Abstract Background Antigen testing using lateral flow devices (LFDs) plays an important role in the management of the novel coronavirus pandemic of 2019 (COVID-19) by rapidly identifying individuals who are asymptomatically carrying high levels of the virus. By January 2021, LFD community testing sites were set up across English local authority areas to support the management and containment of regional COVID-19 cases, initially targeting essential workers unable to work from home during the national lockdown. This study aimed to examine the characteristics and motivations of individuals accessing community LFD testing across two local authority areas (LAAs) in the South West of England. Methods Data were collected as part of a service evaluation from December 22nd 2020 until March 15th 2021 for two LAAs. Demographic and postcode data were collected from an online test appointment booking platform and the National Health Service testing service online system, with data accessed from Public Health England. An online survey was sent to individuals who made a testing appointment at an LAA1 site using the online booking platform, consisting of 12 questions to collect data on individual’s motivations for and experiences of testing. Results Data were available for individuals who completed 12,516 tests in LAA1 and 12,327 tests in LAA2. Most individuals who engaged with testing were female, working age, white, and worked as early years or education staff, health and social care staff, and supermarket or food production staff. 1249 individuals completed the survey with 60% of respondents reported getting tested for work-related reasons. Individuals first heard about LFD testing through various channels including work, media, and word of mouth, and decided to get tested based on the ease and convenience of testing, workplace communications, and to identify asymptomatic cases to help stop the spread. Most tests were completed by individuals living in less deprived areas based on national deciles of deprivation. Conclusions While national and local COVID-19 testing strategies have evolved, community and personal LFD testing remains a crucial pillar of the testing strategy. Future studies should collect quantitative and qualitative data from residents to most effectively shape testing offers based on the needs and preferences of their population.

This entry establishes the contested nature of ‘queer’ as a term and provides an overview of some significant theoretical perspectives on queerness. It then outlines the emergence of queer Christian theology as it developed from both theological reflections on LGBT+ sexuality and gender variance and a number of other theoretical traditions. In doing so, it highlights how contemporary queer theology can be grouped into two clusters: those queer theologians who are committed to advancing LGBT+ inclusion within Christian spaces and communities, and those queer theologians who are more sceptical of such a project. This is followed by an overview of the different approaches that queer theologians have taken in negotiating biblical passages that have historically been used to condemn LGBT+ identities and gender nonconformity, using 1 Cor 6:9–10 as an exemplar. Queer engagement with scripture has a much broader scope than just these passages, and this entry provides an illustrative overview of some of the more creative directions of queer biblical scholarship. Moving from scripture to doctrine, this entry uses the differing queer perspectives on Christology to highlight how the project of queering Christian doctrine is diverse and wide-ranging. This entry then turns towards the practical implications of queer theology for Christian communities. LGBT+ marriage is foregrounded as the most prominent avenue through which queer theology has been felt in the everyday practice of Christians, before highlighting some of the different dimensions of queer Christian practice that do not relate to marriage. This entry concludes by describing the contributions queer theology has made to ongoing debates within queer studies and exploring some of the key issues surrounding the contested relationship queer theology has with the theological mainstream.

This journal review article focuses on the use of assistive and rehabilitative exoskeletons as a new opportunity for individuals with diminished mobility. The article aims to identify gaps and inconsistencies in state-of-the-art assistive and rehabilitative devices, with the overall goal of promoting innovation and improvement in this field. The literature review explores the mechanisms, actuators, and sensing procedures employed in each application, specifically focusing on passive shoulder supports and active soft robotic actuator gloves. Passive shoulder supports are an excellent option for bearing heavy loads, as they enable the load to be evenly distributed across the shoulder joint. This, in turn, reduces stress and strain around the surrounding muscles. On the other hand, the active soft robotic actuator glove is well suited for providing support and assistance by mimicking the characteristics of human muscle. This review reveals that these devices improve the overall standard of living for those who experience various impairments but also encounter limitations requiring redress. Overall, this article serves as a valuable resource for individuals working in the field of assistive and rehabilitative exoskeletons, providing insight into the state of the art and potential areas for improvement.

