Arthropods are often ignored or under-sampled in biodiversity and conservation assessments because of their large diversity, small size and lack of taxonomic guides. Rapid biodiversity assessment programmes have been established to assess these groups accurately. A COBRA (Conservation Oriented Biodiversity Rapid Assessment) protocol consists of an intense sampling of a habitat using the optimal combination of sampling methods. We set a basis for future protocols of measuring spider biodiversity in exotic pastures in New Zealand. Overall, 28 spider species were collected. There was variation in species discovery for each collection method, i.e. pitfall traps (86.6% of total species found), ground hand collection (95.4%), suction sampling (85.7%), and sweeping (25%). The various collection methods were complementary in species that were found. Of the four sampling methods used pitfall traps and ground hand collection were far more efficient at collecting spider species in pastures per sample. These findings are relevant for the future development of these protocols and ultimately, these tools will be used for assessing and monitoring biodiversity on farms and the impacts of farming methods. info:eu-repo/semantics/publishedVersion

The marbled cat Pardofelismarmorata is a Near Threatened small felid. The cat’s presence in Nepal is based on an anecdote. A camera trap-based study to access diversity and abundance of terrestrial mammals in eastern Nepal accumulated 3,014 camera trap days and resulted in 5,176 photographs of 17 medium-large sized mammal species. Amongst them, a marbled cat was captured at a single camera trap station in January 2018. The camera trap-capturing the marbled cat was located in the secondary forest at an altitude of 2,750 m a.s.l., dominated by free-ranging cattle close to a permanent human settlement (1.4 km) and a temporary cattle herding camp (0.4 km). This is the first photographic evidence of a marbled cat in Nepal. In this survey, we also recorded three other felid species: common leopard Pantherapardus, Asiatic golden-cat Catopumatemminckii and leopard cat Prionailurusbengalensis. We recommend detailed year-round camera trap surveys in the mid-hills of eastern Nepal along with research on adaptation of the small felids to human-dominated areas and assessment of immediate threats for preparing sound conservation management plans of the marbled cat and its sympatric species. Initiation of conservation programmes engaging local dokpa (herders) is necessary.

The common brushtail possum (Trichosurus vulpecula), introduced from Australia in the mid-nineteenth century, is an invasive species in New Zealand where it is widespread and forms the largest self-sustained reservoir of bovine tuberculosis (Mycobacterium bovis) among wild populations. Conservation and agricultural authorities regularly apply a series of population control measures to suppress brushtail possum populations. The evolutionary consequence of more than half a century of intensive population control operations on the species’ genomic diversity and population structure is hindered by a paucity of available genomic resources. This study is the first to characterise the functional content and diversity of brushtail possum liver and brain cerebral cortex transcriptomes. Raw sequences from hepatic cells and cerebral cortex were assembled into 58,001 and 64,735 transcripts respectively. Functional annotation and polymorphism assignment of the assembled transcripts demonstrated a considerable level of variation in the core metabolic pathways that represent potential targets for selection pressure exerted by chemical toxicants. This study suggests that the brushtail possum population in New Zealand harbours considerable variation in metabolic pathways that could potentially promote the development of tolerance against chemical toxicants.

This study develops multi-item scales of the financial wellbeing of customers of a major Australian bank using self-reported survey data that are matched with the customers' financial records. Using Item Response Theory (IRT) models, the study develops: First a Reported Financial Wellbeing Scale that is formed from responses to 10 questions about people's experiences and perceptions of financial outcomes, and second an Observed Financial Wellbeing Scale that is formed from five financial-record measures of customers' account balances, net spending, and payment problems. The IRT models show that each scale reliably differentiates between a wide range of outcomes and that the components within each scale have similar power to discriminate. We validate the scales by estimating Least Absolute Shrinkage and Selection Operator machine-learning models of how they correlate with other measurable characteristics. Savings habits, spending habits, credit card behavior, household income, education, difficulties with housing payments, and the use of and access to social or government support are each associated with both types of financial wellbeing.

