Furthermore, pharmacokinetic, biodistribution, and liver toxicity analyses were performed in Sprague–Dawley rats, and antitumor efficacy was evaluated in Michigan Cancer Foundation-7 (MCF-7) and squamous cell carcinoma-7 (SCC-7) cells in vitro and in tumour-bearing BALB/c mice in vivo. Advanced analyses including survival rate, immunohistopathology, and terminal deoxynucleotidyl transferase dUTP nick end labelling (TUNEL) assays were performed to evaluate apoptosis in vivo.Results: The average particle size of the HePC-NLCs was 143 ± 16 nm, with a narrow PDI (0.104 ± 0.002), and the incorporation efficiency was found to be 91 ± 7%. The NLCs released HePC in a sustained manner, and this release was significantly lower than that of free drug. The in vitro hemolytic assay demonstrated a significantly reduced hemolytic potential (∼ 9%) of the NLCs compared to that of the test formulations. The HePC-NLCs demonstrated enhanced pharmacokinetic behaviour over free drug, including extended blood circulation and an abridged clearance rate in rats. Furthermore, the HePC-NLCs exhibited higher cytotoxicity than the free drug in MCF-7 and SCC-7 cells. Moreover, the HePC-NLCs showed significantly enhanced (P < 0.005) antitumor activity compared to that of the control and free drug-treated mouse groups. Tumour cell apoptosis was also confirmed, indicating the antitumor potential of the HePC-NLCs.Conclusion: These findings demonstrate the ability of NLCs as a drug delivery system for enhanced pharmacokinetic, antitumor, and apoptotic effects, most importantly when loaded with HePC.Keywords: breast cancer, miltefosine, nano lipid carriers, bioavailability, pharmacokinetics, antitumor efficacy

Abstract Background Casuarina equisetifolia (C. equisetifolia) is a woody species with many excellent features. It has natural resistance against drought, salt and saline-alkali stresses. WRKY transcription factors (TFs) play significant roles in plant response to abiotic stresses, therefore, molecular characterization of WRKY gene family under abiotic stresses holds great significance for improvement of forest trees through molecular biological tools. At present, WRKY TFs from C. equisetifolia have not been thoroughly studied with respect to their role in salt and saline-alkali stresses response. The current study was conducted to bridge the same knowledge gap. Results A total of 64 WRKYs were identified in C. equisetifolia and divided into three major groups i.e. group I, II and III, consisting of 10, 42 and 12 WRKY members, respectively. The WRKY members in group II were further divided into 5 subgroups according to their homology with Arabidopsis counterparts. WRKYs belonging to the same group exhibited higher similarities in gene structure and the presence of conserved motifs. Promoter analysis data showed the presence of various response elements, especially those related to hormone signaling and abiotic stresses, such as ABRE (ABA), TGACG (MeJA), W-box ((C/T) TGAC (T/C)) and TC-rich motif. Tissue specific expression data showed that CeqWRKYs were mainly expressed in root under normal growth conditions. Furthermore, most of the CeqWRKYs were up-regulated by NaCl and NaHCO3 stresses with few of WRKYs showing early responsiveness to both stresses while few others exhibiting late response. Although the expressions of CeqWRKYs were also induced by cold stress, the response was delayed compared with other stresses. Transgenic C. equisetifolia plants overexpressing CeqWRKY11 displayed lower electrolyte leakage, higher chlorophyll content, and enhanced tolerance to both stresses. The higher expression of abiotic stress related genes, especially CeqHKT1 and CeqPOD7, in overexpression lines points to the maintenance of optimum Na+/K+ ratio, and ROS scavenging as possible key molecular mechanisms underlying salt stress tolerance. Conclusions Our results show that CeqWRKYs might be key regulators of NaCl and NaHCO3 stresses response in C. equisetifolia. In addition, positive correlation of CeqWRKY11 expression with increased stress tolerance in C. equisetifolia encourages further research on other WRKY family members through functional genomic tools. The best candidates could be incorporated in other woody plant species for improving stress tolerance.

Background: The human voice is a prominent feature and the most susceptible to environmental changes, lifestyle, and age. Voice problems tend to be significantly under-reported and consequently less likely to be clinically treated. It is yet to be established if the cause lies in the lack of awareness or the impermanent nature of most voice disorders. Objective: To evaluate the patterns and perceptions about vocal use and misuse among young adults in Pakistan. Method: The study used a cross-sectional survey design. A convenient sampling technique was employed to collect the data from n=242 healthy young adults aged 18-25 years studying in colleges and universities of Rawalpindi and Islamabad. A 32-item survey questionnaire titled “patterns and Perceptions about voice use and Misuse” was employed, with a portion for demographic data and five sections dedicated to general awareness, self-perception about voice, and habits of vocal use and misuse. Results: The results regarding the perception and patterns of vocal use and misuse reflects that 88.3% of the participants agreed that precautionary measure would help them to avoid voice disorder. In contrast, 83.3% of the participants reported screaming and yelling as part of their vocal pattern. Participants showed a strong association with the positive attributes related to their voice such as beautiful as 82.2% of the participants reported their voice as pleasant. Conclusion: The study concluded that the outcome of participants’ perceptions and their patterns of vocal behavior were significantly atypical. Screaming and smoking were the most frequently reported response by adults but on the other hand, they did not seek professional help and did not perceive their voice as unpleasant voice. Low temperature is a limiting factor affecting plant growth and development. Casuarina equisetifolia, a typical tropical and subtropical tree important for the ecological restoration of coastal beaches, is sensitive to cold stress. By comparing cold tolerance between C. equisetifolia and Arabidopsis, we investigated the molecular basis underlying the cold sensitivity of C. equisetifolia. Transcriptomic analysis showed that the number of cold-induced genes in C. equisetifolia was significantly less than that in Arabidopsis, and notably, the response of cold-induced genes was also delayed in C. equisetifolia. Among the cold-induced genes, C-repeat binding factors (CBFs), the major transcription factors in cold acclimation in Arabidopsis, showed a delayed cold-induced expression in C. equisetifolia, despite that C. equisetifolia CBFs could restore the low temperature-sensitive phenotype of Arabidopsis cbfs triple mutants. Interestingly, some key cold-responsive genes (e.g., COR15A and COR15B) targeted by Arabidopsis CBF were absent in the C. equisetifolia genome and many cold-responsive genes in C. equisetifolia lacked the DRE element (i.e., CBF binding cis-element). Moreover, like in C. equisetifolia, many COR genes in other tropical/subtropical plants lacked the DRE element or were directly missing. These two factors could be the underlying reasons for the low-temperature sensitivity of C. equisetifolia and other tropical/subtropical plants.

