Abstract Background Urine-derived stem cells (UDSCs) can be easily isolated from urine and possess excellent stem cell characteristics, making them a promising source for cell therapeutics. Due to their kidney origin specificity, UDSCs are considered a superior therapeutic alternative for kidney diseases compared to other stem cells. To enhance the therapeutic potential of UDSCs, we developed a culture method that effectively boosts the expression of Klotho, a kidney-protective therapeutic factor. We also optimized the Good Manufacturing Practice (GMP) system to ensure stable and large-scale production of clinical-grade UDSCs from patient urine. In this study, we evaluated the in vivo safety and distribution of Klotho-enhanced UDSCs after intravenous administration in accordance with Good Laboratory Practice (GLP) regulations. Methods Mortality and general symptoms were continuously monitored throughout the entire examination period. We evaluated the potential toxicity of UDSCs according to the administration dosage and frequency using clinical pathological and histopathological analyses. We quantitatively assessed the in vivo distribution and retention period of UDSCs in major organs after single and repeated administration using human Alu-based qPCR analysis. We also conducted long-term monitoring for 26 weeks to assess the potential tumorigenicity. Results Klotho-enhanced UDSCs exhibited excellent homing potential, and recovered Klotho expression in injured renal tissue. Toxicologically harmful effects were not observed in all mice after a single administration of UDSCs. It was also verified that repeated administration of UDSCs did not induce significant toxicological or immunological adverse effects in all mice. Single and repeated administrated UDSCs persisted in the blood and major organs for approximately 3 days and cleared in most organs, except the lungs, within 2 weeks. UDSCs that remained in the lungs were cleared out in approximately 4–5 weeks. There were no significant differences according to the variation of sex and administration frequency. The tumors were found in the intravenous administration group but they were confirmed to be non-human origin. Based on these results, it was clarified that UDSCs have no tumorigenic potential. Conclusions Our results demonstrate that Klotho-enhanced UDSCs can be manufactured as cell therapeutics through an optimized GMP procedure, and they can be safely administered without causing toxicity and tumorigenicity.

This study aimed to enhance the data write performance in measurements with exceptionally high sampling rates, such as acoustic emission measurements. This is particularly crucial when employing conventional hard disk drives to store data. This study introduced algorithms for handling binary formats, thereby reducing the data file size, increasing write rates, and ultimately shortening data write times during measurements. The suggested approaches included utilizing specialized binary formats and implementing self-created buffers. These approaches resulted in a remarkable write time reduction of up to 40×. Furthermore, employing multiple drives for writing significantly enhanced performance compared with that of using a single drive. Therefore, the proposed algorithms offer promising results for managing large amounts of data in real time.

Low-pressure gas-admission marine two-stroke engines have significant potential for reducing emissions in the shipping industry. However, they exhibit poor combustion quality under low-load conditions, leading to lower thermal efficiency and high levels of HC and CO emissions. This study numerically evaluated the impacts of the gas admission location and timing on engine performance and exhaust gas emissions under low load conditions. The study revealed that different gas admission locations form distinct NG-air stratifications. Lower gas admission locations resulted in longer ignition delays and shorter combustion durations. The unburned CH4 and CO emissions were significantly reduced by a maximum of 20.14% and 42.29%, respectively. A trade-off exists between NOx and N2O emissions owing to the different temperature conditions of their formation. The NOx emissions increased continuously as the gas admission location was lowered to a maximum of 3.4218 g/kWh; a value slightly above the Tier III emission standards. Advancing the timing of gas admission further decreased the ignition delay time, NOx, and unburned CH4 emissions. This study revealed that in-cylinder natural gas mixture stratification due to varying gas admission locations and timings can improve the combustion efficiency and exhaust gas emissions of a natural gas-diesel dual-fuel engine under low-load conditions.

The injection timing of fuel in a diesel engine affects the combustion condition. Advanced fuel injection prolongs the ignition delay, positively impacting the increase in maximum combustion pressure and improving output. However, excessively advanced fuel injection can cause knocking. Moreover, premature ignition results in increased compression work when the maximum combustion pressure occurs before top dead center (TDC). This study aimed to diagnose and rectify starting failures, noise, and vibrations in a commercially operated ship engine by measuring the combustion pressure during low load operation. The target engine was a 4-stroke diesel, and the fuel injection system was mechanically controlled by a camshaft. The measured engine exhibited a 4.5 °CA error between the TDC, determined by the flywheel mark and the actual TDC. This discrepancy was influenced by excessively advanced fuel injection timing. It was confirmed that fuel injection and ignition were excessively advanced in all cylinders. After readjusting the engine by delaying the fuel injection timing by approximately 10 °CA, the combustion pressure was remeasured. The ignition was delayed by approximately 6.5 °CA at the same load, and the ignition intervals were uniformly adjusted. As the ignition timing was retarded, the compression work decreased and the expansion work increased in each cylinder, resulting in improved output across all cylinders. The amplitude of crankshaft angular velocity variation significantly decreased, improving uneven rotational force.

