9. Dissection of cell wall assembly dynamics during early embryogenesis in the brown alga Fucus

Abstract

Zygotes from Fucus species have been used extensively to study cell polarization and rhizoid outgrowth, and in this model system cell wall deposition aligns with the establishment of polarity. Monoclonal antibodies are essential tools for the in situ analysis of cell wall glycans, and here we report the characteristics of six monoclonal antibodies to alginates (BAM6–BAM11). The use of these, in conjunction with monoclonal antibodies to brown algal sulfated fucans, has enabled the study of the developmental dynamics of the Fucus zygote cell walls. Young zygotes are spherical and all alginate epitopes are deposited uniformly following cellulose deposition. At germination, sulfated fucans are secreted in the growing rhizoid wall. The redistribution of cell wall epitopes was investigated during treatments that cause reorientation of the growth axis (change in light direction) or disrupt rhizoid development (arabinogalactan-protein-reactive Yariv reagent). Alginate modeling was drastically impaired in the latter, and both treatments cause a redistribution of highly sulfated fucan epitopes. The dynamics of cell wall glycans in this system have been visualized in situ for the first time, leading to an enhanced understanding of the early developmental mechanisms of Fucus species. These sets of monoclonal antibodies significantly extend the available molecular tools for brown algal cell wall studies.

Abstract

Introduction: After early successful experience with transcatheter aortic valve replacement (TAVR), concept of transcatheter implantation of a new valve within a failing bioprosthetic valve emerged. Valve-in-valve (ViV) implantation seems to be a simpler option for high risk surgical patients. Methods: We performed five ViV procedures in different valve positions. We included patients with failing bioprosthetic valves with high surgical risk due to concomitant comorbidities. We performed 2 transapical ViV procedures for failing mitral bioprosthetic valves, 1 transfemoral procedure for failing pulmonary valve and 2 transfemoral ViV implantation for failing tricuspid bioprosthetic valves. Results: The procedures were successfully completed in all 5 cases with initial excellent fluoroscopic and echocardiographic verification. There was no valve embolization or paravalvular leakage in any of the cases. Transcatheter valve function was appropriate with echocardiography. Post procedural clinical adverse events like pleural effusion and transient ischemic attack were managed successfully. In midterm follow up all cases remained in appropriate functional class except from the transcatheter pulmonary valve which became moderately stenotic and regurgitant. Conclusion: As the first Iranian all-comers case series with midterm follow up for ViV implantation, we had no mortality. Interestingly none of our patients had neurologic sequelae after the procedure. Midterm follow up for our patients was acceptable with good functional class and appropriate echocardiographic findings. Due to high surgical risk of the redo procedure after failing of a bioprosthetic valve especially in elderly patients with comorbidities, ViV implantation would be a good alternative to surgery for this high risk group.

Despite this, surgeries have been performed using HAC to close nasal/sinus cerebrospinal fluid (CSF) leaks or in the treatment of encephaloceles. Methods This study retrospectively documented four patients in whom HAC was used for closure of ethmoid and sphenoid sinus CSF leaks. Results This study showed that exposed hydroxyapatite could be extremely problematic resulting in scarring, chronic granulation, infection, and prolonged healing. Oftentimes, revision surgery was necessary to help control the reaction to hydroxyapatite. Conclusion Because of the frequent complications occurring with use of hydroxyapatite in the paranasal sinuses, its use should be limited to selected individuals in whom difficulty in achieving closure of the CFS leak could be anticipated and in whom adequate tissue coverage could be provided.