Plants adjust their stomatal aperture for regulating CO2 uptake and transpiration. S-type anion channel SLAC1 (slow anion channel-associated 1) is required for stomatal closure in response to various stimuli such as abscisic acid, CO2, and light/dark transitions etc. Arabidopsis slac1 mutants exhibited defects in stimulus-induced stomatal closure, reduced sensitivity to darkness, and faster water loss from detached leaves. The global transcriptomic response of a plant with defective stimuli-induced stomatal closure (particularly because of defects in SLAC1) remains to be explored. In the current research we attempted to address the same biological question by comparing the global transcriptomic changes in Arabidopsis slac1-3 mutant and wild-type (WT) under dark, and dehydration stress, using RNA-sequencing. Abscisic acid (ABA)- and dark-induced stomatal closure was defective in Arabidopsis slac1-3 mutants, consequently the mutants had cooler leaf temperature than WT. Next, we determined the transcriptomic response of the slac1-3 mutant and WT under dark and dehydration stress. Under dehydration stress, the molecular response of slac1-3 mutant was clearly distinct from WT; the number of differentially expressed genes (DEGs) was significantly higher in mutant than WT. Dehydration induced DEGs in mutant were related to hormone signaling pathways, and biotic and abiotic stress response. Although, overall number of DEGs in both genotypes was not different under dark, however, the expression pattern was very much distinct; whereas majority of DEGs in WT were found to be downregulated, in slac1-3 majority were upregulated under dark. Further, a set 262 DEGs was identified with opposite expression pattern between WT and mutant under light–darkness transition. Amongst these, DEGs belonging to stress hormone pathways, and biotic and abiotic stress response were over-represented. To sum up, we have reported gene expression reprogramming underlying slac1-3 mutation and resultantly defective stomatal closure in Arabidopsis. Moreover, the induction of biotic and abiotic response in mutant under dehydration and darkness could be suggestive of the role of stomata as a switch in triggering these responses. To summarize, the data presented here provides useful insights into the gene expression reprogramming underlying slac1-3 mutation and resultant defects in stomatal closure.

Abstract Background Cuticular wax plays important role in protecting plants from drought stress. In Arabidopsis WRI4 improves drought tolerance by regulating the biosynthesis of fatty acids and cuticular wax. Cyperus esculentus (yellow nutsedge) is a tough weed found in tropical and temperate zones as well as in cooler regions. In the current study, we report the molecular cloning of a WRI4-like gene from Cyperus esculentus and its functional characterization in Arabidopsis. Results Using RACE PCR, full-length WRI-like gene was amplified from yellow nutsedge. Phylogenetic analyses and amino acid comparison suggested it to be a WRI4-like gene. According to the tissue-specific expression data, the highest expression of WRI4-like gene was found in leaves, followed by roots and tuber. Transgenic Arabidopsis plants expressing nutsedge WRI4-like gene manifested improved drought stress tolerance. Transgenic lines showed significantly reduced stomatal conductance, transpiration rate, chlorophyll leaching, water loss and improved water use efficiency (WUE). In the absence of drought stress, expression of key genes for fatty acid biosynthesis was not significantly different between transgenic lines and WT while that of cuticular wax biosynthesis genes was significantly higher in transgenic lines than WT. The PEG-simulated drought stress significantly increased expression of key genes for fatty acid as well as wax biosynthesis in transgenic Arabidopsis lines but not in WT plants. Consistent with the gene expression data, cuticular wax load and deposition was significantly higher in stem and leaves of transgenic lines compared with WT under control as well as drought stress conditions. Conclusions WRI4-like gene from Cyperus esculentus improves drought tolerance in Arabidopsis probably by promoting cuticular wax biosynthesis and deposition. This in turn lowers chlorophyll leaching, stomatal conductance, transpiration rate, water loss and improves water use efficiency under drought stress conditions. Therefore, CeWRI4-like gene could be a good candidate for improving drought tolerance in crops.

Casuarina equisetifolia is widely used in agroforestry plantations for soil stabilization, ecosystem rehabilitation, reclamation, and coastal protection. Moreover, C. equisetifolia has remarkable resistance to typhoons, desert, low soil fertility, drought, and salinity, but not cold. Therefore, it is significant to breed high-quality Casuarina varieties to improve the tolerance and adaptability to cold weather by molecular techniques. The establishment of a rapid and efficient callus induction and regeneration system via tissue culture is pre-requisite for the genetic transformation of C. equisetifolia, which is so far lacking. In this study, we reported an efficient and rapid regeneration system using stem segment explants, in which callus induction was found to be optimal in a basal medium supplemented with 0.1 mg⋅L–1 TDZ and 0.1 mg⋅L–1 NAA, and proliferation in a basal medium containing 0.1 mg⋅L–1 TDZ and 0.5 mg⋅L–1 6-BA. For bud regeneration and rooting, the preferred plant growth regulator (PGR) in basal medium was 0.5 mg⋅L–1 6-BA, and a combination of 0.02 mg⋅L–1 IBA and 0.4 mg⋅L–1 IAA, respectively. We also optimized genetic a transformation protocol using Agrobacterium tumefaciens harboring the binary vector pCAMBIA1301 with β-glucuronidase (GUS) as a reporter gene. Consequently, 5 mg L–1 hygromycin, 20 mg L–1 acetosyringone (As), and 2 days of co-cultivation duration were optimized to improve the transformation efficiency. With these optimized parameters, transgenic plants were obtained in about 4 months. Besides that, Agrobacterium rhizogenes-mediated transformation involving adventitious root induction was also optimized. Our findings will not only increase the transformation efficiency but also shorten the time for developing transgenic C. equisetifolia plants. Taken together, this pioneer study on tissue culturing and genetic transformation of C. equisetifolia will pave the way for further genetic manipulation and functional genomics of C. equisetifolia.

Synthetic dyes are widely used as colorant compounds in various industries for different purposes. Among all the dyestuffs, azo dyes constitute the largest and the most used class of dyes. These dyes and their intermediate products are common contaminants of ground water and soil in developing countries. Biological methods have been found to be promising for the treatment and degradation of these compounds. In the present study, we focused on the biological removal of azo dyes (Reactive orange 16 and Reactive black 5) under aerobic conditions using an indigenous bacterial strain isolated from contaminated industrial areas. The bacterial isolate was identified as Bacillus cereus strain ROC. Degradation experiments under agitation with both free and immobilized cells indicates that this strain degrades both azo- dyes in 5 days. The immobilized cells were more proficient than their free cell counterparts. The toxicity of the biotransformation products formed after decolorization were assessed by conducting bacteriotoxic and phytotoxic assays. All the toxicity assays indicate that the dyes’ degraded products were non-toxic in nature, as compared to the dyes themselves. The kinetics of the azo dyes’ degradation was also studied at various initial concentration ranges from 50 mg/L to 250 mg/L by growth independent kinetic models. Zero-order kinetics were fit to the experimental data, producing values of least squares regression (R2) greater than 0.98, which indicates that the bacterial strain degrades both dyes by co-metabolism rather than utilizing them as sole energy source. These results indicate that the Bacillus cereus ROC strain has great potential to degrade dye-contaminated water and soil.