Although hydraulic accumulators play a vital role in the hydraulic system, they face the challenges of being broken by continuous abnormal pulsating pressure which occurs due to the malfunction of hydraulic systems. Hence, this study develops anomaly detection algorithms to detect abnormalities of pulsating pressure for hydraulic accumulators. A digital pressure sensor was installed in a hydraulic accumulator to acquire the pulsating pressure data. Six anomaly detection algorithms were developed based on the acquired data. A threshold averaging algorithm over a period based on the averaged maximum/minimum thresholds detected anomalies 2.5 h before the hydraulic accumulator failure. In the support vector machine (SVM) and XGBoost model that distinguish normal and abnormal pulsating pressure data, the SVM model had an accuracy of 0.8571 on the test set and the XGBoost model had an accuracy of 0.8857. In a convolutional neural network (CNN) and CNN autoencoder model trained with normal and abnormal pulsating pressure images, the CNN model had an accuracy of 0.9714, and the CNN autoencoder model correctly detected the 8 abnormal images out of 11 abnormal images. The long short-term memory (LSTM) autoencoder model detected 36 abnormal data points in the test set.

The vessel speed reduction program (VSRP) was first introduced in the Port of Los Angeles in 2001 to improve air quality. In this study, an algorithm was developed to calculate ship emissions with a bottom-up approach based on ship activity using automatic identification system (AIS) data. The target vessel applied to the emission calculation was a vessel participating in Korea’s VSRP. Factors considered for the calculation of emissions were ship type, speed, gross tonnage, engine power, load, sulfur content of fuel, and fuel consumption rate by engine age. The algorithm is designed to calculate the reduction amount by VSRP by simultaneously calculating the emission amount of the actual speed and the emission amount of the cruise speed when not participating in VSRP. The emission results of ships that participated in the VSRP in 2020 revealed that their speed was reduced by 47% and carbon dioxide emissions by 71.9%. These results were verified through comparison with the fuel consumption report of the container fleet presented by the International Maritime Organization. Our findings can be used to monitor the air pollutant emissions of ships entering major ports in Korea and to develop policies envisaged at reducing the production and effect of greenhouse gases and air pollutants. Moreover, we recommend that this model be replicated in other countries for monitoring ship-induced emissions.

Autoimmune disease: the importance of estrogen Estrogen signaling may play a crucial role in mediating autoimmune responses and preventing diseases such as rheumatoid arthritis. Estrogen plays vital roles in the female endocrine and reproductive systems, and its function is partly mediated by the estrogen receptor alpha (ERα). ERα-deficient mice have more severe autoimmune disease, suggesting a link between estrogen signaling and immune responses. Je-Min Choi at Hanyang University in Seoul, South Korea, and co-workers generated mice with T cells lacking ERα and followed their immune system development. They were particularly interested in monitoring follicular helper T cells (TFH), which help B cells generate antibodies in response to pathogen attack as well as self-antigen. The ERα-deficiency in T cells spontaneously developed mild autoimmune symptoms in mice, with increased levels of TFH and autoantibodies. Their results suggest ERα-mediated estrogen signaling in CD4 T cells is needed to suppress TFH activity and limit autoimmune disease development in female.

T cells express specific T cell receptors (TCR) to recognise antigens and initiate adaptive immune responses. Here the authors show, in a mouse model of autoimmune encephalomyelitis, that memory-like CD4 T cells expressing unrelated TCR can also infiltrate the spinal cord and contribute to autoimmunity.

The measurement of shaft bending (whirling) moment can be performed via a telemetry system including strain gauges which can obtain the bending stress. By using a single bridge of strain gauge, it was possible to obtain only the nominal bending moment. However, in case of the propeller shaft vibration measurement, the true dynamic bending moment is needed to evaluate the effect of propeller forces on the stability of the bearings. To deal with this, typically two bridges of strain gauge at 90 degrees are needed. This research presents a novel reliability assessment method in establishing the true dynamic bending moment using only one bridge of strain gauge. This is achieved by using the actual bending stress measured by strain gauge combined with its own phase lag stress at an angle of 90 degrees. To validate this technique, the experiments were performed under the rapid turning transient states during a sea trial of a 50,000 DWT oil/chemical tanker. As a result, great fluctuations in propeller force were detected, resulting in a non-uniform oil film in the bearings. The displacement sensor was also installed and confirmed the established true dynamic bending moment.

