effects of FOS supplementation at different stages on the growth performance and diarrhea rate of weaned piglets. A split-plot experimental design was used with sow diet effect in the whole plot and differing piglet diet effect in the subplot. Fifty-two multiparous sows (223.24 ± 14.77 kg) were randomly divided into 2 groups (0 or 0.2% FOS). The experiment lasted from day 85 of gestation to day 21 of lactation. Reproductive performance, glucose tolerance, placental angiogenesis, and intestinal flora of sows were assessed. At weaning, 192 weaned piglets were grouped in 2 × 2 factorial designs, with the main effects of FOS supplemental level of sow diet (0 and 0.2%), and FOS supplemental level of weaned piglet diet (0 and 0.2%), respectively. The growth performance and diarrhea rate of the weaned piglets were analyzed during a 28-d experiment. Objective To statistically analyze the perioperative results of patients with ABO-incompatible kidney transplantation (ABOi-KT), in order to explore the changes in blood group antibody of type-A/B recipients. Methods A total of 33 cases of blood group A/B ABOi-KT recipients in our hospital from January 2021 to October 2023 were recruited and divided into two groups of group A(n=18) and group B(n=15) according to the different blood types of recipient. The effects of preoperative plasmapheresis on antibody titer, antibody rebound and renal function after operation(serum urea nitrogen, creatinine and estimated glomerular filtration rate on the 1st, 3rd, 7th and 14th day) were analyzed between the two groups. According to the postoperative rebound of blood type antibodies, 33 recipients were divided into antibody rebound group(n=7) and non rebound group(n=26), and the differences in initial blood type antibody titers between the two groups were analyzed. Results There was no significant difference in the clearance rate of IgM with preoperative plasma exchange between the two groups (Z=-0.26, P>0.05); Levels of serum urea nitrogen and creatinine on the 1st, 3rd, 7th and 14th day after operation between group A and group B were not statistically significant(P>0.05), the same as eGFR. Group B was more prone to rebound antibody compared with group A (P0.05) between the two groups was found. Conclusion The patients type B receiving type AB kidney donors are more prone to rebound antibody after ABOi-KT operation compared to the the patients type A receiving type AB.

The influences of aging temperatures (160–200 °C) and 8% pre-rolling on the mechanical properties, and microstructures of the creep-aged AA2050 alloys were investigated. The results show that for the solid-solutioned alloys after the creep aging, the strength increased with an increase of aging temperatures. The ultimate tensile strength (UTS) and yield strength (YS) reached the highest value of 527 and 484 MPa at 200 °C, respectively, although the elongation (EL) reduced to 10.8%. At expense of σ phases, the temperature increase promoted the dislocation accumulation and the dense precipitation of θ′ and T1 phases, which increased the strength. It is noted that the strong oriented precipitation at 200 °C lost some strengthening effect. After the 8% pre-rolling, the strength of creep-aged alloys increased firstly and then decreased with the increase of temperature. The high strength (UTS = 574 MPa, YS = 535 MPa) at 180 °C was attributed to the dense precipitation of T1 phases under the action of diffusion kinetic and thermodynamic driving force. When the temperature reached 200 °C, the activated ledge mechanism significantly thickened T1 phases, thus decreasing the strength. The work revealed the critical roles of aging temperature and pre-rolling in tailoring precipitation behaviors and mechanical performance of Al–Li alloys, paving a way in designing creep-aged Al–Li alloys with the superb mechanical properties.

Abstract Background In light of the aging population, increasingly suffering from the metabolic syndrome (MS), strategies need to be developed to address global public health challenges known to be associated with MS such as arthritis. As physical activity (PA) may play a crucial role in tackling those challenges, this study aimed to determine the association between the number of MS risk factors, PA and arthritis in people ≥ 50 years old. Methods Data from the Survey of Health, Ageing, and Retirement in Europe (SHARE) were used to estimate the prevalence of arthritis and MS risk factors in the European population ≥ 50 years and to evaluate the associations between MS risk factors, PA and arthritis. Binary logistic regression was performed to calculate the odds ratio of different factors. Results 73,125 participants were included in the analysis. 55.75% of patients stated at least one of the three MS risk factors. The prevalence of rheumatoid arthritis (RA) and osteoarthritis (OA)/other rheumatism among ≥ 50 years population was 10.19% and 19.32% respectively. Females showed a higher prevalence of arthritis than males. Prevalence did not differ between groups with different levels of PA. Arthritis prevalence was positively correlated with the number of MS risk factors (P 0.05). Conclusion Middle-aged and older Europeans with multiple comorbidities suffered from RA, OA or other rheumatism more frequently than participants with fewer comorbidities, while the level of physical activity was not associated with the number of metabolic risk factors in patients with RA and OA/other rheumatism.

Machine learning models have been increasingly popular in landslide susceptibility mapping based on the correlations among landslides and their inducing factors. However, mislabeled data in model training sets would deteriorate model accuracy. This study employed a Bayesian network to analyze influencing factors on landslides in Fujian Province, China, prone to typhoons and landslides. An inventory of 5,992 historical landslides informs Bayesian network modeling, with ten geoenvironmental factors as predictors. We introduced a progressive noise filtering method to mitigate the mislabeling effects of non-landslide points. The results show that altitude, wind speed, and lithology are the most important factors of landslides in the study area. The accuracy of the resultant landslide susceptibility map was verified using the area under the receiver operating characteristic curve (AUC) and Moran’s I index. The AUC value was improved from 0.838 to 0.931 during the progressive noise filtering. The correlation between historical landslide number density (LND) and resultant landslide susceptibility index (LSI) was evaluated. The Local Indicators of Spatial Association based on Moran’s I index shows consistent distribution patterns for high LND and high LSI regions. This study provides a useful reference for reliable landslide susceptibility mapping in the study area and similar areas.