The present study reports the synthesis, photocatalytic decolorization of reactive black 5 dye and phytotoxicity of graphene quantum dots (GQDs) and iron co-doped TiO2 photocatalysts via modified sol gel method. GQDs were synthesized by direct pyrolysis of citric acid (CA). Scanning electron microscopy (SEM) and energy dispersion spectroscopy (EDS), Raman spectroscopy, atomic force microscopy (AFM), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), diffuse reflectance spectroscopy (DRS), Brunauer–Emmett–Teller (BET) and photoluminescence spectroscopy (PL) were used to determine the physicochemical properties of the best performing photocatalysts. The results indicated improved physicochemical properties of GQD-0.1Fe-TiO2-300 with root mean square roughness (Rz) (33.82 nm), higher surface area (170.79 m2 g−1), pore volume (0.08 cm3 g−1), and bandgap (2.94 eV). Moreover, GQD-0.1Fe co-doping of TiO2 greatly improved the photocatalytic decolorization efficiency for RB5 dye. The photocatalytic reaction followed the pseudo first order reaction with gradual decrease in Kapp values for increment in RB5 concentration. The KC value was obtained as 2.45 mg L−1 min−1 while the KLH value was 0.45 L mg−1 indicating the heterogeneous reaction system followed the Langmuir-Hinshelwood isotherm and simultaneously occurring adsorption and photocatalytic processes. Photocatalytic reaction mechanism studies exhibited the holes and OH radicals as the main active species in the GQD-0.1Fe-TiO2-300 responsible for the decolorization of RB5. The proposed reaction pathway showed that both Fe-TiO2 and GQDs play important role in generation of electrons and holes. Additionally, GQD-0.1Fe-TiO2-300 were durable up to four cycles. Phytotoxicity assay displayed that treated water and best performing photocatalysts had no effect on Lycopersicon esculentum seed germination. Therefore, the proposed system can pave a viable solution for safe usage of dye loaded wastewater and effluent for irrigation after treatment.

Background Photosynthesis is a key process in plants that is compromised by the oxygenase activity of Rubisco, which leads to the production of toxic compound phosphoglycolate that is catabolized by photorespiratory pathway. Transformation of plants with photorespiratory bypasses have been shown to reduce photorespiration and enhance plant biomass. Interestingly, engineering of a single gene from such photorespiratory bypasses has also improved photosynthesis and plant productivity. Although single gene transformations may not completely reduce photorespiration, increases in plant biomass accumulation have still been observed indicating an alternative role in regulating different metabolic processes. Therefore, the current study was aimed at evaluating the underlying mechanism (s) associated with the effects of introducing a single cyanobacterial glycolate decarboxylation pathway gene on photosynthesis and plant performance. Methods Transgenic Arabidopsis thaliana plants (GD, HD, OX) expressing independently cyanobacterial decarboxylation pathway genes i.e., glycolate dehydrogenase, hydroxyacid dehydrogenase, and oxalate decarboxylase, respectively, were utilized. Photosynthetic, fluorescence related, and growth parameters were analyzed. Additionally, transcriptomic analysis of GD transgenic plants was also performed. Results The GD plants exhibited a significant increase (16%) in net photosynthesis rate while both HD and OX plants showed a non-significant (11%) increase as compared to wild type plants (WT). The stomatal conductance was significantly higher (24%) in GD and HD plants than the WT plants. The quantum efficiencies of photosystem II, carbon dioxide assimilation and the chlorophyll fluorescence-based photosynthetic electron transport rate were also higher than WT plants. The OX plants displayed significant reductions in the rate of photorespiration relative to gross photosynthesis and increase in the ratio of the photosynthetic electron flow attributable to carboxylation reactions over that attributable to oxygenation reactions. GD, HD and OX plants accumulated significantly higher biomass and seed weight. Soluble sugars were significantly increased in GD and HD plants, while the starch levels were higher in all transgenic plants. The transcriptomic analysis of GD plants revealed 650 up-regulated genes mainly related to photosynthesis, photorespiratory pathway, sucrose metabolism, chlorophyll biosynthesis and glutathione metabolism. Conclusion This study revealed the potential of introduced cyanobacterial pathway genes to enhance photosynthetic and growth-related parameters. The upregulation of genes related to different pathways provided evidence of the underlying mechanisms involved particularly in GD plants. However, transcriptomic profiling of HD and OX plants can further help to identify other potential mechanisms involved in improved plant productivity.

Summary Photovoltaic (PV) energy is highly promising because of its renewable, green, and environment‐friendly nature. In this article, the design and analysis of an isolated PV system using a push‐pull converter with a fuzzy logic‐based maximum power point tracking (MPPT) algorithm is presented. Furthermore, DC‐DC converters, along with intelligent controllers fed with MPPT algorithms, are used to ensure the maximum extraction of incident energy. The proposed methodology utilizes fuzzy logic MPPT techniques based on an isolated push‐pull boost converter to optimize the power output of PV modules, as well as to achieve isolation and high DC gain for DC/AC inversion. This work also presents a single‐phase inverter with fuzzy logic close loop control analysis with LCL filter design. A Canadian solar panel of 250 W is assumed in this research work, which has an open circuit voltage 59.9 V, short circuit current 5.49 A at 25°C temperature, and 1000 W/m2 irradiance. The voltages are tracked, through the MPPT algorithm. These voltages represent a boost to 340 V DC through push‐pull boost converter and are inverted up to 220 V AC through fuzzy logic voltage source inverter. In addition, a unipolar switching technique is used to remove the total harmonic distortion under linear load. The proposed methodology is simulated in MATLAB/Simulink. The simulation results verify that the proposed methodology can efficiently track the MPPT. Finally, the hardware prototype of the proposed system has been experimentally validated.

Background Salinity is one of the most lethal abiotic stresses which affect multiple aspects of plant physiology. Natural variations in plant germplasm are a great resource that could be exploited for improvement in salt tolerance. Eruca sativa (E. sativa) exhibits tolerance to abiotic stresses. However, thorough evaluation of its salt stress tolerance and screening for traits that could be reliably applied for salt tolerance needs to be studied. The current study was designed to characterize 25 E. sativa accessions, originating from diverse geographical regions of Pakistan, for the salt stress tolerance. Methods Salt stress (150 mM NaCl) was applied for 2 weeks to the plants at four leaf stage in hydroponics. Data of the following morpho-physiological traits were collected from control and treated plants of all the accessions: root length (RL), shoot length (SL), plant height (PH), leaf number (LN), leaf area (LA), fresh weight (FW), dry weight (DW), chlorophyl content (SPAD), electrolyte leakage (EL), relative water content (RWC), gas exchange parameters and mineral ion content. Salt tolerance was determined based on membership function value (MFV) of the tested traits. Results Compared with control, the salt-stressed group had significantly reduced mean SL, RL, PH, LN, LA, FW, DW and SPAD. NaCl treatment triggered a slight increase in EL in few accessions. Mean RWC of control and treated groups were not significantly different although few accessions exhibited variation in this trait. Salt stress caused a significant reduction in photosynthesis rate (PR), transpiration rate (TR) and stomatal conductance (SC) but intercellular CO2 (Ci) was not significantly different between control and treated groups. Compared with control, the salt-stressed plants accumulated significantly higher Na+, K+ and Ca2+ while significantly lower Mg2+. K+/Na+ ratio was significantly decreased in salt-stressed plants compared with control. Importantly, significant inter-accession variations were found for all the tested traits. The principal component analysis identified SL, RL, PH, LN, LA, FW, DW and PR as the most significant traits for resolving inter-accession variability. Based on MFV of the tested traits, accessions were categorized into five standard groups. Among 25 accessions, one accession was ranked as highly tolerant, four as tolerant while 15 accessions were ranked as moderately tolerant. Of the remaining five accessions, four were ranked as sensitive while one accession as highly sensitive. Conclusion E. sativa accessions were found to exhibit significant genetic diversity in all the tested traits. A few most significant traits for dissecting the genetic variability were identified that could be used for future large-scale germplasm screening in E. sativa. Salt tolerant accessions could be a good resource for future breeding programs aiming to improve salt stress tolerance.

