Abstract

In humans and model species, alterations of sperm DNA methylation patterns have been reported in cases of spermatogenesis defects, male infertility and exposure to toxins or nutritional challenges, suggesting that a memory of environmental or physiological changes is recorded in the sperm methylome. The objective of this study was to ascertain if early life plane of nutrition could have a latent effect on DNA methylation patterns in sperm produced post-puberty. Holstein-Friesian calves were assigned to either a high (H) or moderate (M) plane of nutrition for the first 24 weeks of age, then reassigned to the M diet until puberty, resulting in HM and MM groups. Sperm DNA methylation patterns from contrasted subgroups of bulls in the HM (ejaculates recovered at 15 months of age; n = 9) and in the MM (15 and 16 months of age; n = 7 and 9, respectively) were obtained using Reduced Representation Bisulfite Sequencing. Both 15 and 16 months were selected in the MM treatment as these bulls reached puberty approximately 1 month after the HM bulls. Hierarchical clustering demonstrated that inter-individual variability unrelated to diet or age dominated DNA methylation profiles. While the comparison between 15 and 16 months of age revealed almost no change, 580 differentially methylated CpGs (DMCs) were identified between the HM and MM groups. Differentially methylated CpGs were mostly hypermethylated in the HM group, and enriched in endogenous retrotransposons, introns, intergenic regions, and shores and shelves of CpG islands. Furthermore, genes involved in spermatogenesis, Sertoli cell function, and the hypothalamic–pituitary–gonadal axis were targeted by differential methylation when HM and MM groups were compared at 15 months of age, reflecting the earlier timing of puberty onset in the HM bulls. Abstract

Somatic cells are present in the milk throughout lactation and consist of leukocytes and epithelial cells exfoliated from the mammary epithelium. Our objective was to determine the efficacy of using somatic cells from goat milk for dynamic studies of gene expression in the mammary gland. Over a 4-wk interval, cells were isolated from daily morning milk samples and samples taken 30 min after milking. They were characterized by direct cell counts and by flow cytometry analysis after immunostaining with antibodies directed against cytokeratin and CD45, a common leukocyte antigen. Epithelial cell counts within the morning milk ranged from 15 to 45% of total milk somatic cells. After-milking samples contained twice as many cells as did morning milk samples. The RNA was extracted from the somatic cells of both types of milk samples with equivalent efficiency (a mean of 1.2 [micro]g RNA/mL of milk). Four mRNA variants of the [alpha]-S1 casein gene were detected by Northern blot analysis and the amount of each mRNA in milk cells was related to protein concentration in milk. The comparison between mRNA from the mammary gland and from congruently collected milk cells showed that relative amounts of mRNA for each milk-protein ([alpha]-S1 and [kappa]-casein and [alpha]-lactalbumin) were conserved. In a third experiment, daily milk cell RNA preparations were extracted to assess the effect of growth hormone (GH) on mammary gene expression; four goats were separated into two groups in order to perform a switch-back design consisting of three treatment weeks: Control, GH-Control or GH-Control-GH. In this study, treatment of goats with GH increased milk yields by 5%. Throughout the control and GH treatments, the expression of the three milk-protein genes studied were highly and significantly correlated (r = 0.949 and r = 0.958, P < 0.001 for, respectively, [alpha]-S1 and [kappa]-casein and for [alpha]-S1 casein and [alpha]-lactalbumin). During GH treatment, the three milk-protein mRNA abundances increased with the same pattern. In conclusion, the opportunity to use milk somatic cells for RNA preparation and analysis provides a significant improvement over the use of biopsy samples in assessing gene activity in the mammary gland and allows easy and repetitive sampling without damaging mammary tissue. Furthermore, we propose that this method could be used to investigate the transcriptional status of the mammary gland of an animal in relation to its genotype, nutritional and pathologic status, and under influence by hormonal factors. Abstract

cDNA clones coding for rabbit prolactin were isolated from a pituitary library using a rat prolactin RNA probe. One cDNA contained 873 bases including the entire coding sequence of rabbit prolactin, its signal peptide and the 5′ and 3′ untranslated regions of 44 and 145 nucleotides respectively. The deduced amino acid sequence of the cloned prolactin cDNA presented a 93–78% identity with mink, porcine and human prolactins. The prolactin gene transcription was investigated by RT-PCR analysis in several organs of midlactating New Zealand White rabbits. The ectopic transcription of the prolactin gene was examined in more detail in the mammary gland. A strong PCR signal was detected in the mammary gland of virgin does and was also observed during pregnancy and at the beginning of lactation. This PCR signal was very weak in mid-lactating and absent in post-weaning mammary gland.