Ōtamahua/Quail Island is an 85 ha Recreation Reserve in Lyttelton Harbour, New Zealand. It is being ecologically restored and cats, hedgehogs, rabbits, mustelids and rats have been eradicated. However, the house mouse (Mus musculus) survived two eradication attempts, a bait station operation in 2002 and an aerial operation in 2009. Data on aerial bait deposition, bait degradation, mouse resurgence and non-target mortalities were collected. Brodifacoum bait deposition in many areas was considerably lower than the prescribed rate of 8 kg/ha. Mouse sign was simultaneously detected at seven sites around the island 6 months after the aerial operation. Tails were collected from trapped mice on Quail Island, the adjacent mainland and on the stepping-stone island (King Billy) to undertake DNA analysis. Despite being classified as critically endangered, little work has been done on leopard protection in Pakistan. Once widely present throughout this region, leopards are now sparsely distributed, and possibly extinct from much of their previously recorded habitat. While leopards show morphological and genetic variation across their species range worldwide, resulting in the classification of nine different subspecies, the leopard genetic structure across Pakistan is unknown, with previous studies including only a very limited sampling. To clarify the genetic status of leopards in Pakistan we investigated the sequence variation in the subunit 5 of the mitochondrial gene NADH from 43 tissue samples and compared it with 238 sequences available from online databases. Phylogenetic analysis clearly separates the Pakistani leopards from the African and Arabian clades, confirming that leopards from Pakistan are members of the Asian clade. Furthermore, we identified two separate subspecies haplotypes within our dataset: P. p. fusca (N = 23) and P. p. saxicolor (N = 12).

Silver nanoparticles (AgNPs) from industrial use, discharged via the land application of sewage sludge, are interacting with soil biota, including earthworms. In affected organisms, excessive production of reactive oxygen species can result in lipid peroxidation, shifting the balance between oxidants and antioxidants to cause oxidative stress. We determined selected lower-tier biomarkers such as antioxidant responses and lipid peroxidation in Aporrectodea caliginosa earthworms exposed to soils spiked with AgNPs or silver nitrate (AgNO3 ). Aporrectodea caliginosa were exposed to AgNPs at 0 (control), 0.3, 3, 30, and 300 mg/kg or Ag+ (as AgNO3 ) at 0, 0.03, 0.3, 3, and 10 mg/kg in soil for 4 wk. At 1, 2, 3, and 4 wk, the activity of the antioxidant enzymes superoxide dismutase, catalase, glutathione peroxidase, glutathione S-transferase, as well as lipid peroxidation (malondialdehyde content), increased as a function of concentration, with a much larger response for Ag+ than AgNPs. Given the likelihood of ever-increasing AgNP concentrations in soil, where AgNPs can transform to ionic Ag (Ag+ ), our findings of antioxidant response to oxidative stress in a common indicator organism even at an environmentally realistic exposure concentration of 0.03 mg/kg demonstrate that AgNPs may affect soil fertility and, thus, agricultural production. Silver nanoparticles (AgNPs) are now widely used in many industry applications. Because there are no regulations on discharge limits, improper discharge of waste from these industries can lead to environmental contamination and damage to ecosystem organisms. In this study Aporrectodea caliginosa earthworms were exposed to 0 (control), 0.3, 3, 30, 300 mg/kg Ag as AgNPs and 0, 0.03, 0.3, 3, 10 mg/kg of Ag as AgNO3 in soil for 4 weeks and select biochemical and neurotoxicity studies were conducted weekly. The lipid peroxidation (measured using thiobarbituric acid reactive substances; TBARS) and activities of antioxidant enzymes (catalase, glutathione peroxidase, superoxide dismutase, glutathione S transferase, lipid peroxidation), and nerve conduction velocity (NCV) of the medial giant fibers (MGF) using a novel non-invasive electrophysiological technique were measured in earthworms weekly. The TBARS and antioxidant enzyme activities were elevated by both AgNO3 and AgNPs and this was most evident in earthworms at 4 weeks >3 > 2 > 1. In neurotoxicity studies, MGF NCV progressively decreased in A. caliginosa exposed to both AgNPs and AgNO3. The findings highlight oxidative stress and neurotoxic effects of Ag compounds on earthworms and the importance of government authorities to have legislations in place to prevent excessive soil contamination by AgNPs produced by the expanding nanotechnology industries.