Pasteurella multocida toxin (PMT) is one of the most important virulence factors of Pasteurella multocida type D. Pasteurella multocida infection has caused enormous economic losses in the pig farming industry. Although it is well known that this bacterial infection causes progressive atrophic rhinitis, its effects on other organ tissues in pigs are unclear. In this study, PMT was expressed and purified, and the cytotoxic effects of PMT on four types of swine cells, LLC-PK1, PAM, IPEC, and ST, were investigated. LLC-PK1 exhibited the highest sensitivity to the cytotoxic effects of PMT. Our studies revealed that a PMT concentration of 0.1 μg/kg can lead to weight loss, whereas a PMT concentration of 0.5 μg/kg can lead to death in mice. PMT causes damage to the intestines, kidneys, lungs, livers, and spleens of mice. Furthermore, PMT caused acute death in pigs at treatment concentrations greater than 5 μg/kg; at PMT concentration of 2.5 μg/kg, weight loss occurred until death. PMT mainly caused damage to the hearts, lungs, livers, spleens and kidneys of pigs. The organ coefficient showed that damage to the heart and kidneys was the most severe and caused the renal pelvis and renal pyramid to dissolve and become cavitated. Pathology revealed hemorrhage in the lungs, liver, and spleen, and the kidneys were swollen and vacuolated, which was consistent with the damaged target organs in the mice. In conclusion, these findings demonstrate that PMT is extremely toxic in vitro and in vivo, causing damage to various organs of the body, especially the kidneys and lungs. This study provides a theoretical basis for the in-depth exploration of the cytotoxic effects of PMT on target organs.

Abstract The continued miniaturization of chips demands highly thermally conductive materials and effective thermal management strategies. Particularly, the high‐field transport of the devices built with 2D materials is limited by self‐heating. Here a systematic control of heat flow in single‐side fluorinated graphene (FG) with varying degrees of fluorination is reported, revealing a superior room‐temperature thermal conductivity as high as 128 W m−1 K−1. Monolayer graphene/FG lateral heterostructures with seamless junctions are approached for device fabrication. Efficient in‐plane heat removal paths from graphene channel to side FG are created, contributing significant reduction of the channel peak temperature and improvement in the current‐carrying capability and power density. Molecular dynamics simulations indicate that the interfacial thermal conductance of the heterostructure is facilitated by the high degree of overlap in the phonon vibrational spectra. The findings offer novel design insights for efficient heat dissipation in micro‐ and nanoelectronic devices.

Summary: Adjustments in foliar photoprotective pigments are crucial for plant adaptation to harsh environments, serving as indicators of environmental stress. However, understanding when and where these adjustments occur across diverse biomes remains unclear due to challenges in large-scale observation. Here, we propose a novel approach to assess dynamics in photoprotective pigments at the canopy level using a new index derived from space-borne optical sensors. This approach generates a global map depicting the daily mean shortwave radiation threshold at which adjustments typically occur under prevailing climatic conditions. The global average of this threshold is 262 ± 50 W m⁻2, with lower values at high latitudes and peaks near 40° in both hemispheres. Temperature exerts a stronger influence on this latitudinal pattern than humidity. Future projections suggest a decrease in this threshold over northern high latitudes, implying exacerbated vulnerability under identical radiation levels due to negative warming responses. Based on this threshold, a high-stress zone around 60°N is identified and is predicted to shift southward in the future. These findings bridge critical gaps in photoprotection research and offer a new perspective on understanding the biogeochemical cycles of global ecosystems. This framework can also enhance our ability to predict the fate of diverse ecosystems under future climate.

In this study, a six-month pot experiment was conducted to explore the effects of nanoparticles (NPs), including CeO2, TiO2 and SiO2 NPs at 200 and 800 mg/kg, on the growth and quality of model medicinal plant Salvia miltiorrhiza. A control group was implemented without the application of NPs. Results showed that NPs had no significant effect on root biomass. Treatment with 200 mg/kg of SiO2 NPs significantly increased the total tanshinone content by 44.07 %, while 200 mg/kg of CeO2 NPs were conducive to a 22.34 % increase in salvianolic acid B content. Exposure to CeO2 NPs induced a substantial rise in the MDA content in leaves (176.25 % and 329.15 % under low and high concentration exposure, respectively), resulting in pronounced oxidative stress. However, TiO2 and SiO2 NPs did not evoke a robust response from the antioxidant system. Besides, high doses of CeO2 NP-amended soil led to reduced nitrogen, phosphorus and potassium contents. Furthermore, the NP amendment disturbed the carbon and nitrogen metabolism in the plant rhizosphere and reshaped the rhizosphere microbial community structure. The application of CeO2 and TiO2 NPs promoted the accumulation of metabolites with antioxidant functions, such as D-altrose, trehalose, arachidonic acid and ergosterol. NPs displayed a notable suppressive effect on pathogenic fungi (Fusarium and Gibberella) in the rhizosphere, while enriching beneficial taxa with disease resistance, heavy metal antagonism and plant growth promotion ability (Lysobacter, Streptomycetaceae, Bacillaceae and Hannaella). Correlation analysis indicated the involvement of rhizosphere microorganisms in plant adaptation to NP amendments. NPs regulate plant growth and quality by altering soil properties, rhizosphere microbial community structure, and influencing plant and rhizosphere microbe metabolism. These findings were beneficial to deepening the understanding of the mechanism by which NPs affect medicinal plants.

Salinity is an important abiotic factor in aquaculture because of its significance in the multiple physiological progress of marine fish. The liver is the main organ involved in energy metabolism, supplying energy to other body tissues and organs. Greater amberjack (Seriola dumerili) is characterized by fast growth and is an important aquatic export product in China. In this study, the greater amberjack was cultured in varying salinity levels, i.e., from 5 to 40 ppt, for 72 h. Notably, all fish under 5 ppt salinity died within 24 h because of limited tolerance ability. A comparative transcriptome analysis was performed to explore the molecular response mechanism of the liver, a non-osmoregulatory organ, when fish was subjected to hypo- (10 ppt) and hyper-salinity (40 ppt) stress. A total of 100 and 5 DEGs were identified under hypo- (10 ppt) and hyper-salinity (40 ppt) stress, respectively. The DEGs were involved in lipid metabolism, glycine, serine, and threonine metabolism, and in various signaling pathways, including Calcium, PPAR and adipocytokine, and adrenergic signaling in cardiomyocytes. The mRNA expression levels of acsl5, lipin1, and apoeb in the liver were down-regulated under 10 ppt salinity, suggesting an elevation of fatty acids oxidation and energy requirement in response to the hypo-salinity stress. Similarly, there was a decrease in the expression levels of atp1b1b and cftr, responsible for the reduced Na+, K+ movement and Cl- ion secretion, under hypo-salinity stress. This study provides important basic data for the optimal artificial breeding of greater amberjack, and reveals the osmotic regulation mechanism of the liver in response to salinity stress.