En el Centro de Acuicultura Continental Hardap se implementó un programa de desarrollo de la acuicultura a partir de la formación de tres bancos de reproductores, dos de tilapia mossambica, uno tomado de la presa HARDAP (246 y 216 , y el otro (250 y 215 a partir de alevines seleccionados del segundo ciclo de desove de esta especie logrado en dicho centro. El tercer banco de reproductores formado fue de carpa común (200 y 236 capturado del medio natural. Se estableció un flujo productivo de ambas especies por ciclos que permitió obtener alevines destinados a las granjas de peces estatales, privadas, y a las comunidades situadas en las regiones de: Karas, khomas, Omaheke y Hardap. En el primer cuatrimestre del año 2011 se llevaron a crecimiento en tierra 135 650 alevines, se inicio la siembra de 14 350 ejemplares a densidades de 3 ej./m2 en los 48 acuatorios previamente seleccionados en condiciones de monocultivo, o en policultivo en una proporción de tilapia al 60 % y 40 % de carpas. La cosecha se realizó entre los 9-11 meses de cultivo con pesos entre 200-300 g y supervivencia de 70-85 %. Miembros de la comunidad previamente capacitados, se incorporaron a la atención del cultivo integrado de peces con vegetales (zanahoria, remolacha, cebolla) y animales domésticos (cabras, chivos y carneros). Para promocionar el consumo de pescado y métodos de preparación se impartió un taller a la comunidad de Hardap. Se desarrolló una labor divulgativa sobre la actividad acuícola con vista a orientar el perfil vocacional de los alumnos del último año de secundaria de las regiones de Mariental y Rehoboth. Community members previously trained, joined the growing attention of integrated fish farming vegetables (carrots, beets, onions) and domestic animals (goats, sheep and goats). Fish consumption and preparation methods were promoted in a workshop to the community of Hardap. Abstract

Background: Aromatase inhibition has been proposed as a potential approach for growth enhancement in children with short stature, but detailed animal studies are lacking. Aim: To assess the effect and potential adverse effects of aromatase inhibition on growth in female rats. Methods: Prepubertal Wistar rats received intramuscular injections with placebo or the aromatase inhibitor exemestane at a dose of 10, 30 or 100 mg/kg/week (E10, E30, E100) for 3 weeks. A control group was ovariectomized (OVX). Weight and length gain, tibia and femur length, growth plate width, organ weights, insulin-like growth factor I (IGF-I) levels, and histology of the ovaries, uterus and brain were analyzed. X-ray microtomography of femora was performed. Results: E100 significantly increased weight gain and growth plate width, but less prominently than OVX. Trabecular number and thickness were decreased in E100 and OVX in the metaphysis and epiphysis. E100 significantly decreased ovarian weight and multiple cysts were seen upon histological evaluation. No significant effects were found on IGF-I levels and brain morphology in E100. E10 and E30 had no effects on growth. Background: Estrogen deficiency induced by aromatase inhibitors may be a novel treatment modality for growth enhancement in short children, but may have adverse effects on bone, brain and reproduction. Aim: To assess growth effects and potential adverse effects of aromatase inhibition in male rats. Methods: 26-day-old prepubertal rats received intramuscular injections with placebo or the aromatase inhibitor exemestane at a dose of 10, 30 or 100 mg/kg/week [E10, E30, E100(6)] for 6 weeks, completely covering the sexual maturation phase, or with 3 weeks E100 followed by 3 weeks placebo [E100(3)]. Growth parameters and histology of the testis, seminal vesicle and brain were analyzed. Bone architecture was studied with X-ray microtomography. Results: Exemestane dose-dependently decreased body weight and tail length gain, as well as liver and seminal vesicle weights, but did not affect nose-anus length gain, growth plate width or radial growth. E100(6) decreased trabecular thickness (epiphysis and metaphysis) and number (metaphysis). Normal IGF-I levels and brain, testis and seminal vesicle morphology were observed. E100(3) resulted in decreased tail length gain only. Background: There was a need for a simple method of predicting the severity of gallstone pancreatitis soon after admission, to guide the clinician for intensive monitoring, or for transfer to a specialist center. Aim: To validate the role of simplified criteria [white blood cell count (WBC) ⩾ 14.5 x 1 03/dl; blood urea nitrogen (BUN) ⩾ 12 mg/dl; random blood sugar (RBS) ⩾ 150 mg/dl; pulse ⩾ 1 00/mm] in predicting the severity of gallstone pancreatitis at admission. Setting and Design: This prospective study was performed on 54 patients with gallstone pancreatitis admitted in the Department of Surgery of a tertiary hospital. Material and Methods: Fifty-four patients with gallstone pancreatitis were followed for development of complications. Sensitivity analysis of Biliary Ranson ⩾ 3, modified lmrie ⩾ 3, acute physiology and chronic health evaluation II (APACHE-Il) score of ⩾ 5, WBC ⩾ 14.5 x 1 03/dl, BUN ⩾ 12 mg/dl, RBS ⩾ 150 mg/dl, pulse ⩾ 1 00/mm and combinations of the four simplified criteria were compared using the Z4est. P < 0.05 was considered statistically significant. Results: Both Biliary Ranson ⩾ 3 and modified lmrie ⩾ 3 had sensitivity of 96.15% and specificity of 96.43%. Both APACHE-Il ⩾ 5 and RBS ⩾ 150 mg/dl had sensitivity of 88.46%, which is comparable to Biliary Ranson ⩾ 3 and modified lmrie ⩾ 3. Conclusion: RBS > 150 mg/dl can be used as a simplified prognostic indicator at admission in patients with gall stone pancreatitis and is comparable to Biliary Ranson ⩾ 3, modified lmrie⩾3, and APACHE-Il of⩾5. 37. Efficiency of population structures for mapping of Mendelian and imprinted quantitative trait loci in outbred pigs using variance component methods