Carbamates are synthetic pesticides, extensively used throughout the world due to their broad specificity against various insect pests. However, their enormous and inadequate use have made them a potential threat to the environment. At low temperature, degradation of carbamates becomes difficult mainly because of low biological activity. In the present study, we isolated a bacterial strain from a low temperature climate where the N-methylated carbamates are used for crop protection. The bacterium, was identified as Pseudomonas plecoglossicida strain (TA3) by 16S rRNA analysis. Degradation experiments with both free and immobilized cells in minimal salt medium indicated that the strain TA3 utilized carbaryl, carbofuran and aldicarb as both carbon and nitrogen source. TA3 can grow well at 4 °C and demonstrated the ability to degrade three carbamates (50 μgml−1) at low temperature. The immobilized cells were found more efficient than their free cells counter parts. Immobilized cells has ability to degrade 100% of carbamates at 30 °C while 80% at 4 °C but incase of their free cells counter parts the efficiency to degrade carbamates was less which was 60% at 4 °C and 80% at 30 °C. TA3 free cellsextract also depicted high activity against all the three carbamates even at 4 °C indicating a possible enzymatic mechanism of degradation.

Programmed cell death (PCD) is a process intended for the maintenance of cellular homeostasis by eliminating old, damaged, or unwanted cells. In plants, PCD takes place during developmental processes and in response to biotic and abiotic stresses. In contrast to the field of animal studies, PCD is not well understood in plants. Calcium (Ca2+) is a universal cell signaling entity and regulates numerous physiological activities across all the kingdoms of life. The cytosolic increase in Ca2+ is a prerequisite for the induction of PCD in plants. Although over the past years, we have witnessed significant progress in understanding the role of Ca2+ in the regulation of PCD, it is still unclear how the upstream stress perception leads to the Ca2+ elevation and how the signal is further propagated to result in the onset of PCD. In this review article, we discuss recent advancements in the field, and compare the role of Ca2+ signaling in PCD in biotic and abiotic stresses. Moreover, we discuss the upstream and downstream components of Ca2+ signaling and its crosstalk with other signaling pathways in PCD. The review is expected to provide new insights into the role of Ca2+ signaling in PCD and to identify gaps for future research efforts.

Background: Chronic myeloid leukaemia (CML) is a type of blood cancer that begins in the hematopoietic stem cells. It is primarily characterized by a specific chromosomal aberration, the Philadelphia chromosome. While the fusion gene is a major contributor to CML, several other genes including ADGRE2, that are reported as highly expressed in hematopoietic stem cells and could be utilized as a therapeutic marker in leukemic patients are implicated in the disease's progression. Until recently, little research had been conducted to identify single nucleotide polymorphisms (SNPs) associated with CML. Therefore, this study aims to investigate the influence of non-synonymous variants on the structure and function of the gene encoding adhesion G protein-coupled receptor E2, ADGRE2, and to evaluate their association with CML and its clinical and pathological characteristics., Methods: Non-synonymous SNPs of ADGRE2 were retrieved from the ENSEMBL, COSMIC, and gnomAD genome browsers, and the pathogenicity of deleterious variants was assessed using several established computational tools, including SIFT, CADD, REVEL, PolyPhen, and MetaLR., Results: Various in silico analyses explored the impact of damaging SNP on the function, stability, and structure of EGF-like modules containing mucin-like hormone receptor-like2 (EMR2) protein encoded by the ADGRE2 gene. Genotype analysis was performed on collected blood samples, revealing that altered genotype TT of variant rs765071211 (C/T) was associated significantly with CML patients compared to the control. Objectives . This research investigated the performance properties of protective gloves alongside improvement in thermo-physiological comfort properties. Methods . Knitted gloves prepared from filament stainless steel, filament Kevlar, staple spun Kevlar and filament glass were used in the core, while 100% viscose rayon and 50/50% polyester/viscose rayon were used in the sheath. Gloves were tested for cut resistance, tear resistance, puncture resistance and abrasion resistance as the prime focus, and thermo-physiological comfort properties were also tested. In this research, a multi-response optimization technique, i.e., principal component analysis, was applied to identify the best yarn combination for gloves based on the aforementioned properties. Thermal images were also taken in constant ambient conditions for temperature distribution maps across the hand's surface. Results . All of the results were evaluated statistically with analysis of variance, and concluded that the effects of the core yarn on thermo-physiological properties were less significant. Conclusion . The results revealed that samples having dual-core yarn exhibited better in terms of overall properties. The sample having dual-core filament Kevlar and staple spun Kevlar ranked the best. In addition, developed samples exhibited better comfort properties than the control sample.