Background: Ras association domain family member 1 (RASSF1) encodes the RASSF1A protein, serving as a scaffold protein situated at the intersection of a complex signalling network. Aims: To evaluate the immunological and prognostic significance of RASSF1 expression in various types of human cancers, with a specific focus on lung cancer. Methods: Differential expression analysis of RASSF1 was conducted based on data from The Cancer Genome Atlas, Genotype-Tissue Expression, and Cancer Cell Line Encyclopaedia databases. Prognostic analysis was performed using the Cox regression test and Kaplan-Meier test. Spearman's test was utilized for correlation analysis. Gene Ontology (GO) and Kyoto Encyclopaedia of Genes and Genomes (KEGG) gene sets were employed to enrich the associated signaling pathways. Immunohistochemical staining and quantitative real-time PCR were employed to detect protein and mRNA expression levels, respectively. Results: RASSF1 expression was significantly lower in tumour tissues than in normal tissues in most cancers, and Cox regression analysis demonstrated a significant correlation between RASSF1 expression and the prognosis of over 12 types of cancer. Specifically, high RASSF1 expression was associated with poor OS in nine cancer types, including GBMLGG (HR = 4.98, P = 1.2e-31), LGG (HR = 3.72, P = 2.5e-10), and LAML (HR = 1.48, P = 2.4e-3). Further analysis showed that RASSF1 expression was significantly correlated with immune checkpoint- and immune-related genes. Moreover, RASSF1 expression is involved in tumour microenvironment (TME), RNA modification, genomic heterogeneity, and tumour stemness. GO and KEGG analyses showed that RASSF1 was closely related to tumour immune-related pathways. Finally, RASSF1A was moderately correlated with PD-L1 (R = 0.556), and RASSF1A overexpression significantly affected the expression of several genes involved in the Th17 cell differentiation signalling pathway in lung cancer. Conclusions: RASSF1 was differentially expressed in 29 human cancers and played a critical role in tumour immunity. Thus, RASSF1 has the potential to be used as a prognostic marker and reference for achieving more precise immunotherapy, particularly in lung cancer.

Heat stroke (HS) is a critical condition with extremely high mortality. Heat acclimation (HA) is widely recognized as the best measure to prevent and protect against HS. Preventive administration of oral rehydration salts III (ORSIII) and probiotics have been reported to sustain intestinal function in cases of HS. This study established a rat model of HA that was treated with probiotics-based ORS (ORSP) during consecutive 21-day HA training. The results showed that HA with ORSP could attenuate HS-induced hyperthermia by regulating thermoregulatory response. We also found that HA with ORSP could significantly alleviate HS-induced multiple organ injuries. The expression levels of a series of heat-shock proteins (HSPs), including HSP90, HSP70, HSP60, and HSP40, were significantly up-regulated from the HA training. The increases in intestinal fatty acid binding protein (I-FABP) and D-Lactate typically seen during HS were decreased through HA. The representative TJ proteins including ZO-1, E-cadherin, and JAM-1 were found to be significantly down-regulated by HS, but sustained following HA. The ultrastructure of TJ was examined by TEM, which confirmed its protective effect on the intestinal barrier protection following HA. We also demonstrated that HA raised the intestinal levels of beneficial bacteria Lactobacillus and lowered those of the harmful bacteria Streptococcus through 16S rRNA gene sequencing. These findings suggest that HA with ORSP was proven to improve intestinal thermotolerance and the levels of protective gut microbiota against HS.

Summary: Chemical warfare agents (CWAs), epitomized by the notoriously used mustard gas (HD), represent a class of exceptionally toxic chemicals whose airborne removal is paramount for battlefield safety. This study integrates high-throughput computational screening (HTCS) with advanced machine learning (ML) techniques to investigate the efficacy of metal-organic frameworks (MOFs) in adsorbing and capturing trace amounts of HD present in the air. Our approach commenced with a comprehensive univariate analysis, scrutinizing the impact of six distinct descriptors on the adsorption efficiency of MOFs. This analysis elucidated a pronounced correlation between MOF density and the Henry coefficient in the effective capture of HD. Then, four ML algorithms were employed to train and predict the performance of MOFs. The Random Forest (RF) algorithm demonstrates strong model learning and good generalization, achieving the best prediction result of 98.3%. In a novel exploratory stride, we incorporated a 166-bit MACCS molecular fingerprinting (MF) to identify critical functional groups within adsorbents. From the top 100 MOFs analyzed, 22 optimal functional groups were identified. Leveraging these insights, we designed three innovative substructures, grounded in these key functional groups, to enhance HD adsorption efficiency. In this work, the combination of MF and ML could provide a new direction for efficient screening of MOFs for the capture of HD in the air. The outcomes of this study offer substantial potential to revolutionize the domain of CWA capture. This represents a significant stride toward developing practical solutions that enhance both environmental protection and battlefield security.

Vision Transformer (ViT) models with strong performance are widely popular in the vision field. However, they are often limited by computation and model size, making the study of lightweight models critical. Existing lightweight ViT models often exhibit a weak dependence on high-frequency information and lack an effective multi-frequency fusion mechanism. This limitation typically necessitates the use of complex computational resources and the expansion of model scale to compensate for these shortcomings. In response to this situation, this paper proposes the DSCAFormer model, which develops a novel architecture that includes a local convolutional block with attention-like algorithms and a global spatial transformer block. These components use a channel splitting strategy with a gating mechanism to merge local and global information, forming a multi-frequency spatial feature extractor capable of effectively integrating both local and global information from images. Furthermore, a channel aggregation method is introduced to enhance the extraction of spatial information within the channel space, thereby enabling spatial feature perception within the context and the allocation of multi-feature computation. A series of comprehensive experiments were conducted in the domains of classification, detection, and instance segmentation, which demonstrated the DSCAFormer model’s remarkable scalability and competitiveness. For instance, on the ImageNet-1K dataset, the DSCAFormer model, which utilized 2.5M, 4.3M, and 7.4M parameters, achieved top-1 accuracies of 72.5%, 76.7%, and 79.5%, respectively, with 0.4 G, 0.7G, and 1.2G FLOPs.These results outperform the MobileViTv2-0.5/0.75/1.0 models, with respective accuracy improvements of 2.3%, 1.1%, and 1.4%, and reductions in FLOPs of 20%, 30%, and 33%. In addition, DSCAFormer has also shown competitive performance in downstream tasks.

