Hydrogel beads containing purified, crude extract and commercial bromelain preparations were developed using encapsulation technique with two carrier agents, sodium alginate and gum Arabic in various ratios of 2:0, 1.5:0.5 and 1:1 (w:w). Stability assessment of bromelain-loaded hydrogel beads was conducted using in vitro gastrointestinal digestion models. Beads produced with 2 % (w/v) sodium alginate (S2) and 1.5 % (w/v) sodium alginate combined with 0.5 % (w/v) of gum Arabic (SG1.5) demonstrated high production yield in a range of 90.99 %–95.92 % and loading capacity in a range of 76.03 %–76.97 %. All bead samples showed high encapsulation efficiency in a range of 93.71 %–98.49 % with spherical shapes. The formulation of 1.0 % (w/v) sodium alginate combined with 1.0 % (w/v) gum Arabic (SG1) resulted in a higher wall thickness. SEM image revealed numerous small crystals on the surface of SG1.5 and SG1, which were formed by crude extract and commercial bromelain. Crude bromelain extract-loaded beads exhibited optimal protein release across pH 3–7 at 25 °C, with enhanced release following thermal treatment. Encapsulation in SG1 resulted in the highest protein release (105.38 %) and antioxidant activity (36.41 mmol Trolox/g bead for FRAP, 6.34 mmol Trolox/g bead for ABTS) after intestinal digestion. Conversely, the encapsulated purified bromelain yielded the lowest release of protein and antioxidant activity after simulated digestion due to the structural formation between bromelain and wall materials. The hydrogel beads have potential to enhance the stability and bioaccessibility of bromelains.

Freshwater crab shells (Somanniathelphusa dugasti) are abundant in inland areas and rich in chitosan, which can be extracted and utilized. This study investigated the characteristics of chitosan from freshwater crab shells (FC-chitosan) and its impact on extending the shelf life of Nam Dok Mai mangoes (Mangifera indica L.). The FC-chitosan had a high degree of deacetylation (99.15 %), thereby enhancing solubility and reactivity for a uniform coating, and a low ash content (0.75 %), minimizing the interference effects on mechanical strength and transparency, both of which could contribute to its potential use as a well-adhering coating material. The FC-chitosan film exhibited superior properties as a coating film, with relatively higher transparency (25.09 % T/mm), tensile strength (21.57 MPa), and elongation (76.09 %) compared to commercially available marine crab chitosan. Mango fruits coated with 2.0 % (w/v) FC-chitosan demonstrated appropriate postharvest quality, with lower weight loss and maintained peel color, fruit firmness, titratable acidity, and ascorbic acid during storage at room temperature (30 ± 5 °C) for up to 11 days. The results suggest that FC-chitosan is effective as a coating material for delaying ripening and extending the shelf life of mangoes. The novelty of using chitosan derived from freshwater crab shells lies in its unique properties and its sustainable use of underutilized byproducts.

This study aimed to determine the effectiveness of a professional development program on the use of a STEM-based 5E Inquiry Learning Model in enhancing the learning of STEM-based Inquiry Learning Model among 78 in- and pre-service science teachers. The topic electric circuit was used as the context of the study. Given the aim and the exploratory nature of this study, a triangulation mixed-methods design was employed: One-group pretest-posttest design which involved a single group that was pre-tested, exposed to the STEM-based 5E Inquiry Learning Model, and post-tested. This was triangulated by the qualitative responses from the participants' reflections. The analysis of the quantitative dataset using the paired samples t-test indicated that the post-test mean score was statistically significantly higher than the pre-test mean score and that the effect size obtained was educationally significant. Further analyses by gender and participants' teacher status, indicated a non-significant difference, giving credence that this result was truly the outcome of the intervention (treatment) effect instead of any disguised effect. The analysis of qualitative data through a recursive process resulted in the crystallization of three overarching key themes: Enhanced knowledge on the content used, empowered pedagogical skills on STEM-based Inquiry Learning, and heightened classroom management skill on promoting collaboration in the classroom.