This work investigates the effect of varying the knitting structure and stitch length (SL) on various thermo-physiological and ergonomic comfort properties of the occupational graduated compression socks. Thermo-physiological comfort, ergonomic comfort and dimensional stability of theses stockings were analysed in a comparative manner. Obtained results were evaluated statistically using the technique of analysis of variance (ANOVA). A Fisher's multiple comparison test was commissioned to analyze the relationship between the alteration of stitch length (SL) on various utility functions and properties desired in the occupational compression socks. In order to examine whether the difference of stitch length is significant, p values were determined. Further the influence of knitting structures e.g., plain, 2 × 2 Rib and 1 × 3 Rib was analysed on the selected properties. The interactive effect of both stitch length (SL) and knitting structure was studied using statistical techniques. It was concluded that knitting structure has a stronger impact on thermo-physiological and ergonomic comfort properties. Results showed a significant variation in thermo-physiological and ergonomic comfort by altering stitch length by means of the statistical analysis. An innovative approach for the manufacturers has been developed for optimizing performance in compression stockings. Limited efficiency, lower durability, moisture absorbance, and pest/fungal/bacterial interaction/growth are the major issues relating to porous nonwovens used for acoustic and thermal insulation in buildings. This research investigated porous nonwoven textiles composed of recycled cotton waste (CW) fibers, with a specific emphasis on the above-mentioned problems using the treatment of silicon coating and formation of nanofibers via facile-solution processing. The findings revealed that the use of an economic and eco-friendly superhydrophobic (contact angle higher than 150°) modification of porous nonwovens with silicon nanofibers significantly enhanced their intrinsic characteristics. Notable improvements in their compactness/density and a substantial change in micro porosity were observed after a nanofiber network was formed on the nonwoven material. This optimized sample exhibited a superior performance in terms of stiffness, surpassing the untreated samples by 25-60%. Additionally, an significant enhancement in tear strength was observed, surpassing the untreated samples with an impressive margin of 70-90%. Moreover, the nanofibrous network of silicon fibers on cotton waste (CW) showed significant augmentation in heat resistance ranging from 7% to 24% and remarkable sound absorption capabilities. In terms of sound absorption, the samples exhibited a performance comparable to the commercial standard material and outperformed the untreated samples by 20% to 35%. Enhancing the micro-roughness of fabric via silicon nanofibers induced an efficient resistance to water absorption and led to the development of inherent self-cleaning characteristics. The antibacterial capabilities observed in the optimized sample were due to its superhydrophobic nature. These characteristics suggest that the proposed nano fiber-treated nonwoven fabric is ideal for multifunctional applications, having features like enhanced moisture resistance, pest resistance, thermal insulation, and sound absorption which are essential for wall covers in housing.

Herein we designed, optimized, and characterized the Metformin Hydrochloride Transethosomes (MTF-TES) and incorporate them into Chitosan gel to develop Metformin Hydrochloride loaded Transethosomal gel (MTF-TES gel) that provides a sustained release, improved transdermal flux and improved antidiabetic response of MTF. Design Expert® software (Ver. 12, Stat-Ease, USA) was applied for the statistical optimization of MTF-TES. The formulation with Mean Particle Size Distribution (MPSD) of 165.4 ± 2.3 nm, Zeta Potential (ZP) of -21.2 ± 1.9 mV, Polydispersity Index (PDI) of 0.169 ± 0.033, and MTF percent Entrapment Efficiency (%EE) of 89.76 ± 4.12 was considered to be optimized. To check the chemical incompatibility among the MTF and other formulation components, Fourier Transform Infrared (FTIR) spectroscopy was performed and demonstrated with no chemical interaction. Surface morphology, uniformity, and segregation were evaluated through Transmission Electron Microscopy (TEM). It was revealed that the nanoparticles were spherical and round in form with intact borders. The fabricated MTF-TES has shown sustained release followed by a more pronounced effect in MTF-TES gel as compared to the plain MTF solution (MTFS) at a pH of 7.4. The MTF-TES has shown enhanced permeation followed by MTF-TES gel as compared to the MTFS at a pH of 7.4. In vivo antidiabetic assay was performed and results have shown improved antidiabetic potential of the MTF-TES gel, in contrast to MTF-gel. Conclusively, MTF-TES is a promising anti-diabetic candidate for transdermal drug delivery that can provide sustained MTF release and enhanced antidiabetic effect.

This paper investigates the development of fabric materials using several blends of inherently fire-resistant (FR) fibers and various knitted structures. The samples are evaluated with respect to their performance and comfort-related properties. Inherently fire-resistant fibers, e.g., Nomex, Protex, carbon and FR viscose, were used to develop different structures of knitted fabrics. Cross-miss, cross-relief, and vertical tubular structures were knitted by using optimum fiber blend proportions and combinations of stitches. Several important aspects of the fabric samples were investigated, e.g., their physical, mechanical and serviceability performance. Thermo-physiological and tactile/touch-related comfort properties were evaluated in addition to flame resistance performance. An analysis of mechanical performance indicated that the knitted structure has a significant influence on the tensile strength, bursting strength and pilling resistance. The cross-relief structure proved to be the strongest followed by the cross-miss and vertical tubular structures. The FR station suits made from 70:30 Protex/Nomex exhibited the best combination of tensile and bursting strength; therefore, this material is recommended for making a stable and durable station suit. Interestingly, it was also concluded from the experimental study that knitted samples with a cross-relief structure exhibit the best fire-resistance performance. Fiber blends of 70:30 Protex/Nomex and 70:30 Nomex/carbon were found to be optimum in terms of overall performance. The best flame resistance was achieved with Nomex:carbon fiber blends. These results were confirmed with vertical flammability tests, TGA, DTGA and cone calorimetry analysis. The optimization of blend composition as well as knitting structure/architecture is a crucial finding toward designing the best FR station suit in terms of mechanical, dimensional, thermal, thermo-physiological and flame resistance performance.

This paper presents an experimental study on the influence of alkaline environments on natural fibers of plant and mineral origin in concretes. The durability of concrete-based composite materials is influenced by the properties of the reinforcing fiber, and the serviceability of concrete is dependent on its durability. The aim of the present study is to investigate the strength, weight loss %, and surface degradation of jute, sugarcane, coconut, sisal, as well as basalt fibers through an accelerated aging method when used as reinforcements in concrete. The samples were immersed in an alkaline environment of sodium and calcium hydroxide at two different levels of pH for one week. Further, the fibers were immersed in NaOH and Ca(OH) 2 solutions of 1 M, 2 M, 4 M, and 6 M concentrations for 48 h in order to investigate the gradual effect of an alkaline environment on the mechanical properties of the fiber. It was concluded that the weight loss % was greatest for jute fibers when used in concrete composite, while there was no significant effect on the basalt fiber samples. The strength of jute fiber in the concrete sample was also most severely affected by the aging process, compared to other fibers. The strength of basalt fibers in a concrete composite was least affected by the aging process. In some cases, the sisal fiber sample showed an increase in fiber tenacity after the aging process due to fibrillation, which might have increased the interfacial area. The fiber microstructure before and after the aging was evaluated through the use of scanning electron microscopy (SEM). SEM analyses of different fibers were carried out to investigate surface degradation. The fiber pull-out strength was found to be the greatest for basalt fiber, followed by jute and sisal. This is indicative of the excellent adhesion of such fibers with cement in a concrete composite. In these cases, the use of sisal fiber results in defibrillation and increased specific surface area. Sugarcane and coconut fibers ruptured due to their inherent weakness and provided only a small increment in the mechanical performance of the concrete. Basalt fiber-reinforced concrete offered the greatest compressive strength, followed by jute and sisal. These observations provide crucial information regarding the durability and aging of natural fiber-reinforced concrete.