The use of telepathology for clinical applications in Canada has steadily become more attractive over the last 10 years, driven largely by its potential to provide rapid pathology consulting services throughout the country regardless of the location of a particular institution. Based on this trend, the president of the Canadian Association of Pathologists asked a working group consisting of pathologists, technologists, and healthcare administrators from across Canada to oversee the development of guidelines to provide Canadian pathologists with basic information on how to implement and use this technology. The guidelines were systematically developed, based on available medical literature and the clinical experience of early adopters of telepathology in Canada. While there are many different modalities and applications of telepathology, this document focuses specifically on whole-slide imaging as applied to intraoperative pathology consultation (frozen section), primary diagnosis, expert or second opinions and quality assurance activities. Applications such as hematopathology, microbiology, tumour boards, education, research and technical and/or standard-related issues are not covered.

Most protected area impact research that uses counterfactuals draws heavily on quantitative methods, data, and knowledge types, making it valuable in producing generalizations but limited in temporal scope, historical detail, and habitat diversity and coverage of ecosystem services. We devised a methodological pluralistic approach, which supports social science qualitative methods, narratives, mixed methods, and interdisciplinarity, to fully unlock the potential of counterfactuals in ensuring a place-based and detailed understanding of the socioecological context and impacts of protected areas. We applied this approach to derive possible counterfactual conditions for the impact of a montane protected area on 40 years of vegetation change in the Cape Floristic Region-a global biodiversity hotspot and UNESCO World Heritage Site in South Africa. We incorporated diverse methods, knowledge, and information sources, drawing on before-after protected area comparisons for inside and outside the protected area. A significant increase in shrubland vegetation (17-30%) was observed and attributed primarily to a decline in frequent burning for grazing. This also occurred outside the protected area and was driven by socioeconomic drivers and not by concerns over biodiversity conservation or land degradation. Had the protected area not been established the area would have seen intensification of cultivation and increased road networks, buildings, and water storage in dams. Our approach increased historical temporal coverage of socioecological change and contextualized assumptions around causality. Protected area impact evaluation should reengage in place-based research that fully incorporates pluralism in methodologies for constructing counterfactuals in a way that builds regional and global understanding from the local level upward. We devised 10 key principles for deriving counterfactuals grounded in methodological pluralism, covering aspects of collaboration, cocreation, inter- and transdisciplinarity, diverse values and lived experiences, multiple knowledge types, multiple possible causal mechanisms, social science qualitative methods, perceptions, perspectives, and narratives., (© 2024 The Authors. Around the world today, universities are expected to play a unique role as creators of regional growth and innovation. While there appears to be a consensus that the role of universities has been expanded, critiques show that the contribution of universities to their regions is still not well defined. There have been some developments in the literature on the concept of modern universities such as the triple helix, entrepreneurial university and engaged university. However, those concepts focus on enforcing universities' roles in a single domain such as entrepreneurship, innovation, or civic engagement. Little is known about how universities can facilitate regional growth that goes beyond knowledge transfer activities such as spin-off creation, licensing, and patenting. This paper contributes to a more comprehensive understanding of universities' role in regional growth through the theoretical lens of ambidexterity. Using ambidexterity, universities with a regional focus were distinguished from those engaged in research commercialization and traditional third-mission roles. Through two case studies, this study found that teaching, research, and engagement should not be separated, since they can serve both economic and social missions. As a result, a new model of multidextrous universities is proposed where universities meet both economic and social missions through teaching, research, and engagement. Contrary to previous contributions which presented universities as ambidextrous organizations where tension appears only between research commercialization and research publication or between teaching and research, this study suggests that universities need to overcome tensions and incorporate a sense of place in all activities to successfully contribute to regional growth., (© The Author(s) 2023.)

Background: Conifer populations appear disproportionately threatened by global change. Most examples are, however, drawn from the northern hemisphere and long-term rates of population decline are not well documented as historical data are often lacking. We use a large and long-term (1931-2013) repeat photography dataset together with environmental data and fire records to account for the decline of the critically endangered Widdringtonia cedarbergensis. Eighty-seven historical and repeat photo-pairs were analysed to establish 20th century changes in W. cedarbergensis demography. A generalized linear mixed-effects model was fitted to determine the relative importance of environmental factors and fire-return interval on mortality for the species., Results: From an initial total of 1313 live trees in historical photographs, 74% had died and only 44 (3.4%) had recruited in the repeat photographs, leaving 387 live individuals. Juveniles (mature adults) had decreased (increased) from 27% (73%) to 8% (92%) over the intervening period. Our model demonstrates that mortality is related to greater fire frequency, higher temperatures, lower elevations, less rocky habitats and aspect (i.e. east-facing slopes had the least mortality)., Conclusions: Our results show that W. cedarbergensis populations have declined significantly over the recorded period, with a pronounced decline in the last 30 years. Individuals that established in open habitats at lower, hotter elevations and experienced a greater fire frequency appear to be more vulnerable to mortality than individuals growing within protected, rocky environments at higher, cooler locations with less frequent fires. Climate models predict increasing temperatures for our study area (and likely increases in wildfires). If these predictions are realised, further declines in the species can be expected. Urgent management interventions, including seedling out-planting in fire-protected high elevation sites, reducing fire frequency in higher elevation populations, and assisted migration, should be considered.