Aims: To influence educational psychology practice by sharing key themes from research and work capturing young peoples' experiences of engaging with youth offending services (YOS). Rationale: The prioritisation of the voice of the child in educational psychology practice is central to inclusivity (Davie, 1993; Messiou, 2002). Professionals capture and respond to the voices of children and young people, with varying degrees of authenticity, accountability and longevity (Hart, 1992; Hart et al., 2004). It is important that adults facilitate and support meaningful participation (Hart, 2008) by employing appropriate methods to enable young people to share their narratives. This paper seeks to influence educational psychology practice through the authentic voice of young people; focusing on what they say works when they are engaging with youth offending services, and highlighting accessible methods used to elicit voices. Method: The author discusses their experience of using free associative methods; the Grid Elaboration Method (GEM) (Joffe & Elsey, 2014) and the Free Association Narrative Interview (FANI) (Hollway & Jefferson, 2008) as part of their qualitative doctoral research with five young people recruited through a local authority YOS, answering the research question 'What do participants talk about when asked about their engagement experiences with youth offending services?' The author describes how these approaches facilitated accessible means to hearing young peoples' voices, capturing their views through codes and themes illuminated through thematic analysis. Prioritisation of child voice during this formative experience inspired a co-production project creating a whiteboard video describing young peoples' experience of working with a YOS. It provided space, voice, audience and influence to young people involved (Lundy, 2007) and through it, an accessible resource exists for those who may engage with YOS in the future. Limitations: The small number of participants recruited for the original research study (five) and the co-production project in YOS (six), impedes generalisability. Further limitations relate to the interpretivist nature of the original piece of research and following co-production project. The psycho-social approach, inherently acknowledges the subjective nature of analysis undertaken, suggesting that each individual researcher may analyse the data from a subjective perspective and therefore may come to different themes than those illuminated by this researcher. Implications and discussion: Themes that emerged through the script and visual imagery during the co-production project are linked back to key themes from the author's original research. Key considerations for educational psychologists working in YOS are shared, focusing on the role educational psychologists can play in supporting young peoples' meaningful engagement through supporting relationship and identity development and the structural processes that serve psychological functions. Proposals for educational psychology roles in YOS that provide consultative and supervisory support as well as in strategic shaping of services are also shared.

This paper examines the teaching patterns of experts employed in the information technology (IT) industry. The objective was to study the teaching patterns that are used to develop new IT workers in organizations. Thematic content analysis was conducted on interviews with thirty-four IT professionals working in IT and non-IT industries with experience of mentoring new workers. The study investigated the use of adult-learning theory in four teaching patterns: time-based, function-based, technical-task-based, and case-based. Indications for the applications of the various patterns are described in detail. The researchers suggest the use of the patterns identified to optimize the process of enhancing the ability of IT students involved in work-integrated learning projects.

Abstract Poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) [P(3HB-co-3HHx)] is a bacterial copolymer in the polyhydroxyalkanoates (PHAs) family, a next-generation bioplastic. Our research team recently engineered a newly P(3HB-co-3HHx)-producing bacterial strain, Cupriavidus necator PHB−4/pBBR_CnPro-phaC Rp . This strain can produce P(3HB-co-2 mol% 3HHx) using crude palm kernel oil (CPKO) as a sole carbon substrate. However, the improvement of P(3HB-co-3HHx) copolymer production by this strain has not been studied so far. Thus, this study aims to enhance the production of P(3HB-co-3HHx) copolymers containing higher 3HHx monomer compositions using response surface methodology (RSM). Three significant factors for P(3HB-co-3HHx) copolymers production, i.e., CPKO concentration, sodium hexanoate concentration, and cultivation time, were studied in the flask scale. As a result, a maximum of 3.6 ± 0.4 g/L of P(3HB-co-3HHx) with 4 mol% 3HHx compositions was obtained using the RSM optimized condition. Likewise, the higher 3HHx monomer composition (5 mol%) was obtained when scaling up the fermentation in a 10L-stirrer bioreactor. Furthermore, the produced polymer's properties were similar to marketable P(3HB-co-3HHx), making this polymer suitable for a wide range of applications.

This paper presents research related to the work-integrated learning (WIL) model. The objectives were to explore the characteristics and the techniques associated with it. The focus of this research was on model linked to the IT industry or IT department in non-IT industries. A systematic literature review was applied as research methodology and the collected data were from 2006 to 2016 from eight databases. There were 24 articles which matched the search criteria. The WIL models reported can be separated into four groups. The two key features are activities created by universities and activities offered by industry. The model techniques are knowledge, methods and tools for acquiring research problems and developing solutions. The existing models are very broad and need to be narrowed down so that prospective careers and the needs of the digital workforce are prioritized. Researchers suggest future trends in WIL models in the digital economy.

