Plants have long been exploited as a sustainable source of food, flavors, agrochemicals, colors, therapeutic proteins, bioactive compounds, and stem cell production. However, plant habitats are being briskly lost due to scores of environmental factors and human disturbances. This necessitates finding a viable alternative technology for the continuous production of compounds that are utilized in food and healthcare. The high-value natural products and bioactive compounds are often challenging to synthesize chemically since they accumulate in meager quantities. The isolation and purification of bioactive compounds from plants is time-consuming, labor-intensive, and involves cumbersome extraction procedures. This demands alternative options, and the plant cell culture system offers easy downstream procedures. Retention of the metabolic cues of natural plants, scale-up facility, use as stem cells in the cosmetics industry, and metabolic engineering (especially the rebuilding of the pathways in microbes) are some of the advantages for the synthesis and accumulation of the targeted metabolites and creation of high yielding cell factories. In this article, we discuss plant cell suspension cultures for the in vitro manipulation and production of plant bioactive compounds. Further, we discuss the new advances in the application of plant cells in the cosmetics and food industry and bioprinting.

Sweet sorghum is the present target and cheap source for biofuel production. Tissue culture recalcitrance and low levels of transformation reproducibility of protocols are main constrains for transgenic sweet sorghum development. Production of a transient transgenic sweet sorghum for the development of disease resistant transgenics was the aim of our studies. In this study we have developed an efficient regeneration and transformation system for in vitro culture of sweet sorghum from the immature embryo callus. Histological studies of sweet sorghum embryogenic calli revealed that the development of embryogenic shoots. Murashige and Skoog nutrient agar medium with different concentrations of 2mg/l 2,4-D and combination of 0.2 mg/l NAA and 2 mg/l Kinetin gave higher frequency of callus induction and shoot regeneration. Embryogenic callus was competent to accept the DNA with GV2600 strain. These tissues are susceptible to Agrobacterium mediated transformation carrying pCAMBIA1301 with gus gene construct as well as for shoot multiplication. For effective transformation, GV2600 strain carrying pCAMBIA1301 at 0.6 O.D. was found to be compatible in giving sweet sorghum transgenics. Molecular confirmation was done by Polymerase Chain Reaction and Southern blotting for the putative plants. The regenerated plantlets survived during acclimatization were growing similar to the normal plants. All the major parameters that affecting transformation frequency were optimized for the development of perfect sweet sorghum transgenics.

Systems genetics is key for integrating a large number of variants associated with diseases. Vitamin K (VK) is one of the scarcely studied conditions in lieu of ascertaining either the differentially expressed genes (DEGs) or variants in an individual subpopulation of diseased phenotypes associated with VK, viz. myocardial infarction, renal failure, prostate cancer, thrombosis, thrombocytopenia, coagulation related diseases to name a few. In this work, we have screened characteristic DEGs common to three VK-related diseases, viz. myocardial infarction, renal failure and prostate cancer and asked whether or not any DEGs in addition to pathogenic variants are common to these conditions. We attempt to bridge the gap in finding characteristic biomarkers and discuss the role of long noncoding RNAs (lncRNAs) in the biogenesis of VK deficiencies.

Livestock provides an additional source of income for marginal cropping farmers, but crop residues that are used as a main source of animal feed are characteristically low in digestibility and protein content. This reduces the potential livestock product yield and quality. The key trait, which influences the quality and the cost of animal feed, is digestibility. In this study, we demonstrate that sorghum breeding can be directed to achieve genetic gains for both fodder biomass and digestibility without any trade-offs. The genotypic variance has shown significant differences for biomass across years (13,035 in 2016 and 3,395 in 2017) while in vitro organic matter digestibility (IVOMD) showed significant genotypic variation in 2016 (0.253) under drought. A range of agronomic and fodder quality traits was found to vary significantly in the population within both the control and drought conditions and across both years of the study. There was significant genotypic variance (σg2) and genotypic × treatment variance (σgxt2) in dry matter production in a recombinant inbred line (RIL) population in both study years, while there was only significant σg2 and σgxt2 in IVOMD under the control conditions. There was no significant correlation identified between biomass and digestibility traits under the control conditions, but there was a positive correlation under drought. However, a negative relation was observed between digestibility and grain yield under the control conditions, while there was no significant correlation under drought population, which was genotyped using the genotyping-by-sequencing (GBS) technique, and 1,141 informative single nucleotide polymorphism (SNP) markers were identified. A linkage map was constructed, and a total of 294 quantitative trait loci (QTLs) were detected, with 534 epistatic interactions, across all of the traits under study. QTL for the agronomic traits fresh and dry weight, together with plant height, mapped on to the linkage group (LG) 7, while QTL for IVOMD mapped on to LG1, 2, and 8. A number of genes previously reported to play a role in nitrogen metabolism and cell wall-related functions were found to be associated with these QTL.