Abstract

In a simulation study different designs for a pure line pig population were compared for efficiency of mapping QTL using the variance component method. Phenotypes affected by a Mendelian QTL, a paternally expressed QTL, a maternally expressed QTL or by a QTL without an effect were simulated. In all alternative designs 960 progeny were phenotyped. Given the limited number of animals there is an optimum between the number of families and the family size. Estimation of Mendelian and parentally expressed QTL is more efficient in a design with large family sizes. Too small a number of sires should be avoided to minimize chances of sires to be non-segregating. When a large number of families is used, the number of haplotypes increases which reduces the accuracy of estimating the QTL effect and thereby reduces the power to show a significant QTL and to correctly position the QTL. Dense maps allow for smaller family size due to exploitation of LD-information. Given the different possible modes of inheritance of the QTL using 8 to16 boars, two litters per dam was optimal with respect to determining significance and correct location of the QTL for a data set consisting of 960 progeny. The variance component method combining linkage disequilibrium and linkage analysis seems to be an appropriate choice to analyze data sets which vary in marker density and which contain complex family structures.

Abstract

The characteristics of detected wave form, and output power of the oscillations of a Gunn diode, with an oscillation frequency of approximately 4 GHz mounted inside a coaxial type resonant cavity, at varied cavity lengths, were measured when the temperature was lowered from room temperature to -120degreesc.
Heat is generated when a Gunn diode is operated at relatively high output power. Generally, because of this heat, the quality characteristics of the diode get worse. That is why that the characteristics of the Gunn diode at higher temperatures, above room temperature, has practical importance and a relatively large amount of research has been done on it. On the other hand, we know that theoretically the output power and efficiency of oscillation in the diode increases gradually as the temperature decreases from room temperature. Abstract