MoS2 nanoscrolls that have inner core radii of ∼250 nm are generated from MoS2 monolayers, and the optical and transport band gaps of the nanoscrolls are investigated. Photoluminescence spectroscopy reveals that a MoS2 monolayer, originally a direct gap semiconductor (∼1.85 eV (optical)), changes into an indirect gap semiconductor (∼1.6 eV) upon scrolling. The size of the indirect gap for the MoS2 nanoscroll is larger than that of a MoS2 bilayer (∼1.54 eV), implying a weaker interlayer interaction between concentric layers of the MoS2 nanoscroll compared to Bernal-stacked MoS2 few-layers. Transport measurements on MoS2 nanoscrolls incorporated into ambipolar ionic-liquid-gated transistors yielded a band gap of ∼1.9 eV. The difference between the transport and optical gaps indicates an exciton binding energy of 0.3 eV for the MoS2 nanoscrolls. The rolling up of 2D atomic layers into nanoscrolls introduces a new type of quasi-1D nanostructure and provides another way to modify the band gap of 2D materials.

It is known that a ship’s shafting system can be adversely affected by hull deformation, variations in the engine power, the propeller load, and eccentric propeller thrusts, thereby increasingly affecting the behavior of the shaft’s movement. A deformed shafting system may also lead to a potential risk of bearing damage by causing a change in the local load of the rear part of the after-stern tube bearing of the propeller shaft. With this concern, a series of previous studies were focused on optimizing the effects of hull deformation by securing a proper level of propulsion shaft stability and optimizing the relative inclination angle and oil film retention based on a quasi-static state, that is, Rules for the Classification of Steel Ships and experiences of shipyards. However, despite our efforts to resolve this issue, marine accidents involving stern tube bearing damage have continued to occur under relatively unattended ship motions in a transient state, that is, a quasi-static state that can possibly cause sudden stern flow field changes. Therefore, to improve the stability of the propulsion shaft, it is necessary to understand ship motions and the conditions of shafting systems in a dynamic state and a transient state when the system is designed. From this point of view, this study investigated the effect of changes in eccentric propeller forces on the motion of the propeller shaft in a representative transient state of a 50,000 deadweight tonnage tanker by means of the strain gauge method and a displacement sensor. The research findings demonstrate that propeller thrust fluctuations have a direct effect on the shaft stability by significant changes in the shaft motion that can lead to unbalanced supporting loads on the stern tube bearing. These results clearly reproduce the cause of damage to the ship in which the accident occurred at a reliable level and will be a reference for establishing pragmatic guidelines for preventing damage to similar ships in the future.

Exploring the ground state of two-dimensional (2D) magnetic systems enriches the fundamental understanding of magnetism and boosts the applications. The authors here show the suppression of magnetic order and 2D XY behavior in XXZ-type antiferromagnet NiPS3 when approaching the monolayer limit by Raman spectroscopy.

Nowadays, damage to ships due to marine debris at sea is increasingly reported. Specifically, a piece of rope or fishing net can wrap around the propeller shaft, stopping it from rotating. Although various efforts have been made, there are still numerous practical challenges. A rope cutter system, which was designed to cut suspended objects wrapped around the propeller shaft, offers a great advantage in protecting the propulsion system. Since the cutting action produces the transient torsional vibration, the connecting bolts should be stable and sufficiently rigid under normal conditions. However, in the event of an encounter with an object that is too hard to cut, the bolts must be broken so that the rope cutter is released. Those kinds of objects may not be long enough to wrap around the shaft, such as a piece of wood. Therefore, this research presents the novelty in maintaining the safety of the propulsion system installed with a rope cutter. In this study, a method for obtaining the maximum diameter of the connecting bolt as a safety device is described, and the torques required to cut ropes of various thicknesses were determined based on actual measurements. Finally, a series of experiments in the laboratory-scale and shipboard test on an actual ship show that the rope cutter does not significantly reduce the ship’s power performance or increase the structural vibration.

Global warming and air pollutants are in general major worldwide concerns including for the marine shipping industry. Equipped with new technologies, the onboard DC grid has proven several advantages, including up to 27% reduction in specific fuel consumption with reduced emissions. That can be achieved by installing an optimized variable speed diesel generator. The engine speed is adjusted according to the required power, which allows to always keep the best efficiency of the combustion process. However, it also exposes some changes in the behavior of the structure vibrations. Measurements on an experimental variable speed diesel generator show that vibration increases when trying to slow down the engine for the same load. This behavior is closely related to the resonance in low rev range that usually occurs with general gensets. In other words, we can conclude that the DC grid’s variable speed generator may be beneficial for fuel efficiency, but not for mechanical life and safety. Several measures had been given, of which the alternation of the natural frequency is presented as an economical and efficient solution. The ultimate goal is to maintain operational safety while respecting reduced fuel consumption.