Compression stockings/socks are one of the most essential materials to treat vascular disorders in veins. However, the comfort of wearing such stockings over prolonged period of time is a major problem. There is limited research in the area of comfort optimization while retaining the compressional performance. The current work is carried out with an aim to determine the optimum level of the input factors e.g., knitting structure, plaiting yarn linear density and main yarn linear density for achieving desired stretch recovery percentage and thermo-physiological comfort properties of compression socks used in treatment of vascular disorders. Their optimum combination was determined by using Taguchi based techniques for order of preference by similarity to ideal solution i.e., TOPIS. In this study, thickness, areal density, air permeability, thermal resistance, over all moisture management capacity (OMMC), stretch and recovery % were optimized simultaneously by using Taguchi-TOPSIS method. The results showed that linear density of plaiting and main yarn has significant influence on all the comfort related properties for compression stockings/socks. The optimum sample had linear density 20 denier for Lycra covered by 70 denier of nylon 66 in the plaiting yarn. It also suggested 120 denier nylon 66 in the main yarn knitted into a plain single jersey structure. The percentage contribution of the factors i.e., structure, plaiting yarn linear density and main yarn linear density was obtained by using ANOVA which are 7%, 31% and 42% respectively. It is worth mentioning that in case of compression stockings, the main yarn linear density has more significant effect on comfort properties as compared to other independent parameters. The results were verified by experiment, and the accuracy was relatively high (maximum error 8.533%). This study helped to select suitable knit structure with the change of linear densities of plaiting yarn and main yarn for comfortable compression stocking/sock and will fulfill the potential requirement for treatment of venous/vascular disorders. The novel methodology involving TOPSIS method helped in analyzing the cumulative contribution of the input parameters to achieve optimum compression as well as comfort performance. This modern approach is based on contemporary scientific principles and statistical approximations. The paper deals with research focused on the use of fillers in the field of polymeric materials produced by additive technology SLA (stereolithography). The aim of the research is to evaluate 3D printing parameters, the mechanical properties (tensile strength, hardness), and the interaction of individual phases (polymer matrix and filler) in composite materials using SEM analysis. The tested fillers were cotton flakes and ground carbon fibres in different proportions. For the photosensitive resins, the use of cotton flakes as filler was found to have a positive effect on the mechanical properties not only under static but also under cyclic loading, which is a common cause of material failure in practice. The cyclic stress reference value was set at an amplitude of 5-50% of the maximum force required to break the pure resin in a static tensile test. A positive effect of fillers on the cyclic stress life of materials was demonstrated. The service life of pure resin was only 168 ± 29 cycles. The service life of materials with fillers increased to approximately 400 to 540 cycles for carbon fibre-based fillers and nearly 1000 cycles for cotton flake-based fillers, respectively. In this paper, new composite materials suitable for the use of SLA additive manufacturing techniques are presented. Research demonstrated the possibilities of adding cotton-based fillers in low-cost, commercially available resins. Furthermore, the importance of material research under cyclic loading was demonstrated.

The current work is focused on numerical and experimental studies of woven fabric composites modified by hybridisation with biological (cellulosic) filler materials. The mechanical performance of the composites is characterized under tensile, bending and impact loads and the effect of hybridisation is observed with respect to pure and nonhybrid composites. Numerical models are developed using computational tools to predict mechanical performance under tensile loading. The computational prediction results are compared and validated with relevant experimental results. This research is aimed at understanding the mechanical performance of basalt-epoxy composites partially reinforced with micro-/nano-sized bio-fillers from cellulose and intended for various application areas. Different weave structures, e.g., plain, twill, matt, etc., were investigated with respect to the mechanical properties of the hybrid composites. The effects of hybridizing with cellulose particles and different weave patterns of the basalt fabric are studied. In general, the use of high-strength fibres such as basalt along with cellulosic fillers representing up to 3% of the total weight improves the mechanical performance of the hybrid structures. The thermomechanical performance of the hybrid composites improved significantly by using basalt fabric as well as by addition of 3% weight of cellulosic fillers. Results reveal the advantages of hybridisation and the inclusion of natural cellulosic fillers in the hybrid composite structures. The material developed is suitable for high-end applications in components for construction that demand advanced mechanical and thermomechanical performance. Furthermore, the inclusion of biodegradable fillers fulfills the objectives of sustainable and ecological construction materials.

Background: Aplastic anaemia is a rare bone marrow failure syndrome and is defined by pancytopenia associated with a hypo-cellular bone marrow with no increase in reticulin and the absence of any abnormal infiltrate. The objective of the study was to determine the frequency of Aldehyde Dehydrogenase type 2 (ALDH2) deficiency in patients with Aplastic Anaemia and investigate its correlation with patient and disease characteristics. It was a descriptive cross-sectional study conducted at Armed Forces Bone Marrow Transplant Centre Rawalpindi from 01-08-2022-01-02-2023, over 6 months., Methods: A total of 56 patients who were diagnosed with aplastic anaemia during this period, fulfilling inclusion criteria were enrolled. Patients were genotyped as GG (homozygous) and GA (heterozygous). GG had normal ALDH2, while GA were patients with ALDH2 deficiency. Data was collected on the patient's demographics, type and severity of anaemia, type of hematopoietic stem cell transplant (HSCT) and frequency of ALDH2 deficiency. Results were analyzed for ALDH2 deficiency and its correlation with patient and disease characteristics was investigated., Results: A total of 56 patients were included in the study. The median age of the patients was 28 years (20-39). According to the type of aplastic anaemia, 2 (3.6%) had Fanconi anaemia and 54 (96.4%) had acquired aplastic anaemia. In our study, 18 (32.1%) patients had undergone HSCT while the remaining 38 (67.9%) could not undergo HSCT. The frequency of the presence of ALDH2 deficiency was 2 (3.6%). There was no statistically significant correlation between the frequency of ALDH2 deficiency with variables like gender, age distribution, type of aplastic anaemia, the severity of aplastic anaemia and hematopoietic stem cell transplant., Conclusions: We concluded from our study the frequency of ALDH2 was rare in patients with aplastic anaemia. There was no statistically significant correlation between the frequency of ALDH2 deficiency with variables like gender, age distribution, type of aplastic anaemia, the severity of aplastic anaemia and hematopoietic stem cell transplant.