Background: Windthrow, the uprooting of trees during storms associated with strong winds, is a well-established cause of mortality in temperate regions of the world, often with large ecological consequences. However, this phenomenon has received little attention within arid regions and is not well documented in southern Africa. Slow rates of post-disturbance recovery and projected increases in extreme weather events in arid areas mean that windthrow could be more common and have bigger impacts on these ecosystems in the future. This is of concern due to slow rates of post-disturbance recovery in arid systems and projected increases in extreme weather events in these areas. This study investigated the spatial pattern, magnitude and likely causes of windthrown mortality in relation to other forms of mortality in Aloe dichotoma, an iconic arid-adapted arborescent succulent and southern Africa climate change indicator species., Results: We found that windthrown mortality was greatest within the equatorward summer rainfall zone (SRZ) of its distribution (mean = 31%, n = 11), and was derived almost exclusively from the larger adult age class. A logistic modelling exercise indicated that windthrown mortality was strongly associated with greater amounts of warm season (summer) rainfall in the SRZ, higher wind speeds, and leptosols. A statistically significant interaction term between higher summer rainfall and wind speeds further increased the odds of being windthrown. While these results would benefit from improvements in the resolution of wind and substrate data, they do support the hypothesised mechanism for windthrow in A. dichotoma. This involves powerful storm gusts associated with either the current or subsequent rainfall event, heavy convective rainfall, and an associated increase in soil malleability. Shallow rooting depths in gravel-rich soils and an inflexible, top-heavy canopy structure make individuals especially prone to windthrown mortality during storms., Conclusions: Results highlight the importance of this previously unrecognised form of mortality in A. dichotoma, especially since it seems to disproportionately affect reproductively mature adult individuals in an infrequently recruiting species. Smaller, more geographically isolated and adult dominated populations in the summer rainfall zone are likely to be more vulnerable to localised extinction due to windthrow events.

Sperm motility is regulated by protein phosphorylation. We have shown that the signaling kinase, glycogen synthase kinase-3 alpha (GSK-3 alpha), is present in spermatozoa. In somatic cells, GSK-3 is regulated by serine and tyrosine phosphorylation. In this report, we document that both GSK-3 alpha and GSK-beta isoforms are present in spermatozoa, with GSK-3 alpha being the predominant isoform. The relationship between GSK-3 serine phosphorylation and motility was investigated. Serine phosphorylation of GSK-3 increases significantly in spermatozoa during their passage through the epididymis. Initiation and stimulation of motility in vitro by isobutyl-methyl-xanthine, 2-chloro-2'-deoxy-adenosine, and calyculin A lead to a dramatic increase in GSK-3 serine phosphorylation. The concentration-dependent induction of motility by calyculin A is closely associated with GSK-3 serine phosphorylation. Immunoprecipitation of GSK-3 alpha and GSK-3 beta shows that both of the GSK-3 isoforms are more active in caput than in caudal spermatozoa. Calyculin A treatment decreased the activity of both isoforms. Column chromatography was used to purify inactive GSK-3 alpha from the caudal sperm extracts. This GSK-3 alpha species was phosphorylated at amino acid residues serine 21 and tyrosine 214. Inactive GSK-3 alpha is present in caudal but not in caput epididymal spermatozoa. The enzymes protein kinase B (PKB; also known as cAkt) and phosphoinositide 3-kinase (PI3-kinase), the upstream signaling proteins involved in GSK-3 phosphorylation, are both present in spermatozoa. Fluorescence immunocytochemistry showed that GSK-3 is present in the head and tail regions of sperm. Our work suggests a novel role for the signaling system involving GSK-3 in the regulation of sperm motility.