In this study, the performance of a near-infrared (NIR) fiber-optic probe coupled with stability competitive adaptive reweighted sampling (SCARS) was investigated for the analysis of acetic acid, ethanol, total soluble solids, caffeic acid, gallic acid, and tannic acid in the broth of pineapple vinegar during fermentation. The NIR spectra of the broth samples in the region of 11,536–3956 cm−1 were collected during vinegar fermentation promoted by Acetobacter aceti. This continuous biological process led to changes in the concentrations of all analytes studied. SCARS provided optimized and stabilized NIR spectral variables for the construction of a partial least squares (PLS) model for each analyte using a small number of optimal variables (under 88 variables). The SCARS-PLS model outperformed the conventional PLS model, and achieved excellent accuracy in accordance with ISO 12099:2017 for the four prediction models of acetic acid, ethanol, caffeic acid, and gallic acid, with root-mean-square error of prediction values of 0.137%, 0.178%, 0.637 μg/mL and 0.640 μg/mL, respectively. In contrast, only an acetic acid content prediction model constructed via the conventional PLS method and using the whole spectral region (949 variables) could pass with acceptable accuracy. These results indicate that the NIR optical probe coupled with SCARS is an appropriate method for the continuous monitoring of multianalytes during vinegar fermentation, particularly acetic acid and ethanol contents, which are indicators of the finished fermentation of pineapple vinegar.

This research aims to explore on quality assessment and develop a participatory knowledge management (KM) approach through a competency development model based on the concepts of "CommonKADS", needs assessment, and CIPP model. Data were collected by the use of questionnaires, interview formats, and focus group discussion items for the model's empirical test. They were analyzed by frequency distribution, mean and standard deviation, t-test, and data analysis. The approach's participatory characteristic was determined for the stakeholders, followed by its components, consisting of the input and process factors with technical conditions and driving mechanism. After their learning needs were assessed and prioritized, a model was developed as a conceptually formulated pattern of a KM's functional guidance with five dimensions-language competencies, coordination techniques, planning and administration competency, teamwork commitment and academic competency. The findings indicated that (1) an empirical test of the approach was conducted with the foreign affairs officers and university lecturers successfully and satisfactorily, (2) the key informant experts verified that the appropriateness of this participatory approach to knowledge management. This approach was confirmed as a series of continuing KM procedures involving the stakeholders' physical and mental participation in creating, thinking, planning, and making the decision.

Bromelain is a mixture of proteolytic enzymes derived from pineapple (Ananas comosus) fruit and stem possessing several beneficial properties, particularly anti-inflammatory activity. However, the molecular mechanisms underlying the anti-inflammatory effects of bromelain are unclear. This study investigated the anti-inflammatory effects and inhibitory molecular mechanisms of crude and purified rhizome bromelains on lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 macrophage cells. RAW264.7 cells were pre-treated with various concentrations of crude bromelain (CB) or purified bromelain (PB), and then treated with LPS. The production levels of pro-inflammatory cytokines and mediators, including nitric oxide (NO), interleukin (IL)-6, and tumor necrosis factor (TNF)- were determined by Griess and ELISA assays. The expressions of inducible nitric oxide synthetase (iNOS), cyclooxygenase (COX)-2, nuclear factor kappa B (NF-B), and mitogen-activated protein kinases (MAPKs)-signaling pathway-related proteins were examined by western blot analysis. The pre-treatment of bromelain dose-dependently reduced LPS-induced pro-inflammatory cytokines and mediators, which correlated with downregulation of iNOS and COX-2 expressions. The inhibitory potency of PB was stronger than that of CB. PB also suppressed phosphorylated NF-B (p65), nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor alpha, extracellular signal-regulated kinases, c-Jun amino-terminal kinases, and p38 proteins in LPS-treated cells. PB then exhibited potent anti-inflammatory effects on LPS-induced inflammatory responses in RAW264.7 cells by inhibiting the NF-B and MAPKs-signaling pathways.

Natural compounds represent the great capability to stimulate several cell types. Macrophage plays an important role for an effective immune response for infection and inflammation. Isoquinoline alkaloid, sanguinarine, and chelidonine are active compounds that exhibit activity on various tumor cells and immune cells. However, the effect of these compounds on the endosomal toll-like receptor (enTLR) and type I interferon (IFN) are still unclear. The monocyte-derived macrophages (MDMs) were cultured and were determined their cell viability and phagocytic activity to Staphylococcus aureus DMST8840. The nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression were also examined. The expression of enTLRs, type I IFN, and cytokines were determined by real-time PCR. Result shows that the compounds did not affect on MDM cell viability. Sanguinarine and chelidonine enhance phagocytic activity of MDM against Staphylococcus aureus DMST8840 by revealing a higher number of bacterial survival than the MDM treated by polyI:C, and the cell control after co-culture for 3 h. The production of NO has no difference amount but iNOS mRNA production was down-regulated in sanguinarine, chelidonine and their mixed treated MDM. The expressions of enTLRs and IFN-β1 mRNA were up-regulated in both compounds and their combination. Additionally, these compounds also enhance M1-liked cytokine by up-regulated IL-6 and down-regulated IL-10 and TGF-β1, respectively. Therefore, sanguinarine and chelidonine enhance enTLR and IFN-β1 expression and trend to stimulate the cell into M1-liked MDM.