New Zealand fur seals are one of many pinniped species that survived the commercial sealing of the eighteenth and nineteenth centuries in dangerously low numbers. After the enforcement of a series of protection measures in the early twentieth century, New Zealand fur seals began to recover from the brink of extinction. We examined the New Zealand fur seal populations of Banks Peninsula, South Island, New Zealand using the mitochondrial DNA control region. We identified a panmictic population structure around Banks Peninsula. The most abundant haplotype in the area showed a slight significant aggregated structure. The Horseshoe Bay colony showed the least number of shared haplotypes with other colonies, suggesting a different origin of re-colonisation of this specific colony. The effective population size of the New Zealand fur seal population at Banks Peninsula was estimated at approximately 2500 individuals. The New Zealand fur seal (Arctocephalus forsteri) is one of many pinniped species that has shown a remarkable recovery from the brink of extinction after cessation of commercial sealing during the eighteenth and nineteenth centuries. It is commonly believed that this species competes with recreational and commercial fisheries. We identified prey items using massive parallel sequencing from New Zealand fur seal faecal samples that were collected throughout the species distribution. The data support generalist feeding behaviour for this species. The diet composition showed significant geographical and inter-seasonal variation. As many as 46 species of fish and 18 species of cephalopod were identified from a single colony. The data suggest cartilaginous species (sharks, rays, and skates) constitute an important part of the New Zealand fur seal diet. Approximately 10 % of the species identified in the seal diet were of significant commercial value, which indicates some qualitative food competition between New Zealand fur seals and commercial fisheries in exploiting marine species.

All multicellular organisms host a wide diversity of microorganisms in and on their bodies, which are collectively known as their microbiome. Characterising microbial communities that inhabit different body niches in wild animals is critical to better understand the dynamics of microbiome diversityand its functional significance. The current study is the first to apply massively parallel sequencing of 16S rRNA to characterise the microbial diversity and functional content of oral microbiota in two of New Zealand’s most important invasive mammals, the omnivorous common brushtail possum (Trichosurus vulpecula) and the carnivorous stoat (Mustela erminea). In total, strains of bacteria belonging to 19 different phyla, 27 classes, 52 orders, 103 families, 163 genera and 51 known species were identified from the oral cavities of the study species. Strains of the phyla Proteobacteria, Firmicutes, Bacteroidetes, Fusobacteria, and Actinobacteria dominated the core oral microbial diversity in both species, while other taxa were comparatively less abundant. Despite invasive populations typically demonstrating limited genetic variation, intraspecific variation of the core bacterial taxa in the oral microbiota was considerable. This suggests that a complex interaction between genetic, physiological, and environmental factors determines the diversity of the study species’oral microbiome.

Rats and mice cause severe biodiversity impacts worldwide, including in New Zealand, where poisoning campaigns are undertaken on the mainland and offshore islands using cereal-based baits. However, bait stations are often irregularly serviced so wildlife managers require a bait that would remain palatable to rodents for at least 6 months in the field. One potential way to improve the field life of baits is by coating them in materials that reduce water uptake and subsequent mould growth. However, these coatings must be palatable to the rodents. This report investigates the palatability to captive Norway rats (Rattus norvegicus) and mice (Mus musculus) of both weathered and unweathered rodent food pellets coated with various polymer solutions. When compared with uncoated alternatives, fresh baits coated with shellac or polyvinyl butyral (PVB) in ethanol solvent were preferred by the rats. The use of acetone rather than ethanol solvent did not affect bait palatability. A combination of shellac and PVB reduced palatability compared with shellac alone. Weathered shellac-coated baits remained more palatable than uncoated fresh pellets to rats for up to 6 months and outperformed the PVB and combination coatings. Overall, the palatability of fresh-coated baits was lower for mice. The PVB-coated baits had lower mean palatability scores than the shellac-coated baits but none of the bait types was significantly less palatable than uncoated baits. The shellac-coated baits remained palatable to mice for up to 4 months. Shellac is recommended as a coating for multi-species rodent baits.