Carrageenans are major gel forming polysaccharides in the extracellular matrix of the red macroalga Chondrus crispus. These galactans are made of linear chains of repetitive disaccharide motifs based on d-galactose residues alternately linked by β-1,4 and α-1,3 glycosidic bonds. A definite number of disaccharide motifs are known, based on their regular sulfations and the presence of a 3,6-anhydro bridge. While these motifs are variable as a function of species, life cycle phases, or seasons, our understanding of the in vivo regulation of carrageenan fine structures is still limited. Characterized hydrolytic enzymes (κ-, ι- and λ-carrageenases) are powerful tools for identifying glycan structures in extracted ECMs. Their use, combined to chromatography and high-resolution mass spectrometry, allowed us to refine our understanding of carrageenan variability in the phases of C. crispus. We provide the first demonstration that κ/ι carrabiose motifs are not limited to gametophytes, but are also present in tetrasporophytes, together with over- and under-sulfated λ-motifs. Our findings highlight a more complex carrageenan composition than previously described in this model system. These results are further discussed in the light of recent transcriptomic data and suggest that the historical hypotheses on the biosynthetic pathway of carrageenans in red algae may need revision. Statement of the Problem: Dental caries and periodontal disease are the most common oral problems. Chemical antibacterial agents often have side effects; thus, researchers have long been in search for organic and herbal products to prevent dental caries and periodontal disease., Purpose: The present study has aimed to assess the effects of Dracocephalum moldavica essential oil on Streptococcus mutans , Streptococcus salivarius , Streptococcus sobrinus , and Lactobacillus acidophilus compared to Chlorhexidine (CHX)., Materials and Method: In this in vitro study, the plants were collected from Zanjan Province, Iran. Analysis of the essential oil was carried out by gas chromatography/mass chromatography. Micro-broth dilution and disc diffusion methods were used for assessment of the antimicrobial activity, and minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were evaluated., Results: The mean diameter of the growth inhibition zones in the well plate method for Dracocephalum moldavica showed that it had greater antimicrobial activity against Lactobacillus acidophilus  than others ( p Value= 0.01). Furthermore, Dracocephalum moldavica had higher antimicrobial activity than CHX. The results of MIC and MBC showed that Dracocephalum moldavica had lower antibacterial activity than CHX., Conclusion: Dracocephalum moldavica essential oil demonstrated antibacterial properties against cariogenic bacteria. Introduction: Hospitals as the main providers of healthcare services play an essential role in the management of disasters and emergencies. Nurses are one of the important and influential elements in increasing the surge capacity of hospitals. Accordingly, the present study aimed to assess the effect of surge capacity enhancement training for nursing managers on hospital disaster preparedness and response., Methods: All nursing managers employed at Motahari Hospital in Tehran took part in this interventional pre- and post-test action research study. Ultimately, a total of 20 nursing managers were chosen through a census method and underwent training in hospital capacity fluctuations. The Iranian version of the "Hospital Emergency Response Checklist" was used to measure hospital disaster preparedness and response before and after the intervention., Results: The overall hospital disaster preparedness and response score was 184 (medium level) before the intervention and 216 (high level) after the intervention. The intervention was effective in improving the dimensions of hospital disaster preparedness, including "command and control", "triage", "human resources", "communication", "surge capacity", "logistics and supply", "safety and security", and "recovery", but had not much impact on the "continuity of essential services" component., Conclusion: The research demonstrated that enhancing the disaster preparedness of hospitals can be achieved by training nursing managers using an action research approach. Encouraging their active participation in identifying deficiencies, problems, and weaknesses related to surge capacity, and promoting the adoption and implementation of suitable strategies, can enhance overall hospital disaster preparedness., (© 2024. Abstract