Abstract

Conférence invitée présentée au 25th Annual Meeting of the Brasilian Embryo Technology Society "The impact of reproductive biotechnologies on animal health and production" Cumbuco, Brésil, du 18 au 20 Aout 2011; Background: Since the first success in sheep, the production of viable cloned offspring by somatic cell nuclear transfer (SCNT) in various mammals has increased significantly. The incidence of pregnancy failure and fetal death, however, is still very high, whatever the species, and impairs the commercial development of this technology, even in the bovine species where the success rates are highest compared to other species. Review: In cattle, most gestation losses are initially due to abnormal implantation and poor placental development leading to fetal demise during the early post-implantation period (30 to 70 days of pregnancy). Thereafter, in continuing pregnancies, los ses usually occur in the last third of gestation and affect about 25% of the on-going pregnancies, with very large differences according to phenotype. These are currently referred to as the Large Offspring Syndrome (LOS), Large Placenta Syndrome or Abnormal Offspring Syndrome. In all cases, the placenta appears to be central to the onset of the pathology, with placentomegal y and hydrallantois being the most common features. Clinically, transabdominal ultrasound monitoring of fetal and placental development as well as the masurement of maternal plasma concentrations of pregnancy associated glycoproteins (PAG) are recommended in order to monitor the pregnancies. Humane termination of the pregnancies by Caesarian section or slaughtering of the affected animals is recommended when the pathology onset is diagnosed more than 2 weeks prior to term. Underlying mechanisms include abnormal placental vascularization, which is present early in SCNT placental development. Enzymatic response to oxidative stress is also modified. In the first trimester, several genes expressed in the trophoblast have been found to be differentially expressed between SCNT and control conceptuses, including placental lactogen (PL), the PAG, prolactin related protein-1 (PRP-1) and Dickkopf-1(DKK-1), to name a few. All these proteins are expressed in the Binucleate cells (BNC) of the trophoblast and thus, indicate that BNC function may be affected in SCNT from very early in gestation, thereby compromising placental development. Later in pregnancy, it has been shown that transplacental exchanges are disturbed, in particular those related to glucose metabolism. Moreover, endocrine function is altered compared to controls, with decreased estrogen secretion and modifications in PAG secretion, resulting in largely elevated maternal plasma concentrations. Gene expression patterns are affected, with most prominent functional effects involving cell cycle, cell signaling pathways, molecular transport, DNA replication, recombination and repair. Most of the affected genes are downregulated. Finally, many of the pathologies reported with SCNT pregnancies resemble abnormalities reported with either mutations or deletions of imprinted genes or dysregulation of imprinted gene expression, and the expression of several imprinted genes have been shown to be abnormal in SCNT placenta. Conclusions: In conclusion, pregnancy failure after SCNT is due to multiple factors affecting, implantation, placental development, morphology, vascularization, responses to oxidative stress and the epigenetic control of gene expression. If abnormal nuclear reprogramming may induce long term effects in bovine SCNT, these effects may also be due to fetal programming due to abnormal placental function and perturbed fetal development.

Abstract

Workshops are an important part of the IFPA annual meeting. At IFPA Meeting 2010 there were twelve themed workshops, six of which are summarized in this report. 1. The immunology workshop focused on normal and pathological functions of the maternal immune system in pregnancy. 2. The transport workshop dealt with regulation of ion and water transport across the syncytiotrophoblast of human placenta. 3. The epigenetics workshop covered DNA methylation and its potential role in regulating gene expression in placental development and disease. 4. The vascular reactivity workshop concentrated on methodological approaches used to study placental vascular function. 5. The workshop on epitheliochorial placentation covered current advances from in vivo and in vitro studies of different domestic species. 6. The proteomics workshop focused on a variety of techniques and procedures necessary for proteomic analysis and how they may be implemented for placental research.

Abstract

Abstract. To understand more closely the structural requirements of the LH molecule necessary to stimulate adenylate cyclase, we studied the modulation of this enzyme in partially purified plasma membranes prepared from isolated interstitial cells of rat testis submitted to oLH and to some oLH derivatives and natural analogues. The role of Mg2+was also investigated in relation to the structural modifications of oLH. Some new facts appeared in this study: 1. Methyl oLH, which exhibited the same ability as native oLH to stimulate cAMP accumulation and steroidogenesis in isolated cells, cannot induce the same level of maximal stimulation of adenylate cyclase as native oLH in plasma membranes. This phenomenon is related to the Mg2+concentration, and the differences between maximal activation induced by methyl oLH and oLH were more apparent at a free Mg2+concentration of 3.3 mmol/l than at lower concentrations. The maximal activity (in terms of native oLH) of other alkyl derivatives, such as ethyl or isopropyl oLH, on the contrary, was similar in isolated plasma membranes and in intact cells suggesting that the differential behaviour of the membranes specifically concerns the methyl derivative. 2. Guanidyl oLH and guanidyl porcine LH, which were able to induce cAMP accumulation in intact cells, did not exhibit any stimulating activity in plasma membranes. 3. Among the natural analogues, hCG and pLH are distinguished by a lower maximal activity (by comparison with oLH) particularly at high Mg2+concentration. This work shows that changes in the LH structure have an impact not only on the parameters of the adenylate cyclase complex but also on the transduction of the hormone signal and its modulation by Mg2+.