The New Zealand fur seal (Arctocephalus forsteri) passed through a population bottleneck due to commercial sealing during the eighteenth to nineteenth centuries. To facilitate future management options, we reconstructed the demographic history of New Zealand fur seals in a Bayesian framework using maternally inherited, mitochondrial DNA sequences. Mitogenomic data suggested two separate clades (most recent common ancestor 5000 years ago) of New Zealand fur seals that survived large-scale human harvest. Mitochondrial haplotype diversity was high, with 45 singletons identified from 46 individuals although mean nucleotide diversity was low (0.012 ± 0.0061). Variation was not constrained geographically. Analyses of mitogenomes support the hypothesis for a population bottleneck approximately 35 generations ago, which coincides with the peak of commercial sealing. Mitogenomic data are consistent with a pre-human effective population size of approximately 30,000 that first declined to around 10,000 (due to the impact of Polynesian colonization, particularly in the first 100 years of their arrival into New Zealand), and then to 100-200 breeding individuals during peak of commercial sealing.

BACKGROUND: Invasive mammalian pests have inflicted substantial environmental and economic damage on a worldwide scale. CONCLUSION: In this article we discuss recent progress and trends in a number of areas of research aimed to achieve long-term population suppression or eradication of mammalian pest species. In New Zealand, sodium fluoroacetate (1080) is used for the control of unwanted terrestrial vertebrate pests, some of which are carriers of bovine TB, and for the conservation of native flora and fauna. One key advantage of 1080 used for animal pest control is that it is biodegradable. In this short review, we focus on the persistence of 1080 in carcasses and the risk of 1080-poisoned carcasses to dogs, and the possible risk of toxicity in other non-target species via trophic transfer. The slower breakdown of 1080 in poisoned carcasses under certain conditions (e.g. cold, dry conditions), and the risks of secondary poisoning, have to be understood and managed following use of this toxin in conservation or as part of TB eradication programs. The use of first generation anticoagulants by the Department of Conservation (DOC) for rodent control has increased in recent years. This study estimates the likely hepatic persistence time of diphacinone in red deer, pigs and cattle exposed to a single sublethal dose, as well as coumatetralyl in red deer. Red deer were given an initial dose of either 1.5 mg/kg diphacinone or 8.25 mg/kg coumatetralyl, which equated to a similar quantity of commercially available bait for these anticoagulants. At these initial doses, the mean hepatic elimination half-life of diphacinone in red deer is estimated as 6.0 days whereas the mean estimated hepatic elimination half-life of coumatetralyl is estimated as 18.9 days. In pigs given an initial dose of 1.5 mg/kg, the mean hepatic elimination half-life is estimated as 12.4 days. Cattle were dosed with 1.5 mg/kg diphacinone in two similar trials. The complete mitochondrial genome of three mustelid species, stoats (Mustela erminea), weasels (Mustela nivalis) and ferrets (Mustela furo), and the New Zealand fur seal (Arctocephalus forsteri) were sequenced using direct mitochondrial DNA extraction and overlapping long PCRs. The usual 37 mammalian mitochondrial genes (13 protein coding genes, 22 t-RNA and 2 r-RNA) were identified in all four mitogenomes. The divergence of stoats from other members of the sub-family Mustelinae was dated 4.5 million years ago. The mitogenomic data were consistent with a bear-like origin of seals.