We propose the graphene film with trapezoid-shaped nanoparticles (GTNAs) to transport particles. In our design, the conversion of plasmon surface resonances can be realized without changing the excitation light source. By sequentially activating three closely packed potential wells, nanoparticles can be transported between adjacent traps in a creeping manner. Three adjacent potential wells form a linearly repeating array structure, forming a nano-optical conveyor belt. When the resonant wavelength is 5.5 μm, and the power density is 0.4 mW/μm2, we verified that the target particle can move along the direction of the hot spots. In addition, the movement of nanoparticles in a liquid environment will be interfered with by viscous resistance and the random Brownian motion process. Since particles produce hysteresis or derailment during transmission, we also analyzed the time interval of switching the Fermi level to manipulate the particle in real-time. The three-dimensional finite-difference time-domain method has been used to verify that the design of this paper provides a conveyor belt in tunable graphene without rotating the polarization angle of the light source and has broad application prospects in biomedical diagnostics.

Person re-identification (ReID) has made significant progress in recent years, but Domain Generalization person re-identification (DG-ReID) remains a challenging task. Current DG-ReID methods update feature extractors and classifier parameters based on single-instance features. Consequently, these methods might not learn the common features between different instances of each identity, thus limiting its generalizability in the unseen target domain. To address this issue, we propose the Cross Intra-Identity Instance (CI3) learning framework based on Transformer. The CI3 framework leverages the mixed cross-attention (MCA) module and dynamic memory bank to extract random intra-identity instances during each iteration to learn additional information beyond the visual features between them. Furthermore, we introduce a task separation layer to eliminate the detrimental information in the backbone network. To bridge the domain gap between the source and target domains, we designed an online generalization feature constraint (OFC) strategy that utilizes constructed pseudo-label information to learn general cross-domain features. Our method effectively enhanced the performance of the DG-ReID model, achieving competitive results in extensive experiments on four benchmark datasets.

Aflatoxin B1 (AFB1) is a potent toxin in food, necessitating rapid, instant, and sensitive detection. We have engineered an electrochemical sensor to monitor AFB1 using a system composed of Fe3O4-NH4/AuNPs/apt-S1. The aptamer specifically recognizes AFB1, while ‘S1’ is functionalized with methylene blue to enhance the current. The RecJf exonuclease promotes the formation of the electrochemical strategy. The Fe3O4 component, with its magnet properties, enables a rapid separation of solids and liquids without the need for instrumentation. The sensor exhibits a linear range for AFB1 ranging from 1 ng to 10 μg. The regression equation is I(nA) = 446.8 × logc+2085 (where I and c represent the peak current and AFB1 concentration, respectively). The correlation coefficient is 0.9508, and the detection limit is 3.447 nM. The relative standard deviation of AFB1 in peanut oil ranges from 4.80% to 6.80%. These results demonstrate that the sensor has high sensitivity, stability, repeatability, and specificity for AFB1 detection.

Hereditary hemochromatosis (HH) is a disease characterized by disordered iron metabolism. It often involves mutations of the HFE gene, which encodes the homeostatic iron regulator protein (HFE), as well as mutations affecting hepcidin antimicrobial peptide, hemojuvelin, or transferrin receptor 2. Historically, HH has been observed primarily in European and European diaspora populations, while classical HH is rare in Asian populations, including in China. In this article, we report a rare case of HH in a Chinese man that could be attributed to a heterozygous C282Y/H63D HFE mutation. Based on clinical examination, liver biopsy, and genetic testing results, the patient was diagnosed with HH. Clinical signs and symptoms and serum iron-related test results were recorded for a period of two years after the patient began treatment. Over this observation period, the patient was subjected to 25 phlebotomies (accounting for a total blood loss of 10.2 L). His serum ferritin levels decreased from 1550 μg/L to 454 μg/L, his serum iron concentration decreased from 40 μmol/L to 24.6 μmol/L, and his transferrin saturation decreased from 97.5% to 55.1%. Early diagnosis is essential for patients with HH to obtain good outcomes. Regular phlebotomy after diagnosis can improve HH symptoms and delay HH disease progression.

With the ever-increasing role that nuclear power is playing to meet the aim of net zero carbon emissions, there is an intensified demand for understanding the thermal hydraulic phenomena at the heart of current and future reactor concepts. In response to this demand, the development of high-resolution flow analysis instrumentation is of increased importance. One such under-utilised and under-researched instrumentation technology, in the context of fluid flow analysis, is fibre Bragg grating (FBG)-based sensors. This technology allows for the construction of simple, minimally invasive instruments that are resistant to high temperatures, high pressures and corrosion, while being adaptable to measure a wide range of fluid properties, including temperature, pressure, refractive index, chemical concentration, flow rate and void fraction—even in opaque media. Furthermore, concertinaing FBG arrays have been developed capable of reconstructing 3D images of large phase structures, such as bubbles in slug flow, that interact with the array. Currently a significantly under-explored application, FBG-based instrumentation thus shows great potential for utilisation in experimental thermal hydraulics; expanding the available flow characterisation and imaging technologies. Therefore, this paper will present an overview of current FBG-based flow characterisation technologies, alongside a systematic review of how these techniques have been utilised in nuclear thermal hydraulics experiments. Finally, a discussion will be presented regarding how these techniques can be further developed and used in nuclear research.

Abstract Background Enucleation, a surgical procedure, is commonly used to treat large jaw cysts, unicystic ameloblastomas and keratocysts. However, it remains unclear to what extent the jaw bone regenerates after enucleation. We aimed to evaluate the percentage and the survival analysis of jaw bone regeneration, in terms of cavity volume residual (CVR), in patients who underwent enucleation of large jaw cysts, unicystic ameloblastomas and keratocysts. Methods We collected data longitudinally from 75 patients who underwent jaw cystic lesions enucleation at the Stomatological Hospital of Xi’an Jiaotong University, between January 2015 and June 2021. All patients had both preoperative and postoperative cone-beam computed tomography (CBCT) imaging data. CBCT images were analyzed using Image J. Changes in the CVR were assessed at various follow-up time points, and the Kaplan-Meier method was utilized to evaluate the CVR over time. Results The patients had a mean age of 31.7 years (range: 5.5–72 years) with 58.66% of them being male. The postoperative CVR was 32.20% at three months, 21.10% at six months, 15.90% at 12 months, and 5.60% at 24 months. The percentage of CVR during follow-up periods for the initial size Quartile (Q)1 (212.54-1569.60 mm3) was substantially lower than those of Q2 and Q3 at and after seven months of follow-up and became statistically significant at the 12-month mark. Conclusion This study demonstrates that spontaneous bone regeneration can occur after enucleation of large jaw cysts, unicystic ameloblastomas and keratocysts, even without the use of filler materials. The initial size of the lesion had a significant impact on the outcome of cystic lesion enucleation over time. To minimize the risks associated with radiation exposure and expenses, we recommend reducing the frequency of CT imaging follow-ups for patients with small initial cavity sizes (ranging from 212.54 to 1569.60 mm3).

