Various bioactive coatings are created to improve the biocompatibility of substrates used for bone regeneration, which includes formulation of thin zirconia coatings with pro-osteogenic properties. The aim of this study was to assess the biological properties of ZrO2 thin films grown by Atomic Layer Deposition (ALD) technology (ZrO2ALD).Methodology: The cytocompatibility of the obtained layers was analysed using the mice pre-osteoblastic cell line (MC3T3) characterized by decreased expression of microRNA 21-5p (miR-21-5p) in order to evaluate the potential pro-osteogenic properties of the coatings. The in vitro experiments were designed to determine the effect of ZrO2ALD coatings on cell morphology (confocal microscope), proliferative activity (cell cycle analysis) and metabolism, reflected by mitochondrial membrane potential (cytometric-based measurement). Additionally, the influence of layers on the expression of genes associated with cell survival and osteogenesis was studied using RT-qPCR. The following genes were investigated: B-cell lymphoma 2 (Bcl-2), Bcl-2-associated X protein (Bax), p53 and p21, as well as osteogenic markers, i.e. collagen type 1 (Coll-1), osteopontin (Opn), osteocalcin (Ocl) and runt-related transcription factor 2 (Runx-2). The levels of microRNA (miRNA/miR) involved in the regulation of osteogenic genes were determined, including miR-7, miR-21, miR-124 and miR-223.Results: The analysis revealed that the obtained coatings are cytocompatible and may increase the metabolism of pre-osteoblast, which was correlated with increased mitochondrial membrane potential and extensive development of the mitochondrial network. The obtained coatings affected the viability and proliferative status of cells, reducing the population of actively dividing cells. However, in cultures propagated on ZrO2ALD coatings, the up-regulation of genes essential for bone metabolism was noted.Discussion: The data obtained indicate that ZrO2 coatings created using the ALD method may have pro-osteogenic properties and may improve the metabolism of bone precursor cells. Abstract

Structural and magnetic properties of GaAs thin films with embedded MnAs nanoclusters were investigated as function of the annealing temperature and layers composition. The presence of two kinds of nanoclusters with different dimensions and structure were detected. The fraction of Mn atoms in each kind of cluster was estimated from the extended X-ray absorption fine structure analysis. This analysis ruled out the possibility of the existence of nanoclusters containing a hypothetic MnAs cubic compound - only (Mn,Ga)As cubic clusters were detected. Change of the layer strain from the compressive to tensile was related to the fraction of zinc blende and hexagonal inclusions. Thus the zinc blende inclusions introduce much larger strain than hexagonal ones. The explanation of observed thermal induced strain changes of the layers from the compressive to tensile is proposed. The magnetic properties of the samples were consistent with structural study results. Their showed that in sample containing solely cubic (Mn,Ga)As inclusions Mn ions inside the inclusions are still ferromagnetically coupled, even at room temperature. This fact can be explained by existence in these clusters of GaMnAs solid solution with content of Mn higher than 15 % as was found in theoretical calculations., Comment: 12 pages, 16 figures, 4 tables

Abstract

Topical Area: Nutrient-Gene InteractionsTopical Area: Nutrient-Gene Interactions; Objectives : We have previously shown that 12 months (mo.) high-fat diet (HFD) in pigs led to pathophysiological alterations, incl. fattening and increased femoral artery intima-media-thickness, which were partly reversed after 3 mo. return to control diet (Zabek et al., PLoS One 2017). The aim of this study was to decipher underlying mechanism of action of these dietary interventions on the arteries by nutrigenomics analyses of intima and media of aorta.Methods : 32 female pigs were divided into 3 groups: Control diet (CD) for 12 mo.; HFD for 12 mo.; 3) Reversal diet group (RD): HFD for 9 mo. followed by CD for 3 mo. After 12 mo. animals were killed and abdominal aorta collected. RNA was isolated from aorta intima and media for whole genome microarray analyses followed by bioinformatics analyses.Results :HFD compared to CD group significantly affected gene expression profile in intima with genes belonging to the chemotaxis, inflammation or endothelial permeability. RD induced gene expression profile was distinct from the CD group. This suggests that 3 mo. of reversal to CD is not sufficient to correct gene expression changes induced by HFD. Comparison of RD profile with that of HFD group revealed a group of genes with opposite expression, e.g. genes regulating inflammation, toll-like cell signaling pathway or cytoskeleton organization involved in the regulation of cell permeability. This suggests that return to the RD only partly restored gene expression alterations due to the HFD. Significant changes in expression of genes in media following HFD were also observed, such as genes involved in cytoskeleton organization and migration MAPK signaling. As for intima, the expression profile of media of pigs on RD was different on that of these on CD diet. Compared to HFD, a group of genes involved in PI3K or MAPK pathways presented opposite expression suggesting that RD can partly correct the changes in genomic effect induced by HFD.Conclusions : This study revealed genomic modifications induced by long-term HFD consumption on arterial intima and media. The return to normal diet for 3 mo. was not sufficient to counteract the genomic effect of long-term HFD consumption.Funding Sources : WROVASC Integrated Cardiovascular Centre, co-financed by the European Regional Development Fund.

The European Synchrotron and FEL User Organisation (ESUO) represents about 22 000 users of the European synchrotron radiation (SR) and Free-Electron Laser (FEL) facilities, which offer various kinds of experiments ranging from physics over life sciences up to cultural heritage. Established in 2010, ESUO is composed of national delegates from 30 European member states and associated countries and is headed by an executive board of eight members. Our vision is to support a thriving (European) synchrotron and FEL user community with equal opportunities of access and participation for all scientists, based solely on the scientific merit of their ideas.