Through the efforts of research we have bucked this trend and retained and developed new tools. Three new toxins have been extensively researched and registered, namely para-aminopropiophenone (PAPP) in 2011 for stoats and feral cats; zinc phosphide for possums in 2012; and encapsulated sodium nitrite (ESN) in 2013, for possums and feral pigs. The development of PAPP and ESN, coined red blood cell toxins, developed for humaneness, represent the first new vertebrate pesticides registered for field control of mammalian pests anywhere in the world for g30 years. Research on rodenticides including diphacinone with the additive cholecalciferol (D+C) and a palatable form of norbormide continues. A New Zealand EPA application for D+C was filed in June 2015. More effective killing systems are being researched, and the first successful field trials of resetting toxin delivery devices for possum and stoat control were completed in 2013 and 2014. Improved deployment strategies, integration of humane and selective toxins, lures of greater potency, and improved killing devices aided by species’ recognition will transform ground control for endangered species protection. Sodium fluoroacetate (1080) and other important tools have been retained as new tools are emerging from a research and development pipeline. It is important for the future of New Zealand’s biodiversity that this focused research continues and we continue to learn and advance new technologies. Our goals are shifting to enable reduction in density of rat, stoat, and possum populations to zero over large scales (i.e., elimination at landscape scale), and to hold these at zero through detection and response, including the use of new technologies for perimeter control as part of barrier systems for conservation.

A new low-concentration formulation of Feracol® paste containing 0.4% concentration cholecalciferol was tested for efficacy against possums in cage trials, and against possums and rodents in a single unreplicated field trial. A new low-concentration 0.4% cholecalciferol cereal pellet was also tested in the field against possums and rodents. 20 wild-caught possums, held in individual cages, were presented with 15 g of 0.4% cholecalciferol paste. Possum body weight, amount of paste eaten and days until death were recorded. The field trial occurred on Banks Peninsula, Canterbury, from September to November 2009, using a treatment site of c. 100 ha and a non-treatment site of c. 200 ha. The treatment site was divided into two even blocks, one for paste and one for pellets. Pre- and post-monitoring were carried out using leg-hold traps for possums, and tracking tunnels for rodents. In New Zealand, around half of the lowland water bodies do not meet water quality standards, primarily due to widespread changes in land use. Riparian restoration has been occurring in New Zealand for over 30 years in an effort to minimize this impact, however, little monitoring of restoration effectiveness has been undertaken. This study evaluated the impact of riparian plantings on water quality of lowland streams in the Lake Ellesmere catchment. A paired-catchment design on four river reaches was used to compare restored riparian buffers with control sites upstream. Chemical water quality sampling was used in conjunction with a macroinvertebrate community assessment. Equivocal benefits of riparian restoration were observed, with improvements in some variables but not in others. Riparian restoration had a positive effect on water quality in terms of increasing dissolved oxygen and decreasing turbidity. However, conductivity increased at planted sites and no changes were observed in other variables. These findings may be affected by the insufficient width of all four buffers, and gaps in the buffer system contributing to poor water quality. Although variable responses were seen in invertebrate community composition, this research suggests that even narrow planted buffer strips may be effective in improving some water quality parameters. The overall mixed results present a fair assessment of the benefits of riparian restoration that may be expected within a relatively short time following vegetation establishment. Even when no baseline data have been established prior to restoration, this research showed monitoring can demonstrate the effectiveness of riparian restoration.

1. Physical structure and species composition of vegetation determine spider diversity through habitat availability. Here, we assess, for the first time, the role of plant structure on spider communities in New Zealand native alpine tussock grasslands. We investigate the specific associations between spider assemblages and plant communities and test the hypothesis that spider diversity increases with plant diversity and tussock cover. 2. Spiders were sampled using density- and abundance-activity-dependent methods. Data on physical characteristics and species composition of plants revealed gradients in vegetation, driven by distinct intra-ecosystem plant communities. Effects of vegetation on spider diversity and composition were assessed through linear mixed-effects models. Redundancy analysis was used to determine and visualise the spider species–level response to gradients in plant compositions. 3. There was a positive relationship between plant diversity and spider diversity, while the effects of tussock cover varied with spider taxa. Overall, physical structure and species composition of vegetation had effects on the abundance of a similar number of spider species and families. 4. Gradients in vegetation were matched to gradients in spider communities, whose diversity and composition varied according to their habitat preference. The family Orsolobidae was associated with wetland vegetation, and Linyphiidae with shrubs. The abundance of certain spider families and species, such as Lycosidae and Anoteropsis hilaris, was consistently affected by vegetation. 5. Environmental factors, such as soil moisture, may affect plant species composition and physical structure in tussock grasslands, which in turn determine spider assemblages. Lycosidae were identified as potential indicators of structural changes in tussock grassland plants and could be valuable for ecological monitoring in conservation management.