This work demystifies the role that packaging boundary conditions (both physically and electromagnetically) can play in a nematic liquid crystal (NLC)-based inverted microstrip (IMS) phase shifter device operating at the 60 GHz band (from 54 GHz to 66 GHz). Most notably, the air box radiating boundary and perfect electric conductor (PEC) enclosing boundary are numerically examined and compared statistically for convergence, scattering parameters, and phase-shift-to-insertion-loss ratio, i.e., figure-of-merit (FoM). Notably, the simulated phase tunability of the radiating air box boundary structure is 8.26°/cm higher than that of the encased (enclosed) PEC boundary structure at 60 GHz. However, the maximum insertion loss of the encased PEC structure is 0.47 dB smaller compared to that of the radiant air box boundary structure. This results in an FoM increase of 29.26°/dB at the enclosed PEC limit (relative to the less-than-optimal airbox radiation limit). Arguably, the NLC-filled IMS phase shifter device packaging with metals fully enclosed (in addition to the default ground plane) enhances the symmetry of the structure, both in the geometry and the materials system. In electromagnetic parlance, it contributes to a more homogenously distributed electric field and a more stable monomodal transmission environment with mitigated radiation and noise. Practically, the addition of the enclosure to the well-established NLC-IMS planar fabrication techniques provides a feasible manufacturing (assembling) solution to acquire the reasonably comparable performance advantage exhibited by non-planar structures, e.g., a fully enclosed strip line and rectangular coaxial line, which are technically demanding to manufacture with NLC.

Background and aimLithium is considered to be the first-line treatment for bipolar disorder, and paliperidone was approved for the treatment of schizophrenia and acute bipolar manic/mixed episodes. However, both agents have been associated with thyroid dysfunction and cardiovascular adverse effects like subclinical hypothyroidism, bradycardia, and sinus arrest, even at therapeutic doses.Case presentationHere, we reported a case of a 17-year-old Han Chinese female who developed symptomatic hypothyroidism, sinus bradycardia, and sinus arrest while being treated with lithium and paliperidone for bipolar disorder with psychotic features including auditory hallucinations. Her workup suggested that these adverse effects might be related to the combined lithium and paliperidone treatment, although other causes could not be ruled out. After discontinuing both medications, her thyroid function and heart rhythm normalized over 20 days.ConclusionTo our knowledge, hypothyroidism, sinus bradycardia, and sinus arrest associated with the combined use of lithium and paliperidone had not been reported previously. Further research is warranted to elucidate the potential risks and benefits of this combination therapy for bipolar disorder with psychotic symptoms.

Abstract Continuous satellite measurements of the Earth's magnetic field have advanced the characterization of spatial‐temporal variations of the main field over the past two decades. To comprehend the underlying mechanism responsible for the geomagnetic field variations, we develop a novel core surface flow inversion scheme based on physics‐informed neural networks. The inversion method can account for the secular variation contributed by the interaction between the core flow and undetectable small‐scale magnetic fields. Based on the novel inversion framework, we derive a time‐dependent core surface flow model between 2000 and 2022 from the CHAOS‐7 core field model. The inverted core flow is then analyzed using the dynamic mode decomposition to extract wave‐like fluid motions. By calculating the magnetic secular acceleration contributed by each dynamic mode, we identify that the dynamic modes with period of about 10 and 7 years are responsible for geomagnetic jerks in the Atlantic and Pacific equatorial regions.

ABSTRACT The failure of COVID-19 vaccines to prevent SARS-CoV-2 infection and transmission, a possibly critical reason was the lack of protective mucosal immunity in the respiratory tract. Here, we evaluated the effects of mucosal and systemic immunity from a novel simian adenovirus-vectored COVID-19 vaccine (Sad23L-nCoV-S) in mice in comparison with Ad5-nCoV-S by intranasal (IN) drip and intramuscular (IM) injection vaccinations. As good as the well-known Ad5-nCoV-S vaccine, a single-dose IN inoculation of 1 × 109 PFU Sad23L-nCoV-S vaccine induced a similar level of IgG S-binding antibody (S-BAb) and neutralizing antibody (NAb) and higher IgA in serum, while IN route raised significantly higher IgG and IgA S-BAb and NAb in bronchoalveolar lavage (BAL), and specific IFN-γ secreting T-cell response in lung compared with IM route, but lower T-cell response in spleen. By prime-boost vaccination regimens with different combinations of IN and IM inoculations of Sad23L-nCoV-S vaccine, the IN-involved vaccination stimulated higher protective mucosal or local immunity in BAL and lung, while the IM-involved immunization induced higher systemic immunity in serum and spleen. A long-term sustained mucosal and systemic NAb and T- cell immunity to SARS-CoV-2 was maintained at high level over 32 weeks by prime-boost vaccination regimens with IN and IM routes. In conclusion, priming or boosting immunization with IN inoculation of Sad23L-nCoV-S vaccine could induce effective mucosal immunity and in combination of IM route could additionally achieve systemic immunity, which provided an important reference for vaccination regimens against respiratory virus infection. IMPORTANCE The essential goal of vaccination is to generate potent and long-term protection against diseases. Several factors including vaccine vector, delivery route, and boosting regimen influence the outcome of prime-boost immunization approaches. The immunization regimens by constructing a novel simian adenovirus-vectored COVID-19 vaccine and employing combination of intranasal and intramuscular inoculations could elicit mucosal neutralizing antibodies against five mutant strains in the respiratory tract and strong systemic immunity. Immune protection could last for more than 32 weeks. Vectored vaccine construction and immunization regimens have positively impacted respiratory disease prevention.

Abstract The effect of temperature on the dynamic parameters of silty clay in a seasonally frozen region was assessed using a GDS dynamic triaxial test system. The strength parameters, dynamic elastic modulus, damping ratio, and other dynamic parameters of the soil samples were analyzed under different temperature conditions. The results demonstrated that the shear strength parameters (internal friction angle and cohesion) of the silty clay under a dynamic load increased significantly with decreasing temperature, and the internal friction angle increased sharply below 0 °C. The dynamic elastic modulus increased as the temperature decreased and changed significantly in the ice-water phase change region. The slope of the dynamic stress–strain curve of the soil sample increased significantly with decreasing temperature. As the temperature decreased, the damping ratio reduced, and the ability of the soil to absorb seismic waves declines. The research results provide new data and information to guide construction projects in seasonally frozen region.