Laminarin, a β(1,3)-glucan, serves as a storage polysaccharide in marine microalgae such as diatoms. Its abundance, water solubility and simple structure make it an appealing substrate for marine bacteria. Consequently, many marine bacteria have evolved strategies to scavenge and decompose laminarin, employing carbohydrate-binding modules (CBMs) as crucial components. In this study, we characterized two previously unassigned domains as laminarin-binding CBMs in multimodular proteins from the marine bacterium Christiangramia forsetii KT0803 T , thereby introducing the new laminarin-binding CBM families CBM102 and CBM103. We identified four CBM102s in a surface glycan-binding protein (SGBP) and a single CBM103 linked to a glycoside hydrolase module from family 16 (GH16&#95;3). Our analysis revealed that both modular proteins have an elongated shape, with GH16&#95;3 exhibiting greater flexibility than SGBP. This flexibility may aid in the recognition and/or degradation of laminarin, while the constraints in SGBP could facilitate the docking of laminarin onto the bacterial surface. Exploration of bacterial metagenome-assembled genomes (MAGs) from phytoplankton blooms in the North Sea showed that both laminarin-binding CBM families are widespread among marine Bacteroidota. The high protein abundance of CBM102- and CBM103-containing proteins during phytoplankton blooms further emphasizes their significance in marine laminarin utilization., (© 2024 The Authors. Introduction: The preparedness of Emergency Medical Services (EMS) plays a crucial role in managing health disaster risks. This study was conducted to assess the disaster preparedness of EMS facilities in Iran, using a nationally reliable tool., Methods: A cross-sectional study was conducted in May 2021 to evaluate the disaster preparedness of EMS facilities in Iran using a national standard tool. The census sampling approach was utilized to select the samples, and descriptive statistics, as well as analytical statistics like the independent t-test and Pearson's correlation test, were used to analyze the data using SPSS-18 software., Results: The findings of this study revealed that the majority of EMS facilities in Iran possess a moderate level of preparedness in dealing with disasters. Also, the results indicate that EMS facilities have an average level of preparedness against disasters in all dimensions except for "coordination and cooperation" and "surge capacity"., Conclusion: According to this study, the majority of EMS facilities in Iran are exposed to different disasters. Furthermore, the findings indicate that while EMS facilities are generally prepared to respond to disasters at an average level, there are some important preparedness gaps in dimensions like response capacities, coordination, and cooperation. Abstract

Carrageenans are linear sulfated galactans synthesized in the Gigartinales, Rhodophyceae species with a varied range of biological properties that are of value to the pharmaceutical and cosmetic sectors. It is unknown how the fine structure of carrageenans dictates their capacity to affect molecular and cellular responses important to wound healing, or the ability to mitigate oxidative, hemostatic and inflammatory processes. Here we use specific endo-carrageenases, from the marine bacterium Zobellia galactanivorans, to produce enzymatically defined neo-series oligosaccharides from carrageenans with 3,6-anhydro-D-galactose on the non-reducing end. Further enzymatic modification of the oligosaccharides was done by treating with the 3,6-anhydro-D-galactosidases from the same bacterium which hydrolyze non-reducing end 3,6-anhydro-D-galactose moieties from neo-carrageenan oligosaccharides. Using the enzymatically produced oligosaccharides, we demonstrate binding to natural human serum antibodies and a monoclonal anti-αGal Ab (m86). The significant interactions with the Galα(1,3)Gal reactive antibodies produced by humans makes them potential potent inducers of complement-dependent reactions and attractive for therapeutic applications. Abstract

The carrageenophyte red alga Chondrus crispus produces three family 16 glycoside hydrolases (CcGH16-1, CcGH16-2, and CcGH16-3). Phylogenetically, the red algal GH16 members are closely related to bacterial GH16 homologs from subfamilies 13 and 14, which have characterized marine bacterial β-carrageenase and β-porphyranase activities, respectively, yet the functions of these CcGH16 hydrolases have not been determined. Here, we first confirmed the gene locus of the ccgh16-3 gene in the alga to facilitate further investigation. Next, our biochemical characterization of CcGH16-3 revealed an unexpected β-porphyranase activity, since porphyran is not a known component of the C. crispus extracellular matrix. Kinetic characterization was undertaken on natural porphyran substrate with an experimentally determined molecular weight. We found CcGH16-3 has a pH optimum between 7.5 and 8.0; however, it exhibits reasonably stable activity over a large pH range (pH 7.0-9.0). CcGH16-3 has a K M of 4.0 ± 0.8 μM, a k cat of 79.9 ± 6.9 s -1 , and a k cat /K M of 20.1 ± 1.7 μM -1  s -1 . We structurally examined fine enzymatic specificity by performing a subsite dissection. CcGH16-3 has a strict requirement for D-galactose and L-galactose-6-sulfate in its -1 and +1 subsites, respectively, whereas the outer subsites are less restrictive. CcGH16-3 is one of a handful of algal enzymes characterized with a specificity for a polysaccharide unknown to be found in their own extracellular matrix.