Layered materials have strikingly anisotropic mechanical properties. Here, the authors use Raman spectroscopy and first-principles calculations to unveil that MoS2, an archetypal layered material, possesses a coupling between in-plane uniaxial strain and interlayer shear, enabling derivation of an unexplored off-diagonal elastic constant.

The enlargement of ships has increased the relative hull deformation owing to draft changes. Moreover, design changes such as an increased propeller diameter and pitch changes have occurred to compensate for the reduction in the engine revolution and consequent ship speed. In terms of propulsion shaft alignment, as the load of the stern tube support bearing increases, an uneven load distribution occurs between the shaft support bearings, leading to stern accidents. To prevent such accidents and to ensure shaft system stability, a shaft system design technique is required in which the shaft deformation resulting from the hull deformation is considered. Based on the measurement data of a medium-sized oil/chemical tanker, this study presents a novel approach to predicting the shaft deformation following stern hull deformation through inverse analysis using deep reinforcement learning, as opposed to traditional prediction techniques. The main bearing reaction force, which was difficult to reflect in previous studies, was predicted with high accuracy by comparing it with the measured value, and reasonable shaft deformation could be derived according to the hull deformation. The deep reinforcement learning technique in this study is expected to be expandable for predicting the dynamic behavior of the shaft of an operating vessel.

Cyclodextrins (CDs) have drawn a lot of attention from the scientific communities as a model system for host–guest chemistry and also due to its variety of applications in the pharmaceutical, cosmetic, food, textile, separation science, and essential oil industries. The formation of the inclusion complexes enables these applications in the condensed phases, which have been confirmed by nuclear magnetic resonance (NMR) spectroscopy, X-ray crystallography, and other methodologies. The advent of soft ionization techniques that can transfer the solution-phase noncovalent complexes to the gas phase has allowed for extensive examination of these complexes and provides valuable insight into the principles governing the formation of gaseous noncovalent complexes. As for the CDs’ host–guest chemistry in the gas phase, there has been a controversial issue as to whether noncovalent complexes are inclusion conformers reflecting the solution-phase structure of the complex or not. In this review, the basic principles governing CD’s host–guest complex formation will be described. Applications and structures of CDs in the condensed phases will also be presented. More importantly, the experimental and theoretical evidence supporting the two opposing views for the CD–guest structures in the gas phase will be intensively reviewed. These include data obtained via mass spectrometry, ion mobility measurements, infrared multiphoton dissociation (IRMPD) spectroscopy, and density functional theory (DFT) calculations.

The reliability of propulsion shafting systems is a major concern for ocean-going vessels because mid-ocean repairs can be time-consuming and spare parts must be available. To address this concern, vibration modeling and experimental measurements were conducted on a propulsion shafting system with a Z-drive propeller, with the objective of identifying the source of failure for the flexible rubber coupling connecting the diesel engine with the intermediate shaft. The torsional fluctuations in the flexible coupling dramatically increased and then abruptly ceased. The modeling results revealed that the frictional losses during power transmission through the universal joints could act as an excitation force for self-excited vibration. The coupling connected to the intermediate shaft did not have sufficient radial flexibility to dampen these vibrations. To avoid the effects of the self-excited torsional vibration, it is recommended that this coupling is replaced with one that is capable of absorbing the radial shaft displacement.

The nuclear factor of activated T cells (NFAT) family of transcription factors, which includes NFAT1, NFAT2, and NFAT4, are well-known to play important roles in T cell activation. Most of NFAT proteins are controlled by calcium influx upon T cell receptor and costimulatory signaling results increase of IL-2 and IL-2 receptor. NFAT3 however is not shown to be expressed in T cells and NFAT5 has not much highlighted in T cell functions yet. Recent studies demonstrate that the NFAT family proteins involve in function of lineage-specific transcription factors during differentiation of T helper 1 (Th1), Th2, Th17, regulatory T (Treg), and follicular helper T cells (Tfh). They have been studied to make physical interaction with the other transcription factors like GATA3 or Foxp3 and they also regulate Th cell signature gene expressions by direct binding on promotor region of target genes. From last decades, NFAT functions in T cells have been targeted to develop immune modulatory drugs for controlling T cell immunity in autoimmune diseases like cyclosporine A, FK506, etc. Due to their undesirable side defects, only limited application is available in human diseases. This review focuses on the recent advances in development of NFAT targeting drug as well as our understanding of each NFAT family protein in T cell biology. We also discuss updated detail molecular mechanism of NFAT functions in T cells, which would lead us to suggest an idea for developing specific NFAT inhibitors as a therapeutic drug for autoimmune diseases.