For a sustainable environment and to tackle the pollution problem, industrial wastes can be used in concrete composite materials. This is especially beneficial in places prone to earth quack and lower temperature. In this study, five different types of waste fibres such as polyester waste, rubber waste, rock wool waste, glass fibre waste and coconut fibre waste were used as an additive in 0.5% 1%, and 1.5% by mass in concrete mix. Seismic performance related properties of the samples were examined through evaluation of compressive strength, flexural strength, impact strength, split tensile strength, and thermal conductivity. Results showed that, impact strength of the concrete significantly improved by the addition of fibre reinforcement in concrete. Split tensile strength and flexural strength were significantly reduced. Thermal conductivity was also influenced by addition of polymeric fibrous waste. Microscopic analysis was performed to examine the fractured surfaces. In order to get the optimum mix ratio, multi response optimization technique was used to determine the desired level of impact strength at an acceptable level of other properties. Rubber waste was found to be the most attractive option followed by coconut fibre waste for the seismic application of concrete. The significance and percentage contribution of each factor was obtained by Analysis of variance ANOVA (α = 0.05) and pie chart which showed that Factor A (waste fibre type) is the main contributor. Confirmatory test was done on optimized waste material and their percentage. The order preference similarity to ideal solution (TOPSIS) technique was used for developed samples to obtain solution (sample) which is closest to ideal as per given weightage and preference for the decision making. The confirmatory test gives satisfactory results with error of 6.68%. Cost of reference sample and waste rubber reinforced concrete sample was estimated, which showed that 8% higher volume was achieved with waste fibre reinforced concrete at approximately same cost as pure concrete. Concrete reinforced with recycled fibre content is potentially beneficial in terms of minimizing resource depletion and waste. The addition of polymeric fibre waste in concrete composite not only improves seismic performance related properties but also reduces the environmental pollution from waste material which has no other end use., (© 2023. Current research deals with thermoplastic polyamide (PA6)-based composites reinforced with basalt and Kevlar fabrics. Hybrid composites were developed by altering the stacking sequence of basalt and two kinds of Kevlar fabrics. Pure-basalt- and pure-Kevlar-based samples were also developed for comparison purposes. The developed samples were evaluated with respect to mechanical and thermomechanical properties. Mechanical tests, e.g., tensile, flexural, and impact strength, were conducted along with thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC) to ascertain the load-bearing and high-temperature stability of the hybrid composite samples vis-à-vis pure-basalt- and Kevlar-based samples. Scanning electron microscopy (SEM) was carried out to study the nature of fracture and failure of the composite samples. The pure-basalt-based PA6 thermoplastic composites exhibited the best mechanical performance. Hybridization with basalt proved to be beneficial for improving the mechanical performance of the composites using Kevlar fabrics. However, a proper stacking sequence and density of Kevlar fabric has to be selected. The thermogravimetric analysis showed minimal weight loss for basalt-based composites. Furthermore, the thermal stability of the composites using Kevlar fabric was improved by hybridization with basalt fabric. The thermomechanical characteristics of hybrid composites may be altered by changing the stacking order of the reinforcements. Differential scanning calorimetry further established that the hybrid composites with alternate layers of basalt and Kevlar can improve the heat flow rate and enable survivability at extreme temperatures. Such novel hybrid composites can be used for high-load-bearing and high-temperature applications, e.g., defense, aerospace, automotives, and energy applications.

This paper presents a study conducted on prepregs manufactured by a novel method for the impregnation of a thermoplastic matrix. Different composite prepregs based on polypropylene and reinforced with natural fibers (e.g., basalt and jute fibers) were developed. The mechanical and dynamic mechanical properties were investigated. DMA tests were conducted at 1 Hz frequency and properties such as storage modulus and damping (tan δ) were evaluated. The overall mechanical properties of the basalt fiber composites were found to be superior to that of the jute fiber-based samples. Thermo-gravimetric analysis (TG/DTG) of the composite samples showed that the thermal degradation temperatures of the basalt-based composites shifted to higher temperature regions compared to the PP or jute fiber composites. The addition of basalt fiber considerably improved the thermal stability of the composite samples. Microscopic images of the tensile fractured composite samples illustrated better fiber-matrix interfacial interaction due to the novel technology of prepregs. Single-ply and 2-ply prepregs showed significantly superior mechanical, thermal, and thermo-dynamical performance compared to the control sample (pure PP). 2-Ply composites demonstrated higher modulus, tensile strength, and storage modulus due to the higher fiber volume fraction. Basalt-based samples showed a minimum weight loss of about 57% up to 700 °C in contrast to 96.05% weight loss in the jute-based samples and 98.4% in the case of pure PP. The heat resistance index (T HRI ) is more than twice for basalt compared to jute and PP. Furthermore, the superior thermal stability of basalt is reflected in its DSC curves, showing the highest endothermic peak. The technique of using the resin in the form of thermoplastic yarns offers cost effective and efficient alternatives for composite manufacturing.

Current treatment options for cutaneous leishmaniasis are associated with myriad limiting factors including low penetration, poor efficacy, and drug toxicities. Herein, we reported imiquimod and terbinafine co-loaded mannosylated transethosomes (IMQ-TER-MTES) with enhanced cutaneous retention, macrophage targeting, anti-leishmanial potential, and dermal immunomodulation. IMQ-TER-MTES were optimized using Design Expert® followed by their loading into chitosan gel. Moreover, the antileishmanial response against amastigotes-infected macrophages and Leishmania-infected BALB/c mice was evaluated. Finally, the safety and immunomodulation activity of IMQ-TER-MTES gel was performed using BALB/c mice. Optimized IMQ-TER-MTES showed nano-sized particles with low poly-dispersibility index (PDI) and high drug entrapment. Mannosylation has augmented macrophage targeting and the internalization capability of TES. IMQ-TER-MTES showed significantly reduced IC 50 value (19.56 ± 3.62 μg/ml), higher selectivity index (29.24), and synergism against Leishmania major (L. major) amastigotes. In L. major infected BALB/c mice, the cutaneous lesion healing potential of IMQ-TER-MTES was also elevated with reduced lesion size (1.52 ± 0.43 mm). Superior safety of IMQ-TER-MTES was observed in BALB/c mice along with adequate stimulation of dermal immune cells, in contrast to the ALDARA®. Achievement of better comfort properties in station suits of fire fighters without compromising flame retardancy is an utmost necessity. Inherently fire-resistant fibers play vital role in this scope. In this work twenty-three plain single jersey knitted fabric samples were developed by using five inherently fire-resistant fibers. The fibers used were meta-aramid Nomex, fire resistant (FR-Viscose, modacrylic (Protex), FR-polyester (Recron) and carbon fibers. All the fibers were blended in different blend ratios. Vertical flammability test was performed to investigate the flammability properties. Thermal characterization of the samples was done using thermogravimetric analysis (TGA). Thermophysiological and wearing comfort properties of all samples were evaluated. Scanning electron microscopy (SEM) was carried out for the evaluation of surface morphology of the fibers after charring. Results of vertical flammability test revealed that sample containing 100% Nomex fibers produced minimum char length. Nomex and carbon fiber blended fabric provided better moisture management along with better flame retardant behavior. Statistical tool named as Principal Component Analysis (PCA) was utilized for the optimization of all responses. All the samples were ranked as per principal component analysis. Aims: Herein, we investigate the potential of levosulpiride-loaded nanostructured lipid carriers (LEVO-NLCs) for effective brain delivery with anti-psychotic and antidepressant effects., Main Methods: Micro-emulsion method was used to prepare LEVO-NLCs, followed by its optimization using Design Expert®, investigation of the particles properties and entrapment efficiency (%EE). Moreover, in-vitro release, in-vivo plasma and brain kinetic studies of LEVO-NLCs were executed. Anti-psychotic activity of LEVO-NLCs was accomplished in LPS-induced psychosis mice model. Additionally, expressions of neuro inflammatory mediators, neurodegeneration and neuro-inflammation in brain tissues was investigated., Key Findings: The optimized LEVO-NLCs were rounded shaped nanoparticles (157.2 nm) with suitable zeta potential (-29.6 mV), low PDI (0.395) and high EE (83.67 %). No chemical interactions were found, however, the crystalline drug was changed to amorphous. LEVO-NLCs displayed sustained drug release behavior when compared with drug suspension. Moreover, a meaningfully higher AUC (106,642.27 ± 876.44 ng.h/mL) and Cmax (38,534.72 ± 2344.10 ng/mL) of the LEVO-NLCs in brain was observed as compared to the AUC (15,684.33 ± 1005.49 ng.h/mL) and Cmax (7717.56 ± 871.23 ng/mL) of LEVO-Suspension. Similar profiles of both the formulations were perceived in plasma pharmacokinetic studies. Furthermore, LEVO-NLCs exhibited a meaningfully improved anti-psychotic activity in LPS-induced psychosis mice model with reduced immobility time and enhanced struggling time. Likewise, treatment with LEVO-NLCs showed reduced levels of neuro inflammatory markers (p-NF-κB and COX-2) in LPS-induced mice. Cricket is one of the most popular global sports, and cricket pads are important personal protective gear used for shock absorption and peak deceleration of the impact forces of the cricket ball for both batsmen and wicket keepers. The materials selection of the padding should be considered according to requirements. In the present study, flexible composites were manufactured using knitted unidirectional thermoplastic composite prepregs. Prepregs were fabricated using thermoplastic yarns, e.g., High Density Polyethylene (HDPE), Polypropylene (PP), and Low Melting Polyester (LMPE). Para-aramid (Kevlar) and Flax yarns were used as inlay. The structures were stacked in three and five layers, and hot compression was used to convert thermoplastic yarn into matrix. A total of twelve samples were prepared, and their mechanical properties were evaluated. Tensile and flexural properties, short beam strength, and impact properties were optimized using the multi-criteria decision-making (MCDM) technique for order performance by similarity to ideal solution (TOPSIS). This approach was used to select the best material for use in cricket pads. The candidate samples were ranked using statistical techniques. The optimum sample was found to be FP5, i.e., Flax with polypropylene using five layers, which exhibited the maximum impact strength. The results showed that the mechanical properties were improved in general by increasing the number of layers. The significance and percentage contribution of each factor was obtained by ANOVA (α = 0.10) and pie chart, which showed Factors A and C (inlay yarn and number of layers) to be the main contributors. The optimal samples showed superior impact-related performance compared to a market sample cricket pad.