Abstract Background The objective of this study was to examine the correlation between serum Klotho protein concentration and postmenopausal hypertension. Methods A cross-sectional study design was used, in which 1713 postmenopausal women who participated in the National Health and Nutrition Examination Survey (NHANES) 2013–2016 were included. Multivariate logistic regression models were applied to assess the association between serum Klotho concentration and postmenopausal hypertension. Results A weighted analysis was executed, revealing a noteworthy hypertension prevalence rate of 53.44% among the study participants. Participants with lower quartile of serum Klotho concentration had a higher prevalence of hypertension than those in higher quartiles (Q1:62.29% vs. Q2: 48.52% vs. Q3: 47.33% vs. Q4: 55.02%, p

Abstract There are few studies focus on post-neoadjuvant treatment in hormone receptor-positive (HR+)/human epidermal growth factor receptor 2-negative (HER2-)/lymph node-positive (LN+) breast cancer, a multi-center, open-label, randomized, controlled phase III trial was conducted to evaluate pathological response-guided non-cross-resistant adjuvant chemotherapy in patients with HR+/HER2-/LN+ breast cancer who were non-responsive to primary chemotherapy. Patients received four cycles of non-cross-resistant adjuvant chemotherapy plus endocrine therapy (ET), or ET alone. Forty patients responsive to neoadjuvant chemotherapy and with Miller and Payne G4 or G5 and LN- status were assigned to the observation group. Distant disease-free survival was the primary endpoint. The final intention-to-treat analysis comprised 379 patients. After a median follow-up period of 72.4 months, the 5-year distant disease-free survival was 92% and 90% in the chemotherapy plus ET and ET-alone groups, respectively. Comparatively, the observation group showed a trend towards better distant disease-free survival. For patients non-responsive to neoadjuvant chemotherapy, adjuvant non-cross-resistant chemotherapy did not significantly improve distant disease-free survival compared to ET alone.

Detection of nitro pollutants is an important topic in environmental protection. A total of 3 Cd (II) complexes (1–3) based on 3 soft organic isomers, n-(3,5-dicarboxylato benzyloxy) benzoic acid (n = 2, 3 or 4-H3DBB), and a linear N-donor ligand, 3-bis(imidazole-l-ylmethyl) benzene (3-bibz), have been synthesized hydrothermally. Structural diversity of Complexes 1–3 displays the architectural 2D or 3D change: Complex 1 exhibits a 2D network featuring tri-nuclear metal units, Complex 2 is a 3D framework based on similar tri-nuclear metal units, and Complex 3 shows a 3D network with binuclear units. Fluorescent sensing properties exhibited in all these complexes have been discovered to detect nitrobenzene (NB) selectively and sensitively. In particular, Complex 3 possesses high sensitivity for NB with the lowest detection limit of 1.15 × 10−10 M. The results of the theoretical calculation verified the fluorescence detection mechanism of NB by these Cd-based complexes. Therefore, these Cd-based complexes might be used as excellent luminescent sensors for NB.

Pasteurella multocida, a zoonotic pathogen that produces a 146-kDa modular toxin (PMT), causes progressive atrophic rhinitis with severe turbinate bone degradation in pigs. However, its mechanism of cytotoxicity remains unclear. In this study, we expressed PMT, purified it in a prokaryotic expression system, and found that it killed PK15 cells. The host factor CXCL8 was significantly upregulated among the differentially expressed genes in a transcriptome sequencing analysis and qPCR verification. We constructed a CXCL8-knockout cell line with a CRISPR/Cas9 system and found that CXCL8 knockout significantly increased resistance to PMT-induced cell apoptosis. CXCL8 knockout impaired the cleavage efficiency of apoptosis-related proteins, including Caspase3, Caspase8, and PARP1, as demonstrated with Western blot. In conclusion, these findings establish that CXCL8 facilitates PMT-induced PK15 cell death, which involves apoptotic pathways; this observation documents that CXCL8 plays a key role in PMT-induced PK15 cell death.

Common rust (CR), caused by Puccina sorghi, is a major foliar disease in maize that leads to quality deterioration and yield losses. To dissect the genetic architecture of CR resistance in maize, this study utilized the susceptible temperate inbred line Ye107 as the male parent crossed with three resistant tropical maize inbred lines (CML312, D39, and Y32) to generate 627 F7 recombinant inbred lines (RILs), with the aim of identifying maize disease-resistant loci and candidate genes for common rust. Phenotypic data showed good segregation between resistance and susceptibility, with varying degrees of resistance observed across different subpopulations. Significant genotype effects and genotype × environment interactions were observed, with heritability ranging from 85.7% to 92.2%. Linkage and genome-wide association analyses across the three environments identified 20 QTLs and 62 significant SNPs. Among these, seven major QTLs explained 66% of the phenotypic variance. Comparison with six SNPs repeatedly identified across different environments revealed overlap between qRUST3-3 and Snp-203,116,453, and Snp-204,202,469. Haplotype analysis indicated two different haplotypes for CR resistance for both the SNPs. Based on LD decay plots, three co-located candidate genes, Zm00001d043536, Zm00001d043566, and Zm00001d043569, were identified within 20 kb upstream and downstream of these two SNPs. Zm00001d043536 regulates hormone regulation, Zm00001d043566 controls stomatal opening and closure, related to trichome, and Zm00001d043569 is associated with plant disease immune responses. Additionally, we performed candidate gene screening for five additional SNPs that were repeatedly detected across different environments, resulting in the identification of five candidate genes. These findings contribute to the development of genetic resources for common rust resistance in maize breeding programs.