Bennett's wallaby (Macropus rufogriseus rufogriseus) are an introduced pest requiring control in New Zealand. Historically, sodium fluoroacetate (1080) has been used, but there is political resistance to this method of control from specific interest groups. Previous trials with captive animals have shown that Feratox® cyanide pellets are efficacious for Bennett's wallaby and the aim of this study was to field test Feratox®. A 300-ha treatment block was pre-fed non-toxic bait for 2 weeks followed by the presentation of 200-mg Feratox® pellets. Rates of faecal pellet accumulation were markedly higher in the non-treatment block following control and the Guildford score of wallaby abundance reduced from 5 to 2 in the treatment block. In total, 110 dead wallabies were found less than 50 m from the baiting sites. These results suggest that Feratox® is an effective ground-based alternative to 1080 for Bennett's wallaby control.

The Youth Risk Behavior Surveillance System (YRBSS) monitors priority health-risk behaviors among US high school students. To better understand the ramifications of changing the YRBSS from paper-and-pencil to Web administration, in 2008 the Centers for Disease Control and Prevention conducted a study comparing these two modes of administration. Eighty-five schools in 15 states agreed to participate in the study. Within each participating school, four classrooms of students in grades 9 or 10 were randomly assigned to complete the Youth Risk Behavior Survey questionnaire in one of four conditions (in-class paper-and-pencil, in-class Web without programmed skip patterns, in-class Web with programmed skip patterns, and ''on your own'' Web without programmed skip patterns). Secondary metabolites in fruit are compounds that are not directly associated with plant growth; some are directly related to plant reproductive processes, specifically seed protection and dispersion. There is a complex and species-specific interaction between these plant compounds and their avian seed dispersers. To determine whether two important secondary metabolites in wine grapes – tannins and colour pigments – might be important cues to two of the avian species that forage on wine grapes, and how species-specific this interaction might be, comparative field experiments were run with Australasian silvereyes (Zosterops lateralis) and European blackbirds (Turdus merula). Both species were offered a glucose/fructose artificial nectar with increasing concentrations of grape tannins. In a second experiment, they were offered both green and purple artificial grapes where only the colour differed. Both species showed aversion to tannins; silvereyes appeared to have greater tolerance than blackbirds of tannin concentrations above 5%. In summer no preference was shown between green and purple coloured artificial grapes, but in late autumn blackbirds took only purple grapes whereas silvereyes pecked mostly at green. Links between tannin for seed protection and colour signals to birds are discussed. Colour may cue the two species to different species-specific aspects of fruit nutritive value.

The risks of vertebrate pesticides to non-target animals are determined by intrinsic susceptibility, the toxicokinetics of the compounds used, and the degree and frequency of exposure. Metabolism and persistence studies coupled with field surveys have provided us with an improved understanding of the toxicokinetics and non-target effects of different anticoagulants. This has enabled improved choice of tools for island versus mainland use in New Zealand, and has stimulated the development of low-residue tools. Brodifacoum is a potent second-generation rodenticide used worldwide for commensal rodent control, and has been widely used to eradicate rodents from islands. The risks associated with ‘one-off’ application of baits containing second-generation anticoagulants for rodent eradication on islands are considered to be outweighed by the potential benefits to their ecosystems.