Composites are a promising material for high-specific strength applications; specifically, fiber-reinforced polymer composites (FRPCs) are in the limelight for their extraordinary mechanical properties. Amongst all FRPCs, carbon fiber reinforcements are dominant in the aerospace and automotive industry; however, their high cost poses a great obstacle in commercial-scale manufacturing. To this end, we explored alternate low-cost inorganic fibers such as basalt and rockwool as potential replacements for carbon fiber composites. In addition to fibrous inclusions to polymers, composites were also fabricated with inclusions of their respective particulates formed using ball milling of fibers. Considering automotive applications, composites' mechanical and thermo-mechanical properties were compared for all samples. Regarding mechanical properties, rockwool fiber and basalt fiber composites showed 30.95% and 20.77% higher impact strength than carbon fiber, respectively. In addition, rockwool and basalt fiber composites are less stiff than carbon and can be used in low-end applications in the automotive industry. Moreover, rockwool and basalt fiber composites are more thermally stable than carbon fiber. Thermogravimetric analysis of carbon fiber composites showed 10.10 % and 9.98 % higher weight loss than basalt and rockwool fiber composites, respectively. Apart from better impact and thermal properties, the low cost of rockwool and basalt fibers provides a key advantage to these alternate fibers at the commercial scale.

Herein, Imiquimod (IMQ) was incorporated in nanotransethosomes (nTES) to develop the IMQ-nTES nano-drug delivery system. IMQ-nTES was optimized using 2 3 factorial design. The optimized formulation was expressed with a particle size of 192.4 ± 1.60 nm, Poly-dispersibility of 0.115 ± 0.008, and IMQ percent entrapment efficiency of 91.05 ± 3.22%. Smooth and round morphology of IMQ-nTES vesicles was confirmed by TEM micrographs. Moreover, FTIR results have shown drug-excipient compatibility. The IMQ-nTES was laden inside the low molecular weight chitosan gel, which exhibited easy application, spreadability and no irritation to the applied skin. The release pattern has clearly exhibited improved dissolution properties of IMQ with the provision of the sustain release pattern. Higher IMQ content was deposited in deeper epidermis and dermis with IMQ-nTES gel, in contrast to ALDARA. In vivo, comparative toxicity study on BALB/c mice has shown significantly reduced (p < 0.001) psoriatic area severity index (PASI) score and less increment in ear thickness. Epidermal hyperplasia was an obvious finding with ALDARA which was, providentially, minimal in IMQ-nTES gel-treated skin. FTIR analysis of skin tissue has shown an enhancement of lipid and protein content in the ALDARA group, however, in the IMQ-nTES group no such change was observed. With ALDARA application, CD4 + T-cells and constitutive NF-κβ expression were significantly elevated, in comparison to the IMQ-nTES gel treated group. Moreover, the adequate expression of IFN-γ and cytotoxic CD8 + T-cells were suggesting the preserved IMQ efficacy with IMQ-nTES gel. Quantification of cutaneous as well as systemic inflammatory markers has also suggested the reduced psoriatic potential of IMQ-nTES gel. Herein, Trifluralin (TFL) laden transfersomes (TFS) were investigated against Cutaneous Leishmaniasis (CL), via localized and targeted dermal delivery of TFL. Designed TFL-TFS were optimized utilizing 2 3 full factorial design on the basis of desired response factors including Particle size (P.S), Polydispersity index (PDI), TFL entrapment (%EE) and deformability index (DI). Optimized formulation was found to display P.S of 140.3 ± 2.3, PDI of 0.006 ± 0.002, %EE of 86 ± 0.5 and 43.5 ± 1.0 DI. Results of TEM and XRD analysis have shown intact spherical structure of TFL-TFS and alteration in TFL crystallinity, respectively. Moreover, the optimized TFL-TFS were loaded in Carbopol-940 gel to attain protracted skin retention. TFL-TFS were found to exhibit sustain TFL release profile for up to 24 h. Ameliorated skin permeation of TFL-TFS, even in absence of permeation enhancers, has shown its suitability for cutaneous application. Macrophage uptake assay demonstrated higher intracellular penetration, evidenced by intense reddish fluorescence of rhodamine loaded TFS in comparison to rhodamine-solution. In vitro anti-leishmanial assessment was showing 2.86-folds and 3.07-folds decrement in IC 50 -value of TFL-TFS against L. tropica KWH23 amastigotes and promastigotes, respectively.