Exploding global population, rapid urbanization, salinization of soils, decreasing arable land availability, groundwater resources, and dynamic climatic conditions pose impending damage to our food security by reducing the grain quality and quantity. This issue is further compounded in arid and semi-arid regions due to the shortage of irrigation water and erratic rainfalls. Millets are gluten (a family of proteins)-free and cultivated all over the globe for human consumption, fuel, feed, and fodder. They provide nutritional security for the under- and malnourished. With the deployment of strategies like foliar spray, traditional/marker-assisted breeding, identification of candidate genes for the translocation of important minerals, and genome-editing technologies, it is now tenable to biofortify important millets. Since the bioavailability of iron and zinc has been proven in human trials, the challenge is to make such grains accessible. This review encompasses nutritional benefits, progress made, challenges being encountered, and prospects of enriching millet crops with essential minerals.

Genes encoding bacterial cold shock proteins A (CspA, 213 bp) and B (CspB, 216 bp) were isolated from Escherichia coli strain K12, which showed 100% homology with gene sequences isolated from other bacterial species. In silico domain, analysis showed eukaryotic conserved cold shock domain (CSD) and ribonuclease-binding domain (RBD) indicating that they bind to RNA and are involved in temperature stress tolerance. Overexpression of these two genes in E. coli resulted in higher growth in presence of 200 mM NaCl and 300 mM mannitol. Western blot confirmed the translational products of the two genes. Seedlings of indica rice were transformed with Agrobacterium tumefaciens containing pCAMBIA1301 CspA and CspB genes. Transgene integration was confirmed by β-glucuronidase (GUS) histochemical assay, polymerase chain reaction (PCR) amplification, and gene copy number by Southern blotting. Chlorophyll, proline, Na+, and K+ contents were higher in transgenics exposed to 150 mM NaCl and drought (imposed by withholding water) stresses during floral initiation stage. Catalase (CAT), superoxide dismutase (SOD), and guaiacol peroxidase (GPX) activities increased, while malondialdehyde (MDA) content was low in transgenics. Transgenics displayed increased root, shoot, and panicle lengths, root dry mass, and a distinct stay-green (SGR) phenotype. Higher transcript levels of CspA, CspB, SGR, chlorophyllase, isopentenyl adenine transferase 1 (IPT1), 9-cis-epoxycarotenoid dioxygenase (NCED), SOD, and sirtuin 1 (SIRT1) genes were observed in transgenics compared to wild type plants (WT) under multiple stresses. Present work indicates that bacterial chaperone proteins are capable of imparting SGR phenotype, salt and drought stress tolerance alongside grain improvement.

Sorghum is widely grown as a failsafe crop in semi-arid regions particularly in post rainy season. Though the effect of drought on crop performance is studied widely there are few studies illustrating the association of fodder quality and agronomic traits under drought. To study the interactions we evaluated a set of 24 cultivars under drought for three years in post rainy season. The effect of drought was evident in delayed flowering (by 2 days) and reduce plant height (by 0.98 cm) compared to control. The fodder digestibility traits were reduced (in vitro organic matter digestibility by 2.25 times) under drought. Genomic DNA of good quality is a prerequisite in molecular biology studies involved in deciphering the information contained in the molecules that are chosen for studying the evolutionary interrelationships among taxa. Genomic DNA should be free of contaminants which may otherwise interfere while performing PCR. This is particularly essential while isolating genomic DNA from gastropods due to the presence of large amounts of mucopolysaccharides in their tissues. In the present study, we have developed a new method for obtaining high quality genomic DNA from a pulmonate snail Achatina fulica, which secretes a large amount of mucus. In this method, we have obtained good quality genomic DNA, compatible for downstream molecular biology applications like PCR and sequencing of 16S rDNA. The yield of the genomic DNA was 2790 µg/ml. This method may be extended for isolating high quality genomic DNA from different gastropods native to Indian sub-continent, which serves as a starting point for studying their phylogenetic affinities.

Bacteria produce and sense chemical signal molecules, communicate with closet neighbors. Chemical signals are diverse and comprise cyclic and linear peptides, short and long chain γ-quinolones, N-acyl-homoserine lactones (AHL) and unsaturated fatty acids. These signaling molecules are collectively called autoinducers. Certain signals are readily diffusible small molecules, while others are hydrophobic and can be vesicle or membrane -associated. Several chemical signals are vastly genus or species specific, while LuxS gene product furanosyl borate diester, a more universally synthesized and recognized molecule. It is assumed that bacteria use these auto inducer molecules not only to identify their neighbors and cell density, but also to govern some aspects of their environment, such as confinement and diffusion. In a broad sense, quorum sensing allows harmonization of cell density wide activities, together with virulence factor production, biofilm dynamics, bioluminescence and swarming motility on surfaces. Swarming motility is a flagella-driven movement of bacterial cells through it can spread as a biofilm over a surface. Different chemical signals produced either by bacteria (AHL) may persuade the QS regulated swarming activities in bacteria. This review emphasizes the role of AHL and other low-molecular-mass signal molecules involvement of in swarming motility of bacteria.