Pineapples are a tropical fruit with high nutritional value and high vitamin and sugar contents. In this study, low-grade pineapples were fermented to produce vinegar using surface culture fermentation (SCF), which involved the addition of dragon fruit juice, to compare the quality and antioxidant activity of different preparations of vinegar. The highest acetic acid concentration (7.35%) was obtained from pineapple vinegar after 20 days of incubation. Vinegar made from mixed pineapple and dragon fruit juice without peel and vinegar with pineapple and dragon fruit juice with peel had acetic acid concentrations of up to 6.20% and 4.50%, respectively. The mixed-fruit vinegar of pineapple and dragon fruit juice with peel displayed the highest antioxidant activity at 210.74 µg/g TE, while no significant difference was found between the other two vinegars (189.52 vs. 187.91 µg/L TE). Notably, the volatile compounds detected in the vinegars were alcohols and esters, which may contribute to the distinct aroma. Overall, the addition of dragon fruit juice with peel to pineapple vinegar increased the phenolic content and antioxidant activity; however, fermentation was slightly slower than that of the other two test materials.

Background: The constructive and specific feedback in guiding long jump athletes to improve their performance in each phase is part of the critical process for achieving desired long jump distance. However, to date, the potential approach for assisting a coach in capturing long jump movement and transferring their knowledge to long jump students is not well-established.Objectives: To investigate the performance of long jump students and evaluate transferring knowledge from coaches to long jump students using a Knowledge-Based Smart Trainer (KBST) System.Methods: Twenty-two participants (fifteen males, mean age = 15.33 ± 1.95 years; seven females, mean age = 14.57 ± 2.07 years) participated in the study. All participants were recruited from eleven sports schools in Thailand. Each participant was instructed to perform the long jump movement, including running, take-off, and landing, for three attempts (Test 1, Test 2, and Test 3). Test 1 was the conventional approach (coaches provided the feedback based on their experience). Test 2 and Test 3 were the KBST system approach (coaches provided the feedback based on the results from KBST system). Two cameras were used to record the participant movement from the starting position to the landing position. The capture data were analyzed by KBST system program. The outcome measures were starting position, maximum velocity, maximum velocity position, and take-off angle. Repeated-Measures ANOVA was conducted to compare the long jump performance across the three trials. The statistical significance was set at p-value < 0.05.Results: There was a statistically significant difference between Test 1 and Test 3 for long jump distance (mean difference = 0.292; Std. Error = 0.129; Sig. = 0.34). However, the mean of take-off angle was similar across the three trials (Test 1 = 12.16°, Test 2 = 12.71°, and Test 3 = 12.95°, respectively).Conclusion: The KBST system was effective in improving long jump students’ performance and also transferring knowledge from the coach to long jump students.

Oil palm empty fruit bunches (EFBs) are an attractive lignocellulosic material that can be used as a cheap renewable feedstock to produce organic acids and many other value-added products. This research is aimed at investigating the potential of steam-exploded oil palm EFBs for the production of fumaric acid, a food additive widely used for flavor and preservation, through a separate hydrolysis and fermentation process using the selected fungal isolate Rhizopus oryzae K20. To develop an efficient method for the recovery and purification of fumaric acid from fermented oil palm EFBs, a two-stage precipitation protocol was employed, followed by an activated carbon-mediated polishing step to remove contaminants. After these two processes were accomplished, a recovery yield of 81.2% and a purity of 83.5% were achieved.

This study used Fourier transform-near-infrared (FT-NIR) spectroscopy equipped with the liquid probe in combination with an efficient wavelength selection method named searching combination moving window partial least squares (SCMWPLS) for the determination of ethanol, total soluble solids, total acidity, and total volatile acid contents in pineapple fruit wine fermentation using Saccharomyces cerevisiae var. burgundy. Two fermentation batches were produced, and the NIR spectral data of the calibration samples in the wavenumber range of 11,536–3952 cm−1 were obtained over ten days of the fermentation period. SCMWPLS coupled with second derivatives searched and optimized spectral intervals containing useful information for building calibration models of four parameters. All models were validated by test samples obtained from an independent fermentation batch. The SCMWPLS models showed better predictions (the lowest value of prediction error and the highest value of residual predictive deviation) with acceptable statistical results (under confidence limits) among the results achieved by using the whole region. The results of this study demonstrated that FT-NIR spectroscopy using a liquid probe coupled with SCMWPLS could select the optimized wavelength regions while reducing spectral points and increasing accuracy for simultaneously monitoring the evolution of four chemical parameters in pineapple fruit wine fermentation.