This work proposes and examines the feasibility of next-generation 0.3 THz phase shifters realized with liquid crystals (LCs) as tunable dielectrics coaxially filled in the transmission line. The classic coaxial transmission line topology is robust to electromagnetic interference and environmental noise, but is susceptible to higher-order modes from microwave to millimeter-wave towards terahertz (THz) wavelength ranges, which impedes the low-insertion-loss phase-shifting functionality. This work thus focuses primarily on the suppression of the risky higher-order modes, particularly the first emerging TE11 mode impacting the dielectric loss and metal losses in diverse manners. Based on impedance matching baselines at diverse tuning states of LCs, this work analytically derives and models two design geometries; i.e., design 1 for the coaxial geometry matched at the isotopically referenced state of LC for 50 Ω, and design 2 for geometry matched at the saturated bias of LC with the maximally achievable permittivity. The Figure-of-Merit for design 1 and design 2 reports as 35.15°/dB and 34.73°/dB per unit length, respectively. We also propose a constitutive power analysis method for understanding the loss consumed by constitutive materials. Notably, for the 0.3 THz design, the isotropic LC state results in an LC dielectric loss of 63.5% of the total input power (assuming 100%), which becomes the primary constraint on achieving low-loss THz operations. The substantial difference in the LC dielectric loss between the isotropic LC state and saturated bias state for the 0.3 THz design (35.76% variation) as compared to that of our past 60 GHz design (13.5% variation) indicates that the LC dielectric loss’s escalating role is further enhanced with the rise in frequency, which is more pronounced than the conductor losses. Overall, the results from analytical and finite-element optimization in this work shape the direction and feasibility of the unconventional THz coaxial phase shifting technology with LCs, actioned as continuously tunable dielectrics.

In satellite remote sensing (SRS), there is a demand for large-power microwave components. A Butler matrix is essential to a transmitting antenna array in SRS. This article illustrates the electrical and mechanical design, simulation, and test results of a large-power planar beamforming network for SRS at C-band. It is a 4 × 4 Butler matrix based on square coaxial lines. Short-ended stubs are used in the Butler matrix to broaden its bandwidth by 10%, support inner conductors, and enhance heat transfer in vacuum environments. The simulation results are consistent with the measured results. The reflection coefficient is less than −18 dB, and the isolation is more than 23 dB from 3.8 GHz to 4.2 GHz. The insertion losses are less than 0.6 dB, and the phase errors are better than ±6°. The measured peak microwave power of the proposed Butler matrix is 9 kW. Its size is 440 × 400 × 40 mm3. The proposed Butler matrix beamforming network can be applied to SRS systems.

Earth’s inner core is heterogeneous and anisotropic. A new study based on computational simulation reveals the presence of ionic hydrogen flux in iron crystals, driven by the dipole geomagnetic field, which promotes the formation of observed inner core structure.

Renal ischemia-reperfusion injury (IRI) is a non-negligible clinical challenge for clinicians in surgeries such as renal transplantation. Functional loss of renal tubular epithelial cell (TEC) in IRI leads to the development of acute kidney injury, delayed graft function (DGF), and allograft rejection. The available evidence indicates that cellular oxidative stress, cell death, microvascular dysfunction, and immune response play an important role in the pathogenesis of IRI. A variety of immune cells, including macrophages and T cells, are actively involved in the progression of IRI in the immune response. The role of B cells in IRI has been relatively less studied, but there is a growing body of evidence for the involvement of B cells, which involve in the development of IRI through innate immune responses, adaptive immune responses, and negative immune regulation. Therefore, therapies targeting B cells may be a potential direction to mitigate IRI. In this review, we summarize the current state of research on the role of B cells in IRI, explore the potential effects of different B cell subsets in the pathogenesis of IRI, and discuss possible targets of B cells for therapeutic aim in renal IRI.

Purpose: This study aimed to screen biomarkers to predict the efficacy of programmed cell death 1 (PD-1) blockade immunotherapy combined with chemotherapy as neoadjuvant therapy for esophageal squamous cell carcinoma (ESCC). Methods: In the first stage of the study, the baseline concentrations of 40 tumor-related chemokines in the serum samples of 50 patients were measured to screen for possible biomarkers. We investigated whether the baseline concentration of the selected chemokine was related to the therapeutic outcomes and tumor microenvironment states of patients treated with the therapy. In the second stage, the reliability of the selected biomarkers was retested in 34 patients. Results: The baseline concentration of macrophage migration inhibitory factor (MIF) was negatively correlated with disease-free survival (DFS) and overall survival (OS) in patients treated with the therapy. In addition, a low baseline expression level of MIF is related to a better tumor microenvironment for the treatment of ESCC. A secondary finding was that effective treatment decreased the serum concentration of MIF. Conclusion: Baseline MIF levels were negatively correlated with neoadjuvant therapy efficacy. Thus, MIF may serve as a predictive biomarker for this therapy. The accuracy of the prediction could be improved if the serum concentration of MIF is measured again after the patient received several weeks of treatment.

Abstract To determine the current evidence on artificial neural network (ANN) in prognostic studies of musculoskeletal diseases (MSD) and to assess the accuracy of ANN in predicting the prognosis of patients with MSD. The scoping review was reported under the Preferred Items for Systematic Reviews and the Meta-Analyses extension for Scope Reviews (PRISMA-ScR). Cochrane Library, Embase, Pubmed, and Web of science core collection were searched from inception to January 2023. Studies were eligible if they used ANN to make predictions about MSD prognosis. Variables, model prediction accuracy, and disease type used in the ANN model were extracted and charted, then presented as a table along with narrative synthesis. Eighteen Studies were included in this scoping review, with 16 different types of musculoskeletal diseases. The accuracy of the ANN model predictions ranged from 0.542 to 0.947. ANN models were more accurate compared to traditional logistic regression models. This scoping review suggests that ANN can predict the prognosis of musculoskeletal diseases, which has the potential to be applied to different types of MSD.

In industrial areas where anomalous samples are lacking, unsupervised methods with high accuracy are especially important to ensure product quality and stability. An unsupervised method based on a feature extractor and a distribution estimation module has been applied in the industrial field and has achieved good performance. Flow models are usually used as distribution estimation modules. But traditional flow models only focus on feature information in two dimensions, the channel dimension, and the space dimension, while ignoring the cross-dimensional connection between them. To solve this problem, we proposed a flow model with a global attention mechanism for anomaly detection of images. It embeds combined modules of global attention mechanism and convolutional layer in the structure of a reversible neural network, which is capable of global cross-dimensional extraction of image features. In the comparison experiments, our method achieves average image and pixel-level AUC of 0.997 and 0.987 on the MVTec AD dataset and 0.968 and 0.984 on the BTAD dataset, respectively, which outperforms the other traditional methods. In addition, in the detection task, our method also possesses faster inference speeds. This shows that our method has excellent anomaly detection and localization performance, which meets the industrial demand for high-precision anomaly detection and localization.

Abstract Background Lotus (Nelumbo Adans.) is used as an herbal medicine and the flowers are a source of natural flavonoids. ‘Da Sajin’, which was firstly found in the plateau area, is a natural mutant in flower color with red streamers dyeing around white petals. Results The LC–MS-MS results showed that eight anthocyanin compounds, including cyanidin 3-O-glucoside, cyanidin 3-O-galactoside, malvidin 3-O-galactoside, and malvidin 3-O-glucoside, were differentially enriched in red-pigmented tissues of the petals, whereas most of these metabolites were undetected in white tissues of the petals. Transcriptome profiling indicated that the relative high expression levels of structural genes, such as NnPAL, NnF3H, and NnANS, was inconsistent with the low anthocyanin concentration in white tissues. Members of the NnMYB and NnbHLH transcription factor families were presumed to play a role in the metabolic flux in the anthocyanin and proanthocyanidin biosynthetic pathway. The expression model of translational initiation factor, ribosomal proteins and SKP1–CUL1–F-box protein complex related genes suggested an important role for translational and post-translational network in anthocyanin biosynthesis. In addition, pathway analysis indicated that light reaction or photo destruction might be an important external cause for floral color determination in lotus. Conclusions In this study, it is supposed that the natural lotus mutant ‘Da Sajin’ may have originated from a red-flowered ancestor. Partial loss of anthocyanin pigments in petals may result from metabolic disorder caused by light destruction. This disorder is mainly regulated at post translation and translation level, resulting in a non-inherited phenotype. These results contribute to an improved understanding of anthocyanin metabolism in lotus, and indicate that the translational and post-translational regulatory network determines the metabolic flux of anthocyanins and proanthocyanidins under specific environmental conditions.

Abstract In this study, we numerically simulate the evolution of the orbital angular momentum (OAM) spectrum of a vortex laser beam in the optical parametric chirped pulse amplification (OPCPA) process, which is an effective technical method to realize ultra-intense and ultra-short vortex laser amplification. The results show that the proportion of the vortex laser beam with 100% topological charge (TC) of 1 decreases to 97.44% with the enhancement of the saturation amplification after amplification by a 15 mm length LBO pumped by a 526.5 nm laser with a pump intensity of 1.74 GW/cm2. Conversely, the beams with other topological charges generate and increase with the amplification. The simulation results are consistent with our previous experimental results. Meanwhile, compared with non-collinear OPCPA, collinear OPCPA can maintain well the proportion of TC $$l = 1$$ l = 1 .

Earth’s magnetic field is generated in the fluid outer core through the dynamo process. Over the last decade, data assimilation has been used to retrieve the core dynamics and predict the evolution of the geomagnetic field. The presence of model errors in the geomagnetic data assimilation is inevitable because current numerical geodynamo models are still far from realistic core dynamics. In this paper, we investigate the effect of model errors in geomagnetic data assimilation based on ensemble Kalman filter (EnKF). We construct two dynamo models with different control parameters but exhibiting similar force balance and magnetic morphology at the core surface. We then use one dynamo model to generate synthetic observations and the other as the forward model in EnKF. Our test experiments show that the EnKF approach with the pre-setting model errors can nevertheless recover large-scale core surface flow and make a rough short-term (5-year) prediction. However, the data assimilation in the presence of model errors cannot keep improving the core state even though new observations are available. Motivated by the planned Macau Science Satellite-1, which is expected to provide improved internal geomagnetic field model, we also perform a test experiment using synthetic observations up to spherical harmonic degree \begin{document}$\ell=18$\end{document}. Our results indicate that high-resolution observations are crucial in reconstructing small scale flow.

Abstract Background A high-risk prevention strategy is an effective way to fight against human immunodeficiency virus (HIV) and acquired immunodeficiency syndrome (AIDS). The China AIDS Fund for Non-Governmental Organizations (CAFNGO) was established in 2015 to help social organizations intervene to protect high-risk populations in 176 cities. This study aimed to evaluate the role of social organizations in high-risk population interventions against HIV/AIDS. Methods This study was based on the CAFNGO program from 2016 to 2020. The collected data included the number and types of social organizations participating in high-risk group interventions and the amount of funds obtained by these organizations each year. We explored the factors influencing the number of newly diagnosed AIDS cases using a spatial econometric model. Furthermore, we evaluated the effectiveness of intervention activities by comparing the percentages of the individuals who initially tested positive, and the individuals who took the confirmatory test, as well as those who retested positive and underwent the treatment. Results Overall, from 2016 to 2020, the number of social organizations involved in interventions to protect HIV/AIDS high-risk populations increased from 441 to 532, and the invested fund increased from $3.98 to $10.58 million. The number of newly diagnosed cases decreased from 9128 to 8546 during the same period. Although the number of cities with overall spatial correlations decreased, the spatial agglomeration effect persisted in the large cities. City-wise, the number of social organizations (direct effect 19.13), the permanent resident population (direct effect 0.12), GDP per capita (direct effect 17.58; indirect effect − 15.38), and passenger turnover volume (direct effect 5.50; indirect effect − 8.64) were the major factors influencing new positive cases confirmed through the testing interventions performed by the social organizations. The initial positive test rates among high-risk populations were below 5.5%, the retesting rates among those who initially tested positive were above 60%, and the treatment rates among diagnosed cases were above 70%. Conclusions The spatial effect of social organizations participating in interventions targeting high-risk populations funded by CAFNGO is statistically significant. Nevertheless, despite the achievements of these social organizations in tracking new cases and encouraging treatment, a series of measures should be taken to further optimize the use of CAFNGO. Working data should be updated from social organizations to CAFNGO more frequently by establishing a data monitoring system to help better track newly diagnosed AIDS cases. Multichannel financing should be expanded as well.

Antibody-mediated rejection (AMR) has a strongly negative impact on long-term renal allograft survival. Currently, no recognized effective treatments are available, especially for chronic antibody-mediated rejection (CAMR). Donor-specific antibodies (DSAs) secreted by long-lived plasma cells and memory B cells are acknowledged as biomarkers of AMR. Nevertheless, it may be too late for the DSA routine examination production since DSAs may have binded to graft vascular endothelial cells through complement-dependent or complement-independent pathways. Therefore, methods to effectively monitor memory B cells and long-lived plasma cells and subsequently prevent DSA production are key to reducing the adverse effects of AMR. Therefore, this review mainly summarizes the production pathways of memory B cells and long-lived plasma cells and provides suggestions for the prevention of AMR after transplantation.
